Summary Eleven clones obtained from a cross between cv. Katahdin and fusion products betweenSolanum tuberosum and the non tuber-bearing speciesS. brevidens, were backcrossed again withS. tuberosum (clone AR80-127-5). Small tubers harvested from 583 seedlings of these second backcross (BC2) populations were screened for tuber tissue resistance toErwinia carotovora subsp.atroseptica under aerobic conditions. After multiplication in the field. BC2 clones again were screened for soft rot resistance under aerobic as well as anaerobic conditions. In general, the resistance of the BC2 populations was reduced in comparison with the BC1. Variation for resistance was found within the BC2 populations, but there was no correlation between the results of the three tests performed. Four BC2 populations were also screenned for resistance to blackleg in the field. Significant differences were found between populations for mean percentage of diseased plants, but these differences could not be explained by the resistance of the parental clones.  相似文献

Summary Expression of the pectate lyase (PL) isoenzyme 3 in transgenic potato lines of cv. Désirée mediates an enhanced resistance of tuber tissue toErwinia carotovora (Ec) soft rot due to a pre-activation of plant defence mechanisms. Therefore, theSolanum tuberosum cvs Agave and Adretta with a moderate level of soft rot resistance were crossed with such PL-expressing potato lines. The resulting progenies were assessed with respect to plant/tuber characteristics over a period of four years and then tested for PL3-expression as well as for soft rot resistance. 71% of the selected progeny lines exhibited a stable production of the PL3 enzyme. Statistical analysis revealed differences between the transgenic and the non-transgenic progeny concerning the soft rot resistance of tuber tissue. Compared with the PL-inactive progeny, extension of Ec-rotting on the wound surface of PL-transgenic potatoes was diminished on average by 51.8%. Similarly, the degree of cell lysis caused by bacterial maceration was significantly (P<0.05) reduced in tuber tissue of PL-expressing progeny lines. The latter also revealed an enhanced PPO and PAL activity in their tuber tissue indicating an active plant defence. It is concluded therefore that the PL-mediated soft rot resistance introduced into potatoes by means of molecular techniques is heritable.  相似文献

Summary Potato tubers were inoculated with two biotypes ofPhytophthora infestans then stored at 3,7, 10 and 15°C. Image analysis quantified average reflective intensity (ARI) of diseased tissue from cut surfaces of sample tubers. Tuber tissue infection and infection rate were measured by calculating Mean ARI of samples. Average tuber tissue infection and infection rate was minimal at 3°C (P.i.-US8 orP.i.-US1). Tuber tissue infection increased at temperatures >3°C, from 220 Mean ARI seven days after inoculation (dai) to 190–150 Mean ARI 50 dai (depending on cultivar and biotype ofP. infestans). Rate of tuber tissue infection caused byP.i.-US1 at 7°C was about zero in cv. Snowden but greater than −0.2 ARI day−1 (cvs Russet Burbank and Superior). Rate of late blight infection in tuber tissue generally increased with temperature from −0.2 ARI day−1 (at 7°C) in all cultivars to a maximum of −0.8 ARI day−1 (10°C).  相似文献

Summary Various methods of screening for resistance to root-knot nematodes were compared and evaluated. Seedling populations ofSolanum spp., grown in clay pots and plastic tubes with silver sand and inoculated with juveniles ofMeloidogyne chitwoodi andM. hapla, showed large differences in the number of egg masses on roots 7 weeks after inoculation. The differences were reproducible when re-testing was done with cuttings and plants from tubers. No resistance toMeloidogyne spp. was observed with ten potato cultivars when grown in clay pots, plastic tobes or closed containers. Plants from tubers in growth pouches developed a large two-dimensional root system, and after inoculation with juveniles the infection process could be observed over 8 weeks. A method of infecting potato tuber tissue withMeloidogyne is described, using tuber slices in Petri dishes as a potential screening test for tuber resistance.  相似文献

The influence of tuber-borne pathogenicStreptomyces on potato common scab incidence in daughter tubers was examined under micro-plot conditions. Visually healthy tubers, surface-disinfested healthy tubers, and tubers with 25% scab coverage were planted in pasteurized soil. At 30, 57, and 93 days after planting, soil and belowground plant parts were sampled from various zones around the mother tuber. The total population of actinomyctes was determined by plating on a semiselective medium, and colonies of pathogenicStreptomyces were identified using polymerase chain reaction (PCR) targeting thetxtA gene. At day 30, pathogenicStreptomyces populations were below detectable levels, whereas by day 93, measurable levels were present on the belowground plant parts and in soil adjacent to scabby mother tubers at 104 to 105 CFU g?1 soil. Incidence of scab in the progeny tubers was 89% at harvest. Progeny tubers produced by visually healthy seed tubers had an incidence of 60% scab and substantial populations of pathogenicStreptomyces were present in the zone near the mother tuber (104 CFU g?1 soil). Plots planted with surface disinfested seed tubers had a very lowStreptomyces population in the tuber zone and yielded 100% marketable tubers. A perfect correlation (r = 1.00) was found between the population densities of pathogenicStreptomyces in the root zone and daughter tuber disease incidence indicating that measurement of such populations in the field can serve as an excellent predictor of scab disease. After harvest, populations of pathogenicStreptomyces (106 to 107 CFU g?1 lesion tissue) remained high on the mother tubers.  相似文献

Enrichment techniques and serological strain typing methods were used to determine the source of stem and progeny tuber populations ofErwinia carotovora (Ec) in 1980 and 1981. Plots were established in a field that had been planted with potatoes since 1972. Seed tubers of cultivars Russet Burbank and Norgold Russet were assayed forErwinia carotovora pv.carotovora (Ecc) andErwinia carotovora pv.atroseptica (Eca) by tuber peeling and direct plating and plating following enrichment in a pectate-based broth. Levels of seed tuber infestation ranged from undetectable to 2.4 × 104 colony-forming units per g peel tissue. Prior to planting Ecc was detected in soil samples from field plots by enrichment in a pectate-based broth. Ec was isolated from symptomless plants with increasing frequencies from 8 weeks following planting regardless of the level of Ec populations in seed tubers. At harvest progeny tuber recontamination was low and not related to levels of seed tuber Ec populations. In 1980, serological typing of representative Ecc isolates indicated serogroups III, XXIX, and unknown strains isolated from the soil but not the seed tubers prior to planting were isolated from stems and progeny tubers. Serogroup XXIX was isolated from soil, seed, stems and progeny tubers in 1981. Throughout both years, Ecc was isolated more frequently than Eca from seed, soil, stems, and progeny tubers. These data emphasize the potential for reinfestation of seed stocks and infection of plants by populations of Ecc that overwinter in soil under Wisconsin conditions.  相似文献

Summary Calcium levels in the periderm of tubers (cv. Sebago) growing in irrigated sands were influenced by the stem position at which the tuber grew, but the effect was not consistent over two seasons. Calcium levels were increased at all tuber positions by addition of calcium to soil; combined treatments of gypsum (1400 kg Ca2+/ha) pre-plant and calcium nitrate (37 kg Ca2+/ha) side dressings gave the greatest response. Percentage incidence of soft rot lesions caused byErwinia chrysanthemi pv.zeae was not influenced by periderm calcium content. The severity of infection (weight of tissue macerated within 48 h at 30°C) was significantly reduced only by the pre-plant gypsum treatment, but there was no correlation with calcium content of the periderm. Calcium levels in periderm tissue (60–208 mg Ca2+/100g) are within the range of published data for other cultivars and results support previous reports that the correlation between tuber susceptibility toErwinia spp. and periderm calcium content is inconsistent.  相似文献

Summary Tubers of clones from a diploid hybrid population ofSolanum phureja andS. stenotomum were screened for resistance to tuber soft rot caused by strains ofErwinia carotovora subsp.atroseptica (Eca),E. carotovora subsp.carotovora (Ecc) andE. chrysanthemi (Ech). Significant, positive correlations between resistance to the different strains and species were observed, indicating that screening for resistance to tuber soft rot could be accomplished by using just one of the three pathogens. Strains of Ecc and Ech were found to be much more virulent than strains of Eca. Inoculating with Ecc and Ech resulted in more distinct differences among susceptible, moderately resistant and resistant clones than inoculating with Eca.  相似文献

Nine experiments were run at three hot tropical sites (5–12°S, 180–800 m) within Peru to quantify the influence of plant population on soil temperature and growth and yield of the potato.Radiation interception was greatest at the highest plant populations and soil cooling was directly proportional to the amount of crop cover over the soil, but no appreciable effect on the timing of tuber initiation was apparent. More stems per unit land area leading to a higher leaf area index (lai) were primarily responsible for greater interception of radiation at the higher plant populations, although some compensation in stem number per unit land area and in lai at lower populations was evident later in the season.In general, tuber yield increased linearly with increases in planted population over the range studied (2.7–12.5 plants m−2, and in one experiment to 31.7 plants m−2), and was proportional to increases in the amount of intercepted radiation. Tuber yields ranged from 8 to 60 t ha−1 over sites and populations. Vigorous clones with Solanum tuberosum spp. andigena in their genetic background constituted the exceptions to this linear trend, and for these clones yields declined at the highest populations, particularly when the rectangularity of planting vastly deviated from square patterns. Tuber yield of Solanum tuberosum spp. tuberosum and Neotuberosum (S. tuberosum spp. andigena selected for tuberization under long-day conditions) clones did not respond to variations in rectangularity of planting and, probably due to their small stature and early maturity, did not demonstrate signs of intense between-plant competition for tuber yield as measured with the Kira competition density index. In contrast, for clones with Solanum tuberosum ssp. andigena in their genetic background, maximum tuber yield at populations greater than 5.5 plants m−2 was dependent on the rectangularity of planting, and declined as the latter deviated from squareness.Since the proportion of marketable tubers was scarcely affected by the planting densities, plant population of S. tuberosum ssp. tuberosum clones planted in hot climates should be as close as possible without limiting the amount of soil available for hilling-up.  相似文献

新疆伊犁地区是小麦条锈病的常发区和重发区,其年平均发生面积超过100×104 hm2以上。为明确新疆伊犁地区小麦条锈菌群体的遗传结构,2021年,从新疆伊犁地区的新源县、巩留县、察布查尔县和伊宁县共采集到320份小麦条锈菌标样,利用16对小麦条锈菌SSR引物进行了群体遗传研究。结果表明,新疆伊犁地区条锈菌群体遗传多样性水平较高,群体间存在一定的遗传分化。群体间遗传变异仅占总变异的12%,群体内遗传变异占88%,不同群体间存在频繁的菌源交流,其中察布查尔县与新源县群体菌源交流最为频繁(Nm=4.717)。  相似文献

Since the environment of the Mediterranean regions allows offseason production, potatoes are planted in autumn-winter and harvested in spring. During this period, potatoes are subjected to low temperatures and short day lengths which modify the growth characteristics of plants. For this reason, our analysis of competition response was conducted to better clarify the biological relationship between yield and plant density. Field trials were conducted in Sicily (south Italy), a highly representative area of early potato crop in the Mediterranean Basin, with the aim of studying effects of intraspecific competition on tuber yield and yield components. Ten planting densities (ranging from 3.0 to 8.0 plants m?2) were studied on cv. Spunta using “tuberpieces” with a different number of eyes (one eye or all the eyes in 1996; one eye, two eyes, or all the eyes in 1997). Intraspecific competition reduced the tuber yield of individual plants, which became gradually less evident with increasing plant density. Competition affected the number of tubers per plant in the lower plant populations only (from 3.0 to 5.8 plants m?2), whereas effects on average tuber weight were at times more marked in the higher populations (from 5.8 to 8.0 plants m?2) and at other times in the lower densities (from 3.0 to 5.8 plants m?2). As a consequence of increased plant density, and notwithstanding the higher intraspecific competition, the yield of tubers per unit area increased linearly. Regardless of the number of eyes per tuber-piece, when passing from the lower to the higher plant density, yield increased from 34.0 to 54.11 ha?1 in 1996 and from 39.9 to 56.7 t ha?1 in 1997.  相似文献

Summary Potato stems and tubers grown in the field from seed tubers inoculated withClavibacter michiganensis subsp.sepedonicus, which causes bacterial ring rot, were tested by indirect, sandwich enzyme-linked immunosorbent assay (ELISA) in five laboratories. Correlation between values for each experimental treatment from the five laboratories was greater (r=0.86) than correlation between values for individual samples (r=0.71). When three or more laboratories obtained ELISA values of ≥0.200 for a sample, that sample was presumed to be positive. Conversely, when three or more laboratories obtained ELISA values <0.200, the consensus determination was regarded as negative. The percentage of stem and tuber samples that were in agreement with the consensus ELISA determination varied from 65.5 to 96.7%. Indirect immunofluorescence tests, conducted on the same samples in two laboratories, were consistent with 83.4–91.9% of the consensus ELISA determinations. Presence or absence ofC.m. sepedonicus was confirmed in some samples by an eggplant bioassay and direct isolatiion of the bacterium. The ELISA procedure was well suited for screening large numbers of samples and this study confirms it to be a promising procedure in routine indexing of seed potatoes forC.m. sepedonicus.  相似文献

Summary Four potato lines of cv. Désirée that express the pectate lyase (PL) isoenzyme 3 ofErwinia carotovora subsp.atroseptica were examined in a 4-year field experiment with respect to plant development, tuber yield and resistance of tuber tissue toErwinia soft rot. The PL3 degrades plant cell wall pectin into unsaturated oligogalacturonates eliciting plant defence responses. In one line, enzyme expression was controlled by the CaMV 35S promoter (C) and in three lines it was driven by the potato patatin B33 promoter (D). Plant development of the D-lines in field plots was not distinguishable from that of the non-transgenic counterpart. Also tuber yield was not too different. By contrast, plants of the C-line were smaller than those of the nontransformed counterpart and also showed reduced tuber yield. There were no significant differences in dry mass, starch and protein content of tuber tissue between PL transgenic and non-transgenic potatoes. But compared with the latter, field-grown tubers expressing the PL displayed an enhanced resistance toErwinia soft rot. Thus, average rotting caused byEc-bacteria was diminished in tubers of PL-transgenic lines by 34.1%. The resistance of tubers toEc soft rot was significantly correlated with the PPO activity in tuber tissue.  相似文献

Summary The use of crop simulation models to predict yield, associated with decision support systems such as Decision Support System for Agrotechnology Transfer (DSSAT), are useful tools to test different management strategies. The potato growth model included in DSSAT is SUBSTOR-potato. To evaluate its performance in Argentina it was calibrated and validated using experimental results from different sites and years. Cultivar-specific coefficients were obtained during calibration. Validation based on several independent sets of field data, including cvs Huinkul, Kennebec, Mailén and Spunta showed good agreement (R2=0.915; n=24) between observed and simulated values in normal ranges of tuber yields. However, when the input parameter maturity date was not taken into account, tuber yields were overvalued due to an overestimation of LAI values during maturation. To solve this problem, a genetic coefficient for the duration of tuber filling needs to be included in the model.  相似文献

Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.  相似文献

Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.  相似文献

Summary Photosynthetic characteristics, carbon allocation and partitioning of newly-fixed14C among the various carbohydrate fractions were studied in leaves of potato plants (Solanum tuberosum L. cv. Désirée) of different ages. The higher photosynthetic rate of young leaves was associated with higher levels of starch and reducing sugars. After 24 hours, in young leaves the fixed14C had been partitioned mainly to the residue and starch fractions, while in older leaves there was an increase in partitioning to sucrose and reducing sugars. The activities of the various sink organs were found to be dependent upon their distance from the source leaf. Tuber activity, however, was affected similarly by all fully expanded leaves. Although young fully expanded leaves are thought to be the major source of assimilates for tuber growth, it seems that a vast potential for contributing to plant productivity and tuber yield lies also in the older leaves, which translocate most of their photoassimilates to the tubers.  相似文献

Summary Plants were grown in the field from seed pieces of potato cultivars injected with ring rot bacteria (Corynebacterium sepedonicum). Symptoms were produced with both 18 and 180 colony forming units (cfu) on cvs Norchip and Red Pontiac and with 180 cfu on cv. Belrus plants. Tuber symptoms were detected in all cultivars except Belrus and Teton. Tuber progeny produced plants that developed top symptoms on all cultivars except those of cv. Nooksack, Russet Burbank, and Teton. Tuber progeny of the second crop produced both plants and tubers with symptoms developing only in the cv. Nooksack. In another 3-year experiment, variability in the disease response of selected cultivars and lines was examined following knife-inoculation of tuber seed with a high level of ring rot bacteria. Significant correlations between top and tuber symptoms were detected, but they were not high enough to make unnecessary the examination of both top and tuber symptoms in ring rot disease selection studies. Summary Offspring were produced from a cross between two long-day-adaptedSolanum phureja clones which carried resistance to tuber soft rot (Erwinia carotovora subsp.atroseptica). In tests carried out on the produce of field-grown plants raised from tubers, over fifty per cent of the 173 offspring were found to be highly resistant. Assessments were also carried out of tuber yield, mean tuber weight, tuber number, shape, regularity, flesh colour, texture of the steamed flesh, fry colour, after-cooking blackening, sprout length after storage and overall dormancy. There were statistically significant differences between clones for all characters (P<0.001). Twelve of the clones were selected on the basis of high resistance, yield, tuber weight, regularity of shape and absence of after-cooking blackening. The value of resistant long-day-adapted diploid material for commercial breeding is discussed.  相似文献
