The aim of this study was to clarify the hormonal and immunological changes in periparturient dairy cows with excessive weight loss following calving. We analyzed leukocyte populations and the peripheral blood mononuclear cell (PBMC) mRNA of IFN-gamma, IL-4, growth hormone (GH)-receptor (R) and insulin -R and measured the GH and insulin concentrations. Ten dairy cows fed in one herd were used in this study and were divided into two groups. Five cows that experienced loss of body weight following calving of over 10% comprised the Weight Loss Group, and the remaining five cows, which experienced loss of body weight of 5% or less following calving, served as the Control Group. The Weight Loss Group had high serum cholesterol and low blood urea nitrogen levels compared with the Control Group throughout the period of observation. In regard to the leukocyte populations, there were significantly fewer CD4(+)CD45R-T cells in the Weight Loss Group 1 to 2 months after calving compared with the Control Group. The percentage of IFN-gamma/IL-4 among PBMCs in the Weight Loss Group tended to be lower than in the Control Group throughout the observation period. In the Weight Loss Group, the levels of GH-R during month 2 were lower than in the Control Group. We concluded that excessive loss of body weight in dairy cows following calving is associated with depressed cellular immune function.  相似文献

Most of the metabolic diseases of dairy cows occur within the first 2 wk after calving, and cows with a metabolic disease are prone to infectious diseases. Although metabolic diseases are generally recognized as a risk factor for infectious diseases owing to the associated decrease in immune function, the difference in immune status between cows with milk fever (MF) or displaced abomasum (DA) during the lactation period has not been clarified. Therefore, the peripheral blood leukocyte populations in 38 multiparous Holstein cows from 1 herd were analyzed after calving. The cows were divided into 3 groups according to health: 21 cows that remained clinically healthy throughout the experimental period (control group), 9 cows that had MF on the day of calving, and 8 cows with an onset of DA within 4 wk after calving. The T- and B-cell numbers were lowest at week 0, and they increased gradually after calving. There was no significant difference between the 3 groups in the number of each subset of leukocytes on the day of calving, but the number of CD8+ T-cells was significantly lower in the MF and DA groups than in the control group at week 1. The numbers of CD4+, CD8+, and WC1+ T-cells tended to be lower in the DA group than in control group from weeks 4 to 12, a tendency not observed in the MF group. These data suggest that when cows have DA around the time of calving, their lymphocyte numbers remain lower until 12 wk after calving.  相似文献

To compare the changes in the insulin reaction of Holstein dairy cows and Japanese Black cows (JB) during the periparturient period, the insulin resistance test in vivo and lymphocytes proliferation with insulin in vitro were performed. Ten healthy Holstein dairy cows (Holstein group) and 10 healthy JB cows (JB group) used in this study were observed on days 60, 40, and 20 before calving and days 7 and 20 after calving. In insulin resistance reaction in vivo and in vitro, a low insulin-stimulated glucose disposal rate and lymphocyte proliferation with insulin were observed in the Holstein group compared with the JB group during the experimental period. An analysis of the lymphocytes cultured with insulin showed that the percentage of CD4+CD45R- T cells in the Holstein group was significantly lower than that of the JB group before day 20. These findings indicate that T cells reaction to insulin in healthy periparturient Holstein cows is lower than that in Japanese Black.  相似文献

In this study, we investigated whether ozonated autohemoadministration (OAHA) influences leukocyte populations in cows with clinical inflammatory disease. Eleven cows with inflammatory disease (Inflammatory Group) and three healthy cows (Control Group) were used for this study. The CD4(+)/CD8(+) ratio in the Inflammatory Group increased significantly compared to that in the Control Group 3 to 4 days after OAHA treatment. In the Inflammatory Group, the number of CD14(+) cells decreased gradually after OAHA, but CD14(+) levels remained stable in the Control Group. The number of MHC class-II(+) cells decreased gradually in the Inflammatory Group, but increased gradually in the Control Group, and the difference between the groups was significant on day 14 after OAHA. These findings suggest a possible difference in the activation of immune response after OAHA in infected cows compared to healthy cows.  相似文献

Changes in immune factors expressed by milk somatic cells from Holstein cows with hypocalcemia after calving were investigated in this study. Fourteen multiparous Holstein cows after their 3rd or 4th calving in one farm were used. The cows were divided into 2 groups: 7 cows needing treatment due to onset of hypocalcemia (hypocalcemia group; age = 5.53 ± 0.27 years, parity = 3.14 ± 0.14) and 7 cows without health problems (control group; age = 5.88 ± 0.31 years, parity = 3.57 ± 0.26). Milk samples were collected aseptically using a cannula and mRNA of immune factors expressed by milk somatic cells were analyzed. Milk samples (50 mL) were collected from the right rear mammary gland of cows before milking at day 1 and weeks 1, 2, 4, and 8 after calving. All milk samples showed a negative reaction to the California Mastitis Test. Levels of relative interleukin (IL)-6 and cathelicidin in the hypocalcemia group were lower than those in the control group in weeks 1 to 8. A significant difference in relative IL-6 levels was found in week 4 (P < 0.05). These results suggest that levels of IL-6 expressed by milk somatic cells may be affected by hypocalcemia in dairy cows.  相似文献

Increased disease rates are commonly reported among high-yielding dairy cows in the transition period, extending from 3 weeks before to 3 weeks after calving, and characterized by the occurrence of an inflammatory response in terms of both positive and negative acute phase proteins (APP+ and APP-). To determine the above inflammatory response, the authors had developed the Liver Functionality Index (LFI), which defines the above condition on the basis of some APP- responses (albumin, cholesterol sensu stricto+bilirubin) during the first month of lactation. In this respect, low LFI values are associated to a high inflammatory response and vice versa. The relationship between LFI and inflammatory cytokine response was investigated from day -28 to day +28 with respect to calving in 12 periparturient dairy cows showing the six highest and six lowest LFI values within a cohort of 54 high-yielding dairy cows. The hypothesis being tested was that LFI and APP- on the whole could be used as readout of successful vs. non-successful adaptation to the transition period, with a strong association to disease occurrence. In fact, low LFI cows experienced many more disease cases (13 vs. 3 in high LFI Group) and related drug treatments till day +28. Interleukin-6 (IL-6) serum concentrations were always higher in low LFI cows (P<0.05 on day +28). The greater IL-6 levels were correlated with higher ceruloplasmin (APP+) and lower lysozyme serum concentrations (P<0.05 and <0.1, respectively). This latter finding was correlated with a clear role in vitro of lysozyme in a dose-dependent modulation of the inflammatory response of swine intestinal epithelial cells and bovine peripheral blood mononuclear cells. Hematological examinations showed no significant differences between the two groups under study. On the whole, our results indicate that LFI and LFI-related parameters could be used to identify cows at risk in the transition period toward an improved farm management. Also, our study indicates that disease cases in periparturient, high-yielding dairy cows are correlated with signs of accentuated IL-6 response and other markers of inflammatory phenomena. These likely start in the late lactation period or around dry-off, as suggested by our prepartal data, and proceed at much greater levels after calving.  相似文献

During the postpartum period there is a high incidence of mastitis in dairy cows. The reason for this increased risk of mastitis still remains unclear. Since leukocytes in colostrum have an important role in preventing the onset of mastitis, we investigated the leukocyte populations, which express CD4, CD8, CD14, CD21 or WC1, in colostrum as well as in blood obtained from 14 Holstein cows. Eight cows developed mastitis within a week after calving and the other 6 remained healthy. The percentage of CD14+ cells in colostrum was significantly lower in mastitic cows than in healthy cows. There were no significant differences in other marker positive cells either in the colostrum or in the blood. The CD14+ cells in colostrum play an important role of defense against invading microorganisms in the mammary glands. Our results suggested that the lower percentage of CD14+ cells in colostrum might predict the incidence of mastitis in the following period.  相似文献

The leukocyte populations of periparturient dairy cows were analyzed after administration of active egg white/Clostridium butyricum Miyairi additive. Sixty-eight Holstein milking cows were divided into 3 groups. Group A was administered active egg white product (AEWP)/Clostridium butyricum Miyairi 588 (Miyairi 588) additive (n=23). Group B was administered Miyairi 588 only (n=23), and Group C was the control group (n=22). The challenged groups were administered 100 g of AEWP + Miyairi 588, or Miyairi 588 alone, daily for 60 days from 1 month before until 1 month after paturition. Blood samples were collected from all groups three times (1 month before, 1 week after and 1 month after parturition) for analysis of the peripheral leukocyte population. The results showed significantly higher numbers of CD4+ cells in Group A compared with Group C 1 week after paturition. AEWP/Miyairi 588 additive may enhance the number of CD4+ T cells in periparturient dairy cows.  相似文献

试验旨在探明皮下注射褪黑素(MT)对荷斯坦奶牛配种妊娠率及血清生殖激素的影响。用计步器法确定自然发情的首次配种荷斯坦奶牛150头,对其中70头进行颈部皮下肌内注射褪黑素30 mg,12 h后进行人工输精;选择170头产后首次配种的荷斯坦奶牛进行同期排卵-定时输精处理,其中90头荷斯坦奶牛最后一次注射促性腺激素释放激素(gonadotropin-releasing hormone,GnRH)的同时进行颈部皮下肌内注射褪黑素30 mg,16 h后进行人工输精。在进行二次配种的荷斯坦奶牛中选择153只进行皮下注射褪黑素。荷斯坦奶牛输精后20~35 d进行妊娠检查,详细记录首次配种妊娠母牛头数、二次配种妊娠母牛头数、产犊数。选取同期排卵-定时输精的荷斯坦奶牛25头,皮下注射褪黑素8 h后用放射免疫法检测其血清中褪黑素、促卵泡素(FSH)、促黄体素(LH)、雌二醇(E2)的含量及35 d妊检时妊娠母牛血清中孕酮(P4)含量。结果显示,与自然发情和同期排卵对照组相比,其对应的皮下注射褪黑素组荷斯坦奶牛的妊娠率及产犊率均显著提高(P<0.05);皮下注射褪黑素组的双犊率显著提高(P<0.05),首次配种妊娠率和产犊率均显著提高(P<0.05),二次配种妊娠率和产犊率均差异不显著(P>0.05)。血清激素检测结果表明,与对照组相比,皮下注射褪黑素组血清中褪黑素、LH、E2含量均显著增加(P<0.05);35 d妊检时,皮下注射褪黑素的妊娠母牛P4含量显著增加(P<0.05)。本试验结果表明,皮下注射褪黑素能够提高荷斯坦奶牛的妊娠率、产犊率及血清中LH、E2和P4含量,说明皮下注射褪黑素能够促进卵母细胞成熟和排卵,并提高配种妊娠率。  相似文献

试验旨在研究当归提取物对围产期奶牛生产性能及健康状况的影响。选择60头荷斯坦奶牛按胎次、上一泌乳周期产奶量、预产期相近的原则随机分成4组,对照组饲喂粉状精料,试验一组将精料中的部分蛋白饲料制成颗粒,试验二、三组分别在蛋白颗粒饲料中添加0.4%和0.8%的当归提取物。试验测定采食量、奶产量、乳成分、血液指标及胎衣不下、子宫炎等健康指标。结果表明:添加当归提取物能显著提高奶牛采食量及产奶量(产后45 d内)(P<0.05);对乳成分无显著影响(P>0.05),血液生化指标变化较小;对部分产后疾病的预防效果较为明显,产后首次发情日期提前。研究显示,在日粮中以颗粒料为载体添加当归提取物对提高围产期奶牛的生产性能、改善健康状况有显著作用。  相似文献

AIMS: To assess the efficacy of a feed supplement designed to reduce the dietary availability of calcium when fed during the last 2-3 weeks of pregnancy, on the incidence of postparturient hypocalcaemia (milk fever) and on milksolids (MS) production in pasture-fed dairy cows in early lactation. METHODS: In Experiment 1, late-pregnant Holstein/Friesian cows, 4-10 years old, grazing pasture, were either fed the supplement (1 kg/cow/day) for 2-4 weeks prior to calving (n=11) or no supplement (n=10). Plasma calcium concentrations were measured immediately before and after a 20 h fast intended to induce hypocalcaemia at the end of this period, 1-7 days before the cows calved. In Experiment 2, mixed-breed dairy cows, 3-10 years old in 7 commercial dairy herds were fed the same supplement (1 kg/cow/day) for 1-3 weeks immediately prior to calving (n=565) or no supplement (n=614), and incidences of clinical milk fever and MS production at 6-9 weeks post calving were compared between groups. RESULTS: In Experiment 1, plasma calcium concentrations were higher (p=0.01) in supplemented than in non-supplemented cows after fasting, indicating reduced susceptibility to fasting-induced hypocalcaemia. In Experiment 2, the incidence of milk fever was lower (6.4% vs 17.1%, p=0.001) and MS production was higher (p=0.01) in supplemented than in non-supplemented cows. CONCLUSIONS: These preliminary results demonstrate that feeding a supplement designed to reduce dietary calcium availability for 2-4 weeks immediately prior to calving reduced the susceptibility of cows to fasting-induced hypocalcaemia, reduced the incidence of clinical milk fever and increased MS production in early lactation. CLINICAL RELEVANCE: The concept and product reported here have potential to provide dairy farmers with a practical means to prevent hypocalcaemia and improve milk production during early lactation.  相似文献

A high incidence (about 70%) of alteration in endometrial epidermal growth factor (EGF) profile, i.e., loss of 2 peaks on days 2–4 and 13–14, has been linked to a reduced fertility in multiparous repeat breeder Holstein cows. However, the EGF profile in Holstein heifers and other breeds (types) of cattle has not been investigated. In study 1, EGF concentrations were determined using endometrial tissues obtained by biopsy on days 3, 7 and 14 from 84 fertile Holstein heifers to obtain a normal range and 53 repeat breeder Holstein heifers to estimate incidence of alterations in the EGF profile. In repeat breeder heifers, EGF concentrations were similar to fertile controls on 3 days and five animals (9.4%) had an altered EGF profile with EGF concentrations below the normal range on days 3 and 14. In study 2, EGF concentrations on day 3 were repeatedly examined from the nulliparous period to the third postpartum period in 28 Holstein (dairy) and 47 Japanese Black (beef) cattle. The effect of parity on EGF concentrations on day 3 was different between Holstein and Japanese Black cattle. In Japanese Black cows, the EGF concentrations were consistently high throughout the study period, while in Holstein cows, the EGF concentrations decreased after the second calving. In conclusion, unlike multiparous repeat breeder Holstein cows, an altered EGF profile may not be a major cause of repeat breeding in Holstein heifers, and the peak EGF concentrations around day 3 may decrease even in fertile populations of multiparous dairy cows, but not in beef cows.  相似文献

OBJECTIVE: To determine effects of parturition on glucocorticoid receptor (GR) expression in neutrophils, serum cortisol concentration, and total blood leukocyte and neutrophil counts in periparturient dairy cows. ANIMALS: 23 Holstein cows. PROCEDURE: Blood samples were collected from 8 multiparous and 5 primiparous periparturient cows at various times from 28 days before parturition until 14 days after parturition. Glucocorticoid receptor expression in neutrophils, serum cortisol concentration, and total blood leukocyte and neutrophil counts were determined. Results were compared with results from control samples obtained from 5 multiparous and 5 primiparous Holstein cows in midpregnancy. RESULTS: Neutrophils from periparturient cows had 49% reduction in GR expression at calving, compared with GR expression 2 to 4 weeks before calving, and 39% reduction, compared with neutrophils from cows in midpregnancy. Reduction in neutrophil GR expression began 1 week before calving and was most severe at calving and 24 hours after calving; a significant difference in GR expression was detected between primiparous and multiparous cows. Serum cortisol concentrations and total leukocyte and neutrophil counts were significantly increased at calving and returned to baseline values by 24 hours after calving. Significant negative correlations were detected between neutrophil GR expression and serum cortisol concentration, total leukocyte count, and neutrophil count. CONCLUSIONS AND CLINICAL RELEVANCE: Reduced GR expression in blood neutrophils of periparturient dairy cows was associated with increased serum cortisol concentrations, leukocytosis, and neutrophilia. Thus, GR down-regulation in neutrophils may be involved in periparturient neutrophil dysregulation and may cause increased susceptibility to mastitis.  相似文献

A single injection of a synthetic analogue of prostaglandin F2 alpha (fenprostalene; Syntex) on the day of calving or between days 14 and 21 after calving did not affect the calving to first service interval, the number of services per conception or the conception rate of dairy cows. In a second trial, cows calved more than 45 days were injected once weekly with prostaglandin F2 alpha (dinoprost; Upjohn) or fenprostalene until served at the first detected oestrus. The calving to first service interval and the conception rate were not affected. Following injection of either prostaglandin, the spread in the pattern of onset of oestrus and the range in the proportion of cows (75 to 98 per cent) seen in oestrus within seven days were similar. This variation in the time of onset of oestrus after the administration of prostaglandin precludes limiting oestrus detection to four or five days per week, and is a major limitation to the use of prostaglandins in breeding programmes in dairy cows.  相似文献

Fifteen Holstein dairy cattle were monitored for lymphocyte subsets and expression of adhesion molecules on cells in milk and blood at parturition and at intervals up to 21 days post-partum. Using flow cytometry, we determined percentages of T cells (CD4+, CD8+, gammadelta) and B cells from milk and blood of these cows. We also measured expression of adhesion molecules (CD62L, LFA-1, LPAM-1, and CD44) on lymphocytes in milk and blood. Significantly higher percentages of CD8+ cells were found in milk than in blood at all time points while significantly higher percentages of B cells were found in blood than in milk at all time points. There were minimal to no significant differences in percentages of CD4+ or gammadelta+ cells between milk and blood. Expression of adhesion molecules was consistently higher on all subsets of milk lymphocytes compared with blood lymphocytes. These differences were most pronounced and statistically significant at calving and in the first week following calving. CD62L, LPAM-1 and CD44 were expressed on a significantly higher percentage of lymphocytes in milk at calving than in milk at subsequent sampling times, while LFA-1 expression on lymphocytes in milk was significantly lower at calving than at subsequent times.  相似文献

The occurrence of multiple metabolic and inflammatory diseases in dairy cows is higher during the periparturient period, which may be triggered by bacterial components, but not a viable bacterium. This study aimed to determine the association of endometritis and ovarian follicular cyst (OFC) with mastitis in dairy cows. Ninety-eight Holstein dairy cows were clinically examined for endometritis and OFC approximately 30–50 days after calving. Blood and milk samples were collected for the determination of milk somatic cell count (SCC); milk interleukin-1β (IL-1β), tumor necrosis factor-α (TNFα), and interleukin-8 (IL-8) concentrations; and plasma haptoglobin (Hp) and lipopolysaccharide-binding protein (LBP) concentrations. Of the 98 dairy cows included in this study, 12 were diagnosed with endometritis and 37 cows were identified as OFC-positive, whereas the remaining 49 cows were healthy (without endometritis or OFC). The average and maximum SCCs and plasma Hp and LBP concentrations were not significantly different between the healthy cows and those with endometritis or OFC. However, when the maximum SCC was classified as <300, 300–1,000, or >1,000 × 103 cells/ml, the percentage of cows with the maximum SCC <300 × 103 cells/ml was significantly lower in the endometritis and OFC-positive groups than in the healthy group. These results suggested that cows with endometritis and OFC during the postpartum period exhibit high SCC, indicating that some bacterial components can be transferred between organs.  相似文献

This study was to determine the effect of anionic salts on health and production performance of Holstein cows during the peripartum period. Twenty-four Holstein cows during the peripartum period were selected and divided into control group and experimental group. Twelve Holstein dairy cows in control group were fed diets with original concentrate,while twelve cows in experimental group were fed diets with concentrates adding anionic salts for the last 21 days before expected calving. All animals were ad libitum fed with the same diets after calving. The results showed that urine pH maintained 5.7 to 6.9,and blood pH was decreased significantly compared to control group (P<0.05). However,the contents of plasma Ca,glucose and alkaline phosphatase (ALP) activity in experimental group were significantly increased in 1 d after calving (P<0.05). What's more, the incidences of hypocalcemia,milk fever,retained placenta,displaced abomasum and mastitis were reduced in experimental group and the incidences of retained placenta was significantly decreased (P<0.05). No significant difference was detected in milk yield between two groups (P>0.05). In summary,supplementation of anionic salts concentrate could help to keep the health and plasma Ca homeostasis of Holstein cows.  相似文献

The transition period is a critical time for dairy cows as the animal is subjected to the physiological stress accompanying parturition. Immunosuppression and health status were examined during this period in 80 Holstein cows. Blood samples were taken from each cow 3, 2 and 1 week before and after calving, and at calving (0 day). RNA was extracted and subjected to real‐time PCR to determine mRNA levels for the immune‐related genes TLR 2, 4, 6, 7 and β‐defensin 5 in addition to the reproduction‐related genes prolactin and IGF‐I. Results showed significant up‐regulation of pro‐inflammatory‐selected genes, TLR 4, 6 7 and β‐defensin 5 at the third‐week post‐calving; however, earlier periods had lower expression of such genes. In contrast, the immunosuppression biomarker TLR2 gene was up‐regulated at calving and 1 week after parturition and then down‐regulated again at second and third week. Prolactin and IGF‐I genes expression levels were significantly and gradually increased mainly post‐partum. This research highlights that the expression patterns of TLRs, BNBD5, PRL and IGF‐I could be biomarkers to follow up immune and reproductive status of dairy cow at peri‐parturient period to predict the most susceptible risk time for disease incidence and to build up management protocol.  相似文献
