The effect of different larval dose level and dosing regimens on the course of Cooperia oncophora infection in calves was studied. Four groups each of 4 calves were experimentally infected either with 50,000 or 200,000 C. oncophora larvae (L3) given either as single infections or as daily trickle infections. An additional group of calves remained as uninfected controls. The animals were necropsied on week 4 after infection. Mild to moderate clinical signs of parasitic gastroenteritis developed among calves given high doses of larvae, but liveweight gains were not significantly different from those of the uninfected controls. Serum pepsinogen levels of dosed animals were within normal ranges but rose slightly, and on day 14 p.i. they differed significantly from those of the controls. On that occasion, the levels of serum pepsinogen in the trickle infected groups significantly exhibited the levels of the single infected groups. Hypoalbuminaemia was not a feature on any occasion. The various groups did not differ significantly with regard to total worm counts and adult worm counts, but the groups receiving high larval dose harboured significantly more fourth stage larvae than the group receiving low doses of larvae, both in terms of absolute counts and in terms of percentages of total worm burdens. Within the same dose level, there was a tendency of a more even distribution of worms along the small intestine when the infections was given as a single infection compared with a trickle infection. The results indicate that C. oncophora larval dose and dosing regimens may influence the pathogenic effects and to some extent the distribution of the parasite in the small intestine.  相似文献

OBJECTIVE: To report the prevalence of bovine viral diarrhea virus (BVDV) in calves and calf groups (ie, calves from the same farm) in beef breeding herds and evaluate the ability of biosecurity risk assessment questionnaires to identify calf groups with positive results for BVDV. DESIGN: Nonrandom survey. ANIMALS: 12,030 calves born in spring from 102 operations. PROCEDURES: Cow-calf producers that voluntarily enrolled in a screening project submitted ear notch specimens from calves and answered a 29-question survey instrument. Ear notch specimens were tested for BVDV with an antigen-capture ELISA (ACE), and ear notch specimens with positive ACE results for BVDV were immediately retested by performing immunohistochemistry (IHC). Follow-up testing, 3 to 4 weeks after initial positive ACE results, was done by use of a second IHC test and virus isolation on a subsequently submitted ear notch specimen from the same calves to identify those that were persistently infected (PI). RESULTS: 102 producers submitted ear notch specimens for BVDV screening. Initially, 24 of 12,030 calves had positive ACE results for BVDV. A second ear notch specimen was submitted for 20 of these 24 calves. Of 20 retested calves, 12 had positive ICH results for BVDV, confirming PI status. The 12 PI calves came from 4 calf groups (3 singletons and 1 calf group with 9 PI calves). CONCLUSIONS AND CLINICAL RELEVANCE: Prevalence of BVDV in calf groups was low, and questions designed to identify high-risk biosecurity behaviors had little value in identifying calf groups with positive results for BVDV.  相似文献

Holstein calves were managed from less than 1 wk of age as in the special-fed veal industry but subjected to three dietary regimens (n = 10/group): low dietary iron (LI = approximately 5 mg iron/kg dry milk replacer) throughout 16 wk; 2) high then low dietary iron (H-LI = 140 mg/kg through d 18, then approximately 5 mg/kg through wk 16; a typical industrial scheme) or 3) high dietary iron (HI = approximately 105 mg/kg throughout the study). Several physiologic, behavioral and health indicators of welfare were monitored at various times. From wk 7 on, blood hemoglobin concentration was higher in HI calves than in LI, whereas that in H-LI calves was intermediate. Blood red cell count was higher in HI calves than in LI from wk 11 to 16, and was higher in HI than in H-LI from wk 14 to 16. Ratio of blood segmented neutrophils to lymphocytes (an indicator of stress) did not differ due to dietary regimen. Between wk 2 and 16, lying time increased from 69.5 to 76.6% of total time. Oral behaviors (e.g., licking and gnawing) occupied less than 15% of total time. Dietary regimen did not affect time spent either lying or engaging in oral behaviors. Calves in all dietary-regimen and slaughter-age groups experienced high frequencies of pneumonia, digestive-tract maladies and trichobezoars, but neither disease nor medical-treatment frequency was related to dietary regimen. Live, hot-dressed carcass and liver weights of the five calves/group slaughtered at 16 wk were not affected by dietary regimens, but carcass grade was highest for LI calves and lowest for HI.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Two different feeding regimens for calves (age up to 21 days) suffering from acute diarrhea were evaluated with respect to course of illness and change in body weight. The patients of the experimental group were given whole milk ad libitum three times a day. The patients of the control group were offered whole milk at a rate of 13% their body mass per day divided into three meals. In addition, both groups were offered an oral rehydration solution at a rate 9.5% of body mass per day, also divided into three portions. Data from 60 calves (30 in both groups) were available for statistical analysis. The duration of illness in the experimental group was 10.9 +/- 6.0 days vs. 10.0 +/- 5.2 days in the control group. The daily increase of body mass in the experimental group averaged 0.607 +/- 0.452 kg, the one in the control group 0.230 +/- 0.231 kg (p = 0.0002). The calves of the experimental group required 13.16 +/- 11.18 I milk per kg weight gain, the calves of the control group 21.39 +/- 12.80 I. Six calves of the experimental group and two calves of the control group subsequently had a relapse. On the basis of the results obtained, a feeding regimen for calves suffering from acute diarrhea is suggested.  相似文献

Bovine viral diarrhea virus (BVDV) affects cattle populations causing clinical signs that range from subclinical immunosuppression to severe reproductive and respiratory problems. Detection and removal of persistently infected (PI) calves is the single most important factor for control and eradication of BVDV. Current testing strategies to detect PI calves rely heavily on immunohistochemistry (IHC) and a commercially available antigen capture ELISA (ACE) assay. These viral assays depend on 1 or 2 monoclonal antibodies which target the E(rns) glycoprotein of BVDV. The sensitivity and specificity of these two tests have been reported previously. The purpose of this research was to characterize a strain of BVDV (AU501) that was undetectable using IHC and ACE based on a single monoclonal antibody, but was consistently detected in samples from a Holstein steer using virus isolation and PCR testing. Sequencing of this AU501 viral isolate revealed a unique mutation in the portion of the genome coding for the E(rns) glycoprotein. This unique field strain of BVDV demonstrates the risk of relying on a single monoclonal antibody for detection of BVDV. Multiple testing strategies, including polyclonal or pooled monoclonal antibodies that detect more than one viral glycoprotein may be necessary to detect all PI calves and facilitate eradication of BVDV.  相似文献

Profiles of blood cell counts were evaluated for 15 calves from three different farms. These calves showed petechia in the mucous membranes and in the skin and prolonged secondary bleeding after puncture. The clinical course of the disease could be observed in eleven calves. With exception of one case, the blood cell counts indicated a severe anaemia, leukocytopenia and thrombocytopenia. Out of these 15 calves, six calves survived and the other nine calves died or had to be euthanized due to the severity of the disease. Necropsy of these nine calves revealed petechia in the skin, subcutis, muscles, in inner organs and all serous membranes. Pathohistological examination showed a depletion of the bone marrow and lymphatic tissue in eight calves. These findings confirmed the diagnosis of bovine neonatal pancytopenia (BNP) for eight of these nine calves. Bluetongue virus serotype 8 was tested negatively using PCR. Bovine virus diarrhoea virus (BVDV) was negatively tested using immunofluorescence and cell culture and salmonella species were negatively tested in seven dissected calves. A cluster of toxins was negatively tested in one of the dissected calves. All 15 calves had high antibody titres for BVDV. The BVDV-antibody titres from twelve dams with affected calves were positive in six cases and not detectable in the other six cases. In three of the six dams with not detectable BVDV-antibody titres, calves were fed with colostrum of a further dam with high BVDV-antibody titres. In the further three dams without detectable BVDV-antibody titres, we could not ascertain which colostrum has been fed to the calves. BVDV-specific antigen could not be detected in any of the samples from the calves and dams tested. Using the activity of the gamma-glutamyl-transferase, we assumed a sufficient supply with colostrum for the examined calves.The cause for the occurrence of these BNP cases was due to bone marrow depletion.The reason for the bone marrow depletion remained unclear. However, it was obvious that the BNP described here is highly likely caused by colostrum from cows with positive BVDV-antibody titres.  相似文献

Influence of isoprinosine on bovine herpesvirus type-1 infection in cattle   总被引:1,自引:0,他引:1  
A study was conducted to determine the in vivo efficacy of isoprinosine (ISO) in calves infected with bovine herpesvirus type-1 (BHV-1). Calves were infected with BHV-1 on day 0 and received ISO daily for 14 days. Clinical signs of disease, shedding of BHV-1, lymphocyte proliferative responses to mitogens, interleukin-2 production, and alveolar macrophage bactericidal activity were monitored during the study. Rectal temperatures were increased (P less than 0.05) in BHV-1 and ISO-BHV-1 calves at days 3 to 7 postinfection (PI). Isoprinosine did not influence BHV-1 shedding in calves. Lymphocyte proliferative responses to phytohemagglutinin (PHA) were lower (P less than 0.01) in BHV-1 calves when compared to control or ISO calves at day 4 PI, but ISO did not ameliorate this effect. Interleukin-2 activity was greater (P less than 0.05) in ISO-BHV-1 calves on days 4 and 8 PI in PHA-stimulated lymphocytes and on day 8 PI in concanavalin A-stimulated lymphocytes when compared to control, ISO or BHV-1 calves. Isoprinosine treatment of BHV-1-infected calves tended to decrease alveolar macrophage bactericidal activity. These data suggest that ISO does not reverse BHV-1 suppression of lymphocyte proliferation, but may enhance IL-2 production in BHV-1 infected calves.  相似文献

Reasons for performing study: Studies in man have shown a correlation between Angiotensin I‐converting enzyme (ACE) genetic polymorphisms, ACE activity in the blood and superior athletic performance in sports requiring endurance. It has been hypothesised that the same correlation occurs in horses. There is no information in the literature concerning the effects of training on ACE activity in equine plasma. Hypothesis: Exercise training influences the activity of circulating ACE and the response observed is dependent on the exercise protocol. Methods: Thirteen horses of mixed breeds were randomly allocated 2 different training protocols to be carried out for a period of 15 weeks. Blood samples were collected from each horse before the beginning of training to determine baseline values. Subsequent sampling took place every 15 days throughout the training phase and for 8 weeks of paddock rest. Angiotensin I‐converting enzyme activity was determined by automated spectrophotometry. Results: Training for 15 weeks significantly increased plasma ACE activity, irrespective of training protocol. Differences observed in ACE activity pattern between the 2 training protocols were not statistically significant. Increase in ACE activity peaked with maximum workload. As soon as training was interrupted, ACE levels significantly decreased. Conclusions and discussion: Exercise training affects levels of ACE activity in equine plasma. The mechanism for this is not yet elucidated, but cardiovascular adaptation to exercise and blood pressure changes might be involved in this regulation. Potential relevance: Exercise training produced a gradual increase in enzymatic activity and might warrant the use of ACE as a tool for fitness monitoring. Angiotensin I‐converting enzyme enzymatic activity in the plasma might be directly correlated to a change in genetic expression and that variability must be taken into account when evaluating results from horses undergoing a physical training programme.  相似文献

The effects of a modified-live infectious bovine rhinotracheitis virus vaccine (administered ocularly or intranasally) on experimentally induced infectious bovine keratoconjunctivitis were evaluated. The modified-live infectious bovine rhinotracheitis virus vaccine was administered to 13 male Holstein calves (intranasally in 4 and ocularly in 9; day 0). Five calves were not vaccinated and served as controls. Calves were examined daily and, starting on day 4, Moraxella bovis was administered ocularly to all 18 calves once daily for 4 days. The eyes of all calves were assigned a clinical score, and the ocular secretions were evaluated for presence of infectious bovine rhinotracheitis virus and M bovis daily until day 19. The severity of the ocular lesions was estimated by scoring the lesions clinically and by determining the protein concentration, myeloperoxidase activity, and WBC count in the tears. By day 5, conjunctivitis, chemosis, and epiphora were observed in all of the calves vaccinated ocularly. The calves vaccinated intranasally developed conjunctival plaques, but did not develop chemosis or photophobia. All of the calves developed keratitis after inoculation with M bovis. The median lesion scores were greater in both groups of vaccinated calves than in the controls. Corneal perforations developed exclusively in the vaccinated calves. The frequency of M bovis isolation from ocular secretions was significantly (P less than 0.05) greater in the vaccinated calves than in the controls. The tears from the intranasally vaccinated calves contained the highest myeloperoxidase activity and WBC count. Polioencephalomalacia (PEM) was induced in calves by feeding a semipurified, low-roughage diet of variable copper and molybdenum composition. Two formulations resulting in Cu-insufficient and Cu-sufficient forms of the diet were fed (n = 10 and 4 calves, respectively); both diets induced PEM. Clinical signs of disease developed as early as 15 days after transition to the experimental diets and included impaired vision, decreased response to external stimuli, and abnormal gait. Grossly evident cerebrocortical lesions consisted of laminar areas of cavitation and/or autofluorescence seen under UV illumination. Hepatic Cu concentration was decreased in calves fed the Cu-insufficient diet, but not below normal range. During the course of feeding either diet, rumen pH decreased, rumen volatile fatty acid concentrations increased, rumen and blood lactic acid concentrations increased, and rumen and plasma thiamine concentrations increased. The thiamine pyrophosphate effect on erythrocyte transketolase activity was unaltered in calves of either diet group. This nutritionally induced form of PEM does not appear to be related to Cu deficiency or reduction in plasma or rumen thiamine concentration.  相似文献

The effect of clenbuterol hydrochloride on the course of disease in calves with a natural bovine respiratory syncytial virus infection was examined. Six calves (three to nine months of age) originating from four herds with respiratory tract disease and serological evidence of a bovine respiratory syncytial virus infection were used in this study. The calves were injected intravenously with clenbuterol hydrochloride. The effect of clenbuterol on the course of disease was measured using the PO2 in blood taken from an indwelling canula inserted in the caudal auricular artery and by clinical signs. Clenbuterol did not improve clinical signs. After clenbuterol administration arterial PO2 values decreased significantly in five out of six patients. Six to eight hours after medication the mean arterial PO2 values were higher than initial values. The moderate positive effect of clenbuterol after six to eight hours may be caused by enhancing ciliary activity and by the secretolytic activity of clenbuterol.  相似文献

The prevalence of bovine viral diarrhea virus (BVDV) in persistently infected (PI) cattle in beef breeding herds was determined using 30 herds with 4530 calves. The samples were collected by ear notches and tested for BVDV antigens using immunohistochemistry (IHC) and antigen capture enzyme-linked immunosorbent assay (ACE). Animals with initial positives on both IHC and ACE were sampled again using both tests and serums were collected for viral propagation and sequencing of a viral genomic region, 5′-untranslated region (5′-UTR) for viral subtyping. Samples were also collected from the dams of PI calves. There were 25 PI calves from 4530 samples (0.55%) and these PI calves were from 5 of the 30 herds (16.7%). Two herds had multiple PI calves and 3 herds had only 1 PI calf. Only 1 of the 25 dams with a PI calf was also PI (4.0%). The subtype of all the PI isolates was BVDV1b. Histories of the ranches indicated 23 out of 30 had herd additions of untested breeding females. Twenty-four of the 30 herds had adult cowherd vaccinations against BVDV, primarily using killed BVDV vaccines at pregnancy examination.  相似文献

The effects of weaning on beef calves of different ages were investigated. Forty male and 40 female calves were either weaned at 6 (W6, n = 40) or 8 (W8, n = 40) months of age. The vocalization activity and behaviors (feeding, lying and standing/walking) were observed during the first 3 days following weaning. Body weight was recorded at 6 and 8 months and 2 weeks following weaning. W6 animals vocalized more often than W8 calves and females more than males on day 1 (P < 0.05). The vocalization activity decreased at a higher rate in W8 than in W6 from days 1 to 3. On day 2, W6 calves spent less time lying and more standing/walking than W8 calves (P < 0.05). The behaviors did not differ between males and females. The average daily gain after weaning did not differ between weaning ages (P > 0.05), but W8 calves gained more from 6 to 8 months of age. In conclusion, weaning distress was more pronounced in W6 than in W8 calves. W8 calves also had a superior growth performance during 6 to 8 months of age. The effect of the calves' sex was less pronounced. Therefore, it is recommended to wean beef calves later than 6 months of age.  相似文献

Estradiol plus progesterone (EP) implants have been shown to favorably alter the time course or decrease the severity of many of the clinical manifestations associated with coccidiosis and endotoxemia in calves. This study evaluated the effect of EP treatment on plasma tumor necrosis factor-alpha (TNF), thromboxane (TXB), prostacyclin (PRC), nitrite and nitrate (NO[x]), and cortisol. Holstein steer calves were divided into four groups: control, EP, endotoxin (LPS), and EP + LPS (n = five/group). Estradiol/progesterone pellets (Synovex-S) were implanted subcutaneously when calves reached 20 wk of age. One week after implantation, calves were injected i.v. with endotoxin (i.e., lipopolysaccharide; LPS, 0.6 microgram/kg of BW) or nonpyrogenic saline placebo. Body temperature was measured and blood was collected before injection and at 1, 2, 3, 4, 6, and 8 h thereafter. Plasma concentrations of TNF, cortisol, TXB, PRC, NO[x], were measured. Body temperature increased in both LPS and LPS-EP calves, but had returned to normal by 6 h in the LPS-EP group (P < 0.05). Plasma TNF and cortisol increased after LPS (P < 0.01), but were not differentially affected by EP treatment. Likewise, EP did not affect the magnitude of increase in LPS-induced PRC, but EP decreased the magnitude of increase in TXB (P < 0.05). Plasma NO[x]) levels were increased (P < 0.01) in calves after LPS; treatment with EP attenuated the LPS-associated increase in plasma NO[x] levels. These results suggest that EP exerts specific effects on different components of the proinflammatory cytokine cascade. Although the initiation of responses mediated by TNF, cortisol, and PRC do not seem to be differentially affected by EP, components of the nitric oxide- and TXB-axis responses to LPS are decreased in calves pretreated with EP.  相似文献

A severe outbreak of respiratory tract disease associated with bovine respiratory syncytial virus (BRSV) on a large beef-fattening farm is described. The outbreak started two days after five- to seven-month-old calves were vaccinated with a modified live BRSV vaccine. The disease ran a very severe course among five- to seven-month-old vaccinated calves, but disease was absent in eight-month-old an older non-vaccinated calves. The presence of IgM antibodies in sera of non-vaccinated calves indicated that BRSV was spreading on the farm between two to 15 days before the day of vaccination. The data indicate that vaccination with modified live vaccine during the course of a natural infection may enhance the severity of disease. The possible pathogenesis of the disease is discussed.  相似文献

新生犊牛通过初乳获得被动免疫,但是关于热应激对新生犊牛被动免疫的影响程度尚不清楚。本试验旨在研究热应激牛初乳对新生犊牛被动免疫是否有显著影响,并与非热应激生产条件下进行比较。试验动物为对应于采食热应激和非热应激母牛初乳的新生犊牛各4头,在其采食母牛初乳后的不同时间内采血用于免疫指标分析。结果表明:热应激奶牛哺乳犊牛血清中各成分含量在144h内均低于非热应激奶牛哺乳犊牛血清中相对应成分,其中IgG、T3、T(472h内)变化差异显著(P<0.05),INS和IGF-Ⅰ变化差异不显著(P>0.05),热应激在一定程度上影响新生犊牛被动免疫,有关机理有待于进一步研究。  相似文献

Newborn, male, Holstein calves, were continuously housed for three weeks in calf hutches at 17 degrees C or in a thermal environment which varied rhythmically on a daily basis either between -20 degrees C and -8 degrees C (experiment A) or between -30 degrees C and -18 degrees C (experiment B). Compared to warm-housed calves, cold-housed calves in experiment A had metabolic rates which were significantly higher (p less than 0.001) in a standing position but which were not significantly different (p less than 0.05) in a recumbent position. Recumbent and standing cold-housed calves in experiment B had an increased (p less than 0.05) metabolic rate compared to warm-housed controls. Heat loss was less (p less than 0.05) for recumbent cold-housed calves in experiment B than for standing calves in a thermoneutral environment. Localized subcutaneous hemorrhages of hindlimbs were a consistent necropsy finding among all cold-housed calves. Average daily gains of cold-housed calves were not significantly different from warm-housed controls. Clinical, physiological and pathological findings indicated that cold treatments used in the present study did not cause serious harm to calves. It was concluded that calves housed in properly managed hutches are remarkably cold tolerant.  相似文献

Angiotensin-I converting enzyme (ACE) is a key regulator of blood pressure, electrolytes and fluid homeostasis through conversion of angiotensin I into angiotensin II. Recently, a genetic polymorphism of the ACE gene, which accounts for 47% of the variation of ACE activity in blood, has been advocated as a biomarker of athletic aptitude. Different methods of analysis and determination of ACE activity in plasma have been used in human and equine research without a consensus of a "gold standard" method. Different methods have often been used interchangeably or cited as being comparable in the existing literature; however, the actual agreement between assays has not been investigated. Therefore, in this study, we evaluated the level of agreement between three different assays using equine plasma obtained from 29 horses. Two spectrophotometric assays using Furylacryloyl-phenylalanyl-glycyl-glycine as substrate and one fluorimetric assay utilizing o-aminobenzoic acid-FRK-(Dnp)P-OH were employed. The results revealed that the measurements from the different assays were not in agreement, indicating that the methods should not be used interchangeably for measurement of equine ACE activity. Rather, a single method of analysis should be adopted to achieve comparable results and critical appraisal of the literature is needed when attempting to compare results obtained from different assays.  相似文献

Effects of short-chain fatty acids on pancreatic exocrine secretion were studied under anesthesia in calves within 2 weeks of age (2-wks calves) given only whole milk and milk replacement and in which rumen fermentation has not begun yet, and in calves at 13 weeks of age (13-wks calves) weaned at 40 days of age and in which rumen fermentation has already begun. Basal rate of juice flow and protein concentration and amylase activity in pancreatic juice under basal condition were significantly lower in the 2-wks calves than those in the 13-wks calves. Intravenous administrations of acetate, propionate and butyrate stimulated pancreatic juice secretion and protein and amylase output in both groups of calves. Those responses were increased with increasing carbon number in the molecule of fatty acids. Although the response of amylase output (/kg of body weight) in the 2-wks calves was significantly less than that in the 13-wks calves, the response of juice flow and protein output (/kg of body weight) in the 2-wks calves were equivalent to or greater than those in the 13-wks calves. These results indicate that the characteristic of pancreas, being stimulated by short-chain fatty acids, in calves and probably in other ruminants is not generated on the process of postnatal development, but has been already acquired before rumen fermentation begins.  相似文献
