Changes in the regional cerebral blood flow (rCBF), a measure for regional cerebral metabolism, have been reported in dogs suffering from behavioral problems like shadow chasing, anxiety, and compulsive disorders. The rCBF can be measured with single-photon emission computed tomography. Although the acquisition is performed afterward under general anesthesia, the distribution of technetium-99m-ethylene-diylbis-l-cysteine diethyl ester or 99mTc-ECD, a tracer which becomes trapped in the brain, represents the rCBF at the moment the tracer was administered to the awake dog (fixed or frozen image). The aim of this study was to examine the in vivo stability and the duration of the fixed distribution of this tracer in the canine brain. Three acquisitions were performed 15, 40, and 65 minutes after tracer injection. Total counts and perfusion indices, normalized to the total brain counts and to the cerebellum, were calculated (semiquantification). At T65, significant differences in the perfusion indices occurred compared with at T15. This study shows that in dogs, regional differences in the clearance of 99mTc-ECD result in significant alterations of the perfusion indices from 65 minutes onward after tracer injection. Therefore, it is recommended to start the acquisition between 15 and 40 minutes postinjection in dogs when this technique is used for studying rCBF alterations in dogs with behavioral problems.  相似文献

Background: Among various feed additives currently used in poultry nutrition, an important role is played by bioactive substances, including prebiotics. The beneficial effect of these bioactive substances on the gastrointestinal tract and immune system give rise to improvements in broiler health and performance nutrition, thus increasing the productivity of these birds. An innovative method for introducing bioactive substances into chickens is the in ovo injection into eggs intended for hatching. The aim of the study was to evaluate the development of histomorphological parameters of the duodenum and productivity in chickens injected in ovo with the prebiotic DiNovo~?(extract of Laminaria species of seaweed, BioA tlantis Ltd., Ireland) on d 12 of incubation, under large-scale, high density poultry production conditions.Results: There was no significant impact of the injection of DiNovo~? prebiotic on the production parameters of broiler chickens(body weight, FCR, EBI and mortality) obtained on d 42 of rearing. No significant impact of the DiNovo~? injection on the duodenum weight and length was observed, as wel as on the CSA, diameter and muscular layer thickness of the duodenum. The in ovo injection of DiNovo~? significantly increased the width of the duodenal vil i(P 0.05) and crypt depth(P 0.01) of chickens on d 21 of rearing. Other histomorphological parameters of duodenal vil i at d 42 of chickens rearing such as: the height, width, and cross section area of vil i were significantly greater in chickens from the control group compared to those from the DiNovo~? group(P 0.05 and P 0.01).Conclusions: In conclusion, this study demonstrates that injection of DiNovo~? prebiotic into the air chamber of egg significantly influences the histomorphological parameters on d 21 of rearing without negatively affecting productivity in chickens at the end of rearing.  相似文献

1. The aim of this study was to determine if the pharmacokinetics (PK) of florfenicol (FF) undergo age-dependent changes in broilers. Since drug elimination depends on cardiovascular functions, a haemodynamic study was performed in parallel.

2. Broilers of 0.68, 1.27, 2.45 and 5.13 kg were administered FF in a single intravenous dose of 30 mg/kg body weight. Plasma drug concentrations were determined using high-performance liquid chromatography and PK parameters were calculated using a non-compartmental model. Echocardiography was used to measure haemodynamic functions.

3. During growth, the area under the drug concentration-time curve (AUCinf) increased from 25.7 ± 2.9 to 39.0 ± 8.0 mg h/l. Total body clearance (ClB) gradually decreased from 1.19 ± 0.14 to 0.80 ± 0.15 l/h/kg. Elimination half-life increased from 0.73 ± 0.08 to 1.07 ± 0.07 h, whereas volume of distribution (Vss) remained unchanged. Haemodynamic measurements revealed an increase in cardiac output, from 495 ± 65 to 1303 ± 306 ml/min, in the respective body weight groups.

4. Allometric models for PK and haemodynamic parameters were developed and validated. All models proved to be statistically significant; however, only models for ClB and Vss met stringent validation criteria. Model for ClB was used to calculate an optimal dose for a given age group that provides uniform AUCinf.

5. Age-dependent change in FF kinetics may cause variability in therapeutic response under clinical conditions. A novel approach to the dosing protocol was proposed as a means of optimising therapeutic efficacy.  相似文献

Pulmonary function tests were performed in six healthy calves. Prostaglandin F2 causes severe narrowing of both upper and lower airways (total lung resistance increased, dynamic compliance decreased). Clenbuterol administered intravenously fifteen minutes prior to prostaglandin F2 aerosol, and in increasing doses (0, 0.4, 0.8, 1.2 g/kg), on days 1, 2, 4 and 6 of the experiment, effectively but not entirely suppressed these responses.These data indicate that -adrenergic receptors are present in the bovine airways and that the use of clenbuterol (0.8 g/kg) may be effective in treating clinical respiratory disease such as bronchopneumonia in calves.  相似文献

Muscarinic and -adrenoceptors were identified in airway epithelium, smooth muscle and lung parenchyma from Holstein-Friesian calves and cows and were characterized with [3H]quinuclidinyl benzilate and [3H]dihydroalprenolol, respectively. The muscarinic receptor density in the smooth muscle of cows (B max=4803±245 fmol/mg protein) was 33% greater (p<0.01) than in calves. Low receptor numbers were detected in the epithelium and parenchyma. In both calves and cows, the density of epithelial -adrenoceptors was twice as high as in smooth muscle and parenchyma. The quantity of -adrenoceptors in the tracheal epithelium (B max=994±83 fmol/mg protein) and smooth muscle (B max=492±41 fmol/mg protein) in cows was respectively 37% (p<0.001) and 35% (p<0.01) lower than in calves. Adenylate cyclase (AC) assays indicated that the basal and the (–)-isopropylnoradrenaline- (ISO-) stimulated cAMP production were not significantly different between the calves and cows. After stimulation with NaF, significantly higher cAMP production was found in all tissues from cows. Significant correlations were found between absolute AC responses to NaF and -adrenoceptor density in epithelium (r=–0.75,p<0.001) and smooth muscle (r=–0.63,p<0.01). Few tests have been developed to evaluate the cognitive and motor capabilities of domestic cats, despite the suitability of cats for specific studies of neuroanatomy, infectious diseases, development, aging, and behavior. The present study evaluated a T-maze apparatus as a sensitive and reliable measure of cognition and motor function in cats. Eighteen purpose-bred, specific pathogen-free, male, neutered domestic short-haired cats (Felis catus), 1-2 years of age, were trained and tested to a T-maze protocol using food rewards. The test protocol consisted of positional discrimination training (left arm or right arm) to reach a predetermined criterion, followed by 2 discrimination reversal tests. The 2 reversal tests documented the ability of the subjects to respond to a new reward location by switching arms of the T-maze. Data were collected on side preference, number of correct responses, and latency of the responses by the subjects. Aided by a customized computer program (CanCog Technologies), data were recorded electronically as each cat progressed from the start box to the reward arm. The protocol facilitated rapid training to a high and consistent level of performance during the discrimination training. This learning was associated with a decrease in the latency to traverse the maze to a mean of 4.80 ± 0.87 seconds, indicating strong motivation and consistent performance. When the rewarded side was reversed in the test phase, the cats required more trials to reach the criterion, as expected, but again showed reliable learning. The latency to the reward in the first session of reversal increased 86% from the first to the last trial, indicating that it may provide a useful index of cognitive processing. Latencies subsequently decreased as the new reversal paradigm was learned. This paradigm provides a relatively rapid and reliable test of cognitive–motor performance that can be used in various settings for the evaluation of feline cognitive and motor function.  相似文献

Summary

Following concomitant intravenous administration of Tomanol® and sodium penicillin G to six Dutch Friesian dairy cows a significant decrease in total body clearance of penicillin (34.7%) and a prolongation of the elimination half‐life of penicillin (17.2%) was observed. Tomanol® did not affect other pharmacokinetic parameters such as rate constants of drug transfer (k12/k21, α en β), distribution volume of the central compartment (V1), and extrapolated serum drug levels. Intravenous or intramuscular administration of Tomanol® had no effect on the tissue distribution of penicillin G, because neither a change in the ratios of muscle to serum and of kidney cortex to serum nor a change in an induced steady state level of low penicilline G serum concentrations was observed. From the data obtained it is concluded that concomitant Tomanol® administration with penicillin induces an elevation of the serum penicillin concentration and prolongs the persistence of penicillin residues in carcass meat and organs.  相似文献

Abstract

Sequential growth measurements were made on successive populations of Themeda triandra and Sporobolus fimbriatus tillers initiated at different times of the year. The independent influences of tiller age and time of year on tiller growth were isolated by deriving growth indices for tillers of the same age at different times of the year and for tillers of all ages at each time of the year respectively.

Isolating the effect of plant age on growth showed that T. triandra is most productive under a regime of long periods of undisturbed growth, while S. fimbriatus is better suited to frequent interruptions in growth. Isolating the effect of time of year shows that both species are particularly sensitive to perturbations in spring. However, the photosynthetic efficiency of T. triandra is strongly influenced by time of year while S. fimbriatus seems to be a more opportunistic assimilator of sunlight.  相似文献

The influence of pretreatment with ketoconazole [cytochrome P450 3A (CYP3A) + P-glycoprotein (P-gp) inhibitor], elacridar (selective P-gp inhibitor) and rifampicin (CYP3A + P-gp inducer) on oral morphine pharmacokinetics and pharmacodynamics was investigated in experimental dogs. Seven beagles were used in a four-way crossover design. Morphine hydrochloride was administered orally (2.5 mg/kg) alone (control group CON) or after pretreatment with ketoconazole (group KETO), elacridar (group ELA) or rifampicin (group RIF). Morphine plasma concentrations were analysed by liquid chromatography-tandem mass spectrometry. Sedation scores (none, mild, moderate or severe) were evaluated subjectively. Dogs were significantly (P < 0.05) more sedated after ketoconazole pretreatment. There were no significant differences between group CON and the other pretreatment groups in pharmacokinetic parameters taking both sexes into account. Sex differences were apparent in some pharmacokinetic parameters of morphine. The area under the plasma concentration time curve (AUC(0-∞) ) was significantly higher, and the total body clearance was significantly lower in male compared to female dogs in all treatment groups. Ketoconazole, rifampicin and elacridar pretreatment had no significant effects on morphine pharmacokinetics, although dogs in the ketoconazole group showed higher sedation scores.  相似文献

In injection site sarcoma (ISS) in cats lateral as well as deep margins should be correctly planned for a successful surgical outcome. The discrepancy between clinical and computed tomography (CT) measurements of dimension in resectable tumour has led to possible bias that affects the subsequent surgical dose. The aim of this study was to prospectively investigate the agreement between clinical and CT measurements of dimension in newly diagnosed ISS in cats. Fifty‐three client‐owned cats that underwent both clinical and CT measurements of the length and width of ISS were included. CT measurements showed a tendency towards being larger than clinical dimensions, and this difference increased with increasing tumour size. Based on our results, in further studies focusing on ISS in cats, the kind of assessment used to define tumour dimensions (CT versus clinic) should be declared and specified to properly consider surgical results and prognostic impact of this variable.  相似文献

Palatants may be added to equine feed and medication either during or after manufacturing to enhance product acceptance. Prior studies have examined a variety of palatants but results have been limited and inconsistent. Therefore, the objective of this research was to evaluate topically applied oil–based palatants on feeding preferences in horses. Stock-type horses (n = 10) were used in this paired preference test across a two-phase study. Phase one compared six palatants (banana, anise, peppermint, apple, spearmint, and orange) to a control (corn oil), whereas phase two compared preference among palatants (anise, apple, and peppermint). Feeding stocks were utilized and horses were allowed 15 seconds for olfaction followed by 3 minutes for consumption. Variables recorded included first diet sniffed and consumed, first action, aversive behaviors, excessive salivation, and consumption. Each trial was also video-recorded and number of chews were counted. Data were analyzed using chi-square and t tests in SAS version 9.4 with P < .05 established as significant. Findings from phase one reveal excessive salivation was observed less frequently (P < .05) for most palatants with the exception of peppermint and orange. Orange negatively impacted palatability indicated by less consumption when compared with the control (P = .02), although there was no impact on chews per gram. No difference between control and treatment diets for first sniff or first consumed was observed when analyzed individually in either phase one or two, although there was a moderate positive correlation (ф = 0.39, P = .04) between olfaction and consumption during the peppermint and anise comparison. Consumption as the first action was consistent across all trials (P < .05). Anise was preferred over apple and peppermint as indicated by higher total consumption (P < .05) in phase two. These data indicate that oil-based palatants can affect feed preferences in horses with increased palatability from anise and decreased palatability from orange flavors.  相似文献

The effect of genistein (GEN) on the gene expression level of stromal cell-derived factor-1/CXCL-12 and early growth response gene-1 was studied in ovarian tissue of young and initially ageing (early stages in the ageing process) female rats. Forty, young female Sprague Dawley (SD) rats of 2–3 months old (200 ±20 g) and forty, initially ageing female SD rats of 10–12 months (490 ± 20 g) old were selected. According to the weight, rats were divided into control group, low-dose group (L), medium-dose group (M) and a high-dose group (H) and were given 15, 30 and 60 mg/kg GEN respectively. The positive control (Oestrogen) group was given 0.5 mg/kg diethylstilbestrol. The treatment lasted for 30 days. The mRNA expression of C-X-C motif chemokine ligand 12 (CXCL-12) and early growth response factor-1 (EGR-1) was measured by real-time PCR, and protein expression of EGR-1 was detected by Western blot. When compared to the negative control group (NC), the ovary/body weight ratio in the young rats decreased in the GEN group, but the difference was not significant. Similarly, compared with NC, the ovary/body weight ratio in the initially ageing rats also decreased with the increase in GEN concentration, but the decrease was significant in M and H groups (p < .01). The administration of GEN enhanced both the gene and protein expression levels of CXCL-12 and EGR-1 in the ovary. Pearson's correlation analysis showed a synergistic effect between CXCL-12 and EGR-1. Thus, we conclude that the effect of GEN on CXCL-12 and EGR-1 in the initially ageing group was obvious than that in the younger group.  相似文献

Trypanosoma evansi infection typically produces wasting disease, but it can also develop into a neurological or meningoencephalitis form in equids. Trypanosomiasis in horses was treated with quinapyramine sulfate, and all the 14 infected animals were recovered clinically. After clinical recovery, four animals developed a neurological form of the disease at various intervals. Two of these animals treated with diminazene aceturate recovered temporarily. Repeated attempts failed to find the parasite in the blood or the cerebrospinal fluid (CSF), but all of the animals were positive in enzyme-linked immunosorbent assay. The calculation of the antibody index (AI) in the serum and the CSF and polymerase chain reaction (PCR) analysis of the CSF and brain tissue were carried out to confirm the neuro-infection. We found PCR and AI analyses of the CSF to be useful tools in the diagnosis of the neurological form of trypanosomiasis when the organism cannot be found in the blood or CSF. The increased albumin quotient is indicative of barrier leakage due to neuroinflammation. The biochemical changes in the CSF due to nervous system trypanosomiasis include increases in the albumin quotient, total protein, and urea nitrogen. It seems to be the first report on relapse of the nervous form of trypanosomiasis in equids even after quinapyramine treatment in endemic areas.  相似文献

Animal welfare issues associated with whip use during Thoroughbred races in Australia are of increasing importance as the racing industry seeks to promote the well-being of its equine athletes. Recent data have questioned the justification of whip use as either an accelerator or for steering. The aim of this study was to investigate associations between whip use and prerace variables, including jockey experience, starting price, weight carried, and barrier drawn. If the whip is genuinely used in response to a given horse’s performance, there should be no consistent predictors of whip use. We explored the influence of these variables on official whip counts for the race section 400-200 m from the finish (S2), and the section 200 m to the finish (S1), and found that, for S1 alone and when whip use in S1 and S2 was summed (i.e., to summarize the final 400 m of a race), there were no significant predictors on whipping. However, in S2, apprentice jockeys whipped horses on average >3 times as compared with the non-apprentice jockeys (1.48 vs. 0.45; P = 0.022). These findings suggest that rider inexperience in Thoroughbred racing influences the distribution of whippings imposed on horses as they tire in the penultimate 200 m section and may contribute to the growing debate surrounding whip use in the sport of horse racing. This preliminary report involved only small numbers of horses and jockeys and thus should be viewed as a trigger for larger scale investigation of the prerace predictors of whip use in Thoroughbred racing.  相似文献
