The aim of this study was to determine the effects of pre‐slaughter transport during summer and subsequent water shower spray on stress, postmortem glycolysis, energy metabolism and adenosine monophosphate‐activated protein kinase (AMPK) in Pectoralis major (PM) muscle of broilers. Results indicated that transport during high ambient temperature induced the release of plasma corticosterone, which significantly affected stress conditions. Moreover, we found a lower energy status in the early postmortem period compared to the control group. AMPK was activated in this situation, following by the rapid glycolysis and accumulation of lactic acid, leading to a high incidence of pale, soft, exudative (PSE)‐like meat. Water shower spray with resting after transport relieved the stress situation, recovered energy homeostasis and lessened the deterioration of meat quality. As a key molecular target for the control of energy status, AMPK has a similar potential in regulating postmortem muscle glycolysis of broilers as in mammals.  相似文献

Effects of water‐misting sprays with forced ventilation on post mortem glycolysis, adenosine monophosphate‐activated protein kinase (AMPK) and meat quality of broilers after transport during summer were investigated in the present paper. A total of 105 mixed‐sex Arbor Acres broilers were divided into three treatment groups: (i) 45 min transport without rest (T); (ii) 45 min transport with 1 h rest (TR); and (iii) 45 min transport with 15 min water‐misting sprays with forced ventilation and 45 min rest (TWFR). Each treatment consisted of five replicates with seven birds each. The results indicated that the water‐misting sprays with forced ventilation could mitigate the stress caused by transport under high temperature conditions during summer, which reduced the energy depletion in post mortem Pectoralis major (PM) muscle. This resulted in a higher energy status compared to the T group, which would decrease the expression of phosphorylation of AMPK (p‐AMPK). Furthermore, decreased the expression of p‐AMPK then slowed down the rate of glycolysis in post mortem PM muscle during the early post mortem period, which in turn lessened the negative effects caused by transport on meat quality. In conclusion, water‐misting sprays with forced ventilation may be a better method to control the incidence of the pale, soft and exudative meat in broilers.  相似文献

ABSTRACT: The objective of this study was to investigate the response of sarcoplasmic proteins in bovine LM to low-voltage electrical stimulation (ES; 80 V, 35 s) after dressing and its contribution to meat tenderization at an early postmortem time. Proteome analysis showed that ES resulted in decreased (P < 0.05) phosphorylation of creatine kinase M chain, fructose bisphosphate aldolase C-A, β-enolase, and pyruvate kinase at 3 h postmortem. Zymography indicated an earlier (P < 0.05) activation of μ-calpain in ES muscles. Free lysosomal cathepsin B and L activity increased faster (P < 0.05) in ES muscles up to 24 h. Immunohistochemistry and transmission electron microscopy further indicated that lysosomal enzymes were released at an early postmortem time. Electrical stimulation also induced ultrastructural disruption of sarcomeres. In addition, ES accelerated (P < 0.05) the depletion of ATP, creatine phosphate, and glycogen, as well as a pH decline and the more preferred pH/temperature decline mode. Finally, ES accelerated meat tenderization, resulting in lesser (P < 0.05) shear force values than the control over the testing time. A possible relationship was suggested between a change in the phosphorylation of energy metabolic enzymes and the postmortem tenderization of beef. Our results suggested the possible importance of the activation of μ-calpain, phosphorylation of sarcoplasmic proteins, and release of lysosomal enzymes for ES-induced tenderization of beef muscle.  相似文献

Washed red blood cells from normal cows were incubated as 10 and 20 per cent suspensions in media containing 0, 2.5 and 25 mM phosphate. The results showed that the rate of glycolysis was dependent on the inorganic phosphate concentration. In the absence of phosphate, the consumption of glucose and the production of 2,3-diphosphoglycerate, lactate and adenosine 5'-triphosphate (ATP) were decreased. Incubation without added phosphate also greatly increased the production of fructose-1,6-diphosphate, glyceraldehyde-3-phosphate and dihydroxyacetone phosphate. Moderate hypophosphataemia was induced in two pre-ruminant calves. Washed red cells from the blood of these animals showed a depletion of ATP when compared with red cells from a control calf. The results indicate that phosphorus deficiency, leading to hypophosphataemia, may be a mechanism of post parturient and related syndromes of haemoglobinuria by decreasing red cell glycolysis and resultant ATP synthesis. Subnormal concentrations of ATP would predispose red cells to altered structure and function, a loss of normal deformability, and an increase in fragility and haemolysis with resultant haemoglobinuria.  相似文献

The effects of alpha-lipoic acid (ALA) on the growth, body composition, postmortem AMP-activated protein kinase (AMPK) activation, and 24-h muscle pH were investigated. Thirty male C57BL/6J mice were fed diets containing 0, 0.5, or 1.0% ALA (DM basis). At the end of the 3-wk feeding trial, carcass weights decreased (P < 0.05) 14 and 30% for mice fed 0.5 and 1.0% ALA, respectively, compared with the 0% group, with decreases in BW as the levels of dietary ALA increased. This change in carcass weight occurred because carcass fat content for mice receiving 0.5 and 1.0% ALA was 7.32 and 8.09% lower (P < 0.05), respectively, than for the 0% ALA treatment, and because gonadal fat decreased (P < 0.05) 85% in mice fed 1.0% ALA compared with those fed 0% ALA. Dietary ALA caused a slight increase (P < 0.05) in carcass moisture content, with no (P = 0.07) effect on protein and ash content. Furthermore, ALA supplement decreased (P < 0.05) ADFI (DM basis) from 4.3 g/d for 0% ALA-fed mice to 3.4 g/d for 1.0% ALA-fed mice. At 20 min postmortem, pH was greater (P < 0.05) in muscle of mice fed 1.0% ALA than in muscle of mice fed 0% ALA. Ultimate (24-h) pH values differed (P < 0.05) among treatments, and mean values were 5.83, 6.08, and 6.29 for 0, 0.5, and 1.0% ALA, respectively. Phosphorylation of AMPK alpha subunit at Thr172, an indicator of AMPK activation, was decreased (P < 0.05) in muscle of ALA-treated mice at 20 min postmortem. Because AMPK has a crucial role in the control of glycolysis, the reduction in AMPK activation decreases glycolysis, and thereby increases the ultimate pH of postmortem muscle. In summary, dietary ALA supplement can decrease fat accumulation in mice, and because ALA increased muscle pH at 20 min and 24 h postmortem, these results suggest that dietary ALA supplementation might decrease carcass fatness and prevent the development of PSE pork and poultry. However, further research is required to test the effects of ALA in swine and poultry.  相似文献

:对 3 0 1头瘦肉型新品系苏钟 系和 系仔猪进行氟烷测验 ,阳性率为 1 .3 3 %。随机抽取氟烷阴性猪 3 4头进行基因型检测 ,有 6头为杂合子 ,由此得知 NN、Nn和 nn3种基因型的频率分别为 81 .2 6%、1 7.4 1 %和 1 .3 3 %,N和 n2种基因的频率分别为 89.97%和 1 0 .0 3 %。对 3种基因型猪的肉质进行评定结果显示 ,4头氟烷阳性猪 ( nn)肉质最差 ,出现 3头严重 PSE肉和 1头轻度 PSE肉 ,而氟烷阴性猪 ( NN或 Nn)未发生 PSE肉。2 0头苏钟 系氟烷阴性猪用阳性公猪测交 ,对产出的 2 0 8头后代进行氟烷测验 ,共检出 4窝中的 2 1头阳性猪。对测交后代进行肥育性能和肉质测定 ,发现阳性猪的日增重和饲料报酬等较低 ,屠宰率、眼肌面积和瘦肉率较高 ,肉质较差  相似文献

一磷酸腺苷激活的蛋白激酶(AMPK)广泛存在于真核细胞中,AMPK的活性受一磷酸腺苷(AMP)/三磷酸腺苷(ATP)的调节,应激反应可通过ATP的产生减少或消耗使细胞内AM P/ATP增加,从而激活AMPK。激活的AMPK可通过磷酸化下游靶蛋白,改变脂类和碳水化合物代谢,使其朝着抑制ATP消耗过程、促进ATP生成反应的方向进行,即抑制脂肪酸和糖原合成,促进脂肪酸氧化分解和葡萄糖的吸收,从而迅速恢复细胞中的能量,因此AMPK被称为"细胞能量调节器",在动物适应环境的过程中发挥着重要的作用。本文在国内外已有文献报道基础上,对AMPK的结构、分布、活性调节及对糖脂代谢的调节作用进行综述。  相似文献

Glycolytic potential of red, soft, exudative pork longissimus muscle   总被引:7,自引:0,他引:7  
Red, soft, exudative (RSE) pork is red as "normal" pork and exudative as PSE pork. The origin of RSE quality is unknown. In the present study, we determined whether the occurrence of RSE pork is related to the presence of the RN (Rendement Napole) gene. A glycolytic potential [= 2 x (glucose + glycogen + glucose-6-phosphate) + lactate] of > 180 micromol lactate/g of meat was used as indicator for presence of the RN gene. At 1 d postmortem, pork loin samples were collected at a commercial slaughter plant, and meat quality characteristics and glycolytic potential were determined. Glycolytic potential of RSE pork (n = 42) was higher (P < .01) than that of RFN (red, firm, nonexudative) pork (n = 25): 137 vs 110 micromol lactate/g of muscle. The glycolytic potential of PSE pork (n = 28) was 161 micromol lactate/g of muscle, which was higher (P < .01) than the glycolytic potential of RFN or RSE pork. Using a glycolytic potential of 180 micromol lactate/g of muscle as suggestive for the presence of the RN gene, four (10%) of the RSE samples were from RN carriers. By the same criterion, the PSE group contained nine (32%) RN gene carriers. These results suggest that the occurrence of RSE quality is not related to the presence of the RN gene.  相似文献

The purpose of the paper is to describe research carried out in Denmark in areas associated with PSE.In the carcass, meat colour is used as an expression of meat quality. Meat quality deteriorated until 1972 but has improved substantially during the last 6–7 years.The incidence of halothane susceptibility in the Danish Landrace breed is low. Approximately 60% of the halothane-susceptible pigs had PSE carcasses compared to 22% for the non-susceptible pigs. However, pigs heterozygous at the Hal locus are intermediate in meat quality, and elimination of the gene for halothane susceptibility will lead to a substantial improvement in meat quality. A proportion of halothane-susceptible pigs as well as heterozygous carriers can be detected indirectly by using the H blood group system alone or in combination with the PHI system. This is due to linkage disequilibrium between the three systems within the Danish Landrace breed. Use of the H and PHI systems and the halothane test in combination with full sib information on KK index may increase the genetic gain by 5–30%. However, a greater extra gain is easily obtained by a slight increase in selection intensity based on the KK index alone. Thus, it does not seem profitable to use blood and enzyme types or halothane test as a routine in selection against PSE under Danish conditions. In contrast, these tests are useful within herds with a high frequency of PSE.  相似文献

The purpose of the present study was to investigate the variations in histochemical characteristics of muscle samples segregated according to metabolic rates (MR) and pork quality attributes. A total of 231 crossbred Duroc x (Yorkshire x Landrace) pigs was evaluated. Samples of the LM were taken to evaluate histochemical characteristics, postmortem MR, and meat quality. Samples were classified into fast, normal, and slow MR groups based on muscle pH at 45 min and R-value. Drip loss and lightness (L*) were used to assign samples to 1 of 4 quality classes. Pale, soft, and exudative pork belonging in the fast group had the greatest (P < 0.05) percentage of type IIb fibers, and RSE (reddish-pink, soft, and exudative) pork belonging in the fast group had a similar tendency. Additionally, RFN (reddish-pink, firm, and nonexudative) pork belonging in the normal group showed a lower (P < 0.05) percentage of type IIb fibers than PSE or RSE, regardless of MR, and DFD pork had the lowest (P < 0.05) percentage of type IIb fibers. In general, the fast-glycolyzing PSE pork with the lowest pH at both 45 min and 24 h had greater percentages of type IIb fibers than the fast-glycolyzing RFN pork. There were more fiber-type composition differences between quality classes in pork undergoing a fast rate of metabolism compared with pork undergoing a normal rate of metabolism. It can be concluded that muscle histochemical characteristics are associated with early postmortem MR, the extent of glycolysis, and, thereby, pork quality; however, these effects are limited to the pigs exhibiting a fast glycolytic rate.  相似文献

Crossbred pigs, heterozygous for the halothane gene, were used to determine the effects of long-term dietary supplementation of magnesium mica (MM) and short-duration transportation stress on performance, stress response, postmortem metabolism, and pork quality. Pigs were blocked by weight, penned in groups (six pigs per pen), and pens (three pens per diet) were assigned randomly either to a control corn-soybean meal diet or the control diet supplemented with 2.5% MM (as-fed basis; supplemented at the expense of corn). Diets were fed during the early-finisher (0.95% lysine, as-fed basis; 43.7 to 68 kg) and late-finisher (0.85% lysine, as-fed basis; 68 to 103 kg) periods. At the conclusion of the 71-d feeding trial, 12 pigs from each dietary treatment were selected randomly and subjected either to no stress (NS) or 3 h of transportation stress (TS). Dietary MM had no effect (P > or = 0.40) on ADG or ADFI; however, G:F was improved (P < 0.05) during the early-finisher period when pigs were fed MM-supplemented diets. Plasma glucose concentrations were increased in TS pigs fed the control diet, but transportation did not affect plasma glucose in pigs fed 2.5% MM (diet x transportation stress; P = 0.02). Dietary MM did not affect blood lactate, cortisol, insulin, NEFA, Ca, or Mg concentrations in response to TS (diet x transportation stress; P > or = 0.13); however, circulating lactate, cortisol, and glucose concentrations increased in TS pigs (transportation stress x time; P < 0.01). The LM from TS pigs fed MM had higher initial (0-min) and 45-min pH values than the LM from NS pigs fed the control diet (diet x transportation stress x time; P = 0.07). Lactic acid concentration and glycolytic potential were greater in the LM of TS pigs fed MM than TS pigs fed control diets (diet x transportation stress; P < or = 0.01). Although some trends were identified, neither MM (P > or = 0.15) nor TS (P > or = 0.11) altered the color or water-holding capacity of the LM and semimembranosus. The transportation model elicited the expected changes in endocrine and blood metabolites, but dietary MM did not alter the stress response in pigs. Conversely, although pork quality traits were not improved by dietary MM, delaying postmortem glycolysis and elevating 0- and 45-min muscle pH by feeding finishing diets fortified with MM may benefit the pork industry by decreasing the incidence of PSE pork in pigs subjected to short-duration, routine stressors.  相似文献

The objective of this study was to determine the effects of oral administration of sodium citrate (CIT) or acetate (ACE) to pigs on blood parameters, postmortem glycolysis, pH decline, and quality attributes of pork. Previous studies have shown that CIT has the potential to inhibit phosphofructokinase (PFK), a key enzyme in postmortem muscle glycolysis. In Exp. 1, CIT, ACE, or water was orally administered (0.75 g/kg of BW) to 24 pigs. After a 30-min rest, pigs were exercised, and blood samples were taken at 45 and 75 min after oral treatment. Citrate and ACE tended (P = 0.08) to increase blood pH and increased (P = 0.02) bicarbonate levels immediately after exercise. After a 30-min rest, blood pH of pigs administered ACE tended (P = 0.09) to remain higher, whereas blood pH of CIT-treated pigs was similar to that of control pigs. Bicarbonate levels in ACE- and CIT-treated pigs were still greater (P < 0.05) than those of control pigs at 75 min after oral treatment. In Exp. 2, 30 pigs were administered CIT, ACE, or water 45 min before stunning (electric plus captive bolt). Antemortem treatments had no effect (P > 0.10) on muscle pH or postmortem concentrations of the glycolytic metabolites of glucose-6 phosphate, fructose-6 phosphate, fructose-1,6 bisphosphate, glyceraldehyde-3 phosphate, dihydroxyacetone phosphate, or lactate. Minor, but inconsistent, differences in quality attributes were found in LM chops, and no differences in quality attributes were found between control and CIT- or ACE-treated pigs for inside and outside semimembranosus muscles (P > 0.10). There was no significant inhibition of the PFK enzyme by orally administered CIT or ACE; however, the PFK glycolytic metabolite data analysis indicated that PFK was a main regulatory enzyme in postmortem muscle.  相似文献

Weaning stress may cause reduced energy intake for maintenance of mucosal structure.Gln,Glu,and Asp are major energy sources for the small intestine.This study investigated whether Gln,Glu,and Asp improve the intestinal morphology via regulating the energy metabolism in weaning piglets.A total of 198 weaned piglets were assigned to 3 treatments:Control(Basal diet+1.59%L-Ala);T1(Basal diet+1%L-Gln+0.5%L-Glu+0.1%L-Asp);T2(Low energy diet+1%L-Gln+0.5%L-Glu+0.1%L-Asp).Jejunum and ileum were obtained on d 5 or 21 post-weaning.T1 enhanced growth performance.T1 and T2 treatments improved small intestinal morphology by increasing villus height,goblet cell number and decreasing crypt depth.Days post-weaning affected the efficacy of T2,but not T1,on energy metabolism.At normal energy supplementation,Gln,Glu,and Asp restored small intestinal energy homeostasis via replenishing the Krebs'cycle and down-regulating the AMPK(adenosine monophosphate activated protein kinase)pathway.As these are not sufficient to maintain the intestinal energy-balance of piglets fed with a low energy diet on d 5 post-weaning,the AMPK,glycolysis,beta-oxidation,and mitochondrial biogenesis are activated to meet the high energy demand of enterocytes.These data indicated that Gln,Glu,and Asp could restore the energy homeostasis of intestinal mucosa of weaning piglets under normal energy fed.Low energy feeding may increase the susceptibility of piglets to stress,which may decrease the efficacy of Gln,Glu,and Asp on the restoration of energy balance.These findings provide new information on nutritional intervention for insufficient energy intake in weaning piglets.  相似文献

Depression induces anorexia, leading to suppressed feeding behaviors and energy intake. Previously, we revealed that chronic social defeat induced a mild suppression of feeding in rats with elevated levels of hypothalamic malonyl‐CoA which regulates feeding. Therefore, we attempted to elucidate the effects of chronic mild food restriction on behavior and on hypothalamic malonyl‐CoA. The chronic mild food restricted rats were fed a restricted diet approximately 80% to 90% amount of diet compared to the control for 5 weeks. Ratios of restriction were adjusted with feed consumption in the chronic social defeat stressed rats. Chronic mild food restricted rats exhibited a suppression of body weight gain similar to that of the chronic social defeat stressed rats. Also these rats showed increased time spent in the center area of an open field (OF), prolonged immobility time in forced swim, increased phosphorylation of hypothalamic adenosine monophosphate‐activated protein kinase (AMPK) and acetyl‐CoA carboxylase and a decreased concentration of hypothalamic malonyl‐CoA. Weight of the adrenal glands, locomotion in an OF, mitogen‐activated protein kinase cascade and calcium/calmodulin‐dependent protein kinases II in the hippocampus were not affected by chronic mild food restriction. Our findings suggest that chronic mild food restriction activates AMPK following a decreased hypothalamic malonyl‐CoA.  相似文献

The objective of this study was to compare carcass characteristics of a newly introduced breed, the Waguli (Wagyu x Tuli), with the carcass characteristics of the Brahman breed. Brahman cattle are used extensively in the Southwest of the United States because of their tolerance to adverse environmental conditions. However, Brahman carcasses are discounted according to the height of their humps because of meat tenderness issues. The Waguli was developed in an attempt to obtain a breed that retained the heat tolerance of the Brahman but had meat quality attributes similar to the Wagyu. Twenty-four animals were used. Six steers from each breed were fed a 94% concentrate diet and 6 steers from each breed were fed an 86% concentrate diet. Eight steers, 2 from each group, were harvested after 128 d, after 142 d, and after 156 d on feed. Waguli steers had larger LM, greater backfat thickness, greater marbling scores, and greater quality grades than the Brahman steers (P < 0.05). The Japanese Wagyu breed is well known for its highly marbled and tender meat, and these traits are also present in the Waguli. The Waguli had significantly lower Warner-Bratzler shear force values than the Brahman steers after 7 and 10 d of postmortem aging (P < 0.05); this difference decreased after 14 d postmortem (P = 0.2), when tenderness of the slower aging Brahman had increased to acceptable levels. Toughness of the Brahman has been associated with high levels of calpastatin in Brahman muscle, and the Waguli LM had significantly less calpastatin activity (P = 0.02) at 0 h postmortem than the Brahman LM. At 0-h postmortem, the total LM calpain activity did not differ between the Brahman and Waguli (P = 0.57). Neither diet nor days on feed had any significant effect on the 0-h postmortem calpain or at 0-h postmortem calpastatin activity, nor an effect on Warner-Bratzler shear-force values. In conclusion, LM muscle from the Waguli steers had a high degree of marbling, lower shear force values, and low calpastatin activity, all of which are related to more tender meat.  相似文献

The objective of the present experiment was to investigate the effects of transportation, lairage, and preslaughter stressor treatment on glycolytic potential and pork quality of the glycolytic longissimus and the oxidative supraspinatus (SSP) or serratus ventralis (SV) muscles. In a 2 x 2 x 2 factorial design, 384 pigs were assigned randomly either to short (50 min) and smooth or long (3 h) and rough transport, long (3 h) or short (< 45 min) lairage, and minimal or high preslaughter stress. Muscle samples were taken from the LM at 135 min and from the SSP at 160 min postmortem for determination of the glycolytic potential and rate of glycolysis. At 23 h postmortem, pork quality was assessed in the LM and the SV. Effects of transport and lairage conditions were similar in both muscle types. Long transport increased (P < 0.01) the glycolytic potential and muscle lactate concentrations compared with short transport. Both long transportation and short lairage decreased (P < 0.01) redness (a* values) and yellowness (b* values) of the LM and SV. In combination with short lairage, long transport decreased (P < 0.05) pork lightness (lower L* values), and electrical conductivity was increased (P < 0.05) after long transport. Several interactions between stress level and muscle type (P < 0.001) were observed. High preslaughter stress decreased (P < 0.001) muscle glycogen in both the LM and SSP, but this decrease was greater in the LM. Lactate concentrations were increased (P < 0.001) only in the LM by high preslaughter stress. Increases in ultimate pH (P < 0.001) and decreases in a* values (P < 0.01) were greatest in the SV, whereas increases in electrical conductivity (P < 0.001) were greatest in the LM. The lack of interactions among transportation, lairage, and muscle type was attributed to the relatively minor differences in stress among treatments. It was concluded that, in glycolytic muscle types such as the LM, the high physical and psychological stress levels associated with stress in the immediate preslaughter period have a greater effect on the water-holding capacity of the meat and may promote PSE development. Conversely, oxidative muscle types tend to have higher ultimate pH values and produce DFD pork in response to intense physical activity and/or high psychological stress levels preslaughter.  相似文献

以蒙古羊为研究对象,探究了传统掏心式屠宰、电击晕方式屠宰及抹脖子屠宰对宰后肌肉AMPK活性、糖酵解及食用品质的影响。结果表明:经抹脖子屠宰的肌肉中肌酸激酶、AMPK、乳酸脱氢酶活性均高于掏心式和电击晕方式屠宰组;掏心式屠宰后的蒙古羊羊肉滴水损失、熟肉率及嫩度高于其他2个组。研究结果说明,掏心式与电击晕方式屠宰有利于降低肉羊屠宰过程的应激反应,而掏心式屠宰更有助于提高羊肉食用品质。  相似文献
