Root regeneration in shoot tip cuttings responds to graduated nitrogen (N) fertilization of stock plants. In pelargonium cuttings, reduced carbohydrate reserves caused by high N absorption by the donor plants and post-harvest storage of cuttings limit adventitious root formation, especially in low-light environments. In contrast, in chrysanthemum, similar carbohydrate reserves do not have this dominant effect on rooting capacity. The positive correlation between rooting capacity and internal N status is stable across a wide range of environments and is genotypically consistent for this species. However, the influence of N and carbohydrates on adventitious rooting of Euphorbia pulcherrima is unknown. We investigated the consequences of different N fertilization regimens applied to E. pulcherrima stock plants and cold and dark storage of the cuttings on N absorption, carbohydrate distribution, and rooting capacity of cuttings. Increasing time of stock plant cultivation with graduated N nutrition produced cuttings with N contents, ranging from 19 to 51 mg N g−1 dry mass (DM). High N absorption resulted in low carbohydrate concentrations in cuttings, and subsequent storage decreased carbohydrate concentrations further, particularly in stems. Lower sucrose contents in leaves were correlated with reduced rooting of stored cuttings at a particular harvest date. However, despite the lower carbohydrate levels, root numbers and lengths correlated positively with internal N concentrations. These relationships remained stable in unstored and stored cuttings, even when overall rooting intensity was reduced under lower natural light during autumn. Multivariate regressions accounting for nitrogen content, sucrose content and daily light integral during rooting highlighted these relationships and explained up to 79% of rooting variances. We conclude N nutrition of stock plants and N absorption by cuttings are the dominant factors determining the rooting capacity of poinsettia when rooting occurs under sufficient light, as is commonly available during propagation. To maximize rooting capacity of poinsettia cuttings their nitrogen content should exceed a threshold of 40 mg N g−1 DM.  相似文献

Summary

Cuttings from trees of jujube (Ziziphus jujuba Mill.), particularly mature ones, root with great difficulty. A greenhouse experiment assessed the effects of two strains of Agrobacterium rhizogenes (A4 and TR105) on softwood cuttings from two sources: ten year old trees of cv. Li and a tree approximately 40 years old of the cv. Contorta. The cuttings were collected and trimmed to 7.5 cm. Ten weeks after inoculation, both strain and source of cutting influenced visible symptoms of infection. Jujube cuttings were more responsive to TR105 than to A4, and the ‘Li’ cuttings were more responsive than ‘Contorta’ cuttings. Strain TR105 was very effective in increasing rooting percentages and root number. ‘Li’ cuttings had better rooting percentages, greater root number, and greater root length than did the ‘Contorta’ cuttings. Inoculation with A. rhizogenes may also possess great potential for promoting adventitious root formation in other difficult woody ornamental or fruit tree species.  相似文献

The effect of light quality on in vitro rooting of cherry rootstock “Colt” was studied to evaluate adventitious root development, ex vitro acclimatization and plant growth performance. Adventitious rooting was dependent on the light quality to which the microcutting were exposed. Highest number of roots per microcuttings was recorded under dichromatic light (blue + red). This response could be ascribed to the control of a strong synergistic interaction between phytochromes and blue light photoreceptors. Red light was more effective on root elongation than blue light. In this response a strong synergistic interaction between phytochromes and blue light photoreceptors was suggested. The effect of light quality on the number of root/explant affected the plant during acclimation, scoring the highest level of plant survival in the blue-, red- and blue + red-exposed plantlets. The light quality effect was also observed under greenhouse culture conditions, as shown by growth parameters at the end of six months in plants growth. No specific role was observed for the photoequilibrium of phytochrome. The results reported in this work show that plantlets exposed to different light quality, during the in vitro rooting phase, retain the light quality effects in the subsequent plant acclimatization and greenhouse plant growth phase.  相似文献

Current year shoot cuttings were collected in October and August from three growth habits of peach (Compact, Pillar, and Standard) and treated with one of four concentrations of indole butyric acid (0, 250, 1250, and 2500 mg L−1 IBA). Rooting response was measured after 5 weeks in the greenhouse. Little or no rooting occurred with cuttings from any growth habit that was collected in October or in August when treated with 0 and 2500 mg L−1 IBA. In August, the number of shoots that rooted was greater in cuttings from Pillar (79 and 45%) than Compact (13 and 3%) treated with 250 and 1250 mg L−1 IBA, respectively. Cuttings from Standard trees had intermediate rooting of 56 and 6% at 250 and 1250 mg L−1 IBA, respectively. Pillar trees consistently grew more roots with greater root length per cutting than the other growth habits. It is proposed that differences in rooting response among the growth habits may be associated with differences in endogenous auxin concentration that had been found in previous studies. Within peach and possibly other fruit trees, the capacity of shoot cuttings to develop adventitious roots can vary by cultivar and successful root induction with exogenous plant growth regulators may depend, in part, on endogenous hormone levels.  相似文献

Stem cuttings of oregano (Origanum vulgare L.) with pre-formed roots responded to a range of IBA and IAA concentrations with increased root emergence and without damage to the cuttings. No similar promotion of rooting was observed in cuttings lacking pre-formed root primordia. The data are discussed with respect to the differentiation and elongation phases of adventitious root formation. Root emergence from oregano cuttings and the continuity of stem vessels into the growing root are described.  相似文献

The capacity of many microorganisms for antagonism towards pathogens is unknown and previously uncharacterized species may be potentially useful in this respect. This work presents the evaluation of the capacity of a previously uncharacterized Ascomycete, denominated here as UA to protect chili pepper (Capsicum annuum) against Phytophthora capsici, and other soil-borne plant pathogens and establishes the role of UA as a biocontrol agent. Inoculation of UA 2 days before the inoculation of P. capsici led to 77.8% survival of pepper plants. Simultaneous confrontation of both microorganisms in vitro led to 53.1% growth inhibition of P. capsici, while the inoculation of P. capsici 3 days after the inoculation with UA improved growth inhibition up to 73%. Simultaneous confrontation in vitro of UA with Fusarium oxysporum and Fusarium solani led to 41.2 and 50% growth inhibition, respectively, but had no effect on Rhizoctonia solani or a binucleate Rhizoctonia isolate. Moreover, formation of zoosporangia and the germination of zoospores were completely inhibited by exposure to undiluted filter sterilized filtrate. UA produces septate mycelia, but could not be classified in detail due to a lack of spores or reproductive structures. However, sequencing of Internal Transcribed Spacer 1, 2 and the 5.8S genes indicated that this fungus is a member of the Ascomycetes.  相似文献

Inoculation of growing media with plant growth promoting rhizobacteria (PGPR) has a number of potential benefits for the production of ornamental plants. Certain rhizobacteria synthesise the enzyme ACC deaminase, which cleaves ACC, the precursor of the plant hormone ethylene. Bacterial metabolism is now known to lead to a reduction in [ACC] in the plant transpiration stream and bacteria are hypothesised to act on ACC exuded from roots. This in turn reduces the ethylene generated in plants growing in growing media inoculated with these bacteria. Here we tested if applications of the ACC deaminase containing rhizobacteria Variovorax paradoxus5C–2could be of benefit to ornamental growers by reducing ethylene generation in stressed plants. Ethylene is produced at levels that are inhibitory to growth and development under a number of abiotic stresses. The propagation and production of hardy ornamentals is deleteriously affected by abiotic stresses that involve ethylene signalling, including drought and physical wounding. Inoculation of growing media with V. paradoxus 5C-2 lowered ethylene emission from mature leaves of Cytisus × praecox experiencing drought stress. In addition, bacterial inoculation of the growing media resulted in significantly reduced abscission of the mature leaves under drought treatment. Beneficial effects of inoculation where also found in the wounding response of Fargesia murielae following divisional propagation and late season senescence in Aquilegia × hybrida in response to drought stress. Together these results demonstrate that V. paradoxus5C–2has real potential for use on ornamental nurseries in situations where plant stresses are unavoidable.  相似文献

The effect of the mature leaf retention and the exogenous application of flavonoids (naringenin, quercetin and rutin at 30 μM for 12 h) was studied for adventitious rooting of Ilex paraguariensis cuttings. Softwood cuttings harvested from young 3-year-old plants and adult 10- and 20-year-old plants were rooted under intermittent fog. A strong correlation (r2 = 0.72) between leaf retention and rooting was noted. The highest percentage of adventitious root formation (40%) was obtained when the leaf was artificially removed after 42 days of incubation. This data was supported by the histological analysis which provided anatomical evidence that cuttings have initiated root primordia by 21 days and the regenerated roots emerge through the epidermis after 35 days of incubation. A strong correlation between the position of the leaf and the site of roots regeneration was observed. A 100% of the rooted cutting with a single leaf only formed roots along the leaf axis at the base of the cutting. Quercetin increased the rooting percentage more than three times compared to the control and all flavonoids tested improved the distribution of roots around the stem without impacting the number of regenerated roots per rooted cutting from 20-year-old plants.  相似文献

In this study we investigated the potential usefulness of different substrates produced from gorse (Ulex europaeus L.) for propagating cuttings of Thuja plicata ‘Atrovirens’. The gorse was composted by different methods, to produce four types of compost. These composts were then used as such (100% compost) or in mixes with coarse sand and pine bark compost (25% compost, 25% coarse sand and 50% pine bark compost, v/v/v) to prepare eight different substrates. The substrates were compared in cutting propagation trials carried out in a greenhouse, with a standard rooting medium consisting of 25% Sphagnum peat moss, 25% coarse sand and 50% pine bark compost (v/v/v). Cuttings were taken from a garden hedge in March 2003, and comprised shoots of the previous year's growth. The bases of the cuttings were treated with a 0.4% solution of IBA rooting hormone, then inserted into cells filled with the corresponding substrate and placed on a propagation bench. Rooting percentage and visual rooting score were determined in July. The quality of roots was evaluated, as total root length and number of root tips, in July, September and November 2003, by use of a Delta-T Scan system. The rooting success of the cuttings ranged from 75% to 90%, except when the substrate comprised the pure composted gorse, obtained after the shorter period in an enclosed composting system with addition of fertilizer and reuse of leachates. Most of the cuttings in this substrate died and only 4% of the cuttings rooted successfully. Physicochemical characteristics of this rooting medium included very high electrical conductivity, low C/N ratio and high water retention. Addition of coarse sand and pine bark compost to the composted gorse significantly increased the rooting success. The root quality variables differed depending on the substrate considered and date of harvest. The results showed that substrates made from gorse compost are of potential use as a peat substitute for propagating cuttings of T. plicata ‘Atrovirens’. The physicochemical properties of the composted gorse should be determined before its use to establish whether it can be used pure as a substrate for rooting or whether it should be mixed with other substances.  相似文献

Despite advances in tissue culture techniques, propagation by leafy, softwood cuttings is the preferred, practical system for vegetative reproduction of many tree and shrub species. Species are frequently defined as ‘difficult’- or ‘easy-to-root’ when propagated by conventional cuttings. Speed of rooting is often linked with ease of propagation, and slow-to-root species may be ‘difficult’ precisely because tissues deteriorate prior to the formation of adventitious roots. Even when roots form, limited development of these may impair the establishment of a cutting.  相似文献

The influence of position of cuttings, auxin treatments and seasonal variation on adventitious rooting of Dendrocalamus asper, a multipurpose edible bamboo of horticultural value, was examined. Single node proximal (culm) and distal (culm-branch) cuttings were collected during the months of April–June and treated for 24 h with water or 2 mM auxin (indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and naphthalene acetic acid (NAA)). The treated cuttings were planted horizontally in sand beds and maintained under misting conditions for 2 months. Culm cuttings exhibited better adventitious rooting than culm-branch cuttings and the month of May proved to be the best for rooting of both types of cuttings. Auxin treatments also influenced adventitious rooting and their effectiveness was in the order: NAA>IBA>IAA. The interaction among auxin, position of cuttings and month of collection was also found to be significant on root induction and differentiation. Single node culm cuttings, without auxin treatment, resulted in 98% plantlets in May and 67% plantlets in June, and emerged as a suitable material for large-scale vegetative propagation. Administration of IAA or NAA to abundant culm-branch cuttings of low commercial utility (collected in May) also produces ≥50% plantlets.  相似文献

Hairy roots were induced from leaf-derived calli of lavandin (Lavandula × intermedia Emeric ex Loisel.) by infection with wild-type strains of Agrobacterium rhizogenes, A-5 (MAFF 02-10265) and A-13 (MAFF 02-10266). A-5-inoculated calli formed hairy roots more efficiently than A-13 ones. The transgenic shoots could be obtained from hairy root segments mediated by each Agrobacterium strain. However, different plant growth regulators were required for efficient adventitious shoot formation in each strain. In A-5, the most efficient adventitious shoot formation rate of 23.8% was observed in a medium with 4.4 × 10−6 M of 6-benzylaminopurine. On the other hand, a significantly higher rate of 13.2% was detected in a medium with 4.0 × 10−7 M of N-(2-chloro-4-pyridyl)-N′-phenylurea in A-13. Most of the regenerated plants showed dwarfism with closed internodes and extensive lateral branching, which were typical characteristics of ‘hairy root syndrome’. On the other hand, only nine of the 45 regenerated plants formed flower buds in early June, a delay of about one month compared with nontransgenic regenerated plants. The floral stalks and spikes of these plants were very short, resulting in a compacted form. Many regenerants showed a significantly lower productivity of essential oil than nontransgenic regenerants. Moreover, the relative percentage of the linalyl-cation-derived compounds, linalool and linalyl acetate, decreased in most of the regenerated plants. Compact plants with the ability of flower bud formation are assumed to be valuable not only for lavandin breeding, but also for clarifying the interaction between rol genes expression and essential oil production.  相似文献

Domestication of the recently discovered and highly endangered Wollemi pine has relied almost entirely upon serial propagation of cuttings from a very small conservation collection. This study assessed the requirement for applied auxin to induce rooting in tip cuttings and lower segment cuttings of Wollemi pine. Both types of cuttings proved easy-to-root, with mean rooting of 71% for tip cuttings and 82% for lower segments. Auxin application (at 1.5, 3.0 or 8.0 g indole-3-butyric acid/L) did not accelerate root protrusion from propagation tubes or affect final rooting percentages.  相似文献

Summary

Semi-hardwood cuttings of Cornus alba ‘Sibirica’, Deutzia ‘Mont Rose’, Forsythia × intermedia ‘Lynwood’, Ligustrum vulgare ‘Liga’, Philadelphus × virginalis, Potentilla fruticosa ‘Goldfinger’, and Spiraea × vanhouttei were planted on seven dates from July to October in two years and at three locations to investigate the effect of planting date on root formation, axillary bud growth, and plant survival. Cuttings were planted directly in the field and covered with polyethylene. Generally, root formation, bud growth, and plant survival were similar both years and at the three locations. All species except Deutzia had relatively constant rooting percentages at planting dates until mid August. For all species rooting percentages declined from mid August to October. Except for Deutzia, plant survival the following spring was constant or decreasing with planting date. For all species except Potentilla axillary bud growth and survival of cuttings planted in late September or October tended to increase while rooting percentages continued to decrease. In all species there was a close relationship between axillary bud growth and survival. Results revealed that many roots per cutting accelerated axillary bud growth.  相似文献

The effect of substrate, medium composition, irradiance and ventilation on rooting percentage, and root morphology and functioning with respect to mineral uptake was investigated in jojoba [Simmondsia chinensis (Link) Schneider]. Plantlets grown on Sorbarod plugs embedded in perlite accumulated more biomass and showed better rooting than plantlets grown on Sorbarod plugs embedded in agar. In plants grown on the latter substrate, the addition of sucrose to water or salts media promoted rooting and accumulation of shoot and root biomass. On perlite, rooting was high in all treatments, while root growth was enhanced only by 2% sucrose. Sucrose and irradiance had a positive synergistic effect on plantlet growth. Doubling the irradiance from 58 to 116 μE m−2 s−1 and using high ventilation (22-mm membrane) failed to improve rooting but promoted the ability of the stomata to close, thereby reducing water loss under ambient conditions. High irradiance and ventilation lowered the chlorophyll content, but without impairing biomass accumulation. Plantlets grown under enhanced ventilation and 0.5% sucrose formed fine roots within the cellulose plug and thick roots on the vessel floor. With the 2% sucrose supplementation, the roots were generally thicker, with very few thin roots being formed. 36Cl uptake into plantlets and transport to the leaves decreased in plants exposed to a high sucrose concentration. Acclimatization of jojoba plantlets was improved by enhancing ventilation.  相似文献

为了探明吲哚丁酸(Indole-3-butyric acid,IBA)对辣椒(Capsicum annuum L.)下胚轴不定根发生的诱导机制,采用IBA浸蘸后扦插的方法,研究了外源IBA处理后辣椒下胚轴不定根发生进程、糖类物质转化与不定根发生相关基因表达情况。结果表明:外源IBA诱导辣椒下胚轴不定根发生具有浓度依存效应,150μmol·L~(-1) IBA处理后不定根发生区段长度、不定根数及平均根长分别是对照的3.47倍、3.28倍和1.91倍;150μmol·L~(-1) IBA上调了不定根启动相关基因Ca ARL1和Ca Cyc B1的表达,下胚轴的不定根原基发生区域明显增加;此外,150μmol·L~(-1) IBA还上调了下胚轴不定根发生区段糖代谢相关基因Ca CWI和Ca STP的表达,可溶性糖、淀粉含量及细胞壁转化酶活性显著提高。结果表明适宜浓度IBA处理可以促进辣椒下胚轴不定根发生区段糖类物质的积累和转化,诱导辣椒下胚轴不定根的发生。  相似文献

Field and pot experiments were conducted to evaluate the effect of co-cultivation and crop rotation on the growth and corm rot disease of gladiolus (Gladiolus grandiflorus sect. Blandus) cv. Aarti caused by Fusarium oxysporum f.sp. gladioli (Massey) Snyd. and Hans. In the field experiment, gladiolus was co-cultivated with 10 agricultural/horticultural crops viz. Allium cepa L., Brassica campestris L., Capsicum annuum L., Eruca sativa Mill., Helianthus annuus L., Tagetes erectus L., Zea mays L., Vinca rosea L. and Rosa indica L., in a soil infested with F. oxysporum. All the crops except V. rosea and R. indica reduced disease incidence. The effect of H. annuus and T. erectus was significant and more pronounced than other co-cultivated crops. In general, root and shoot dry biomass, corm fresh weight, number of cormlets and number of flowers per spike decreased as compared to the un-inoculated monoculture gladiolus treatment (negative control) but these parameters enhanced as compared to the F. oxysporum inoculated monoculture gladiolus treatment (positive control). In a pot experiment, all the crops of the field experiment except V. rosea and R. indica were sown in rotation with gladiolus. Pot grown plants of different species were harvested at maturity and the soil was inoculated with F. oxysporum. Gladiolus was cultivated 1 week after inoculation. Disease incidence was significantly suppressed in all the treatments ranging from 29% to 53%. The highest suppression of disease incidence was recorded in T. erectus (53%) followed by B. campestris (49%). The effect of preceding crops on various vegetative parameters was similar in the pot experiment to that of the field experiment. The present study suggests that corm rot disease of gladiolus can be managed by mixed cropping of H. annuus and T. erectus or cultivation of T. erectus and B. campestris in rotation.  相似文献

In order to reduce unnecessary amount of P-fertilizer and severity of Phytophthora root rot in citrus orchards, the experiment was set up. Thirteen indigenous arbuscular mycorrhiza (AM) fungi species were isolated from rhizosphere soil of citrus orchards in Thailand and were then propagated into three host plants [sorghum (Sorghum bicolor), maize (Zea mays), and leek (Allium cepa)] by trap culture. We also tested whether indigenous AMF species (13 different species) could colonize into three cultivars of citrus scions and rootstocks (Shogun: Citrus reticulata Blanco cv. Shogun; Tangerine: C. reticulata; and C-35 citrange: Citrus sinensis × Poncirus trifoliata). With root colonization rates, the results indicated that Acaulospora tuberculata and Glomus etunicatum provided the best colonization in all citrus cultivars. We selected, therefore, those AMF species to verify their influences on citrus growth and Phytophthora root rot resistance. Three cultivars of citrus scions and rootstocks, Shogun, Tangerine and C-35 citrange, were inoculated with two effective indigenous AMF species, G. etunicatum or A. tuberculata in order to determine the influences on citrus growth. The plants were investigated to determine the mycorrhizal efficiency index (MEI), AM colonization, P content, and other parameters. Co-inoculation of AMF species (G. etunicatum or A. tuberculata) with Phytophthora nicotianae was also carried out in Shogun scion/C-35 citrange rootstocks. The results of citrus growth revealed that Shogun and Tangerine inoculated with G. etunicatum produced the highest MEI. Tangerine and C-35 citrange amended with fertilizers and G. etunicatum showed the highest P content in leaves. This indicated that G. etunicatum has an influence on citrus growth and P uptake, suggesting it to be the highly effective strain. Shogun scion/C-35 citrange rootstock combinations that were inoculated by both P. nicotianae and different AM fungi (G. etunicatum or A. tuberculata) showed root injury at low level of root rot symptom. However, the part of Shogun scions grafted on rootstocks showed severe symptom of shoot die back in treatment inoculated with P. nicotianae alone, while treatment inoculated with different AM species (G. etunicatum or A. tuberculata) and P. nicotianae rendered lower shoot die back symptoms than that of Phytophthora treatment. The low level of shoot die back symptom was shown at first, then healthy young shoot was restored. Our results indicated the facts that different host plants and different AMF species produced different outcomes of growth and pathogen resistance. The application of both AM isolates, therefore, has an enormous potential to be produced the inoculum for citrus orchards.  相似文献

The genus Ptilotus has immense potential for ornamental horticulture but its commercial development has been hindered by propagation limitations. Poor seed quality and germination are reported. Cutting propagation is limited by cutting supply as the juvenile phase of Ptilotus is short. Micropropagation has been used in an attempt to overcome these difficulties but explants become floral in vitro and this causes plantlets to elongate. Ethephon has been used to control flowering of stock plants of many ornamental species. This study investigated the effect of ethephon applied to young (3-week-old, deflasked from tissue culture) and mature (1-year-old) Ptilotus plants in a greenhouse. A system of applying gaseous ethylene at 0, 100, 200 and 300 mg l−1 to the headspace of in vitro plantlets in glass jars was developed and the response of in vitro plantlets to ethylene studied. One-year-old Ptilotus plants were treated with 500 mg l−1 ethephon 2 days before pruning or 1 or 2 weeks after pruning. Ethephon application 2 days before pruning decreased the number of inflorescences and increased the number of shoots (compared to the control) but was phytotoxic. Ethephon applications of 150 or 300 mg l−1 applied weekly or fortnightly to 3-week-old plants deflasked from tissue culture reduced plant height and number of inflorescences and at low concentrations increased the number of new shoots. A fortnightly application at 150 mg l−1 is recommended. Previous reports on the effects of ethylene on inflorescence production on plantlets in vitro are limited. Our study showed that exposure of in vitro plantlets of P. nobilis to ethylene gas at 100 mg l−1 for 1 h significantly increased the number of shoots and plant height but this did not occur for plantlets of P. spicatus. Plantlets of P. spicatus exposed to transient ethylene at 200 and 300 mg l−1 showed significantly greater rooting (52.4%) than the control (13.6%).  相似文献
