Recently, there has been interest in programs that certify pork production practices that minimize the risk of exposure of pigs to Trichinella spiralis. Certification might be useful for reducing the risk of human trichinellosis from pork in Argentina, but more information is needed on pig production practices and sources of Trichinella infection in Argentinian pigs. In this study, 21 pig farms were assessed for Trichinella infection including some farms using total and partial confinement management, and others with pigs raised exclusively outdoors. A total of 3224 muscle samples were collected from pigs raised on these farms and tested to determine the presence of T. spiralis larvae by artificial digestion. Serum samples from the same 3224 pigs were tested for antibodies to T. spiralis by ELISA. For each farm, a questionnaire was completed summarizing information about management factors and this information was used to assess risk factors for exposure of T. spiralis. Based on the results, pigs raised outdoors were more likely to be infected than pigs raised in total or partial confinement (p  0.05). Pigs fed waste products containing meat were 12.5 times more likely to be infected than pigs not fed waste containing meat (p < 0.01). The role played by rats in transmission of Trichinella is unclear; however, on farms with evidence of wild animals and access of pigs to wildlife carcasses, the prevalence of Trichinella infection was significantly higher. All pigs raised under good hygienic and sanitary conditions were negative for Trichinella infection by both artificial digestion and ELISA.  相似文献

In Argentina, there are three known species of genus Trichinella; however, Trichinella spiralis is most commonly associated with domestic pigs and it is recognized as the main cause of human trichinellosis by the consumption of products made with raw or insufficiently cooked pork meat. In some areas of Argentina, this disease is endemic and it is thus necessary to develop a more effective programme of prevention and control. Here, we developed a quantitative risk assessment of human trichinellosis following pork meat sausage consumption, which may be used to identify the stages with greater impact on the probability of acquiring the disease. The quantitative model was designed to describe the conditions in which the meat is produced, processed, transported, stored, sold and consumed in Argentina. The model predicted a risk of human trichinellosis of 4.88 × 10?6 and an estimated annual number of trichinellosis cases of 109. The risk of human trichinellosis was sensitive to the number of Trichinella larvae that effectively survived the storage period (r = 0.89), the average probability of infection (PPinf) (r = 0.44) and the storage time (Storage) (r = 0.08). This model allowed assessing the impact of different factors influencing the risk of acquiring trichinellosis. The model may thus help to select possible strategies to reduce the risk in the chain of by‐products of pork production.  相似文献

No systematic studies of the occurrence of Trichinella in wildlife have been carried out in Northern Ireland (NI) in recent years, and the last reports of trichinellosis in livestock and human outbreaks in NI date back to 1979 and 1945, respectively. In this study, covering the period 2003/2004 and 2007/2008, a total of 443 red foxes (Vulpes vulpes) were collected throughout the country and screened for trichinellosis using a modified muscle digest method. One examined animal was found to be infected with larvae from Trichinella spiralis, indicating a national prevalence in NI of Trichinella in foxes of 0.2%. This prevalence compares well to the findings reported from the bordering Republic of Ireland [Rafter, P., Marucci, G., Brangan, P., Pozio, E., 2005. Rediscovery of Trichinella spiralis in red foxes (Vulpes vulpes) in Ireland after 30 years of oblivion. J. Infect. 50, 61–65] and could be a further indication for a sylvatic Trichinella life cycle existing independently from the domestic cycle.  相似文献

Human trichinellosis is a foodborne disease caused by ingestion of infective Trichinella muscle larvae via pork or meat of other food animals which are susceptible to this zoonotic parasite. There are new approaches for a risk-oriented meat inspection for Trichinella in pigs which are accompanied by monitoring programmes on herd level to control freedom from this parasite. For this purpose, testing schemes utilizing serological tests with a high sensitivity and specificity are required.This study aimed at the evaluation of an ELISA and a Western Blot (WB) for the detection of anti-Trichinella-IgG in terms of sensitivity and specificity taking results of artificial digestion as gold standard. For this purpose, 144 field sera from pigs confirmed as Trichinella-free as well as 159 sera from pigs experimentally infected with T. spiralis (123), T. britovi (19) or T. pseudospiralis (17) were examined by ELISA (excretory–secretory antigen) and WB (crude worm extract). Sera from pigs experimentally infected with four other nematode species were included to investigate the cross-reactivity of the antigen used in the WB. For all Trichinella-positive pig sera, band pattern profiles were identified in the WB and results were analysed in relation to ELISA OD% values.Testing of pig sera revealed a sensitivity of 96.8% for the ELISA and 98.1% for the WB whereas the methods showed a specificity of 97.9 and 100%, respectively. WB analysis of Trichinella-positive pig sera revealed five specific band patterns of 43, 47, 61, 66, and 102 kDa of which the 43 kDa protein was identified as the predominant antigen. The frequency of the band pattern profile was irrespective of the dose and the period of infection as well as the Trichinella species investigated.In conclusion, monitoring in swine farms for Trichinella antibodies should be based on screening pig sera by means of ELISA followed by confirmatory testing through WB analysis.  相似文献

The aim of the present work was to determine the presence of human and porcine trichinellosis in an area of Argentina historically regarded as Trichinella-free. Human blood donors (n = 216) and swine destined for consumption (n = 57) were evaluated by serological techniques (ELISA, immunofluorescence, and/or Western Blot). Muscle tissues from 26 of the pigs were evaluated for the presence of Trichinella larvae by the artificial digestion method. A questionnaire was used to collect and evaluate data on eating habits of the human population under study and on swine-raising conditions. The survey showed that 98.1% of the individuals (n = 212) were regular consumers of pork in the form of stuffed products such as sausages produced by local butchers. The seroprevalence (positive sera by at least two of the three methods) was 8.3% (n = 18) for human trichinellosis and 24.5% (n = 14) for porcine trichinellosis. Trichinella spiralis larvae were found in 2 of the 26 pigs (7.7%) with parasite loads of 0.33 and 2.4 muscle larvae per gram. Twelve swine found positive by serological and/or parasitological tests were raised under poor sanitary conditions (presence of rubbish in the surroundings, with cannibalism and scavenging behaviors, presence of rodents, etc.). Our study confirms the existence of porcine trichinellosis in an area regarded as Trichinella-free, provides supporting serological evidence of human infection in this area, and indicates that failure to report cases of trichinellosis based on inadequate surveillance can result in incorrect prevalence classification of an area.  相似文献

Trichinella infections in humans and pigs have been documented in Greece since 1945 and a high prevalence of infection in pigs occurred in the 1950s. Up to 1984 only sporadic infections in humans were documented, and this zoonosis was not considered as a public health problem until 2009 when a human outbreak caused by the consumption of pork from an organic pig farm occurred. In the present study, we describe the re-emergence of Trichinella spp. infections in free-ranging pigs from organic farms of 3 counties (Dramas, Evros and Kavala) in Northern-Eastern Greece during the period 2009–2012. Totally 37 out of 12,717 (0.29%) free-ranging pigs which were tested during the period in question, were positive for Trichinella spp. larvae. The etiological agent was identified as Trichinella britovi. The average larval burden was 13.7 in the masseter, 6.2 in the foreleg muscles and 7.5 in the diaphragm. The 37 positive animals originated from seven free range pig farms. The practice of organic pig production systems in Greece has grown in popularity over the last years due to the increasing interest of consumers for products considered as traditional. However, this type of pig production increases the risk for Trichinella spp. infections, since animals can acquire the infection by feeding on carcasses or the offal of hunted or dead wild animals. The awareness and education of hunters and farmers is extremely important to reduce the transmission among free ranging pigs and the risk for humans.  相似文献

The wild boar is an important source of trichinellosis for people in European countries as a large number of hunted animals escape veterinary control. In November 2012, uncooked sausages made with meat from wild boar were consumed by 38 persons in a village of the Lucca province (Tuscany region, Italy). Of them, 34 were serologically positive, 32 developed clinical signs and symptoms of trichinellosis, and two were asymptomatic. Trichinella britovi larvae were detected in vacuum‐packed sausages made with the same batch of sausages consumed raw which had been prepared with meat from wild boar hunted in the Lucca province. As no case of trichinellosis had been reported in this region during the last 20 years, the regional public health authority considered the risk for this zoonosis to be negligible and put in place a surveillance programme on Trichinella spp. in indicator animals (mainly foxes and including wild boar for private consumption), by testing only a percentage of heads. The experience from this outbreak shows that the definition of a region with a negligible risk for Trichinella infection is not applicable to wild boar and stresses the need to test all Trichinella‐susceptible wild animals intended for human consumption and to implement risk communication to consumers and hunters.  相似文献

The aim of this study was to determine the presence of trichinellosis in backyard-farmed pigs and the risk factors associated with the infection in Zaria, Kaduna State. Serum samples were collected from 120 pigs selected at random from 50 small backyard farms, and the presence of Trichinella spp. antibodies was determined using a commercially available enzyme-linked immunosorbent assay (ELISA) kit. Data on farm management practices from the farms were obtained through the use of a structured questionnaire. The overall seroprevalence of Trichinella spp.-specific antibodies was 40 % (48/120) by ELISA. All the extensive farms sampled had at least one Trichinella-positive animal. The age and sex of the animals were not significantly (p?>?0.05) associated with the infection; however, the management systems, presence of rodents, rodent control, and access to dead pigs showed significant (p?<?0.05) association with Trichinella spp.-infected pigs on the farm. In conclusion, there was a high prevalence of antibodies to trichinellosis in backyard raised pigs in Zaria, and intensive pig farming with the adoption of proper biosecurity measures is advocated to prevent the transmission and spread of trichinellosis.  相似文献

In Slovakia, monitoring the prevalence of Trichinella spp. in wildlife was performed since 2000 in the main reservoir animals, the red fox (Vulpes vulpes) and wild boar (Sus scrofa), using artificial digestion method as recommended by International Commission on Trichinellosis. The results of investigation performed in 5270 red foxes showed that Trichinella infection is widespread across Slovakia and prevalence increased significantly from 4.9% in 2000 to 20.5% in 2007. Recently, a higher Trichinella prevalence (0.11%) in wild boars was also demonstrated. The results indicate that foxes and wild boars are involved in the spread of Trichinella, although the latter host species seems to play a secondary role in the maintenance of the sylvatic cycle in Slovakia. Trichinella britovi is the predominant species circulating in Slovakia, both in foxes and wild boars, and Trichinella spiralis occurs only sporadically. Mixed infections of T. britovi and Trichinella pseudospiralis were recorded in 2005 in one wild boar from Eastern Slovakia and in 2006 in one red fox from the same region. These findings are important with respect to an outbreak caused by T. pseudospiralis in a pig farm in the same district 3 years ago. This study provides a complex picture on Trichinella occurrence in all regions of Slovakia and may be a good basis for evaluating the risk of parasite transmission to the domestic cycle and human beings.  相似文献

Because of its role in human disease, there are increasing global requirements for reliable diagnostic and control methods for Trichinella in food animals to ensure meat safety and to facilitate trade. Consequently, there is a need for standardization of methods, programs, and best practices used in the control of Trichinella and trichinellosis. This review article describes the biology and epidemiology of Trichinella, and describes recommended test methods as well as modified and optimized procedures that are used in meat inspection programs. The use of ELISA for monitoring animals for infection in various porcine and equine pre- and post-slaughter programs, including farm or herd certification programs is also discussed. A brief review of the effectiveness of meat processing methods, such as freezing, cooking and preserving is provided. The importance of proper quality assurance and its application in all aspects of a Trichinella diagnostic system is emphasized. It includes the use of international quality standards, test validation and standardization, critical control points, laboratory accreditation, certification of analysts and proficiency testing. Also described, are the roles and locations of international and regional reference laboratories for trichinellosis where expert advice and support on research and diagnostics are available.  相似文献

Seropositivity among pregnant woman in Austria has decreased from 48% to 50% at the end of the 1970s to 35% in recent years. Despite this decrease, knowledge of possible sources and risk factors for Toxoplasma infection remains important. We reviewed seroepidemiological studies that were undertaken to assess the roles of undercooked meat and oocysts in cat faeces as potential sources of infection in pregnant women. Improved management and hygiene in pig herds raised in confinement have resulted in less contact of pigs with cats and a decrease of infected pigs within one decade from 14% in 1982 to 0.9% in 1992. In Austrian wild boar populations, however, seroprevalences remained essentially unchanged during the same decade (18% in 1983 and 19% in 1990–1993). Austrian sheep and goats are usually kept on small farms where cats abound and are predominantly seropositive (66% in sheep and 69% in goats). The seroprevalence in cats has decreased from approximately 81% in 1987 to 59% in 1996; presumably because of cats’ increased consumption of processed food. Despite the decrease of infection in pregnant women via the cat‐to‐pig pathway, it may be offset by a recent concomitant increase in mutton consumption. Free‐ranging chickens are a good indicator of the prevalence of T. gondii oocysts in the soil because chickens are ground feeders. Antibodies to T. gondii, as evaluated by the modified agglutination test, were found in 36% of chickens from 12 Austrian ‘biofarms’. Because Austrians rarely consume raw pork, the fraction of human T. gondii infections associated with pig meat consumption is likely small. As meat consumption and lifestyle patterns change in Austria, the risk of human infection with T. gondii via different pathways needs re‐evaluation and targeted educational efforts to control transmission.  相似文献

Abstract

AIMS: To determine the annual likelihood of exposure to an infectious dose of Trichinella spiralis from consuming imported pork meat from New Zealand to Singapore.

METHODS: Input values specific for chilled pork meat imported into Singapore from New Zealand were used in a quantitative risk-assessment model. The model, designed to allow any combination of importing and exporting countries, was divided into two components, viz the release assessment, and the exposure assessment that assessed the annual risk of exposure to the consumer (ARC). The former estimated the likelihood that a contaminated fresh meat product from New Zealand would arrive at Singapore's border, and took into consideration the prevalence of disease on different types of farms. The latter determined the likelihood over a year that a person in Singapore would consume one or more servings of imported fresh meat from New Zealand that contained a burden of greater than or equal to one larva(e) of T. spiralis per gram after preparation for consumption.

RESULTS: The ARC for offal was 2.41 × 10?7, which was below the pre-selected safety threshold of 1.00 × 10?6. The ARC for lean meat was 2.39 x 10?5, which was above the acceptable safety threshold.

CONCLUSIONS: The study demonstrated that continued routine testing at slaughter is unnecessary for pig offal produced commercially, and provided a model with which to further assess management of the risk of exposure to T. spiralis in lean meat.

CLINICAL RELEVANCE: The potential of Trichinella species to cause disease in humans is a public health concern, and has created adverse effects on the international trade of fresh lean meat without regard to the surveillance measures employed by particular pork-producing countries.  相似文献

Endemic trichinellosis is re-emerging in Serbia and it is a serious problem both from the perspective of human health and animal husbandry. The widespread appearance of human trichinellosis is attributed to a high prevalence of Trichinella infection in domestic animals, especially swine. Epidemiological data presented in this paper were collected during a 12-year period (1995–2006) at small private swine farms in the region of Branicevo, Serbia where a high Trichinella prevalence in slaughter pigs (0.57%) has been detected. To further monitor Trichinella prevalence in swine, a serological survey, using ELISA, was performed in 2006. Of 916 swine tested by ELISA, Trichinella specific antibodies were detected in 15 (1.64%), while suspect results were obtained in 10 (1.09%). Positive or suspect animals originated from all parishes except one (Pozarevac). Our results point to the need for systematic monitoring in pigs to achieve a better control of trichinellosis in Serbia.  相似文献

Trichinellosis is a zoonotic disease due to the ingestion of raw or undercooked meat from animals infected with the larvae of nematodes belonging to the genus Trichinella. In January–February 2015, an outbreak of trichinellosis occurred in Genoa, Northern Italy. The epidemiological link was traced back to a dinner served at an agritourism farm on 31 December 2014, where a majority of the 52 guests had consumed the ‘beef’ steak tartare. The source of infection was not traced; however, it was noted that the amount of beef purchased officially for providing at the dinner did not correspond with that served, suggesting that meat of a different origin had been added to the beef to prepare the steak tartare. Clinical and laboratory data of 30 individuals out of the 52 (57.7%), of which four were hospitalized, were consistent with that of the case definition of trichinellosis. Western blot patterns of the sera from patients with confirmed trichinellosis were similar to the diagnostic pattern identified for the reference sera of Trichinella pseudospiralis but different from those of the control sera tested for patients infected with Trichinella spiralis and Trichinella britovi. Identification of T. pseudospiralis as the aetiological agent responsible for the outbreak of trichinellosis using an indirect tool represents an advancement in the epidemiological investigation of this zoonotic disease.  相似文献

Epidemiology of trichinellosis in Mexico, Central and South America   总被引:6,自引:0,他引:6  
Trichinella species are widely distributed throughout the world and are found in a large number of carnivorous animals, humans and incidental hosts. The data presented in this review show that Trichinella infection has been reported in both humans and animals in Mexico, Argentina and Chile since the end of the 19th century, and more recently in Bolivia. This parasitic infection is still a public health problem in countries such as Argentina and Chile. Although efforts have focused on the control and prevention of trichinellosis in these countries, there were still human cases and outbreaks of trichinellosis reported. Diagnosis of infection in animals such as pigs still includes, in many endemic areas, the use of trichinoscopy. In Argentina, however, artificial digestion has been recently introduced in slaughterhouses to detect Trichinella infection in pigs. In some endemic areas in Mexico, the use of serological assays for human trichinellosis and pig infections have resulted in improved detection. Most of the outbreaks of human trichinellosis in Mexico, Argentina and Chile have occurred as a result of the consumption of undercooked pork or pork products from animals raised under poor hygienic conditions and which are clandestinely slaughtered. In several studies, rats, dogs and cats have been found to be infected with Trichinella and may be considered a risk for transmission of the infection to pigs or other animals intended for human consumption. Another potential source of transmission of Trichinella to humans is horsemeat; however, information related to horse trichinellosis in Latin-American countries is scarce. In most cases the etiological agent of human trichinellosis in Central and South America has been reported to be Trichinella spiralis; however, only in a few cases has the parasite species been properly identified. Based on the reports available, it is clear that there is a need to carry out better controlled epidemiological studies to determine the true prevalence of the infection in this region of the world. Also, more sensitive methods of diagnosis and improvements in conditions for pig production as well as better sanitary inspection systems, are needed for controlling and preventing transmission of trichinellosis in these countries.  相似文献

Trichinella spiralis larvae establish chronic infections in skeletal muscles of immunocompetent hosts. Muscle infection is crucial to transmission and survival of the parasite in nature. Chronic infections by this highly immunogenic parasite are associated with modulation or escape from potentially destructive immune responses. Increasing demands for cost-effectiveness in surveillance for human health hazards from animal origins can be met by introducing risk-based principles. This e.g. implies targeting subpopulations with higher risk of infection compared to the whole population. Furthermore, historical data from surveillance can be collated and used to assess future risk of infection. To demonstrate the effectiveness of combining these two approaches, we used a model called “Discounting historical evidence”. It depends mainly on the annual risk of introduction (PIntro) and the surveillance system sensitivity (SSe) (ability to detect infection if present). The model implies simulations that reiterate for a number of years. For each year the output is updated with the confidence on absence in infection. Trichinella spiralis infection in pigs is used as an example. In Denmark, more than 20 million pigs are tested annually. Despite more than 70 years of testing no pigs have been found positive for Trichinella. Hence, PIntro is low. SSe can be estimated from the maximum number of infected carcasses expected under the specified design prevalence, and the sensitivity of the test applied. According to our assessment, the current prevalence of Trichinella in Danish pigs is less than one case per million, which we interpret as negligible risk. Based on this, a risk-based surveillance programme for Trichinella is designed that targets all out-door reared pigs as well as all sows and boars (current total 610,000 slaughtered annually). These subpopulations are judged to have higher risk of getting Trichinella. Again, SSe and PIntro are estimated and the model results show that risk-based surveillance can be applied without jeopardizing human health. Finally, we incorporate wildlife surveys and test quality assurance in the programme. The results of the simulation model were included in an application to the European Commission concerning Denmark's status as a region with negligible risk of Trichinella. In July 2007, the European Commission granted status as “negligible risk” to Danish pigs and pork.  相似文献

Trichinellosis is a major food-borne zoonosis with health, social, and economic impacts. Epidemiological data on swine trichinellosis in China from 2005 to 2009 were obtained from seven Provinces/autonomous regions/Municipalities (P/A/M) and analyzed and sero-epidemiological data were acquired from five P/A. The seroprevalence ranged from 0.01% to 29.95% as determined by enzyme-linked immunosorbent assay or an immunochromatographic strip method. The prevalence of Trichinella infection in swine slaughtered at abattoirs varied from 0% to 5.75% in five P/A. Between 2005 and 2009, endemic areas of swine trichinellosis were located mainly in the Western (Guangxi and Qinghai), central (Henan and Hubei), and North-eastern parts (Heilongjiang) of China.Swine trichinellosis in China is transmitted mostly through garbage. Pigs infected with Trichinella are predominately from small backyard farms where animals are raised under poor hygienic conditions, and from rural and mountainous areas where they range freely at pasture. The prevalence of Trichinella in pork sold at the market was reported in four P/A, and varied from 0.06% to 5.6% as determined by trichinoscopy or the digestion method. From 2005 to 2009, 15 outbreaks of human trichinellosis, with 1387 cases and 4 deaths, were recorded in three P/A of South-western China. Twelve (85.71%) of these 15 outbreaks were caused by the eating of raw or undercooked pork, which remains the predominant source of trichinellosis in humans. Pig-rearing practices must be improved, and mandatory inspection of pork further strengthened in rural and mountainous areas in Western China for the control of the disease.  相似文献

Two species of Trichinella were identified from China by means of PCR amplification of the mitochondrial small subunit ribosomal DNA and the expansion segment V region of the ribosomal DNA. Seven isolates originating from domestic pig and one isolate originating from dog showed identical DNA banding pattern to Trichinella spiralis, and two isolates from dog showed DNA banding pattern identical to Trichinella nativa. Sequence analysis of the 5S rDNA inter-gene spacer region from the ten Trichinella isolates confirmed the existence of only two Trichinella species and revealed the inner species genetic variation within T. spiralis and T. nativa.  相似文献

Trichinellosis is one of the most important zoonotic diseases in Romania. Even though the disease is a serious public health concern, only a limited number of Trichinella isolates have been identified at the species level; in the past, all larvae were assumed to be Trichinella spiralis. The present study was conducted to identify Trichinella spp. circulating among wild and domestic animals in Romania, using PCR-based methods. Trichinella spp. larvae originating from 54 wild and 23 domestic mammals were examined. No Trichinella spp. larvae were detected in muscle samples of 182 birds. T. spiralis and Trichinella britovi were the only two species identified in the 40 isolates that yielded a positive PCR result. Overall, T. britovi was more prevalent (n = 26; 65%) than T. spiralis (n = 14; 35%). T. spiralis was the predominant species found in domestic animals (n = 9; 75%), while T. britovi was more prevalent in wildlife (n = 24; 86%). No mixed infections were found. The highest prevalence of Trichinella infection was detected in wolves (11/35; 31%), in European wild cats (4/28; 14%), and in red foxes (5/71; 7%). The distribution of Trichinella spp. in Romania does not show a species-specific clustering; both of the two species found were present over the entire range of counties studied.  相似文献
