Genetic effects on cold‐stress tolerance were assessed for red drum (Sciaenops ocellatus), an economically important sciaenid fish in the southern USA. Thirty‐five families were generated via ‘natural’ spawning of multiple sets of five breeders (three dams × two sires) in individual brood tanks. Offspring from the 35 families were transferred abruptly from an acclimation temperature of ~24 °C to 5.7 °C and maintained subsequently at an average temperature of 6.5 °C. Offspring were assigned a posteriori to individual broodfish (dam and sire) based on genotypes at nuclear‐encoded microsatellites. Heritability of the survival–time probability function was estimated using a proportional hazard approach and an animal‐additive model. The estimated heritability was 0.20 (95% CI: 0.07–0.40), indicating a significant genetic component to acute cold‐stress tolerance in red drum.  相似文献

Heritability of juvenile growth rate was estimated for red drum (Sciaenops ocellatus), an economically important sciaenid fish in the southern USA. Thirty‐eight families were generated via ‘natural’ spawning of multiple sets of five breeders (three dams × two sires) in individual brood tanks. Offspring were individually tagged with Passive Integrated Transponder (PIT) tags and mixed for grow‐out in replicate ‘common‐garden’ tanks. Juvenile growth was followed from 166.4±18.6 to 254.0±27.0 mm (total length). Offspring were assigned a posteriori to individual brood fish (dam and sire) based on genotypes at nuclear‐encoded microsatellites. Heritability (h2) of a thermal growth coefficient was estimated using an animal‐additive model and a restricted maximum‐likelihood algorithm. Estimates of h2 were 0.33±0.08 and 0.31±0.08 for thermal growth coefficient based on length and weight respectively. These results indicate a significant genetic component in juvenile growth rate in red drum. Estimates of h2 for condition coefficient (K) at various measurement dates averaged 0.38, suggesting a genetic component to shape in juvenile red drum.  相似文献

Two experiments were conducted to determine the nutritional value of various dietary proteins for juvenile red drum. In the first 8-week feeding trial, diets containing similar quantities of lipid, carbohydrate, available energy and ash with 35% crude protein from either lyophilized whole-body croaker (Micropogon undulatus), striated beef muscle, red drum processing waste or commercially processed menhaden (Brevoortia tyrannus) fish meal were fed to juvenile red drum in brackish (6 ppt) water along with a control diet containing lyophilized muscle of red drum. The control diet produced significantly (P<0.05) greater weight gain (WG), feed efficiency (FE), and protein efficiency ratio (PER) values than all other diets; intermediate responses were observed for fish fed diets containing protein from red drum waste and whole-body croaker, while diets containing striated beef muscle and menhaden fish meal yielded the lowest values. Some differences in tissue indices and body composition of red drum including hepatosomatic index, whole-body ash and lipid, as well as liver lipid and glycogen were induced by the various diets. In the second 8-week feeding trial, the control diet containing red drum muscle was compared with similar diets containing protein from whole-body croaker and menhaden fish meal. Again the control diet produced the greatest WG, FE, and PER values followed by whole-body croaker and then menhaden fish meal. Effects of the dietary proteins on tissue indices and body composition were limited. The excellent protein quality and low-temperature processing of lyophilized red drum muscle resulted in superior performance of red drum relative to the other evaluated protein products, and lyophilized whole-body croaker provided better performance than commercially processed menhaden fish meal.  相似文献

Thirty new microsatellite loci were isolated from a microsatellite‐enriched genomic library of walking catfish Clarias macrocephalus. The CA motif was the most abundant, although several other motifs were also isolated. Two gynogenetic diploid families, each consisting of a female and 50 offspring, were genotyped at 56 microsatellite loci, including 30 loci developed in the present study and 26 loci reported in the previous study. Overall, 33 anonymous microsatellites derived from genomic library and 11 EST‐linked microsatellites were mapped to their centromeres. High levels of chiasma interference were apparent in the walking catfish genome as indicated by an average frequency of second‐division segregation (y) of 0.643 ± 0.248. Twenty‐six loci (59%) showed a high microsatellite–centromere recombination, with a frequency >0.67, and three loci had recombination frequencies >0.9. This study demonstrated that gene–centromere mapping provided a rapid method for the expansion of the initial linkage map of walking catfish.  相似文献

This study evaluated various by‐catch and by‐product meals of marine origin with red drum (Sciaenops ocellatus L.). Four different kinds of by‐catch or by‐product meals [shrimp by‐catch meal from shrimp trawling, Pacific white shrimp (Litopenaeus vannamei (Boone)) processing waste meal, red salmon (Oncorhynchus nerka (Walbaum)) head meal, and Pacific whiting (Merluccius productus (Ayres)) meal] were substituted for Special Select? menhaden fish meal at 33% or 67% of crude protein in diets formulated to contain 40% crude protein, 12% lipid, and 14.6 kJ digestible energy g?1. Each of these diets and three additional diets consisting of shrimp processing waste meal formulated on a digestible‐protein basis and two Pacific whiting diets containing reduced levels of ash were also evaluated in two 6‐week feeding trials with juvenile red drum (initial weight of 4–5 and 1–2 g fish?1 in trials 1 and 2). Red drum fed by‐catch meal at either level of substitution performed as well as fish fed the control diet; whereas, fish fed shrimp processing waste meal diets had significantly (P≤0.05) reduced weight gain and feed efficiency ratio values compared with the controls, even when fed on a digestible‐protein basis. The diets containing Pacific whiting at either levels of substitution and regardless of ash level supported similar performance of red drum as those fed the control diet. Fish fed the red salmon head meal diet fared poorly, probably owing to an excessive amount of lipid in the diet that became rancid. Overall, by‐catch meal associated with shrimp trawling and Pacific whiting appear to be suitable protein feedstuffs for red drum.  相似文献

Two separate feeding trials examined the effects of dietary supplementation of the prebiotics GroBiotic®‐A and inulin on growth performance and gastrointestinal tract microbiota of the red drum, Sciaenops ocellatus. In the first feeding trial, fish meal‐based diets without prebiotics or supplemented with either GroBiotic®‐A or inulin at 1% of dry weight were fed to triplicate groups of juvenile red drum (initial weight of 2.6 g) in 110‐L aquaria operated as a brackish water (7 ppt) recirculating system for 8 wk. In the second feeding trial, soybean meal/fish meal‐based diets supplemented with either GroBiotic®‐A or inulin at 1% of dry weight were fed to triplicate groups of red drum (initial weight of 15.8 g) in 110‐L aquaria operated as either a common recirculating water system or closed system with individual biofilters (independent aquaria) for 6 wk. Supplementation of the prebiotics in either feeding trial did not alter weight gain, feed efficiency ratio, or protein efficiency ratio of red drum fed the various diets. In the second feeding trial, the culture system significantly affected weight gain, feed efficiency ratio, and protein efficiency ratio although there were no effects of dietary treatments on fish performance or whole‐body protein, lipid, moisture, or ash. Denaturing gradient gel electrophoresis (DGGE) analysis of the gastrointestinal tract microbial community showed no effect of the dietary prebiotics as the microbial community appeared to be dominated by a single organism with very low diversity when compared with other livestock and fish species. DGGE of the microbial community in the biofilters of the independent aquariums showed a diverse microbial community that was not affected by the dietary prebiotics.  相似文献

Abstract  The ability of a commercial fisher to limit red drum, Sciaenops ocellatus (Linnaeus), bycatch was evaluated by comparing the catch of directed sets (net site chosen by the commercial fisher) vs randomly placed gillnets during 70 days of gillnetting ( n  = 691 sets). Catch rate did not differ between directed sets and nets set randomly. Thus, fisher experience did not lead to reduced bycatch; however, this may have been influenced by overall low red drum bycatch (0.41 fish net−1 for 12 h sets and 0.59 fish net−1 for 24 h sets). Catch rates of southern flounder were much higher at 3.97 and 3.37 fish net−1 for 12 h and 24 h sets, respectively. Fishing 10 nets per day, the catch of red drum exceeded the daily bag limit of seven red drum on 23% of the days. Of the 326 red drum retrieved from gillnets, 32.5% were dead at the time of retrieval; but only 15 of the 106 dead red drum had to be discarded due to size limits, and six were discarded dead based on bag limits. Current fisheries management regulations (bag limit and window limit of 457–686 mm) are supported by these findings given the conditions (e.g. red drum densities) at which the study was performed.  相似文献

Gillnets fished in North Carolina, USA, estuaries have high rates of bycatch relative to the target catch of southern flounder, Paralichthys lethostigma Jordan & Gilbert. This study tested whether rectangular‐mesh gillnets would maintain catch rates of southern flounder and reduce fish bycatch relative to conventional diamond‐mesh gillnets in two North Carolina estuaries. In the Neuse River estuary, catch rates of legal southern flounder were not different between the two mesh shapes, but the bycatch of red drum, Sciaenops ocellatus (Linnaeus) and other fish species was reduced with rectangular‐mesh net. In the Newport River estuary, southern flounder and red drum catches were reduced in rectangular‐mesh net, but the decrease was greater for red drum. Catches of sublegal southern flounder were reduced in the rectangular‐mesh net in both estuaries. Reduced catch rates of sublegal southern flounder and bycatch species suggest rectangular mesh may help manage stocks of estuarine fish species in areas where gillnets are used to target flatfishes.  相似文献

Genetic linkage maps were constructed with amplified fragment length polymorphism (AFLP) and microsatellite markers for the pearl oyster, Pinctada martensii (Dunker), the main bivalve used for marine pearl production in Asia. Twenty‐four AFLP and 84 microsatellite primer pairs were used for linkage analysis in a full‐sib family with two parents and 78 offspring. Of the 2357 AFLP fragments generated, 394 (16.7%) were polymorphic and segregating. Most (340 or 86.2%) of the markers segregated according to expected Mendelian ratios. Female and male linkage maps were constructed using 230 and 189 markers, including 15 and 10 microsatellites respectively. The female map consisted of 110 markers in 15 linkage groups, covering 1415.9 cM, with an average interval of 14.9 cM. The male map consisted of 98 markers in 16 linkage groups, with a total length of 1323.2 cM and an average interval of 16.1 cM. When unlinked doublets were considered, genome coverages were 78.5% for the female and 73.5% for the male map. Although preliminary, the genetic maps constructed here should be useful for future linkage and quantitative trait loci mapping efforts.  相似文献

The dietary lysine requirement of juvenile red drum Sciaenops ocellatus was reevaluated in two separate experiments. In the first experiment, lyophilized red drum muscle was combined with an L-form crystalline amino acid premix to yield diets containing approximately 35% protein on a dry-matter basis. Gradations (0.25%) of L-lysine HCl were added to the basal diet containing approximately 1.0% lysine. Each diet was fed to triplicate groups of juvenile red drum initially weighing 6–7 g for 8 weeks. Based on growth and feed efficiency data, the lysine requirement (±SE) was determined to be 1.55% (±0.079%) of dry diet or 4.430% of dietary protein. In a second experiment, the lysine requirement of red drum was investigated with diets containing 35% intact protein from zein and red drum muscle. Each of the zein-based diets containing incremental levels of lysine was fed to triplicate groups of juvenile red drum initially weighing 1–2 g for 8 weeks. A crystalline amino acid test diet serving as a control in this experiment significantly outperformed the zein-based diets, and palatability of those diets was questionable due to an obvious change in feeding behavior of the fish. Due to the inferior performance of the fish fed the zein-based diets, the authors were not successful in determining a requirement with these diets. Thus, a lysine requirement of 1.55% of dry diet (4.43% of dietary protein) as determined in the first experiment is recommended for juvenile red drum.  相似文献

Median lethal concentrations of un-ionized ammonia-nitrogen to red drum (Sciuenops ocellarus) were 0.9 ± 0.14 mg/L (mean ± SE) after 24 h and 0.8 ± 0.16 mg/L after 48 h (salinity = 4.0%, temperature = 20 C, pH = 6.8–7.1). Ammonia moved quickly from the environment into the plasma with 90% of steady state concentrations being reached after 33 minutes. When ammonia-exposed fish were moved to ammonia-free water, 90% of the ammonia which had accumulated in the plasma was no longer present after 3.3 minutes. The sensitivity of red drum to environmental ammonia appears to be similar to that of most other fishes tested. Based on the median lethal concentrations for red drum observed in this study and the acute-chronic concentration ratios for other species of fish, it is suggested that red drum fingerlings be chronically exposed to no more than 0.05 mg/L un-ionized ammonia-nitrogen.  相似文献

We assessed the effects of a fishmeal (FM)‐ and two soya‐based diets [without or with prebiotic (GroBiotic® ‐A; GBA) supplementation] on the production performance, non‐specific immunity, blood parameters and gut microbiota diversity of red drum, Sciaenops ocellatus. All diets were formulated to contain 450 g/kg crude protein (CP) and 120 g/kg lipid. A combination of soya bean meal and soya protein concentrate (SP) was used to replace 88% of the FM in the FM‐based diet (REF) originating the first soya‐based diet (SOY). The second soya‐based diet (SOY + GBA) was designed identically to SOY except for the supplementation of GBA (20 g/kg) substituting for wheat flour. After feeding the experimental diets to triplicate groups of 25 (~3.4 g/fish) and 90 (~4.1 g/fish) red drum juveniles for 9 (Trial I) and 16 weeks (Trial II), our results showed that (i) red drum was highly sensitive to the soya‐based diets in the first week of feeding; (ii) replacing 88% of FM with SP reduced the production performance of red drum without affecting intestinal micromorphology; (iii) whole‐blood leucocyte oxidative radical production was unaffected while lower haematocrit and higher plasma lysozyme activity were found in fish fed REF; (iv) plasma osmolality was unaffected while lower glucose and higher lactate levels were observed in fish fed SOY and SOY + GBA; (v) 16S rRNA gene‐based diversity of fish gut microbiota was affected both by SP and by GBA. This study expands the information on red drum responses to soya‐based diets and prebiotic supplementation.  相似文献

The continued expansion of intensive aquaculture activities requires that substitutes for fish meal‐based protein be identified. In this study, we evaluated partial (50 and 75%) replacement of fish meal in red drum, Sciaenops ocellatus, diets with soy protein concentrate (S) and barley protein concentrate (B). Growth, feed efficiency, survival, tolerance to low temperature, and tolerance to handling in warm, hypersaline water were evaluated. Only the diet with a 50% replacement of fish meal with S yielded results comparable to fish fed a nonsubstituted fish meal diet. However, the low‐temperature studies were complicated by differences in mean fish weight among the groups. The results of this study indicate S may be partially substituted for fish meal in red drum diets.  相似文献

A feeding protocol was developed for red drum larvae based on combining a commercial microparticulate diet (Kyowa Fry Feed) with live prey (rotifers) in a closed, water reuse system. In five trials, growth and survival were measured on larvae reared on a combination of live and microdiet for 1–5 d and then microdiet alone. Results in each trial were compared to control larvae reared on live rotifers Brachionus plicatilis and brine shrimp nauplii Artemia salinas. The most satisfactory combination was feeding live food and microdiet together for the first five days and then completely discontinuing live prey, eliminating the need to feed brine shrimp to the larvae. Growth rates of larvae fed progressively larger sizes of the microdiet were as good as larvae reared on live prey. Both groups metamorphosed to the juvenile stage at less than one month. Survival rates on the five day live food and microdiet combination were a remarkable 60% from egg to the juvenile stage. The successful weaning of red drum to microdiets paves the way to produce a semipurified diet to test nutrient requirements of larval fish.  相似文献

A feeding trial was conducted to evaluate the dietary copper requirement of red drum (Sciaenops ocellatus) and compare the bioavailability of copper sulphate (CuSO4) and copper‐ethanolamine. A basal diet was formulated using semi‐purified ingredients and analysed to contain 3 mg Cu/kg. Both copper sources were supplemented to the basal diet at either 5, 10 or 20 mg Cu/kg of dry diet. No significant differences were observed in growth performance of fish fed the various diets. However, red drum fed all copper‐supplemented diets retained more copper in liver and whole‐body tissues compared to fish fed the basal diet. Within both inorganic and organic copper treatments, the highest tissue copper concentrations were observed in fish fed diets supplemented with 10 mg Cu/kg. No significant differences were detected in net copper retention regardless of the nature of the copper source; hence, the bioavailability of copper sulphate and copper‐ethanolamine complex was not different in the diets for juvenile red drum. Furthermore, the minimum copper requirement for growth performance of juvenile red drum appeared to be satisfied when fish were fed the basal diet containing 3 mg Cu/kg diet, and no detrimental effects were observed in red drum fed diets supplemented with 20 mg Cu/kg.  相似文献

This study was designed to evaluate the effects of dietary supplementation with brewers yeast (Brewtech®) and nucleotides (Optimûn), either singularly or in combination, on red drum growth, body composition, stress responses and possible resistance to Amyloodinium ocellatum infection. Juvenile red drum (~1 g initially) fed practical fish‐meal‐based diets for 6 weeks had similar weight gain regardless of whether the diet was unsupplemented (basal) or supplemented with brewers yeast (2% of diet), nucleotides (Optimûn at 0.2% of diet) or both brewers yeast (2% of diet) and nucleotides (Optimûn at 0.2% of diet). Dietary effects on hepatosomatic index, intraperitoneal fat ratio or whole‐body composition were not significant, except that fish fed the singular nucleotide supplement had a significantly higher whole‐body lipid content compared with fish fed the basal diet. No significant dietary effects on cortisol response were observed after a 15 min confinement stress test perhaps because of the extreme variation among individual fish. Subsequently, in situ challenge by co‐habitation with A. ocellatum caused 100% mortality regardless of dietary treatment within a 48 h period. It is concluded that dietary supplementation of brewers yeast and nucleotides did not promote enhanced growth or improved cortisol response and resistance to uncontrolled amyloodiniosis in juvenile red drum, at least at the tested dosages.  相似文献

The effect of chitosan coating enriched with 6-gingerol on the quality and shelf life of red drum fillets during refrigerated storage was investigated in this study. Samples were applied with chitosan coating (CH), 6-gingerol (GI), and chitosan coating with 6-gingerol (CH+GI) before storing at 4°C for 20 days. The data in this study showed that treatment with CH+GI could significantly decrease pH value, K-value, thiobarbituric acid value, and total volatile basic nitrogen in red drum fillets during refrigerated storage. The counts of microorganisms, such as Enterobacteriaceae, psychrotrophic, mesophilic bacteria, and lactic acid bacteria, were also significantly lower in the CH+GI group compared with those in the CH and GI groups. Moreover, the combination of chitosan coating and 6-gingerol was more effective in relieving the deterioration of tissue texture and sensory characteristics in the red drum fillets. In addition, the results suggested that chitosan coating and 6-gingerol had a synergistic effect on the quality improvement of red drum fillets. This work demonstrates that chitosan coating enriched with 6-gingerol may be a promising natural preservative for red drum during refrigerated storage.  相似文献

This study was conducted to determine if relatively high dietary levels of aluminum and iron might affect growth, mortality, and mineral content of red drum, Sciaenops ocellatus, because algae coproduct may have rather high aluminum and iron levels due to the algae harvesting and extracting processes. Experimental diets were prepared by supplementing different levels of aluminum (1000, 2000, 4000, 8000, and 3000 mg/kg diet) from aluminum sulfate, or iron (1000, 2000, 4000, and 8000 mg/kg diet) from ferric sulfate, or a combination of aluminum and iron (4000 mg Al/kg and 4000 mg Fe/kg) to a basal diet which was formulated from practical ingredients and analyzed to contain an average of 526 mg Fe/kg and 1940 mg Al/kg diet. The feeding trial was conducted in a recirculating system consisting of 38‐L aquaria maintained at 7 ppt salinity with synthetic seawater and sodium chloride. Each diet was fed to triplicate groups of 12 juvenile red drum initially averaging 4.5 g/fish for 7 wk. At the end of the feeding period, there were no significant differences in average weight gain, feed efficiency, or hepatosomatic index of fish fed the various diets. Significant differences were found in liver iron concentration (P = 0.045) but not iron concentration of whole body or muscle tissues. In regard to aluminum content of the liver, whole body, and muscle tissues, detectable levels were not recorded. Based on these results, it does not appear that extremely high levels of aluminum or iron adversely affect juvenile red drum, and thus elevated levels of these minerals should not limit the use of algal coproducts in diet formulations.  相似文献

Two separate comparative feeding trials were conducted to evaluate if supplemental dietary guanidinoacetic acid (GDA), either singly or in combination with creatine, could enhance growth performance of red drum. The basal diet for both trials was formulated with practical ingredients but was not supplemented with creatine or GDA. For the experimental diets, creatine (0 or 20 g/kg) and GDA (0, 5 or 10 g/kg) were added to the basal diet in a 2 × 3 factorial arrangement for trial 1. Another 2 × 3 experimental design was adopted in trial 2 to further evaluate creatine (0 or 20 g/kg) and GDA (0, 10 or 20 g/kg) supplementation. Each diet was fed to juvenile red drum in either quadruplicate (trial 1) or triplicate (trial 2) aquaria twice daily for a total of 8 weeks. After each feeding trial, fish were sampled for body condition indices and whole‐body composition analysis, as well as determination of plasma, liver and muscle creatine concentrations. In trial 1, dietary creatine supplementation alone significantly (p < .05) improved weight gain and feed efficiency of red drum. An interaction between creatine and GDA was seen in whole‐body protein and lipid in the two trials. In trial 1, the highest levels of whole‐body protein and lipid were observed in fish fed 10 g GDA/kg, and in trial 2, supplementation of the diet with 10 g GDA/kg increased muscle protein and muscle lipid although there was a trending decline in fish fed the 20 g/kg GDA‐supplemented diet. In both trials of the present study, dietary GDA significantly (p < .05) enhanced liver creatine content. Dietary creatine also significantly (p < .05) enhanced plasma and muscle creatine content of red drum. Based on the results of this study, creatine was effective in enhancing weight gain and feed efficiency of red drum as seen in previous studies; however, GDA was not effective in influencing growth performance but limited synthesis of creatine from dietary GDA was apparent.  相似文献

Quadruplicate groups of juvenile red drum (initial mean weight 2.7 g; 20 fish per replicate) were fed experimental diets containing 35% crude protein and graded levels of lysine for eight weeks. Lysine concentration in the basal diet was 1.2% and was supplied by a combination of peanut meal and shrimp-head meal. The basal diet was supplemented with lysine-HCI to provide 1.6, 2.0, and 2.4% lysine. Each of these diets contained an essential amino acid (EAA) premix. Two additional diets were formulated to contain 1.2 and 2.4% lysine without the EAA premix. Weight gain and feed efficiency (FE) data indicated between 1.2 and 1.6% dietary lysine was adequate. However, serum lysine concentrations indicated 1.6–2.0% lysine was required. Fish fed 1.2 or 2.4% lysine, without the EAA premix, exhibited reduced weight gain and feed efficiency. Results indicated that red drum were able to utilize crystalline amino acids when incorporated into diets containing intact protein and, when lysine was adequate, the proteins were deficient in at least one other essential amino acid. It is recommended that a dietary lysine level of 5.7% of the dietary protein be used in formulating red drum diets.  相似文献
