Background: DNA methylation plays a vital role in reproduction.Entire genome DNA methylation changes during the oestrous phase(ES) and dioestrous phase(DS) in the ovaries of Guanzhong dairy goats were investigated using bisulphite sequencing to understand the molecular biological mechanisms of these goats' oestrous cycle.Results: We discovered distinct genome-wide DNA methylation patterns in ES and DS ovaries.A total of 26,910 differentially methylated regions were upregulated and 21,453 differentially methylated regions were downregulated in the ES samples compared with the DS samples(P-values ≤0.05 and fold change of methylation ratios ≥2).Differentially methylated region analysis showed hypomethylation in the gene body regions and hypermethylation in the joining region between upstream regions and gene bodies.The methylation ratios of the STAR,FGF2,FGF12,BMP5 and SMAD6 genes in the ES samples were lower than those of the DS samples(P-values≤0.05 and fold change of methylation ratios ≥2).Conversely,the methylation ratios of the EGFR,TGFBR2,IGF2 BP1 and MMD2 genes increased in the ES samples compared with the DS samples.In addition,223 differentially methylated genes were found in the GnRH signalling pathway(KO04912),ovarian steroidogenesis pathway(KO04913),oestrogen signalling pathway(KO04915),oxytocin signalling pathway(KO04921),insulin secretion pathway(KO04911) and MAPK signalling pathway(KO04010).Conclusions: This study is the first large-scale comparison of the high-resolution DNA methylation landscapes of oestrous and dioestrous ovaries from dairy goats.Previous studies and our investigations have shown that the NR5 A2,STAR,FGF2 and BMP5 genes might have potential application value in regulating caprine oestrus.  相似文献

The majority of pregnancy loss in ruminants occurs during the first three weeks after conception, particularly during the period of conceptus elongation that occurs prior to pregnancy recognition and implantation. This review integrates established and new information on the biological role of ovarian progesterone(P4), prostaglandins(PGs),interferon tau(IFNT) and cortisol in endometrial function and conceptus elongation. Progesterone is secreted by the ovarian corpus luteum(CL) and is the unequivocal hormone of pregnancy. Prostaglandins(PGs) and cortisol are produced by both the epithelial cells of the endometrium and the trophectoderm of the elongating conceptus. In contrast, IFNT is produced solely by the conceptus trophectoderm and is the maternal recognition of pregnancy signal that inhibits production of luteolytic pulses of PGF2αby the endometrium to maintain the CL and thus production of P4. Available results in sheep support the idea that the individual, interactive, and coordinated actions of P4, PGs, IFNT and cortisol regulate conceptus elongation and implantation by controlling expression of genes in the endometrium and/or trophectoderm. An increased knowledge of conceptus-endometrial interactions during early pregnancy in ruminants is necessary to understand and elucidate the causes of infertility and recurrent early pregnancy loss and provide new strategies to improve fertility and thus reproductive efficiency.  相似文献

This experiment was conducted to investigate the effects of dietary sodium acetate on lactation performance and rumen microbiota of dairy goats. Fifteen healthy Xinong Saanen dairy goat ewes with similar body weight of (53.54±3.07) kg, the same parity (2 parities) and in mid-lactation [(60±5) d] were randomly divided into 3 groups with 5 replicates in each group. Dairy goats in the control group were fed a basal diet, and those in the experimental groups were fed the basal diet supplemented with 30 (experimental group I) and 60 g/ (d.head) (experimental group II) sodium acetate, respectively. The pre-trial period lasted for 7 days and the experimental period lasted for 21 days. The milk yield of dairy goats was recorded during the experiment. At the end of the experiment, milk samples were collected to analyze the changes in milk composition, and rumen contents of dairy goats were collected from two groups (control group and experimental group II) with significant differences in milk fat percentage. Gas chromatography and 16S rDNA sequencing technology were used to analyze the rumen fermentation parameters and rumen microbial composition and structure of dairy goats. The results showed as follows: 1) compared with the control group, dietary sodium acetate extremely significantly increased the daily milk yield of dairy goats (P<0.01), and the daily milk yield in experimental groups I and II was increased by 0.29 and 0.22 kg, respectively (P<0.01); in experimental group II, the milk fat percentage was significantly increased by 1.30 percentage points (P<0.01), and the non-fat solid content in milk was increased by 3.10% (P>0.05). 2) Compared with the control group, the rumen butyric acid content of dairy goats in experimental group II was significantly decreased (P<0.01), and there was no significant difference in rumen pH, the other volatile fatty acid content and acetic acid to propionic acid ratio (P>0.05). 3) Compared with the control group, the Sobs index, Chao1 index and Ace index of rumen microbiota in experimental group II were significantly increased (P<0.05); the results of similarity analysis (ANOSIM) also showed that there were significant differences in the structure of rumen microbiota between the two groups (P<0.05). 16S rDNA sequencing results showed that at the phylum level, Bacteroidetes and Firmicutes were the dominant phyla in the rumen of the two groups of dairy goats; at the genus level, Prevotella_1 was the dominant genus in the rumen. Compared with the control group, the relative abundances of Ruminococcus_1, Ruminococcaceae_UCG-005, Prevotellaceae_UCG-004 and Lachnoclostridium_12 in rumen of dairy goats in experimental group II were significantly increased (P<0.05), while the relative abundances of Succiniclasticum and Methanobrevibacter in rumen was significantly decreased (P<0.05). 4) The results of correlation analysis showed that rumen microbiota was correlated with daily milk yield, milk fat percentage, milk protein percentage and non-fat solid content of dairy goats. In conclusion, dietary 60 g/ (d.head) sodium acetate can significantly increase the daily milk yield and milk fat percentage, significantly reduce rumen butyrate content, and improve the lactation performance of dairy goats by improving rumen microbial composition and structure. © 2023 Authors. Background: Haemonchosis is a major economic problem in goat production in humid, tropical and subtropical regions. The disease is caused by an abomasal nematode, Haemonchus contortus, which is highly pathogenic in small ruminants. The aim of this study was to identifying single-nucleotide polymorphisms(SNP) that were associated with fecal egg counts(FEC) and could be used as markers to identify resistance to H. contortus in goats.Results: Ten novel variants in the CI TA, ATP2 A3, HSPA8, STAT5 B, ESYT1, and SERPING1 genes were associated with FEC in goats with a nominal significance level of P 0.05. Two missense mutation in the exon region of the caprine CI TA gene resulted in replacement of arginine with cysteine at position 9473550(R9473550 C) and aspartic acid with glutamic acid at position 9473870(D9473870 E). Chinese goat breeds had significantly higher FEC than Bangladeshi goat breeds within their respective genotypes. Polymorphism information content(PIC), effective al ele number(Ne), and heterozygosity(He)were greatest for the STAT5 B_197_A G SNP locus in al goat breeds. Pairwise coefficients of linkage disequilibrium(D′, r~2)revealed complete LD(r~2= 1) between significant SNP polymorphisms in CI TA and SERPING1 and strong LD(r~2= 0.93 and0.98) between polymorphisms in HSPA8 and ATP2 A3, respectively. Correlation coefficient(r) between FEC and body weight(BW) was significantly positive(r = 0.56***, P 0.001) but that between FEC and packed cel volume(PCV) was negatively significant(r =-0.47**, P 0.01) in the total population of goats. On the other hand, correlation coefficient(r)between BW and PCV was not significant in total population of goats. Association analysis revealed that haplotypes within ATP2 A3, HSPA8, and SERPING1 were significantly associated with FEC. Quantitative real-time PCR revealed that the relative expression of m RNA was higher(P 0.001) for resistant, compared to susceptible, groups of goats for al candidate genes except CI TA.Conclusions: This study identified SNP markers that can potential y be used in marker-assisted selection programs to develop goat breeds that are resistant to H. contortus.  相似文献
