Background: Hematologic and plasma biochemical evaluations are routinely used in evaluating the chelonian patient, but appropriate processing techniques have been minimally defined.
Objectives: This study was designed to compare the effects of temperature, time, anticoagulant, and species on sodium and potassium values in the Aldabra tortoise ( Geochelone gigantea ) and the Burmese mountain tortoise ( Manouria emys ).
Methods: Blood samples from 7 Aldabra tortoises and 8 Burmese mountain tortoises were collected into tubes without anticoagulant and tubes containing lithium heparin. Sodium and potassium concentrations were measured by flame photometry in serum and plasma harvested immediately after collection and from aliquots of whole blood stored at 4°C and 25°C for 5 to 120 minutes.
Results: In Aldabra tortoises, storage time and temperature had no significant effect on potassium concentrations in heparinized blood and in blood without anticoagulant. However, sodium concentrations in serum and plasma decreased significantly in samples without anticoagulant stored at 4°C and 25°C and in heparinized samples stored at 4°C. In Burmese mountain tortoises, potassium concentrations in serum and plasma increased significantly with time in samples without anticoagulant and in heparinized samples stored at 4°C and 25°C, but the increases were less at 4°C. Sodium concentrations in serum and plasma decreased significantly in blood without anticoagulant and heparinized blood stored at 4°C and 25°C.
Conclusions: Storage of blood samples with and without anticoagulant at 4°C significantly improved the stability of potassium and sodium concentrations in both species of tortoises. Early separation of red cells from serum or plasma after blood collection is especially important to ensure the reliability of potassium measurements. 相似文献

BACKGROUND: The tawny owl (Strix aluco) is a protected species in Italy. Orphaned, injured, and ill owls often are sheltered and treated in rehabilitation centers, where hematologic and biochemical analyses would be helpful to evaluate and monitor the status of their health. OBJECTIVES: The major aim of this work was to assess hematologic and biochemical constituents together with protein electrophoretic fractions in healthy tawny owls. In addition, we compared laboratory methods for determining hemoglobin (Hgb), total protein, and albumin concentrations. METHODS: Heparinized blood samples were collected from 10 clinically healthy adult captive tawny owls between March 2001 and November 2003 for CBC, routine biochemical analysis, and protein electrophoresis. Alternate methods for Hgb (estimation as HCT/3 vs spectrophotometry), total protein (biuret vs refractometry), and albumin (bromcresol green vs electrophoresis) concentrations were compared in 34 samples from 16 unhealthy adult owls and 8 nestlings. RESULTS: Results were reported as mean, median, and range (minimum-maximum). Significant differences and poor concordance were observed between methods for Hgb, total protein, and albumin. CONCLUSIONS: Hematologic and plasma biochemical values in captive tawny owls may be useful in evaluating and monitoring the health of this species in captivity. 相似文献

Background: Bearded dragons are one of the most popular pet lizard species, and biochemical reference values are useful for health management of these reptiles. Objectives: The objectives of this study were to measure plasma biochemical values in healthy captive bearded dragons, determine reference values, and evaluate the effects of sex and season on the results. Methods: Blood samples were collected from 100 captive healthy bearded dragons in Tokyo during the summer and winter. Plasma biochemical measurements were performed using a dry‐slide automated biochemical analyzer. The data were then compared based on sex and season using 2‐way ANOVA. Results: Globulin, cholesterol, and calcium concentrations of females were higher in both summer and winter compared with the values obtained for males. Both males and females had higher uric acid concentrations in winter than in summer. When compared with males, females had a higher chloride concentration in summer and a higher total protein concentration and aspartate aminotransferase activity in winter. Potassium concentration in males was lower in winter than in summer, whereas in females cholesterol concentration was lower in winter than in summer. Conclusions: Biochemical values that differed based on sex and season in bearded dragons were similar to those in other lizards. These differences reflect physiologic differences in reproductive status in females and seasonal changes in temperature and hydration status. Plasma biochemical values established for bearded dragons in this study will be useful in the diagnostic assessment of captive animals. 相似文献

Behavioural frequencies and time budgets for male and female Chersina angulata were recorded in spring, September 2004. The daily activity of the population was 10.51 ± 0.42 h (mean ± CI), but individual males and females were in the open for 2.57 ± 1.12 h and 1.58 ± 1.44 h, respectively. Both sexes spent nearly 3.5 h per day basking with 90% of the basking time in the cover of sparse vegetation. Feeding and walking were the most frequent active behaviours. The activity pattern of the sexes differed between 10:00 and 12:00 in the morning when males spent more time out of cover on active behaviours, particularly feeding, than females did. The daily activity pattern of the population peaked between 10:00 and 12:00, reflecting the activity pattern of the more active sex, males. Females were out of cover, active, and feeding at higher temperatures than were males. The high temperatures probably allowed females to complete activities more efficiently, reducing their exposure time and predation risk. Although fighting and mating represented small fractions of male time budgets, the extended physical presence through walking and feeding may help males to establish dominance hierarchies and enhance mating success. 相似文献

BACKGROUND: The rock hyrax (Procavia capensis) is an herbivore prevalent from South Africa to Turkey, and a most common zoo animal. Although many studies of hyrax diseases and physiology are available, clinicopathologic data are limited. OBJECTIVES: The purpose of this study was to establish comprehensive hematologic and biochemical reference intervals for trapped, apparently healthy, free-ranging rock hyraxes using modern laboratory methods and to assess differences related to sex, gestation, and age. METHODS: Blood samples were obtained from 27 healthy, free-ranging hyraxes under anesthesia. Gender, body weight, and gestational status were recorded. Hematologic (n = 25) and serum biochemical (n = 22) analyses were performed using standard automated methodology. Data for male vs female, adult vs juvenile, and pregnant vs nonpregnant female hyraxes were compared using the Mann-Whitney U-test. Associations between variables were assessed using Pearson's or Spearman rank correlation tests. RESULTS: Significant age- and sex-related, but not gestation-related differences were observed in several variables. Serum alkaline phosphatase activity and phosphorus concentration were significantly higher in juveniles compared with adults. A unique type of monocyte comprised 1-3% of leukocytes in 4 hyraxes. Markedly high serum creatine kinase (CK) activity was observed in most hyraxes. CONCLUSIONS: The large number of animals and the availability of sex, age, and gestational data in this study will be useful to zoo and wildlife veterinarians working with rock hyraxes. High serum concentrations of betahydroxybutyric acid in the rock hyrax, compared with dogs, cats, and ruminants, may be related to its unique digestive system. High CK activity may have been the result of a capture myopathy-like syndrome. The unique monocytes in hyraxes resemble those of elephants and are a novel finding in this species. 相似文献

Abstract: Tilapia are a commonly aquacultured fish yet little is known about their normal physiology and response to disease. In this study we determined the results of complete hematologic (n = 40) and plasma biochemical profiles (n = 63) in production tilapia ( Oreochromis hybrids). The fish were raised in recirculating systems with a high stocking density (120 g/L), and were in the middle of a 15-month production cycle. Blood was analyzed using standard techniques, and reference intervals were determined using nonparametric methods. Non-production tilapia (n = 15) from low-density tanks (4 g/L) also were sampled; the clinical chemistry results were compared to reference intervals from the fish raised in high-density tanks. Differences were noted in plasma protein, calcium and phosphorus concentrations, such that reference intervals for high-density production tilapia were not applicable to fish raised under different environmental and management conditions. 相似文献

BACKGROUND: Increasing interest in wildlife care leads to the need for new tools to evaluate animal health. Laboratory investigations require reference intervals against which to compare the results obtained. For common buzzards, only a few studies have been performed to establish hematologic and biochemical reference intervals. OBJECTIVES: The aim of this work was to develop reference values for routine hematologic and biochemical constituents and protein electrophoretic fractions and evaluate possible seasonal differences in values for healthy common buzzards. METHODS: Heparinized blood samples were collected from 23 captive, clinically healthy common buzzards between February 2001 and June 2003. A CBC, routine biochemical analysis, and protein electrophoresis were performed. Data distribution was assessed and results from birds sampled in spring, summer, and winter were compared. Results from alternative methods for hemoglobin (Hgb; estimated as HCT / 3 vs spectrophotometry), total protein (biuret vs refractometry), and albumin (bromcresol green vs electrophoresis) concentrations also were compared. RESULTS: Reference intervals were calculated as 10-90th percentiles. In spring and summer, total WBC and heterophil counts, and urea, total protein, prealbumin, and beta- and gamma-globulins concentrations were significantly different from winter values. Results obtained by alternative methods for Hgb, total protein, and albumin concentrations were significantly different from those obtained by standard methods, although estimated and spectrophotometric Hgb values were significantly correlated. CONCLUSIONS: The reference values obtained in this study for hematologic and plasma biochemical constituents and their seasonal variation in healthy, captive common buzzards will be useful in the clinical evaluation of these birds in rehabilitation settings. 相似文献

Background: Alterations in cellular and chemical constituents in blood are indicators of physical or chemical restraint in deer. Objectives: The aim of this study was to determine the effects of physical and chemical restraint on hematologic and biochemical analytes in captive brown brocket deer (Mazama gouazoubira). Methods: Six adult deer were physically restrained and randomly anesthetized in a crossover design using 3 protocols (P). In P1, anesthesia was induced directly with isoflurane using a face mask and was maintained with isoflurane delivered by endotracheal tube. In P2, oral premedication with midazolam was followed 1 hour later by induction and maintenance of anesthesia with isoflurane, as in P1. In P3, intravenous anesthesia with a combination of ketamine, xylazine, and atropine was followed by anesthesia with isoflurane, as in P1. Blood was collected immediately after physical restraint (T0) and during anesthesia at 20‐minute intervals (T15, T35, and T55). Hematologic and biochemical constituents were determined using manual methods or automated analyzers, with cortisol measured by enzyme immunoassay. Results: During physical restraint, following food and water restriction, significant hemoconcentration, increased WBC count, and lower cortisol concentration were observed compared with results at T15, T35, and T55. During anesthesia, lymphocyte count was lower in P2 than P1, urea concentration was decreased at T15 using P2, and plasma glucose concentration was increased after T0 (P<.01). Among the anesthetic protocols, there was a trend for decreased cortisol concentration using P2. Conclusions: Physical restraint and anesthesia affect some blood constituents in captive brown brocket deer, and midazolam premedication may modify the intensity of the stress response. 相似文献

Toxoplasma gondii consists of three genotypes, namely genotype I, II and III. Based on its virulence, T. gondii can be divided into virulent and avirulent strains. This study intends to evaluate an alternative method for predicting T. gondii virulence using hierarchical cluster analysis based on complete coding sequences (CDS) of sag1, gra7 and rop18 genes. Dendrogram was constructed using UPGMA with a Kimura 80 nucleotide distance measurement. The results showed that the prediction errors of T. gondii virulence using sag1, gra7 and rop18 were 7.41%, 6.89% and 9.1%, respectively. Analysis based on CDS of gra7 and rop18 was able to differentiate avirulent strains into genotypes II and III, whereas sag1 failed to differentiate. 相似文献
