The present study was to examine effect of soy leaf powder (SLP) and soy leaf ethanol extract (SLEE) on serum lipoproteins in hamsters. The control group was fed a semisynthetic diet containing 0.1% cholesterol, while the tested groups were maintained on the same diet but supplemented with 3% SLP or the equivalent amount of SLEE derived from 3% SLP for 4 weeks. SLP supplementation led to a trend of lowering serum total cholesterol (TC) and nonhigh density lipoprotein cholesterol (non-HDL-C), with HDL-C being unaffected, whereas incorporation of SLEE into the diet led to an elevated level of HDL-C and a lower level of non-HDL-C with TC being unchanged. Both SLP and SLEE supplementation caused favorably a decrease in the ratio of non-HDL-C to HDL-C. The present results demonstrate that not only soybean seeds but also soy leaves are cardioprotective, by favorably modulating serum lipid profile.  相似文献

The present study examined the cholesterol-lowering activity of algal powder (AP), algal lipids (AL), and algal residue (AR) and their interaction with genes of transporters, receptors, and enzymes involved in cholesterol absorption and metabolism. In this experiment, 48 hamsters were fed either control diet or one of the three experimental diets containing 2% AP, 1.0% AL, or 1.0% AR for 6 weeks. Plasma total cholesterol (TC) and non-high-density-lipoprotein-cholesterol (non-HDL-C) were significantly decreased in the AP and AL groups but not in the AR group compared with those in the control hamsters. It was found that the cholesterol-lowering activity of AP and AL was associated with down-regulation of hepatic 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase, low-density lipoprotein receptor (LDLR), and intestinal Niemann-Pick C1-like 1 (NPC1L1) transporter. It was concluded that the alga possessed the cholesterol-lowering activity and its lipids were the active ingredients. The mechanisms underlying the cholesterol-lowering activity of algae were mediated most likely by increasing the sterol excretion and decreasing the cholesterol absorption and synthesis.  相似文献

Seven legume extracts containing phytoestrogens were analyzed for estrogenic activity. Methanol extracts were prepared from soybean (Glycine max L.), green bean (Phaseolus vulgaris L.), alfalfa sprout (Medicago sativa L.), mung bean sprout (Vigna radiata L.), kudzu root (Pueraria lobata L.), and red clover blossom and red clover sprout (Trifolium pratense L.). Extracts of kudzu root and red clover blossom showed significant competitive binding to estrogen receptor beta (ERbeta). Estrogenic activity was determined using an estrogen-dependent MCF-7 breast cancer cell proliferation assay. Kudzu root, red clover blossom and sprout, mung bean sprout, and alfalfa sprout extracts displayed increased cell proliferation above levels observed with estradiol. The pure estrogen antagonist, ICI 182,780, suppressed cell proliferation induced by the extracts, suggesting an ER-related signaling pathway was involved. The ER subtype-selective activities of legume extracts were examined using transiently transfected human embryonic kidney (HEK 293) cells. All seven of the extracts exhibited preferential agonist activity toward ERbeta. Using HPLC to collect fractions and MCF-7 cell proliferation, the active components in kudzu root extract were determined to be the isoflavones puerarin, daidzin, genistin, daidzein, and genistein. These results show that several legumes are a source of phytoestrogens with high levels of estrogenic activity.  相似文献

OBJECTIVE: To compare serum lipids and their changes during a family-based health education in children aged 6-17 years with or without the epsilon4 allele of the gene encoding apolipoprotein E (apoE). DESIGN: An intervention study. SETTING: A family-based prevention of risk factors of coronary heart disease in Eastern Finland. The programme consisted of two counselling meetings at children's schools and three at children's homes. SUBJECTS: Four hundred and thirty-nine children with a family history of cardiovascular diseases (CVD) participated in a family-based health education. The children were divided into two groups according to apoE genotype. The risk group consisted of 143 children having apoE epsilon4 allele (genotype epsilon3/4 or epsilon4/4) and the non-risk group of 296 children without apoE epsilon4 allele (epsilon2/3 or epsilon3/3). The final sample of the follow-up study included 354 (81%) children (114 and 240, respectively). RESULTS: Baseline differences were found in low-density lipoprotein cholesterol (LDL-C) (P=0.007) and LDL-C/high-density lipoprotein cholesterol (HDL-C) ratio (P=0.030) among boys and in total cholesterol (TC)/HDL-C (P=0.008) and LDL-C/HDL-C ratios (P=0.006) among girls. Differences between groups in changes during the follow-up were observed only for TC/HDL-C ratio (P-value adjusted for age=0.049) among boys. CONCLUSIONS: At baseline, children with apoE epsilon4 allele had on average a more unfavourable lipid profile than those without apoE epsilon4 allele. However, the effect of about 33 months' family-based health education on plasma lipids did not depend on apoE genotype in children with a family history of CVD.  相似文献

Dietary phytoestrogens may be involved in the occurrence of chronic diseases. Reliable information on the phytoestrogen content in foods is required to assess dietary exposure and disease risk in epidemiological studies. However, existing analyses have focused on only one class of these compounds in plant-based foods, and there is only little information on foods of animal origin, leading to an underestimation of intake. This is the first comprehensive study of phytoestrogen content in animal food. We have determined the phytoestrogen content (isoflavones: biochanin A, daidzein, formononetin, genistein, and glycitein; lignans: secoisolariciresinol and matairesinol; coumestrol; equol; enterolactone; and enterodiol) in 115 foods of animal origin (including milk and milk-products, eggs, meat, fish, and seafood) and vegetarian substitutes using liquid chromatography-mass spectrometry (LC-MS) with (13)C-labeled internal standards. Phytoestrogens were detected in all foods analyzed; the average content was 20 microg/100 g of wet weight (isoflavones, 6 microg/100 g; lignans, 6 microg/100 g; equol, 3 microg/100 g; and enterolignans, 6 microg/100 g). In infant soy formula, 19 221 microg/100 g phytoestrogens were detected (compared to 59 microg/100 g in non-soy formula). Our study shows that all foods analyzed contained phytoestrogens and most foods (except for fish, seafood, and butter) contained mammalian phytoestrogens (enterolignans and equol). This is the first comprehensive study of phytoestrogen content of foods of animal origin and will allow for a more accurate estimation of exposure to dietary phytoestrogens.  相似文献

The effect of rice bran ethanolic extract from Superjami, a new blackish‐purple pigmented cultivar, on the lipid and glucose metabolisms in a postmenopausal‐like model of ovariectomized rats was investigated. Sprague‐Dawley female rats were subjected to either bilateral ovariectomy or a sham operation and randomly divided into three dietary groups (n = 6): sham‐operated fed with normal diet (SHAM), ovariectomized fed with normal diet (OVX‐AIN76), and ovariectomized fed with normal diet supplemented with Superjami bran extract (OVX‐SJ). The OVX‐AIN76 group showed significantly higher body weight, triglyceride, total cholesterol, blood glucose, and insulin than the SHAM group. On the other hand, supplementation of Superjami bran extract markedly reduced the body weight gain and improved the lipid and glucose profiles in ovariectomized rats through inhibition of hepatic lipogenesis and adipokine production and modulation of glucose‐regulating enzyme activities. These findings demonstrate that extracts from Superjami bran may be useful in the prevention and management of postmenopause‐induced hyperlipidemia and hyperglycemia.  相似文献

Nutrient levels in buckwheats that were maximized in day 8 sprouts (D8SP) included total phenolics, quercetin, and l-ascorbic acid, whereas those of oxalic, malic, tartaric, and citric acids, rutin, and gamma-aminobutyric acid (GABA) were found to reach maximum levels on day 10. Ethanolic extract of D8SP (2.5 mg/mL) revealed potent free-radical scavenging (FRS) and antioxidative (ANO) capabilities. However, its Fe2+-chelating capability was only moderate. To further study the hypolipidemic activity of D8SP, 36 Syrian hamsters were grouped into six groups and fed for 28 days, respectively, with (i) control meal, (ii) high fat plus high cholesterol meal, (iii) high fat plus high cholesterol plus 2.5% of buckwheat seeds, (iv) high fat plus high cholesterol plus 25% of buckwheat seeds, (v) high fat plus high cholesterol plus 2.5% of D8SP, and (vi) high fat plus high cholesterol plus 25% of D8SP. High seed meal prominently enhanced body weight gain, whereas high sprout meal exhibited the highest feed efficiency. Ratios of liver/body weight (L/B) were significantly lowered by all BS meals. Although low seed meal reduced serum total cholesterol (TC) levels (p<0.05), its effect was still inferior to the high seed and sprout meals (p<0.01). In contrast, serum triglyceride (TG) levels were lowered only by the high seed and sprout meals (p<0.05). Alternatively, levels of serum low-density lipoprotein cholesterol (LDL-C) were significantly suppressed by all buckwheat meals (p<0.01). Serum high-density lipoprotein cholesterol (HDL-C) levels were increased, however, insignificantly. Nutraceutically more meaningful is that both LDL-C/HDL-C and TC/HDL-C ratios were significantly lowered (p<0.01). Apparently, hepatic TC levels were significantly reduced, whereas hepatic TG levels were totally unaffected. Conclusively, sprouting triggers a variety of nutritional changes in buckwheats. Day 8 sprouts, consisting of high polyphenolic and moderate quercetin contents, are nutraceutically maximized when hypocholesterolemic, hypotriglyceridemic, and antioxidative activities are concerned.  相似文献

The effect of olive oils on lipid metabolism and antioxidant activity was investigated on 60 male Wistar rats adapted to cholesterol-free or 1% cholesterol diets. The rats were divided into six diet groups of 10. The control group (control) consumed the basal diet (BD) only, which contained wheat starch, casein, cellulose, and mineral and vitamin mixtures. To the BD were added 10 g/100 g virgin (virg group) or Lampante (Lamp group) oils, 1 g/100 g cholesterol (chol group), or both (chol/virg group) and (chol/Lamp group). The experiment lasted 4 weeks. Plasma total cholesterol (TC), LDL-cholesterol (LDL-C), HDL-cholesterol (HDL-C), triglycerides (TG), total phospholipids (TPH), HDL-phospholipids (HDL-PH), total radical-trapping antioxidative potential (TRAP), malondialdehyde lipid peroxidation (MDA), and liver TC were measured. Groups did not differ before the experiment. In the chol/virg and chol/Lamp vs chol group, the oil-supplemented diets significantly (P < 0.05) lessened the increase in plasma lipids due to dietary cholesterol as follows: TC (25.1 and 23.6%), LDL-C (39.3 and 34.7%), TG (19.3 and 17.0%), and TC in liver (36.0 and 35.1%) for the chol/virg and chol/Lamp group, respectively. The chol/virg and chol/Lamp diets significantly decreased the levels of TPH (24.7 and 21.2%; p < 0.05 in both cases) and HDL-PH (22.9 and 18.0%; p < 0.05 in both cases) for the chol/virg and chol/Lamp group, respectively. Virgin and Lampante oils in rats fed basal diet without cholesterol did not affect the lipid variables measured. Virgin, and to a lesser degree Lampante, oils have increased the plasma antioxidant activity in rats fed BD without cholesterol (an increase in TRAP, 20.6 and 18.5%; and a decrease in MDA, 23.2 and 11.3%, respectively). In the rats of chol/virg and chol/Lamp vs Chol diet groups the added oils significantly hindered the decrease in the plasma antioxidant activity (TRAP, 21.2 and 16.7%; and MDA, 27.0 and 22.3%, respectively). These results demonstrate that virgin, and to less degree Lampante, oils possess hypolipidemic and antioxidant properties. It is more evident when these oils are added to the diets of rats fed cholesterol. These positive properties are attributed mostly to the phenolic compounds of the studied oils.  相似文献

This study was designed to test the lipid-lowering and antioxidative activities of triacetylated hydroxytyrosol compared with its native compound, hydroxytyrosol, purified from olive tree leaves. Wistar rats fed a standard laboratory diet or a cholesterol-rich diet for 16 weeks were used. The serum lipid levels, the thiobarbituric acid-reactive substances (TBARS) level, as an indicator of lipid peroxidation, and the activity of superoxide dismutase (SOD) as well as that of catalase (CAT) were examined. The cholesterol-rich diet induced hypercholesterolemia that was manifested in the elevation of total cholesterol (TC), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C). Administration of hydroxytyrosol and triacetylated hydroxytyrosol (3 mg/kg of body weight) decreased the serum levels of TC, TG, and LDL-C significantly and increased the serum level of high-density lipoprotein cholesterol (HDL-C). Furthermore, the content of TBARS in liver, heart, kidney, and aorta decreased significantly when hydroxytyrosol and its triacetylated derivatives were orally administered to rats compared with those fed a cholesterol-rich diet. In addition, triacetylated hydroxytyrosol and hydroxytyrosol increased CAT and SOD activities in the liver. These results suggested that the hypolipidemic effect of triacetylated hydroxytyrosol and hydroxytyrosol might be due to their abilities to lower serum TC, TG, and LDL-C levels as well as to their antioxidant activities preventing the lipid peroxidation process.  相似文献

Phytoestrogens are secondary plant metabolites that have received increasing attention for their bioactivity, in particular due to their structural and functional similarity to 17beta-estradiol. Although urinary and plasma phytoestrogens can be used as biomarkers for dietary intake, this is often not possible in large epidemiological studies or in the assessment of general exposure in free-living individuals. Accurate information about dietary phytoestrogens is therefore important, but there are very limited data concerning food contents. In this study was analyzed a comprehensive selection of tea, coffee, alcoholic beverages, nuts, seeds, and oils for their phytoestrogen content using a newly developed sensitive method based on LC-MS incorporating (13)C 3-labeled standards. Phytoestrogens were detected in all foods analyzed, although the contents in gin and bitter (beer) were below the limit of quantification (1.5 microg/100 g). Lignans were the main type of phytoestrogens detected. Tea and coffee contained up to 20 microg/100 g phytoestrogens and beer (except bitter) contained up to 71 microg/100 g, mainly lignans. As these beverages are commonly consumed, they are a main source of dietary lignans. The results published here will contribute to databases of dietary phytoestrogen content and allow a more accurate determination of phytoestrogen exposure in free-living individuals.  相似文献

本试验研究饲料中添加不同水平的DHEA对肉鸡脂类代谢和相关激素的影响。将180羽1日龄AA肉鸡随机分为饲料中不添加DHEA的对照组,添加20 mg•kg-1和5 mg•kg-1DHEA的高、低剂量组,饲喂至42日龄时分别从各组随机取12只公鸡和母鸡,采集血样、肝脏、腹脂、左侧胸肌和腿肌,分别测定血清葡萄糖(BG)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、非酯化脂肪酸(NEFA)和三碘甲腺原氨酸(T3)、甲状腺素(T4)、游离T3(FT3)、游离T4(FT4)、胰高血糖素(glucagon)、瘦素(leptin)含量。结果表明:与对照组相比,DHEA显著降低肉鸡体重、腹脂重和腹脂率,显著降低TG、TC、HDL-C、FT3、FT4含量,显著升高瘦素含量,而对血糖和LDL-C没有影响;DHEA使公鸡血清NEFA含量升高,母鸡血清NEFA含量降低;DHEA使公鸡血清LPL活性和胰高血糖素含量显著升高,血清T4显著降低。提示,DHEA可能通过提高肉鸡体内脂类分解代谢相关酶的活性及激素水平,对脂类代谢起调节作用。  相似文献

Postmenopausal women have an increased risk of coronary heart disease. Oxidation of low-density lipoprotein (LDL) has been implicated in atherogenesis, and the presence of modified LDL (LDL(-)) in plasma appears to represent LDL oxidation in vivo. Because previous studies have demonstrated a strong antiatherogenic effect of estrogen due to its antioxidant activity and similar antioxidant activity was found for specific isoflavones derived from soy extract, the antioxidant activity of a phytoestrogen extract derived from soy and alfalfa was studied. Copper-mediated LDL oxidation was inhibited in the presence of soy and alfalfa extracts, and this effect was further enhanced in the presence of acerola cherry extract, which is rich in ascorbic acid. Male rabbit aortic endothelial cells pretreated with soy extract were resistant to the toxic effects of high levels of LDL and LDL(-), and a lesser, but significant protection, was also afforded by alfalfa extract. Cell-mediated oxidation of LDL, measured by LDL(-) formation, was inhibited in the presence of soy extract but not alfalfa extract. However, in the presence of acerola cherry extract, both soy and alfalfa extracts potently inhibited the formation of LDL(-). These findings show that acerola cherry extract can enhance the antioxidant activity of soy and alfalfa extracts in a variety of LDL oxidation systems. The protective effect of these extracts is attributed to the presence of flavonoids in soy and alfalfa extracts and ascorbic acid in acerola cherry extract, which may act synergistically as antioxidants. It is postulated that this synergistic interaction among phytoestrogens, flavonoids, and ascorbic acid is due to the "peroxidolitic" action of ascorbic acid, which facilitates the copper-dependent decomposition of LDL peroxides to nonradical products; this synergy is complemented by a mechanism in which phytoestrogens stabilize the LDL structure and suppress the propagation of radical chain reactions. The combination of these extracts markedly lowers the concentrations of phytoestrogens required to achieve significant antioxidant activity toward LDL.  相似文献

Type 1 diabetes (T1D) damages pancreatic islets, gradually causing chronic complications. This study investigated the berberine effect on T1D in vivo. Nonobese diabetic (NOD) mice were grouped and administered 50, 150, and 500 mg of berberine/kg of body weight over 14 weeks using consecutive tube feeding. Changes in pancreatic islets, serum insulin, berberine, and lipid levels were determined. The results showed that berberine supplementation significantly (P < 0.05) increased the number of decreased islets and raised serum berberine levels in dose-dependent manners in experimental mice. Berberine supplementation also increased insulin levels, but decreased the ratio of low-density lipoprotein cholesterol (LDL-C)/total cholesterol (TC). Furthermore, serum berberine levels showed a significantly positive correlation with high-density lipoprotein cholesterol (HDL-C) levels and the HDL-C/TC ratio, but a negative correlation with the LDL-C/HDL-C ratio. This study suggests that berberine administration in vivo protects pancreatic islets and serum lipids in NOD mice.  相似文献

The aim of this study was to evaluate the antiatherosclerotic effect of commercially available phenolic-rich extracts from grape seeds (ExGrape seeds, EGS; grape seed extract, GSE) and marc (ExGrape total, EGT) in cholesterol-fed hamsters and to investigate possible operating mechanisms. These extracts fed at a moderate dose mimicking two glasses of red wine per meal reduced plasma cholesterol (-11% on average) but did not affect plasma antioxidant capacity of hamsters. The extracts prevented the development of aortic atherosclerosis by 68% (EGS), 63% (EGT), and 34% (GSE). Elsewhere, in an ex vivo experiment using rat aortic rings, EGS (7 microg/mL) induced 77% endothelium-dependent relaxation, whereas EGT and GSE (30 microg/mL) induced 84 and 72%, respectively. These results suggests that phenolic extracts from grape seeds and marc are beneficial in inhibiting atherosclerosis by indirect mechanism(s).  相似文献

Groups of hamsters were fed diets containing tristearin (TS), a baker's shortening (BS), soybean oil (SO), a blend (BL) of TS and SO, or a hard fat (HF). Only BS contained trans fatty acids. After four weeks, serum total cholesterol (T-CH) levels were most elevated in hamsters fed HF. In comparison, hamsters fed BS, BL, or SO showed 13.3, 15.4, or 23.7% reduction in serum T-CH, respectively. Reduction in serum T-CH was even more pronounced in hamsters fed TS. However, this group also showed poor weight gains, apparently because only 16.2% of ingested TS was absorbed. Reduction in T-CH occurred due to reduction of both high-density-lipoprotein (HDL)-CH and non-HDL-CH, but the pattern of reductions still favored lower T-CH-to-HDL-CH ratios. Like T-CH, serum TG levels were also most elevated in hamsters fed HF, with other groups showing significant reductions. Liver CH responses did not conform to responses observed for serum CH and TG levels. Tested in selected baked products prepared in-house, blended fat (BL) was found as functional as regular BS, and it contained no trans fatty acids.  相似文献

Policosanol is a group of long chain primary alcohols and has been shown to reduce blood cholesterol levels and to inhibit the oxidation of low-density lipoprotein (LDL). The present study examined (i) the effect of policosanol supplementation in the diet on the fecal excretion of neutral and acidic sterols in hamsters and (ii) the antioxidant activity of policosanol in human LDL. Golden Syrian hamsters were divided into four groups (n = 12/each) fed one of the four diets containing 0 (control), 0.38, 0.75, and 1.50 g kg(-1) policosanol for 6 weeks. It was found that hamsters given 0.38-1.5 g kg(-1) diets had a serum total cholesterol level lowered by 15-25% and had a high-density lipoprotein cholesterol elevated by 7-16.8%. It was found that policosanol increased the excretion of acidic sterols by 25-73%. Contrary to that in previous reports, policosanol had no apparent anti-LDL oxidation activity when 1-tetracosanol, 1-hexacosanol, and 1-octacosanol were incubated in human LDL. Policosanol also possessed no scavenging activity on the free radical2,2-diphenyl-1-picrylhydrazyl. These data provide evidence that in addition to the effect of HMG-CoA reductase, the cholesterol-lowering activity of policosanol is partially mediated by its inhibition on the absorption of bile acids, but these data disprove the claim that policosanol is an antioxidant.  相似文献

Obesity plays a critical role in dyslipidemia and related disorders. Mulberry water extracts (MWEs) contain polyphenols, including gallic acid, chlorogenic acid, rutin, and anthocyanins. In this study, using 6-week-old male hamsters, we investigated the anti-obese effect of MWEs. After 12 weeks of treatment, MWEs lowered high-fat diet (HFD)-induced body weight and visceral fat, accompanied with hypolipidemic effects by reducing serum triacylglycerol, cholesterol, free fatty acid, and the low-density lipoprotein (LDL)/high-density lipoprotein (HDL) ratio (n=8 for each group). MWEs decreased hepatic lipids, thus protected livers from impairment. The hepatic peroxisome proliferator-activated receptor α and carnitine palmitoyltransferase-1 were elevated, while fatty acid synthase and 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase were reduced by MWEs, indicating that MWEs regulated lipogenesis and lipolysis, which exerted the anti-obese and hypolipidemic effects. Noticeably, MWEs showed both efficacy and safety in vivo. In concluson, MWEs can be used to reduce body weight, serum, and liver lipids.  相似文献

Phytoestrogens have been implicated as promising therapeutic agents to treat the vascular impairment seen in menopausal women. The present study investigated the long-term effects of phytoestrogens from Curcuma comosa Roxb. on vascular relaxation of isolated thoracic aorta from ovariectomized (OVX) rats. Treatment of OVX rats for 12 weeks with C. comosa powder, hexane extract, and a novel phytoestrogen, diarylheptanoid-D3, [(3R)-1,7-diphenyl-(4E,6E)-4,6-heptadien-3-ol] prevented impairment of the endothelium-dependent relaxation response to acetylcholine in OVX, but not the endothelium-denude aortic ring relaxation in response to sodium nitroprusside. These data suggest that the vascular relaxation effect of C. comosa is mediated via endothelial cells. Treatment with D3 also increased endothelial nitric oxide synthase (eNOS) and estrogen receptor-α (ERα) protein expression in the aorta of OVX rats and suppressed elevated tumor necrosis factor-α (TNF-α) expression in OVX aortic rings. These results indicate that C. comosa treatment prevents impairment of vascular relaxation in estrogen-deficient animals via the ER-eNOS pathway as well as through its ability to promote an anti-inflammatory response.  相似文献
