Samples of serum and diaphragm muscle were collected from 100 pigs, and serum samples and oesophagi were collected from 100 sheep. The diaphragm muscle and oesophageal tissues were examined for the presence of macroscopic and microscopic Sarcocystis cysts by compression between trichinoscope plates as well as by tissue digestion with pepsin solution. The sera were examined by the indirect haemagglutination test (IHA), using antigens from Sarcocystis gigantea. With these methods, 95% of the sheep and 43% of the pigs were found to be infected with Sarcocystis sp.  相似文献

Sarcocystis cruzi, S. hirsuta and S. hominis are apicomplexan parasites that affect cattle worldwide with variable prevalence. The aim of the present study was to evaluate the prevalence of Sarcocystis spp. in Argentinean cattle comparing microscopic fresh examination and molecular methods. Blood, myocardium and loin samples were collected in five slaughterhouses from a total of 380 bovines. Origin of animals was representative of the major beef cattle production area of Argentina. Samples were analyzed by fresh microscopical examination, transmission electron microscopy (TEM), IFAT and PCR-RFLP. Thin walled sarcocysts corresponding with S. cruzi were found in 99.5% of heart samples. Sarcocysts were detected in 73.1% of loin samples; 71.5% had S. cruzi cysts and 23.1% had thick walled sarcocysts (S. hirsuta or S. hominis). TEM observation revealed the presence of characteristic S. hominis and S. hirsuta cyst walls in 7 and 1 loin samples respectively. Using IFAT, 379/380 animals had titers 25 or higher, showing a full agreement with fresh examination. Amplification products were detected in 35.5% (135/380) of loin samples; however Sarcocystis species could only be determined by RFLP in 29 samples. Agreement between fresh examination and PCR was low (Kappa value=0.262). This is the first report of S. hominis and S. hirsuta in Argentina. Further studies are needed to improve the sensitivity of molecular methods for species identification, especially for differentiation of S. cruzi and S. hirsuta from the zoonotic species S. hominis. The results of the present study and others focusing on sensitivity and specificity of Sarcocystis spp. diagnostic methods should contribute to improve food safety.  相似文献

Sarcocystis zoites were found in pepsin digests of 68.8% of 157 pigs from Hissar, Haryana. Sarcocystis-infected meat was fed to 4 young dogs and 2 cats. The dogs shed Sarcocystis sporocysts in their faeces 12 days after eating infected meat whereas cats did not shed sporocysts.  相似文献

Muscles from heart, tongue, oesophagus, neck and abdomen from 502 adult water buffaloes (Bubalus bubalis) slaughtered in Ho Chi Minh City, Vietnam, between 1996 and 1997 were examined for Sarcocystis cysts by a combination of ocular and histological examination. Sarcocysts were present in 396 (79%) of the animals and the prevalence increased with age from a 57% infection rate among 2-3 year old animals to 93% among 6-7 year olds. The prevalence was higher in animals originating from the northern part (89%) than in those from the southern part (69%) of the country. Four species of Sarcocystis were identified. S. levinei (74%) was the most common species found, followed by S. fusiformis (41%), S. buffalonis (33%) and S. dubeyi (12%). All four species were present in 8% of the infected animals. The most common site for sarcocyst location was oesophagus, followed by cervical muscles, tongue and heart.  相似文献

The prevalence of Sarcocystis spp. infection was investigated in 605 sheep, 826 goats, 1080 cattle, 580 water buffaloes and 36 camels slaughtered from 1992 to 1996 in the Baghdad area (Iraq) using naked eye examination for macroscopic sarcocysts, and peptic digestion, muscle squash, squeezing methods and indirect fluorescent antibody test (IFAT) for microscopic types. The intestinal stages of the parasite were also studied in dogs experimentally fed with tissues containing microscopic cysts. The percentage prevalence of macroscopic cysts were 4.1, 33.6, 0.2, 15.6 and 0, and of the microscopic type, 97.0, 97.4, 97.8, 82.9 and 91.6 for the above-mentioned hosts, respectively. Among the different organs examined, macroscopic cysts were found to be highest in the oesophagus and the lowest in the heart. Peptic digestion method gave the highest rate (93.3%) followed by indirect fluorescent antibody test (IFAT) (88.6%), squeezing (81.3%), and muscle squash (81.2%). Each infected dog shed a total of about 150-200 million sporocysts. Histologically, developmental stages of the parasite were detected in the small intestinal mucosa of the dogs on Days 7 and 13 post-infection.  相似文献

Pepsin: hydrochloric acid digestion and histological examination of the myocardium of 51 hearts from five to nine month-old lambs from the southern half of the North Island, showed that 47 (92%) were infected with microscopic sarcocysts that are presumed to be dog-derived. Of 157 sarcocysts detected histologically, 32 corresponded morphologically with Sarcocystis ovicanis and 120 with Sarcocystis tenella (Syn. S. arieticanis). A total of 938 faecal samples (461 cattle calves, 264 buffalo calves, 55 lambs, 116 kids and 42 piglets) from different livestock farms and individual small holdings in six targeted states of India were collected and screened by modified Ziehl–Neelsen staining technique to determine the prevalence of Cryptosporidium spp. and its association with age, sex, season and faecal consistency in domesticated animals. Overall, 16.2 % of the animals were positive for Cryptosporidium infection with prevalence of 16.3, 24.2, 1.8, 3.5 and 19.1 % in cattle calves, buffalo calves, lambs, kids and piglets, respectively. The prevalence of infection was significantly higher (p?<?0.05) in bovines (19.3 % cattle and 33.7 % buffalo) below 1 month of age than in animals between 1 and 3 months of age. But in piglets, it was higher in the age group of 1 to 3 months (22.6 %) than in younger animals (9.1 %). Also, higher prevalence (p?>?0.05) was recorded in females than in males. Seasons had a significant effect (p?<?0.05) on the prevalence of infection in large ruminants, with the highest prevalence in monsoon (cattle 28.8 % and buffalo 36.6 %) followed by pre-monsoon and post-monsoon season. However, in case of sheep and goats, the prevalence was higher (p?>?0.05) in post-monsoon than in monsoon season. A high degree of association was noticed between Cryptosporidium infection and diarrhoea in ruminants screened during the present study. But, in case of pigs, the prevalence was higher in non-diarrhoeic than in diarrhoeic animals. Genotyping of Cryptosporidium spp. based on nested PCR amplification of partial 18S rRNA and its subsequent digestion with SspI, VspI and MboII restriction enzymes revealed prevalence of Cryptosporidium parvum in representative number of positive samples of cattle, buffalo and goats.  相似文献

Feral goats (Capra hircus) are commonly found on farmland in certain parts of New Zealand and, in recent years, many have been captured and slaughtered for export. Fresh preparations of microisolated sarcocysts from striated muscle of several domestic reindeer from northern Norway were examined by light microscopy. In cardiac muscle, cysts of S. grueneri were found. In skeletal muscle, cysts of S. rangiferi, S. tarandi and S. tarandivulpes were found in all samples examined. In the abdominal muscles of some reindeer, one or two other types of cysts were found.Cysts of one type were macroscopic in size, and ovoid to cylindrical in shape. The cysts were surrounded by a 8–12 µm thick layer of fibrous material, and measured 1682×910 µm. The cysts had relatively few and irregularly distributed, 20–35 µm long, and 3–5 µm wide, linguiform cyst wall protrusions, which could only be seen after removal of the fibrous layer. These cysts were classified as cysts of S. hardangeri, a species previously described from wild reindeer in southern Norway.Cysts of the other type were long and slender, measuring 5460–12700 (8994 ± 2575) × 95–280 (180 ± 50) µm. The cysts had numerous very fine, flexible, hair-like cyst wall protrusions, which were 8–10 [xm long and less than 0.5 µm thick. These cysts are considered to belong to a new Sarcocystis species of reindeer, for which the name Sarcocystis rangi n, sp. is proposed. The reindeer is recorded as the intermediate host for 6 different species of Sarcocystis.  相似文献

Tissue samples comprising the oesophagus and diaphragm were collected from 400 sheep and 400 goats slaughtered at the abattoirs in the study area. Out of this number, 36 were positive for Sarcocystis cysts (sarcocysts) in sheep and 56 in goats. The sarcocysts in sheep measured 35.7 to 500 microns lengthwise and the cyst-wall 2.4 microns. They were identified to be Sarcocystis tenella. The cysts in goats measured 98 to 700 microns and the cyst-wall 2.7 microns. They were identified to be Sarcocystis capracanis. In both animals species, the sarcocysts were more frequent in the oesophagus than in the diaphragm. All sarcocysts seen were microscopic.  相似文献

Fourteen hundred and ninety-six domestic pigs, Sus scrofa domestica L., from a sub-tropical and high-rainfall area of India were examined over a period of 18 months to ascertain their nematode parasite spectrum. The eleven species recovered, arranged in descending order of prevalence, are as follows: Ascaris suum, Oesophagostomum dentatum, Bourgelatia diducta, Stephanurus dentatus, Globocephalus connorfilii, Physocephalus sexalutus, Ascarops dentata, A. strongylina, Pseudocruzia orientalis, Setaria bernardi and Gnathostoma hispidum. This is also the first report of the occurrence of Setaria bernardi in pigs in India. A. Suum, the most prevalent species (51.67%) found in the pigs of the region, is presumably of zoonotic importance. Another species of public health significance that was encountered in the survey was Gnathostoma hispidum. The overall infection rate was considerably higher (76.42%) in the low-altitude region than in the high-altitude one (62.50%). The highest level of infection (73.2%) was observed during autumn and the lowest (63.0%) in winter.  相似文献

Faecal specimens from 162 piglets, aged 1-5 months, from four farms around Harare were examined for coccidia during 1989-1990. Oocyst count per gram (OPG) of faeces was determined by the McMaster technique. Identification of Eimeria species was done after sporulation of oocysts in 2.5% potassium dichromate solution. Measurements of sporulated oocysts were taken from 20 positive cases by using calibrated eye micrometer. Overall incidence was 25.3% (41/162) varying between 15 and 39.5% at different farms. The OPG ranged between 150 and 11,300. Eight species of Eimeria were identified, namely, Eimeria debliecki, Eimeria perminuta, Eimeria suis, Eimeria polita, Eimeria neodebliecki, Eimeria porci, Eimeria scabra and Eimeria spinosa, shown in the order of their prevalence. Eimeria debliecki was the most predominant, showing highest oocyst counts in 45% of 20 pigs, followed by E. suis (25%) and E. polita (20%). On two occasions only one Eimeria sp. was found while in the other 18 (90%) cases, mixed infections of two to six species were recorded.  相似文献

The prevalence of Treponema hyodysenteriae in faecal samples from healthy pigs of various ages in different farrowing units was investigated.Samples from herds designated as Category I were processed within 2 hrs. of sampling. Samples from herds designated as Category II were transported 2 to 3 days before cultivation procedures started. T. hyodysenteriae was demonstrated in 53.7 % to 93 % of the samples collected from Category I herds. No marked difference in the frequency of positive samples from the various age groups of pigs was found. In Category II herds, the organism was demonstrated in 10 % of the samples.The degree of beta-haemolysis shown by isolated strains was grouped into 3 groups: weak, moderate and strong. Strongly betahaemolytic strains, supposedly enteropathogenic, were demonstrated in all Category I herds. Such strains were found in 4.6 % to 25 % of the positive samples in these herds. In Category II herds, 2 out of 17 positive samples harboured strongly beta-haemolytic strains of T. hyodysenteriae.The amount of growth of T. hyodysenteriae on primary plates inoculated with sample material originating from the 2 categories of herds was mostly moderate or abundant. Strongly beta-haemolytic isolates originating from Category I herds produced abundant growth on primary plates in approx. 60 % of samples harbouring such strains. In samples from Category I herds with strains producing weak or moderate beta-haemolysis sparse and moderate amount of growth of the organism was predominant.  相似文献

Effects of non-lethal Sarcocystis miescheriana infections on the blood coagulation system were investigated. Nine pigs were inoculated orally with 2 X 10(5) sporocysts (Group A) and nine pigs (Group B) served as non-infected controls. Blood samples were taken from the vena jugularis externa every 2 or 3 days until 19 days post-infection (dpi). The following parameters were investigated: partial thromboplastin time (PTT), prothrombin time (PT), thrombin time (TT), thrombin coagulase time (TCT), fibrinogen (FIB), factor (F) VIII, F XI, F XII, antithrombin III (AT III), alpha 2 macroglobulin (alpha 2 MG), alpha 2 antiplasmin (alpha 2 AP), pre-kallikrein (PK), and the number of circulating thrombocytes. All infected pigs suffered from acute sarcocystiosis between 12 and 19 dpi. Clinical illness was most severe from 14 to 17 dpi. At this time, PTT and FIB increased, and TT and TCT decreased slightly. The activities of the clotting factors increased at 17 and 19 dpi. However, only F VIII activity was significantly higher in the infected pigs than in the controls at 17 and 19 dpi. PK was significantly lower in the infected pigs at 12, 14, and 17 dpi. Thrombocyte counts were reduced with the onset of the acute phase of illness and some pigs had marked thrombocytopenia. These results indicate low-grade disseminated intravascular coagulation (DIC) in the course of mild S. miescheriana infections in pigs.  相似文献
