Incidence of seroconversion to caprine arthritis-encephalitis virus (CAEV) was determined for 1,194 goats on 11 dairies, using 2 repeated annual herd tests for CAEV. Current life table methods were used to compare age-specific incidence of seroconversion for pasteurized milk-raised and unpasteurized milk-raised goats. Logistic regression models were used to determine the risk factors associated with CAEV seroconversion, and to estimate odds ratios for seroconversion for various factor levels. Goats raised by unpasteurized milk-feeding methods were 2.5 to 6.7 times more likely to seroconvert than were goats raised by pasteurized milk-feeding methods, depending on the method of comparison. Similarly, 61.6 to 85.0% of seroconversions in yearling goats possibly were attributable to unpasteurized milk feeding. Among yearling goats, CAEV seroconversion was associated with feeding method, breed, and source of goat (herd of origin) when the effect of dairy was considered. In addition to the 6.7 times greater risk of seroconversion for unpasteurized milk-raised goats, yearling goats of the Saanen and Toggenburg breeds were 2.2 and 3.3 times, respectively, more likely to seroconvert than were Alpine yearling goats. Yearling goats purchased from another source were less likely to seroconvert than were yearlings raised on the dairy where they were studied. Among goats > 1 year old, age was associated with risk of seroconversion. Goats that were 3 years old or were > or = 4 years old were 1.7 and 3.2 times, respectively, more likely to seroconvert than were 2-year-old goats, when adjusted for effect of dairy. The effects of dairy were significant (P < or = 0.001) in yearling and older goats.  相似文献

Log-linear methodology was used to examine relations among caprine arthritis-encephalitis virus (CAEV) seroreactivity and host/management factors in a cross-sectional study of 2,826 goats on 13 California dairies. The CAEV serologic status was associated with age and feeding method (pasteurized/unpasteurized milk), but not with breed. Data from a prevalence survey of 321 goats from 2 additional dairies demonstrated very good fit of the selected log-linear model (P = 1.00), indicating that the model was very appropriate to describe the relations. Odds of seropositivity and odds ratios were generated by use of a logit model derived from the log-linear model. Goats raised by the unpasteurized feeding method were estimated to have been 3.3 times more likely to be CAEV-seropositive than goats fed by the pasteurized method, when adjusted for the effects of age. Goats aged 2, 3, 4, and 5 or greater years were estimated to have been 1.7, 2.6, 4.5, and 5.7 times, respectively, more likely to be CAEV-seropositive than were yearling goats when ratios were adjusted for pasteurization status. Breed, gender, and herd of origin were not associated with CAEV seroreactivity when the effects of other factors were considered. Estimated odds of CAEV seroreactivity and the associated odds ratios for combinations of factor levels are reported. In this study, the magnitude and direction of the associations among CAEV serologic status, age, and pasteurized feeding methods were demonstrated.  相似文献

An investigation was conducted to determine if depriving goat kids of colostrum and rearing them away from the herd would prevent transmission of caprine retrovirus infection. Twenty-four newborn goat kids were deprived of colostrum and reared on cow's milk away from their dams from an endemically infected goat herd. Twenty-three colostrum-deprived kids had no evidence of retrovirus infection at birth. One kid had sucked briefly and obtained some colostrum resulting in passive transfer of antibody but it did not develop evidence of infection. Nineteen showed no serological evidence of infection over the 370 days of the study. One colostrum-deprived, segregated goat was subsequently found to be infected and developed arthritis-synovitis. Three had doubtful positive response in one or 2 serological tests during the period but no evidence of infection in leucocyte co-cultures. Cells centrifuged from colostrum of infected goats were co-cultivated with foetal goat synovial membrane cultures. Caprine retrovirus was isolated from cells in the colostrum from the 3 goats examined.  相似文献

A prospective observational cohort study of 361 dairy goat kids was conducted to compare 2 methods of controlling caprine arthritis-encephalitis virus infection under commercial dairy conditions. To compare effectiveness of feeding kids pasteurized milk vs serologic testing and segregation in addition to pasteurized milk feeding, goats were monitored up to the age of 30 months by use of monthly agar gel immunodiffusion testing. Survival analysis methods were used to determine whether age at seroconversion differed between the 2 groups. Significantly lower rates of seroconversion were observed in the segregated group (P < 0.001), compared with the nonsegregated group. Of 193 goats in the pasteurized milk-only group, 146 (75.6%) seroconverted within the 30-month study period, whereas infection was detected in 39 (23.2%) of 168 goats in the test/segregated group. Nonsegregated goats were 3.37 times more likely to seroconvert by 24 months of age, and 70.3% of seroconversions by 24 months of age could be attributed to nonsegregation. For age-specific intervals beyond 180 days of age, 70 to 100% of seroconversions could be attributed to lack of segregation. Cohort life tables for age at seroconversion were reported for each group. Type of colostrum fed, sex, and weaning group (season) were not significantly associated with age at seroconversion. Saanen goats had lower age-specific risk of seroconversion in the nonsegregated group alone and overall. Non-Saanen goats wee 1.5 times more likely to seroconvert than were Saanen goats, when adjusted for a possible confounding effect of weaning group. Results indicate that pasteurized milk feeding and routine test and segregation would be a substantially more effective means of control of the disease in dairy goat herds than would pasteurized milk feeding alone.  相似文献

Somatic cell counts, the bacteriological condition of the milk and antibodies against caprine arthritis encephalitis virus (CAEV) were measured monthly throughout lactation in 121 lactating goats of the Murcia-Granada breed in four commercial dairy goat herds. The prevalence of bacterial intramammary infection was 5.6 per cent and the prevalence of CAEV infection was 20.6 per cent. An analysis of variance revealed a significant effect of herd, intramammary infection and the interaction between intramammary infection and CAEV on the somatic cell count. In udder halves free of intramammary infection, the somatic cell counts were significantly lower in seronegative goats than in seropositive goats (P<0.05), but the difference was not significant in udder halves persistently infected by bacteria. There was a significant increase in somatic cell counts due to bacterial intramammary infection (P<0.01) in the seronegative goats, but this effect was not present in the seropositive animals.  相似文献

In 30 female goats hyperimmunized with bovine insulin, antibody concentrations in serum were determined by radioimmunoassay. Blood samples were taken from the newborn lambs immediately after birth and during the 12 weeks thereafter. Twin lambs were separated and reared in either a colostrum-fed or a colostrum-deprived group. Bovine insulin was added to the milk rations of colostrum-deprived lambs. At the age of 10 weeks, these kids were given a single dose of 1 IU of insulin in combination with complete Freund's adjuvant. Colostrum from nanny goats was also secured for further investigations. Most of the hyperimmunized adult goats responded with the development of insulin antibodies. Their kids were born without these specific immunoglobulins, but several hours after they were allowed to suckle colostrum, the antibody concentration reached values comparable with the values measured in the nanny's blood. Colostrum-deprived kids did not show an increase in binding capacity. The addition of insulin to the milk rations of bottle-fed kids did not act as a primer in antibody production. Immunoglobulins in blood serum of nanny goats and kids and in colostral whey belong mainly to the IgG class. These results indicate that in goats, maternal insulin antibodies are transferred from nanny to offspring only by colostrum.  相似文献

Goat kids from a herd endemically infected with caprine arthritis encephalitis (CAE) virus were raised according to 3 methods. One group of ten goat kids was removed from infected does at birth before suckling or licking by the doe could occur (snatch birth technique). Kids were fed on goat colostrum, which had been heated to 57 degrees C for ten minutes and then held in a thermos flask for one hour. Subsequently the kids were fed reconstituted spray dried cows' milk powder. They were raised apart from infected goats with separation maintained by a wire fence. Contact occurred across-the-fence. Passively acquired serum antibody to CAE virus was detected in some kids at two to three months of age. Nine of the ten goats were negative for serum antibody to CAE virus when tested at 5-6, 9 and 12 months of age. One goat was positive at three and nine months of age but was negative when tested at 12 months of age. A second group of four kids was removed at birth and fed heat-treated goat colostrum, followed by milk from CAE virus-infected does. All four kids became infected with CAE virus; they developed serum antibody to CAE virus between 5-6 and 9 months of age. A third group of two kids was not removed from their infected dams. The present experiments on goats were conducted with the aim of verifying (a) whether the identity of the goat kids can affect the characteristics of milk let-down during suckling (Study 1) and (b) whether the presence of the goat kids may improve the milk let-down during hand milking (Study 2). In Study 1, 16 lactating goats with twins and 12 with single kids were used. Dams with single goat kids ejected more milk when suckling their kids at Days 5 and 20 of lactation, than when suckling aliens. Dams with twins ejected more milk when suckling their own kids than aliens only at Day 5 of lactation. Taking into account all the dams, they ejected more milk with more fat, protein, and lactose during suckling their own litter than when suckling aliens. In Study 2, when dams (n = 13) were hand milked in the presence of their own litter or in its absence, they ejected more milk with more fat, protein, and lactose than when milked in the presence of alien kids. We conclude that milk let-down in goats can be influenced by the type of stimuli applied or to which they are exposed to during suckling and milking.  相似文献

SUMMARY A study of the prevalence, spread and control of caprine arthritis-encephalitis virus (CAEV) in dairy goat herds in New South Wales (NSW) during 1986–1988 found that 56.8% of 1484 goats in 14 dairy herds were infected with CAEV. The prevalence of CAEV infection within most herds not implementing control measures increased during the study. At the end of the study, 59.7% of 1322 goats were infected. The prevalence of CAEV increased with age. Differences between breeds were less apparent. Within seven herds with a high standard of identification of goats, 149 of 812 goats seroconverted in an ELISA. Of these newly infected goats, 142 (95.3%) were > 1 yr of age and 96 (64.4%) were > 2 yr suggesting lateral spread of the virus. Most of the goats > 2 yr of age had been in the milking herd for a minimum of 3 to 6 months. The high seroconversion rate within the milking herd suggested that factors other than the ingestion of infected colostrum and milk before weaning were important for the spread of CAEV. Observations indicated that behaviour of goats, particularly reproductive behaviour among lactating does, and milking herd management practices are important in the spread of CAEV. A high density of livestock, poor livestock control and contamination of feed, water, equipment and personnel were implicated in transmission. Poorly functioning milking machines may also be involved. CAEV was eradicated from 3 herds by the implementation of strict control measures.  相似文献

Caprine arthritis-encephalitis virus was isolated from goat milk and transmitted most efficiently to kids through both milk and colostrum. In addition, it appeared that transmissions through other secretions of the doe occurred, but were less important than transmission in milk and colostrum. Intrauterine infection may have occurred in 2 of 32 cesarean-derived goats, but postpartum horizontal transmission could not be ruled out. Transmission by the aerosol route was not demonstrated, and even short-term direct contact between virus-infected bucks and virus-free does during breeding did not result in transmission. Prolonged direct contact for over 12 months between weaned cesarean-derived goats and virus-infected goats was necessary before horizontal transmission could be demonstrated under nondairy conditions. However, when uninfected does were milked with infected does, a high percentage became infected in less than 10 months. Heat inactivation (56 C) reduced approximately 10(5) median tissue culture infective doses of caprine arthritis-encephalitis virus to below titratable levels, and virus was not transmitted to kids fed virus-infected colostrum that had been heated at 56 C for 1 hour. A program of eradication is discussed in which kids are removed from their dams at birth, fed safe sources of colostrum and milk, and isolated from other goats until weaning.  相似文献

Sir, — The isolation of caprine arthritis-encephalitis virus (CAEV) from the carpal joint of a yearling goat with chronic interstitial pneumonia is reported in this issue of the Journal.(3) We wish to report on the experimental infection of sheep and goats with this isolate of CAEV. Two 20-month-old Romney lambs (483, 485) and two 2-month-old feral goat kids (43, 45) were inoculated with CAEV suspension (104.0 median tissue culture infective doses [TCID50ml]). Each animal was inoculated with 1.0 ml of virus suspension by each of the following routes: intravenous, intracerebral into the right cerebral hemisphere and intra-articularly into the right radio-carpal joint.(4) An additional control lamb (490) and goat kid (44) were inoculated in identical fashion with cell culture fluid from uninfected goat synovial membrane cells.  相似文献

The objective of this study was to elucidate, whether the impaired insulin sensitivity with regard to glucose utilisation in ruminating goats compared with suckling goat kids may be due to a reduced expression of the glucose transporters GLUT1 and GLUT4 in skeletal muscle. Muscle samples were removed from red, oxidative muscles (M. masseter, diaphragm) and white, glycolytic muscles (M. longissimus dorsi, and M. semitendinosus) of five goat kids fed with milk exchanger and nine adult goats of different stages of life. Samples were analysed for their GLUT1 and GLUT4 contents. The muscles were characterised metabolically by measuring the activities of isocitrate dehydrogenase (ICDH) and of lactate dehydrogenase. In all four analysed muscles the GLUT4 contents of adult goats were significantly (p < 0.05) lower than in goat kids. Significant differences concerning the GLUT4 contents in skeletal muscles were not detected in adult goats of different stages of life. The GLUT1 contents differed to a lower extend between goat kids and adult goats. The results of this investigation indicate that the impaired insulin sensitivity of adult compared with suckling ruminants is accompanied by or leads to a decreased GLUT4 expression.  相似文献

[目的]探讨放养云岭山羊羔羊在哺乳期的生长发育规律。[方法]选取待产母羊24只,共产羔36只(公羔17只,母羔19只),测定0~8周龄羔羊的体重(每周测定1次)和体尺(隔1周测定)。采用Logistic、Gompertz和Von Bertalanffy 3种非线性模型对0~8周龄公羔、母羔的体重进行生长曲线拟合,并对初生体重以及试验末体重进行相关性分析。[结果]采用的3种非线性模型均能够很好地拟合哺乳期云岭山羊羔羊生长曲线,拟合度均大于0.99。Gompertz和Von Bertalanffy模型拟合效果较优,公羔的生长发育曲线拐点时间分别为3.212周和2.862周,拐点体重分别为5.691、5.352 kg;母羔拐点时间分别为1.927周和1.325周,拐点体重分别为4.532、3.960 kg;公、母羔之间拐点周龄和拐点体重存在差异,公羔平均在3.037周龄体重增长速度达到最大,而母羔平均在1.626周龄体重增长速度达到最大。初生重对羔羊试验末体重有极显著(P<0.01)影响。[结论]该研究结果可为今后对哺乳期放养云岭山羊羔羊进行合理补饲提供依据。  相似文献

ABSTRACT: The aim of this study was to determine, using immunofluorescence and in situ hybridization, whether CAEV is capable of infecting goat uterine epithelial cells in vivo. Five CAEV seropositive goats confirmed as infected using double nested polymerase chain reaction (dnPCR) on leucocytes and on vaginal secretions were used as CAEV positive goats. Five CAEV-free goats were used as controls. Samples from the uterine horn were prepared for dnPCR, in situ hybridization, and immunofluorescence. The results from dnPCR confirmed the presence of CAEV proviral DNA in the uterine horn samples of infected goats whereas no CAEV proviral DNA was detected in samples taken from the uninfected control goats. The in situ hybridization probe was complementary to part of the CAEV gag gene and confirmed the presence of CAEV nucleic acids in uterine samples. The positively staining cells were seen concentrated in the mucosa of the lamina propria of uterine sections. Finally, laser confocal analysis of double p28/cytokeratin immunolabelled transverse sections of CAEV infected goat uterus, demonstrated that the virus was localized in glandular and epithelial cells. This study clearly demonstrates that goat uterine epithelial cells are susceptible to CAEV infection in vivo. This finding could help to further our understanding of the epidemiology of CAEV, and in particular the possibility of vertical transmission.  相似文献

Caprine arthritis-encephalitis virus (CAEV) infection of dairy goats was shown by virus isolation and serology to be widespread in South Australia. CAEV was isolated at necropsy from 24 of 27 dairy goats with swollen joints from 13 herds, and from 9 of 30 liver dairy goats in 7 herds. Virus was isolated most frequently from synovial membranes, and occasionally from mammary glands, mammary lymph nodes, choroid plexus, lungs, spleen, bone marrow, salivary glands, leucocytes, synovial fluid and milk. Antibody to CAEV was detected in the serum of 13 of 17 of the necropsied goats tested in a single-line gel diffusion test, and in another 3 retested with a modified double-line technique. Serum antibody was also demonstrated in 61 of 77 dairy goat herds, many with histories of arthritis. In 1984 to 1986 the annual number of serologically positive serums and proportions of the numbers tested were 134 (40%), 121 (45%) and 42 (18%), respectively. CAEV was isolated from leucocytes of 8 live goats in 6 of these herds. In fibre goats antibody was detected in the serum of 25 Angora and 19 crossbreds (0.1%) from the 33,279 Angora, 1,705 Cashmere, 8,715 crossbred and 904 feral goats tested.  相似文献

A lentivirus was isolated from 2 goats in Mexico that were seropositive to caprine arthritis encephalitis virus (CAEV) by the agar gel immunodiffusion (AGID) test. The lentivirus was identified as CAEV by the observation of giant multinucleated cells (syncytia) in goat synovial membrane (GSM) monolayers co-cultivated with blood mononuclear (BMN) cells from the seropositive goats, and by amplifying a DNA segment of the CAEV gag gene using the polymerase chain reaction (PCR) technique. Subsequently, cell supernatants from the GSM cells co-cultivated with BMN cells were used to infect 2 CAEV-seronegative goats. These goats seroconverted to CAEV as determined by the AGID test, and CAEV was re-isolated from these goats. One of the goats developed polyarthritis 8 mo after inoculation. Previous serological surveys indicate that infection with CAEV is prevalent among goats in Mexico. To our knowledge this is the first report of CAEV isolation in Mexico. Because of globalization of markets and increased trading among nations, the rapid identification and reporting of diseases such as CAEV are important to prevent the dissemination of these diseases.  相似文献

The prevalence of infection with caprine arthritis-encephalitis virus was assessed using an indirect ELISA. Antiviral antibodies could be detected in 42% of 5974 goat serum specimens included in this study (Switzerland except Ticino and Graubünden 61%). Infected animals were found in all breeds of goats and in all regions of the country, with the lowest prevalence in the Canton Ticino in southern Switzerland. A comparison of clinical and serological data showed that only 20-30% of the infected animals develop carpitis or mastitis. We did not see any cases of encephalitis in young kids, indicating that this manifestation occurs only infrequently. According to the goat owners, each year, approximately 5-10% of the total goat population are eliminated because of arthritis. The decrease in milk production in infected animals is estimated at 10-15%.  相似文献

Faecal samples from 222 healthy dairy goats on 12 farms in Spain, as well as bulk tank milk samples of these farms, were screened for the presence of verotoxin-producing Escherichia coli (VTEC) and enteropathogenic E. coli (EPEC). VTEC and EPEC were isolated in 47.7 and 7.7% of the animals, respectively. VTEC were isolated more frequently from adults and replacement animals than from goat kids. In contrast, EPEC were detected more frequently from goat kids than from replacement animals and adults. VTEC or EPEC strains were not detected in the bulk tank milk samples. Although a selective enrichment protocol was used, the serotype O157:H7 was not detected. The most frequent serotypes among the 106 VTEC strains isolated from goats were O5:H-, O76:H19, O126:H8, O146:H21, ONT:H- and ONT:H21. None VTEC strain was eae-positive. The absence of the eae gene in the VTEC strains could indicate that these strains are less virulent for humans that the classical eae-positive enterohaemorrhagic E. coli types. However, 16% of VTEC strains isolated from healthy goats belonged to serotypes associated with haemolytic uraemic syndrome in humans. The ehxA gene was detected in 84.9 and 52.9% of the VTEC and EPEC from goats, respectively. The beta1, theta/gamma2 and zeta were the most frequent intimin types among the 17 EPEC strains studied and the most prevalent serotypes of these strains were O156:H25 and O177:H11. Our data show that in Spain healthy goats are an important reservoir of VTEC and EPEC, and a potential source of infection for humans.  相似文献

During epidemic outbreaks in two goatherds clinical symptoms and deaths occurred in five (14%) of the 3-week-old goat kids in farm A, and in six (33%) of those in farm B. In the latter farm, three female goats aborted before the clinical symptoms in the kids emerged. Mycoplasma could be isolated from both healthy and sick goat kids and from female goats, which had diseased kids or had aborted. Three goat kids (one from herd A and two from herd B) were sent for post-mortem examination. In all these cases septicaemia caused by Mycoplasma was diagnosed. Based on the bacteriological examination the Mycoplasma strains proved to be Mycoplasma mycoides subsp. capri (Mmc). This was confirmed by the PCR examination. Mmc was isolated from several locations including from the rectum of one healthy female goat, and from two diseased kids. In addition, bacteria were detected in the small intestine in two of the necropsied kids by bacteriological and/or immunohistochemical methods. The finding suggests that Mmc may be transmitted via faeces in goatherds, kept under conventional conditions.  相似文献

Orf (contagious ecthyma) is a viral disease of small and wild ruminants, humans, and less frequently other species. In sheep and goats, the disease is characterized by the formation of vesiculo-proliferative lesions in the skin of lips and nostril. Here, a form of generalized orf in 16 goat kids from 2 different locations in west Texas is described. The disease was characterized by multifocal, severe, proliferative dermatitis that persisted from about 2 months of age until the goat kids were euthanized 3 months later. All affected goats were Boer or Boer crosses under 1 year of age. The mean immunoglobulin concentration in sera of affected goats was elevated compared with healthy control goats. Severe to moderate lymphadenomegaly of the nodes draining the areas of the skin affected with orf lesions was present in all 16 goat kids. Suppurative arthritis, chronic fibrinous pneumonia, and premature thymic involution were found in 3, 5, and 7 of the goat kids, respectively. The skin lesions of 3 goat kids were infested with larvae of the opportunistic black garbage fly (Ophira sp.). The orf virus was identified in skin lesions by isolation in Marbin-Darby ovine kidney cells, electron microscopy, and amplification of viral DNA by polymerase chain reaction. The orf virus was not detected in peripheral blood or lymph node mononuclear cells of any of the goats. Cross-neutralization experiments showed that an ovine orf virus antiserum raised in sheep was more effective in neutralizing a sheep orf virus isolate than a caprine orf virus isolate. The clinical and epidemiological characteristics of these orf cases may be the result of susceptibility factors within some individuals of the Boer breed of goats.  相似文献
