The glutathione (GSH) -oxidant defence system protects the erythrocytes and leucocytes from oxidative damage. Leucocyte -superoxide dismutase (SOD), GSH-peroxidase (GSH-px), GSH-reductase (GR), GSH-S-transferase (GSH-S-t) and arginase were examined in samples from buffaloes infected with Anaplasma marginale. All the enzymes, except arginase, were also studied in the red cell haemolysates from these animals. GSH-S-t, GSH- and glutathione-reductase (GR) levels in leucocytes decreased in infected animals suggesting a decline in the efficiency of the GSH-oxidant defence system. SOD levels increased but there was no change in leucocyte-arginase activity due to infection. Infection caused no significant changes in red cell SOD, GSH-px, GR and GSH. However, GSH-S-t significantly decreased (P less than 0.05).  相似文献

In the present study the level of enzyme hydrolases (alkaline phosphatase, myeloperoxidase, elastase, arginase, lysozyme and β-galactosidase) of polymorphonuclear cell (PMN) granules in different ruminant species and their release in response to activation was studied. Buffalo PMN alkaline phosphatase activity was higher (P < 0.01) than in PMNs of cattle and goats. Interestingly, myeloperoxidase was higher in cattle PMNs and least in goat PMNs (P < 0.01), a similar pattern was observed in the distribution of enzyme arginase. As far as lysozyme is concerned, its activity was significantly higher (P < 0.01) in PMNs of buffaloes than in the case of cattle and goat PMNs. On activation, these cells released MPO and elastase, in all the species studied, while lysozyme was secreted only in buffalo PMN cells. Activity of certain enzymes related to oxidant defence systems such as glutathione peroxidase and glutathione reductase were higher in cattle and goats compared to that in buffaloes. These observations are likely to have bearing on immunodefense roles played by PMNs and reflected differences among the ruminant species studied.  相似文献

选择日本大耳白兔,按年龄分为幼年、青年、老年三组,分别测定动脉血血清SOD、MDA、GSH—px活性水平,以探讨日本大耳白兔抗氧化物酶活性增龄性变化。结果显示:随着年龄的增长,血清SOD活性在中年期以前是随增龄而增加,青年组显著高于幼年CR(P〈0.05,P〈0.01),在中年期以后是随增龄而降低,老年组明显低于青年组(P〈0.05,P〈0.01);血清MDA含量是随增龄而增加,老年组明显高于青年组和幼年组(P〈0.05,P〈0.01),青年组明显高于幼年CR(P〈0.05);血清GSH—px活性随增龄而降低,老年组显著低于青年组和幼年组(P〈0.01),青年组显著低于幼年组(P〈0.05,P〈0.01)。机体在衰老过程中体内自由基生成增加。抗氧化物酶活性全面降低可能是加速机体衰老的原因之一。  相似文献

试验旨在探讨鸡血藤总黄酮(TFSD)对猪圆环病毒2型(PCV2)感染小鼠脾脏氧化应激的影响。将70只昆明小鼠随机分为7组,对照组、TFSD组(100 mg/kg体重)、PCV2组、PCV2+维生素C (VC)组、PCV2+不同浓度TFSD (25、50和100 mg/kg体重)组,每组10只,连续3 d采用灌胃和腹腔注射PCV2病毒液的方法建立小鼠氧化应激模型,第4~6天每天按上述分别灌胃给予生理盐水、VC或TFSD。第7天剖杀小鼠,取脾脏分析黄嘌呤氧化酶(XOD)、髓过氧化物酶(MPO)和超氧化物歧化酶(SOD)活力,并检测还原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG)水平,计算GSH/GSSH。结果显示,PCV2感染小鼠后,脾脏中XOD与MPO的活力及GSSG水平显著上升(P<0.05),SOD活力、GSH水平及GSH/GSSG显著下降(P<0.05)。TFSD处理小鼠脾脏的SOD活力、GSH水平和GSH/GSSG比值均显著高于PCV2组(P<0.05),而XOD、MPO活力和GSSG水平则显著低于PCV2组(P<0.05),对PCV2引起的氧化应激相关酶活力与相关分子水平变化的抑制作用优于VC。结果表明,鸡血藤总黄酮对PCV2诱导的小鼠脾脏氧化应激有良好的调节作用。  相似文献

The aim of this study was to investigate the effect of total flavonoids of Spatholobus suberectus Dunn (TFSD) on porcine circovirus type 2 (PCV2) induced oxidative stress in mice spleen.70 Kunming mice were divided into 7 groups:Control group, TFSD group (100 mg/(kg·BW)), PCV2 group, PCV2+vitamin C (VC) group, and PCV2+various concentrations of TFSD groups (25, 50 and 100 mg/(kg·BW)). Mice were continuously treated with PCV2 via both intragastric administration and intraperitoneal injection for 3 d to establish oxidative stress models. From the 4th to 6th day, mice were intragastric administrated with saline, VC or TFSD, respectively, according to the grouping method. At the 7th day, the activities of xanthine oxidase (XOD), myeloperoxidase (MPO) and superoxide dismutase (SOD), the levels of glutathione (GSH) and oxidized glutathione (GSSG), and the ratio of GSH to GSSG in the mice spleen were analyzed. The results showed that PCV2 infection significantly upregulated the XOD and MPO activities and GSSG content(P <0.05), and dramatically downregulated the SOD activity, GSH level and the ratio of GSH to GSSG (P <0.05) in the mice spleen.Compared to PCV2 group, the SOD activity, GSH content and the ratio of GSH to GSSG in mice treated with TFSD were significantly increased (P <0.05), while the activities of XOD and MPO and the level of GSSG were significantly decreased (P <0.05), showing better performance in the inhibition of PCV2 induced changes of oxidative stress associated enzyme activities and moledule levels than VC.In conclusion,TFSD had regulative effect on the oxidative stress induced by PCV2 in mouse spleen.  相似文献

1. Tissue-specific profiles of the expression of the antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) and the concentrations of reduced glutathione (GSH) during the development of the chick embryo were investigated. 2. The liver, brain, yolk sac membrane (YSM), kidney, lung, heart and skeletal muscles were collected at the following days of embryo development: 10, 11, 13, 15, 17, 19, 21 and 22 (day-old chicks). 3. The different tissues of the embryo displayed distinct development strategies with regard to the acquisition of antioxidant capacity. In the liver the specific activity of SOD increased between days 10 and 11 of development, then significantly decreased up to day 15 and remained at the same value during the rest of the developmental period. GSH-Px specific activity increased through the time of development. CAT had 2 peaks of specific activity at day 10 of the development and in day-old chicks. 4. The brain was characterised by comparatively high SOD-specific activity especially during the last days of incubation. The specific activities of GSH-Px and CAT were low throughout development. 5. In the YSM maximal GSH-Px and CAT-specific activities were found on day 15 of incubation. In the kidney and heart GSH-Px-specific activity increased at hatching time. CAT-specific activity in the kidney increased just after hatching. 6. It is concluded that each tissue studied expressed a profile of antioxidant defence mechanisms to deal with oxidative stress at hatching time.  相似文献

利用自然病便,采取分组(血尿组、低磷组、对照组)对比的方法,对低磷奶牛红细胞超氧化物歧化酶(SOD)1、谷胱甘肽过氧化物酶(GSH-PX)活性和丙二醛(MDA)含量进行了测定。结果表明,低磷组及血尿组奶牛红细胞平均SOD、GSH-PX活性明显低于对照组,而MDA含量显著高于对照组及血尿组奶牛红直线相关分析表明,血清磷含量与SOD、GSH-Px活性呈极显著正相关(相关系数及回归方程分别为r=0.94  相似文献

The oxidant/antioxidant equilibrium of trained thoroughbred horses (n = 40) was assessed on three occasions during a period of three months under field conditions by blood antioxidant markers analysis, i.e. plasma ascorbic acid (AA), plasma antioxidant capacity of water-soluble components (ACW), whole blood (GSH) and oxidised (GSSG) glutathione, plasma alpha-tocopherol and beta-carotene, plasma antioxidant capacity of lipid-soluble components (ACL), red blood cell superoxide dismutase (SOD) and glutathione-peroxidase activity (GPx) and plasma trace-elements, i.e. selenium (Se), copper (Cu), zinc (Zn). A control group of ten horses receiving a placebo and an antioxidant group of 30 horses orally supplemented with an antioxidant mixture were randomly formed. An antioxidant imbalance was observed after three months in the control group, reflected by a significant decrease in GSH, SOD, GPx, Se (P < 0.05) and a significant increase in GSSG (P < 0.05). The antioxidant supplement prevented GPx and Se decrease and significantly increased ACW, alpha-tocopherol, beta-carotene and ACL (P < 0.05). Significant sex- or age-related differences were found for AA, ACW, alpha-tocopherol, SOD, GPx and Se, and there were significant correlations between ACW-AA, ACL-alpha-tocopherol, GPx-Se, CPK-Se, CPK-alpha-tocopherol and CPK-Cu. This field study has shown that trained thoroughbred horses undergo significant changes of several blood antioxidant markers and that oral antioxidant supplementation might partially counterbalance these changes by improving the hydrophilic, lipophilic and enzymatic antioxidant blood capacity.  相似文献

以0.2mg/kg体重剂量给健康仔猪肌注亚硒酸钠溶液后,系统研究了仔猪血硒浓度与抗氧化系统动态变化。实验结果表明,健康仔猪体内抗氧化系统随机体硒水平的变化而变化,但与后者相比有一个明显的滞后期。给予一定量的硒后,体内的抗氧化能力并没有立即增强,SOD活力及T-AOC均经过较长一段时间后才开始显著上升,而GSH-Px活性则是先降低而后又逐渐升高,MDA的变化也明显滞后于硒浓度的变化。机体内抗氧化系统与硒水平之间存在着一定的规律性。  相似文献

The aim of the study was the evaluation of cadmium effects on the activity of antioxidant enzymes in rat hepatocytes. The studies were conducted with isolated rat hepatocytes incubated for 1 or 2 hours in a modified (deprived of carbonates with phosphates) Williams' E medium (MWE) in the presence of cadmium chloride (25, 50 and 200 microM). Hepatocytes incubated in the MWE medium without cadmium chloride were used as a control. The application of the modified Williams' E medium allowed for the appearance of cadmium compounds in a soluble form that is indispensable for suitable estimation of its toxic action. There were evaluated markers of the oxidative stress such as: concentration of thiobarbiturate reactive substances (TBARS)--proportional to the level of lipid peroxidation, concentration of reduced glutathione (GSH), and the activity of antioxidant enzymes, including superoxide dismutase (SOD1 and SOD2), catalase (CAT), total glutathione peroxidase (GSHPx), selenium--dependent glutathione peroxidase (SeGSHPx), glutathione transferase (GST) and glutathione reductase (GSHR). Alterations of antioxidant enzymes activity, the level of TBARS and GSH in isolated rat hepatocytes caused by cadmium in vitro, were shown to depend on the concentration and time of exposure of cells to this metal. The increased level of TBARS and GSH was observed as well as changes in the activity of antioxidant enzymes. The activity of SOD isoenzymes and CAT was increased, whereas GSHPx and GST were decreased. These results indicate that cadmium induces oxidative stress followed by alterations in the cellular antioxidant enzyme system in isolated rat hepatocytes.  相似文献

本试验以12周龄的多年生黑麦草(Lolium perenne ‘cuttle’)幼苗为试验材料,用铅(Lead,Pb),Pb+还原型谷胱甘肽(Glutathione,GSH)和Pb+丁硫氨酸-亚砜亚胺(L-Buthionine-sulfoximine,BSO)处理1周,研究GSH对Pb胁迫下多年生黑麦草抗氧化系统的调控机理,寻求缓解植物Pb胁迫的有效措施。结果表明:Pb胁迫下,外源GSH能够显著提高多年生黑麦草Pb的吸收和转运,增加抗氧化酶超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GPX)和谷胱甘肽还原酶(Glutathione reductase,GR)活性,显著增加抗氧化剂GSH含量及GSH/氧化型谷胱甘肽(Oxidized glutathione,GSSG)比率,提高了植物的还原力。外源GSH也能够显著降低活性氧超氧阴离子(Superoxide anion,O2·-)和过氧化氢(Hydrogen peroxide,H2O2)的产生量、膜的相对透性(Relative electric conductivity,EC)和丙二醛(Malondialdehyde,MDA)含量,保持了细胞的稳定性,减轻了膜脂的过氧化程度。外源BSO作用基本上与GSH相反。相关分析表明:GSH可通过提高SOD和GPX等抗氧化酶的活性,促进GSSG和GSH的相互转化,降低活性氧产生量,提高植物抵抗逆境胁迫的能力。  相似文献

Antioxidants secreted by the reproductive tract protect spermatozoa against the toxic effects of reactive oxygen species (ROS) after ejaculation. This study aimed at characterizing the level of antioxidant protection in boar cauda epididymidal spermatozoa and fluids of the cauda epididymidis, vesicular and prostate glands. Also, this study investigated the effect of a 5-h period of dialysis on the antioxidant capacity of boar seminal plasma. Superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), glutathione transferase (GST) and phospholipid hydroperoxide glutathione peroxidase (PHGPx) activities were monitored in the cauda epididymidal spermatozoa or reproductive tract fluids. Also, the concentrations of total glutathione (GSH + GSSG), L-ergothioneine (ERT) and l-ascorbate and the total antioxidant status (TAS) of the fluids were measured. It was found that the cauda epididymidal spermatozoa exhibited high SOD activity and relatively low activity of PHGPx. The relative amounts of GPx, GR and GST activities in the cauda epididymidal spermatozoa were negligible, whereas CAT activity was undetectable. Greater SOD activity was found in the fluids of the cauda epididymidis and prostate gland. Furthermore, the prostate gland fluid appeared to be the main source of CAT activity in the seminal plasma, whereas the highest level of GPx activity was derived from the cauda epididymidal fluid. The reproductive tract fluids exhibited negligible amounts of GR and GST activities. It seemed that the significant amounts of GSH + GSSG, ERT and L-ascorbate in the reproductive tract fluids could have an ameliorative effect on the level of TAS in the seminal plasma. Dialysis had a marked effect on the total antioxidant capacity of the seminal plasma, which was manifested in greater activity of SOD and GPx. The findings of this study confirmed that the scavenging potential of the seminal plasma is dependent on the contributions of different antioxidants, originating in various fluids of boar reproductive tract.  相似文献

分别用小承气汤和利血平复制大鼠脾虚证模型,通过检测血清和心、肝、脾、肾等组织中抗氧化酶SOD和GSHP-Px的活性及脂质过氧化反应终产物MDA含量,研究脾虚证与自由基代谢的相关性及四君子汤对其影响。结果表明:两种方法复制脾虚证模型与对照组比较均表现为SOD活性显著升高(P<0.05),GSH-Px活性显著降低(P<0.05);四君子汤反证治疗组与脾虚证组比较表现为SOD活性显著下降(P<0.05),GSH-Px活性显著升高(P<0.05),MDA含量显著降低(P<0.05),与正常对照组接近。表明大鼠虚证动物模型与体内自由基代谢紊乱,脂质过氧化作用增强存在一定关系,健脾益气方药四君子汤能够提高体内抗氧化酶活性,拮抗自由基对生物膜的过氧化损伤,具有抗脂质过氧化损伤的作用。  相似文献

The objective of this study was to make the preliminary characterization of the antioxidant defence systems of the yellow fraction (YF) of red deer's (Cervus elaphus L.) semen during the rutting period. The semen was collected using artificial vagina (AV). The studies included spectrophotometric determination of antioxidant enzymes activities such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). We also analysed the contents of low‐molecular antioxidants such as L‐glutathione (GSH + GSSG), L‐ascorbate (ASC) and total antioxidant status (TAS). Additionally, the samples were subjected to PAGE and stained for SOD and GPx activities. It was demonstrated that the yellow fraction exhibited activities of SOD and GPx, with the highest activities in September and October. CAT activity was not detected. Staining for the SOD and GPx activities confirmed three protein bands with SOD activity and one protein band with GPx activity. The content of GSH + GSSG was similar in trials dating from October to December contrary to the content of ASC which was high in samples from September and October. The stable rate of TAS was observed during the whole rutting period. The results of this study showed that the YF of red deer semen is equipped with basic battery of antioxidant enzymes comprising SOD and GPx, with the supporting role of GSH + GSSG and ASC. Moreover, the samples obtained at the peak of the rutting period occurring from September to October had the highest enzymatic activity in comparison with remaining months of the rutting period, which contributed to the high quality of the semen by preventing it from the formation of oxidative stress during the short period of intense sexual activity of male red deer. The better understanding of the mechanisms of antioxidant defence systems in the YF of deer's semen may contribute to the potential use of this fraction in technology of wild ruminant semen preservation.  相似文献

Oxidant/antioxidant equilibrium disturbance has already been reported in trypanosome infections by several authors. The present study was aimed to explore the possible oxidant/antioxidant disturbance in surra of naturally infected horses before and after treatment. Fifteen naturally infected horses were chosen to analyse erythrocytic indices, platelet counts, lipid peroxides (LPO), nitric oxide (NO), reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) while six healthy animals acted as control. There was a highly significant (P<0.001) reduction in red blood cell (RBC) count, haemoglobin (Hb), packed cell volume (PCV) and platelet levels and a significant reduction in mean corpuscular haemoglobin concentration (MCHC) (P<0.05) was noticed. A highly significant increase in NO (P<0.001), a significant increase in LPO (P<0.05) and a significant decrease in GSH, SOD and CAT (P<0.05) were found. A negative correlation of RBC count with LPO (r=-0.844) and nitrate (r=-0.702) while a positive correlation with GSH (r=0.489), SOD (r=0.580) and CAT (r=0.689) was observed. All the animals were treated with Quinapyramine sulphate (3mg/kg s.c.) only once. Nine animals recovered completely without any side effects. The recovered animals were monitored and samples were collected every seven days for up to 21 days and parameters were analysed. After treatment, a significant increase in haematological parameters was noticed whereas the oxidative indices varied without any statistical significance. To conclude, the increase in oxidant parameters and decrease in antioxidant enzymes in infected horses indicates the disturbance of oxidant/antioxidant indices. There was a significant increase in post therapy haematological values, while the oxidant/antioxidant indices changed insignificantly indicating that antioxidants might be supplemented in the therapeutic regimen.  相似文献

通过检测鸡胚在早期发育过程中的抗氧化能力指数、脂质过氧化产物丙二醛、维生素C及谷胱甘肽的含量和超氧化物歧化酶活性,探讨内源性抗氧化活性物质变化,旨在为胚胎的抗氧化系统形成提供参考.结果显示,鸡胚在发育初期,胚胎内维生素C、谷胱甘肽、丙二醛含量及超氧化物歧化酶活性均有不同程度的提高,而卵黄内的抗氧化能力指数水平则逐渐降低.此外,试验结果还提示每克组织内的抗氧化指标在孵育前期呈递增趋势,发育至13日龄时随着体内抗氧化功能的健全而单位组织内的抗氧化水平趋于下降.以上结果说明,鸡胚内的抗氧化系统是在早期发育过程中逐渐形成并日益完善的,可能在胚胎发育过程中发挥着重要的生理意义.  相似文献

Oxidative stress is thought to contribute to the pathogenesis of HIV infection in humans. For example, CD4(+) T cells are particularly affected in HIV patients and oxidative stress may also contribute to impairment of neutrophil function in HIV/AIDS patients. Since cats infected with FIV develop many of the same immunological abnormalites as HIV-infected humans, we investigated effects of acute FIV infection on oxidative stress in cats. Cats were infected with a pathogenic strain of FIV and viral load, changes in neutrophil number, total blood glutathione, malondiadehye, antioxidant enzyme concentrations, and reduced glutathione (GSH) concentration in leukocytes were measured sequentially during the first 16 weeks of infection. We found that superoxide dismutase and glutathione peroxidase concentrations in whole blood increased significantly during acute FIV infection. In addition, neutrophil numbers increased significantly during this time period, though their intracellular GSH concentrations did not change. In contrast, the numbers of CD4(+) T cells decreased significantly and their intracellular GSH concentration increased significantly, while intracellular GSH concentrations were unchanged in CD8(+) T cells. However, by 16 weeks of infection, many of the abnormalities in oxidative balance had stabilized or returned to pre-inoculation values. These results suggest that acute infection with FIV causes oxidative stress in cats and that CD4(+) T cells appear to be preferentially affected. Further studies are required to determine whether early treatment with anti-oxidants may help ameliorate the decline in CD4(+) T cell number and function associated with acute FIV infection in cats.  相似文献

Phenylbutazone (PBZ) is widely used in equine medicine, and its side effects on the gastrointestinal tract are well known. The inhibition of prostaglandins and the oxidative stress induced by nonsteroidal anti‐inflammatory drugs (NSAIDs) are described as mechanisms of gastric mucosal injury in humans. In horses, only the secondary effect of changes in cyclooxygenases is related to gastric mucosal injury. The objective of this study was to evaluate the effect of PBZ on certain antioxidative/oxidative parameters of the gastric mucosa. The concentrations of antioxidants and oxidants (superoxide dismutase, SOD; catalase, CAT; nitric oxide, NO; total glutathione, GSH; myeloperoxidase, MPO; and malondialdehyde, MDA), PGE2 levels, and the ulcerative lesions score were assessed. The results demonstrated decreased levels of antioxidant variables, increased levels of oxidant variables, and alterations in the prostaglandin E2 (PGE2), myeloperoxidase (MPO), and glutathione (GSH) levels. In conclusion, PBZ induces oxidative stress in the gastric glandular mucosa of horses by changing the antioxidant–oxidant balance of this surface, which might be regarded as another mechanism of injury in the horse stomach.  相似文献

Antioxidant vitamins are commonly used as colorant, preventive, sweetener, nutritive and antioxidant materials in food industry. Thus, many studies have been conducted in recent years. We have performed the present study to understand the effects of α-lipoic acid (ALA), vitamin C and E, added into rations of Japanese quails, on chronic heat stress. This study was carried out in five groups as: control, stress, ALA, vitamin E and vitamin C groups. Heat stress is applied to all groups except the control group. Superficial pectoral muscles tissues were used for biochemical determinations. During the research, it is found that heat stress exerted undesired effects such as increasing lipid peroxidation (LPO) level indicating oxidative stress in Japanese quails. In addition, superoxide dismutase (SOD) and catalase (CAT) enzyme activities and glutathione (GSH) amounts were studied to understand the behaviour of defence mechanism. While stress increased LPO ratio, it was determined that all antioxidant added into the ration decreases LPO significantly (p < 0.05). In addition, it was determined that all of antioxidants added into ration effectively restored SOD activity and LPO ratio which heat stress affected negatively. Interestingly, vitamin C did not adjust GSH ratio in contrast to vitamin E and ALA, where vitamin E and C did not perform any positive effect on heat stress decreased CAT activity. Finally, it can be thought that antioxidant vitamins relatively ameliorated these undesired affects caused by stress factors given.  相似文献
