In the current work attempts were made to investigate culture of leaf explants derived from in vitro seedlings of two sweet orange (Citrus sinensis (L.) Osbeck) cultivars, Bingtangcheng and Valencia. Effects of several factors, including culture medium, lighting condition, explant age and genotype on regeneration response were examined based on three parameters, percentage of explants producing shoots, mean number of shoots per explant and shoot forming capacity. Culture of the explants on shoot-inducing media (SIM) composed of MT salts supplemented with different growth regulators gave rise to disparate shoot regeneration, in which SIM1 (MT + 0.5 mg L−1 BA + 0.5 mg L−1 Kinetin + 0.1 mg L−1 NAA + 3% sucrose + 0.8% agar, pH 5.8) was shown to be the most effective medium for direct induction of shoots from leaf explants. Highly significant difference in the response of shoot bud regeneration was noted between the two cultivars, with Bingtangcheng being more responsive than Valencia. Culture of explants from fully developed leaves led to better shoot regeneration capacity in comparison to undeveloped ones. However, the two lighting conditions used herein did not cause significant difference in shoot regeneration. Phenotypic observation and randomly amplified polymorphic DNA (RAPD) analysis confirmed that all the regenerated plants from both genotypes were genetically identical to their donor plants, suggesting absence of detectable genetic variation in the regenerated plants. The data presented here demonstrated that direct initiation of plants from leaf explants has been successfully accomplished. To our knowledge, this is the first report on direct regeneration of shoots from leaf explants in Citrus, which will provide an alternative source for citrus genetic manipulation in the future.  相似文献

Sweet orange (Citrus sinensis L. Osb.) ‘Hamlin’ is a canker (Xanthomonas axonopodis pv. citri: Xac) susceptible citrus genotype grown commercially worldwide. Canker causes severe economic losses and restricts the marketability of crop for export. Little is known about the role of oxidative stress in canker development. In the present investigation, sweet orange ‘Hamlin’ leaves were artificially inoculated with Xac to determine the impact of Xac infection on hydrogen peroxide (H2O2) metabolism. Characteristic symptoms following artificial inoculation were water soaking of the infiltrated zone between 2 and 8 days after inoculation (dai); raised epidermis accompanying tiny yellow colored bacterial colonies at 8 dai; and yellowing and necrosis of the infected zone by 12–16 dai. In planta Xac population increased 1000 fold by 14 dai from an initial population of 7.3 × 106 cfu cm−2 (0 dai). Peak concentrations of H2O2 were observed at 24 h and between 8 and 10 dai and coincided with higher activity of total superoxide dismutase (SOD). Lower levels of H2O2 in infected leaves were maintained by Xac induced higher activities of catalase (CAT), ascorbate peroxidase (APOD), and guaiacol peroxidase (POD). It appears Xac altered H2O2 metabolism in C. sinensis L. Osb. ‘Hamlin’ to enhance survival and growth.  相似文献

Inter-section hybrids were obtained in the reciprocal crosses between Primula filchnerae (2n = 2x = 24) of Sect. Pinnatae and P. sinensis ‘Fanfare’ (2n = 2x = 24) of Sect. Auganthus by rescuing ovules on half-strength (1/2) Murashige and Skoog's (MS) medium supplemented with 50 g l−1 sucrose, 2.5 g l−1 gellan gum, 0.1 mg l−1 α-naphthaleneacetic acid (NAA), 0.1 mg l−1 6-benzyladenine (BA) and 50 mg l−1 gibberellic acid (GA3). In ovule culture, germination occurred with radicle elongation but no plumule was observed. The radicle kept on the initial medium showed root proliferation with callus formation. When the calluses were transferred to (1/2)MS media containing 30 g l−1 sucrose and 3 g l−1 gellan gum, without plant growth regulators (PGRs) or with 1 mg l−1 zeatin and 0.1 mg l−1 NAA, plantlets were regenerated. The plants thus obtained were confirmed to be hybrids through flow cytometry (FCM) and random amplified polymorphic DNA (RAPD) analyses. The hybrid obtained when P. filchnerae was used as the maternal parent was diploid, whereas hexaploid hybrid was obtained when using P. sinensis as the maternal parent. The hexaploid hybrid might be produced through chromosome doubling of a triploid originated from the fertilization of P. sinensis with unreduced pollen of P. filchnerae.  相似文献

Inter simple sequence repeats (ISSR) polymorphism in Citrus indica Tanaka (Rutaceae), an endemic and threatened wild species, was examined along with three other closely related wild taxa (C. medica L., C. latipes (Swingle) Tanaka, and C. sp. ‘Memang athur’) by analyzing 53 representative accessions sampled from North-east India. Jaccard's similarity values among 53 accessions of Citrus ranged from 0.46 to 0.97 (average = 0.75). Genetic similarity values among all the 34 accessions of C. indica were found in the range of 0.82 to 0.97 with an average of 0.90. Heterozygosity (Ht = 0.123) and Shannon's information index (I = 0.188) values estimated for C. indica revealed significantly low level of genetic variation within the species. UPGMA dendrogram grouped all 53 accessions of Citrus into four major clusters: Cluster I – C. latipes; Cluster II – C. medica; Cluster III – ‘Memang athur’ and Cluster IV – C. indica. The dendrogram placed all the 34 accessions of C. indica in five sub-clusters under Cluster IV. The placement of C. indica accessions in various sub-clusters and groups in the dendrogram was based on molecular differentiation of individual accessions rather than their geographical origin. Very low genetic diversity and destruction of its natural habitat pose serious threat to C. indica even in the Citrus Gene Sanctuary in Nokrek Biosphere Reserve (NBR) in Meghalaya. Low genetic variability, heterozygosity and Shannon's information index in C. medica, C. latipes and ‘Memang athur’ are also concerns that need to be addressed for developing appropriate strategies to conserve the genetic diversity extant in these valuable genetic resources.  相似文献

This study was initiated to investigate the differences in germination percentages and rates between Corylopsis coreana Uyeki and Corylopsis sinensis var. calvescens Rehder & E.H. Wilson following a warm stratification (WS) and cold stratification (CS), and to study the effect of different WS temperatures interacting with different durations of CS. Warm stratification at 10 °C, 15 °C, 20 °C, and 25 °C was given for 1 month (1 M 10 °C, 15 °C, 20 °C, and 25 °C WS) followed by 0 M, 1 M, 2 M, and 3 M of CS at 5 °C (0 M, 1 M, 2 M, 3 M CS) and seeds were germinated in an air conditioned greenhouse maintained at 18.5 °C/18 °C. On average, less than 1% of C. coreana seeds germinated when sown without any WS and CS or with 1 M 15 °C, 20 °C, and 25 °C WS without CS treatment. However, 26% C. coreana seeds germinated after 1 M 10 °C WS without any CS treatment. Germination was not affected by WS temperatures when followed by 2 M 5 °C CS. It is concluded that C. coreana exhibited low seed germination at 10 °C and that this temperature could be considered the upper limit of CS for C. coreana. Only 2 M CS was required for more than 90% seeds to germinate. However, C. sinensis var. calvescens required longer than 3 M CS for more than 29% seeds to germinate. This clearly shows that there is an interspecific variation in optimum dormancy-breaking requirements.  相似文献

Transgenic Citrus sinensis (L.) Osb. cv. Hamlin plants expressing the hrpN gene were obtained by Agrobacterium tumefaciens (Smith and Towns) Conn-mediated transformation. hrpN encodes a harpin protein, which elicits the hypersensitive response and systemic acquired resistance in plants. The gene construct consisted of gst1, a pathogen-inducible promoter, a signal peptide for protein secretion to the apoplast, the selection genes nptII or aacC1 and the Nos terminator. The function of gst1 in citrus was evaluated in transgenic C. sinensis cv. Valencia harboring the reporter gene uidA (gus) driven by this promoter. Histochemical analysis for gus revealed that gst1 is activated in citrus leaves by both wounding and inoculation with Xanthomonas axonopodis Starr and Garces pv. citri (Hasse) Vauterin et al. Genetic transformation was confirmed by Southern blot hybridization in eight cv. Hamlin acclimatized plants. RT-PCR confirmed hrpN gene expression in seven cv. Hamlin transgenic lines before pathogen inoculation. Some hrpN transgenic lines showed severe leaf curling and abnormal growth. Six hrpN transgenic lines were propagated and evaluated for susceptibility to X. axonopodis pv. citri. RT-PCR confirmed gene expression in all six hrpN transgenic lines after pathogen inoculation. Several of the hrpN transgenic lines showed reduction in susceptibility to citrus canker as compared with non-transgenic plants. One hrpN transgenic line exhibited normal vegetative development and displayed very high resistance to the pathogen, estimated as up to 79% reduction in disease severity. This is the first report of genetic transformation of citrus using a pathogen-inducible promoter and the hrpN gene. Further evaluations of the transgenic plants under field conditions are planned. Nevertheless, the evidence to date suggests that the hrpN gene reduces the susceptibility of citrus plants to the canker disease.  相似文献

Paphiopedilum armeniacum, Paphiopedilum micranthum and Paphiopedilum delenatii are endangered orchid species. These three Paphiopedilum species and their hybrids are difficult to distinguish morphologically. In this study, rDNA-ITS (internal transcribed spacer) sequences were used to design species-specific SCAR (sequence characterized amplified regions) markers to distinguish P. armeniacum, P. micranthum, P. delenatii and their respective hybrids. The developed markers efficiently amplified 600 bp DNA product for P. armeniacum and its hybrids (SCAR-600armF/Pap-ITS2R), 300 bp product for P. delenatii and its hybrids (SCAR-300delF/Pap-ITS2R) and 700 bp product for P. micranthum and its hybrids (SCAR-700micF/Pap-ITS2R). The effectiveness of designed species-specific markers was also confirmed by using multiplex polymerase chain reaction amplification with a combination of developed three SCAR markers.  相似文献

Agrobacterium tumefaciens-mediated transformation has been one of the methods used to generate transgenic plants in bell pepper. An alternate transformation method that avoids/minimizes tissue culture would be beneficial for the improvement of bell pepper due to its recalcitrant nature. In this report, transgenic bell pepper plants have been developed by a tissue-culture-independent A. tumefaciens-mediated in planta transformation procedure. In the present study, two open pollinated varieties viz., Arka Gaurav and Arka Mohini were used for transformation. Agrobacterium strain EHA105 harboring the binary vector pCAMBIA1301 that carries the genes for β-glucuronidase (uid A) and hygromycin phosphotransferase II (hpt II) was used for transformation. GUS histochemical analysis of T0 and T1 plants at various stages of growth followed by molecular analysis using PCR, Southern analysis and RT-PCR allowed selection of transgenics. The method resulted in 17.8% and 11.4% of the T0 plants in Arka Gaurav and Arka Mohini being selected as chimeric and 35.0% and 29.7%, respectively, were identified as stable transformants in the T1 generation based on PCR analysis.  相似文献

This work investigated the fingerprinting and phenotyping of Citrus germplasm; species selected were of historical importance belonging to Citrus limonimedica Lush. and its supposed ancestors, along with some other species of the Citrus genus. An integrated approach based on the exploitation of nuclear DNA content, morphological traits and molecular markers, such as RAPD fingerprints and ITS-based SNPs, was employed. We studied a core collection of 54 distinct accessions, including 43 genotypes of the Citrus species (18 species or supposed species) and 11 genotypes of the Poncirus genus, which was used as the reference outgroup. Morphological trait analysis and statistical analysis of DNA content and markers were useful for reconstructing a Citrus phylogeny. In particular, our experiments aimed at estimating the genetic variation within and the genetic relatedness among C limon (L.) Burm., C. limonimedica and C. medica L. to shed light on the hybrid origin hypothesis of C. limonimedica. The results of the multidisciplinary analyses allowed us to confirm a remarkable differentiation between Poncirus and Citrus genera and to highlight a close relationship among the three investigated Citrus species but a distinct difference between these three species and other species in the Citrus genus. RAPD fingerprints and ITS polymorphisms enabled us to point out a variation gradient between lemon and citron, with C. limonimedica as a possible intermediate species. Some accessions of C. medica and C. limonimedica that deviate from such a trend suggest recurrent introgression and/or hybridisation with other species of Citrus.  相似文献

Seven chilli landraces of Manipur belonging to three cultivated species of Capsicum (Capsicum annuum, Capsicum frutescens, and Capsicum chinense) form economically important food crops of the region. The genotypes were characterized using ten random amplified polymorphic DNA (RAPD) markers. The cluster analysis based on Jaccard's similarity coefficient calculated by UPGMA method differentiated the genotypes into two main cluster groups. One cluster represented the C. annuum genotypes while the other cluster represented the C. frutescens and the C. chinense genotypes. C. chinense genotypes were more close to C. frutescens genotypes. Genetic variation between the C. frutescens genotypes was more than among the C. annuum genotypes and the C. chinense genotypes were the least similar ones.  相似文献

The regenerability of three ornamental species—Lysimachia christinae, Lysimachia rubinervis and Lysimachia nummularia ‘Aurea’, were investigated using in vitro leaves and shoot tips. 6-Benzylaminopurine (BAP) and α-naphthalene acetic acid (NAA) added to Murashige and Skoog (MS) medium were tested for their effect on organogenesis. On the medium, shoot regeneration occurred directly without callus formation. In these species, L. christinae developed the highest regeneration rate and numbers of shoots/explant from shoot tips (100%, 12.25) and leaf bases (100%, 13.01) on the MS medium containing 3.0 mg l−1 BAP and 0.1 mg l−1 NAA. For L. rubinervis, the highest shoot induction rate and number of shoots/explant were obtained from shoot tip (100%, 16.87–17.20) on the MS medium with 0.1 mg l−1 NAA and 3.0–5.0 mg l−1 BAP. L. nummularia ‘Aurea’, however, showed the highest regeneration rate and number of shoots/explant (100%, 12.73) from leaf bases on MS medium supplemented with 1.0 mg l−1 BAP and 0.1 mg l−1 NAA. All in vitro shoots rooted well on half macronutrient MS medium containing 0.1 mg l−1 NAA. After acclimatization, transplanted plantlets grew normally and flowered in the field.  相似文献

Prunus microcarpa C.A. Mey. subsp. tortusa is a deciduous shrub well adapted to severe winter and dry-hot summer conditions. As the first step to explore the genetic and horticultural potential of P. microcarpa C.A. Mey. subsp. tortusa, we used SSRs to elucidate the genetic variation within its populations dispersed in upper Mesopotamia. We also investigated its phylogenetic relationship with economically important Prunus species; almond, apricot, sweet cherry, peach and plums. Using 47 amplifying SSR primer pairs, 63 P. microcarpa C.A. Mey. subsp. tortusa genotypes sampled from five locations and 15 cultivars belonging to other Prunus species were assayed. The cross-species transportability of SSRs was 96% indicating a high degree of homology between P. microcarpa C.A. Mey. subsp. tortusa and the other Prunus species. The genetic distance between P. microcarpa C.A. Mey. subsp. tortusa genotypes belonging to a particular geographic site was lower than that between genotypes of different geographic origins. Cluster analysis differentiated P. microcarpa C.A. Mey. subsp. tortusa genotypes according to their geographic sites and separated them from the other Prunus species. P. microcarpa C.A. Mey. subsp. tortusa and sweet cherry, the subgenus Cerasus, were located in the same major cluster, the other Prunus species, belonging to the subgenera Amygdalus and Prunus, were located in another one. The analysis of molecular variance (AMOVA) revealed that genetic variation among individuals within populations (59.10%) was much higher than among Prunus groups (29.28%) and among P. microcarpa C.A. Mey. subsp. tortusa populations of different geographic sites (11.61%). The results indicate a substantial genetic diversity in P. microcarpa C.A. Mey. subsp. tortusa and the need of exploring a wider area to increase the chance of finding a particular genotype.  相似文献

The essential oils of aerial parts of Achillea gypsicola Hub.-Mor., Hypericum scabrum L., Satureja hortensis L., and Origanum acutidens (Hand.-Mazz.) Letswaart were analyzed in this study by GC and GC–MS and their oils were tested for toxicity against broadbean weevil (Bruchus dentipes). A. gypsicola oil contained camphor (40.17%), 1,8-cineole (22.01%), piperitone (11.29%), borneol (9.50%) and α-terpineol (1.56%) as major components. A total of 74 components were identified by GC–MS in H. scabrum oil, including α-pinene (9.26%), terpinen-4-ol (5.12%), camphor (5.94%), δ-cadinene (4.52%), pulegone (4.45%), γ-muurolene (4.12%), pinocarvone (3.97%) and β-caryophyllene (3.42%) as predominant components. The essential oils of O. acutidens and S. hortensis were characterized by high contents of carvacrol (86.99% and 55.74%), γ-terpinene (0.71% and 20.94%), p-cymene (1.95% and 12.30%), α-terpinene (0.13% and 2.04%) and β-caryophyllene (1.30% and 1.08%). All of the essential oils were toxic to adults of B. dentipes and insect mortality increased with increasing concentration of each oil. The oils (20 μl dose) brought about 100% mortality in 36 h. Although desirable insecticidal activities against the pest were achieved with the oils from all four plant species, S. hortensis and O. acutidens oils were more effective, particularly after 6 h of treatment. The current results concluded that the essential oils, in particular O. acutidens and S. hortensis oils, may be used as potential botanical insecticides against B. dentipes.  相似文献

The aim of this work was to assess the relation between carbohydrate levels and flower and fruit production, as well as the role of carbohydrates on CO2 fixation activity, by analysis of leaves, twigs and roots from the alternate bearing ‘Salustiana’ cultivar of sweet orange (Citrus sinensis [L.] Osbeck). A heavy crop load (on year) did not affect photosynthesis activity when compared to non-fruiting trees (off year). Fruiting trees accumulated most of the fixed carbon in mature fruits, whilst no accumulation was observed in roots before harvest. Non-fruiting trees transported part of the fixed carbon to the roots and mobilize it for growth processes and, at the end of the season (December), store it as reserves. Reserve carbohydrates accumulation in leaves started by early December for both tree types, showing the same levels in on and off trees until spring bud sprouting. A heavy flowering after an off year caused the rapid mobilization of the stored reserves, which were exhausted at full bloom. We found no evidence on carbon fixation regulation by either fruit demand or carbohydrate levels in leaves. Carbohydrate reserves played little or no role over fruit set, which actually relied on current photosynthesis.  相似文献

A study was carried out to investigate the influence of desiccation and freezing followed by various presowing rehydration procedures on the desiccation sensitivity of the seed of Citrus suhuiensis cv. limau madu. The freshly harvested seeds of limau madu were desiccated under a broad range of relative humidity (RH) to various equilibrium water contents (g H2O g−1 dw). The desiccated and desiccated–frozen seeds were either directly sown under germination conditions or subjected to presowing rehydration procedures: seed preheating, prehumidification and osmoconditioning. The hydrated and desiccated seeds were sown in controlled germination conditions and the survival was evaluated 4–6 weeks after sowing. The results showed that desiccation progressively reduced the percentage of normal seedling of the seeds of limau madu and the viability is almost lost at water contents below 0.08 g H2O g−1 dw. The estimated desiccation sensitivity was substantially high (WC50 = 0.143 g H2O g−1 dw) when the desiccated seeds were rapidly rehydrated (uncontrolled rehydration). In contrast, seed prehumidification, preheating and osmoconditioning (controlled rehydration procedures) markedly enhanced normal seedling percentages decreasing the estimated values of WC50 (between 0.08 and 0.127 g H2O g−1 dw). While the rapidly rehydrated desiccated–frozen seeds were almost killed at water content of 0.15 g H2O g−1 dw, prehumidification and preheating have noticeably increased percentage of frozen seeds survival at the same water content. However, at water content of 0.21 g H2O g−1 dw preheating significantly (P < 0.05) increased percentage of normal seedling of the frozen seeds. Seed desiccation markedly reduced the percentages of germinated seeds with multiple seedlings. Seed controlled rehydration remarkably increased the survival of polyembryos. The beneficial effect of seed controlled rehydration on the survival of the desiccated seeds was pronounced at medium water contents (0.08–0.25 g H2O g−1 dw).  相似文献

Analysis of polymorphism in ISSR amplicons revealed diverse genetic relationship between Citrus indica and five other citrus species. In a consensus UPGMA dendrogram, based on Nei and Li's distance matrices, the C. indica samples from three different sites of Meghalaya, India were clustered together with 99% bootstrap support. C. reticulata, C. sinensis, C. aurantifolia formed a cluster with 67% bootstrap separation. C. macroptera and C. maxima samples from two sites of Meghalaya formed separate clusters with respectively 100% and 98% bootstrap supports. Principal Components Analysis projected C. indica to be more closely related to C. aurantifolia than to the other citrus species of the study. PCA also exposed variations within the C. maxima and C. macroptera samples.  相似文献

Oil spotting or oleocellosis, is a major problem in citrus crops. As the rootstock and fertilization play important roles in citrus growth and fruit development, we investigated the influence of different rootstocks on the growth, mineral nutrition metabolism, water relations, and fruit oleocellosis of eight-year-old field ‘Hamlin’ sweet orange trees. Trees grafted onto Lichi16-6 trifoliata (Poncircus trifoliate) had the greatest rate of oleocellosis (RO), and trees grafted onto Goutou orange (Citrus aurantium) had the greatest degree of oleocellosis (DO). In contrast, trees grafted onto Rangpur lime (Citrus limonia Osbeck) had the lowest RO and DO. Trees were the most vigorous on Rangpur lime rootstocks, followed by Lichi16-6 trifoliata, and then Goutou orange. In addition, because the scion/stock girth ratio showed significant correlations with the RO and DO, oleocellosis parameters can be a good indicator of scion/stock affinity. The total N, total P, Ca2+, and Mg2+ in leaves from trees on Rangpur lime were significantly lower than in leaves from trees on Goutou orange or Lichi16-6 trifoliata. In addition, the RO showed a significant correlation with the leaf Ca2+ and S concentrations and with the peel Mg2+ concentration. The DO was significantly correlated with the total peel N and S concentrations. In addition, the RO showed a significant correlation with the net assimilation of CO2 (ACO2), stomatal conductance (GS), transpiration rate (ET), and water-use efficiency (WUE). However, the DO showed a significant correlation with the GS, ACO2, and WUE. Taken together, these results indicate that rootstocks affect the development of oleocellosis in ‘Hamlin’ sweet orange due to their effects on the mineral nutrition balance and water relations.  相似文献

This work reports the transferability and polymorphism of previously reported SSRs in 10 Prunus species. The availability of a large number of SSRs in the genus Prunus makes marker choice random, while preventing comparison of results in fingerprinting studies. The availability of SSR markers, polymorphic in a wide sample of Prunus species, would facilitate marker choice, while allowing the comparison of results. In this work, microsatellite markers useful for analyzing 10 different Prunus species (P. persica, P. dulcis, P. armeniaca, P. domestica, P. insititia, P. salicina, P. cerasifera, P. avium, P. cersus and P. mahaleb) were searched through screening SSRs previously reported to be conserved and/or polymorphic in more than one Prunus species. A selected group of 13 SSRs, transferable to the 10 species, was analyzed in terms of their usefulness for analyzing these species. The amplification range, polymorphism and variability detected by these loci are reported. The information provided will be useful for Prunus genetic studies as well as conservation and management of Prunus germplasm resources.  相似文献

Apple cultivars from Canary Islands can possibly be valuable genetic resources for subtropical areas. We localised 31 accessions considered by growers to be local, and confirmed by historical references that apple crop was introduced in XV century. These accessions were compared with 77 Spanish and 26 commercial cultivars in order to detect synonyms. A set of 10 SSRs were studied, and 2 of them presented 2 loci. Cultivars from La Palma (Canary Islands) presented five specific alleles not found in other Spanish regions. Those polymorphisms allowed detecting one introgressant in La Palma from non-native cultivars, and the other 30 accessions were classified into 14 genotypes. Some accessions derived from non-native cultivars such as Golden Delicious. A main cultivar could be detected, Del País, with 14 accessions. Secondary ones were Camuesa and Pero. Genetic differentiation was small between regions (Fst = 0.057) but significant, confirmed by analysis of molecular variance (AMOVA). Major genetic differentiation was found between non-native cultivars and cultivars from Asturias and Basque Country. Bayesian method and admixture analysis reconstructed three ancestral groups (RPP), Asturian and Basque cultivars grouped in RPPI (mainly those used for cider production), a mixture of cultivars from Galicia and La Palma in RPPII and non-native cultivars were in RPPIII. This genetic differentiation was also confirmed by factorial correspondence analyses (FCA). AMOVA over RPPs increased the genetic differentiation. Allelic variation found in this study showed that Spanish local cultivars represent a differentiated genetic pool that will provide original genotypes to diversify the reduced number of cultivars used in commercial production. The objective of this study was to produce interspecific hybrids between an Ogura-cytoplasmic male sterile (CMS) line of zicaitai (Brassica campestris var. purupurea, 2n = 20) and cultivars of ornamental kale (Brassica oleracea var. acephala, 2n = 18) to develop a CMS system for hybrid seed production. Pollination with pollen grains of ornamental kales irradiated at a power output of 9.0 mW with a He–Ne laser for 3 min could overcome the cross-incompatibility between the species concerned. Intact hybrids could be efficiently produced from ovules cultured on Murashige and Skoog media supplemented by 0.2 mg l−1 6-benzyladenine. Chromosome number of hybrids was confirmed to be 2n = 19. Hybrids resembled ornamental kales in leaf morphology and in vernalization response. Pollens of hybrids had a sterile appearance. Moreover the hybridity of the putative hybrids was confirmed by RAPD data on a DNA fragment of 820 bp.  相似文献
