One- to six-month-old calves, housed in the large-capacity calf house of the Nová Bystrice state farm in the district of Jindrich?v Hradec were studied for the occurrence of coccidia in the period from January 1974 to April 1975. The coccidia found in these animals included nine species of the genus Eimeria: E. zuernii, E. bovis, E. ellipsoidalis, E. subspherica, E. auburnensis. The species E. alabamensis, E. cylindrica, E. wyomingensis and E. bukidnonensis occurred just sporadically. Further, the findings included oocysts of Isopora spp. The oocysts of the coccidia E. wyomingensis and Isopora spp. were found for the first time in Czechoslovakia. The overall extensity of invasion (EI) and intensity of invasion (II) grow with age as follows: (chart: see text) The results suggest that even modern large-capacity calf houses are not free from invasion by coccidia; the calves in the house under study were invaded by ten species. Transition to vegetable feeding is accompanied by a marked rise of the EI of all the species of coccidia found in the animals.  相似文献

Over the years 1983 to 1984 two-hundred and sixty-five bulls were examined during mass occurrence of bovine cysticercosis in one farm; out of this number 67 animals (25.3%) were confirmed autopsically as infected with cysticercosis. Counterimmunoelectrophoresis was used to examine the serum of slaughtered animals by help of two antigens--Taenia saginata and Taenia crassiceps. With T. saginata the sensitivity amounted to 44.8% and the specificity to 86.4%, with T. crassiceps to 28.3% and to 89.9%.  相似文献

A cross-sectional study was conducted from October 2005 to April 2006 on bovine cysticercosis in cattle slaughtered at Awassa municipal abattoir with the objective of determining the prevalence of Taenia saginata cysticercosis, cyst viability, distribution and its public health implication. Questionnaire survey involving 120 respondents was also conducted on human taeniasis. A total of 400 carcasses were examined during the study period, of which 105 (26.25%) were infected with T. saginata metacestodes. From a total of 3200 samples inspected, 500 cysticerci were detected in 141 samples, of which 221 (44.2%) were alive. The anatomical distribution of cysticerci were 65 (29.2%) heart, 56 (25.3%) shoulder muscle, 59 (26.7%) masseter, 23 (10.4%) tongue, 12 (5.4%) diaphragm, three (1.4%) liver, two (0.9%) lung and one (0.5%) kidney samples. The prevalence varied significantly between local and crossbred animals (OR = 3.15, P < 0.05), but not varied between sex, age groups and origin of the animals. T. saginata taeniasis was a widespread public health problem in the town with an overall prevalence of 64.2% (77 of 120). The potential risk factors for disease contraction were raw meat consumption, religion and occupational risks. In conclusion, the study revealed high prevalence of T. saginata metacestodes throughout the edible organs together with existence of deep-rooted tradition of raw meat consumption. This may magnify the public health hazards of T. saginata in the study area. As a result, the disease deserves due attention to safeguard the public health and further promote beef industry in the country.  相似文献

Serum IgG response of cattle with cysticercosis caused by Taenia saginata was studied in an enzyme-linked immunosorbent assay (ELISA) where a T. saginata metacestode surface extract was used as antigen. In experimentally infected calves, a sharp rise in specific antibody levels was found 3-4 weeks after the infection followed by a logical level of detection corresponded to about 25 cysts. The ELISA was employed in cattle herds where cysticercosis outbreaks had occurred and also in supposedly uninfected herds. Significantly increased antibody levels were found in the herds with massive cysticercosis cases. The test was not adapted for individual diagnosis as some animals of the uninfected herds, especially within the older age groups, had elevated antibody values. The ELISA was, however, useful in the investigation of outbreaks to determine the extent and pattern of the infection in the herd. The rate of decline in antibody levels in these herds was studied by follow up sampling. The increased antibody levels in the infected herds were also reflected in colostrum-fed calves. This observation was employed to estimate the time of infection.  相似文献

A sero-epidemiological study of Taenia saginata cysticercosis was carried out in adult cattle in Zambia to determine the prevalence and study the influence of the farming system on the infection rate. Serum samples were examined for circulating parasite antigen by a monoclonal-based sandwich ELISA. Thirty-eight of 628 serum samples were found positive (prevalence 6.1%). Cysticercosis was significantly more prevalent in feedlots and in traditional farming systems than in dairy farms. It is suggested that the continuous man to animal contact and the use of casual workers in feedlots may be factors that are conductive to T. saginata transmission.  相似文献

In spite of the EU directives that regulate meat inspection for bovine cysticercosis, Taenia saginata is still present in Europe and causes economic losses due to condemnation, refrigeration and downgrading of infected carcasses. The main reasons for this persistence include the low sensitivity of current meat inspection protocols, the dissemination and survival of eggs in the environment and cattle husbandry systems, which allow grazing on pastures and drinking from water streams. It is assumed that water streams and surface water are potentially contaminated with T. saginata eggs. Furthermore, current wastewater management not only fails to halt, but rather contributes to the dissemination of eggs in the environment. Here, the authors discuss an integrated approach for control of this food-borne zoonosis, as well as the potential use of serological methods as a way of improving detection of bovine cysticercosis.  相似文献

The recombinant vaccines that have been developed against cysticercosis and hydatidosis in sheep and cattle are remarkable for their effectiveness and are prominent as examples of the very few non-living vaccines against parasitic diseases. Their development has been through practical application of molecular parasitology, utilising immunochemical techniques in antigen identification and recombinant DNA methods in antigen production. This brief overview discusses the contribution of molecular techniques to the successful development of recombinant vaccines against Taenia ovis, Taenia saginata and Echinococcus granulosus as well as the immunological and genomic studies that have arisen from their development.  相似文献

The larval stage (syn Cysticercus bovis) of the human tapeworm Taenia saginata causes cysticercosis in cattle, which has both aesthetic and food safety implications to consumers of beef. A monoclonal antibody-based immunohistochemical (IHC) assay developed to improve postmortem diagnosis of this parasite and a standard histological method were assessed to determine their fitness for intended use. Sections from 169 known-positive specimens of T. saginata from experimentally or naturally infected cattle, and from 30 known-negative specimens and lesions of various etiologies from non-infected cattle, were tested. The IHC assay identified significantly more known positive bovine cysticerci than the histological method (91.7% and 38.5%, respectively). Positive IHC staining occurred on sections from other cestode species, but should not affect the diagnostic specificity of this assay for bovine cysticercosis, due to the different host and/or tissue preferences amongst these parasites. Use of the IHC assay should improve the reliability of diagnosing lesions caused by degenerated cysticerci, facilitating more effective and efficient control of bovine cysticercosis.  相似文献

Immunity in Taeniids is predominantly antibody mediated and thus many serological immuno-determinants will have potential in both protection and diagnosis. The antigenicity of six peptides derived from four potentially protective molecules cloned from a Taenia saginata oncospheres cDNA library have been evaluated as targets for the specific diagnosis of bovine cysticercosis. The six peptides consist of: two peptides (HP6-2 and HP6-3) derived from the sequence of the 18 kDa surface/secreted oncospheral adhesion antigen identified by McAb-HP6, two peptides (Ts45W-1 and Ts45W-5) derived from the sequence of the T. saginata homologue of the T. ovis 45W protective gene family, one peptide (TS45S-10) derived from a T. saginata sequence with significant similarity to the T. ovis 45S protective antigen, and one peptide (TEG-1) derived from the sequence of the T. saginata homologue of Echinococcus spp. main surface protein. Longitudinal studies indicate that T. saginata infected cattle respond to all six peptides by 3-4 weeks post-infection and that the antibody levels remain high for at least 12 weeks post-infection. As protection against Taeniid parasites is predominantly antibody mediated, some of these six peptides may be of value as immuno-prophylactic tools and hence also in assays to determine resistance to infection with the parasite. For diagnosis, on the other hand, only three peptides (HP6-2, TEG-1 and Ts45S-10) performed with the necessary sensitivity and specificity to determine exposure to infection with T. saginata, and now merit an exhaustive evaluation prior to employment as routine diagnostic tools.  相似文献

The diagnostic value of intradermal tests was verified on 126 bulls (out of this number 32 autopsically positive) subject to intravital diagnostics of bovine cysticercosis. As allergen were used homologous antigens from adult larvae of Taenia saginata and heterologous antigens from larvae of Taenia crassiceps, as well as hydatid fluid from Echinococcus granulosis. With the antigen T. saginata the sensitivity reached 53.1% and specificity 90.4%, with the antigen T. crassiceps 53.1% and 80.9% and with the antigen E. granulosus 16.7% and 89.5%.  相似文献

Fifteen metacestode antigens from Taenia saginata were defined by Laurell crossed immunoelectrophoresis and investigated for their potential use in immunodiagnosis of bovine cysticercosis. Several antigens cross reacted with those of some common cattle parasites. Three of the antigens, designated as numbers 4, 8 and 11, were selected on the basis of their restricted cross reactions and were isolated by affinity chromatography. These antigens showed high sensitivity and specificity values in the enzyme-linked immunosorbent assay (ELISA) for serodiagnosis of bovine cysticercosis.  相似文献

In March, 1973, a large feedlot near Phoenix, AZ, reported an increased incidence of bovine cysticercosis. Approximately 10% of cattle sent to slaughter from January to April, 1973, were infected with the cysticercus stage of Taenia saginata. One employee who worked at the feed mill and loaded hay in the fields was also found to be infected with T saginata. Recommendations for control of similar epizootics included educating employees about mode of transmission and improved personal hygiene, the inauguration of a surveillance program that included examination of employees prior to employment and periodically thereafter, maintenance of animal source and destination records, restriction of unauthorized personnel to critical areas, and periodic microbiologic assay of water supply.  相似文献

Risk factors in light T. saginata cysticercosis in cattle herds were investigated in a case-control study. The case group consisted of farms from which cattle with low-grade cysticercosis had been detected at slaughter. The control group consisted of farms where no cases were detected during the same period. The major risk factor identified was allowing cattle access to drink from streams carrying effluent from sewage treatment plants (odds ratio = 3.6). Spreading of septic tank sludge through deliberate or accidental mixing with animal slurry, deposition of sewage sludge, and proximity to railways, camping sites or sewage treatment plants seemed to be of minor importance.  相似文献

A 'dipstick' immunoassay for bovine cysticercosis, using an antigen isolated from Taenia hydatigena cyst fluid, was evaluated in cattle experimentally infected with Taenia saginata. The assay correctly identified six out of seven infected cattle, including an animal in which only 12 living cysticerci were found. Cattle became seropositive as early as 3 weeks post-infection. A false-negative reaction was found for one very lightly infected animal, from which only four living cysticerci were recovered at necropsy. The assay was also used to detect circulating antibodies in experimentally infected cattle before and after therapeutic treatment with anthelminthics. The results suggest that praziquantel-treated animals gradually revert to being seronegative after the cysticerci are killed.  相似文献

Data from slaughter plants (n = 3) and feedlots (n = 18) in eastern Washington were analyzed to characterize occurrence patterns of cysticercosis in Washington during 1984. Three concurrent peaks in cysticercosis rates (0.6/1,000 to 5/1,000 slaughtered cattle) were detected at 3 slaughter plants. Peaks were observed at 8 feedlots from December 1983 to March 1984, at 6 feedlots from April to July 1984, at 2 feedlots from August to October 1984, and at 3 feedlots from November 1984 to February 1985. Affected feedlots were not closely associated geographically and were feeding cattle from many, predominantly northwestern, origins. For 3 feedlots for which time in the feedlot was available for each slaughter shipment, an increase in cysticercosis rate with increasing time in the feedlot was noticed. Within these 3 feedlots, cases of cysticercosis were widely scattered spatially. The pattern of cysticercosis indicated human fecal contamination of a regionally available feed source. Of feedstuffs in use, potato waste, a byproduct of the processed potato industry, appeared to be the most likely source of Taenia saginata ova.  相似文献

Urban sewage production is increasing and its agronomical use as a fertiliser has been advocated. Considerable defiance is prevalent in consumers and among farmers on the use of such fertilisers due to unknown pathological or environmental risks. The aim of the present review was to consider which pathological risk is major. Cysticercosis due to Taenia saginata appears to be one of the major pathological threats when sewage sludge is used to fertilise cattle pastures in temperate areas. The situation is different in Africa (Taenia solium and T. saginata are both highly prevalent) and Asia (Taeniasaginata-like are prevalent). The processing of sludge and the delay between its application onto a pasture and grazing are probably major risk factors. Little data are available on the influence of processing, delay between processing and the use of sludge on the pathogenic risk. Producers and consumers will be more confident on the use of sludge if objective data are gained on risk. Most of the cases of cysticercosis (North America, United-Kingdom, Germany or Denmark) are related to poor human hygiene or accidental overflooding of sewage plants onto pastures. The standard application of sludge on pastures is apparently at low risk. This low risk does not mean that surveillance should cease since outbreaks of cysticercosis have been reported. Future investigations should concentrate on the most sustainable means of reducing risk (length of storage before use, composting, other treatments).  相似文献

Taenia saginata cysticercosis in an Ohio cattle feeding operation   总被引:1,自引:0,他引:1  
In January to March 1981, 37 slaughter cattle from a single Ohio feeding operation were determined, at postmortem inspection, to be infected with Taenia saginata cysticerci. A subsequent outbreak on this same farm in March 1983 involved 7 slaughter cattle. An epidemiologic investigation was conducted of possible sources of the T saginata ova; these included leakage of raw sewage onto the pasture after a flood in 1980, municipal sewage sludge application on the farm, defecation in feed or water by farm workers, and other off-farm sources. Temporal and spatial observations implicated raw sewage contamination of pastures as the most likely source of infection in the 1981 outbreak. The outbreak in 1983 was more likely associated with sludge application. The possibility of an infected worker exposing the cattle to infected feces was not excluded definitely as a possible source.  相似文献

The paper describes the epidemiological investigation carried out on two dairy farms with cattle infected with Taenia saginata cysts. On the first affected farm it was estimated using Bayesian techniques that approximately 65% of 1400 mixed-age cattle were infected with Taenia saginata cysts. The investigation aimed to determine potential exposure pathways of cattle to Taenia saginata with a view to finding the human source of infection and to describe the epidemiology of the outbreak on the affected farms. In order to determine potential exposure pathways, investigation was centred on how feed or water could have been contaminated with eggs. The plausibility of pathways was determined by examining the spatial and temporal association between factors related to the pathway and the prevalence of infection in cattle strata. This study reports on the purification of an antigen predominant within the vesicular fluid (VF) of T. crassiceps metacestodes and shown to share identity with the major vesicular fluid protein of the T. saginata larval stage. Purification was achieved by gel filtration of the VF on Sephacryl S-300 superfine, followed by ion exchange HPLC. The antigen represents a single polypeptide chain (Mr appr. 37,000) with carbohydrate moieties without affinity to ConA. Isoelectric focusing of the electrophoretical and immunological homogeneous antigen resulted in five bands focusing at pH 4.0, 4.2, 4.4, 4.6 and 4.8, respectively. In ELISA, the purified antigen detected serum IgG-antibodies in all 21-35 weeks old calves (n = 10) with experimental cysticercosis (70 to 6,000 viable larvae recovered). When compared to T. saginata metacestode VF the antigen was the better reagent for discriminating between infected and non-infected animals. As shown by immunodiffusion and ELISA the antigen is also common to the T. saginata adult stage and obviously to other taeniid metacestodes where it is accumulated in the VF or hydatid fluid.  相似文献

Two lungworm outbreaks in dairy herds were investigated in order to estimate the resulting economic costs. On the two farms, with 110 and 95 cows, total costs were estimated at ?59 and ?67 per cow, respectively. Overall, milk production reduced by 15 to 20 per cent during the outbreaks. Five cows died on one farm, while on the other farm seven cows died as a result of the lungworm outbreak. On one farm, 51.7 per cent of the total costs was due to reduced milk production and 33.1 per cent was due to disposal of dead animals. On the other farm, it was 36.3 and 50.9 per cent, respectively. The remaining 13 to 15 per cent of the total costs were due to extra inseminations, laboratory diagnosis and treatments. The history and development of the outbreaks are described. One lesson from these outbreaks is that recognising that potentially lungworm-na?ve animals are to be introduced into the adult herd allows for timely measures (for example, vaccination) to prevent a lungworm outbreak.  相似文献
