Summary Among the nematodes infesting chickpea (Cicer arietinum L.) plants in Syria, cyst nematode (Heterodera ciceri Vovlas, Greco et Di Vito) is the most important. It is uneconomical to grow chickpea in fields infested with cyst nematode and to control this nematode with nematicide. Therefore, investigations were conducted at ICARDA, Syria from 1987 to 1991 to identify sources of resistance to cyst nematode in 7258 lines of C. arietinum and 102 lines of eight annual Cicer species including C. bijugum K.R. Rech. (13 lines), C. chorassanicum (Bge) M. Pop. (3 lines), C. cuneatum Hochst. ex Rich. (3 lines), C. echinospermum P.H. Davis (8 lines), C. judaicum Boiss. (18 lines), C. pinnatifidum Jaub. & Sp. (18 lines), C. reticulatum Ladiz. (36 lines), and C. yamashitae Kitamura (3 lines). All lines were grown in a greenhouse at 15–25°C in pots containing soil infested with 20 eggs of the nematode g-1 soil. Nematode infestation was evaluated on a 0 to 5 scale based on number of females and cysts on roots. Resistance was found in one line of C. bijugum, six lines of C. pinnatifidum, and one line of C. reticulatum. No lines of C. arietinum, C. chorassanicum, C. cuneatum, C. echinospermum, C. judaicum, or C. yamashitae was resistant to cyst nematode. We examined variation at four isozyme and seven microsatellite loci in natural populations of Brassica oleracea L. on the coast of Dorset. All loci were polymorphic, and the diversity index of isozyme loci was similar to that of the microsatellites. Both microsatellites and isozymes showed significant spatial differentiation of genetic variation, but the scale differed between the marker types. Significant non-random mating at isozyme loci was detected within small groups of about five plants, in which adjacent individuals were separated by less than 5 m. On the other hand, non-random mating at microsatellite loci was only detected within the Dorset region as a whole. FST was significant for all loci and there was evidence of isolation by distance at both microsatellite and isozyme loci. From these data we infer that there is low but significant amounts of gene flow among B. oleracea populations in Dorset. The differences in the genetic structure between the two types of marker may be due to higher mutation rates at the microsatellite loci; however, the data suggest that stepwise gain or loss of single repeat units is not the principal mechanism. 相似文献

Information on genetic diversity and genetic relationships among genotypes of Brassica carinata is currently limited. The objectives of this study were to evaluate patterns and levels of genetic diversity in B. carinata based on amplified fragment length polymorphisms (AFLP) as compared with Brassica juncea and Brassica nigra, and to evaluate agronomic and seed quality data for plants grown in the field in western Canada. A total of 296 AFLP bands
were generated from four primer pair combinations and scored for presence/absence in 66, 20 and 7 accessions of B. carinata, B. juncea and B. nigra, respectively. B. carinata was less genetically diverse than the other two species. Differences in diversity were evident in the proportion of polymorphic
loci within each species: 23, 35 and 50% for B. carinata, B. nigra and B. juncea, respectively. Pair-wise similarity measures based on the Jaccard coefficient were highest among accessions of B. carinata and showed the narrowest range: 0.911 (0.810–0.981) compared to B. nigra: 0.569 (0.438–0.660) and B. juncea: 0.715 (0.345–0.951). AFLP-based genetic distance information can be used by plant breeders to select diverse genotypes.
AFLPs are also useful for fingerprinting cultivars and two primer pair combinations were sufficient to uniquely identify all
the accessions of B. carinata. More variation among accessions was identified in the agronomic trial than had previously been described in studies of B. carinata in western Canada, but the data were too limited to draw conclusions regarding specific accessions. Overall, the findings
were in agreement with other published work describing the favourable agronomic potential of this species. 相似文献

An isolate ofPuccinia hordei (ND89-3) originally collected in Morocco is virulent on most barley genotypes reported to possess resistance, except cultivar
Estate (CI 3410), which possesses theRph3 gene and exhibits a low to intermediate level of resistance (infection type 12). Isolate ND89-3 possesses one of the widest
virulence spectrums reported forP. hordei. Accessions ofHordeum vulgare (1,997 in total) andH. spontaneum (885 in total), mostly originating from the Mediterranean region and parts of North Africa, were evaluated with isolate ND89-3
at the seedling stage to identify new sources of leaf rust resistance. Fifty-eight accessions ofH. vulgare, and 222 accessions ofH. spontaneum exhibited low infection types to this isolate. Further evaluations of these resistant accessions with isolates ofP. hordei virulent forRph3,Rph7, andRph12 suggested that most of the resistantH. vulgare accessions possess theRph3 gene. Data suggested additional sources of effective resistance inH. vulgare are rather limited. FiveH. vulgare accessions and 167H. spontaneum accessions were identified as possible sources of new genes for leaf rust resistance. These accessions likely possess resistance
genes that are different fromRph1 toRph12, or gene combinations thereof based on their reaction to four leaf rust isolates. Utilization of these accessions in barley
breeding will broaden the germplasm resources available for genetic control ofP. hordei.

The domestication of cultivated flax (Linum usitatissimum L. subsp.usitatissimum) is briefly discussed.
Using data documented as a matter of routine in genebank work, 63 accessions of cultivated flax from the flax germplasm collection
of the Gatersleben Genebank are compared with 73 accessions of its wild progenitor pale flax (subsp.angustifolium (Huds.) Thell.), which have been observed in systematic field trials. Range of variation, genetically based variation, heritability
and correlation of several characters are considered, especially with respect to the influence of domestication.
Cultivated flax shows higher variation in the characters of generative plant parts, while pale flax varies more in the vegetative
parts of the plant.
The character correlations are similar in both subspecies. Of the 29 characters studied in pale flax the parameters describing
tillering, height of plant, weight of seeds, width of petals and width of leaves are especially suitable for distinguishing
between different accessions. In spite of the high heritabilities of most of the characters, the influence of the environment

The crossing data do not give any further information as to
the genetic differentiation ofB. oleracea during the course of domestication. Introgression between wild and cultivated forms has probably occurred frequently. All
members of theB. oleracea cytodeme belong to the primary and secondary gene pools of the 18 chromosomic crops and are as such of great importance for

Lentil is one of the important cool-season food legumes grown in many countries in the Mediterranean region. But a substantial
yield loss is observed every year due to various biotic stresses. The Sitona weevil (Sitona crinitus Herbst) is a major insect pest limiting lentil productivity mainly in the countries of West Asia and North Africa region.
The adult insects feed on the leaflets at seedling stage, and the plant suffers due to reduced photosynthesis. The larvae
feed on the root systems and on the nodules, thus decreasing the ability of the plant to fix atmospheric nitrogen. Since sources
of resistance to this pest in the cultivated lentil Lens culinaris Medikus subsp. culinaris are lacking, we searched for resistant sources in a collection of wild Lens species available in the ICARDA Gene Bank. We screened 315 accessions of wild lentil covering all known species/sub-species
based on nodule damage at ICARDA’s main experimental station (Tel Hadya, Aleppo), a hot-spot for the pest in the region. Large
variation was observed in the percent nodule damage among accessions across species. Eight accessions, ILWL 110, ILWL 136,
ILWL 166, ILWL 203, ILWL 207, ILWL 245, ILWL 254 and ILWL 258 were identified as resistant, with ≤10% nodule damage, compared
to >56% damage recorded on the cultivated lentil. This is the first report of resistance against Sitona weevil in lentil. One resistant accession ILWL 245 belongs to the species L. culinaris Medikus subsp. orientalis (Boiss.) Ponert, progenitor of the cultivated lentil, which is crossable with the cultivated lentil. This line is being used

When planning optimal conservation strategies for wild and cultivated types of a plant species, a number of influencing biological
and environmental factors should be considered from the outset. In the present study Brassica rapa was used to illustrate this: to develop Scandinavian conservation strategies for wild and cultivated B. rapa, DNA-marker analysis was performed on 15 cultivated and 17 wild accessions of B. rapa plus 8 accessions of the cross compatible B. napus. The B. rapa cultivars were bred in Sweden and Finland in 1944–1997 and the wild B. rapa material was collected from Denmark, Sweden and United Kingdom. The B. napus accessions were bred within the last 20 years in the Scandinavian countries. Results were based on scoring of 131 polymorphic
ISSR markers in the total plant material. A Bayesian Markov chain Monte Carlo (MCMC) approach implemented in NewHybrids demonstrated
a clear distinction of B. rapa and B. napus individuals except for three individuals that seemed to be backcrosses. The backcrossed hybrids descended from two Swedish
populations, one wild and one escaped. The overall pattern of genetic variation and structure in B. rapa showed that cultivated and wild B. rapa accessions formed two almost separated clusters. Geographical origin and breeding history of cultivars were reflected in
these genetic relationships. In addition, wild populations from Denmark and Sweden seemed to be closely related, except for
a Swedish population, which seemingly was an escaped cultivar. The study point to that many processes, e.g. spontaneous introgression,
naturalisation, breeding and agricultural practise affected the genetic structure of wild and cultivated B. rapa populations. 相似文献

Summary During the winters of 1990/91 and 1991/92, 181 accessions of Triticum dicoccon Schrank from the CIMMYT gene-bank were screened in the field for resistance to Russian wheat aphid, Diuraphis noxia (Kurdjumov). Accessions were sown in hill plots of 10 seeds and artificially infested with D. noxia at the two-leaf growth stage. Hills were visually assessed for damage at tillering, booting and heading. Entries differed significantly in their reaction to D. noxia, and severity of symptoms increased with time. Twenty four of the entries were highly resistant to the aphid. In winter 1991/92, 807 accessions of wild and cultivated wheats (26 species) and synthetic hexaploids were screened similarly for resistance to D. noxia. A large number of A-genome species were resistant, while few D-genome species were identified as resistant. These newly discovered sources of resistance can be used to expand the genetic base of resistance to D. noxia in both bread (T. aestivum L.) and durum wheats (T. turgidum L. convar. durum (Desf.) Three hundred and fifty three Triticum accessions, several also classified as Aegilops and comprising 13 diploid, tetraploid or hexaploid species, were screened for seedling and adult-plant resistance to Puccinia triticina Eriks. using a mixture of pathotypes UVPrt2, 3, 9 and 13. Seedlings were spray-inoculated with a suspension of freshly collected urediospores in distilled water containing Tween 20® seven days after planting. Infection types (ITs) were scored 10 days post-inoculation (d.p.i.). Fully expanded flag leaves were inoculated and ITs and leaf rust severity were scored 16 d.p.i. One hundred and eighty two of the accessions were resistant to moderately resistant in the adult stage, whereas 126 were resistant or moderately resistant as seedlings to the pathotype mixture. Hypersensitive adult-plant resistance was particularly apparent in lines of T. timopheevii, T. sharonense, T. longissimum, T. searsii and T. turgidum. In T. turgidum, which comprised 272 accessions, approximately 44% of the adult plants were resistant to moderately resistant compared to 28% of the seedlings. The expression of these adult-plant resistances varied between hypersensitive flecking of flag leaves, and small pustules commonly associated with chlorosis and/or necrosis of leaf tissue. Partial resistance, expressed by small pustules without any apparent chlorosis, was observed in species such as T. tauschii, T. turgidum ssp. durum and T. turgidum ssp. pyramidale. 相似文献

The identification of seedling resistance to white rust of crucifers was performed in a screening of a B. oleracea core collection with 400 accessions representing the genetic and geographic diversity of the species. Fifty seedlings per accession were tested against the Portuguese isolate Ac502 using the methodology and evaluation procedures developed by and . The percentage of resistant seedlings (%R) and the conventional rating criteria of the mean Disease Index (DI) based on the two different evaluation procedures of disease expression used, were compared and adopted as the criteria to rank the accessions for their interest as sources of resistance. A great variability of reactions was found between and within accessions of the core collection, ranging from complete resistance to full susceptibility. Sources of resistance were found namely among the cauliflowers, broccoli and tronchuda cabbages gene pools. Forty-seven accessions presented at least 20% of resistant seedlings. Nine accessions (the kales INRA18 and INRA62, the cauliflowers HRI4856, HRI4866 and HRI5424, the loose-head cabbage HRI11555, the savoy cabbage BRA848, the black broccoli HRI6318 and the Portuguese tronchuda cabbage ISA207) presented 50–78% of resistant seedlings and so they should be considered as potential and useful sources for direct use in breeding programs for white rust resistance. Fourteen inbred lines, representing the full range of disease expression, derived from resistant accessions of the core collection were also tested for resistance to other two Portuguese isolates (Ac503 and Ac504) and to a UK isolate. It is well known that wild carrots may pollute the seed cropsof cultivated carrots, but whether cultivated carrots can alsodisperse pollen and seed, and thereby introduce genes into wildcarrot populations, is only little described. In Denmark, there is nocommercial seed production of carrots, and as biennials they shouldnormally not flower before harvest of the roots. Still, floweringindividuals can be found in most Danish fields, and sometimes in veryhigh numbers. At least 75% of the flowering plants are malefertile, with ca. 83% of the pollen being viable. More thanhalf of the plants produce seeds. Pollen and seed dispersal fromfields into wild carrot populations is probably rather frequent inDenmark. A closer inspection of the morphology of flowering plantsindicate that some of these (2–60%) arebolters of pure cultivar origin, as indicated primarily by orangeroot colour. The remainder is probably first or advanced generationhybrids between wild and cultivated plants, as indicated by theirwhite roots and combinations of morphological characters from eitherplant type. Some of these hybrids are imported to Denmark togetherwith the sowing seed, as indicated by significantly differentfrequencies of bolters with white roots in different carrot cultivarstested in the field. 相似文献

A total of 1,383 accessions of wild barley (Hordeum vulgare ssp. spontaneum) held in the USDA-National Small Grains Collection, Aberdeen were screened for resistance to powdery mildew (Blumeria
graminis f. sp. hordei) in the Czech Republic. They were subsequently tested at the seedling stage for reaction to selected powdery mildew pathotypes and simultaneously for their adult plant resistance in the field. One hundred and twenty-three accessions exhibited resistance to 22 pathotypes in the greenhouse tests (reaction type 2–3 or lower) as well as resistance to the natural population of this pathogen in the field in the Czech Republic during at least 2-year testing. In Sicily and in the small surrounding islands the section Brassica of the genus Brassica comprises five species, B. insularis Moris, B. incana Ten., B. macrocarpa Guss., B. rupestris Raf. and B. villosa Biv. These taxa represent a genetic resource as relatives of kale crops but some populations are endangered or threatened, thus isozyme analyses were performed to assess the genetic diversity degree at population and species levels in order to assist the design of conservation management programs.Eleven loci from five enzyme systems (aconitase, leucine aminopeptidase, 6-phosphogluconate dehydrogenase, phosphoglucoisomerase phosphoglucomutase) were analyzed in sixteen natural population (fifteen from Sicily, one from Calabria). Mean within-population genetic diversity was moderate (P = 41%, A = 1.54, H = 0.16). In some cases a great number of heterozygous individuals were detected, in other cases fixation index (F) deviated significantly from Hardy-Weinberg genotypic expectations.A total of 37 alleles was recognized, six of which resulted exclusive to single populations. The among-population component of the total genetic diversity (Gst mean values) for each species was 0.30–0.37, indicating genetic differentiation among populations.Among B. villosa and B. rupestris populations genetic distance values resulted rather low and they resulted high with B. incana and B. macrocarpa populations.The results are discussed with regard to the distribution of the genetic diversity level and the genetic resources management. 相似文献

Some Arachis species are widely used as commercial plants, e.g. the groundnut A. hypogaea, an important source of good quality protein and oil, and A. pintoi and A. glabrata, that are utilized as forage species. Germplasm of most Arachis species is available in germplasm banks. However, little it is known about the genetic attributes of this germplasm, and mainly about its genetic variability, which is very important for its maintenance. In the present study RAPDs were used to assay the genetic variation within and among 48 accessions of five sections of the genus Arachis and to establish the genetic relationships among these accessions. Ten of 34 primers tested were selected for DNA amplification reactions since they yielded the largest numbers of polymorphic loci. A dendrogram was constructed based on data from the 10 primers selected. Eighty RAPD polymorphic bands were analyzed among the accessions studied. The relationships among species based on RAPDs were similar to those previously reported based on morphological, cytological and crossability data; demonstrating that RAPDs can be used to determine the genetic relationships among species of the different sections of the genus Arachis. In general, wide variation was found among accessions and low variation was found within the accessions that had two or more plants analyzed. However, higher polymorphism was found in the section Trierectoides and in one accession of A. major, indicating that generalizations should be avoided and each species should be analyzed in order to establish collection and maintenance strategies. 相似文献

About 90% of chickpea (Cicer arietinum L.) in the world is grown under rainfed conditions where it is subjected to drought and heat stress. Unlike the cultivated
chickpea, annual wild Cicer species possess sources of resistance to multiple stress; annual wild Cicer species were therefore evaluated for resistance to drought and heat stress. Eight annual wild Cicer species (Cicer bijugum, C. chorassanicum, C. cuneatum, C. echinospermum, C. judaicum, C. pinnatifidum, C. reticulatum, and C. yamashitae) were compared with special checks, the cvs ICC 4958 and FLIP 87-59C (drought resistant) and ICCV 96029 (very early double-podded).
ILC 3279 and 8617 as drought susceptible checks were sown after every 10 test lines. Yield losses due to drought and heat
stress in some accessions and susceptible checks (ILC 3279 and ILC 8617) reached 100%. Accessions were evaluated for drought
and heat resistance on a 1 (free from drought and heat damage)−9 (100% plant killed from drought and heat) visual scale. Four
accessions of C. reticulatum and one accession of C. pinnatifidum were found to be as resistant to drought and heat stress (up to 41.8°C) as the best checks. C. reticulatum should be taken account in short term breeding programs since it can be crossed with the cultivated chickpea. 相似文献

The Hessian fly, Mayetiola destructor (Say), is a major pest of wheat in North Africa, southern Europe, North America, and northern Kazakhstan. It is believed
this pest (like wheat) originated in West Asia. The Syrian Hessian fly biotype has been found to be the most virulent worldwide,
and has been used at the International Center for Agricultural Research in the Dry Areas (ICARDA) for screening wheat and
its wild relatives to identify new sources of resistance. The screening was conducted in an insect rearing room set at 20°C
and 70% RH using a Hessian fly population collected from Lattakia region, Syria. The experimental design was a randomized
complete block with four replications. ‘Nasma’ (bread wheat) and ‘Cando’ (durum wheat) were used as susceptible and resistant
checks, respectively. A total of 623 lines/accessions of wheat and its wild relatives (Aegilops and Triticum) were evaluated. Twenty-nine Aegilops accessions and four synthetic derived bread wheat lines were found resistant. The presence of dead first instars confirmed
the resistance reaction and also showed that antibiosis is the major mechanism of resistance in these materials. These sources
of resistance are used in ICARDA’s wheat breeding programs for the development of Hessian fly-resistant germplasm/varieties. 相似文献
