北方棉区棉花黄萎病菌落叶型菌系鉴定   总被引:19,自引:1,他引:18  
 采用RAPD扩增与温室致病性测定2种方法,以黄萎病菌Verticillium dahliae的5个落叶型、7个非落叶型菌系和V.albo-atrum的2个菌系为对照,对采自北方棉区6省(自治区)的34个棉花黄萎病菌菌系进行致病型鉴定。94.1%的菌系在2种鉴定结果中表现一致,与对照菌系相比较,确定其中26个菌系为落叶型菌系,6个菌系为非落叶型菌系。从而证实了黄萎病菌落叶型菌系在北方棉区河北、河南、山东3省的存在,并发现所确定的26个北方落叶型菌系中的22个与来自美国的对照落叶型菌系T9、V44的关系比与来自江苏的对照落叶型菌系VB、V991的关系更近。本实验还初步筛选到2条用于鉴别V.dahliae落叶型与非落叶型菌系的RAPD特异条带OPB-19966和OPM-201691,将它们用于对34个北方菌系的RAPD扩增鉴定,则与温室致病性测定结果的一致性分别为88.2%和94.1%,证明这2条特异条带在鉴定棉花黄萎病菌落叶型菌系上具有一定的实用价值,并为进一步制作特异探针以形成一套简便、准确、规范化的鉴定技术奠定了基础。  相似文献

A multiplex-nested-PCR procedure was developed for in planta detection of Verticillium dahliae isolates infecting artichoke and assessment of their vegetative compatibility groups (VCGs). PCR markers were identified and assigned to V. dahliae VCGs, including: i) a 334 bp marker amplified from VCG1A or VCG2B334 isolates; ii) a 688 bp marker amplified from VCG2A or VCG4B isolates; and iii) a 688 bp and a 964 bp PCR marker amplified from VCG2B824 isolates. The infecting V. dahliae VCGs were identified in artichoke tissues according to specific patterns of amplified markers after two rounds of PCR. The PCR-based 'molecular tool box' was first optimized using DNA extracted from artichoke plants artificially inoculated with isolates representative of known VCGs. Thereafter, the efficiency of the molecular procedure was tested using DNA extracted from naturally-infected artichoke plants showing a range of symptom severity as well as from symptomless plants. The novel multiplex-nested-PCR assay was clearly superior in detecting the pathogen compared to conventional isolation procedures, and in addition was informative about the VCGs. Moreover, the PCR method allowed the detection and VCG identification of V. dahliae infections in symptomless but infected plants, which had yielded false negatives when checked by microbiological isolation procedures. This 'molecular tool box' has uncovered the presence of several V. dahliae VCGs infecting the same artichoke plants in the Comunidad Valenciana Region. In addition, it is useful for genetic and pathogenicity diversity studies of V. dahliae populations infecting artichoke, and may help in predicting the severity of verticillium wilt epidemics.  相似文献

为探讨河南商丘地区棉花黄萎病菌的致病型群体变异,对该地区棉花上分离的8株单孢菌株的菌落形态、显微结构、致病力、ITS序列、系统进化及菌体蛋白等方面进行了研究。结果表明:这些菌株均属于棉花黄萎病菌Verticilliumdahliae;系统进化树显示8株菌株并没有聚在同一进化枝上;8株黄萎菌菌株存在致病力差异,SQ4菌株致病力最强,属于落叶型,而其它致病力较弱的7个菌株属于非落叶型;不同致病力的菌株间蛋白谱带存在差异。  相似文献

不同植棉省区落叶型黄萎病菌的培养特性及致病力比较   总被引:2,自引:1,他引:1  
近年来落叶型症状的棉花黄萎病对棉花生产的危害越来越大,为明确落叶型棉花黄萎病菌的分化特点,采用特异性分子标记方法,对我国10个主要植棉省区30个落叶型棉花黄萎病菌的培养特性和致病力进行了比较。落叶型黄萎病菌系在我国主要植棉省区已普遍存在,该菌系以菌核型为主,其微菌核呈放射状或环状。63.3%菌系的菌丝发达致密,其余菌系的菌丝较为疏松。落叶型黄萎病菌系之间的产孢量和致病力差异较大,产孢量变幅为3.7×107~18.8×107个孢子/mL;71.9%为强致病力菌系,25.0%为中等致病力。研究表明,不同植棉省区间落叶型黄萎病菌系的产孢量和致病力具有一定差异。  相似文献

以14个黄萎病菌代表菌系为对照,对来自我国北方棉区的34个棉花黄萎病菌菌系进行RAPD分析。选用对所有供试菌系都有扩增条带的14个引物,取其结果中稳定性和多态性均好的65条谱带作类平均法系统聚类分析,建立树状图。将上述48个菌系分为4大类,结果表明北方棉区河北、河南、山东的部分棉田存在黄萎病菌落叶型菌系的危害,而且85.7%的落叶型菌系与对照的美国落叶型菌系T9、V44的亲缘关系比与对照的江苏落叶型菌系V、V991更接近。  相似文献

新疆北部棉区黄萎病菌种群致病性分化及变异   总被引:6,自引:2,他引:4  
采用特异性引物PCR检测技术和鉴别寄主法对新疆北部棉区41个棉花黄萎病菌菌系进行检测,以期明确黄萎病菌种群致病性分化及变异。特异性引物(ND1/ND2和D1/D2)PCR检测结果显示,供试菌系中落叶型菌系16个,占39.0%;非落叶型菌系24个,占58.5%;1个菌系未能检测出致病类型。鉴别寄主法测定结果显示:41个供试菌株中致病性强、中、弱的菌系分别为21、12和8个,分别占51.2%、29.3%和19.5%。落叶型菌系的致病力明显高于非落叶型,平均病情指数分别为39.4和25.8。研究表明,无论落叶型菌系和非落叶型菌系,接种后都可产生落叶症状,但落叶的程度有明显差别,其落叶症状的轻重不仅与菌系的致病类型有关,还与品种的抗病性密切相关。  相似文献

采用Nit硝酸还原酶缺陷型突变体技术,共得到棉花黄萎病菌Nit突变体301个,其中A型155个,B型97个,C型29个。共检测到落叶型棉花黄萎病菌11株,除以前报道的6株外,发现了5个新菌株。针刺接种后,在棉株上均引起典型的落叶型症状。通过聚丙烯酰受凝胶电泳比较落叶型和非落叶型的8个代表菌株的蛋白质电泳图谱的差异,发现在浓度为7.5%的凝胶中,落叶型菌株有两条特异性的蛋白,其迁移率分别为Rf5=0.135,Rf9=0.405。用非落叶型菌株DF4、落叶型菌株SY12菌体的可溶性蛋白制备获得两个抗血清。用间接ELISA法检测,即能区分棉花黄萎病菌的立枯、枯萎、炭疽病菌,落叶型和非落叶型黄萎病菌均呈阳性反应,但落叶型黄萎病菌的OD值高于非落叶型黄萎病菌。  相似文献

The inheritance of tolerance to the herbicide metribuzin was studied in two durum wheat cultivars, one of which, 'Anton', is resistant and one, 'Nita', susceptible. Parents, F1, F2 and F3 of the crosses 'Anton' × 'Nita' and 'Nita' × 'Anton' were tested for herbicide response. The character evaluated was the increase in weight of plants treated with the herbicide. As there were no significant differences between progenies of reciprocal crosses, cytoplasm was not involved in tolerance. Tolerance was semi-dominant with means values of F1, F2 and F3 progenies in the range of mid-parent. The heritability of this trait estimated by regression of the average of progeny F3 in their parent F2 had a value of 0.23 ± 0.063 and a value of 0.52 ± 0.150 estimated by the relation V G/ V P for full-sib F2 families.  相似文献

The reactions of several tomato cultivars and the genetic basis of resistance to an isolate of Alternaria alternata f. sp. lycopersici were examined. Among the 105 cultivars tested only two, Regal and Toper, were susceptible, while the remainder were highly resistant. The resistant cultivar ACE 55 VF (P;) was crossed with the susceptible cultivar Earlypak No. 7 (P1)- When F1, F2, F3, BC1, BC1 selfed, F2 X P2 and BC1P2 x P1 progenies were inoculated at the two-leaf stage with a suspension of spores of the pathogen, the ratios of resistant to susceptible plants indicated that resistance was conferred by a single dominant gene.  相似文献

A flowing solution culture technique was used to study the effects of N nutrition on banana root infestation by the burrowing nematode Radopholus similis . Paired groups of banana plants were grown in separate compartments containing coarse sand (2–5 mm diameter). Input nutrient solutions had fixed compositions and identical total N concentrations. The two compartments differed in their NH4+/total N concentrations, one receiving 0% NH4+/total N (100% NO3/total N) and the other 60% NH4+/total N (40% NO3/total N). The compartments were connected by a inverted T-shaped tube containing fine sand (200–250  μ m diameter) through which nematodes were inoculated. The pH and N concentrations of the output solution were determined at regular intervals. The pH of the output solution differed markedly between the two compartments, with values of 6.8 and 3.6 in the 0% and 60% NH4+ compartments, respectively. In the compartment receiving 0% NH4+, the number of nematodes in the roots and the growth of banana plants was significantly higher than in the compartment receiving 60% NH4+. Even though they were more infested by nematodes, banana plants receiving 100% NO3 developed better than the less infested plants grown in the medium containing 40% NO3.  相似文献

Soil drench and stem puncture inoculation were compared as methods for selecting cocoa cultivars with resistance to Verticillium dahliae. Disease progress was more rapid and induced symptoms were more severe following stem puncture and, under glasshouse conditions, differences between cultivars were detected 15 days after inoculation. Moreover, using stem puncture, inoculum densities of 104 conidia/ml were sufficient to differentiate resistant and susceptible cultivars, whereas with the soil drench method, inoculum densities of 107 conidia/ml were necessary. Although a substantially higher proportion of plants were affected by stem puncture inoculation, the resistance of cultivar Pound-7 remained effective at high inoculum densities of 108 conidia/ml. With either method, older seedlings were more susceptible to V. dahliae than younger ones. However, with stem puncture, 15-day-old seedlings were sufficiently susceptible for a valid disease assessment. In contrast, with soil inoculation, 60-day-old plants were required. In a nursery trial with 15-day-old seedlings, seven cocoa genotypes previously selected as resistant, moderately resistant or susceptible to Verticillium dahliae , on the basis of root inoculation, were ranked in the same order when stem punctured. Stem puncture inoculation of young seedlings is cost-effective in terms of time and space, and is therefore recommended for screening of cocoa for wilt resistance, especially in large-scale breeding programmes.  相似文献

 为了探究不同年代的大丽轮枝菌(Verticillium dahliae Kleb.)菌株生物学特性和遗传多样性方面的差异,本文以我国黄河流域和长江流域主产棉区6个省的不同年代大丽轮枝菌菌株为研究对象,观察培养性状,测定致病类型(致病力、落叶型),同时采用ISSR指纹图谱分析其遗传多样性。结果显示,不同年代大丽轮枝菌之间菌丝生长速率无显著差异,但菌核型菌株所占比例有减少趋势;2007~2009年和2017年的菌株全部为落叶型菌株,而1983~2000年的菌株中落叶型菌株仅占28.6%,表明随着年代的推移,黄河流域和长江流域的落叶型菌株所占比例呈上升趋势;不同年代菌株之间致病力存在显著差异,1983~2000年、2007~2009年和2017年的菌株中强致病力类型菌株分别占21.4%、25.0%和38.9%,仅有的5株弱致病力类型菌株均为1983~2000年的菌株;与2000年后的菌株相比,1983~2000年的菌株,Nei′s基因多样性指数为0.205 1,Shannon信息指数为0.299 0,表现出更丰富的多样性,利用NTSYS软件和Structure软件对ISSR指纹图谱进行聚类分析,两种方法均将所有供试菌株分为4个类群,且聚类结果与致病力和不同年代之间均具有一定的相关性,与地理来源无明显相关性。本研究结果表明,过去30年间我国黄河流域和长江流域棉田黄萎病菌落叶型和强致病力类型菌株所占比例逐渐升高,且不同年代和不同致病力的菌株在遗传上有差异,为进一步探究大丽轮枝菌的遗传与进化奠定了基础。  相似文献

SSF-126, ( E )-2-methoxyimino-N-methyl-2- (2-phenoxyphenyl)acetamide, inhibited mycelial growth (IC50 7·7 μM) of Pyricularia oryzae. The inhibited mycelium, which was incubated for 30 h in the presence of SSF-126, grew vigorously after transfer to a fungicide-free medium, indicating that SSF-126 is a fungistatic compound. SSF-126 strongly inhibited oxygen consumption by P. oryzae at low concentrations (IC50 0·8 μM). However, the initial inhibition of oxygen consumption was followed by normal mycelial respiration that was insensitive to SSF-126. These results suggest that SSF-126 has a new type of antifungal action.  相似文献

ABSTRACT To better understand the genetic relationships between Verticillium dahliae isolates from lettuce and other phytopathogenic Verticillium spp. isolates from various hosts and geographic locations, the complete intergenic spacer (IGS) region of the nuclear ribosomal RNA gene (rDNA) and the beta-tubulin gene were amplified and sequenced. The sequences of the complete IGS region and the beta-tubulin gene were used alone and in combination to infer genetic relationships among different isolates of Verticillium with the maximum-likelihood distance method. Phylogenetic analyses set sequences into four distinct groups comprising isolates of V. albo-atrum, V. tricorpus, and V. dahliae from cruciferous and noncruciferous hosts. Within the four Verticillium groups, isolates of V. dahliae from cruciferous hosts displayed the closest affinity to V. dahliae from noncruciferous hosts. Isolates of V. dahliae from noncruciferous hosts could be further divided into four subgroups based on sequence similarities within the IGS region. Cross-pathogenicity tests demonstrated that most Verticillium isolates were as virulent on other hosts as on their hosts of origin. A phenogram based on the cross pathogenicity of individual isolates resembled those derived from the IGS and beta-tubulin sequence comparisons. On the basis of the data presented, the potential origin of some isolates of V. dahliae pathogenic on lettuce is proposed.  相似文献

The effects of a range of concentrations of four nutrients – nitrogen, phosphorus, potassium and calcium – in fertilizer solutions on the severity of anthracnose on strawberry cv. Nyoho cultivated under a noncirculation hydroponics system were determined after inoculation with Colletotrichum gloeosporioides . Crop growth and tissue nitrogen, phosphorus, potassium and calcium contents of the entire above-ground parts of the plant were also investigated. Elevated nitrogen and potassium concentrations in the fertilizer solution increased disease severity in contrast to phosphorus and calcium. Treatment with either NH4 or NO3 nitrogen was not significantly different. The dry weight of the strawberry plants increased significantly with elevated concentrations of nitrogen ( R 2 = 0·9078) and phosphorus ( R 2 = 0·8842), but was not influenced by the elevated amounts of potassium ( R 2 = 0·8587) and calcium ( R 2 = 0·6526) concentrations.  相似文献

One hundred and fourteen isolates of Verticillium dahliae obtained from cotton and eggplant in mainland China were successfully assigned to two vegetative compatibility groups (VCGs) except for one self-incompatible isolate. Eleven isolates were strongly compatible with T9, the tester strain of the cotton defoliating pathotype, forming a linear growth of wild type with abundant microsclerotia and dense mycelia between compatible nitrate-nonutilizing mutants. The remaining 102 isolates were grouped into the non-defoliating VCG2, although the strength of the reaction varied; some isolates were strongly compatible with the tester strain while others were only slightly compatible. All VCG1 isolates including T9 showed the same defoliating symptom in greenhouse inoculation tests. This study confirmed the presence of the defoliating pathotype (VCG1) of V. dahliae in mainland China.  相似文献

用一对棉花落叶型黄萎病菌的特异性引物D1和D2进行PCR扩增,对于落叶型黄萎病菌,该对引物可特异性地扩增产生一段550bp的产物,而非落叶型黄萎病菌则不能被扩增.供试的35个新疆黄萎菌系中,有3个菌系扩增出550bp大小的落叶型黄萎病菌特异性片段,表明目前新疆已存在落叶型黄萎病菌,用此技术可快速、准确地检疫和鉴定落叶型黄萎病菌.  相似文献

棉花黄萎病菌致病型的AFLP分析   总被引:16,自引:0,他引:16  
 选用41个棉花黄萎病菌(Verticillium dahliae)代表菌系,在温室条件下,对4个棉花品种鄂荆1号(感)、中棉所12(耐)、文-5(抗)和唐棉2号(抗)进行致病性测定,结果可将供试菌系分为落叶型与非落叶型2类。选取8对AFLP引物PCR扩增的结果中,统计带型稳定、清晰且有多态性的条带,共169条作系统聚类分析,将上述菌系分为2大类,第一类为非落叶型菌系,包括10个非落叶型菌系和1个过渡菌系;第二类为30个落叶型菌系。根据聚类分析建立树状图,发现菌系与地理来源存在一定的相关性,而依据菌系致病力强弱分类则相关关系不大。选用25对EcoRⅠ和MseⅠ引物组合,对供试的41个V.dahliae进行AFLP扩增,筛选到2对引物E64(GACTGCGTACCAATTCGAC)、M53(GATGAGTCCTGAGTAACCG)和E49(GACTGCGTACCAATTCCAG)、M65(GAT-GAGTCCTGAGTAAGAG),能分别扩增出433bp和110bp2条仅为V.dahliae非落叶型菌系独有的特异片段,可将落叶型与非落叶型菌系分开,这2条特异片段被命名为EM433和EM110。  相似文献
