Summary The latent periods, wilting rates and percentages of diseased plants were analyzed for 11 carnation cultivars after root and after stem inoculation with race 2 of Fusarium oxysporum f.sp. dianth. There was no conclusive evidence for the presence of an independent extravascular resistance mechanism, except for Lena plants in which, additional to the vascular resistance components, independent root-bound factors causing retardation of the colonization and wilting process were found. A large variation was observed in the ablity of the cultivars to localize the pathogen in the vascular tissue shortly after infection of the xylem. This ability was positively correlated with the latent period, and negatively with the wilting rate and final disease index. In resistant cultivars, secondary compartmentalization of the fungus higher up in the stem was also observed. After stem inoculation, differences among the cultivars in localization ability and wilt-retarding actors could be identified at an early stage by comparing the precentages of non-colonized plants or the percentages of plants lacking vascular discolouration.  相似文献

Summary Four clones of the Chilota Potato Collection from the germplasm bank of the Universidad Austral de Chile, with potential combined resistance to PVX, PVY and PVS, were mechanically inoculated with these viruses in order to demonstrate wether they were truly resistant or not. Virus coat protein in the inoculated clones was determined by means of NCM-ELISA, DAS-ELISA, Western Blot and NASH.Two of the four inoculated clones were resistant to both PVY and PVS. Summary A procedure for plant regeneration from callus culture of potato, Solanum tuberosum L. is described. Calli were induced from 1–2 mm long shoot apices of potato cultivars Cara and A25/19 on half-strength Murashige and Skoog's medium (half-MS) supplemented with 3.2 mg IAA (indole-3-acetic acid), 1.0 mg kinetin (6-furfurylamino)purine], and 0.5 mg 2,4-D [2,4-dichlorophenoxy)acetic acid]/1. Sixty percent explants produced nodular calli on this medium within 30 days. Calli differentiated into shoot-primordia when subcultured on half-MS medium supplemented with 0.5 mg 2,4-D and 1.0 mg zeatin [6-(4-hydroxy-3-methybut-2 enylamino)amino purine]/1. Differentiated calli on half-MS medium without growth hormones produced complete plantlets which were cloned on the same medium and transferred into soil.  相似文献

Summary A total of 934 potato cultivars and clones from different breeding programmes across the world were tested for their reaction to early blight (Alternaria solani) in Brasilia-DF, Brazil. This field trial was set up in order to identify tetraploid potato genotypes with useful levels of resistance to early blight (EB) that were not correlated with late maturity or lateness. Artificial inoculation (spread ofA. solani infected leaves over plant canopy) was employed even though EB occurs naturally at epidemic levels in the test area. Four disease assessments were made every 10 days, beginning 45 days after planting. Based on the evaluation made 65 days after planting, only 27 clones were identified that had less lesioned leaf area than the resistant standard (cv. Aracy). Area under disease progress curve (AUDPC) was also used as a criterion of resistance. Using AUDPC as a parameter, 22 genotypes were identified presenting values lower than or identical to cv. Aracy. Differences in the commercial yield were recorded in genotypes with identical response to EB, suggesting that distinct levels of tolerance to EB also exist. In spite of the strong correlation between EB resistance and late maturity, some genotypes derived from the parental materials NDD 277-2, Kufri Jyoti, CIP 377888-7, Maine-28, and the clones CFS 69.1 and I-853 were identified as having good levels of EB field resistance not linked with the negative phenotypic trait of lateness. These materials presented medium to medium-early maturity with a vegetative cycle of around 90–95 days. Some selected genotypes presented more acceptable agronomic characteristics including resistance toPhytophthora infestans, potato leaf roll luteovirus (PLRV) and potato virus Y (PVY). The EB resistance sources identified in this work can be recommended as parental materials for widening the genetic base for EB disease resistance in breeding programmes for highland tropical and subtropical areas.  相似文献

In garden asparagus, Fusarium crown and root rot is the main cause of crop decline. Since chemical treatments are inefficient, efforts should focus on the development of resistant cultivars to control the disease. Toxic culture filtrate (TCF) of F. oxysporum has affected asparagus pollen germination and tube growth. Consequently, gametophyte selection was evaluated to ascertain if the application of selective agents at this level could increase selection efficiency. Two susceptible pistillate plants and one tolerant and one susceptible staminate plants were used in controlled crosses. Before pollination, a drop of a germination vehicle with TCF or without it was applied to the stigmas. Some pollinated pistils were fixed and analyzed by fluorescence microscopy; the rest were left on the plant for seed production. Fifty to 200 seeds were obtained per treatment combination (staminate plant x pistillate plant x pollination vehicle). The derived plantlets were inoculated in vitroand evaluated for disease symptoms. The application of TCF to stigmas reduced pollen germination and tube growth compared with untreated controls,regardless of the genotypic combination. Pollen germination and tube growth was poorer for the tolerant staminate genotype than for the susceptible one. When the TCF was applied, the number of seeds per pollination in comparison with the controls diminished only when the susceptible genotype was the pollinator. The percentage of affected root area of the progenies obtained after applying the TCF was lower than in the controls only when the tolerant genotype was the pollinator. Pineapple, Ananas comosus L. Merr., cultivars differing in resistance to fusariose were examined for the phytotoxic effect of culture nitrate (CF), and fusaric acid (FA) isolated from Fusarium subglutinans (Wollew & Reinking) Nelson, Toussoun & Marasas. The possibility of using these substances on F1 hybrids as selection agents for resistance was also evaluated. The phytotoxic effect of these substances was assessed in pineapple plantlets from tissue culture by placing them on wounded leaf segments. The resistance level of the F1 hybrids and cultivars under natural conditions was measured in greenhouse field tests. The susceptible cultivars proved to be the most sensitive in each test, whereas resistant cultivars showed resistance to CF. FA affected all the cultivars independently of the degree of resistance of the cultivars The presence of other metabolites in CF was responsible for the screening effect between the varieties. Using CF as selection criteria allowed the selection of resistant plants with behaviour quite similar to plants where the fungus itself was used as the selection agent.  相似文献

Two pineapple varieties differing in resistance to fusariose were examined for the phytotoxic effect of Fusarium subglutinans culture filtrate. The cultivars were Perolera (more resistant to pathovars of Fusarium subglutinans) and Smooth Cayenne (more susceptible). The phytotoxic effect of culture filtrate was assessed in tissue culture pineapple plantlets (by electrolyte leakage and placing the culture filtrate on wound leaf segments) and callus (inhibition of growth). Smooth Cayenne proved to be the most sensitive cultivar in each test, whereas Perolera showed resistance to the culture filtrate and its callus grew in the presence of high concentrations of culture filtrate that were completely toxic to Smooth Cayenne. Summary Potato plantlets derived from in vitro propagation of three cultivars known for their field resistance (Cruza 148 and BR-63.65) or susceptibility (Désirée) to Pseudomonas solanacearum E.F. Smith were artificially inoculated under controlled conditions. The aim of this work was to determine the optimal inoculum concentration and the best observation period in which the cultivars would show different reactions to bacterial infection as expected on the basis of their field performance.A suitable statistical analysis of disease indices is proposed to distinguish between resistant and susceptible responses, with a particular care for the applicative needs and a univocal interpretation of the results. In order to evaluate the significance of sources of variation related to the observed mean differences, the analysis of variance and a convenient clustering procedure of disease index means were applied.The statistical analysis revealed that, under our conditions, an inoculum concentration of 5×106 cfu/plant was suitable for separating resistant from susceptible responses, in accordance with the reactions already observed in field experiments by other authors. Also, differences among the three cultivars were best observed nine to twelve days after inoculation with the pathogen.  相似文献

Summary The main objective was to determine the reliability of selection in early generations of a potato breeding programme. To that end data were recorded in the seedling generation, an early and a late harvested experiment of the first clonal generation and the late harvested second clonal generation on up to 30 genotypes of 55 populations from crosses of three Solanum tuberosum ssp. tuberosum cultivars and seven selected S. tuberosum ssp. andigena clones, adapted to the northern temperate zone. Phenotypic correlations were computed per population in almost all possible combinations within and between characters between and within generations. The mean across 55 comparable correlation coefficients was considered to be the best estimate of the relationship concerned.In all four experiments data were collected on number of tubers, tuber yield and mean tuber weight; in the three experiments of the two clonal generations in addition on date of emergence, number of stems, plant height, haulm type, and general impression of the underground plant parts and in the two late harvested clonal generations on date of maturity and under water weight.The results confirmed that general impression which is largely decisive for the selection in early generations, is a little reliable measure. However, from the magnitude of the mean correlations between experiments within characters it must be concluded that selection for most of these separate characters is hardly more reliable. An exception was date of maturity for which a mean correlation was determined of r=0.78 between the two clonal generations. The second strongest mean correlations, perhaps surprisingly, were for tuber yield, viz. r=0.52 between the seedling generation and the late harvested first as well as the second clonal generation and r=0.63 between the late harvested clonal generations. From the correlations between characters it appeared that for a high tuber yield, besides its components, in particular an early emergence, a large plant height, an attractive haulm type and a late maturity are of importance.The main conclusion is that, as long as not enough seed tubers are available for the layout of statistically sound experiments, a negative instead of a positive selection should be applied to avoid the rejection of most of the valuable genotypes in an early stage. On the basis of the results directives for the application of such a negative selection are given.  相似文献

Fusarium basal plate rot (FBR), caused by Fusarium oxysporum f. sp. cepae, is an important soil-borne disease of onions worldwide. The causal organism infects the basal stem plate of the bulb and eventually kills the entire plant through degradation of the basal plate. F. o. f. sp.cepae infections in dormant bulbs during storage allow secondary infections to occur. The primary method of infection by F. o. f. sp. cepaeis through direct penetration of the basal stem plate. Infection can also occur through wounded tissue particularly roots and basal portions of bulb scales. The most cost-effective methods of control are crop rotation and host plant resistance. Current research suggests that a single gene, two genes, or multiple genes govern resistance to FBR. Breeding programs have successfully used screening procedures to develop intermediate- and long-day, FBR-resistant cultivars. Summary In two experiments, using different testing methods, the number of newly formed cysts was determined on nine potato genotypes with resistance from various sources. Ten potato cyst nematode (PCN) populations were used in these experiments. Rank correlation between numbers of cysts over potato genotype-PCN population combinations for both experiments was high (rs = 0.90). Dendrograms for PCN populations and potato genotypes were constructed, based on a simultaneous hierarchical clustering procedure for potato genotype-PCN population interaction terms. Several virulence groups could be identified within Globodera rostochiensis as well as within G. pallida. Host genotypes, derived from the same sources of resistance, were clustered in different resistance groups.  相似文献

Summary Fifty-five populations from crosses of three Solanum tuberosum ssp. tuberosum cultivars and seven selected S. tuberosum ssp. andigena clones adapted to the northern temperate zone have been studied in the seedling and the first and second clonal generation. This paper only deals with the effect of seed tuber weight on various important agronomic characters which are selected for in early clonal generations, independently of the type of crosses.The results showed that a distinction has to be made between the effects of differences in seed tuber weight within and between seedlings, i.e. genotypes. The effects on the first clonal generation of differences in weight of the tubers within seedlings and those of the first heaviest tubers between seedlings were similar and comparable to seed tuber weight effects within cultivars. In contrast to the within seedlings effects the between seedlings effects proved to be heritable. This is concluded from the fact that, on average, independent of seed tuber weight seedlings with larger first heaviest tubers performed better in the second clonal generation than those with smaller ones. Notably plant height, general impression, date of maturity and particularly tuber yield and mean tuber weight in the second clonal generation were related to the weight of the first heaviest tubers of the seedlings. The potentialities of these findings for an early generation selection are discussed.  相似文献

Summary Resistance toMeliodogyne chitwoodi races 1 (MC1) and 2 (MC2) andM. hapla (MH) derived fromSolanum bulbocastanum was introduced into the cultivated potato gene pool through somatic fusion. The initial F1 hybrids showed resistance to the three nematodes. Resistance to reproduction on roots by MC1 was accompanied by resistance to tuber damage in F1 clones. Tuber damage sometimes occurred, however, in hybrids of BC1 progeny resistant to reproduction on roots when MC2 and MH were the challenging nematodes. Resistance to reproduction was transferred into BC1 individuals, but a greater proportion of BC1 progeny was resistant to MC1 than to MC2 or MH. Resistance to MC1 appears to be dominant and discretely inherited. F1 and BC1 progeny were pollen sterile, but seed were produced from crosses using cultivated tetraploid pollen sources. Approximately 11 and 33 per cent of pollinations produced berries on F1 and BC1 pistillate parents, respectively. Late blight caused by Phytophthora infestans was monitored in field plots of potato genotypes selected from population A of the International Potato Center (CIP) germplasm collection. Disease severity was measured as percent blighted leaf area and used to compute area under disease progress curves (AUDPC), apparent infection rates (r) and severity at epidemic onset (Yo). AUDPCs revealed more distinct differences among the genotypes than any other disease assessment parameter. Percent disease severity measured 67–77 days after planting (D67-77)explained more variation in AUDPCs than measurements made on any other single day. Increase in percent diseased leaves fit the monomolecular model more closely than the Gompertz, logistic or exponential model. All disease assessment parameters varied among the genotypes and were significantly (p < 0.01) correlated with each other. Genotypes with larger AUDPCs generally had higher DS67-77 and faster rates of disease increase (r). Clones 386191.7 and381403.23 were more susceptible to late blight than all other entries tested. The lowest disease levels were observed on clone 382155.2. Frequency distribution of AUDPCs among genotypes appeared continuous and did not differ significantly (p <0.05)from normal distribution suggesting the observed resistance may be attributable to minor genes. A test to select Fusarium resistant seedlings of the Asiatic hybrid lily is described. Young seedlings of 28 populations, obtained from an incomplete diallel between eight parents with different levels of Fusarium resistance, were tested for resistance. Significant differences in Fusarium resistance between and within populations were detected. The average percentage of selected seedlings ranged from 34% in resistant × resistant crosses to 2% in susceptible × susceptible crosses. Although resistant descendants were obtained in susceptible × susceptible crosses, using at least one resistant parent produced higher percentages of resistant seedlings. The resistance level of the parents correlated highly with the general combining ability for Fusarium resistance based on the seedling test. For eight populations, seedlings selected for Fusarium resistance and non-tested (control) seedlings of the same cross were compared, after propagation, in a clonal test. Variation between and within populations, found at seedling level, was confirmed at clonal level. A positive selection response was found for all eight populations. In the seedling test, approximately 18% of the seedlings were selected as resistant of which 15% (2.7% of seedlings tested) appeared to be susceptible escapes. Comparison between selection at seedling level and at clonal level indicated that approximately 25% of the seedlings tested were missed (rejected resistant plants) in the seedling test. The practical use of a seedling test for Fusarium resistance in lily breeding programs is discussed.  相似文献

Summary Resistance to fusarium wilt, incited by Fusarium oxysporum (Schlecht.) f. sp. lycopersici (Sacc.) Snyder & Hansen race 3 in tomato (Lycopersicon esculentum Mill.) was discovered in LA 716, a L. pennellii accession. A resistant BC1F3 breeding line, E427, was developed from LA 716. E427 was crossed with the susceptible cv. Suncoast and F1, BCP1, BCP2 (to Fla 7155, a susceptible parent) F2, F3, and BCP2S1 seeds were obtained. Segregation for resistance following root dip inoculation over three experiments indicated a single dominant gene controlled resistance. Five of the 12 BCP1S1's segregated more susceptible plants, whereas one of the 12 segregated more resistant plants than expected (P<0.05). Three of 23 F3 lines segregated more susceptible plants than expected while 1 of the 23 had more resistant plants than expected (P<0.05). Segregation in all other lines fit expected ratios. Five of the 23 F3's were homozygous resistant which was an acceptable fit to expectations (P=0.1–0.5). The gene symbol I 3 is proposed for resistance to race 3 of the wilt pathogen. Deviations from expected ratios in data reported here and for other breeding lines indicate an effect of modifier genes and/or incomplete penetrance. Stem decay and root rot of Amaranthus hybridus, caused by Fusarium oxysporum is a serious threat to the commercial production of this crop in South Africa. Five Amaranthus hybridus varieties were examined in vitro for sensitivity to a culture filtrate of Fusarium oxysporum. The phytotoxicity of the culture filtrate was assessed for its inhibitory effect on callus and seeding root growth, as well as on the viability of callus cells. The five varieties exhibited a significant amount of variation in response to the culture filtrate of the pathogen. Variety 17 was the most sensitive variety in each bioassay, whereas variety 20 displayed least sensitivity to the culture filtrate. Callus of variety 20 grew well in the presence of concentrations of culture filtrate that were toxic to another four varieties and the percentage mortality of callus cells after exposure to the filtrate was also the lowest of the five varieties. Root growth of variety 20 was also least affected by exposure to the culture filtrate for up to 6 days. To our knowledge, the presence of resistance in germplasm of A. hybridus to culture filtrates of F. oxysporum has not been demonstrated before. The present study is therefore of significant value to breeding programs aimed at speedily finding amaranth cultivars that are resistant to this important pathogen. Summary Sixty-two 2x families were generated by intermating 16, 2x clones and evaluated for resistance to potato tuber moth (PTM), Phthorimaea operculella under natural infestation in a storage at San Ramon, Peru and in laboratory tests. The following conclusions could be drawn: (1) relatively simple inheritance was observed for resistance derived from Solanum sparsipilum (spl), (2) the high level of resistance of the original spl has been transferred, undiminished, into an advanced 2x population, (3) simple phenotypic selection was successfully applied to transfer resistance into an improved 2x population, (4) there was a strong indication of reciprocal effects, however spl cytoplasm is not essential for the expression of nuclear resistance genes, (5) antibiosis and antixenosis are the mechanism of PTM resistance in this population, and (6) 4x × 2x crosses could be used to transfer the resistance into commercial cultivars.  相似文献

Summary The production of dihaploids is the first step in a potato breeding program at the diploid level. Dihaploid induction ability, dihaploid production ability and seed parent x pollinator interaction were analyzed for 28 × 3, seed parent x pollinator combinations. This is the first report on significant interaction between seed parents and pollinators. Despite this interaction, IVP 101 had a significantly higher dihaploid induction ability than the widely used pollinators IVP 35 and IVP 48. Previous findings on significant differences in dihaploid production ability between seed parents were clearly confirmed.  相似文献
