An enzyme-linked immunosorbent assay (ELISA) using bulk tank milk samples was evaluated as a screening test for bovine tuberculosis (TB), a contagious chronic disease of cattle. An ELISA with MPB70, a major antigen of Mycobacterium bovis was performed using paired sets of milk and sera samples from 33 tuberculin-positive and 43 tuberculin-negative cattle. Anti-MPB70 antibodies were detected in milk samples and there was a significant correlation between seroreactivities of milk and sera samples (R2 = 0.83). Using the tuberculin skin test as the reference test, the sensitivities of ELISA using milk and sera samples were 87.8% and 81.8%, respectively, and the specificities were 97.7% and 100%, respectively.In the screening test using bulk tank milk samples from 931 dairy herds in Whasung, Gyeonggi-do, Korea, the positive rate for anti-MPB70 antibody was 4.5% (42/931) and the tuberculin-positive rate was 2.8% (26/931). Individual milk samples (n = 253) were collected from randomly selected 8 problematic and 3 negative herds (positive and negative in the screening test by MPB70 ELISA using bulk tank milk samples, respectively) and tested by MPB70 milk ELISA. In the problematic herds, positive rates were 10.5% (20/190) for anti-MPB70 antibodies in milk ELISA and 2.1% (4/190) in the tuberculin skin test. More than one dairy cows were positive by milk ELISA among the problematic herds, and all tuberculin-positive dairy cows were positive in the milk ELISA. Further, no positive cows were detected in negative herds both by milk ELISA and tuberculin skin test. These results suggest that an ELISA, using bulk tank milk samples, might be a potential efficient screening test for bovine TB of dairy cows.  相似文献

A milk and a serum ELISA for detection of antibodies against Mycobacterium avium ssp. paratuberculosis (MAP) were evaluated against the complement-fixation test (CFT) and culture of faecal samples from 580 cows collected between August 1996 and December 1996. Milk and serum were obtained concurrently from six dairy herds infected with MAP and from two dairy herds without history of infection with MAP.

A cut-off value of 7 OD% was used in the ELISAs. At this cut-off value, all six culture-positive herds were positive in the serum ELISA but one was negative in the milk ELISA. All six culture-positive herds were positive in the CFT. In the two culture-negative herds, the serum and the milk ELISA deemed all serum samples negative at this cut-off value, whereas four serum samples from one of these herds were positive in the CFT. The highest cut-off value enabling the milk ELISA to record all six culture-positive herds as positive was 4 OD%. The highest cut-off value enabling the serum ELISA to record all six culture-positive herds as positive was 17 OD%. Individual-sample relative sensitivities of the ELISAs ranged from 49 to 64% and relative specificities were 80–96% at the cut-off values of 4, 7 and 17 OD%.  相似文献

A study was conducted to estimate the record keeping, genetic selection, educational, and farm management effects on average milk yield per cow (AYC), milk fat percentage, bacterial score, and bulk tank somatic cell count (BTSCC) of dairy farms in the central region of Thailand. Farms were located in the provinces of Saraburi and Nakhon Ratchisima and were members of the Muaklek dairy cooperative. Records from individual animals were unavailable. Thus, farm records of milk yield, milk fat percentage, bacterial score, and BTCCC were collected from July 1, 2003 through June 30, 2006. Additional record keeping, genetic selection, education, and farm management information was collected through a questionnaire in May of 2006. Data from the Muaklek dairy cooperative and the questionnaire were then merged by a farm identification number. A single trait mixed model was used to analyze AYC, milk fat percentage, and BTSCC, while a log linear model was used to analyze bacterial score. Results showed that farms that kept records on individual animals had higher (P < 0.05) milk fat percentages and lower bacterial scores than farms that did not. Farms that used genetic information (EBV) and phenotypes when selecting sires were higher (P < 0.05) for milk fat percentage than farms that used only phenotypes and personal opinion. Farms milking cows with a single unit milking machine and by hand, had higher (P < 0.05) bacterial scores and BTSCC than farms using only a single or multi unit machine. Overall farms that kept individual animal records, used EBV when selecting sires, used a single method for collecting milk, and used family labor achieved higher performance from their herds than farms that did not.  相似文献

We quantified the effect of Neospora caninum (NC)-serostatus on culling and (re)production in 83 herds randomly selected from the Dutch dairy herd population (random group) and in 17 herds that had experienced an abortion epidemic associated with NC infection (epidemic-abortion group). In the random group, a single whole-herd blood sampling was done during the spring of 2003, while in the epidemic-abortion group whole-herd blood sampling was done repeatedly at least once a year starting after the abortion epidemic during the period 1997–2000 until the summer of 2004. Serological test-results for NC were given as ‘negative’ (N), ‘low-positive’ (LP) and ‘high-positive’ (HP). For analysing the time to culling, calving interval and age of first calving, survival analysis was used. For categorical reproduction parameters either a logistic-regression model (abortion, non-return after 1st insemination) or a Poisson-regression model (number of inseminations per pregnancy) was used. For milk production a linear-mixed model was used. All models were controlled, if applicable, for confounding variables like parity, production, season, year and abortion and adjusted for within-herd clustering.

In random herds, HP serostatus increased the hazard for culling 1.73-fold (95% CI: 1.37–2.19) compared to N and LP serostatus. Compared to N serostatus, LP and HP serostatus in epidemic-abortion herds increased the odds for abortion 1.88-fold (95% CI: 1.41–2.52) and 1.72-fold (95% CI: 1.38–2.14), respectively. No other reproduction parameters were associated with NC-serostatus in the random or epidemic-abortion herds. We found no effect of serostatus on milk production in the random group. In contrast, milk production of LP and HP serostatus in the epidemic-abortion group was respectively, 0.72 kg milk/day (95% CI: 0.15–1.03) and 0.59 kg milk/day (95% CI: 0.13–1.30), less during the first 100 days of lactation in the first year after the abortion epidemic compared with N serostatus.  相似文献

本试验采用随机分组试验设计,选择规模化养殖模式、园区式奶农合作组织养殖模式下的2个奶牛场,每个奶牛场选择80头健康的泌乳奶牛,按照产奶胎次、体重、泌乳阶段和产奶量相近原则随机分为2组,每组40头,就试验组饲粮中添加25g/(头·d)过瘤胃赖氨酸(RPLys)对乳牛产乳量及乳成分的影响进行研究。本试验结果发现,经过饲喂之后,试验组与对照组相比,产奶量有显著提高(P〈0.05)。两种模式下乳蛋白和乳脂肪含量均有提高趋势,但是试验组与对照组没有显著差异(P〉0.05)。乳糖、非脂乳固体两个指标略有提高,但差异不显著(P〉0.05)。规模化和园区式的试验组相对于对照组每天每头奶牛的净增利润分别为1.80元和0.52元。  相似文献

This retrospective study of papillomatous digital dermatitis (PDD) was performed using data for cows that calved between July 1993 and June 1994 on a Mexicali, Mexico, dairy farm in order to calculate incidence rates, and to compare milk yield and reproduction data between affected and nonaffected dairy cows. A total of 190 out of 577 (33%) cows were affected during lactation and six (1%) during the dry period. Fifty-two of 77 (68%) cows that had lesions in the previous lactation and were also present on the farm in the current lactation had lesions in the current lactation. The highest risk for PDD occurred during the first month of lactation (9%). The yearly estimated cumulative incidence risk was 35% and the incidence density rate was 44.6 cases per 1000 cow-months. More animals were affected in summer and fall than in winter and spring. Purchased animals were 3.4 times more likely to be affected than animals born on the farm. Survival analyses indicated healthy cows conceived 93 days after calving (median), but affected cows conceived 113 days after calving (median) (P < 0.01). PDD-affected animals produced less milk than healthy cows, but the difference was not statistically significant in the multiple regression.  相似文献

The objective of the study was to estimate the losses associated with subclinical mastitis (SCM) in crossbred dairy cows in the Central Highlands of Ethiopia. A split udder investigation was performed with 30 cows to determine production losses associated with SCM. Each quarter of the study cows was examined using the California Mastitis Test (CMT) and quarter milk production was measured over a period of 8 days. Production losses were determined for different CMT scores by comparing production of quarters with CMT score 0 to quarters with CMT scores trace, 1, 2 and 3, respectively. Using data from a recently published study, economic losses were determined for different farm sizes and production subsystems by multiplying the prevalence of the respective CMT scores with the production losses associated with these CMT scores. Mean quarter milk production was 0.82 + 0.40 kg per milking in the split udder trial. Milk production was reduced by 1.2%, 6.3%, and 33% in quarters with CMT scores 1+, 2+, and 3+, respectively. Using data from the published study, a quarter with SCM lost an average of 17.2% of its milk production. Production losses associated with SCM were estimated at 5.6% for the Addis Ababa Milk Shed. Stratified losses were highest (9.3%) in urban dairy farms (UDF) and small-scale farms (6.3%). The estimates of the financial losses ranged from US$29.1 in dairy herds in secondary towns (DHIST) to US$66.6 in UDF. A total loss of US$38 was estimated for each cow per lactation. Reducing mastitis in UDF (highest prevalence) to the level of DHIST (lowest prevalence) could reduce the loss by US$35. As this does not include costs associated with treatment or culling of diseased cows, this figure probably underestimates the possible benefits of control measures.  相似文献

The health and housing of the stock on 26 organic dairy herds in four counties in eastern Sweden were studied for one year. The herds ranged in size from 12 to 64 cows, and their milk production from 3772 to 10,334 kg per cow per year. A large-animal practitioner visited the farms three times during the year, and a random sample of a third of the cows in each herd were examined. The calves and young stock and their housing were also studied. The calves were in good condition in all but four herds; their serum immunoglobulins varied from almost none to high levels. The young stock were in good condition and in good housing in 20 herds. No cows with clinical signs of metabolic disorders were found. Body condition scores were adequate or good except in two herds. Acetone was analysed in milk samples from individual cows three to six weeks postpartum, and only sporadic cases with high levels were found. The incidence of diseases treated by a veterinarian was lower in the organic herds than the average for the conventional herds in the local dairy association. The findings at the farm visits supported these data, and it is evident that a good standard of health and welfare can be achieved in organic dairy herds.  相似文献

The objectives of this study were to: (1) characterize Minnesota dairy herds participating in a Johne's disease control program (JDCP) based on herd size, milk production, and clinical Johne's disease (JD) history, (2) evaluate if change in farm management practices, expressed in risk assessment (RA) total score, is associated with the change between the first and most recent ELISA test herd seroprevalence or change in clinical JD culling rate, and (3) identify farm factors associated with ELISA seroprevalence. A total of 1234 RA, performed between January 2000 and February 2004, were available for analysis from 714 dairy herds. ELISA test results from herd sampling between 2000 and 2004 were obtained from the Minnesota board of animal health (MBAH) database, and were available for 474 herds. Both the first and the most recent ELISA test results for herds with more than one RA were available for 262 herds. Mean herd size and mean annual milk production per cow was higher in JDCP dairy herds (161 milking cows) than either all Minnesota dairy herds or Minnesota dairy herd improvement association (DHIA) herds. For herds with more than one RA available, the most recent RA total score was significantly lower (mean 11% less) than the first. The change in RA total score (and any RA subtotal scores) between the first and most recent RA was not associated with the change between the first and the most recent ELISA within-herd seroprevalence or the change in JD culling rate between the first and most recent RA. The most recent ELISA test results were positively associated with postweaned heifer score and JD culling rate. The RA score was not found to be an effective tool for the prediction of ELISA seroprevalence.  相似文献

The objective of this study was to investigate the impact of paratuberculosis sero-status on milk yield, fat, protein, somatic cell count and calving interval in Irish dairy herds. Serum from all animals over 12 months of age (n = 2,602) in 34 dairy herds was tested for antibodies to Mycobacterium avium subsp. paratuberculosis using an ELISA. Herds were categorised by sero-status into positive, non-negative and negative, where a positive herd contained two or more positive cows, a non-negative herd contained only one positive cow and a negative herd contained no positive cows. Data at animal, parity and herd-level were analysed by multiple regression using general linear models. Positive herds (mean herd size = 129 cows) and non-negative herds (81 cows) were larger than negative herds (72 cows) (P < 0.01). Negative herds had the highest economic breeding index (EBI), while positive herds had the highest estimated breeding value (EBV) for milk yield. There was no significant effect of paratuberculosis sero-status at animal, parity or herd-level on milk yield, milk fat or protein production, somatic cell count score (SCCS) or calving interval. Negative herds tended to have a lower SCCS than positive and nonnegative herds (P = 0.087). This study only examined the effects of paratuberculosis sero-status but did not examine the clinical effects of Johne's disease at the farm or dairy industry levels.  相似文献

Present study is the first attempt to evaluate an indigenous milk ELISA with milk culture, standardize milk PCR, estimate lacto-prevalence of Map and genotype Map DNA from milk samples in few Indian dairy herds. In all 115 cows were sampled from 669 lactating cows in six dairy herds from three districts of North India. Fifty milk samples (four herds) were screened by three tests (milk culture, m-ELISA and m-PCR). Lacto-prevalence of Map in four dairy herds was 84.0% (50.0% in fat and 62.0% in sediment). Screening of both fat and sediment increased the sensitivity of culture. Colonies appeared between 45 and 120 DPI. In indigenous m-ELISA, protoplasmic antigen derived from native Map 'Bison type' strain of goat origin was used. Screening of 115 lactating cows by m-ELISA ('herd screening test') detected 32.1% positive lactating cows (lacto-prevalence). Sensitivity of ELISA was 28.5% and 42.8% in single point cutoff and S/P ratio, respectively. Lacto-prevalence of JD was high in dairy herds (66.6-100.0% by culture and 20.0-50.0% by m-ELISA). DDD farm, Mathura had very high (95.8%) and moderate prevalence of Map and lacto-antibodies, respectively. All cows were clinically suffering from JD. Specific IS 900 PCR was standardized in decontaminated fat and sediment of milk samples. DNA isolated from decontaminated pellets was amplified and characteristic 229 bp band was confirmatory for Map. Of the 50 milk samples, 6.0% were positive in m-PCR. The test needs further standardization. Map DNA were genotyped as Map 'Bison type' by IS 1311 PCR-REA. Of the three tests, milk culture was most sensitive followed by m-ELISA and m-PCR. Map DNA isolated from milk samples of dairy cattle were first time genotyped as Map, 'Bison type' in India. High prevalence of Map in milk of dairy herds, posed major health hazard for calves and human beings.  相似文献

Environmental factors that enhance either the survivability or dispersion of Salmonella enterica serovar Typhimurium (S. Typhimurium) could result in a spatial pattern of disease risk. The objectives of this study were to: (1) describe herd status based on antibody response to Salmonella Typhimurium as estimated from bulk tank milk samples and (2) to describe the resulting geographical patterns found among Texas dairy herds. Eight hundred and fifty-two bulk milk samples were collected from georeferenced dairy farms and assayed by an indirect enzyme-linked immunosorbent assay (ELISA) using S. Typhimurium lipopolysaccharide (LPS). ELISA signal-to-noise ratios for each bulk tank milk sample were calculated and used for geostatistical analyses. Best-fit parameters for the exponential theoretical variogram included a range of 438.8 km, partial sill of 0.060 and nugget of 0.106. The partial sill is the classical geostatistical term for the variance that can be explained by the herd's location and the nugget is the spatially random component of the variance. We have identified a spatial process in bulk milk tank titers for S. Typhimurium in Texas dairy herds and present a map of the expected smoothed surface. Areas with higher expected titers should be targeted in further studies on controlling Salmonella infection with environmental modifications.  相似文献

A case-control study was undertaken during the summer of 1989 in central Alberta dairy herds to identify independent predictors of nocardial mastitis. Thirty-seven herds with nocardial mastitis were matched with control herds based on herd size, milk production, and enrolment in Alberta Dairy Herd Improvement Services. Control herds were considered free of nocardial mastitis based on negative cultures of four weekly bulk tank milk samples and one composite milk sample collected during the same period from each lactating cow in the herd. A detailed questionnaire on herd management was completed during farm visits. The use of blanket dry cow therapy was not found to be a risk factor for nocardial mastitis. Dry cow therapy with intramammary products containing neomycin and the use of multidose vials of dry cow medications were the only predisposing factors identified as being significantly associated with nocardial mastitis in central Alberta dairy herds. Use of neomycin as a dry cow therapy increased the odds of nocardial mastitis occurring in these dairy herds by 169 times.  相似文献

A study was conducted to investigate determinates of how milk pricing system, farm location, farm size, and month and year affected farm milk price (FMP), farm milk revenue (FMR) and loss in FMR of dairy farms in the Central region of Thailand. A total of 58,575 milk price and 813,636 milk yield records from 1034 farms were collected from November of 2004 to June of 2006. Farms were located in the districts of Muaklek, Pak Chong, Wang Muang, and Kaeng Khoi. A fixed linear model was used to analyze milk price of farms. Two pricing systems were defined as 1 = base price plus additions/deductions for milk fat percentage, solids-non-fat, and bacterial score, and 2 = same as 1 plus bulk tank somatic cell count (BTSCC). Farm size (small, medium, and large) was based on the number of cows milked per day of farms. Results showed that FMP were lower (P < 0.05) in pricing system 1 than pricing system 2. Most small farms had higher (P < 0.05) milk prices than medium and large farms across both pricing systems. Large farms lost more milk revenue due to deductions from bacterial score and BTSCC than small and medium farms.  相似文献

An observational study was conducted in Wisconsin to compare production and management on organic and conventional dairy farms. Thirty organic dairy herds, where antimicrobials are rarely used for calves and never used for cows, were compared with 30 neighboring conventional dairy farms on which antimicrobials were routinely used for animals of all ages. A seven-page questionnaire regarding milk production, milking practices, housing, incidence of the major dairy diseases and medical treatments was used to assess management and production during 2000-2001. Body condition scores (BCS) of lactating cows and environmental and animal sanitation scores (EASS) were also collected on each of two occasions. The organic herds had significantly fewer cattle than did the conventional herds (P=0.017). The average daily milk production per cow in organic dairy herds (20.2 kg/day) was lower than that of conventional herds (23.7 kg/day). The incidence of clinical mastitis (CM) on organic farms (28 cases per 100 cow-years at risk) was not statistically different from that of on conventional farms (32 cases per 100 cow-years at risk). No significant difference in bulk tank somatic cell count (BTSCC) was observed between organic farms (262,000 cells/ml) and conventional farms (285,000 cells/ml). The average annual cull rate was 18.0 cases per 100 cow-years for the conventional farms and 17.2 for the organic farms (P=0.426). Our paired t-test results indicated significantly higher parasite burden on organic dairy farms; however, no significant difference between the two farm types when controlling for season (March and September), grazing intensity (no grazing, little grazing, grazing with access to housing and grazing only) and herd average milk production per cow. There was little evidence of other fundamental differences between the two farm types in other management and production parameters.  相似文献
