Sixteen wether lambs (25 kg) were fitted with abomasal infusion cannulas and used to study N and endocrine responses to abomasal infusions of arginine (ARG) or ornithine (ORN). Lambs were randomly allotted to four treatment groups and abomasally infused with solutions of water (CON), ARG, ORN or UREA. The ARG solution provided .50 g ARG.HCl/kg BW and was equimolar with ORN.HCl (.40 g/kg). UREA (.28 g/kg) was isonitrogenous with ARG and served as a positive N control. Lambs were housed in metabolism crates for excreta collection and received 729 g DM/d of a 13.7% CP diet in equal portions four times daily. Following a 7-d dietary adjustment period, lambs were infused continuously (2 liters/d) with water for a 5-d preliminary collection period (Period 1), which immediately preceded a 7-d infusion and collection period (Period 2). Sequential blood samples were taken at 15-min intervals for 8 h between 1200 and 2000 on d 4 of both periods. Single samples were obtained at 1500 on remaining days. Nonrepeated measurements were analyzed as a completely randomized design, whereas repeated measurements were analyzed as a split-plot over time. Period 2 measurements were adjusted using covariance techniques if differences among treatment groups were observed for Period 1. Contrasts used in determining treatment effects were: CON vs UREA, CON vs ARG + ORN, and ARG vs ORN. Nitrogen retention was similar for all treatment, suggesting that dietary N was not limiting. Arginine and ORN increased serum ornithine (P less than .05), blood urea N (BUN; P less than .10) and urinary urea N excretion (P less than .01), whereas ARG increased (P less than .05) serum arginine and UREA increased (P less than .01) BUN and urinary urea N. Serum insulin and glucose were not affected by treatment. Compared with CON, ARG and ORN increased (P less than .05) mean somatotropin (STH) concentration (13.8 vs 16.9 and 18.4 ng/ml) and amplitude of STH pulses (9.8 vs 15.1 and 17.8 ng/ml), whereas CON and UREA were similar. Abomasal infusions of ARG and ORN were equally efficacious in stimulating ovine STH secretion when dietary N intake was not limiting.  相似文献

Short-chain fructooligosaccharides (FOS) were supplemented to the diets of nine quarter horses ranging in age from 489 to 539 d with initial BW averaging 400.6 +/- 21.2 kg. The objectives of this study were to determine the effects of dietary FOS on the fecal responses in terms of pH, the microbial population, and VFA concentrations. The horses were used in a 3 x 3 replicated Latin square design, fed according to NRC requirements, and their individual diets were supplemented with no FOS (CON), 8 g of FOS/d (LOW), or 24 g of FOS/d (HIGH) over three 10-d feeding periods. On the last 3 d of each 10-d feeding period, a single fecal sample was collected between 0730 and 0930. Fecal pH decreased linearly (P = 0.01) from 6.48 with the CON diet to 6.38 with the HIGH diet, but there was no change (P = 0.19 for linear effect) in fecal consistency among treatments. A quadratic effect (P < 0.01) was observed for fecal Escherichia coli population, but no difference (P = 0.88 for linear effect) was found in fecal Lactobacilli enumeration among treatments. The presence of fecal Bifidobacteria was unable to be confirmed and was therefore not reported. Fecal acetate concentrations increased linearly (P = 0.03), with means of 2.13, 2.18, and 2.52 mg/g of wet feces for CON, LOW, and HIGH treatments, respectively. Similarly, fecal propionate concentrations increased linearly (P = 0.01), with means of 0.58, 0.64, and 0.73 mg/g for CON, LOW, and HIGH treatments, respectively. Fecal butyrate concentrations also increased linearly (P = 0.02), with means of 0.40, 0.46, and 0.54 mg/g for CON, LOW, and HIGH treatments, respectively. Total VFA (P = 0.01) and lactate (P = 0.02) concentrations increased linearly, with total VFA means of 3.47, 3.69, and 4.25 mg/g for CON, LOW, and HIGH treatments, respectively, and lactate means of 0.36, 0.41, and 0.47 mg/g for CON, LOW, and HIGH treatments, respectively. Supplementing FOS in diets fed to yearling horses altered fecal microbial populations, fecal VFA concentrations, and pH.  相似文献

Crossbred heifers (n = 120; 265 kg, SD = 37) were fed individually (84 d) to determine the effect of supplement type, concentration, and frequency on intake and performance and to estimate the energy value of dry distillers grains plus solubles (DDGS) in a high-forage diet. Treatments were arranged in a 3 x 2 x 2 factorial, with 3 supplements, 2 concentrations, and 2 frequencies of supplementation. Supplements including dry-rolled corn (DRC), DRC with corn gluten meal (DRC + CGM), and DDGS were fed at 0.21% (LOW) or 0.81% (HIGH) of BW daily and were provided daily (DAILY) or 3 times weekly (ALT). Heifers were fed to consume grass hay (8.7% CP) ad libitum. Individual DMI, diet composition, BW, and ADG were used to calculate energy values for DDGS and DRC. Supplement type, concentration, frequency, and interactions were tested using the MIXED procedure of SAS, with BW included as a covariate. Supplement x concentration interactions for gain (P = 0.01) and G:F (P < 0.01) were detected. The effects of dietary energy intake and somatotropin (STH) on growth and puberty were studied in 40 Angus heifers. At an average age of 7 mo (208 +/- 8 d), heifers were assigned to four treatment groups: 1) vehicle (V) + high energy (HE; 2.68 Mcal ME/kg DM), 2) recombinant DNA-derived STH (20.6 mg/d; s.c.) + HE, 3) V + low energy (LE; 2.22 Mcal ME/kg DM) or 4) STH + LE. Animals remained on treatments until 15.5 mo of age. Body weights (BW), hip heights (HH) and areas of pelvic openings (PA) were measured every 28 d and backfat thicknesses (BF) were measured every 56 d. Plasma progesterone was measured in blood samples taken three times per week beginning at 9 mo of age to determine age at first ovulation. Heifers fed HE were heavier (P less than .01), gained faster (P less than .01) and had greater BF (P less than .01) than those fed LE. Animals treated with STH gained faster (P less than .01) and were heavier (P less than .05) between 12 and 15 mo of age than V-treated heifers. Heifers treated with STH also had less BF (P less than .05) and a tendency for a greater (P = .08) increase in HH than in V-treated heifers. Somatotropin interacted with energy (P less than .05) and age (P less than .01) to influence PA. The effects of somatotropin (STH) and energy intake on serum concentrations of glucose (GLU), insulin (INS), nonesterified fatty acids (NEFA), urea nitrogen (UN) and insulin-like growth factor-I (IGF-I) were determined in 40 Angus heifers. At 7 mo (208 +/- 8 d) of age heifers were assigned to four treatment groups: 1) vehicle (V) + high energy (HE; 2.68 Mcal ME/kg DM), 2) recombinant DNA-derived STH (20.6 mg/d; s.c.) + HE, 3) V + low energy (LE; 2.22 Mcal ME/kg DM) or 4) STH + LE. Animals remained on treatments until an average of 15.5 mo of age. Blood samples were taken every 30 min for 4 h at 9, 11, 13 and 15 mo of age to determine circulating concentrations of metabolites and hormones. Serum IGF-I was increased (P less than .01) by STH injections, but this effect appeared to diminish with age (STH x age; P less than .01). Energy intake did not influence IGF-I levels. Somatotropin increased (P less than .01) serum GLU in heifers fed the HE diet but only tended (P = .08) to increase GLU in those fed the LE diet (STH x energy; P = .05). Although STH increased (P less than .01) serum INS in both energy groups, the response in heifers fed the HE diet was greater (P less than .02) than that in heifers fed the LE diet (STH x energy; P less than .05). Apparent absorption and retention of Ca, P, Mg, K, Cu, Mn, Zn, and S were determined in Holstein heifers treated with recombinant bovine somatotropin (bST). Six heifers (initial BW averaged 104 kg) were assigned to various doses of bST in a 6 x 6 Latin square design with six consecutive 21-d periods. Treatments were daily i.m. injections of 0, 6.7, 33.3, 66.7, 100, or 200 micrograms of bST/kg of BW on d 8 to 21 of each period. All heifers were fed a hay-concentrate diet, and nutrient digestibilities and balances were determined on d 14 to 20 of each period. Mineral concentrations in feed, water, excreta, and blood plasma were determined with an inductively coupled argon plasma-emission spectrometer. Exogenous bST did not affect apparent absorption or retention of Ca, P, Mg, K, Cu, Mn, or Zn. Plasma CU concentration was greater (P less than .05) in heifers receiving bST (82 micrograms of Cu/dl) than in those receiving excipient (73 micrograms of Cu/dl); plasma concentrations of other elements were not affected by bST. Injection of bST did not affect S absorption, but urinary S excretion decreased (P less than .05) and S retention increased (P less than .05) as dose of bST increased. The amount of S retained (1.5 g/d) when the heifers received the largest dose of bST was approximately 50% greater than the amount retained when heifers received excipient. These results demonstrate that bST affects the postabsorptive metabolism of S. Generally, bST did not affect nutrient digestibilities. Effects of bST on dietary mineral requirements were not determined because of the relatively short (14 d) treatment periods.  相似文献

Serum samples and BW were obtained from 2-yr-old beef heifers, pregnancy with either single (SF, n = 12) or twin (TF, n = 7) fetuses, at 7-d intervals from d 190 of gestation until calving. Serum insulin-like growth factor I (IGF-I) concentrations of SF heifers gradually declined from d 190 (69.9 +/- 1.0 ng/ml) to d 263 (55.6 +/- .8 ng/ml), then exhibited a slight increase by d 277 (63.4 +/- 1.1 ng/ml). Serum IGF-I concentrations of TF heifers essentially paralleled, yet were lower (P less than .05) than, concentrations in SF heifers for all days tested except d 197 and 205. The SF heifers pregnant with heifer fetuses (n = 6) had higher IGF-I concentrations (P less than .1) than heifers pregnant with bull fetuses (n = 6) for all days tested except d 214 and 235. Instantaneous absolute growth rate (IGR) of SF heifers declined from 1.485 kg/d at d 190 to .257 kg/d by d 277. Rate of decline in IGR of TF heifers was much greater (P less than .0001). Correlations between serum IGF-I concentrations and IGR for SF and TF heifers were .79 (P less than .001) and .59 (P less than .05), respectively. These data suggest that number and sex of fetus influence maternal concentrations of IGF-I and that the combined growth rate of the dam and conceptus during gestation is related to serum IGF-I concentration.  相似文献

The net release of insulin, glucagon and somatostatin by the portal-drained viscera (PDV) and their net uptake by the liver in response to 3-d abomasal infusions of casein were measured in seven multicatheterized beef steers. The steers were fed 4.3 kg DM/d of a high-concentrate diet in 12 equal meals (13.1 Mcal ME/d and 95 g N/d). In two separate experiments, the abomasal infusion of 300 g casein/d (300C) or 150 g casein/d (150C) was compared to a water infusion. Plasma flow was measured by indicator dilution and net flux by venoarterial concentration difference x plasma flow. Arterial plasma concentrations of insulin were increased (P less than .02) by either 300C or 150C. The 300C increased (P less than .03) PDV insulin release but did not affect hepatic uptake, resulting in an increased (P less than .03) total splanchnic (TSP) insulin flux. The 300C increased (P less than .05) plasma concentrations of glucagon as the result of decreased (P less than .06) hepatic extraction ratio and not as the result of increased portal release. The portal and hepatic flux of somatostatin measured as somatostatin-like immunoreactivity (SLI) were highly variable and not affected by casein infusions. Arterial plasma concentrations of somatomedin-C were not responsive to abomasal casein infusions. The abomasal infusion of 300C resulted in increased plasma concentrations of insulin via increased PDV release and increased plasma glucagon via decreased hepatic extraction ratio.  相似文献

Ruminally cannulated Rambouillet wether lambs were used in three 6 x 6 Latin square experiments (n = 6/experiment) to determine which essential AA limit N retention. Lambs (BW = 36.9 +/- 1.9 kg for Exp. 1, 35.1 +/- 1.4 kg for Exp. 2, and 46.0 +/- 1.3 kg for Exp. 3) were housed in metabolism crates and limit-fed (DMI = approx. 1.8% of BW daily) twice daily a soybean hull-based diet low in ruminally undegradable protein. Treatments for Exp. 1 were continuous abomasal infusions of a solution (500 mL/d) containing 1) no AA (CON), 2) a mixture of 10 essential AA and 2 nonessential AA (10EAA), 3) 10EAA with Met removed, 4) 10EAA with Lys removed, 5) 10EAA with His removed, and 6) 10EAA with Thr removed. Treatments for Exp. 2 were abomasal infusions of 1) CON, 2) 10EAA, 3) 10EAA with Leu, Ile, and Val removed (-BCAA), 4) 10EAA with Arg removed, 5) 10EAA with Phe removed, and 6) 10EAA with Trp removed. Treatments for Exp. 3 were abomasal infusions of 1) CON, 2) 10EAA, 3) -BCAA, 4) 10EAA with Leu removed, 5) 10EAA with Ile removed, and 6) 10EAA with Val removed. All lambs received continuous infusions of acetate and propionate into the rumen and dextrose into the abomasum to supply additional energy. Periods were 7 d: 3 d for adaptation to abomasally infused treatments and 4 d for fecal and urinary collections. Blood samples were collected 3 h after feeding on d 7. In all 3 experiments, N retention was greater (P < 0.10) for lambs receiving 10EAA vs. CON, demonstrating that the basal AA supply from CON was limiting. Removal of each of the essential AA from 10EAA decreased (P < 0.10) their concentrations in plasma (except for Trp), indicating that 10EAA supplied these AA in excess of the animal's requirement. In Exp. 1, N retention (g/d) decreased (P < 0.10) in response to the removal of Met and Thr, but was not affected by removal of Lys and His from 10EAA. In Exp. 2, N retention decreased (P < 0.10) in response to removal of all 3 branched-chain AA, Arg, and Trp, whereas the removal of Phe from 10EAA did not affect N retention. In Exp. 3, N retention decreased (P < 0.10) in response to removal of branched-chain AA and Val, but was not affected by the omission of Leu and Ile from 10EAA. The results of this research demonstrated that Met, Thr, Arg, Trp, and Val limited N retention of lambs fed a diet low in ruminally undegradable protein.  相似文献

The objective of this study was to characterize correlated responses in carcass composition, bone mechanical properties, and meat quality indicators to divergent selection for growth rate. Rabbits from low (LOW) or high (HIGH) lines divergently selected over five generations on 63-d BW and a control group (CONT) were used. Rabbits were slaughtered at an average BW of 2,306 +/- 65 g, corresponding to 63, 58, or 52 d of age in LOW (n = 41), CONT (n = 43), and HIGH (n = 44) groups, respectively. Postweaning ADG and G:F increased (P < 0.001) in the order LOW < CONT < HIGH groups. The lengths of the tibia and femur in rabbit legs decreased (P < 0.001) with increasing growth rate. Tibia and femur bone intrinsic stiffness, as assessed by a three-point flexure test, followed the order (P < 0.001) of HIGH < CONT < LOW groups. At the same BW, HIGH rabbits and CONT rabbits showed many similarities for dressing percent, carcass composition, and color and chemical composition of muscles or meat parts. In contrast, carcass yield and the relative proportion of hind part were 3% greater (P < 0.01) in LOW rabbits than in CONT and HIGH rabbits. The meat-to-bone ratio in the hind leg was 11% greater (P < 0.001) in the LOW group compared with the CONT and HIGH groups. Yellow color index and moisture content in LM, a fast-twitch glycolytic muscle, were lower (P < 0.05) in LOW rabbits than in HIGH rabbits, but ultimate pH, WHC, and cooking loss did not differ between the two growth-selected lines. Ultimate pH in semitendinosus, a mixed slow- and fast-twitch oxidoglycolytic muscle, was less (P < 0.001) in HIGH rabbits than in CONT and LOW rabbits. However, lactate dehydrogenase and isocitrate dehydrogenase activities in this muscle (n = 21 per group), assessed at the time of slaughter as markers of glycolytic and oxidative capacities, respectively, did not differ among groups. Growth rate did not modify mean cross-sectional area and type frequency of myofibers in semitendinosus muscle. Finally, Warner-Bratzler shear force and total energy needed to achieve rupture did not differ among groups in semitendinosus muscle (n = 21 per group), whereas they were the least (P < 0.001) in LM (n = 128) from HIGH rabbits. Results from this study indicate that improvement of growth rate by selection has little effect in rabbit carcass and meat quality traits.  相似文献

The 4 yr productivity of 25% (QF; n = 533) and 50% (HF; n = 531) Finnsheep ewes exposed to either Suffolk or Columbia rams in one of three production systems was monitored to test the effects of system, terminal sire breed, maternal line, and their interactions on annual market lamb production. Ewe lambs and yearlings were randomly assigned to either a high-input accelerated lambing system (HIGH), a high-input annual system (MED), or a low-input annual system (LOW). Nursery facilities were available for weak lambs or those born in triplet or more births for the HIGH and MED but not for the LOW systems. Accelerated lambing protocol required early weaning. Sex-adjusted lamb weaning weights were corrected to within-system mean ages of 42 d for HIGH and 70 d for MED and LOW. The HIGH ewes weaned 1.55 lambs per year compared with 1.46 for MED and 1.18 for LOW (P less than .01). However, because of early weaning, HIGH yielded the lowest weight of weaned lamb per year. The MED ewes weaned 5.9 and 11.1 more kilograms of lamb per year than the LOW and HIGH ewes, respectively (P less than .01). The HIGH system may be economically feasible if young lambs could be inexpensively grown to feeder or market lamb weight. The HIGH ewes did not, however, increase lamb numbers in proportion to increased exposures compared with the annual systems. Breed-group effects for ewe productivity (kilogram of lamb weaned per ewe per year) were consistent across management systems, although some interactions among breed group and system were present for components of productivity. Suffolk rams yielded an advantage of 1.6 kg of weaned lamb per exposure over Columbia rams (P less than .05) due to a 3% better lamb survival (P less than .01) and heavier weaning weight, especially in the LOW system. The HF ewes weaned .1 more lambs per exposure than QF ewes (P less than .01); .06 of the lambs were nursery-reared. However, lambs from HF ewes had a 2% lower survival rate (P less than .05) and were 1.4 kg lighter at weaning (P less than .01), so overall productivity among HF and QF ewes was similar.  相似文献

Eight wether lambs (mean BW = 28.8 kg) with ruminal and abomasal cannulas were assigned to either thermally neutral or high ambient temperature treatments. Within each temperature, lambs were randomly allotted to dietary treatments consisting of a basal diet (60% corn and 24% cottonseed hulls) either with (high; 11.4% CP) or without (control; 10.1% CP) added ruminal escape CP as fish meal and with (high) or without (control) 5% added ruminally inert fat in a 2 x 2 factorial treatment arrangement using a Latin square design. Lambs were fed 606 g of DM/d in each period, which consisted of a 10-d adjustment followed by 6 d of sample collection. High temperature increased (P less than .05) respiration rate, evaporative water loss, and rectal temperature. When compared with controls, lambs fed high escape CP retained more N when exposed to high temperatures (2.8 vs 3.6 g of N/d) and less N at neutral temperatures (3.3 vs 3.1 g of N/d; temperature x escape CP; P less than .05). Retention of N was greater (P less than .05) in lambs fed high than in those fed control fat (3.8 vs 2.7 g/d). Lambs fed high vs control escape CP had greater abomasal feed N flow (percentage of intake) when fed high-fat diets (77.3 vs 56.1%) but similar dietary N flow when fed control fat diets (55.8 vs 54.3%; fat x escape CP; P less than .05).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Forty-one primiparous beef heifers were used over 2 yr to evaluate the effect of prepartum administration of a growth hormone-releasing factor analog (GRF-A) or growth hormone-releasing factor (GRF(1-29)-NH2) on somatotropin (ST), insulin-like growth factor I (IGF-I), milk production, heifer BW, and postpartum (PP) return to ovarian activity. Beginning on d -11 +/- 1 from parturition, heifers were administered (s.c.) GRF-A ([desNH2-Tyr1,D-Ala2,Ala15]GRF(1-29)-NH2, 2.5 micrograms/kg; Yr 1) or GRF(1-29)-NH2 (12.5 micrograms/kg; Yr 2) (GRF; n = 17) or vehicle (CON; n = 24) for seven consecutive days. Blood samples were collected at 20-min intervals from -60 to 300 min from the first and fourth injections. Samples were also collected at 20-min intervals for 6 h on d 25 and 69 +/- 1 PP. Area under the curve of ST (nanograms.minute-1.milliliter-1) was greater (P less than .01) in GRF than in CON heifers (9,671 +/- 677 vs 2,611 +/- 237). Increases in ST after GRF-A or GRF(1-29)-NH2 were similar. On d 25 +/- 1 PP, frequency of ST release (pulses per 6 h) was greater (P less than .01) in CON (3.3 +/- .2) than in GRF (2.1 +/- .2) heifers. Milk production was similar (P greater than .1) for the two treatments. Heifer BW loss from d -16 to 81 after parturition was greater (P less than .01) in GRF (88 +/- 5) than in CON (68 +/- 5) heifers. Postpartum return to ovarian activity (progesterone greater than 1 ng/mL for two consecutive weeks) was delayed (P less than .05) in GRF (97 +/- 14) vs CON (71 +/- 8) heifers. After accounting for variation due to treatment and year, a negative (P less than .02) correlation (r = -.39) was detected between concentrations of IGF-I during the first 30 d PP and PP interval to ovarian activity. These results indicate that prepartum administration of GRF altered the release pattern of ST after parturition and was associated with greater PP BW loss and delayed PP return to ovarian activity in heifers.  相似文献

Changes in net portal and hepatic nutrient flux and oxygen consumption in response to 3-d abomasal casein infusions were studied in seven multicatheterized beef steers. Steers were fed 4.3 kg DM/d of a high-concentrate diet in 12 equal meals. Blood flow (para-aminohippurate dilution) and net flux (venoarterial concentration difference x blood flow) across portal-drained viscera (PDV) and hepatic tissues were measured on d 3 of the abomasal infusions. In two experiments, the response to 300 (300C) and 150 (150C) g casein/d were compared, respectively, to a control water infusion. The 300C increased (P less than .05) arterial blood concentrations of alpha-amino N (AAN), urea N and ammonia; 150C increased (P less than .05) arterial urea N. Urinary urea N excretion was increased (P less than .01) by 300C and 150C. Although 300C increased net PDV release of AAN (P less than .07) and alanine (P less than .10), there was no net change in total splanchnic (TSP) flux due to an increased net hepatic uptake of AAN (P less than .01) and alanine (P less than .05). Net PDV glucose flux was decreased (P less than .05) by 300C, but net hepatic glucose flux was not affected by either level of casein. The 150C increased TSP oxygen consumption (P less than .05) and hepatic oxygen extraction (P less than .10). Approximately 26 and 30% of the casein N infused abomasally appeared in the portal blood as AAN for 150C and 300C, respectively. The sum of net PDV ammonia and AAN fluxes accounted for 47 and 88% of the N infused for 150C and 300C, respectively. These data emphasize the importance of intestinal and liver tissues in regulating the flux of nitrogenous compounds absorbed from the diet.  相似文献

Six ruminally cannulated Holstein steers (initial BW = 189 +/- 11 kg) housed in metabolism crates were used in a 6 x 6 Latin square to study effects of ruminal ammonia load on Leu utilization. All steers received a diet based on soybean hulls (2.7 kg of DM/d), ruminal infusions of 200 g of acetate/d, 200 g of propionate/d, and 50 g of butyrate/d, as well as an abomasal infusion of 300 g of glucose/d to provide energy without increasing microbial protein supply and an abomasal infusion of a mixture (238 g/d) of all essential AA except Leu. Treatments were arranged as a 3 x 2 factorial and included Leu (0, 4, or 8 g/d) infused abomasally and urea (0 or 80 g/d) infused ruminally. Abomasal Leu infusion linearly decreased (P < 0.05) both urinary and fecal N excretions and linearly increased (P < 0.05) retained N, but the decreases in urinary N excretion in response to Leu tended (P = 0.07) to be greater, and the increases in retained N in response to Leu were numerically greater in the presence of the urea infusion. Although urea infusions increased (P < 0.05) plasma urea concentrations, urinary N excretions, and urinary urea excretions, retained N also was increased (P < 0.05). The efficiency of deposition of supplemental Leu ranged from 24 to 43% when steers received 0 or 80 g of urea/d, respectively. Under our experimental conditions, increasing ammonia load improved whole-body protein deposition in growing steers when Leu supply was limiting.  相似文献

This work determined the time necessary to stabilize the decrease in urinary N excretion after initiating continuous i.v. glucose infusion and the quantity of glucose required to maximize N balance in growing wether lambs fed a high-protein diet (21.1% CP, DM basis). In the first experiment, six wethers (30 kg) were used in a 10-d crossover design comparing jugular infusion of glucose (600 kcal gross energy/d) plus saline with saline alone. The second infusion experiment was carried out with six wethers (31 kg) assigned to two 3 x 3 plus extra period latin squares, with glucose infusion rates of 0, 300 and 600, and 0, 450 and 900 kcal/d, respectively. Urinary N decreased (P less than .02) by d 2 of glucose infusion, remained stable to the end of the 10-d infusion period, and returned to the preinfusion level within 2 d after glucose infusion was discontinued. Urinary N decreased (P less than .01) and N balance increased (P less than .005) with an increasing level of glucose infusion through 600 kcal/d. Plasma glucose and insulin were elevated (P less than .05) only by infusion of 900 kcal/d of glucose. Glucose was present in the urine of wethers infused with 900 kcal/d of glucose. Glucose infusion had no effect on diet digestibility, hematocrit or plasma urea N. The level of glucose infused into growing wether lambs that maximized reduction of urinary N and was fully utilized for protein deposition without increasing plasma glucose and insulin was about 12 g.Wk-.75.d-1.  相似文献

Two experiments were conducted to assess the endogenous fraction of purine derivative (PD) excretion, urinary recovery, and intestinal digestibility of purines in Nellore heifers. For both experiments, 8 Nellore heifers fitted with ruminal and abomasal cannulas were allocated to two 4 × 4 Latin squares. The diets were based on corn silage and concentrate (60 and 40% DM basis, respectively); feces and urine samples were obtained by total collection, and abomasal DM flow was estimated using indigestible NDF as an internal marker. In Exp. I, 4 of the 8 heifers (BW 258 ± 20 kg) were also fitted with ileal cannula. The planned treatments were 4 different DMI: 1.2, 1.6, 2.0, and 2.4% of BW (DM basis). The endogenous losses and purine recovery as urinary PD were estimated using linear regression between daily urinary PD excretion (Y) and daily abomasal flow of purine bases (X), expressed in millimoles per kilogram of BW(0.75). In Exp. II, the same 8 Nellore heifers (BW of 296 ± 15 kg) were fed at 1.37% BW (DM basis). The treatments were the infusion of purines (RNA from torula yeast, type VI, Sigma) into the abomasum in increasing amounts (0, 33, 66, and 100 mmol/d). All statistical analyses were performed using the PROC MIXED procedure in SAS. In Exp. I, the DMI range was 1.16 to 1.84% of BW and did not affect (P > 0.05) the apparent RNA digestibility in the small intestine, which had a mean of 75.6%, and a true digestibility of 93.0%. The mean ratio of the N-RNA to the total-N in the ruminal bacteria was 0.137. The daily urinary PD excretion (Y, mmol/kg of BW(0.75)) was a function of RNA flow in the abomasum (X, mmol/kg of BW(0.75)): Y = 0.860X + 0.460, where 0.860 and 0.460 were the PD recovery of purines and the endogenous fraction (in mmol/kg of BW(0.75)), respectively. In Exp. II, the daily urinary PD excretion was a function of RNA flow in the abomasum: Y = 0.741X + 0.301, where 0.741 and 0.301 were the recovery of PD in urine of infused purines and the endogenous losses (in mmol/kg of BW(0.75)), respectively. In conclusion, our data suggest that in Nellore heifers the respective values of endogenous PD excretion (mmol/kg of BW(0.75)), urinary recovery of the purines absorbed in the abomasum, and true digestibility of RNA in the small intestine were 0.30, 0.80, and 0.93.  相似文献

Forty-six beef heifers (16 to 23 mo) of two biological types (small = Red Poll-sired, large = Charolais-sired) were individually fed from d 90 of gestation through parturition to evaluate the effects of nutritional restriction on plasma LH and steroid hormone concentrations. Heifers were allotted to one of two nutritional treatments to achieve a BW reduction (loss, fed at 1% of BW/d) or to maintain initial BW (maintenance, fed 1.5% of BW/d) to parturition. Gonadotropin-releasing hormone (100 micrograms) was injected i.m. three times during gestation (d 130; d 200; d 270) and twice after parturition (d 1 to 14; d 23 to 36). Blood samples were collected at 20-min intervals after GnRH for 4 h. Maternal BW change from d 90 to parturition differed (P less than .01) between loss and maintenance heifers. Mean plasma progesterone concentrations were greater (P less than .05) at d 130 and 270 of gestation in small than in large heifers and were greater (P less than .01) at d 23 to 36 postpartum in maintenance than in loss heifers. Mean concentrations of estrone and estradiol were greater (P less than .05) in large than in small heifers at d 200 of gestation. Mean plasma LH concentrations following GnRH injection were greater (P less than .01) in loss than in maintenance heifers at 200 and 270 d of gestation. Sixty male dairy grain-fed calves, raised from 70 to 223 kg BW in individual crates, were used in a 2 X 2 factorial arrangement to determine the effect of administration of human growth hormone-releasing factor (1-29)NH2 (GRF) and(or) thyrotropin-releasing factor (TRF). Calves received twice-daily s.c. injections of .9% NaCl (control), GRF (5 micrograms/kg BW), TRF (1 micrograms/kg BW) or GRF (5 micrograms/kg BW) plus TRF (1 micrograms/kg GTRF). Average daily gain and days on feed were not affected by treatments, but TRF treatment increased (P less than .05) total intake of dry matter (DM) and feed conversion ratio: 3.00, 3.02, 3.08, and 3.22 kg DM/kg weight gain for control, GRF, TRF, and GTRF, respectively. During two 7-d periods, after 66 and 75 d of treatment, feces and urine were collected from 40 calves (5 per treatment per period). Treatment with GRF increased (P less than .05) digestibility of DM, nitrogen (N), and energy and tended (P less than .20) to increase N retention. At slaughter, withers height was increased (P = .05) by GRF and carcass length was increased (P less than .05) by TRF. Pituitary and liver weights were increased (P less than .05) by TRF. The combination of GRF and TRF slightly increased (P less than .10) protein content and decreased (P less than .05) fat content of the 9-10-11th rib section. After d 1, GRF treatment chronically increased (P less than .05) insulin concentrations and also increased (P less than .10) IGF-I concentrations on d 29 and 57. In summary, chronic treatment with GRF and(or) TRF did not improve growth or efficiency, although GRF increased digestibility of DM, N, and energy and the GRF plus TRF combination resulted in slightly leaner carcasses.  相似文献

Twenty-five wether lambs (34 +/- 0.9 kg) fitted with ruminal and abomasal infusion catheters were used in a completely randomized design to determine the effects of differing proportions of ruminal and abomasal casein infusion on N balance in lambs fed a high-concentrate diet (85% corn grain, 1.6% N; DM basis) for ad libitum intake. Wethers were infused with 0 (control) or 10.4 g/d of N from casein with ruminal:abomasal infusion ratios of 100:0, 67:33, 33:67, or 0:100% over a 14-d period. Feed, orts, feces, and urine were collected over the last 5 d. Total N intake and excretion were greater (P < 0.01) in lambs infused with casein than in controls; however, N retention did not differ in lambs infused with casein compared with controls, suggesting that N requirements were met without casein supplementation. Total N intake and total N excretion did not differ among casein infusion treatments. Urinary N excretion decreased linearly (P = 0.07) with decreasing ruminal infusion of casein. Site of casein infusion quadratically (P = 0.06) influenced N retained (g/d), with the greatest retention observed in the 33:67 ruminal:abomasal infusion treatment. Dry matter intake from feed decreased from 1,183 to 945 g/d (P = 0.02) in lambs infused with casein compared with controls, but apparently digested DM did not differ among treatments. These data indicate that decreasing the ruminal degradability of supplemental protein above that required to maximize N retention results in decreased urinary excretion of N without greatly affecting apparent diet digestion.  相似文献
