Zabrotes subfasciatus (Boheman) is the main pest of common beans ( Phaselous vulgaris ). Wild legume seeds from Olneya tesota contain a lectin, PF2, that shows insecticidal activities against this insect. The binding of PF2 to midgut glycoproteins of 20-day-old larvae was evaluated using PF2 affinity chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the proteins retained on the gel revealed several putative glycoproteins, ranging in mass from 17 to 97 kDa. Subsequent protein digestion and analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) provided amino acid fragments that identified an α-tubulin, cytochrome c oxidase subunit I, an odorant receptor, and a lysozyme from available insect sequence databases. The potential of these proteins to serve as part of the mechanisms involved in the insecticidal activity of PF2 to Z. subfasciatus is discussed.  相似文献

Cartap hydrolysis relative to its action at the insect nicotinic channel   总被引:2,自引:0,他引:2  
The insecticide cartap is the bis(thiocarbamate) derivative of 2-(dimethylamino)propane-1,3-dithiol, which on oxidation forms the natural toxicant nereistoxin (NTX) [4-(dimethylamino)-1,2-dithiolane]. Both cartap and NTX are ion channel blockers of the nicotinic acetylcholine receptor (nAChR). Cartap was originally proposed to act only after metabolic conversion to NTX and later suggested to block directly without activation. The present study uses a new approach to differentiate these hypotheses, that is, pH effects on channel-blocking activity and hydrolysis rates. As controls, mecamylamine (the classic channel blocker) and NTX are stable and similar in channel-blocking potency ([(3)H]thienylcyclohexylpiperidine binding assay, honeybee nAChR) at pH 6.1-8.4. In contrast, cartap is >200-fold more effective at pH 7.4 than at pH 6.1, indicating that it undergoes hydrolytic activation. Cartap slowly hydrolyzes to cartap monothiol at pH 6.1 but quickly forms the dithiol and some NTX at pH 7.4. The relationship between potency and hydrolysis products at various pH ranges suggests that cartap dithiol is the most plausible blocking agent.  相似文献

Purpose: Plant-derived essential oils and their encapsulations have recently received increasing interest as an alternative to synthetic biopesticides suitable for integrated pest management and organic farming. In this study, combinations of essential oils (EO) from Rosmarinus officinalis with Cymbopogon citratus and Pelargonium graveolens with Thymus vulgaris in two formulations including encapsulate were tested for their potential for wheat protection against adults and larvae of Oulema melanopus (L.).

Materials and methods: The chemical composition of the essential oils was analyzed by gas chromatography-mass spectrometry (GC-MS). Testing of insecticidal activity of the evaluated substances was conducted with 2.5?ml of tested combinations of EOs sprayed on the paper. Ten adults or larvae of O. melanopus were placed into a vessel. Control of mortality of O. melanopus specimens was assessed after 24?h after establishment of the experiment.

Results and conclusions: Tarsal contact toxicity assay showed the effectiveness of EO in concentration against O. melanopus larvae and adults in both formulations (essential oil and encapsulation) causing 100% mortality within 24?h. These results indicate the great potential of these essential oils and their encapsulations for future use in crop protection against insect pests.  相似文献

Binding of estrogen receptor (ER) to estrogen response element (ERE) induces gene activation and is an important step in estrogen-induced biological effects. Here, we investigated the effects of some dietary phytoestrogens such as the isoflavones genistein and daidzein, its metabolite equol, and the coumestane coumestrol on the binding rate of ERalpha and ERbeta to ERE by a nonradioactive real-time method, the Biacore Technology. ERalpha and ERbeta were able to bind to ERE immobilized on the surface of a sensor chip even in the absence of estrogens. 17beta-Estradiol and phytoestrogens induced an increase in ER binding to ERE in a concentration-dependent manner. 17beta-Estradiol was a more potent activator of binding than the phytoestrogens studied. The concentrations of 17beta-estradiol inducing an increase in the binding response of ERalpha and ERbeta to ERE by 50% (EC(50)) as compared to unliganded ER were 0.03 and 0.01 microM, respectively. Regarding the efficacy of activation of ERalpha, from the most to the least effective compound, the sequence and the EC(50) were as follows: 17beta-estradiol (0.03 microM) > coumestrol (0.2 microM) > equol (3.5 microM) > genistein (15 microM) > daidzein (>300 microM) and for ERbeta 17beta-estradiol (0.01 microM) > coumestrol (0.025 microM) > genistein (0.03 microM) > daidzein (0.35 microM) > equol (0.4 microM). The ratios EC(50)alpha/EC(50)beta were calculated to be for 17beta-estradiol, 3; coumestrol, 8; equol, 8.8; genistein, 500; daidzein > 850. These ratios indicate that genistein and daidzein preferentially activate the binding of ERbeta to ERE. The endogenous hormone 17beta-estradiol as well as coumestrol and daidzein metabolite equol activate the binding of ERbeta to ERE only slightly more effectively than the binding of ERalpha to ERE. Thus, the effect of daidzein can be changed from a specific activator of ERbeta to an activator of both ER isotypes alpha and beta in humans who are able to convert daidzein to equol. While the results of the measurements with ERalpha were in line with the binding affinities of compounds tested for ER, there was a distinct difference between our results and the binding affinities of phytoestrogens for the ERbeta. This leads to the conclusion that phytoestrogens differ not only in their binding affinities for the ER, but also in their potential to increase the rate of receptor binding to the ERE.  相似文献

The essential oils from leaves and inflorescences of Hyptis martiusii Benth were analyzed by GC-MS. Twenty-six compounds representing 93.2% of the essential oil of leaves were characterized; Delta-3-carene (22.5%), 1,8-cineole (24.27%), beta-caryophyllene (6.15%), and bicyclogermacrene (6.32%) were found as the major components. In the essential oil of inflorescences 27 compounds representing 87.7% of the oil were identified. The major components were Delta-3-carene (13.5%), alpha-pinene (5.78%), beta-caryophyllene (6.59%), viridiflorene (8.25%), and germacrene B (5.21%). The essential oil of leaves and 1,8-cineole showed pronounced insecticidal effect against Aedes aegypti larvae and Bemisia argentifolii, the vectors of dengue fever and white fly fruit plague, respectively.  相似文献

A bioactivity-directed investigation of an extract of the New Zealand clubmoss, Lycopodium varium, collected on subantarctic Campbell Island, has led to the isolation of the alkaloid huperzine A (1) as the major antifeedant and insecticidal component. Huperzine A showed insecticidal activity against the Australian carpet beetle, Anthrenocerus australis (LD(50) = 110 ppm), the Australian sheep blowfly, Lucilia cuprina (LD(50) = 2380 ppm), and the webbing clothes moth, Tineola bisselliella (LD(50) = 630 ppm). Feeding by A. australis was reduced by 97% at 63 ppm.  相似文献

This paper describes the purification and characterization of a novel protein from the seeds of Pouteria torta (family Sapotaceae). The protein was purified by a combination of gel filtration, ion-exchange, and reverse phase chromatographies. SDS-PAGE of the purified protein resulted in a single protein band of 14 kDa in the presence and absence of DTT. The lectin-like activity of pouterin was best inhibited by glycoproteins such as fetuin, asialofetuin, heparin, orosomucoid, and ovoalbumin. Pouterin inhibited the growth of the fungi Fusarium oxysporum and Colletotrichum musae and of the yeast Saccharomyces cerevisiae. The incorporation of pouterin into an artificial diet (final concentration = 0.12%, w/w) caused 50% mortality in larvae of the insect Callosobruchus maculatus, whereas 0.08% pouterin produced an ED50.  相似文献

A series of aurones were prepared from various phenols via phenoxy acetic acids and coumaranones and evaluated for insect antifeedant activity against the common cutworm (Spodoptera litura). The naturally occurring aurone was most active at an ED50 of 0.12 micromol/cm2. The synthetic precursor, coumaranones, showed that the introduction of methoxyl and methyl groups to the benzene ring increased insect antifeedant activity. Similarly, the tested aurones showed that the introduction of methoxyl group to the A and/or B rings increased the insect antifeedant activity, but 4,5,6- and 3',4',5'-trisubstituted compounds did not show this activity in this test. The hydroxylation of aurones in the B ring should be disadvantageous for insect antifeedant activity against S. litura. Although the melting points did not correlate well with the insect antifeedant activity, compounds that were nearly inactive had high melting points. A significant correlation was noted between biological activity (pED50) and a hydrogen-bonding parameter calculated from the Rf value obtained from SiOH thin-layer chromatography and a lipophilicity parameter (log k) calculated from the retention time in ODS high-performance liquid chromatography. The respective correlation coefficients (r) were -0.83 and -0.70. The introduction of alkoxy and alkyl groups along with adequate hydrogen bonding seems to contribute to the antifeedant activity of the compounds tested.  相似文献

Starting from precocenes I and II, four of their derivatives with a lactone moiety were obtained. The compounds have been assessed as antifeedants against several diverse insect species including the storage pests the confused flour beetle (Tribolium confusum Duv., larvae and adults), the granary weevil beetle (Sitophilus granarius L., adults), and the khapra beetle (Trogoderma granarium Ev., larvae) and against the herbivorous pest insects Colorado potato beetle (Leptinotarsa decemlineata Say, adults and larvae) and aphids (Myzus persicae Sulz.). Precocenes, especially precocene II, showed a very strong antifeedant effect against all storage pests and aphids. The introduction of a lactone moiety caused a decrease in antifeedant activity against these species. Both precocenes were moderately active against L. decemlineata adults. The best antifeedants to this species were precocene derivatives, especially iodolactones. The introduction of iodine into a molecule had a great effect on the antifeedant activity of those compounds.  相似文献

This study was aimed at determining the effect of raw full-fat soybean in the digestion/absorption of broilers and to evaluate the role of soybean agglutinin (SBA) in the pathogenesis of the runting and stunting syndrome of broilers. Six broiler groups were fed with six experimental diets for 42 days in which the defatted soybean meal of the basal diet was substituted with increasing raw full-fat soybean percentages ranging from 0 to 100%. The results suggested that SBA included in the raw full-fat soybean can play a role in the pathogenesis of the runting and stunting syndrome in broilers since binding and incorporation of SBA to enterocytes were associated with hyperplasia and dysplasia of the intestinal epithelium, as suggested by the progressive increases (P < 0.001) of mitosis/crypt in the duodenum and in the midintestine, as well as by intestinal villi atrophy (P < 0.001) in the duodenum and the midintestine. Indigestion and malabsorption of proteins induced progressive low weight gain (P < 0.001) up to 50% with 100% of raw full-fat soybean in the diet at 42 days. Xanthophylls in serum and skin yellowness were enhanced linearly in the broilers fed 42 days with increasing percentages of raw full-fat soybean in the diet. Increased xanthophylls absorption and decreased protein absorption can be caused by pathological changes in the epithelial intestine triggered by SBA binding and internalization.  相似文献

The bioactivity of caffeine aqueous solutions (0.20-2.00 wt %) and caffeine oleate emulsions (20 vol % oil, 2.00 wt % surfactant, 0.04 wt % caffeine, 0.05 wt % oleic acid) was assessed against two biological models: Drosophila melanogaster and Hypothenemus hampei. The caffeine aqueous solutions showed no insecticidal activity, whereas caffeine oleate emulsions had high bioactivity against both D. melanogaster and H. hampei. By preparing the caffeine oleate emulsions with anionic surfactants (i.e., sodium lauryl sulfate, sodium laureate, and sodium oleate), we obtained a lethal time 50 (LT50) of 23 min. In the case of caffeine oleate emulsions prepared with nonionic surfactants (i.e., Tween 20 and Tween 80), a LT50 of approximately 17 min was observed. The high bioactivity of the caffeine oleate emulsion against H. hampei opens the possibility of using this insecticide formulation as an effective way to control this pest that greatly affects coffee plantations around the world.  相似文献

Under traditional wine-making conditions, this work examines the beta-glycosidic activity of Oenococcus oeni on glycosylated aroma compounds of Tannat wines during malolactic fermentation (MLF) by comparing the changes on selected aglycones liberated. MLF diminished the content of all the glycosylated compounds. The level of the free aroma components was slightly modified by the action of the malolactic fermentation so that the cleavage of the glycosidic linkage by the beta-glycosidic activity of O. oeni did not appear to increase significatively the aglycone contents. The consequences of further chemical rearrangements of the algycones under wine conditions were explored using synthesized glycoconjugates on synthetic medium. Bacteria could also be responsible for the cleavage of aroma glycosylated compounds, being the aglycone adsorbed on polysaccharides or peptidoglycans and was released into the external medium. This hypothesis was studied through the evaluation of a stable arrangement of aroma compounds with polysaccharides produced by lactic acid bacteria. A possible retaining of free-made aroma compounds into the whole cells of O. oeni was also investigated through cell culture analysis. Through the results obtained, we assume stable linkage of aroma compounds with bacterial polysaccharides.  相似文献

Summary The distribution of phosphatase activity and of phosphate fractions of the soil in the proximity of roots was studied in order to evaluate the significance of phosphatases in P nutrition of various plants (Brassica oleracea, Allium cepa, Triticum aestivum, Trifolium alexandrinum). A considerable increase in both acid and alkaline phosphatase activity in all the four soil-root interfaces was observed. Maximum distances from the root surface at which activity increases were observed ranged from 2.0 mm to 3.1 mm for acid phosphatase and from 1.2 mm to 1.6 mm for alkaline phosphatase. The increase in phosphatase activity depended upon plant age, plant species and soil type. A significant correlation was noticed between the depletion of organic P and phosphatase activity in the rhizosphere soil of wheat (r = 0.99**) and clover (r = 0.97**). The maximum organic P depletion was 65% in clover and 86% in wheat, which was observed within a distance from the root of 0.8 mm in clover and 1.5 mm in wheat. Both the phosphatases in combination appear to be responsible for the depletion of organic P.  相似文献

Penitrem G (7), a new indole-diterpenoid compound, has been isolated together with the already known mycotoxins penitrems A-D (1-4) and F (6) from the mycelium of Penicillium crustosum Thom. The structure of penitrem G was established on the basis of spectroscopic data. In addition, paspaline (8), another indole-diterpenoid mycotoxin that has not been previously described in this fungus, was also isolated. These compounds were tested for insecticidal activity against the hemipteran Oncopeltus fasciatus Dallas and the dipteran Ceratitis capitata Wiedemann. Penitrems A-D and F showed convulsive and insecticidal activities against both insect species. In addition, important reductions in the fecundity and fertility of the surviving C. capitata females were observed. In contrast, penitrem G and paspaline did not show any kind of activity. Mortality data and sublethal effects of the treatments have allowed preliminary structure-activity relationships to be proposed.  相似文献

The potato (Solanum tuberosum L.) tuber storage protein, patatin, was purified to homogeneity with a molecular mass of 45 kDa. The purified patatin showed antioxidant or antiradical activity by a series of in vitro tests, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical (half-inhibition concentration, IC(50), was 0.582 mg/mL) scavenging activity assays, anti-human low-density lipoprotein peroxidation tests, and protections against hydroxyl radical-mediated DNA damages and peroxynitrite-mediated dihydrorhodamine 123 oxidations. Using electron paramagnetic resonance (EPR) spectrometry for hydroxyl radical detections, it was found that the intensities of the EPR signal were decreased by the increased amounts of patatin added (IC(50) was 0.775 mg/mL). Through modifications of patatin by iodoacetamide or N-bromosuccinimide, it was found that the antiradical activities of modified patatin against DPPH or hydroxyl radicals were decreased. It was suggested that cysteine and tryptophan residues in patatin might contribute to its antioxidant activities against radicals.  相似文献

In a pre-alpine meadow in the Toggenburg (Switzerland), casts of the earthworm Aporrectodea nocturna, initially detected 20 years ago around a newly planted hedge, now extend 170 m from the hedge. The abundance of A. nocturma decreased between 140 and 170 m from the hedge from 237 (site: An+) to 0 (site: An?) individuals m?2. The worm's activity and the macroporosity it has created have been studied using CAT scanning followed by automated image analysis of pores from 1·5 to 9 mm diameter. Brightness values in images from CAT scanning showed distinct local frequency maxima for stones, soil matrix and macropores. Measured diameters of pores were strongly linearly correlated with diameters of needles used to produce artificial pores. Pores from 1·5 to 3 mm were most abundant. The maximum number of pores from 1·5 to 9 mm (about 1600 m?2) was larger than the maximum number of all earthworms (about 480 m?2). Modelling the cast production of A. nocturna indicated that 23·8 kg m?2 were deposited on the soil's surface within 3 months. The measured macroporosity (1·5–6 mm) was one-quarter to one-sixth of the pore volume corresponding to the removed casts. Thus, net production of pores by earthworms was a result both of the burrowing activity and of the refilling with eroded cast material. Depth distributions of bulk density, total porosity and pores from 1·5 to 9 mm were different at An+ from those at An?. However, diffusion of butane was equal at An+ and An?. Three-dimensional reconstruction suggested that the disposition (continuity, interconnectivity) of pores was more important for gas diffusion than the structure (size distribution, frequency) and bulk soil parameters.  相似文献

Summary Nitrification activity (formation of NO 2 + NO 3 per unit soil weight) was measured in the surface layer of 15 presubmerged soils incubated in petri dishes under flooded but aerobic conditions. soils with pH above 5 nitrified quickly, whereas soils with pH below this level did not nitrify or nitrified slowly. The pH values between 7 and 8.5 were optimal for nitrification. Organic-matter levels in the 15 soils of our study did not influence their nitrification activities. In a follow-up greenhouse pot study, after a period of 3 weeks, 15N-balance measurements showed that the loss of N through apparent denitrification did not follow the nitrification patterns of the soils observed in the petri dishes. Apparent denitrification accounted for 16.8% and 18.9% loss of 15N from a soil with insignificant nitrification activity and a soil with high nitrification activity, respectively. These results, thus, indicate a lack of correspondence between the nitrification activities of soil and the denitrification loss of N when the former was measured in the dark and the latter was estimated in the light. Soils that nitrified in the darkness of the incubator did not nitrify in the daylight in the greenhouse.  相似文献

In this study, a rhamnose-binding lectin from the roe of chinook salmon (Oncorhynchus tshawytscha) was purified and characterized, and its biological activities were examined in several model systems. Chinook salmon roe lectin had a molecular mass of 30 kDa and agglutinated rabbit and bovine erythrocytes. The hemagglutination activity of the lectin was not affected by metal ions. The lectin was stable up to 70 °C and between pH 4 and pH 11. Chinook salmon roe lectin did not exert antifungal activity toward the fungal species tested and did not exhibit mitogenic response toward mouse splenocytes up to a concentration of 5 mg/mL. The lectin had selective antiproliferative activity toward human breast cancer MCF-7 cells and hepatoma Hep G2 cells. It also induced the production of nitric oxide from mouse peritoneal macrophages. This is the first report that demonstrates these biological activities from chinook salmon roe lectin.  相似文献
