Several strobilurin fungicides inhibiting fungal respiration by binding to cytochrome b have been introduced recently. A mechanism of strobilurin resistance identified as active in several plant pathogenic fungi is based on the activation of alternative respiration. Thus far, respective studies have been restricted to single isolates of respective pathogens. Here, we report a study on 250 Venturia inaequalis baseline isolates to the strobilurin kresoxim-methyl having a broad sensitivity distribution characterized by a 50-fold difference in sensitivity of the most- and least-sensitive isolates. For the majority (62%) of these isolates, differences in sensitivity were not caused by the interference of alternative respiration with the full inhibitory potency of kresoxim-methyl. Rather, variable dose-responses with largely different degrees of inhibition achieved at a low dose of kresoxim-methyl were found to be responsible. For 38% of the baseline isolates, alternative respiration was already active during the stage of conidia germination. Activation of this pathway was, again, dependent on the strobilurin dose. Selection of sub-populations of isolates resisting low doses of kresoxim-methyl by multiple mechanisms and the recombination among isolates expressing such mechanisms singly can be expected to be slowed by an anti-resistance strategy based on high strobilurin doses. © 1999 Society of Chemical Industry  相似文献

Up to 2005 the sensitivity of Stemphylium vesicarium (Wallr.) Simm., the causal agent of pear brown spot, to the strobilurin fungicides kresoxim-methyl, trifloxystrobin and pyraclostrobin was still comparable with baseline values associated with good efficacy in the field. During 2006, the first resistant isolates were detected in two commercial pear orchards in the Emilia-Romagna region (Italy), one of which was affected by considerable control failure linked to strobilurin treatments as demonstrated in a field trial. In vitro sensitivity tests with 0.5 mg l−1 of kresoxim-methyl, trifloxystrobin and pyraclostrobin showed that in the population collected in the orchard with control failure the conidial germination was greater than 90% compared to an untreated control both in 2006 and in 2007, i.e. 1 year after the suspension of strobilurin applications. In the other orchard, where only a few symptomatic fruits were found and the strobilurins were still in use, the conidial germination was lower, about 50% in 2006 and 25% in 2007. The molecular analysis of mitochondrial cytochrome b gene of some monospore isolates with different levels of sensitivity confirmed the presence of the mutation causing G143A substitution in all the resistant isolates. In conclusion, both in vitro tests and molecular analysis confirmed the first occurrence of Stemphylium vesicarium resistance to all strobilurin fungicides tested.  相似文献

The apple rust mite Aculus schlechtendali (Nal.) (Acari: Eriophyidae), is a main pest in apple-growing areas in Ankara, Turkey, and chemical control applications have some limitations. Entomopathogenic fungi have a potential for biological control of mites. In this study, an entomopathogenic fungus, Paecilomyces lilacinus (Thom) Samson (Deuteromycota: Hyphomycetes), was first isolated from the mite cadavers on Japanese crab apple leaves and pathogenicity of the fungus was observed in different inoculum densities and relative humidities. The pathogen caused up to 98.22% mortality of the mite population. The effects of some fungicides on the entomopathogenic fungus were determined in in vitro studies. Carbendazim, penconazole and tebuconazole were the most effective fungicides on mycelial growth of P. lilacinus, with EC50 values under 3 μg ml−1. In spore germination tests, captan, mancozeb, propineb were the most effective fungicides, followed by tebuconazole, penconazole, nuarimol and chlorothalonil. Sulphur could not inhibit the conidia germination totally at 5,000 μg ml−1. Copper oxychloride and fosetyl-al prevented conidia formation at concentrations above 1,000 μg ml−1.  相似文献

BACKGROUND: Myclobutanil, a demethylation inhibitor (DMI) fungicide, is an important fungicide for controlling apple scab and powdery mildew. Overuse of this fungicide has led to establishment of scab isolates with reduced sensitivity to this fungicide in several countries. Experiments were conducted to determine the sensitivity of the causal agent of apple scab, Venturia inaequalis (Cooke) Winter, to myclobutanil in the UK, in order to assess whether there is a relationship between fungal insensitivity and the number of DMI applications, and establishing whether fungal sensitivity varied greatly within an orchard. RESULTS: Reduced sensitivity of V. inaequalis to myclobutanil was positively related linearly to the number of DMI applications. ED50 values ranged from 0.028 to 1.017 mg L?1 (average = 0.292) for the baseline population, whereas isolates from two other orchards had much greater ED50 values, ranging from 0.085 to 5.213 mg L?1 (average = 1.852). There was significant variation in fungal sensitivity to myclobutanil among fungal isolates from different locations within a single orchard. CONCLUSIONS: Spatial spread of insensitive isolates of V. inaequalis to myclobutanil is likely to be limited in distance. Conidia may be an important source of primary inoculum. Myclobutanil should still be effective for most field isolates, but its use should be strategically integrated with other groups of fungicides. Copyright © 2009 Society of Chemical Industry  相似文献

Seventeen field isolates of Botrytis cinerea were compared by determining their radial growth on synthetic media containing various amounts of 21 antifungal compounds. Twelve of these compounds were fungicides that are recommended for the control of Botrytis infections. There were marked differences between the isolates in their sensitivity to the compounds. Individual isolates displayed high levels of resistance to some of the fungicides, including benomyl, carbendazim, iprodione, thiabendazole, thiophanate-methyl, vinclozolin and zineb. The most potent growth inhibitors were benomyl and carbendazim (ED95 values for most isolates <0.1 μg fungicide ml?1 media), dichlofluanid, iprodione, nystatin, thiabendazole, thiophanatemethyl and vinclozolin (ED95 values for most isolates < 1.0 μg ml?1), and captan, chlorothalonil, dicloran and thiram (ED95 values for most isolates < 6.0 μg ml?1). Zineb was much less potent than the other recommended anti-Botrytis fungicides; it was no more effective than carboxin, dinocap and mancozeb (ED95 values for most isolates > 25 μg ml?1).  相似文献

No registered fungicide controls Mucor piriformis, a cause of severe postharvest storage rot in pears, but the experimental fungicide RH 886 (active ingredients: 77% 5-chloro-2-methylisothiazol-3-(2H)-one and 23% 2-methylisothiazol-3-(2H)-one) has an ED50 of 23.1 μg ml?1 in 5 min exposure for germination of sporangiospores of M. piriformis and an ED50 of 9.9 μg ml?1 for mycelial growth. Mixing RH 886 into infested, amended soil at 8 mg g?1 soil or mixing copper sulfate into soil at 1 mg g?1 soil prevented sporulation of M. piriformis. Application of RH 886 to pear fruits prior to inoculation, or immersion of fruits in solutions of RH 886 containing sporangiospores of M. piriformis significantly reduced fruit infection.  相似文献

BACKGROUND: Phoma stem canker, caused by the coexisting related fungal pathogens Leptosphaeria maculans (Des.) Ces. & de Not and L. biglobosa Shoemaker & H Brun, is a major disease of winter oilseed rape in the UK. Annually, over 90% of UK crops receive at least one foliar application of fungicide, but little is known about the sensitivity of the more damaging L. maculans and the less damaging L. biglobosa to these fungicides. The effects of flusilazole, tebuconazole and Methyl Benzimidazole Carbamate (MBC) fungicides (benomyl and carbendazim) on the germination of ascospores, conidia and germ tube growth of both species were examined. Isolates collected from different oilseed rape crops in England and Wales were assessed for their mycelial growth on fungicide‐amended medium, and ED50 values were calculated. RESULTS: Leptosphaeria maculans and L. biglobosa differed in their sensitivity to fungicides. Conidial germination of L. maculans was more sensitive to these fungicides than that of L. biglobosa. Isolates of L. maculans had smaller ED50 values for mycelial growth for all fungicides tested than isolates of L. biglobosa. CONCLUSION: These results suggest that fungicide applications might affect the structure of L. maculans/L. biglobosa populations in UK winter oilseed rape crops. The sensitivity of Venturia inaequalis to trifloxystrobin was monitored by both in vitro (spore germination) and in vivo tests (on apple seedlings). There was good correlation between the in vitro and in vivo results. Baseline sensitivity was established with 95 bulk isolates from Europe between 1995 and 1998 which showed a narrow sensitivity range-factor of up to 17 between the least and most sensitive isolates. Monitoring of populations originating from trifloxystrobin-treated and untreated orchards between 1995 and 1999 showed only sensitive isolates and no performance deficiencies were reported. Data also show that trifloxystrobin is not cross-resistant to anilinopyrimidines and triazoles, but is cross-resistant to kresoxim-methyl, another strobilurin which also inhibits the cytochrome bc1 enzyme complex at the Qo-site in the respiration chain of fungal mitochondria (QoI inhibitors). Under experimental conditions from a specific trial site in Switzerland, where trifloxystrobin has been tested since 1994, isolates containing conidia resistant to Qo inhibitors were detected in 1997 and 1999. However no obvious performance deficiencies were reported. Nevertheless these results demonstrate a risk for the development of practical resistance in V inaequalis to Qo inhibitors and therefore a need to apply resistance management principles strictly.  相似文献

采用孢子萌发法和生长速率法测定了2000~2006年间采自浙江衢州、杭州、金华、丽水的65个柑橘绿霉病病菌Penicillium digitatum菌株对嘧菌酯的敏感性。结果表明:嘧菌酯对供试菌株 孢子萌发和菌丝生长的EC50值均呈单峰分布,分别介于0.020 1 ~0.260 0 μg/mL和0.005 3 ~0.079 4 μg/mL 之间,平均值分别为0.042 6 μg/mL 和0.025 0 μg/mL。敏感性频次分析结果表明,该65个菌株孢子萌发和菌丝生长对嘧菌酯的敏感性频率分布均符合正态分布,其EC50平均值0.042 6±0.030 4 μg/mL 和0.025 0±0.012 9 μg/mL可分别作为柑橘绿霉病菌孢子萌发和菌丝生长对嘧菌酯的敏感基线。  相似文献

采用菌丝生长速率法和孢子萌发法测定了烟草赤星病菌Alternaria alternata对6种杀菌剂(氟硅唑、异菌脲、菌核净、嘧菌酯、醚菌酯及多菌灵)的敏感性。结果表明:6种杀菌剂对烟草赤星病菌菌丝生长和分生孢子萌发均表现出了不同程度的抑制活性。其中,对菌丝生长抑制活性最强的是氟硅唑,EC50平均值为(0.34±0.11)mg/L,其次为菌核净[(1.13±0.68)mg/L]和异菌脲[(2.08±0.24)mg/L];对分生孢子萌发抑制活性最强的是嘧菌酯协同100 mg/L水杨肟酸,EC50平均值为(0.28±0.09)mg/L,其次分别为单独处理的嘧菌酯[(5.79±2.76)mg/L]、醚菌酯[(11.93±3.93)mg/L]、菌核净[(19.95±15.94)mg/L]和异菌脲[(23.33±11.33)mg/L]。多菌灵对烟草赤星病菌菌丝生长和分生孢子萌发的抑制活性均最差,不宜用于烟草赤星病的防治,而异菌脲、氟硅唑、嘧菌酯和醚菌酯均有必要进一步针对烟草赤星病进行田间防效试验。  相似文献

Quinone outside inhibitors (QoIs) and succinate dehydrogenase inhibitors (SDHIs) are major groups of agricultural fungicides. However, resistance to some of these fungicides has been reported in a Japanese population of Puccinia horiana, the causal agent of chrysanthemum white rust disease. Because their mechanisms are not well understood, we investigated the existence of mutations in QoI and SDHI target protein-encoding genes. Eight out of nine isolates from cultivated chrysanthemum carried L275F and L299F amino acid substitutions in cytochrome b, the target protein of QoIs. These isolates showed 23- and 17-fold higher EC50 values for the QoI fungicides azoxystrobin and kresoxim-methyl, respectively, in basidiospore germination inhibitory tests, while they were hypersensitive to another QoI, famoxadone. All nine isolates were resistant to SDHI oxycarboxin and carried the I88F substitution in SdhC. This substitution was orthologous to the SdhC-I86F substitution found in some Brazilian isolates of the soybean rust fungus, Phakopsora pachyrhizi, showing reduced sensitivity to some SDHIs. Although the rarity of wild-type sensitive isolates, the subsequent limited number of comparisons between wild types and mutants, and a difficulty in applying reverse genetic analysis to this obligate parasite, are obstacles in making definitive conclusions, L275F and L299F in cytochrome b and SdhC-I88F are suspected to be responsible for the different patterns of sensitivity to QoI and for oxycarboxin-resistance in P. horiana, respectively.  相似文献

The baseline sensitivity ofFusarium graminearum Schwade [teleomorph =Gibberella zeae (Schweinitz) Petch] to the fungicide JS399-19 (development code no.) [2-cyano-3-amino-3-phenylacrylic acetate] and the assessment of risk to JS399-19 resistancein vitro are presented. The mean EC50 values for JS399-19 inhibiting mycelial growth of three populations of wild-typeF. graminearum isolates were 0.102±0.048, 0.113±0.035 and 0.110±0.036 μg ml−1, respectively. Through UV irradiation and selection for resistance to the fungicide, we obtained a total of 76 resistant mutants derived from five wild-type isolates ofF. graminearum with an average frequency of 1.71 × 10−7% and 3.5%, respectively. These mutants could be divided into three categories of resistant phenotypes with low (LR), moderate (MR) and high (HR) level of resistance, determined by the EC50 values of 1.5–15.0 μg ml−1, 15.1–75.0 μg ml−1 and more than 75.0 μg ml−1, respectively. There was no positive cross-resistance between JS399-19 and fungicides belonging to other chemical classes, such as benzimidazoles, ergosterol biosynthesis inhibitors and strobilurins, suggesting that JS399-19 presumably has a new biochemical mode of action. Although the resistant mutants appeared to have comparable pathogenicity to their wild-type parental isolates, they showed decreased mycelial growth on potato-sucrose-agar plates and decreased sporulation capacity in mung bean broth. Nevertheless, most of the resistant mutants possessed fitness levels comparable to their parents and had MR or HR levels of resistance. Apart from its fungicidal effect, the strobilurin kresoxim-methyl (BAS 490 F) was found to induce physiological and developmental alterations in wheat (Triticum aestivum L.) which are seen in connection with improved yield. In a series of biotests including heterotrophic maize and photoautotrophic algal cell suspensions, duckweed, isolated mustard shoots and germinating cress seeds, kresoxim-methyl showed a similar response pattern to standard auxins (e.g. indol-3-ylacetic acid, IAA; 2-(1-naphthyl)acetic acid, α-NAA). Auxin-like activity of kresoxim-methyl was also found when stem explants of tobacco were cultured on a hormone-free medium. Kresoxim-methyl stimulated shoot formation, particularly at 10-7 M . The same effect was induced by 10-8 M IAA. The determination of phytohormone-like substances in shoots of wheat plants foliar-treated with 7×10-4 M kresoxim-methyl revealed only slightly changed levels of endogenous IAA, gibberellins and abscisic acid. In contrast, the contents of dihydrozeatin riboside-type cytokinins increased to 160% of the control, while trans-zeatin riboside- and isopentenyladenosine-type cytokinins remained nearly unchanged. The most remarkable alterations were the reductions in 1-aminocyclopropane-1-carboxylic acid (ACC) levels and ethylene formation which were demonstrated in intact plants, leaf discs and the shoots of wheat subjected to drought stress. Kresoxim-methyl affected the induction of ACC synthase activity which converts S-adenosyl-methionine to ACC in ethylene biosynthesis. In shoots from foliar-treated wheat plants, 10-4 M kresoxim-methyl inhibited stress-induced increases in endogenous ACC synthase activity, ACC levels and ethylene formation by approximately 50%. Reductions in ACC synthase activity and ACC levels of 30% were also obtained at low concentrations of α-NAA (10-6 M ). In contrast, ACC synthase activity in vitro was not influenced by adding the compounds. In wheat leaf discs, the inhibiting effect of kresoxim-methyl, α-NAA and IAA on ethylene formation was accompanied by delayed leaf senescence, characterized by reduced chlorophyll loss. However, in contrast to kresoxim-methyl which showed only inhibitory activity on ethylene synthesis over a wide range of concentrations applied, the auxins stimulated ethylene production at high concentrations of about 10-4 M . The inhibition of ethylene biosynthesis by kresoxim-methyl, together with an increase in endogenous cytokinins could explain the retardation of senescence and the intensified green leaf pigmentation in wheat exposed to this strobilurin. © 1997 SCI.  相似文献

Single-lesion isolates ofCercospora beticola (n=150) were collected in 1998 from sugar beet fields in the area of Serres, N. Greece. In this area, sterol demethylation-inhibiting (DMI) fungicides have been used for almost 20 years to control sugar beet leaf spot. The sensitivity of these isolates to the DMI fungicides flutriafol and difenoconazole (EC50 values) was determined on the basis of inhibition of mycelial growth at several fungicide concentrations. The relative growth (RG) of isolates was correlated at all tested concentrations with the respective EC50 values, indicating that RG provides a reliable estimate for the sensitivity of the isolates. The highest correlation coefficients were obtained for concentrations of 1 μg ml−1 flutriafol and of 0.05 μg ml−1 difenoconazole, respectively. Consequently, they are proposed for monitoring of DMI sensitivity inC. beticola populations, as single discriminatory concentrations in a simplified test method. Based on the RG values at the discriminatory concentration of 1 μg ml−1 flutriafol,C. beticola isolates were classified as either resistant or sensitive. The efficacy of flutriafol, applied at the commercially recommended dose, in controlling Cercospora leaf spot was examined in field experiments conducted during 1999 and 2000. Disease incidence in plots artificially inoculated with resistant isolates and treated with flutriafol was significantly higher than in similar plots inoculated with sensitive strains. These results suggest that poor disease control after application of flutriafol may be based on the presence of resistant strains within the pathogen population in northern Greece. Cucurbit powdery mildew caused by Podosphaera fusca limits crop production in Spain. Since its management is strongly dependent on chemicals, the rational design of control programmes requires a good understanding of the fungicide resistance phenomenon in field populations. Fifty single-spore isolates of P. fusca were tested for sensitivity to three quinone-outside inhibiting (QoI) fungicides: azoxystrobin, kresoxim-methyl and trifloxystrobin. Minimum inhibitory concentration (MIC) values for QoI-sensitive isolates were found to range from 0.25 to 10 μg ml−1 for azoxystrobin to 5–25 μg ml−1 for kresoxim-methyl, using a leaf disc-based bioassay. High levels of cross-resistance to QoI fungicides were found. Eleven isolates showed resistance to the three QoI fungicides tested with MIC and EC50 values >500 μg ml−1 resulting in RF values as high as >715 and >1000 for trifloxystrobin and azoxystrobin, respectively. A survey of P. fusca QoI resistance was carried out in different provinces located in the south central area of Spain during the cucurbit growing seasons in 2002, 2003 and 2004. Examination of a collection of 250 isolates for QoI resistance revealed that 32% were resistant to the three fungicides tested; the provinces of Ciudad Real, Córdoba and Murcia being the locations with the highest frequencies of resistance (44–74%). By contrast, no resistance was found in Badajoz, and relatively low frequencies were observed in Almería and Valencia (10–13%). Nearly 50% of resistant isolates were collected from melon plants. Based on these data, recommendations about the use of QoI fungicides for cucurbit powdery mildew management in the sampled areas are made.  相似文献

氰烯菌酯(2-氰基-3-氨基-3-苯基丙烯酸乙酯,JS399-19) 是一种对禾谷镰孢菌具有专化活性的新型杀菌剂。研究表明,该药在离体条件下对禾谷镰孢菌Fusarium graminearum抗多菌灵菌株及野生敏感菌株的菌丝生长均有很高的抑制活性,平均EC50值分别为0.117±0.036和0.107±0.020 μg/mL 。氰烯菌酯不能抑制禾谷镰孢菌的分生孢子萌发,但能引起抗多菌灵菌株及敏感菌株的分生孢子萌发后的芽管畸形。活体条件下,氰烯菌酯灌根处理对小麦赤霉病有一定防效,在叶片间的输导性较差,不能被小麦穗颈吸收;该药在同一张叶片上仅表现为向上输导,对处理部位的下部几乎没有防效。用400 μg/mL氰烯菌酯和400 μg/mL多菌灵穗部喷雾处理前1 、2 、3 d接种禾谷镰孢菌分生孢子,氰烯菌酯对小麦赤霉病的防效分别为95%、75%和62%;处理后1 、2 、3 d 接种,防效分别为88%、78%和73%,而对照药剂多菌灵的防效较差。说明氰烯菌酯对小麦赤霉病有优异的保护和治疗作用,在禾谷镰孢菌对多菌灵已产生抗性的地区,氰烯菌酯可以作为一种很好的替代药剂用于防治小麦赤霉病。  相似文献

In Calabria (southern Italy), control of crown and root rot of capsicum caused by Phytophthora capsici has relied primarily on soil drenches of metalaxyl. However, severe outbreaks occur every year in glasshouse crops, in which the practice of using plastic mulch and furrow irrigation favours the disease. Single‐hypha isolates of P. capsici collected in Calabria in 1992/1998 were tested in vitro for their level of sensitivity to metalaxyl. Isolates of other species of Phytophthora were used as reference. Fungicide sensitivity was determined by plating mycelial plugs onto potato dextrose agar amended with metalaxyl, at final concentrations ranging from 0.1 to 1000μg mL?1 a.s. Inhibition of radial growth (%) was determined when colonies on unamended medium had covered approximately two‐thirds of the plate. The ED50 values for inhibition of mycelial growth of P. capsici isolates ranged from 1.41 to44.6μg mL?1 a.s. More than 80% of the P. capsici isolates from commercial plastic‐house crops in Calabria showed a moderate level of resistance as they were inhibited less than 60% at 5 μg mL?1 but more than 60% at 100μg mL?1  相似文献

Apple scab caused by Venturia inaequalis is a major disease in apple production. Epidemics in spring are initiated by ascospores produced on overwintering leaves whereas epidemics during summer are driven by conidia produced on apple leaves by biotrophic mycelium. Fungal colonisers of sporulating colonies of V. inaequalis were isolated and their potential to reduce the production of conidia of V. inaequalis was evaluated on apple seedlings under controlled conditions. The four most effective isolates of the 63 screened isolates were tested subsequently under Dutch orchard conditions in 2006. Repeated applications of conidial suspensions of Cladosporium cladosporioides H39 resulted in an average reduction of conidial production by V. inaequalis of approximately 40%. In 2007, applications of conidial suspensions of C. cladosporioides H39 reduced conidial production by V. inaequalis by 69% on August 6 and by 51% on August 16, but no effect was found on August 20. However, viability of available conidia of C. cladosporioides H39 was low at the end of the experiment. Epiphytic and endophytic colonisation by Cladosporium spp. of leaves treated during the experiment with C. cladosporioides H39 was significantly higher than on control leaves sampled 6 weeks after the last application. It is concluded that C. cladosporioides H39 has promising potential as a biological control agent for apple scab control. More information is needed on the effect of C. cladosporioides H39 on apple scab epidemics as well as on mass production, formulation and shelf life of conidia of the antagonist.  相似文献

Field isolates of Botrytis cinerea with moderate levels of resistance to dicarboximide fungicides (ED50 1.0–4.9 μg ml?1) and to dicloran were obtained from glasshouses where vinclozolin and iprodione failed to control grey mould. From sensitive and moderatcly-resistant cultures, laboratory isolates were selected on dicarboximide-amended medium, which were highly resistant to these fungicides (ED50 125->3000 μg ml?1). Conidia of all the resistant isolates germinated well on media amended with 100 μg ml?1 of the dicarboximides vinclozolin, iprodione, procymidone and myclozolin and with 5 μg ml?1 of metomeclan. However, the spores of the moderately resistant isolates did not germinate on 100 μg ml?1 metomeclan while the spores of the highly resistant isolates germinated well. Using media with 100 μg ml?1 of metomeclan to distinguish between the two phenotypes, no highly resistant strain was detected among 312 resistant samples from five cucumber glasshouses with a high frequency of moderately resistant strains. From air-borne inoculum of five glasshouses with 100% resistant populations, 1604 colonies were recovered on vinclozolin-amended (100 μg ml?1) medium and none on metomeclan-amended (100 μg ml?1) medium. It is concluded that strains of B. cinerea highly resistant to dicarboximides are absent from field populations.  相似文献

BACKGROUND: In 2004, field isolates of Botrytis cinerea Pers. ex Fr., resistant to strobilurin fungicides (QoIs), were first found in commercial citrus orchards in Wakayama Prefecture, Japan. Subsequently, QoI‐resistant isolates of this fungus were also detected in plastic strawberry greenhouses in Saga, Ibaraki and Chiba prefectures, Japan. Biological and molecular characterisation of resistant isolates was conducted in this study. RESULTS: QoI‐resistant isolates of B. cinerea grew well on PDA plates containing kresoxim‐methyl or azoxystrobin at 1 mg L?1, supplemented with 1 mM of n‐propyl gallate, an inhibitor of alternative oxidase, whereas the growth of sensitive isolates was strongly suppressed. Results from this in vitro test were in good agreement with those of fungus inoculation tests in vivo. In resistant isolates, the mutation at amino acid position 143 of the cytochrome b gene, known to be the cause of high QoI resistance in various fungal pathogens, was found, but only occasionally. The heteroplasmy of cytochrome b gene was confirmed, and the wild‐type sequence often present in the majority of resistant isolates, indicating that the proportion of mutated cytochrome b gene was very low. CONCLUSION: The conventional RFLP and sequence analyses of PCR‐amplified cytochrome b gene are insufficient for molecular identification of QoI resistance in B. cinerea.
