Porcine circovirus type 1 (PCV1) is considered to be a non-pathogenic virus detected in cell cultures, vaccines or products used for cell culture preparations, all of them of porcine origin. Serological evidence and genetic studies suggested that PCV1 was widespread in domestic pigs. The presence of PCV1 in wild boars in Germany was also described using serological methods. This paper reports the first detection of PCV1 in Hungarian wild boars. Samples were collected at slaughterhouses and processed for polymerase chain reactions. The complete genome of PCV1 detected in the samples was determined and compared with the available PCV1 sequences of the GenBank database. The genomes formed two distinct clusters with minimum differences, where the Hungarian wild boar PCV1 (WB-H8) grouped together with genomes originating from domestic swine from China and Australia and with a genome detected in a porcine pepsin product.  相似文献

To assess the performance of boars derived by somatic cell cloning, we analyzed various aspects of their reproductive characteristics and the expression of two imprinted genes. Cloned boars (cloned Duroc × Jinhua) were analyzed for birth weight, growth rate, age at first ejaculation, semen characteristics and fertility, in comparison with naturally bred control boars of the same strain. The expression of imprinted genes was analyzed using the microsatellite marker SWC9 for the paternally expressed gene insulin‐like growth factor ‐2 (IGF2) and with single nucleotide polymorphisms (SNPs) for the gene maternally expressed 3 (MEG3). The cloned boars had high production of semen and were nearly equal in level of fertility to conventional pigs; they showed similar characteristics as naturally bred boars of the same strains. The expression of IGF2 was partially disturbed, but this disturbed expression was not linked to a change in developmental fate or reproductive performance. These results indicate that use of cloned boars could be highly effective for proliferation of pigs with desirable characteristics, preservation of genetic resources and risk reduction against epidemic diseases, such as foot‐and‐mouth disease, through storage of somatic cells as a precautionary measure for use in regenerating pig populations after a future pandemic.  相似文献

Only a very small amount of physiological data is available about the low fertility (mean litter size is 5.7+/-0.8) of Hungarian native breed, Mangalica (M), sows. The aim of the present paper is to reveal the differences in preovulatory follicle development and intrafollicular oocyte maturation between M and Landrace (L) gilts, with special reference to the peri- and postovulatory secretion and peripheral concentrations of estradiol-17beta (E2), progesterone (P4), and luteinizing hormone (LH). The number of preovulatory follicles was 6.8+/-1.4 and 19.6+/-6.6 in M and L gilts, respectively. A lower degree of cumulus expansion and a lower percentage of mature oocytes (TI/M II) was noted in M. Higher LH and E2 peak levels, a longer E2 to LH peak interval, and lower embryo survival was confirmed. Interestingly, despite the lower number of corpora lutea, a higher peripheral blood level of P4 was shown in M than in L gilts. Both diminished follicular development and protracted oocyte maturation may be involved in low fecundity in M, and the present findings may explain these reproductive phenomena.  相似文献

Thirty-three boars with sexual dysfunctions and twenty-six boars clinically sound as to their reproductive capacity were evaluated for the testosterone levels in the blood plasma before i.v. administration of 500 i.u. of chorionic gonadotropin and two hours after the administration. A group of animals with reproduction disorders comprised boars with an impaired quality of ejaculate and low fertility ability (18 boars) and with sexual dysfunctions (15 boars). No statistically significant difference in the basal concentration of testosterone in the blood was found in the boars with the studied sexual dysfunctions, as compared with the boars with no sexual dysfunctions. Administration of chorionic gonadotropin increased significantly the plasma testosterone levels in both groups. If the effect of chorionic gonadotropin on the studied level of this hormone was compared in boars with sexual dysfunctions and in boars without any disorders, no significant differences were proved. It has been inferred from the above findings that there are no significant disorders of androgen supply and incretion reserve of the gonads in the boars with sexual dysfunctions.  相似文献

Nine blood samples were taken at 30-min intervals from 36 Landrace X Large White boars at each of eight ages (42, 56, 70, 84, 98, 112, 126 and 140 d). Serum concentrations of luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone (T) and estradiol-17 beta (E2) were quantified by radioimmunoassay procedures. The maximum concentration of LH and the age at maximum concentration were predicted for each boar. Variability of LH samples was described for each boar by the pooled within age variance among LH samples and by the number of LH peaks. Measurements of testicular development taken at 140 d of age included: in situ testis width and length, excised testis weights and histological traits of excised testes (seminiferous tubule diameter, percentage of tubules with a lumen and percentage of tubules with active spermatogenesis). Pooled within line correlations were calculated with data from boars selected for either high or low testis weight. Correlations among the testicular traits ranged from .45 to .88. Luteinizing hormone concentration (mean over all ages) was related to measures of testicular development (r = .24 to .49). Concentrations of LH from 42 to 84 d of age were more highly correlated with testicular traits than were the concentrations from 98 to 140 d.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Eleven sexually mature boars and 10 peripubertal boars were used to study the effects of elevated testicular temperature on plasma hormonal levels. The scrotum of the boars was covered with a textile-aluminium foil insulation device for 100 h. Insulation of the scrotum in the peripubertal boars took place at an age of 100 days. Blood samples were drawn 3 times daily for 12 days in the mature boars, starting 3 days before scrotal insulation. In the peripubertal boars, blood sampling was performed once a day for 11 days, starting the first day of scrotal insulation. During scrotal insulation, the plasma levels of testosterone, oestradiol-17 beta and oestrone sulphate decreased continuously in the mature boars. After removal of the scrotal insulation device there was a continuously increase, back to normal levels of oestradiol-17 beta and oestrone sulphate. The plasma levels of testosterone showed an immediate rise of brief duration after removal of the device in 5 of the boars, while in the other 6 boars the rise in testosterone levels came 4 days after removal and lasted for 3 days. In the peripubertal boars, there were no significant differences in the hormone levels between the experimental and control animals during and after scrotal insulation. However, the decrease in testosterone concentration over time, during scrotal insulation, was significant within the experimental group.  相似文献

The objectives of the current experiment were to determine whether boars heterozygous for the mutation in skeletal ryanodine receptors (sRyR), known to cause porcine stress syndrome, differed from wild-type boars in hypothalamic-pituitary-adrenal axis (HPA) function. We have examined basal plasma ACTH, cortisol, and corticosteroid-binding globulin (CBG) concentrations; plasma ACTH and cortisol responses to a nose-snare stressor and at slaughter; dexamethasone suppression of plasma ACTH and cortisol concentrations; and glucocorticoid receptor (GR) density in the pituitary gland, hippocampus, hypothalamus, and frontal cortex. We have also examined carcass yields, composition, and meat quality to determine whether differences in HPA activity were accompanied by an increased incidence of meat quality characteristics associated with pale, soft, exudative (PSE) meat. Thirty boars either heterozygous or wild-type (n = 15 per genotype) for mutated sRyR were tested for HPA function at 7 mo of age. Heterozygous boars had lower basal plasma ACTH (P < .05) and cortisol (P < .04) concentrations. Integrated basal plasma ACTH and cortisol levels were also lower (P < .05 and P < .005, respectively). Genotype had no significant effect on basal CBG, stressor-induced (nose snare or slaughter) or dexamethasone suppression of plasma ACTH or cortisol concentrations. No differences in immunoreactive GR levels were found in the pituitary gland or any brain region examined. We did find a significant, negative correlation (r = -.62, P < .02) between peak (0800) basal plasma ACTH concentrations and hippocampal GR levels. The alterations in basal HPA function in heterozygous boars were accompanied by lighter body weights (P < .03), decreased carcass fat depth (P < .04), and increased carcass lean yields (P < .02). There was a higher incidence of meat quality characteristics associated with PSE meat in heterozygous boars indicated by higher carcass temperatures (P < .04) and meat brightness (P < .0001) with lower carcass pH at slaughter (P < .03) and after chilling (P < .003). In conclusion, we have found differences in basal and not stressor-induced HPA function between boars heterozygous and wild-type for mutated sRyR. This altered basal HPA activity was accompanied by an increased incidence of meat quality aspects associated with PSE meat in heterozygous boars.  相似文献

Vitrification of in vitro matured oocytes of Mangalica and Large White pigs   总被引:1,自引:0,他引:1  
The breeding of Mangalica, a native pig breed in Hungary, had been started in 1833, but this pig breed almost became extinct in Hungary in the past decades. In 1991, the number of sows was only 200. Although in these days the existing Mangalica population consists of more than 6000 animals representing different colour variations, the preservation of this traditional pig breed is still very important. Vitrification is a potential tool for the preservation of gametes and embryos of these animals. The aim of this study was to investigate the effects of vitrification on the developmental competence of Mangalica (M) and Large White (LW) oocytes following fertilisation. The oocytes were vitrified by the Open Pulled Straw (OPS) method using different concentrations of ethylene glycol and dimethyl sulphoxide as cryoprotectants. After rehydration the oocytes underwent in vitro fertilisation; the resultant zygotes were then cultured in vitro for four days to assess embryonic development. In the first experiment, in vitro maturation of M and LW oocytes was compared. No significant difference was observed in the nuclear maturation rate of LW and M oocytes. In the second experiment, the sensitivity of oocytes to vitrification was examined by evaluating oocyte morphology after thawing. A higher percentage of LW oocytes showed normal morphology compared to M oocytes, indicating that Mangalica oocytes are more sensitive to cryoprotectants than Large White oocytes. After warming and in vitro fertilisation, more than 50% of the oocytes started embryonic development and by the end of the incubation period morula stage embryos had developed in both groups. The results show that the OPS vitrification technique is well suited to preserve Mangalica oocytes and from these oocytes morula embryos can be produced.  相似文献

Fifty-eight boars of the “German Landrace” breed were fattened between 70 and 110 kg in individual housing at ambient temperatures of 15°C or 35°C with 60% relative humidity in both cases.The animals under warm conditions showed increased growth of the skeleton and reduced growth of the heart, liver, kidneys, spleen and stomach.The feed intake was strongly restricted by the high temperature, while water consumption showed an extremely high variance. Correspondingly the daily gains were strongly depressed, although the feed conversion ratios of the warm- and cold-housed animals were not significantly different. The fat thickness of the warm-housed animals was significantly lower.  相似文献

Effects of prolactin receptor genotype on the litter size of Mangalica   总被引:1,自引:0,他引:1  
The aim of this study was to detect different alleles of the prolactin receptor (PRLR) gene and to examine their effects on the litter size of the indigenous Hungarian pig, the Mangalica. G1789A single nucleotide polymorphism (SNP) was investigated as a candidate for litter size. Samples from 80 purebred Mangalica sows and data of their 335 litters were provided by Olmos & Tóth Ltd. Hair follicles were used to isolate the required DNA. Allelic discrimination was performed by means of the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method using the AluI restriction enzyme and agarose gel electrophoresis. In the population examined, the A allele was found to be preferable in the Mangalica breed group. The most advantageous AA genotype was the least prevalent (8.75%), while the frequencies of AB and BB were 40% and 51.25%, respectively. Remarkably, the average number of piglets born alive per litter was 1.11 ± 0.39 higher in sows with AA as compared to those with BB genotype. By raising the frequency of the AA genotype, the litter size is likely to increase. However, the effect of PRLR genotypes can differ among pig breeds and even lines. Further studies may be required to observe and estimate possible pleiotropic effects of this polymorphism on other traits.  相似文献

Crossbred boars were used to evaluate the influence of exposure to 8 or 16 hr of light daily from 75 to 175 days of age on growth rate, testicular characteristics and endocrine function. At 160 days of age, concentrations of testosterone in serum (P<.10), the areas under plotted 12 hr testosterone profiles (P<.10) and the number (P<.05) and magnitude (P<.10) of testosterone secretory spikes were increased in boars exposed to 16 hr of light compared to boars in 8 hr light, but concentrations of LH in serum were similar in boars exposed to both treatments. Treatment with GnRH resulted in similar concentrations of LH in serum for both groups of boars. Testosterone in serum after GnRH-mediated LH release was greater at .5 (P<.05) and 1.0 (P<.10) hr following GnRH in boars exposed to 16 hr of light compared to boars at 8 hr, but concentrations of testosterone were similar for both treatments from 1.5 to 4.0 hr after GnRH. Growth rate and testicular and epididymal weights and sperm reserves at 175 days of age were not significantly altered by duration of photoperiod. Boars exposed to 8 hr of light had more hair per unit area than boars exposed to 16 hr of light. We conclude that exposure of prepubertal boars to longer daily photoperiods results in increased concentrations of testosterone in serum at 160 days of age.  相似文献

Investigations were made to identify the causal agent of an acute outbreak of abortions in a domesticated herd of wild boar. Only porcine parvovirus (PPV) was isolated from samples of organs from the still-born sucklings and mummified aborted fetuses. The isolated virus hemagglutinated erythrocytes of guinea pig, murine, rat, and chicken. Identity of the virus, designated the BQ strain, was confirmed by the production of a specific cytopathic effect on susceptible cells and by the results from ELISA, PCR, immunofluorescence assay, and electron microscopy. PPV BQ strain was adapted to growth in a swine testicular cell line. When inoculated into healthy sows, PPV BQ caused the same reproductive disorder observed in the affected herd.  相似文献
