The present work examined the short- and long-term effects of three rearing temperatures on protein metabolism and growth trajectories of Senegalese sole larvae using 14C-labelled Artemia protein as feed. A first feeding trial was performed on larvae reared at 15, 18 and 21 °C (at 26, 17 and 14 days post-hatching (dph), respectively) and a second trial conducted on post-larvae after transfer to the same rearing temperature (~20 °C) (49, 35 and 27 dph, in larvae initially reared at 15, 18 and 21 °C, respectively). Temperature greatly influenced larvae relative growth rate (RGR) and survival, since growth at 15 °C was severely depressed. Protein digestibility and retention was highest at 18 °C during the first trial (85.35 ± 1.16 and 86.34 ± 2.33 %, respectively). However, during the second trial, post-larvae from 15 °C had the highest feed intake and protein digestibility (3.58 ± 1.54 and 75.50 ± 1.35 %, respectively), although retention was similar between treatments. Furthermore, after transfer to 20 °C larvae from 15 °C experienced compensatory growth, which was observed until 121 dph, and confirmed by RGR values, which were significantly higher at 15 ºC than at 21 ºC or 18 ºC. Results from the present study show that Solea senegalensis larval development, survival and protein digestion and retention are highly affected by thermal history.  相似文献

A large effort has been dedicated in the past years to the development of nutritional balanced inert diets for marine fish larvae in order to suppress the nutritional deficiencies of live feed. In this study growth performance, Artemia intake, protein digestibility and protein retention were measured for Senegalese sole (Solea senegalensis Kaup), in order to provide insight into how protein utilization affects growth performance. Three feeding regimes were tested: ST – standard live feed; ArtRL – live feed and 20%Artemia replacement with inert diet (dry matter basis) from mouth opening; ArtRH – live feed and 58%Artemia replacement with inert diet from mouth opening. Artemia intake and protein metabolism were determined at 6, 15 and 21 days after hatching using 14C‐labelled Artemia protein and subsequent incubation in metabolic chambers. At the end of the experiment, sole fed exclusively with live feed were significantly larger than sole from Artemia replacement treatments. Protein digestibility decreased during sole ontogeny, and more sharply in ArtRH sole. Concomitantly retention efficiency increased during ontogeny but with a slight delay in ArtRH sole. Senegalese sole larvae growth and protein utilization is depressed when co‐fed high levels of inert diet and Artemia, mostly during metamorphosis climax.  相似文献

Formulation of diets according to fish amino acid (AA) qualitative requirements may improve fish growth. Two diets with different AA profiles were tested on 16‐day‐old meagre during 20 days. The first diet had an AA profile closer to meagre larval AA requirements (BAL), whereas the second had a different AA profile (UNBAL). Results showed that larvae fed the diet with higher similarities to the AA profile of meagre carcass had a higher final weight (19.8 ± 0.5 mg versus 13.4 ± 1.8 mg) and lower ammonia excretion after being fed (0.6 ± 0.1 ml/(g DW. larvae. h) and 0.8 ± 0.1 ml/(g DW larvae. h)). A higher mean fibre area was obtained in 36 DAH larvae fed the BAL diet (224.8 ± 33.3 μm2 versus 158.8 ± 34.5 μm2) as well as higher larval weight. When fibre area distribution was analysed, a higher frequency of larger fibres was observed in 36 DAH BAL larvae, suggesting that fibre hypertrophy had a higher importance in this treatment. In conclusion, this study shows that meagre larvae fed a diet with an AA profile closer to their requirements had lower nitrogen losses and higher final weight, as result of higher muscle hypertrophy.  相似文献

The present study aimed to determine the effect of feeding time on growth and nitrogen excretion in juvenile sole. An 84‐day growth trial was conducted, in which food was supplied to three triplicate groups of juvenile Senegalese sole (3 g wet weight) at different schedules – diurnal, nocturnal and mixed. At the end of the growth trial, ammonia and urea excretion was assessed during a 24 h cycle. Improved growth (1.3% vs. 0.9% day?1, specific growth rate), higher nitrogen retention (0.35 vs. 0.27 g N kg?1 day?1), lower ammonia excretion (209 vs. 272 mg N‐NH4 kg?1 day?1) and lower total nitrogen excretion (278 vs. 352 mg N kg?1 day?1) were found in daytime‐fed fish compared with night‐fed fish. Fish in the mixed feeding regime showed intermediate values of ammonia and total nitrogen excretion, but did not differ from day‐fed fish regarding the other parameters. Results indicate that juvenile sole at a period of their life cycle appear to use more efficiently dietary protein for somatic growth under a diurnal than under a nocturnal feeding regime. This suggest that at least during a time‐window in the juvenile rearing a diurnal feeding regime might be more effective in the production of this species.  相似文献

Results from three larval Senegalese sole (Solea senegalensis) feeding trials using non-enriched Artemia and Artemia enriched with Super HUFA®, Arasco®, sunflower oil and microalgae are presented and the effects on larval survival, growth and fatty acid (FA) composition are reported. The FA profile of Senegalese sole eggs was analysed to gather information about the nutritional requirements of the early larval stages and a quite high DHA/EPA ratio (4.3) was found. However, there was no evidence of a high dietary demand for DHA or EPA, given that no relationship was found between dietary HUFA concentration and larval growth and survival. When larvae were fed non-enriched Artemia a significantly better growth and comparable survival were obtained than with Artemia enriched with Super HUFA® (containing the highest HUFA level and DHA/EPA ratio). The FA profiles of the larvae generally reflected those of their diets. DHA was an exception, as it was present in high proportions, even in larvae fed DHA-deficient prey. Total FAME concentration decreased during larval development, with SFA, MUFA and PUFA being equally consumed; HUFA appeared to be less used, with its relative concentration being either kept constant (particularly EPA and ARA) or increased (DHA). A specific requirement for ARA in the first larval stages could not be confirmed but it was always present in considerable amounts, even in larvae fed an ARA poor diet.  相似文献

The objective of this study was to evaluate the potential of free amino acids to replace protein‐bound amino acids in diets for Solea senegalensis juveniles. For this purpose, fish with 4.26 g mean initial weight were fed during 55 days with diets containing 500 g kg?1 protein and 100 g kg?1 lipid, based on fish meal and fish oil as main protein and lipid sources (control diet), or diets where 20%, 35% and 50% of fish meal were replaced by crystalline amino acids, using L‐lysine HCl as lysine source. To access the efficiency of the crystalline lysine source, another diet was formulated with 35% fish meal replacement but using L‐lysine sulphate instead of L‐lysine HCl as lysine source. Overall, growth performance, feed utilization, nutrient retention, whole‐body and amino acid composition did not show significant differences among groups. In conclusion, up to 50% protein‐bound amino acids can be replaced by crystalline amino acids in diets for Senegalese sole juveniles without negative effects in fish performance. Additionally, it was shown that L‐lysine sulphate and L‐lysine‐HCl may be used interchangeably by Senegalese sole juveniles, with the potential economic benefits involved.  相似文献

A 74‐day trial was undertaken to evaluate the effects of temperature (16 and 22 °C) and dietary protein/lipid ratio on the performance of juvenile Senegalese sole (mean body weight: 6.4 g). Four experimental diets were formulated to contain two protein levels (550 g kg?1 and 450 g kg?1) combined with two lipid levels (80 g kg?1 and 160 g kg?1). Growth was higher at 22 °C and within each temperature in fish fed diets 55P8L and 45P16L. Feed efficiency, N retention (% NI) and energy retention (% EI) were higher at 22 and at both temperatures in fish fed diet 55P8L. Temperature affected whole‐body composition, with dry matter, protein, lipid and energy being higher and ash lower in fish kept at higher temperature. Independently of temperature, whole‐body lipid, energy and ash were higher and protein was lower in fish fed the high‐lipid diets. Visceral and hepatosomatic indices were not affected by diet composition but were higher in fish kept at 16 °C. Liver glycogen and lipid contents and activities of glutamate dehydrogenase, alanine and aspartate aminotransferases were not affected by diet or water temperature. Malic enzyme (ME) and glucose 6‐phosphate dehydrogenase activities were higher in fish fed the low‐lipid diets. ME activity was higher at lower temperature. In conclusion, increasing water temperature from 16 to 22 °C improves growth and feed efficiency of Senegalese sole juveniles; regardless of water temperature, the diet with 550 g kg?1 protein and 80 g kg?1 lipid promoted the best growth and feed efficiency.  相似文献

Fish held at high stocking densities are generally exposed to chronic stress situations that impose severe energy demands and may predispose the fish to infection. Senegalese sole Solea senegalensis (Kaup) juveniles (78.8±18.9 g body weight) were maintained at low (LSD; 4 kg m?2 at the end of the experiment), medium (MSD; 9 kg m?2) and high (HSD; 14 kg m?2) stocking densities during a period of 63 days. Although disease outbreaks were observed in fish reared at HSD, growth and food consumption did not vary among different treatments. Results from plasma cortisol and free amino acids (FAAs) showed significant differences among different rearing densities pointing to HSD as stressful rearing condition. However, higher plasma glucose and osmolality levels indicated that fish held at MSD may also be under stress. The higher usage of FAAs from the HSD group may be due to the higher demand for energy production in order to cope with stressful rearing conditions, higher rate of protein synthesis or due to synthesis of other important metabolites related to stress response. Therefore, results from the present study point out to HSD as a stressful rearing condition and suggest that crowding stress may affect amino acid requirements.  相似文献

The objective of this work was to evaluate the effect of a dietary amino acid imbalance, originating from the use of a soy protein concentrate (SPC) as the major protein source, on the growth performance and amino acid metabolism of Senegalese sole (Solea senegalensis) postlarvae. Senegalese sole (85.6±24.6 mg wet weight) were fed one of two experimental diets: one based on fish meal (FM) and another based on SPC. Diets were isonitrogenous (around 56% crude protein) and isoenergetic. Diet acceptability was very good and the growth rate was 6.9% day?1 for sole eating the FM diet and 6.0% day?1 for sole eating the SPC diet. Mass‐specific ammonia excretion and the activities of selected amino acid metabolic enzymes (ALAT, ASAT and GDH) did not present significant differences between treatments, although this may have been due to the high variability found for these parameters in the SPC treatment. This variability may suggest different capacities of individual fish to adapt to the possible methionine dietary deficiency. The utilization of amino acids as a substrate for lipogenesis does not seem to be affected by the dietary protein source, since NAPDH‐generating enzymes (G6PD and ME) had similar activities in both treatments. Amino acid metabolism in Senegalese sole postlarvae seems to be slightly affected by the dietary protein source. Nevertheless, the changes induced by the SPC diet do not seem to impair growth, at least at the high dietary protein level used in this experiment.  相似文献

Four isoenergetic (21 kJ/g dry matter, DM) and isolipidic (65 g/kg DM) diets containing different crude protein/total carbohydrate (CHO) ratios: 60/26, 56/30, 52/34 and 48/38, were tested in 22 g Senegalese sole for 104 days. Apparent digestibility coefficients (ADC) were not affected by the treatments, and all groups presented extremely low starch ADC values (22.8%–36.5%). Replacement of dietary protein by CHO did not affect daily growth index (0.9–1.0), but significantly increased voluntary feed intake of fish. Regression analyses demonstrated that digestible protein content, rather than digestible energy, was the main dietary factor influencing such feeding activity (R2 = .952). A significantly increased feed conversion ratio was observed in sole fed increasing CHO contents. The dietary protein/CHO ratio did not influence whole‐body composition. Sole fed the 48/38 diet showed the lowest efficiency in terms of N and energy utilization. PUFA were the most represented fatty acid fraction in fillet, regardless of the dietary protein/CHO ratio, mainly due to the high content of DHA. Senegalese sole increase feed intake under low dietary protein/CHO ratios to ensure an adequate N intake. Such compensatory mechanism seems to be triggered to satisfy a specific protein metabolic requirement for energy purposes as tissue accretion remained unchanged.  相似文献

Three candidate probiotics, which had shown antimicrobial activity in vitro against two fish pathogens, were used in the rearing of Senegalese sole larvae and postlarvae. These probiotics improved the survival of starved sole yolk‐sac larvae. A feeding experiment of sole larvae and postlarvae comprised three treatments: A, B and C. Cultures of a Vibrio sp. (2J18) were added to treatment A, whereas a gram‐positive (J84) and a Shewanella sp. strain (2J27), were added to treatment B, while in a control treatment C no bacteria were added. Addition of bacteria in treatment B increased survival of larvae in the first phase of the experiment [0–20 days after hatching (DAH) (P<0.05)] and decreased the numbers of colony‐forming units (CFU) in larval gut 5 DAH compared with the control treatment (P<0.05). No differences were observed in survival (25–47%) during the second phase of the experiment (20–60 DAH). Nevertheless, the total numbers of CFU in fish gut 40 DAH were significantly lower in treatment B (P<0.05). The colonization rates of the added bacteria were the highest 5 and 11 DAH, and the highest average values reached were 65%, 82% and 17% of the total CFU count for the strains 2J18, 2J27 and J84 respectively.  相似文献

Probiotic supplementation in fish aquaculture has significantly increased in the last decade due to its beneficial effect on fish performance. Probiotic use at early stages of fish development may contribute to better face metamorphosis and weaning stress. In the present work, we studied the influence of Shewanella putrefaciens Pdp11 supplementation on growth, body composition and gut microbiota in Senegalese sole (Solea senegalensis) during larval and weaning development. S. putrefaciens Pdp11 was incorporated using Artemia as live vector (2.5 × 107 cfu mL?1) and supplied to sole specimens in a co-feeding regime (10–86 DAH) by triplicate. Probiotic addition promoted early metamorphosis and a significantly higher growth in length at 24 DAH larvae. S. putrefaciens Pdp11 also modulated gut microbiota and significantly increased protein content and DHA/EPA ratios in sole fry (90 DAH). This nutritional enhancement is considered especially important after weaning, where significantly higher growth in length and weight was observed in probiotic fish. Moreover, a less heterogeneous fish size in length was detected since metamorphosis till the end of weaning, being of interest for sole aquaculture production. After weaning, fish showed significantly higher growth (length and weight) and less variable lengths in fish when supplemented with probiotics. Both the enhancement of nutritional condition and the decrease in size variability associated with probiotic addition are highly interesting for sole aquaculture production.  相似文献

The present study intended to evaluate the effects of early introduction of inert diet in lipid digestibility and metabolism of sole, while larval feed intake, growth and survival were also monitored. Solea senegalensis larvae were reared on a standard live feed regime (ST) and co-feeding regime with inert diet (Art R). Trials using sole larvae fed with Artemia enriched with two different lipid emulsions, containing glycerol tri [1-14C] oleate (TAG) and L-3-phosphatidylcholine-1,2-di-[1-14C] oleoyl (PL), were performed at 9 and 17 days after hatching (DAH) to study lipid utilization. Co-feeding did not affect sole survival rates (ST 59.1 ± 15.9%; Art R 69.56 ± 9.3%), but was reflected in significantly smaller final weight at 16 DAH (ST 0.71 ± 0.20; Art R 0.48 ± 0.14 mg). Higher feed intake was observed in sole larvae fed on Artemia enriched with labeled PL at 9 DAH but not at 17 DAH. At 17 DAH, the smaller larvae (Art R treatment) ingested proportionally more Artemia in weight percentage, independently of enrichment. At 9 DAH lipid digestibility was equal among treatments and higher than 90%, while at 17 DAH it was higher in ST treatment (around 73%) compared to the Art R group (around 66%). Lipid retention efficiency at 9 DAH was higher in the Art R treatment, reaching values of 50%, while these values almost duplicated at 17 DAH, ranging up to 80% in both treatments without significant differences. These results show that co-feeding of live feed and inert diet from first-feeding in Senegalese sole has a toll in terms of growth and lipid digestibility but does not seem to compromise lipid metabolic utilization.  相似文献

Dietary selenium (Se) influences the glutathione peroxidase (GPx) activity and the thyroid hormone system in vertebrates, but these relations are poorly described in fish. The present study aimed to evaluate the effect of Se supplementation on Senegalese sole larvae, focusing on GPx activity and thyroid status. Two groups of larvae were fed on either control (C) or Se‐enriched live feed, from 2 days after hatch (DAH) until 34 DAH. Senegalese sole larvae fed Se‐enriched live feed significantly increased their whole‐body Se, compared to larvae from C group. The activity of GPx was higher in larvae from Se group, suggesting that the levels of this mineral were suboptimal in the control diet. TH levels were also higher in larvae fed Se‐enriched live feed, suggesting that Se may have a positive influence on the TH production in early life stages of marine fish larvae.  相似文献

The present study evaluated effects of the degree of hydrolysis and peptide profile of sardine (Sardinella sp.) protein hydrolysates (SPH) from muscle tissue wastes on productive performance, body composition, hematological parameters, biochemical parameters of metabolism, antioxidant response, and intestinal microbiology tilapia (Oreochromis niloticus). Diets containing three SPH with different degrees of hydrolysis (low: 5.5%; intermediate: 27.9%, and high: 62.5%) were compared to a control diet with fish meal. The experimental design was completely randomized with four treatments and five repetitions. One hundred eighty tilapia juveniles (6.16?±?2.0 g) were distributed in 20 polyethylene tanks of 70 l of useful volume and were evaluated during 56 days. No direct relationship was observed between the degree of hydrolysis of SPH and fish productive performance. Feed consumption was reduced in all diets with SPH (means 53.03 g) compared to the control group (65.91 g), which impaired fish growth and metabolic reserves. Feed utilization was affected by the SPH peptide profile. SPH with a more diverse peptide profile (mean 1.40) had a feed efficiency similar to the control diet (1.22). Changes in biochemical and metabolic parameters in tissues, oxidative response, and microbial populations were observed but may be associated with the low consumption of diets containing SPH. Results suggest that it is necessary to properly characterize hydrolysates and understand their composition before applying fish feed.

Tank bottom surface is an important question to take into account in flatfish aquaculture considering the intimate contact that the fish have with it. The most usual materials in aquaculture tank surfaces are inert materials such as epoxy resins or plastics with null porosity, and cement that presents a high alkalinity, roughness and porosity. To improve the cement characteristics, silica fume admixture is usually added in construction industry. The aim of this study was to evaluate the effect of three types of tank bottom surfaces on growth and welfare over a Solea senegalensis population. Moreover, the degree of fin erosion, the appearance of skin lesions and malpigmentation patterns were also monitored. The bottom surfaces tested were plain cement (CE), the same plain cement blended with silica fume (10%) (SF) and epoxy coated surface (EP). Specific growth rate for each bottom surface were compared. The Image Processing Activity Index (IPAI) was used to evaluate the Senegalese sole welfare. Higher SGR and lower IPAI values were obtained in the surface of cement with silica fume, suggesting a higher welfare. The use of plain cement affected negatively the performance of fish, probably due to the high pH and alkalinity of the water in direct contact with fish skin. The null porosity of EP seems to be related to the main cause of higher activity level of fish kept on it outcoming in lower growth. Silica fume that is frequently use to improve the mechanical characteristics of plain cement, also provides benefits in terms of fish welfare.  相似文献

Morphological changes in Senegalese sole adults (Solea senegalensis Kaup, 1858) reared at two stocking density conditions (low stocking density, 60% of bottom occupation and high stocking density, 180% of bottom occupation) were investigated using geometric morphometrics. Canonical variate analysis on weight matrix scores, including the uniform component (W′), at the end of the experiment revealed differences in body shape between experimental groups. The low density group presented a similar change pattern as that of the high density group but was less intense. Differences were discussed in terms of the effect of a high stocking density on the shape of soles which were very close to commercialization. Results did not indicate a significant size-related shape, likely due to fish age. Our results provide the first promising look at environment-related shape changes in reared sole.  相似文献

To assess how initial size dispersion affects Senegalese sole (Solea senegalensis, Kaup) growth, 128 fish were stocked either under homogeneous (HOM) or heterogeneous (HET) initial size dispersion (standard length, CV 11.8% and 29.7% respectively), and also under low (LD) and high (HD) stocking density (60% and 180% of bottom occupation). After 105 days, growth parameters, evaluated at an individual level using linear mixed‐effects models, were not significantly affected by initial size dispersion, LD fish growing significantly faster than HD fish. Variance of individual growth rate distributions of weight and standard length were lowest for LD‐HOM fish, indicating the most regular growth, and highest for LD‐HET. The diminution of the coefficient of variation along time in HET groups (between 13% and 15.4% for weight, and between 4.3% and 5.8% for standard length), and its slight decrease in the HOM groups (between 1% and 3% for both parameters), is consistent with the absence of strong competition between individuals.  相似文献

Fish tissues, particularly rich in n‐3 PUFA, are prone to lipid peroxidation that can damage cellular membranes, cause severe lesions and subsequently incidences of disease and mortality. However, fish possess antioxidant defences, such as vitamin E (VE) and antioxidant enzymes, to protect them against oxidative damage. This study investigated the effects of an increasing gradient of oxidized dietary lipid on the survival, growth performance, skeletogenesis and antioxidant defensive processes occurring in Senegalese sole (Solea senegalensis) larvae. Four groups of fish were fed live prey enriched with experimental emulsions containing an increasing gradient of oxidized oil: non‐oxidized, NO+VE, 34.5 nmol MDA g?1 w.w.; mildly oxidized, MO+VE, 43.1 nmol MDA g?1 w.w.; highly oxidized, HO+VE, 63.3 nmol MDA g?1 w.w. and highly oxidized without VE, HO‐VE, 78.8 nmol MDA g?1 w.w. The oxidation levels increased in enriched rotifers following the oxidation gradient of the emulsions, but were not affected in enriched Artemia metanauplii. The oxidation status of Senegalese sole larvae increased during development, but this was not related to the dietary treatments. The increasing dietary oxidation levels did not affect the fatty acid profile, survival, growth performance and metamorphosis processes of sole larvae. Senegalese sole seem to activate antioxidant defence mechanisms in response to the increasing amounts of dietary peroxidized lipids, in a manner efficiently enough to prevent detection of any alterations of these physiological processes. Antioxidant systems and detoxification mechanisms appeared to occur through the consumption of dietary α‐tocopherol, the activation of the antioxidant enzymes (catalase, superoxide dismutase, glutathione S‐transferase, glutathione reductase) and the retention of oxidized fat in the intestinal enterocytes for detoxification prior to their utilization. However, fish fed the highest oxidized diet presented a reduction in bone mineralization, but lower incidence of deformities in the vertebral and caudal regions than fish fed the other diets. This study exemplifies the importance of rearing Senegalese sole larvae on non‐oxidized diets during the early larval development to avoid detrimental consequences in older fish, most notably in the process of skeletogenesis.  相似文献
