Pigs obtained from a mycoplasma-free piggery were randomised into 4 groups of 9. Groups 1 and 2 were injected by the intraperitoneal route with liquid culture of the LKR strain of Mycoplasma hyopneumoniae. Group 1 was injected once and group 2 twice. Group 3 was made up of pigs inoculated by the intranasal route with the virulent Beaufort strain of M. hyopneumoniae; they served as the source of infection for the challenge. Group 4 were uninfected, uninjected controls. Six weeks after the last injection, groups from 1 to 4 were placed in contact. Seven of the pigs in the 1-dose group and 6 in the 2-dose group were free of lesions at necropsy 6 weeks after challenge. Of the two pigs with lesions in the 1-dose group one had only a small lesion but the other had extensive lesions; it had not shown an antibody response after injection of culture. The lesions in the 3 pigs in the 2-dose group were all small. All 9 control pigs had lesions which varied from medium to large in size. The difference in the incidence of pneumonia between the injected and control groups was significant (P less than 0.05) and the proportion of severely affected pigs in the vaccinated groups was significantly lower (P greater than 0.01). There was no difference between those given one dose of vaccine and those receiving 2 doses.  相似文献

The objective of this study was to evaluate the efficacy of a new Mycoplasma hyopneumoniae bacterin against a Korean M. hyopneumoniae challenge under experimental conditions. Fifteen pigs were allocated randomly into 3 groups (5 pigs per group) that were designated in 1 of 3 ways: vaccinated-challenged, unvaccinated-challenged, or unvaccinated-unchallenged. The pigs in the vaccinated-challenged group were immunized with an M. hyopneumoniae whole-cell bacterin at a 1.0 mL dose-level at 21 d old. At 42 d old (0 d post-challenge), the pigs in the vaccinated-challenged and unvaccinated-challenged groups were inoculated intranasally with a strain of Korean M. hyopneumoniae. Vaccinated-challenged pigs elicited a strong cell-mediated immunity as measured by M. hyopneumoniae-specific interferon-γ secreting cells when compared with unvaccinated-challenged pigs. Vaccination of pigs with this new M. hyopneumoniae bacterin reduced nasal shedding and lung lesions. The evaluated vaccine was therefore considered effective in controlling M. hyopneumoniae infection.  相似文献

The aim of this study was to establish whether enzootic pneumonia could be induced reliably in piglets by administering an aerosolised culture of Mycoplasma hyopneumoniae. Groups of five M hyopneumoniaefree Landrace x Large White piglets weaned at 11 to 14 days of age were exposed to aerosols of in vitro cultures of a virulent strain of M hyopneumoniae. In three separate trials, 14 of 15 pigs exposed to the bacteria developed pneumonia, but pigs exposed to the culture medium alone did not develop the disease. Lung pathology, both gross and histological, indicated acute disease. Ten of the pigs were tested for seroconversion by Western blot and they were all positive. The growth rates of the infected pigs were significantly reduced and the water consumption of the infected groups was also depressed. M hyopneumoniae was recovered from eight of the 15 infected pigs.  相似文献

In many diagnostic laboratories the diagnosis of mycoplasmal pneumonia in pigs is based on clinical signs and the presence of gross and histopathological lesions. The objective of this study was to evaluate the nested-PCR technique as an adjunct to the histopathological diagnosis of Mycoplasma hyopneumoniae infection. Respiratory disease of 184 swine cases submitted to the Minnesota Veterinary Diagnostic Laboratory between 1 January and 30 June 1998 were used. Bronchial swabs were collected and the nested-PCR performed. Lung samples were graded PCR positive or negative. Histopathological lesions were scored 0-4, depending on the mycoplasma-like characteristics of the lesions, with category 4 demonstrating strong evidence of mycoplasma infection.Nested-PCR correlated well with histopathological lesions characteristic of M. hyopneumoniae in categories 3 and 4 and approximately half of the histopathological categories 1 and 2 were nested-PCR positive. The results demonstrate that the nested-PCR is a valuable adjunct in the diagnosis of M. hyopneumoniae infection when non-diagnostic microscopic lesions of mycoplasmosis are found.  相似文献

The efficacy of an inactivated aqueous vaccine against Mycoplasma hyopneumoniae was evaluated at two M hyopneumoniae-infected farrow-to-finish commercial farms (A and B) in Greece. In a prospective, randomised double-blind study, two groups on each farm received intramuscular doses of either the vaccine or the adjuvant when they were one and four weeks of age. The pigs were observed daily for clinical signs of disease; morbidity and mortality were recorded; and bodyweight was recorded at intervals. At slaughter, the lungs of the animals were examined and the chest cavities were examined for signs of pleuritis. No adverse reactions to the treatments were observed in any of the pigs. On farm A the vaccinated pigs were on average 6 kg heavier at slaughter, and on farm B they were on average 4 kg heavier; on both farms the average daily gain of the pigs was greater than that of the unvaccinated pigs. The prevalence and severity of enzootic pneumonia in the affected lungs were significantly lower in the vaccinated than in the unvaccinated pigs.  相似文献

Nonspecific mitogenicity of Mycoplasma hyopneumoniae membranes for blood lymphocytes (BL) and bronchial lymph node lymphocytes (LNL) from swine was investigated. Additionally, the influence of respiratory tract exposure to the same membrane preparation on responsiveness of these cells was evaluated. Membranes utilized in lymphocyte transformation tests and for inoculation of swine were prepared by osmotic lysis of mycoplasma cells. Conventionally reared and cesarean-derived, colostrum-deprived swine were given membranes intratracheally and responses of BL and LNL to membranes were assessed from postinoculation day 0 to 14. Utilizing a stimulation index of 3 as the cutoff, heated (56 C) M hyopneumoniae membranes exerted moderate nonspecific stimulation of BL 11 of 12 times when BL were collected from normal (control or preinoculation) swine. Similarly, LNL from conventionally reared and control groups of swine were stimulated nonspecifically 4 of 6 times by the same membrane preparations. Exposure of the respiratory tract to membranes appeared to have no influence on stimulation responses of BL at postinoculation days 6 or 13, whereas moderate to marked increases in responsiveness of LNL were detected when collected at necropsy on postinoculation days 7 or 14. These findings indicated that compartmentalization of lymphocyte sensitization in the bronchial lymph nodes resulted from respiratory tract exposure to mycoplasmal membranes. Background

A field trial was carried out with two Mycoplasma hyopneumoniae vaccines in order to investigate the benefit of vaccination under field conditions in modern Danish pig production facilities with pigs being positive for M. hyopneumoniae. The M. hyopneumoniae infection of the herd was confirmed through blood samples that were positive for antibodies against M. hyopneumoniae combined with gross lesions of the lungs related to M. hyopneumoniae at slaughter and detection of M. hyopneumoniae by polymerace chain reaction in these lesions.

Results

A total of 2,256 pigs from two herds were randomly divided into three groups. Group 1 received 2 mL ThoroVAX®VET, Group 2 received 1 mL Ingelvac®MycoFLEX, and Group 3 was a non-vaccinated control group. The vaccination was performed by a person who was not involved in the rest of the trial and vaccination status thereby blinded to the evaluators.The prevalence of lung lesions related to M. hyopneumoniae were significantly lower for pigs vaccinated with ThoroVAX®VET but not for pigs vaccinated with Ingelvac®MycoFLEX®, when compared to non-vaccinated pigs. There was no significant effect of vaccination on growth rate, antibiotic consumption or mortality.

Conclusion

This trial demonstrated that vaccination with Thoro®VAX VET was effective in reducing the prevalence of lung lesion in pig units infected with M. hyopneumoniae.  相似文献

To facilitate the control of enzootic pneumonia (EP) of swine caused by Mycoplasma hyopneumoniae, the complement fixation (CF) test has been used for the detection of M. hyopneumoniae antibodies. However, the CF test is a cumbersome and time-consuming technique and cross-reactivity are major drawbacks associated with this method. To circumvent these drawbacks, we have developed a double-sandwich enzyme-linked immunosorbent assay (ELISA), consisting of purified monoclonal antibody (Mab) against the 46 kDa surface antigen (P46) of M. hyopneumoniae and recombinant P46 protein expressed in Escherichia coli, for the detection of antibodies to M. hyopneumoniae in serum samples from pigs experimentally inoculated with M. hyopneumoniae and from naturally infected pigs, and compared the practical usefulness of ELISA using the CF test. In experimentally inoculated pigs, the CF and ELISA antibodies were detected at almost the same time, and a good correlation was demonstrated between the CF test and the ELISA. In a survey conducted on field samples, the seropositivity by ELISA in pigs of age 2-6 months was increased. At the time of slaughter, approximately 80% of the animals were seropositive for ELISA. However, a gradual decrease in the prevalence of ELISA positive samples was observed in sows with increasing parity. No correlation was seen between the results obtained with the two methods in the clinical samples. The CF test appears to have limited value for the diagnosis of EP in conventional herds because nonspecific reactions were frequently observed. Therefore, this ELISA is a useful alternative to the CF test currently used for the diagnosis of EP.  相似文献

The aim of the study was to investigate the serological reactions of pregnant sows to vaccination with Hyoresp. Further investigations were performed in the offspring of these sows to follow the dynamics of maternal antibodies and the reaction to vaccination at different points in time. The study was conducted in three farrow-to-finish herds endemically infected with M. hyopneumoniae. A total of 30 gilts and 31 sows were vaccinated 8 and 4 weeks ante partum with Hyoresp (Merial GmbH) or given phys. saline solution as a placebo. The offspring was divided into three groups receiving Hyoresp at 1 and 4 or at 4 and 8 weeks of age. The control group was treated with phys. saline solution at 1 and 4 weeks of age. Before vaccination, antibodies against M. hyopneumoniae were detected in 85% of the gilts and 68% of the sows, confirming the endemic infection of the herds. Vaccination of the sows induced a significant increase in the antibody concentration in serum within four weeks and enhanced the concentration of antibodies in the colostrum. As expected, significantly enhanced levels of antibodies were also detected during the first four weeks of life of the offspring of vaccinated sows. The piglets' serological reaction to vaccination at 1 and 4 weeks of age showed marked interferences with maternal antibodies, so that a reaction could be demonstrated only at 8 weeks of age. The serological reaction of piglets vaccinated at 4 and 8 weeks of age was much stronger than that of piglets vaccinated earlier. Surprisingly, the vaccination status of the sow had no effect on the serological response of the piglets in either vaccination scheme. Maternal antibodies are known to reduce the risk of M. hyopneumoniae infections in piglets. Vaccinating the sows against M. hyopneumoniae may thus be an option for farrowing-to-finish herds with an enhanced risk for infections due to ineffective separation of different age groups, poor gilt acclimatisation or high gilt replacement rates.  相似文献

In this study, several cytokine responses were investigated during Mycoplasma hyopneumoniae (Mhp) infection using a gnotobiotic infection model. We found that several inflammatory cytokines (IL-1beta, IL-8, IL-18, and TNF-alpha) and an anti-inflammatory cytokine IL-10 were induced from peripheral blood mononuclear cells (PBMC) of germ-free (GF) piglets stimulated with heat killed Mhp whole antigens, but no IFN-gamma and IL-4 were induced by Mhp. After the intranasal infection of Mhp, IL-1beta, IL-8, IL-18, and IFN-gamma were also detected in the broncho-alveolar lavage fluids (BALF). The antigen-specific IFN-gamma and IL-10 responses after infection of Mhp were gradually suppressed during Mhp infection as well as non-specific immune response to concanavalin A (ConA) and lipopolysacchalide (LPS) at early stage of infection. These results suggested that Mhp infection modulates the immune response of pigs by inducing several cytokines, and causes immuno-suppression of pigs in a gnotobiotic condition.  相似文献

To gain further insight into the pathogenesis of porcine enzootic pneumonia (PEP), cytokine expression in different pulmonary compartments was examined. Mycoplasma hyopneumoniae (Mh) and proinflammatory and immunoregulatory cytokines (IL-1alpha, IL-1beta, IL-2, IL-4, IL-6, IL-8, IL-10 and TNF-alpha) were detected by immunohistochemical methods in porcine lungs experimentally infected with Mh. Ten pigs were inoculated intranasally with Mh and killed in pairs weekly from 1- to 5-week post-inoculation (wpi). Three Mh-free pigs were taken as controls. Mh-antigen was shown in paraffin-wax-embedded tissues by immunohistochemistry in the luminal surface of bronchial and bronchiolar epithelial cells of all Mh-infected pigs. Significant increase in cytokine expression was detected on snap-frozen tissues from the bronchoalveolar exudate of the airways, mononuclear cells of the alveolar septa and macrophages and lymphocytes of the peribronchial and peribronchiolar lymphoid tissue, from 1 wpi onwards, compared to expression in non-pneumonic lungs. The main cytokines in the BALT of Mh-infected animals that showed an increase were IL-2, IL-4, IL-8, IL-10 and TNF-alpha. In the alveolar septa and bronchoalveolar exudate IL-1 (alpha and beta), IL-2, IL-4, IL-8 and IL-10 expression also increased in infected animals.  相似文献

This study evaluated immune cell populations in pigs following weaning and vaccination for Mycoplasma hyopneumoniae. Piglets (n = 24) were weaned (day 0) at 16 (±1) days of age, and randomly assigned to the vaccination group (n = 16) or control group (n = 8). Complete blood cell counts, flow cytometry and serology were completed for blood samples collected on days 0 (within hours of weaning), 3, 7, 14, 30 and 60. The M. hyopneumoniae S:P ratios (sample optical density: positive control optical density) were negative in the vaccination group until days 30 and 60, when the S:P ratios were 1.3 and 1.0, respectively. Control animals remained serologically negative. The percentage of CD4+ T cells was less (P < 0.01) in control pigs than vaccinated pigs at day 3. In contrast, numbers of CD8+ and CD4+CD8+ T cells were greater (P < 0.01) in control pigs than in vaccinated pigs at days 3 and 7. After day 7, few differences in immune cell types were evident between the groups. Differences in lymphocyte populations could not be solely attributed to vaccination, due at least in part, to the confounding influence of weaning. It was difficult to distinguish the influence of vaccination from the impact of weaning on peripheral immune cell populations.  相似文献

A Mycoplasma hyopneumoniae vaccine (Respisure, Pfizer AH) was tested for its effects on antibody formation, daily weight gain (DWG) in different growing periods, lung lesions and quality of meat (chemical composition, physicochemical properties and fatty acid composition). Two groups of conventional piglets were used for the investigation. One group of 11 females and 11 males was vaccinated intramuscularly at the age of 1 and 3 weeks. The other group of 22 piglets was left nonvaccinated as control. The results showed that antibodies against M. hyopneumoniae in the vaccinated group had been formed 14 days after the second vaccination and remained present till the end of the study at 147 days of age. In the nonvaccinated group, seroconversion started at 49 days of age and by the end of the study 10 out of 22 pigs had become seropositive. Vaccinated pigs achieved significantly higher daily weight gain (+30 g) and finishing body weight (+6.04 kg) than the nonvaccinated animals. In addition, the vaccinated pigs showed lesions involving 3.27% of the lung surface in average, while in the nonvaccinated pigs 9.04% of the lung surface was affected. Investigation of meat quality showed that the longissimus dorsi muscle of vaccinated pigs contained significantly lower percentage of fat (-0.63%) and its tryptophan/hydroxyproline ratio was significantly lower (-23.57) in comparison with the control animals. In addition, some other parameters also showed a favourable tendency, e.g. lean meat percentage was 0.91% higher, the protein content of the longissimus dorsi muscle was 0.35% higher, its water-binding capacity was also higher by 0.78%, its monounsaturated fatty acid concentration was 2.97% lower, while its polyunsaturated fatty acid content was 1.65% higher in the vaccinated pigs than in the nonvaccinated animals.  相似文献
