Tying knots and linking microscopic loops of polymers, macromolecules, or defect lines in complex materials is a challenging task for material scientists. We demonstrate the knotting of microscopic topological defect lines in chiral nematic liquid-crystal colloids into knots and links of arbitrary complexity by using laser tweezers as a micromanipulation tool. All knots and links with up to six crossings, including the Hopf link, the Star of David, and the Borromean rings, are demonstrated, stabilizing colloidal particles into an unusual soft matter. The knots in chiral nematic colloids are classified by the quantized self-linking number, a direct measure of the geometric, or Berry's, phase. Forming arbitrary microscopic knots and links in chiral nematic colloids is a demonstration of how relevant the topology can be for the material engineering of soft matter.  相似文献

Enzymes and synthetic chiral catalysts have found widespread application to produce single enantiomers, but in situ switching of the chiral preference of a catalytic system is very difficult to achieve. Here, we report on a light-driven molecular motor with integrated catalytic functions in which the stepwise change in configuration during a 360° unidirectional rotary cycle governs the catalyst performance both with respect to activity and absolute stereocontrol in an asymmetric transformation. During one full rotary cycle, catalysts are formed that provide either racemic (R,S) or preferentially the R or the S enantiomer of the chiral product of a conjugate addition reaction. This catalytic system demonstrates how different molecular tasks can be performed in a sequential manner, with the sequence controlled by the directionality of a rotary cycle.  相似文献

Mycoplasma-like bodies with helical filaments were seen by phase contrast microscopy in juice expressed from tissues of plants infected with corn stunt agent. Each filament is bounded by a "unit membrane" and no cell wall, sheath, envelope, or second membrane has yet been discerned by electron microscopy. The association of these filaments with development of disease, their occurrence in phloem cells as seen by both freeze-etching and thin-section electron microscopy, the diagnosis of infection based on their presence in plants without symptoms, and their absence in noninfected corn are consistent with the hypothesis that these unusual filaments are formed by the mycoplasma-like organism presumed to be the corn stunt agent.  相似文献

The Stokes-Einstein relation relates the diffusion coefficient of a spherical Brownian particle in a viscous fluid to its friction coefficient. For a particle suspended in anisotropic liquid, theory predicts that the drag coefficient should also be anisotropic. Using video microscopy coupled with particle tracking routines, the Brownian fluctuations of micrometer-sized particles were analyzed to yield a quantitative measurement of the diffusion coefficients parallel and perpendicular to the nematic director. The experimental values agree quite well with recent numerical calculations that take into account the distortions of the director field in the vicinity of the particles.  相似文献

Coherently moving flocks of birds, beasts, or bacteria are examples of living matter with spontaneous orientational order. How do these systems differ from thermal equilibrium systems with such liquid crystalline order? Working with a fluidized monolayer of macroscopic rods in the nematic liquid crystalline phase, we find giant number fluctuations consistent with a standard deviation growing linearly with the mean, in contrast to any situation where the central limit theorem applies. These fluctuations are long-lived, decaying only as a logarithmic function of time. This shows that flocking, coherent motion, and large-scale inhomogeneity can appear in a system in which particles do not communicate except by contact.  相似文献

Alkanes adopt extended conformations in solution that minimize steric interactions and maximize surface area. Folding can reduce the amount of hydrophobic surface exposed to solvent, but sterically unfavorable gauche interactions result. However, we found that the alkyl chains of two common surfactants in aqueous solution adopt helical conformations when bound within a synthetic receptor. The receptor recognizes the helical alkane better than the extended conformation, even though 2 to 3 kilocalories per mole of strain is introduced. The proper filling of space and burial of hydrophobic surface drive the molecular recognition between the receptor and the coiled alkane.  相似文献

Fluidization of magnetizable particles by a gas stream in the presence of a uniform applied magnetic field oriented parallel to the flow prevents the hydrodynamic instability that otherwise leads to bubbles and turbulent motion within the medium. The fluidized emulsion expands uniformly in response to gas flow speeds in excess of that at the incipient fluidization point, with transition from the quiescent stable state to bubbling occurring suddenly at a characteristic increased rate of flow. Experimental data demonstrate the dependence of this transition velocity on the intensity of the applied magnetic field, length of the bed, and type of magnetic solids. Data illustrate the pressure distribution through the bed medium, the bedflow characteristics, and other phenomena.  相似文献

Psilocybin, a hallucinogen, formed a blue color with a subfraction of rat-brain mitochondria believed to contain nerve-ending particles. Color formation increased with pH, did not require oxygen, and involved a component that could not be solubilized. The effect was not shown by chemically related neuroactive compounds, such as bufotenine and serotonin, and was antagonized by only tyramine or ethylenediaminetetraacetic acid.  相似文献

The low-density lipoprotein receptor (LDL-R) is a typical example of a multidomain protein, for which in vivo folding is assumed to occur vectorially from the amino terminus to the carboxyl terminus. Using a pulse-chase approach in intact cells, we found instead that newly synthesized LDL-R molecules folded by way of "collapsed" intermediates that contained non-native disulfide bonds between distant cysteines. The most amino-terminal domain acquired its native conformation late in folding instead of during synthesis. Thus, productive LDL-R folding in a cell is not vectorial but is mostly posttranslational, and involves transient long-range non-native disulfide bonds that are isomerized into native short-range cysteine pairs.  相似文献

The stabilization of long-term memories requires de novo protein synthesis. How can proteins, synthesized in the soma, act on specific synapses that participate in a given memory? We studied the dynamics of newly synthesized AMPA-type glutamate receptors (AMPARs) induced with learning using transgenic mice expressing the GluR1 subunit fused to green fluorescent protein (GFP-GluR1) under control of the c-fos promoter. We found learning-associated recruitment of newly synthesized GFP-GluR1 selectively to mushroom-type spines in adult hippocampal CA1 neurons 24 hours after fear conditioning. Our results are consistent with a "synaptic tagging" model to allow activated synapses to subsequently capture newly synthesized receptor and also demonstrate a critical functional distinction in the mushroom spines with learning.  相似文献

Anisotropic particles suspended in a nematic liquid crystal disturb the alignment of the liquid crystal molecules and experience small forces that depend on the particles' orientation. We have measured these forces using magnetic nanowires. The torque on a wire and its orientation-dependent repulsion from a flat surface are quantitatively consistent with theoretical predictions based on the elastic properties of the liquid crystal. These forces can also be used to manipulate submicrometer-scale particles. We show that controlled spatial variations in the liquid crystal's alignment convert the torque on a wire to a translational force that levitates the wire to a specified height.  相似文献

The cell type responsible for inhibition by antigen of migration in vitro of peritoneal exudate cells obtained from tuberculin-hypersensitive guinea pigs was studied. Exudate populations were separated into component cell types, the lymphocyte and the macrophage. Peritoneal lymphocytes from sensitive donors were the immunologically active cells in this system, the macrophages, being merely indicator cells which migrate. Sensitized peritoneal lymphocyte populations, upon interaction with specific antigen in vitro, elaborated into the medium a soluble material capable of inhibiting migration of normal exudate cells.  相似文献
