This study was conducted to investigate the pathophysiology of Haemonchus placei infection in Friesian calves. Seven calves were divided into two groups, three uninfected calves (control group) and four infected animals. The latter group were infected orally with 500 H. placei larvae kg-1 body weight. Five weeks after infection they were all housed in metabolic crates and injected with 125I-bovine albumin. 51Cr-red cells and 59Fe-transferrin, to study albumin metabolism, erythrokinetics and ferrokinetics. The results showed that there was a significant reduction in the mean haematocrit values and reduced weight gains in the infected calves compared with the controls. There was also a change in the distribution of albumin from the extravascular to the intravascular pool and a significant increase in the plasma and blood volumes of infected calves although the blood and albumin loss via the gastrointestinal tract recorded in this study was similar in both groups.  相似文献

The ability of adult Haemonchus placei intestinal homogenate to confer protection against homologous challenge infection was evaluated. Calves were immunized twice with 100 microg H. placei intestinal protein in 5% dextran-sulfate/PBS (vaccinates) or PBS alone (controls) and were challenged with approximately 3300 infective H. placei larvae. There was no significant difference between groups in the total number of nematodes recovered but significantly fewer (p < 0.001) adult females were recovered from vaccinates. The proportion of fourth-stage larvae in vaccinates was significantly greater (p < or = 0.05) than in controls. Lengths of adult male and female nematodes were significantly shorter (p < 0.001) in vaccinated calves, and the numbers of eggs present in the uteri of female nematodes from vaccinates were significantly decreased (p < 0.001). Counts of nematode eggs per gram of feces (EPG) of vaccinates were significantly less than that for controls on Days 29-49 post-challenge (p < or = 0.05). Vaccinates had significant increases in serum IgG1 and IgG2 log(10) titers (p < or = 0.05) but not in serum IgM. EPG, numbers of females, and size of males and females were negatively correlated with increased mean post-challenge IgG1 and IgG2 titers. Reduction in binding of periodate-treated gut homogenate by immune serum indicated a carbohydrate specific component in the immune response.  相似文献

The protective capacity of vaccination with Haemonchus placei whole gut homogenate against challenge with the non-blood-feeding nematode Ostertagia ostertagi was evaluated in calves. Ten helminth-free calves were randomly assigned to two groups. Group 1 received 100microg H. placei intestinal homogenate in the adjuvant 5% dextran sulfate/PBS, while Group 2 received the adjuvant alone. Injections were administered subcutaneously on Days 0 and 28. All calves were challenged with approximately 26,100 O. ostertagi larvae on Day 42. Serum antibody response and counts of nematode eggs per gram of feces (EPG) were determined throughout the study. Calves were necropsied at 5.5 weeks post-challenge for recovery of nematodes. Although significant increases were detected in both serum IgG(1) and IgG(2) of Group 1 calves (p<0.05), there was no significant difference in the total number of O. ostertagi recovered from the two groups (p>0.05). Lengths of adult nematodes were not significantly different between groups nor were the numbers of eggs present in adult females recovered from each group significantly different (p>0.05). There were also no significant differences between groups regarding fecal egg counts (p>0.05). Results suggest either: (1) the antigens targeted by the induced antibodies were not present in O. ostertagi; (2) the antigens targeted by the induced antibodies were present, but not essential to O. ostertagi survival; or (3) the antigen was present and essential, but amount of antibody ingested was insufficient to cause damage to the nematode.  相似文献

Control of Haemonchus placei, one of the most important cattle nematodes in Brazil, relies on the use of anthelmintics. However, there is a need for integrated control, which includes active immunization. The aim of this work was to assess the protection afforded to calves by immunization with adult H. placei extracts against a high-dose challenge infection, a condition frequently found in the tropics. Holstein calves aged 8-10 months were immunized four times with intestinal extracts (Group D) or with a Triton X-100-soluble fraction of adult H. placei (Group A), challenge-infected with 120,000 infective larvae and sacrificed 40 days later. Immunized animals had higher IgG titers than the controls against tested fractions after the 2nd immunization, peaking after the 4th. Sera from both immunized groups recognized bands of similar apparent mass in both antigenic preparations, some of which were similar in molecular weight to Haemonchus contortus antigens with known protective effect to sheep. Egg counts were 49% and 57% lower in Groups A and D than in controls, respectively. High levels of protection were observed in two of the four calves in Group D, as evidenced by very low worm numbers recovered at necropsy, absence of eggs in the uteri of the recovered females and reduced worm length. Group D animals also showed milder signs of anemia than the other infected animals. Results demonstrate that protection against homologous high-dose challenge can be achieved by immunizing calves with H. placei gut antigens.  相似文献

Two studies were conducted in North America to evaluate the persistent activity of doramectin injectable formulation against experimental challenge with Haemonchus placei. In both studies, calves were randomly assigned to 1 of 4 treatment groups (n = 10 per group) or a larval viability group (n = 2). Calves were treated subcutaneously in the lateral midline of the neck with saline (1 ml/50 kg) on Day 0, or with doramectin (200 mg/kg = 1 ml/50 kg) on Day 0, 7, or 14. Animals used to assess larval viability did not receive any treatment. Beginning on Day 14 and continuing through Day 28, each of the 40 treated calves were given approximately 300 infective larvae of H. placei per os. The two larval viability animals received approximately 10,000 larvae as a single dose on Day 28. Approximately two weeks later, all animals were slaughtered and the abomasum from each calf processed for nematode recovery. A 2% aliquot of abomasal contents plus wash was examined for enumeration and identification of nematodes. Geometric mean H. placei counts were calculated from the log (H. placei count +1) and used to estimate percentage reduction. Overall, doramectin was > or =96.9% efficacious in reducing infection with H. placei when challenged daily 14-28 days after treatment.  相似文献

The possible existence of breed differences in the response of sheep to primary infection with Haemonchus contortus was examined by comparing worm establishment and pathogenic effects of the parasite in Awassi and Merino sheep of haemoglobin type B infected with 500 third stage H. contortus larvae per kg body weight. The results showed that the Merino sheep had lower faecal egg counts and worm burdens and suffered less severe clinical disturbances than sheep of the Awassi breed. This suggests that genetic resistance operates primarily at the level of worm establishment.  相似文献

To examine effects on biology and morphology, Haemonchus placei infections were propagated in cattle or sheep and infective larvae were introduced into the same or opposite host. Ovine source larvae had a geometric mean (GM) prepatent period of 26.5 days regardless of host species, compared to 30.6 days for bovine source larvae regardless of host species. Similarly, ovine source H. placei had a higher GM percentage establishment versus bovine source larvae (9.6% versus 3.4%) regardless of host species. The patent daily egg count for ovine source versus bovine source H. placei was 109.1 versus 50.0 FEC regardless of host species. Mean spicule length for ovine source H. placei was 492.5 microm while bovine source measured 496.5 microm. Mean female tail length for ovine source H. placei was 586.5 and 589.5 microm for bovine source H. placei. Using synlophe morphology, all worms that were measured were identified as H. placei. Sixty-three percent of females examined for vulvar flap morphology had knob-like vulvar flaps while the remaining 37% had linguiform vulvar flaps.  相似文献

The effect of time of day, season and stratum of herbage and soil on the availability of Haemonchus contortus and Haemonchus placei third-stage larvae (L3) on pasture was assessed. Feces from infected calves and lambs were placed on pasture plots and samples of upper herbage, lower herbage, mat and soil were collected at five intervals per day throughout the daylight hours on 18 sample days over 12 months. Using recovery rate factors derived from a preliminary investigation on the efficacy of larval recovery from each stratum, the data on larval recoveries were analyzed for the effect of season, time and stratum, and their interactions. Significant (P less than 0.05) differences were found for season, stratum and the season-with-stratum interaction for both parasites. No significant differences were detected for larval counts at different times of the day. Larval recoveries of H. contortus were larger throughout the study than those of H. placei. Most H. contortus L3 were recovered in the summer and autumn, and H. placei in the spring and summer. For both parasites, the recoveries of larvae from the upper and lower herbage were larger than those from the mat and soil. The implications of these findings are discussed in terms of control strategies.  相似文献

The antibody titre of seven calves, naturally infected with Hypoderma bovis, was estimated by an enzyme-linked immunosorbent assay (ELISA) on sera collected from June to early December 1983 following infection in 1982. It was found that the antibody was lost from the circulation of these animals about 14 weeks after the disappearance of the infection. It is suggested that the presence of antibody in the sera of calves sampled in the late autumn and winter indicated that a new infection had been acquired.  相似文献

In four calves given Haemonchus contortus larvae, the serum pepsinogen concentration rose quickly to reach a mean of 3.5 iu tyrosine on day 14 after infection. The mean concentration dropped to 1.8 iu tyrosine by day 23 and remained around 2.0 iu tyrosine until an anthelmintic was given on day 57, when concentrations returned to preinfection values.  相似文献

Cellular changes in the abomasal tissue and draining abomasal lymph nodes were examined after primary infection of lambs with Haemonchus contortus for 3, 5 or 27-36 days.Infection with H. contortus larvae resulted in a rapid and selective increase in the percentage of CD4(+) T-cells in the abomasal lymph node at 3 days post-infection (PI). By 5 days PI, the lymph node weight had increased two-fold; however, the percentage of lymphocyte populations in the abomasal lymph node resembled that seen in uninfected sheep. Lymph node weights remained at increased levels in the adult nematode infected sheep and down-regulation of B-cell surface markers (sIg and MHC Class II) was apparent in this group. Significant increases in the percentage of CD4(+) T-cells co-expressing MHC Class II, but not CD25, were observed in the larval infected groups except in adult nematode infected sheep. Increased numbers of eosinophils, CD4(+), gamma delta(+) T-cells and B-cells were found in the abomasal tissue by 5 days PI, but no further increases in these cell populations were observed in the adult nematode infected group. In contrast, the level of both lamina propria and intraepithelial mast cells observed in the abomasal mucosa was highest in the sheep carrying an adult nematode burden. These findings indicate that sheep are able to generate an early immune response to infection with H. contortus larvae, characterised by the activation of CD4 T-cells and B-cells in the draining lymph nodes and recruitment of eosinophils, CD4(+) and gamma delta-TCR,WC1(+) T-cells and B-cells in larval infected tissues. However, these changes do not seem to be maintained during infection with the adult parasite where increases in mast cell numbers dominate the local response, indicating that different parasite stages may induce distinct and possibly counteractive immune responses.  相似文献

The immunologic response of colostrum-deprived calves to parainfluenza-3 (PI-3) virus given by intranasal inoculation was studied. Inoculation of calves with 3.2 x 10(6) median cell culture infective doses (CCID50) of either virulent (SF-4) virus or a modified strain of PI-3 virus, or with 2.0 x 10(8) CCID50 of SF-4 virus, stimulated development of both serum antibody and nasal secretion (NS) antibody. However, NA antibody decreased in all calves between the 16th and 42nd postinoculation days and was present only at low or moderately low concentrations on the 126th day, when the immunity of the calves was challenged. Generally, calves that were inoculated with 3.2 x 10(6) CCID50 of SF-4 virus developed slightly higher concentrations of serum and NS antibodies than did calves inoculated with modified virus. Calves that were inoculated with 2.0 x 10(8) or 3.2 x 10(6) CCID 50 of SF-4 virus developed comparable concentrations of serum antibody, but large doses of SF-4 virus were less effective than smaller doses of the same virus in stimulating the development of NS antibody. Reinoculation of 3 calves with modified PI-3 virus resulted in a demonstrable increase in serum antibody in 2 calves and an increase and subsequent decrease in NS antibody in all calves. Challenge exposure of inoculated calves to aerosols of SF-4 virus failed to cause clinical signs of disease, and the challenge virus was not isolated from the nasal passages.  相似文献

The objective of the present study was to evaluate the effect of different schemes of calves’ supplementation in a creep feeding system, on the behavior of Bos indicus calves and dams, and also the influence of the calves’ supplementation on dams’ performance. Forty-eight Nellore male calves (147 ± 7 kg body weight and 3 months of age) in the suckling phase and their dams (476 ± 9 kg and 6 years of age) were studied in a completely randomized design. The experiment was divided into two periods of 71 days. The treatments were 5- and 10-g supplement dry matter (DM)/kg BW day offered in periods 1 and 2, respectively (5S/10S); 10- and 5-g supplement DM/kg BW day offered in periods 1 and 2, respectively (10S/5S); 7.5-g supplement DM/kg BW day in both periods 1 and 2 (7.5S); and mineral mix ad libitum in both periods 1 and 2 (MM). No differences (P < 0.05) in body condition score (BCS), final body weight (FBW), and average daily gain (ADG) were found in dams’ performance. Calves from MM treatment spent more time (P < 0.05) grazing than the supplemented calves from 5S/10S and 10S/5S treatments, in the first period. No difference in suckling time was found between the treatments (P > 0.05) in the first evaluated period. Calves from 10S/5S treatment spent more time suckling and less time eating supplements (P < 0.05) than 5S/10S treatment animals, in the second evaluated period. Dams of MM treatment’s calves had more idle time and lower grazing time when compared with the mothers of calves from 5S/10S and 10S/5S treatments. It was concluded that different schedules of Nellore calves’ supplementation on pasture do not affect their mothers’ performance, and supplementation decreases the grazing time of calves in the suckling phase.  相似文献

An ultrasonography was performed in 21 healthy male calves of Nellore (n = 8), Curraleiro Pe-duro (n = 7) and Pantaneiro (n = 6) breeds, between 8 and 12 months, with the objective to display information about size, position and ultrasonographic parenchymal patterns of the liver, in addition to the topography of the gallbladder, right kidney and major vessels. The measurements used were dorsal margin distance (DMD) and ventral margin distance (VMD) of the liver, viewable extension (VE) of the liver, the caudal vena cava (CVC), width (CVCW) and depth (CVCD) and the portal vein (PV), width (PVW) and depth (PVD). The measures DMD, VMD and VE were similar, and the organ’s longest visible extension was in 11th ICS. CVC was more frequently observed in the 11th and 10th ICS, despite being visible from the 12th to the 9th ICS in Nellore and still 8th ICS in Curraleiro Pe-duro and Pantaneiro. CVCD was lower in the 10th and 9th ICS in Nellore. The PV presented higher observation frequency in the 10th ICS. The measures PVD and PVW were higher in Nellore in the 11th, 10th and 9th ICS. To our knowledge, this report is the first to document ultrasonographic findings in three healthy calves. Considering the three breeds, the results show no differences in the ultrasonographic features as regard to the measurements and visualization of the position, size and vasculature of the liver and there was no additional information about the topography of the gallbladder and the right kidney.  相似文献
