Two root-colonizing Fusarium strains, Ls-F-in-4-1 and Rs-F-in-11, isolated from roots of Brassicaceae plants, induced the resistance in Lepidium sativum seedlings against Pythium ultimum. These strains caused an increase in the content of benzyl isothiocyanate, and of its precursor glucotropaeolin, in the roots of the host plants. The increased isothiocyanate content is one of the factors contributing to the resistance of L. sativum against P. ultimum. To be transformed into the fungitoxic compound benzyl isothiocyanate, glucotropaeolin has to be hydrolyzed by myrosinase, which can be produced either by plants or microorganisms. The Fusarium strain Ls-F-in-4-1 has a myrosinase activity but the strain Rs-F-in-11 has not. These results suggest that both strains are able to trigger the metabolic pathway leading to benzyl isothiocyanate production in the plant. In the case of the myrosinase-negative strain Rs-F-in-11, hydrolyzation into isothiocyanate is only due to the myrosinase activity of the plant, and in the other case, the myrosinase produced by the strain Ls-F-in-11 also would contribute to the production of isothiocyanate. Studies of the physiology of the Spirillum lipoferum recognized as the major organism responsible for N2-fixation in the roots of Digitaria decumbens cv transvala were performed in order to improve the methods of culture and help to explain the physiology of this N2-fixing grass bacterial association.Methods for isolation, purification and N2-fixation assays are described. Acetylene concentrations used for N2-ase activity measurements should be at least 12%. the Vmax of cultures in the log phase being at a pC2H2 of 0.12 atm and the apparent Km 0.022 atm. Optimal temperatures for N2-dependent growth are between 32 and 40°C. and little N2-fixation is observed below 24°C. At 42°C the N2-ase is inactivated. When cultures grown at 28 or 36°C are transferred to lower temperatures nitrogenase activity declines rapidly. One hour after transfer to 17°C activity is about half that before transfer and is maintained at this level for at least 8 h. After transfer to 10°C activity ceases after 1 h. Growth is very pH dependent, optimal growth on N2 occurring only between pH 6.8 and 7.8. Nitrogen fixation below pH 5.5 and above 8.0 is less than one-quarter of the optimal. No N2-fixation occurs in the absence of O2 and maximal N2-dependent growth is reached at 1.5% O2 in the gas mixture bubbled through liquid cultures.In contrast to previous reports, several sugars including glucose can be used by the Spirillum for N2-fixation, but only when small amounts of starter nitrogen or organic acids are added to the medium. Efficiencies of N2-fixation on malate and glucose are similar and about 60% of that of cells incorporating NH+4-N. Efficiency of NO-3 incorporation is 74% of that of NH+4-N grown cultures. High observed efficiencies (52 mg N2 fixed g?1 malate or glucose) are attributed to carbon limited growth at optimum or O2 limited conditions, both facilitated by slow diffusion rates through the semi-solid agar medium used.  相似文献

Sulphate sorption on to the surface of short‐range ordered minerals and precipitation of Al‐hydroxy sulphate contribute to the acid neutralizing capacity of soils. The correct measurement of total inorganic sulphate is thus essential in soils that are accumulating SO42– anions. We extracted SO42– by various solutions, namely 0.005 m Ca(NO3)2, 0.016 m KH2PO4, 0.5 m NH4F and 0.2 m acidic NH4‐oxalate (pH 3), from Vitric and Eutric Andosols exposed to prolonged deposition of acid and SO2 from an active volcano (Masaya, Nicaragua). We attributed sulphate extractable by KH2PO4 (20–3030 mg kg?1) to anion‐exchangeable SO42–, which was much smaller than NH4F‐ and oxalate‐extractable SO42– (400–9680 and 410–10 480 mg kg?1, respectively). Our results suggest the occurrence of a sparingly soluble Al‐hydroxy‐mineral phase extractable by both NH4F and oxalate. The formation of Al‐hydroxy minerals would result from the combination of enhanced weathering caused by strong acid loading and simultaneous occurrence of large SO42– concentrations in soil solution. Oxalate extracted slightly more inorganic SO42– than did NH4F, this additional amount of SO42– correlating strongly with oxalate‐extractable Si and Fe contents. Preferential occlusion of SO42– by short‐range ordered minerals, especially ferrihydrite, explains this behaviour. If we exclude the contribution of occluded sulphate then oxalate and NH4F mobilize similar amounts of SO42– and are believed to mobilize all of the inorganic SO42– pool.  相似文献

A gas lysimeter has been designed and used to measure directly the evolution of N2and N2O in a soil profile under field conditions. Concentrations of N2 in the soil atmosphere within the lysimeter as low as 2000–5000 p.p.m. have been achieved by flushing with N-free gas. A flow of gas into the base of the lysimeter forms a barrier against diffusion of soil air into the lysimeter during measurements. After reducing the N2 concentration in the soil core, a low concentration of N2enriched in N-15 is introduced. By monitoring changes in the 8 N value using a high-precision isotope mass spectrometer, rates of N2 evolution down to 6 kg N2-N ha?1 a?1 can be detected. N2O evolution was determined at the same time using the mass spectrometer in the single-beam mode.  相似文献

Agricultural peat soils are important sources of nitrous oxide (N2O). Emissions of N2O were measured from field plots of grass, barley, potatoes and fallow on a peat field in northern Finland during 2000–2002 and in southern Finland in 1999–2002. In the north the mean annual fluxes of N2O (with their standard errors) during 2 years were 4.0 (±1.2), 13 (±3.0) and 4.4 (±0.8) kg N ha?1 from the plots of grass, barley and fallow, respectively. In the north there were no significant thaw periods in the middle of winter. As a result, the thawing in the spring did not induce especially large N2O emissions. Emissions of N2O were larger in the south than in the north. In the southern peat field the mean annual fluxes during 3 years were 7.3 (±1.2), 15 (±2.6), 10 (±1.9) and 25 (±6.9) kg N2O‐N ha?1 for grass, barley, potato and fallow plots, respectively. Here, the largest single episodes of emission occurred during the spring thaw each year, following winter thaw events. An emission factor of 10.4 kg N2O‐N ha?1 year?1 for the N2O emission from the decomposition of the peat results from these data if the effect of fertilization according to the IPCC default emission factor is omitted. The direct effect of adding N as fertilizer on N2O emissions was of minor importance. On average, 52% of the annual N2O flux entered the atmosphere outside the cropping season (October–April) in the north and 55% in the south. The larger N2O fluxes from the peat soil in the south might be due to the more humified status of the peat, more rapid mineralization and weather with more cycles of freezing and thawing in the winter.  相似文献

为了研究生物油/柴油乳化燃料的燃烧特性,利用非离子表面活性剂复配,对热解生物油/柴油混合液进行了乳化,测量了乳化燃料的密度、热值、动力黏度及pH值。在SD1110型柴油机台架上进行4种不同配比的生物油/柴油乳化燃料的发动机台架试验,得出了柴油机燃用生物油/柴油乳化燃料和纯柴油的负荷特性和排放特性曲线,并且对乳化燃料和纯柴油的排放特性进行了对比。研究结果表明:生物油体积分数为20%的乳化燃料当量油耗率最低,乳化燃料CO的排放高于柴油的排放,且生物油含量越高CO排放越大,而乳化燃料的NO及碳烟的排放则优于纯柴油的排放。由于生物油/柴油乳化燃料的理化特性与柴油接近,可以作为普通柴油机的燃油使用。  相似文献

The capacity of different soils of New Caledonia to induce nodulation in Gymnostoma poissonianum was studied. The soils were sampled under five Gymnostoma species, Alphitonia neocaledonica (a non-nodulated endemic Rhamnaceae) and Pinus caribea (an introduced species) growing in various ecological conditions. Using G. poissonianum as trap-host, we observed a higher abundance of Frankia from Gymnostoma spp. in the rhizosphere of A. neocaledonica as compared with bare soils and P. caribea rhizosphere. The nodulating capacity of A. neocaledonica rhizosphere was almost similar to that of the five Gymnostoma species (symbiotic host) studied in the same stations. In comparison, bare soils or rhizosphere of P. caribea had poor nodulating capacities. We isolated fourteen Frankia strains from nodules of G. poissonianum after baiting with the rhizospheric soils of five Gymnostoma and A. neocaledonica. Using the PCR/RFLP method, we confirmed the similarity with those already described. Frankia was abundant in the rhizosphere of A. neocaledonica in all the sites studied. One explanation could be a positive tropism of Frankia towards species belonging to families having nodulated species, which is the case of A. neocaledonica endemic in New Caledonia. We can suppose that the non-nodulated plants belonging to these families can excrete some chemical substances able to attract Frankia and to induce its proliferation.  相似文献

试验研究雷公藤非生物碱对小菜蛾(Plutella xylostella)的生物活性结果表明,雷公藤非生物碱对小菜蛾具有较强的拒食、毒杀以及生长发育抑制作用。雷公藤非生物碱对2龄小菜蛾幼虫的选择性与非选择性拒食中浓度(AFC50)分别为34.89mg/L和42.33mg/L;毒杀中浓度(LC50)为49.86mg/L;100mg/L的雷公藤非生物碱对小菜蛾幼虫的生长发育抑制率达90%以上;但雷公藤非生物碱的杀卵作用不显著,以400mg/L雷公藤非生物碱处理小菜蛾卵后,其孵化率和孵化后幼虫存活率仍分别达53.48%和46%。  相似文献

Alkali soils are poor in sulphur-oxidizing bacteria. By appropriate enrichment, two bacterial strains resembling Thiobacillus thiooxidans and T. novellus have been isolated from an alkali soil. 以粉煤灰(Fly ash,FA)为原料,通过负载壳聚糖和钙离子制备一种新型吸附剂(Ca/CTS/FA)用于酸性大红3R(Acid scarlet 3R,AS 3R)染料的吸附去除,考察了最佳吸附条件和吸附性能,探讨了吸附动力学、吸附热力学及复合吸附剂的再生。结果表明:水体pH、吸附剂投加量、吸附时间和振荡频率均对吸附有影响。在不同温度下,Ca/CTS/FA对AS 3R的吸附动力学均能用准二级吸附速率方程精确描述(R2=1.00)。不同温度下的等温吸附数据分别用Langmuir模型、Freundlich模型和Dubinin-Radushkevich(D-R)模型进行拟合,结果表明等温吸附行为更符合Langmuir模型,同时也较好地符合Freundlich模型和D-R模型;由D-R方程获得的平均吸附能、表观活化能及热力学研究均表明该吸附过程由化学吸附、物理吸附和氢键作用共同控制。热力学参数中吸附自由能变为负值、焓变为正值说明该吸附是吸热性质的自发过程。Ca/CTS/FA复合吸附剂能用0.01 mol·L-1 NaOH溶液再生,至少可循环使用8次以上,再生率均在99%以上。  相似文献

The mineralization of native soil organic matter and the simultaneous diffusion of zero NH+4 and NO?3 to a solution sink of zero N concentration was analysed experimentally and theoretically for a fine sandy loam soil. Experimentally, the NH4 and NO3 ions produced in an incubated unsaturated soil column were allowed to diffuse through a sintered glass plate into a stirred solution sink. The distribution of NH+4 and NO?3 in the soil column was measured after various incubation times. The rate of ammonification was measured directly during incubation and the rate of nitrification modelled from nitrifier growth kinetics. A Freundlich equation was used to describe the equilibrium between soluble and exchangeable NH+4 in the soil. Terms for the microbial transformation of N and the adsorption-desorption of NH+4 were combined with diffusion equations which were solved numerically using finite difference methods. The model constructed was used to predict the NH+4 and NO?3 con-centration distributions in the soil column, and good agreement was obtained between the experimental and predicted concentration profiles. The use of the model for predicting the diffusive flux of mineral N to the outer surfaces of soil peds, where it is vulnerable to leaching, was demonstrated.  相似文献

To understand the process and the kinetics of potassium release from the clay interlayer in natural and arable soils in more detail, I tested the hypotheses that large, monovalent cations, especially NH4+ and Cs+, can reduce the release rates of K+ which is exchanged by Ca2+, even if these monovalent cations are present in concentrations of only a few μm . Percolation experiments were carried out with different illitic soil materials, some containing vermiculite, with 5 m m CaCl2 at pH 5.8 and 20°C, in some cases for over 7000 h. NH4+ and Cs+ both caused a large decrease in the rate at which K+ was released, Cs+ especially. Suppression began at 5 μm NH4+ Blocking by 20 μm NH4+ was easily reversible: the release rates readily increased when NH4+ was omitted from the exchange solution. Blocking by 2 μm Cs+ was equal to approximately 90% of that at 10 μm Cs+. Larger concentrations of Cs+ than 10 μm did not further reduce release but rather caused a slight increase, probably because of enhanced exchange of K+ by Cs+ without exfoliation of the interlayer space. Blocking by Cs+ was not reversible within > 7000 h of percolation by 5 m m CaCl2. The blocking effect was reproduced in several different soil materials using 10 μm Cs+ but was most pronounced in vermiculite-rich samples. As NH4+ is present in most arable soils, at least in concentrations of a few μm , I conclude that the observed effects are of significance in the K dynamics processes in soils, for example near the roots of plants. Further, very small concentrations of Cs+ in exchange solutions containing a large background of Ca2+ appear to be useful for suppressing K+ release from the interlayer in laboratory studies, probably without significantly altering the exchange at outer mineral surfaces.  相似文献

通过间歇式批实验和动力学实验研究了不同pH条件下磷酸盐在镁铝双氢氧化物(Mg-Al-LDH)、钠基膨润土(Na-Mt)及镁铁铝改性膨润土(Mg-Al-Mt、Fe-Mt和Fe-Al-Mt)的吸附特征。结果表明,在pH4.5~9.0范围内,随着pH的升高,Na-Mt,Fe-Mt和Fe-Al-Mt3种矿物对磷的吸附率相应减少,镁铝双氢氧化物对磷的吸附率有所增加;Mg-Al-LDH、Fe-Mt和Fe-Al-Mt的对磷吸附率约为95%,比Mg-Al-Mt高40%,比Na-Mt高80%。用Langmuir方程描述磷的等温吸附过程,最大吸附容量(Qm)大小顺序为Fe-Al-Mt〉Fe-Mt〉Mg-Al-LDH〉Mg-Al-Mt〉Na-Mt,b值大小依次为Mg-Al-LDH〉Fe-Mt〉Fe-Al-Mt〉Mg-Al-Mt〉Na-Mt,最大缓冲容量(Qmb)以Mg-Al-LDH的为最大,Na-Mt的为最小;Freundlich等温吸附方程参数KF代表相对吸附容量,以Mg-Al-LDH的KF值最高,依次是Fe-Mt、Fe-Al-Mt和Mg-Al-Mt,Na-Mt的KF值最小,这与Qmb的结果一致;决定系数(R2)表明,Langmuir等温吸附方程能更好地拟合铁改性蒙脱土和铁铝改性蒙脱土,而Freundlich方程对钠蒙脱土,双氢氧化物和镁铝改性蒙脱土的拟合效果要好。磷酸盐吸附过程分为快反应和慢反应,用动力学实验数据进行拟合,准二级动力学方程能够更好的拟合改性蒙脱土和Mg-Al-LDH对磷的动力学吸附数据,其决定系数为0.999;Elovich方程拟合改性蒙脱土磷酸盐的吸附数据也有很好的相关性(R2为0.89~0.94)。Na-Mt矿物磷酸盐的吸附用抛物线扩散方程描述最为合适,原因可能是磷酸盐镶嵌在矿物层间是一个扩散过程,不涉及化学吸附过程。  相似文献

To evaluate the atmospheric load of reactive gaseous nitrogen in the fast-developing Eastern China region, we compiled inventories of nitrous oxide (N2O), nitrogen oxide (NOx) and ammonia (NH3) emissions from a typical rural catchment in Jiangsu province, China, situated at the lower reach of the Yangtze River. We considered emissions from synthetic N fertilizer, human and livestock excreta, decomposition of crop residue returned to cropland and residue burning, soil background and household energy consumption. The results showed that, for the 45.5 km2 catchment, the annual reactive gaseous emission was 279 ton N, of which 7% was N2O, 16% was NOx and 77% was NH3. Synthetic N fertilizer application was the dominant source of N2O and NH3 emissions and crop residue burning was the dominant source of NOx emission. Sixty-seven percent of the total reactive gaseous N was emitted from croplands, but on a per unit area basis, NOx and NH3 emissions in residential areas were higher than in croplands, probably as a result of household crop residue burning and extensive human and livestock excreta management systems. Emission per capita was estimated to be 18.2 kg N year−1 in the rural catchment, and emission per unit area was 56.9 kg N ha−1year−1 for NH3 + NOx, which supports the observed high atmospheric N deposition in the catchment. Apparently, efficient use of N fertilizer and biological utilization of crop straw are important measures to reduce reactive gases emissions in this rural catchment.  相似文献

为开发一种新型的、功能性天然食品添加剂,该文以绿色木霉菌T-YY发酵液冻干粉(色素粗品)为试验材料,初步研究其紫外-可见光谱性质与稳定性.结果表明,色素粗品中至少含有4种以上不同性质的黄色素,该黄色素易溶于水和醇,色价高,在紫外区及可见光区各有一个吸收峰,最大吸收波长分别为286.5 nm和380 nm;在酸碱环境中色调保持亮黄色,其水溶液在pH<5.0时出现浑浊,在pH≥5.0时呈亮黄色;pH值3.0~9.0色素溶液在室内自然光(25℃)下放置7 d后色调稳定;热稳定性好,100℃加热30 min色素保留率达到90.2%;抗H2O2、HNO3氧化能力强,但不耐Na2SO3还原作用.Fe3 、Cu2 和Mn2 的对色素具有消褪作用, Fe2 、Mg2 、Zn2 、Ca2 、K 、Na 则对其有不同程度的保护和增色作用.试验表明,绿色木霉T-YY黄色素在稳定性上呈现出一定优势,因此具有较大的开发潜力.  相似文献

小麦/玉米套作田棵间土壤蒸发的数学模拟   总被引:7,自引:5,他引:7  
为了探明小麦/玉米套作条件下棵间土壤蒸发规律及内在机制,该文将 Ritchie 模型和间套作群体光能传输模型结合起来模拟了小麦/玉米套作田的棵间土壤蒸发,并用2012年和2013年微型蒸渗仪的实测值对该方法进行了验证。结果表明,2 a模拟值和实测值的变化趋势都非常一致。与实测蒸发相比,该文所建模型2012年模拟结果的均方根误差为0.447 mm/d,平均绝对误差为0.331 mm/d,分别比原Ritchie模型降低16.8%和20.8%。在实测数据的88 d,累计实测蒸发量为107.2 mm,而模型的模拟值为100.5 mm,仅低估实测值6.7%。在2013年实测数据的68 d,累计实测蒸发量为83.1 mm,而模型的模拟值为73.7 mm,低估11.3%。模拟值与实测值的均方根误差和平均绝对误差分别为0.465和0.333 mm/d,略大于2012年。套作群体根系层水量平衡分析结果表明,小麦/玉米套作系统整个生育期土壤蒸发占总蒸发蒸腾的比例高达41.1%,故有必要在该套作农田实施秸秆覆盖等农艺措施,以减少棵间土壤蒸发,提高土壤水分的利用效率。该研究成果可为小麦套作种植模式下田间水分管理提供依据。  相似文献

Humic acids were isolated from nine topsoils in New Zealand tussock grasslands. Cross-polarization 13C NMR spectra of solid samples were used to estimate fractions of carbon contained in different types of chemical functional groups. The degree of oxygen substitution of aromatic rings showed a strong negative correlation with soil development. Aromaticities greater than 0.25 were found in humic acids from only the two least-developed soils.  相似文献
