Two field strains of the fall armyworm, Spodoptera frugiperda (JE Smith), collected from corn in north Florida showed high resistance to carbaryl (626- and 1159-fold) and moderate resistance to parathion-methyl (30- and 39-fold) as compared with a laboratory susceptible strain. A field strain of the diamondback moth, Plutella xylostella (L.) collected from cabbage in north Florida and selected for 20 generations with permethrin showed high resistance to permethrin (987-fold) as compared with a susceptible strain. However, in all instances, no cross-resistance to indoxacarb, a novel oxidiazine insecticide, was observed in these two species. Biochemical studies revealed that, in S. frugiperda, activities of detoxification enzymes (microsomal oxidase, glutathione S-transferase and general esterase) were significantly higher in the field strains than in the susceptible strain, indicating that these detoxification enzymes were not actively involved in the resistance to indoxacarb. The lack of cross-resistance between indoxacarb and permethrin in P. xylostella further supports the notion that the mode of action of these insecticides on the insect sodium channel is different.  相似文献

The behavioral response of larvae of the diamondback moth (Plutella xylostella L.) to permethrin droplets was investigated through computer simulations and videotape analysis of larval movement on treated surfaces. Simulations of pre-contact avoidance responses and post-contact hypersensitivity responses indicated that the different response types would produce different effects on larval speeds and leaving times in the presence of insecticide. Analysis of actual larval movement in treated arenas demonstrated that responses to permethrin consistent with the predictions for pre-contact avoidance did occur. Larval speeds were higher in trials with insecticide droplets than in controls, and these differences arose prior to contact with droplets. When a higher insecticide concentration was used for droplets, the response observed was consistent with a relatively longer distance avoidance response than when droplets had lower permethrin concentrations. Such a response to permethrin should significantly decrease larval exposure to insecticides and increase survival in permethrintreated fields.  相似文献

Base‐line susceptibility for six‐day‐old larvae of the diamondback moth, Plutella xylostella, against Bacillus thuringiensis var kurstaki (Biobit®) was studied by a cabbage leaf disc dip bioassay technique. Diamondback moth from 13 locations in seven different states spread over a distance of about 3000 km longitudinally was used for these studies. Forty‐eight‐hour LC50 values varied from 1.0 to 10.97 mg AI litre−1. Further investigations on the development of resistance under laboratory conditions showed an increase in LC50 from 2.76 (for unselected F1 generation) to 5.28 mg AI litre−1 (for selected F9 generation), using a selection concentration of 6.4 mg AI litre−1. This suggested a possibility of the development of resistance under field conditions if there were to be extensive and indiscriminate use of B thuringiensis. These findings are discussed in relation to integrated pest management and the mechanisms of resistance in resistance management tactics. © 2000 Society of Chemical Industry  相似文献

A field population of Plutella xylostella (L) from Pakistan was found to be highly resistant to deltamethrin (>500-fold) but had little or no resistance to spinosad, fipronil, indoxacarb, abamectin, Bacillus thuringiensis (Bt) var kurstaki, Bt var aizawai or Cry1Ac when compared with a susceptible laboratory population, Lab-UK. A sub-population was selected for six generations (laboratory G3-G8) with deltamethrin (delta-SEL), while a second sub-population was left unselected (UNSEL). Bioassays at G9 found that selection with deltamethrin gave resistance ratios of >230 compared with UNSEL (>6730 compared with Lab-UK). The delta-selected population showed no apparent cross-resistance to spinosad, fipronil or indoxacarb. Logit regression analysis of F1 reciprocal crosses between delta-SEL and Lab-UK indicated that resistance to deltamethrin in the delta-SEL population was inherited as an autosomal, incompletely dominant (D(LC) = 0.67) trait. A direct test of monogenic inheritance based on a back-cross of F1 progeny with delta-SEL suggested that resistance to deltamethrin was controlled by more than one factor.  相似文献

2008-2012年, 采用浸叶法连续5年测定了云南省2个主要蔬菜种植区通海县和弥渡县的小菜蛾田间种群对Bt制剂的抗药性,并进行了盆栽药效评价。结果表明,两地区小菜蛾对Bt制剂均表现为低抗或无抗性,抗药性无明显上升趋势。比较了在光照培养箱和自然条件下3种不同Bt制剂对通海和弥渡县小菜蛾盆栽药效,药后第5天通海和弥渡小菜蛾种群在恒温(25±1 ℃,RH65%~70%)和自然条件下(12~28 ℃,RH50%~75%)的校正死亡率分别为88.3%~95.5%和80.2%~90.6%、78.8%~89.1%和73.4%~85.7%,两者差异不显著。云南小菜蛾种群对Bt制剂均有较高的敏感性,可在十字花科蔬菜生产中有选择地使用。  相似文献

The activities of the acylurea insect growth regulators, chlorfluazuron, teflubenzuron and difubenzuron, and the neurotoxic macrocyclic lactone, abamectin were assessed against a laboratory susceptible (FS) strain and a field (Cameron Highlands, Malaysia (CH)) strain of the diamondback moth, Plutella xylostella L. using a leaf-dip bioassay at 20°C. The time taken to achieve end-point mortality was found to vary considerably (9–17 days), being fastest with abamectin against the FS strain and slowest with difubenzuron against the CH strain. The order of activity (LC50 at F6/7) against second-instar larvae of both strains was: abamectin > chlorfluazuron = teflubenzuron ? difubenzuron. Subsequent assays (F14) with the acylureas, flufenoxuron and hexaflumuron against the FS strain suggested that the former was slightly more active than chlorfluazuron or teflubenzuron, the latter slightly less active. The CH population was found to be 12.6-, 6.7-, 6.4- and 2.3-fold less sensitive to difubenzuron, teflubenzuron, chlorfluazuron and abamectin respectively than the FS strain. Selection of sub-populations of the CH strain with chlorfluazuron (CHL-SEL) and teflubenzuron (TEF-SEL) for six generations (F6-11), resulted in LC50 resistance ratios of 109- and 315-fold respectively when compared with the FS strain, equivalent to an 18- and a 46-fold increase in resistance compared with the unselected CH strain. Marked cross-resistance was also demonstrated between chlorfluazuron and teflubenzuron in both sub-populations. However, there was no evidence of cross-resistance to dijlubenzuron and abamectin and little or no cross-resistance to flufenoxuron and hexaflumuron. Resistance to chlorfluazuron and teflubenzuron appeared to be relatively unstable in the TEF-SEL compared with the CHL-SEL sub-population (over 6–9 generations). However, reselection of the TEF-SEL population with chlorfluazuron (F18–20) led to a very rapid increase in resistance to chlorfluazuron and particularly teflubenzuron. For the latter compound, resistance factors of about 1000000 were obtained (F19, 21). Such values are probably only semi-quantitative, as above a certain level of resistance feeding bioassays with acylureas (compounds which are active to a significant extent by ingestion) are likely to become rate-limiting.  相似文献

A 110-fold acetamiprid-resistant Plutella xylostella (L) strain was established after four selection experiments (in five generations) on a 9.5-fold resistant colony in the laboratory. Inheritance of resistance to the Bacillus thuringiensis Berl. CryIA(b) crystal protein was studied in Plutella xylostella L. (diamondback moth). A field population 50-fold more resistant to CryIA(b) than a control susceptible strain was used. Dose-mortality curves of the resistant population, the susceptible strain and the F1 from the two reciprocal crosses were compared. Resistance transmission to the F1 was dependent on the sex of the resistant progenitor. Sex ratio of the survivors to high doses of CryIA(b) in the F1 of the two reciprocal crosses did not corroborate the preliminary hypothesis of resistance being due to a recessive sex-linked allele. Since, in a previous work, the loss of CryIA(b) binding capacity of resistant insects had been demonstrated, binding to midgut tissue sections from F1 individuals was also analysed. The presence of binding in all of the F1 preparations showed that, at least, a recessive autosomal allele was responsible for the loss of binding capacity in the resistant population.  相似文献

A field population of Plutella xylostella from Malaysia (SERD4) was divided into five sub-populations and four were selected (G2-G5) with the Bacillus thuringiensis insecticidal crystal (Cry) toxins Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da. Bioassay at G6 gave resistance ratios of 88, 5, 2 and 3 for Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da respectively compared with the unselected sub-population (UNSEL-SERD4). The Cry1Ac-selected population showed little cross-resistance to Cry1Ab, Cry1Ca and Cry1Da, (3-, 2- and 3-fold compared with UNSEL-SERD4), whereas the Cry1Ab-SEL sub-population showed marked cross-resistance to Cry1Ac (40-fold), much greater than Cry1Ab itself. In contrast, the Cry1Ca- and Cry1Da-SEL sub-population showed little if any cross-resistance to Cry1Ac and Cry1Ab. The mode of inheritance of resistance to Cry1Ac was examined in Cry1Ac-selected SERD4 by standard reciprocal crosses and back-crosses using a laboratory insecticide-susceptible population (ROTH). Logit regression analysis of F1 reciprocal crosses indicated that resistance to Cry1Ac was inherited as an incompletely dominant trait. At the highest dose of Cry1Ac tested, resistance was recessive, while at the lowest dose it was almost completely dominant. The F2 progeny from a back-cross of F1 progeny with ROTH were tested with a concentration of Cry1Ac that would kill 100% of ROTH. The mortality ranged between 50 and 95% in seven families of back-cross progeny, which indicated that more than one allele on separate loci were responsible for resistance to Cry1Ac.  相似文献

BACKGROUND: The tobacco cutworm, Spodoptera litura (F.), is one of the most destructive polyphagous pests worldwide. The susceptibility of S. litura larvae reared on tobacco, Chinese cabbage, cowpea and sweet potato to phoxim, chlorfenapyr, methomyl, fenvalerate and emamectin benzoate under laboratory conditions was determined. RESULTS: Spodoptera litura larvae reared on tobacco were most tolerant to all insecticides, whereas those that fed on sweet potato were most susceptible. When larvae were reared on each host plant for three generations, the susceptibilities to phoxim of larvae that fed on Chinese cabbage and cowpea were similar, whereas the susceptibility of larvae that fed on sweet potato decreased by the third generation, and on tobacco the susceptibility decreased in each consecutive generation. When nicotine was added to their diet for three consecutive generations, the tolerance of larvae to phoxim increased twofold, and to emamectin benzoate 3.1‐fold, but the tolerance of larvae to fenvalerate and chlorfenapyr did not change. The acetylcholinesterase activities of the larvae that fed on sweet potato and cowpea were greater than the activities of those that fed on Chinese cabbage and tobacco. In contrast, the carboxylesterase activities of the larvae that fed on tobacco and Chinese cabbage were greater than the activities of those that fed on sweet potato and cowpea. The glutathione S‐transferase activities of larvae were highest when they fed on tobacco, followed by Chinese cabbage and cowpea, and the lowest activities were observed when larvae fed on sweet potato. CONCLUSION: Feeding on tobacco or with nicotine added to the diet, the larvae became more tolerant to insecticides, especially to phoxim and emamectin benzoate. BACKGROUND: The pyrethroid resistance of the diamondback moth Plutella xylostella (L.) is conferred by increased gene expression of cytochrome P450 to detoxify the insecticide and/or through gene mutation of the sodium channel, which makes the individual insensitive to pyrethroids. However, no information is available about the correlation between the increased metabolic detoxification and the target insensitivity in pyrethroid resistance. RESULTS: Frequencies of pyrethroid‐resistant alleles (L1014F, T929I and M918I) and two resistance‐related mutations (A1101T and P1879S) at the sodium channel and expression levels of the cytochrome P450 gene CYP6BG1 were examined individually using laboratory and field strains of P. xylostella. Real‐time quantitative PCR analysis using the laboratory strains revealed that levels of larval expression of the resistant strain, homozygous for the pyrethroid‐resistant alleles other than the M918I, are significantly higher than those of the susceptible strain. In the field strains, the expression levels in insects having the same resistant alleles as those of the resistant strains varied greatly among individuals. The expression levels were not significantly higher than those in the heterozygotes. CONCLUSION: Significant correlation between the target insensitivity and the increased metabolic detoxification in pyrethroid resistance of P. xylostella was observed in the laboratory but not in the field. Genetic inheritance of resistance to Bacillus thuringiensis var kurstaki (BTK) was examined in a diamondback moth (Plutella xylostella) population collected from the Melaka region of Malaysia. A BTK‐selected sub‐population (BTK‐SEL) which was more than 100‐fold resistant to BTK compared with a susceptible (ROTH) population of P xylostella was used with standard reciprocal crosses and back‐crosses between ROTH and BTK‐SEL. Logit regression analysis of F 1 reciprocal crosses indicated that BTK resistance was inherited as an incompletely recessive autosomal trait and controlled by a single locus. In contrast, other studies have shown that resistance to Cry1Ac is inherited as an incompletely dominant autosomal trait in a Cry1Ac‐selected sub‐population of the same Melaka population. Topical application of the synergists piperonyl butoxide (PB) and S,S,S-tributyl phosphorotrithioate (DEF) to second-instar larvae of a standard laboratory strain (FS) and an unselected Malaysian field strain (CH) of the diamondback moth Plutella xylostella had no significant effect on the toxicity of the acylurea insecticides, chlorfluazuron and teflubenzuron, in a subsequent leafdip bioassay. In contrast, pre-treatment with PB or DEF in acylurea-selected subpopulations of the CH strain with varying levels of cross-resistance to chlorfluazuron and teflubenzuron significantly increased (up to 34-fold and 28-fold, respectively) the toxicity of both compounds, suggesting that microsomal monooxygenases and esterases may be involved in resistance. The addition of a mineral oil, ‘Sunspray 6E’, to topically-applied chlorfluazuron consistently reduced its LD50 value, and the effect of the oil appeared to be greatest on the most resistant population of P. xylostella. However, the effects of the oil were not significant (P > 0·05) and further studies are necessary to determine whether a penetration factor is present in the CH strain.  相似文献

The efficacy of Bacillus thuringiensis (Bt) subsp. kurstaki HD-1 (‘Dipel’™; Btk; CryIA & CryII) and Bt. subsp. aizawai (‘Florbac’™; Bta; CryIA & CryIC) was assessed against larvae from various field populations of Plutella xylostella (F2 generation) collected in the Cameron Highlands, Malaysia in April 1994 and a lowland population (SERD 2; F10 generation) collected in December 1993. Evidence of resistance to Btk and to a lesser extent Bta is reported in these populations (LC50 Toxicity Ratios [TR]=3–14 and 2–8 respectively), most notably in SERD 2. The first recorded evidence of resistance to abamectin (TR=17–195-fold) in field populations of P. xylostella is also reported. In an unselected sub-population of SERD 2, the TR values for Btk, Bta and abamectin declined 2- to 3-fold (P<0·01) over six generations in the laboratory (F10–F16) while in sub-populations of SERD 2 selected with these products (F11–F15) there was a significant (P<0·01) increase in the TR (15-, 3- and 2·5-fold respectively) when compared with the F10 generation. This suggests the presence of marked resistance to Btk and some resistance to Bta and abamectin. There is also evidence of slight cross-resistance to Btk in the Bta-selected sub-population but no evidence for the reverse selection of resistance or for cross-resistance between Btk and abamectin. Concurrent selection studies (F11–F15) with another sub-population of SERD 2 demonstrated resistance to the acylurea insect growth regulator, teflubenzuron (‘Nomolt’™) (29-fold increase in TR). Based on the selection experiments with SERD 2, estimates of realised heritability (h2) of resistance gave very high values for teflubenzuron and Btk (c.0·7) and moderate values for abamectin and Bta (c.0·3). The results are discussed in relation to integrated pest management (IPM) and insecticide resistance management (IRM) strategies for P. xylostella.  相似文献

使用苏云金芽胞杆菌库斯塔克亚种(Btk)可湿性粉剂对小菜蛾进行继代汰选获得F80代和F100代抗性品系。通过分别测定Cry1Ab、Cry1Ac、Cry1Ah和Cry1Ca四种杀虫晶体蛋白对小菜蛾Btk抗性品系F80代和F100代的室内毒力,明确了小菜蛾Btk抗性品系对四种Bt杀虫晶体蛋白抗性发展规律。研究结果表明,汰选至F80代时,Cry1Ca对小菜蛾的毒力最高,LC50约12.1mg/L,其次为Cry1Ac,LC50约47.7mg/L;而汰选至F100代时,仍以Cry1Ca对小菜蛾的毒力最高,LC50约21.4 mg/L,其次为Cry1Ab,LC50约72.2 mg/L。与相对敏感品系相比,小菜蛾抗性品系对Cry1Ac的抗性发展较快(抗性倍数高达67.3~106.8倍),对Cry1Ab的次之(抗性倍数高达60.0~66.1倍),而对Cry1Ca和Cry1Ah的抗性发展较慢(抗性倍数分别为3.4~6.0倍和1.6~2.5倍)。以上结果说明,在主效杀虫基因为Cry1Ac的Btk药剂选择压力下,小菜蛾对四种Bt杀虫晶体蛋白的抗性发展速度差异较大,且Cry1Ac和Cry1Ab存在交互抗性风险...  相似文献

BACKGROUND: Fufenozide is a novel non‐steroidal ecdysone agonist with good efficacy against diamondback moth (DBM), Plutella xylostella (Lepidoptera: Plutellidae). At present, it is widely applied for the control of a range of lepidopterous pests in China. This study compared the activities of fufenozide and 12 other insecticides against unselected and fufenozide‐selected strains of DBM to examine potential patterns of cross‐resistance. The relative fitness of the fufenozide‐selected strain was assessed to provide information pertinent to insecticide resistance management. RESULTS: Compared with the susceptible strain (JSS), the fufenozide‐resistant strain (JSR) showed high cross‐resistance to dibenzoylhydrazines and benzoylphenylureas, low cross‐resistance to abamectin and no cross‐resistance to organophosphates, carbamates and pyrethroids. JSR had a lower reproductive ability and a relative fitness of 0.5 compared with JSS. CONCLUSION: P. xylostella has the potential to develop resistance to fufenozide, albeit at the expense of fitness. Cross‐resistance between the same and other classes of insecticides is of concern, and should be a key consideration when implementing fufenozide‐based control strategies for this species.
