Are short-day (SD) conditions inductive or permissive for potato tuberization? This question was addressed using anin vitro culture system. Kinetin induction ofin vitro tuberization in potato stolons cultured in the dark can be partially inhibited by light treatments. Photoperiod required for a major inhibition varied with cultivars: 8 hours for cv. Red Pontiac and 16 hours for cv. Kennebec. Short photoperiods decreased kinetininduced tuberization for all cultivars. Stolons cultured on kinetin-free medium generally did not tuberize, regardless of light treatment. Effects of kinetin and photoperiod on stolon and tuber development are observed. Relationships among kinetin, photoperiod and tuber-inducing stimulus produced under short days are discussed.  相似文献

Tuber resistance toPhytophthora infestans was studied in relation to physiological age in tubers of the cultivars Atzimba, Katahdin, Kennebec, and Sebago. Resistance to race1,2,3,4 was evaluated in freshly harvested tubers, and tubers stored at 5 C for up to 215 days. Atzimba tubers were highly resistant when freshly harvested but became increasingly susceptible as they aged physiologically. Tubers of the other three cultivars were susceptible when freshly harvested and also when aged. Further, 57 tuber selections from among 11 crosses initially gave resistant reactions. However, after storage for six months at 5 C, 26.3% of the selections remained resistant, 22.8% were moderately resistant, 31.6% moderately susceptible, and 19.3% fully susceptible.  相似文献

Coumarin readily stimulates tuberization of cultured axillary shoots obtained from etiolated potato sprouts. Adequate concentrations of coumarin must be continuously present in the medium to effect tuber initiation. High nitrogen, gibberellic acid, abscisic acid, indole acetic acid, or naphthalene acetic acid reduced or inhibited coumarin-induced tuberization. Gibberellic acid and high concentrations of nitrogen in medium inhibited the uptake of coumarin-3-14C by the. axillary shoot. Based on the data of physiological activities by coumarin in other plant species and our data on tuberization, we propose that coumarin may represent in part the unknown inhibitor responsible for tuber initiation with reference to the inhibitor/gibberellic acid ratio theory of tuberization.  相似文献

PVX infected plantlets from two potato cultivars grownin vitro with 0.3, 3, 30 or 300 ppm kinetin were exposed to temperatures of 28 or 35 C. After 3 wk, axillary buds were isolated and grown aseptically in organogenic media, followed by PVX testing by ELISA. The serological test was also run on whole plantlets at the end of the kinetin-temperature exposure. No donor plants exposed to 28 C nor the plantlets derived from their buds gave an ELISA (-) reaction, regardless of the kinetin content of the media or that of the cultivar. At 35 C the virus was suppressed to undetectable levels in several whole plantlets. In the cultivar Alpha, 2 out of 6 resulting plantlets after isolation of buds were virus-free in the presence of 3.0 mg/1 kinetin during the treatments. From Atzimba, about 15–40% of the regenerated plants were ELISA (-), without any relationship to the cytokinin content in the media. Heat had a stronger influence on virus elimination than kinetin and the results varied with the cultivar.  相似文献

Potato spindle tuber viroid (PSTV) caused albinism in the new growth of Rutgers tomato under continuous light. The albinism response was obtained with 4 isolates causing severe symptoms in tomato and with 1 mild isolate which is normally symptomless. Rutgers tomato inoculated with the 4 severe strains of PSTV developed typical ‘bunchy top’ symptoms in 12 to 16-hr day lengths. Symptoms developed slower and much less distinctly on plants incubated in 6-hr days. New growth in PSTV-infected plants was green in short days (12-hr illumination) and chlorotic to white in continuous light. Infectivities of extracts from white portions of plants were from 3 to 10 times greater than those from green portions of infected plants. Albino symptoms also developed in other tomato varieties when inoculated with PSTV and kept under continuous light. Albinism was most pronounced under continuous light at 30°C, somewhat less intense at 24°, and at 16°, new growth of infected leaves and stems remained green. Albinism did not develop in otherSolanum species which are symptomless hosts of PSTV nor in seedling potato plants grown in continuous light.  相似文献

The photoperiodic behaviour of 18 potato clones from three taxonomic groups (Andigena, Phureja, and Tuberosum diploid and tetraploid) was studied under controlled environments. Day lengths of 11, 13, 15, and 17 hours were provided with all other environmental conditions common. Six characters were studied: tuber formation, stem height, haulm weight, tuber number, tuber weight, and tuberization efficiency (tuber weight/haulm weight). All clones showed well defined critical daylength values at which tuberization was either absent or irregular. A wide variability for critical day length was observed among clones, the higher values corresponding to Tuberosum tetraploid and the lower values to the Phureja group. Stem height and haulm weight increased as photoperiod increased. Tuber number remained practically the same for all groups but Phureja which showed an inverse relationship. Tuber weight decreased as daylength increased for the groups Andigena, Phureja, and Tuberosum diploid which on the average showed a short day reaction for tuber yield. Tuberosum tetraploid did not show any significant change, behaving as a day neutral type. Tuberization efficiency was, in general, decreased as daylength increased suggesting a short day behaviour for all groups. A regression analysis of tuber yield and tuberization efficiency on photoperiod indicated that, in general, tetraploid genotypes were less sensitive to drastic increases in daylength than diploids.  相似文献

Curing of Sebago potatoes for 10 days at 25°C prior to common storage at 5°C, reduced the responsiveness of tubers to photoinduced glycoalkaloid synthesis. Continuous illumination with 15 and 25-Watt incandescent light for 10 days increased glycoalkaloid content of peelings (12–14% of tuber weight) in uncured potatoes by a factor of 3.2 and 2.8, respectively, while the corresponding factor for cured tubers was only 1.8 for both lights. The peeled tuber portion (86–88% of tuber weight) had negligible amounts of glycoalkaloids, averaging about 1 mg per 100 g of fresh weight. The rise of glycoalkaloid levels in peels of uncured tubers was nearly linear to 164.7 mg/100g (15W light) with no indication of levelling off. In peels of cured tubers, the rise began only after the 4th day of light exposure with an apparent maximum and levelling off at 94.7 mg or approximately 43% lower than the final TGA levels in uncured tubers.  相似文献

Plants from 6 potato cultivars uniformly infected with potato virus X (PVX) were exposed to darkness and gradual temperature increases from 37 to 40 C, during a two-week period. This was followed by shoot tip culture to obtain virus free individuals. The procedure was effective in obtaining PVX-free propagation material from 4 of the 6 varieties. Differences were observed in the response of the cultivars to the heat and to thein vitro culture. At least one plant from every cultivar showed repeated negative serological reaction against PVX after the tissue culture regeneration, except Atzimba and Montsama. Tollocan and Murca were the most resistant to heat, developed better duringin vitro culture and resulted in the largest number of PVX-negative plants. Rosita was intermediate, and Juanita did not adapt to heat stress.  相似文献

The response of diploid potatoes to growing seasons of high temperatures was studied at two locations in eastern North Carolina. The responses measured were survival, tuberization, and yield. One cycle of phenotypic recurrent selection for yield resulted in increases of 3% for survival, 15% for tuberization, and 27% for yield. Heritability of yield, estimated from selection response, was 0.07 for the combined locations. There was a small positive phenotypic correlation between the responses.  相似文献

It is mandatory to assess the allergenic potential of genetically modified (GM) crops before their commercialization. Recently, a transgene [Calcineurin B-like (CBL) protein] has been introduced into tobacco plant to make the crop salt resistance. Therefore, it was felt necessary to assess the allergenic potential of the cbl gene product, which was introduced and expressed in Nicotiana tabacum (tobacco) plant and compared the allergenic effects with the wild-type (WT) counterpart. Bioinformatic analysis revealed that there was no significant sequence homology with known allergens. Also, no difference between the protein digestibility profiles of GM and WT tobacco was found. Rapid digestion of CBL protein (Mol Wt 35 kDa) by simulated gastric fluid (SGF) indicated reduced chances of this protein to induce allergenicity. In addition, BALB/c mice sensitized by intraperitoneal administration of WT and GM tobacco protein showed comparable levels of clinical score, specific IgE, IgG1, histamine level, similar effect on different organs as well as IgE binding proteins. These findings indicate that insertion of cbl gene in tobacco did not cause any additional allergic risk to consumer and the GM and native tobacco proteins behave similarly in both in vitro and in vivo situations even after genetic modification.  相似文献

Gibberellins have been shown to be involved in such important processes as seed germination, flowering, maturation and tuberization in potato. Parents fromSolanum tuberosum Groups Andigena and Tuberosum were found to produce progeny containing dark green, rosette dwarf individuals which could be completely restored to normal appearance by exogenous GA3. Test crosses indicated that this phenomenon can be explained by the action of a single locus, the dwarfing characteristic being conferred by the nulliplex condition. The gene symbol “ga1” is proposed. Group Andigena dwarfs demonstrated much greater tuberization compared to their normal full sibs or dwarfs restored to normal phenotype by exogenous GA3. The possibility that such gibberellin mutants may be involved in the long day adaptation of Gp. Tuberosum is discussed. This character may also be a useful tool for investigating various aspects of the gibberellin status of the potato.  相似文献

In this paper, the surface of magnetic manganese ferrite nanoparticles (MFN) was modified using cetyl trimethylammonium bromide (CTAB). The modified MFN was studied using Fourier transform infrared spectroscopy (FTIR). The adsorption capacity of surface modified MFN (MFN-CTAB) was investigated for dye removal for single and ternary systems. Three anionic dyes, C.I. Direct Red 80 (DR80), C.I. Direct Red 31 (DR31), and C.I. Acid Blue 92 (AB92), were used as model compounds. The effects of operational parameters on dye removal (i.e. adsorbent dosage, dye concentration and salt) and the kinetic and isotherm of dye adsorption were studied. The adsorption kinetic for the dyes was found to be well described by the pseudo-second order model. The maximum dye adsorption capacity (Q 0) of MFN-CTAB for DR80, DR31 and AB92 was 83 mg/g, 59 mg/g and 70 mg/g, respectively. The adsorption isotherm data were analyzed using the Langmuir, Freundlich, and Temkin equations. The results revealed that the Langmuir model fitted the adsorption data better. The results showed that the MFN-CTAB as a magnetic adsorbent might be a suitable alternative to remove dyes from colored aqueous solutions.  相似文献

Field experiments were designed to subject Superior potato plants to various levels of defoliation by the Colorado potato beetle (CPB)Leptinotarsa decemlineata (Say), Coleoptera: Chrysomelidae. Defoliation occurred during each of five consecutive plant growth periods and CPB population, plant leaf area, and tuber weight data were recorded for each period. A visual defoliation rating scheme provided an accurate estimate of actual potato plant leaf area of defoliated plants. Data generated from regression analysis demonstrated a significant dependence of leaf area on CPB numbers per plant during some plant growth periods, but numbers of CPB accounted for very little of the total variation in tuber weight. Plant leaf area was the most important independent variable in the tuber weight regression model.  相似文献

4′-Geranyloxyferulic (GOFA) and boropinic acid have been discovered during the last decade as interesting phytochemicals having valuable pharmacological effects as cancer chemopreventive, anti-inflammatory, neuroprotective, and anti-Helicobacter pylori agents. A reverse-phase HPLC-UV/Vis method for the separation and quantification of the title oxyprenylated ferulic acid derivatives in extracts obtained from peels of nine edible Citrus and Fortunella fruits was successfully applied. Concentration values showed a great variation between the different species, being orange (C. sinensis) the fruit richest in GOFA (0.141?±?0.011 mg/g of exocarp fresh weight) and kumquat (Fortunella japonica) the one in which boropinic acid was recorded as the most abundant phytochemical (0.206?±?0.002 mg/g of exocarp fresh weight). Both secondary metabolites were not detected only in three species. The set-up methodology showed limits of quantification (LOQ) values, that were able to selectively quantify both GOFA and boropinic acid. Results described herein depict a potential chemopreventive dietary feeding role for the Rutaceae spp. under investigation.  相似文献

Potato leafroll virus (PLRV) was purified from potato foliage and stems with an average yield of 0.14 mg of PLRV/kg of potato. Modifications of an existing purification procedure are reported. Five low dosage (38-118 μg of PLRV) intravenous injections were used to produce a PLRV antiserum for use in enzyme-linked immunosorbent assays (ELISA) from tubers. PLRV was readily detected in ELISA testing of potato tubers and leaves and inPhysalis floridana Rybd. Non-specific reactions were low with all tissues. In parallel tests, a Canadian antiserum produced higher nonspecific reactions with tuber and leaf tissue. The results indicated that the use of low dosage-intravenous injections might be necessary methodology for producing PLRV antiserum for use in ELISA diagnostic tests with tuber tissue where high non-specific reactions have been reported.  相似文献

Potato virus A (PVA) was purified fromNicandra physaloides by a simple method that omitted organic solvent clarification and consisted of differential centrifugation followed by equilibrium centrifugation in CsCl. An antiserum was produced that specifically detected PVA in potato leaf sap using either the SDS-agar test or enzyme-linked immunosorbent assay (ELISA). No heterlogous reaction of the antiserum with potato virus Y was detected. Purified PVA was highly infectious; it had an A 258/280 nm absorbance ratio of 1.28. The particles had a normal intact appearance in the electron microscope. Detection of PVA in potato sprouts and foliage by ELISA was compared with the local lesion assay onPhysalis angulata L. plants. ELISA was superior over an indicator plant test when sprout tissue was used. PVA antiserum reacted similarly with mild and crinkle isolates.  相似文献

The growth and tuberization of Norland potatoes were studied under five different temperatures and two photoperiods. Treatment levels included 12, 16, 20, 24, and 28 C with either a 24-h (continuous light) or a 12-h photoperiod at 400 μmol m-2 s-1 PPF. Plants were grown in 6-liter containers and harvested at 56-days-age. Stem length increased with increasing temperature under both photoperiods. The highest tuber yield occurred at 16 C under the 24-h photoperiod (755 g/plant) and at 20 C under the 12-h photoperiod (460 g/plant). Little or no tuber formation occurred at 28 C under either photoperiod or at 24 C under continuous light. As with tuber yield, the highest total plant dry weights also occurred at 16 C under the 24-h photoperiod and at 20 C under the 12-h photoperiod. Harvest index (tuber dry weight to total dry weight ratio) decreased with increasing temperatures and with continuous light. Results indicate that good growth and tuberization can occur under continuous light, and that increasing the photoperiod from 12 to 24 h effectively decreased the optimal temperature for tuber formation from near 20 C to 16 C. Alternatively, the results imply that at cooler temperatures, the potato becomes less obligate for dark period stimulation of tuberization.  相似文献

A total of 23 isolates obtained from scab infected potato tubers representative of six sampling areas in Eastern and Central Canada and five ATCCStreptomyces strains were screened for pathogenicity on the basis of their ability to initiate scab development on aseptically cultured minitubers and plant generated tubers. The results were then correlated with any associated generation of the scab inducing phytotoxin, Thaxtomin A. In all cases a positive correlation was demonstrated between the pathogenicity of variousStreptomyces scabies isolates and their ability to produce the phytotoxin.  相似文献

The potential of the flowering tobacco,Nicotiana alata, to sustain the survival and the development of the Colorado potato beetle (CPB) was assessed. We made food preference, larval development and rearing tests in relation with other plant species, specially with potato. In spite of a clear preference of the CPB for potato, the insects could complete a full life cycle on the flowering tobacco. In comparison with CPBs on potato, those on the flowering tobacco had a slight delay of two days in their development, beginning when larvae reached their tenth day. CPBs on the flowering tobacco had a much lower survival rate (14.0% against 66.7%) and adults were smaller (100.9 mg against 133.0 mg). Nevertheless, the flowering tobacco is the firstNicotiana species which is an acceptable alternative host for the CPB, and could facilitate the survival and the distribution of the insect.  相似文献
