The purpose of this study was to determine the effects which dihydroquercetin (DHQ) has on the natural healing process of suberization and wound periderm initiation in cut seed potatoes. Dihydroquercetin is a phenolic compound extracted from the bark of Douglas Fir,Pseudotsuga menziesii (Mirb.) Franco. Anatomical studies of treated seed potato tissue showed that treatment with bark-383, the cork-rich fraction of the whole bark containing approximately 2% pure DHQ, increases the thickness of the suberized layer formed below the cut surface. Treatment with 4% bark fraction I, which is blended to that concentration with the inert carrier dust Pyrax and contains approximately the same amount of DHQ as does the bark-383, also promoted the formation of a thicker layer of suberized cells. The thickness of the wound periderm did not significantly vary as a result of the use of any treatments containing DHQ. The use of Captan (Orthocide 50W) as a cut seed treatment in these studies was found to inhibit the formation of a suberized layer and subsequently the initiation of a wound periderm. The combination of bark-383 and Orthocide 50W in a treatment mixture reversed the inhibitory affect of the Orthocide 50W alone and therefore allowed for the formation of a well suberized layer and a wound periderm. It was found that wound healing on the cut surface of seed potatoes is largely dependent on the type of tissue present at any given point on that cut surface. Therefore, the response to treatment of the stolon end, center, and blossom end tissues was related to the type of tissue which was present in each of those tuber regions. The fungal mold growth on treated seed pieces incubated in a controlled environment was adequately retarded when Orthocide 50W was combined with bark-383 in a mixture ratio of 1:9 respectively. The longer the treatment material was allowed to remain on the seed piece, the greater was the protection against mold growth afforded by the mixture.  相似文献

Under environmental conditions favorable for wound healing, potato tubers became significantly (P = 0.05) more resistant toA. solani infection during a two-day interval between wounding and inoculation. This period coincided with the time required for wounded tubers to completely seal wounded surfaces by suberization at 15.6°C/98% RH. Suberization of the wounded surfaces provided tubers significant (P = 0.05) protection againstA. solani invasion but wound periderm development was also essential for total exclusion of the pathogen. Wound healing takes place faster at high temperatures and relative humidities. The effect of temperature is more critical in the wound healing process.  相似文献

A survey of selected potato soils in Maine was made to determine the species and populations ofFusarium spp. present.Fusarium solani ‘Coeruleum’ was most often isolated butFusarium roseum ‘Sambucinum’ was not recovered from any soil. Crops or cropping sequences in general showed no direct relationship to observed Fusaria populations or tuber dry rot. Field studies were conducted to determine the effect ofFusarium contaminated seed on soil and daughter tuber contamination. Whole and cut seed were inoculated withFusarium roseum ‘Sambucinum’ and half of each lot treated with thiabendazole (1500 ppm) to controlFusarium seed decay. Soil populations ofF. roseum ‘Sambucinum’ were higher near plants produced from non-treated, cut seed tubers. No increase inFusarium populations was found between plants or near plants produced from whole or thiabendazole treated seed. Daughter tuber contamination was greatest from plants produced from non-treated contaminated cut seed, and lowest from whole or treated seed. Whole non-treated seed produced daughter tubers with contamination equal to treated whole seed. This indicates that the use of whole seed could be a method to reduce daughter tuber contamination and reduce reliance on chemical treatments. Cut seed contaminated withFusarium spp. is an important source of daughter tuber contamination which can result in a high amount of tuber dry rot in storage.  相似文献

A study was conducted to determine the feasibility and advantages of wound-healing potatoes at lower than recommended temperatures. The progress of wound-healing at 6, 9, and 16°C, of tissue taken from Russet Burbank potato tubers, was evaluated by inducing weight loss with high temperature or decay organisms following various curing intervals. Increasing the wound-healing temperature resulted in a greater resistance to weight loss and a decreased susceptibility to infection byErwinia carotovora subsp.carotovora. Resistance to weight loss increased as wound-healing temperature increased following 6 and 16 day intervals. However, weight loss induced after 21 days of healing appeared to level off at 9°C indicating maximum development of a wound barrier. Loss due to rot, after inoculation of fresh cores withE. carotovora, was directly proportional to incubation time and temperature. When wound-healing progress was measured by loss of fresh weight induced by elevated temperatures or rot, the advantages of woundhealing at 16°C compared with 9°C decreased significantly after 21 days of curing. Wound-healing at approximately 9°C for 25 days was found to be the best compromise temperature for keeping rot progress and weight loss at a minimum while allowing the process of healing and maturation to occur.  相似文献

The post-harvest application of chlorine dioxide (ClO2) was evaluated as a disease suppressant for stored potatoes. Chlorine dioxide was prepared by acidifying a buffered sodium chlorite solution with a food grade acid.In vitro studies verified the effectiveness of C1O2 at low concentrations (ED50 = 2 to 122 ppm) againstErwinia carotovora (soft rot),Fusarium spp. (dry rot) andHelminthosporium solani (silver scurf). Evaluations of tubers inoculated withPhytophthora infestans (late blight) andFusarium spp. or infected withH. solani and then treated with ClO2 either going into storage or through the humidification system resulted in a lack of disease suppression. Inconsistent performance of C1O2 in storage appeared to be a result of several contributing factors. Chlorine dioxide concentrations varied greatly (up to six-fold), depending upon the method of activating and diluting sodium chlorite solutions. Chlorine dioxide is a gas soluble in water and, therefore, is easily released from solution (25% –75% loss) into the air when applied as an aqueous spray. Chlorine dioxide reacts quickly with the tuber and associated organic matter, thereby reducing the effectiveness. Applying higher than currently registered rates may be necessary to achieve measurable disease suppression.  相似文献

Progeny tubers from seed potatoes originating from either a traditional clonal selection method or the more modernin vitro tissue culture method of propagation were tested for storage rot response following inoculation of three tuber sites with twoFusarium species. Significant differences were found among disease responses for the twoFusarium species and for the four cultivars tested. Disease symptoms were less severe forF. solani var.coeruleum than forF. sambucinum. Fusarium sambucinum caused less disease in Superior than Kennebec, Russet Burbank and Sebago. Seed propagation method did not significantly affect disease response except for one case;in vitro culture of Kennebec had less disease due toF. sambucinum at the tuber side inoculation site than clonal selection. Kennebec and Superior inoculatedF. sambucinum had significant differences with respect to the number of years of field propagation for the tuber side inoculation site and for the tuber mean rot index. Significant differences in number of years of field propagation were also found for eye-ends of Superior tubers and stem ends of Russet Burbank tubers inoculated withF. solani var.coeruleum.  相似文献

Potato (Solanum tuberosum L. cv. Russet Burbank) tubers were harvested weekly over a 6 week period from non-water-stressed and water-stressed plants. Discs were cut from fresh tubers and from tubers stored for 10 days at 4°C to determine their wound healing ability as evaluated by disc resistance to water loss. Neither harvest date nor water status significantly influenced wound healing while storage significantly increased wound healing ability. Enhanced wound healing associated with storage was accompanied by reduced variability of disc resistance to water loss and increased levels of suberin as reflected by the diol and alcohol components. This work suggests that plant senescence and harvest date are not good indicators of the potential wound healing ability of tubers. Physiological evidence is presented to support the practice of short periods of storage following harvest to improve tuber wound healing ability and thereby reduce shipping and handling losses due to injury.  相似文献

Manipulation of storage environment to provide favorable conditions for wound healing immediately after harvest reducedAlternaria solani tuber infection. Prestorage of tubers at 15.6°C for three weeks before final storage at 10°C normally maintained for commercial storage of potato chip stocks resulted in fewer and smaller lesions than constant storage at 10°C or 4.4°C or prestorage for varying periods at these temperatures. Prestorage of potatoes at a low temperature (4.4°C) for 1–3 months resulted in moreA. solani infection suggesting that low temperatures may inhibit the wound healing process more than they reduce rates ofA. solani infection or disease development. Summary Expression of the pectate lyase (PL) isoenzyme 3 in transgenic potato lines of cv. Désirée mediates an enhanced resistance of tuber tissue toErwinia carotovora (Ec) soft rot due to a pre-activation of plant defence mechanisms. Therefore, theSolanum tuberosum cvs Agave and Adretta with a moderate level of soft rot resistance were crossed with such PL-expressing potato lines. The resulting progenies were assessed with respect to plant/tuber characteristics over a period of four years and then tested for PL3-expression as well as for soft rot resistance. 71% of the selected progeny lines exhibited a stable production of the PL3 enzyme. Statistical analysis revealed differences between the transgenic and the non-transgenic progeny concerning the soft rot resistance of tuber tissue. Compared with the PL-inactive progeny, extension of Ec-rotting on the wound surface of PL-transgenic potatoes was diminished on average by 51.8%. Similarly, the degree of cell lysis caused by bacterial maceration was significantly (P<0.05) reduced in tuber tissue of PL-expressing progeny lines. The latter also revealed an enhanced PPO and PAL activity in their tuber tissue indicating an active plant defence. It is concluded therefore that the PL-mediated soft rot resistance introduced into potatoes by means of molecular techniques is heritable.  相似文献

In a histological study comparing tuber resistance of potato clone B7200-33 and susceptible variety “Russet Burbank” toFusarium roseum ‘Sambucinum’, the formation of suberin and wound periderm was found to be an important resistance mechanism. In B7200-33 tubers a continuous suberin layer was formed whereas in ‘Russet Burbank’ tubers the suberin was formed in clumps so that the pathogen easily could invade the tissue between the clumps but not through a continuous layer. Wound periderm also was formed at a more rapid rate in B7200-33 than in ‘Russet Burbank’ tubers thereby “walling off” the pathogen and reducing decay.  相似文献

The wound response within damaged potato tubers (Solanum tuberosum L.) must be a coordination of suberization with other resistance responses if infection is to be avoided. Previously, we showed that wound healing was affected by wound severity and consequently the type of tissue damaged within the tuber. Using arachidonic acid-induced accumulation of rishitin as a model for phytoalexin accumulation, we now demonstrate that this tuber resistance response is also influenced by wound severity and the type of tissue exposed to the elicitor. Kennebec and Reddale tubers that were superficially wounded by removing a thin tissue slice (0.75 mm thick) from the surface and then treated with the elicitor arachidonic acid produced significantly less rishitin than more severely wounded tubers (e.g., tubers cut in half) and excised tuber tissue disks (17 mm × 4 mm). Excised, elicitortreated tuber tissues accumulated significantly more rishitin in cortical cells than in perimedullary and pith cells. Rishitin accumulation was routinely measured 96 h after wounding and was found to be declining by 144 h regardless of wound severity. Induction of rishitin accumulation was very localized with more than 80 % of the rishitin found within 0.75 mm of the treated wound. These results indicate that excised tuber tissue disks, often used as models in wound research, are not fully representative of intact tubers. Perhaps more important, the results show that superficial wounds of intact tubers, i.e., similar to shallow nick and abrasion type wounds typically incurred during harvest, accumulate very little rishitin. However, rishitin accumulation in the tuber disk model system is much greater than that found in superficially wounded tubers, but is similar to the accumulation that could be elicited in cut (seed) tubers. The complications contributed by these wound-related and tissue-specific interactions must be factored into the model system(s) used in describing the role(s) of phytoalexins in the broad framework of disease resistance for stored potatoes and cut seed.  相似文献

Enrichment techniques and serological strain typing methods were used to determine the source of stem and progeny tuber populations ofErwinia carotovora (Ec) in 1980 and 1981. Plots were established in a field that had been planted with potatoes since 1972. Seed tubers of cultivars Russet Burbank and Norgold Russet were assayed forErwinia carotovora pv.carotovora (Ecc) andErwinia carotovora pv.atroseptica (Eca) by tuber peeling and direct plating and plating following enrichment in a pectate-based broth. Levels of seed tuber infestation ranged from undetectable to 2.4 × 104 colony-forming units per g peel tissue. Prior to planting Ecc was detected in soil samples from field plots by enrichment in a pectate-based broth. Ec was isolated from symptomless plants with increasing frequencies from 8 weeks following planting regardless of the level of Ec populations in seed tubers. At harvest progeny tuber recontamination was low and not related to levels of seed tuber Ec populations. In 1980, serological typing of representative Ecc isolates indicated serogroups III, XXIX, and unknown strains isolated from the soil but not the seed tubers prior to planting were isolated from stems and progeny tubers. Serogroup XXIX was isolated from soil, seed, stems and progeny tubers in 1981. Throughout both years, Ecc was isolated more frequently than Eca from seed, soil, stems, and progeny tubers. These data emphasize the potential for reinfestation of seed stocks and infection of plants by populations of Ecc that overwinter in soil under Wisconsin conditions.  相似文献

Summary Green crop lifting (GCL) for haulm killing was developed in The Netherlands and offers ideal conditions for controlling the blackleg pathogenErwinia carotovora subsp.atroseptica (Eca) by antagonists. Based on the use of mini-tubers or young tubers from field crops, two bioassays for wound protection were developed. GCL was simulated by artificially skinning or wounding tubers, inoculating the damaged skin with Eca, treating with antagonists and incubating in either potting compost or outside in field soil. Mainly fluorescent pseudomonads, pre-screened for in vitro antagonistic activity on agar and high soft rot reducing ability on tuber slices, were tested in the mini-tuber bioassay. Strains giving the highest degree of wound protection were further tested individually and in combination under field conditions in the young tuber bioassay. One individual strain and two combinations, resulting in reduction of contamination levels on skinned surfaces of 85% and between 60% and 70%, respectively, show good potential for biological control of blackleg.  相似文献

A corky patch syndrome that effects potato tuber tissue may be a later manifestation of pink eye disease of potato although an actual cause and effect relationship between the two diseases has not been firmly established. The causal agent for pink eye of potato is unknown but the disease has been associated with the fungiVerticillium andRhizoctonia and the bacteriumPseudomonas fluorescens. Pink eye and corky patch symptoms on the periderm of affected tubers are accompanied by a band of tissue beneath that is intensely auto-fluorescent when viewed under ultraviolet illumination. Studies to determine the cause of the fluorescent reaction were performed using HPLC and a fluorescence monitor. Three phenylpropanoid compounds, chlorogenic acid, esculin and scopoletin, were found to be involved. It appears that these compounds are produced by tuber tissues in response to pathogen invasion and also as part of the wound healing process. Tissues with corky patch syndrome, silver scurf and root-knot nematode contained high concentrations of chlorogenic acid and esculin and low concentrations of scopoletin. Tissues infected with dry rot were visibly less fluorescent, and contained less chlorogenic acid and esculin, and no detectable level of scopoletin.  相似文献

A method has been devised for assessing the potential hazard of soft rot development in potatoes. Tubers are placed in a mist chamber at 20 C (68F), a film of water is maintained on the tuber, and the extent of decay is assessed after 5 days. The soft rot potential determined in this way is intended as apractical indication of potential storage hazard, reflecting the overall effect of the various factors leading to bacterial decay. Tubers that had passed through a flume system and a rinsing spray in a commercial packing plant had a greater decay potential, even after passage through a hot-air dryer, than unwashed tubers. Differences in decay potential could be demonstrated between tubers that had been mechanically harvested and loaded into storage bins and those that had been harvested manually. In addition to the effect of mechanical damage and of passage through flumes, the decay potential was probably affected by the inoculum level ofErwinia carotovora. Pectolytic clostidia were also usually present in tubers rotting in mist chambers, and are probably involved in the decay.  相似文献

Summary Phenolics, for example chlorogenic, caffeic and ferulic acid, and phytoalexins, such as rishitin and phytuberin, were identified in potato tubers cv. Kufri Chandramukhi. The tissue of healthy tubers contained no detectable phytoalexins but did contain phenolics. The levels of these compounds were correlated with soft rot development. The rotting tissue either was free of these groups of compounds or had low concentrations. The wound periderm formed as a result of recovery from injury and infection contained high levels of the compounds. Much higher concentrations were detected at lower storage temperatures when oxygen supply was adequate. Antibacterial properties of the phenolics identified were tested againstErwinia carotovora which was inhibited by chlorogenic, caffeic and ferulic acids. The three phenolics were more effective together, in proportions in which they occurred in wound periderm, than individually. It was observed that none of these phenolics could inhibit pectolytic enzymes ofE. carotovora.  相似文献

The largest difference in the rate of wound healing (suberization and periderm development) of cut tubers was attributable to the difference in temperatures (15.5 vs 23.8 C) tubers were stored at after cutting. Lesser differences were found between the Red Pontiac and Norchief cultivars and between the various regions of the tuber. The least difference was found between the different sources of Norchief. Wound healing occurred most rapidly at 23.8 C, in Red Pontiac and in the cortical region of the tuber. It seems probable that a major cause of the high incidence of seed piece decay and poor stands found in Norchief is due to slow wound healing.  相似文献

The effects of the dormancy releasing chemicals, Rindite and bromoethane, on subsequent tuber response to dry rot were evaluated. Seed tubers of the cultivars Sebago and Acadia Russet were treated with Rindite or bromoethane, and subsequently inoculated with sterile distilled water or spore suspensions ofFusarium sambucinum Fuckel f. 6 Wollenw. After storage for 1 or 11/2 months, tubers were rated for dry rot and dormancy release. Development of seed tuber breakdown byFusarium was not significantly affected by pretreatment with Rindite or bromoethane regardless of any effect on tuber dormancy.  相似文献

研究了巴西橡胶树(Hevea brasiliensis)芽接树的萌条受机械伤害时外施茉莉酸(JA)对2种防卫结构创伤周皮和乳管的形成的影响。结果表明:(1)割伤幼茎树皮直至形成层或刮伤幼茎树皮去掉表皮和2-3层皮层细胞,都能在6d后看到诱导维管形成层分化出次生乳管。在伤口上施用JA或JA生物合成的前体亚麻醉(LA),能够使伤害诱导的乳管增加;而施JA生物合成的抑制剂indomethacin(IND)则产生抑制伤害诱导乳管分化的作用。(2)割伤或刮伤都能诱导形成创伤周皮,而在伤口施用JA和IND,JA可能不参与诱导创伤周皮的形成。(3)施用乙烯利和6-苄基腺嘌呤(BA)都不能引起创伤周皮形成,但BA能促进伤害诱导的创伤周皮细胞层数目增加。  相似文献

Summary The simplest method of inducing tubers to decay in order to detect contamination by the blackleg organism,Erwinia carotovora var.atroseptica, was to wound them and place them in a mist chamber for four days. Incubating the decayed tubers in air for a further two days improved detection of the blackleg organism by immunofluorescence, which was a more sensitive test than the double diffusion method.  相似文献
