A pen trial with 720 Ross 308 male chicks evaluated effects of Paracox-5 vaccination at d 1, dietary mannan oligosaccharide (MOS; Bio-Mos at 2, 1, and 0.5 kg/tonne in starter, grower, and finisher, respectively), and d-15 Eimeria challenge on d-22 lesions and 1 to 42-d live performance. Available floor area in each of 24 floor pens was 2.1 m2, and 30 chicks were placed per pen initially. A 3-phase feeding program was used: starter (0 to 15 d), grower (16 to 22 d), and finisher (23 to 42 d). The 4 dietary and vaccine treatments were as follows: 1) without vaccination, dietary MOS, or coccidiosis challenge; 2) without vaccination and dietary MOS but with a 3-strain pathogenic Eimeria challenge; 3) with Paracox-5 vaccination and Eimeria challenge but no dietary MOS; and 4) with Paracox vaccination, dietary MOS, and Eimeria challenge. The pathogenic Eimeria sporulated oocysts mixture from the French National Institute for Agricultural Research (Paris, France), standardized and well-defined, was given via feed at d 15 (100,000 Eimeria acervulina, 10,000 Eimeria maxima, and 15,000 Eimeria tenella per bird). Coccidiosis lesion scores were recorded at 22 d of age (scored as 0, 1, 2, or 3 with increasing severity). Overall mean lesion scores (P < 0.001) were higher in challenged birds. Paracox-5 alone improved 15-d BW and 1 to 15- and 15 to 22-d average daily gain of Eimeria-challenged broilers (P < 0.05). Dietary MOS improved (P < 0.05) 15 to 42- and 22 to 42-d FCR of Paracox-5 vaccinated, Eimeria- challenged broilers. Mortality was 4.4 to 5.8% by treatment, with no significant differences. It was concluded that dietary MOS improved the feed conversion ratio of Paracox-5-vaccinated (d 1), challenged (d 15) broilers from 15 to 42 d of age.  相似文献

Effects on production and health in broilers vaccinated against coccidiosis (Paracox®, Schering-Plough Animal Health) in combination with an antibacterial organic compound (Orego-Stim®, Meriden Animal Health Ltd.) were investigated. To obtain an environment and infection level similar to practical conditions, experimental floor pens were set up in a commercial broiler chicken house. Orego-Stim® supplement increased live weight and feed intake up to 48 days of age, whilst feed conversion ratio was not significantly affected. Vaccinated birds had a significantly higher live weight between 34 and 48 days, compared to non-vaccinated, but vaccination did not significantly affect feed intake or feed conversion ratio. Coccidial numbers in the litter were low, and only minor intestinal lesions were found. There were no significant differences in lesion scores between the different treatments. Orego-Stim® supplement decreased caecal numbers of Clostridium perfringens at 31 days, but not at 52 days, and numbers were not affected at all by anticoccidial vaccination. Bird health was overall good. The results indicate that vaccination against coccidiosis in combination with an in-feed antibacterial compound may be an alternative control method for intestinal health in organically produced chickens.  相似文献

Increased resistance and loss of sensitivity to traditional anticoccidial agents, as well as interest in antibiotic-free rearing programs, has increased the interest in alternative control measures. Therefore, this study aimed to assess the effect of a DFM (direct fed microbial)/multi-enzyme product (DFM+XAP, providing 2,000 U/kg xylanase, 200 U/kg amylase, 4,000 U/kg protease and 75,000 cfu/g DFM) on broiler performance and profitability under 2 anticoccidial strategies (vaccination and traditional ionophore therapy). The study diets were 1) a nutritionally adequate control based on corn/soy/DDGS (PC), 2) a negative control reduced in energy and amino acids (NC), and 3) NC+DFM+XAP. Diets were divided into 2 subgroups, receiving either ionophores from 0 to 43 d (salinomycin 60 g/t) or a coccidial vaccine (Coccivac B) (6 treatments, n = 8, 50 male birds/n). Performance was recorded on d 22, 35 and 43. The net value of DFM+XAP inclusion was determined for all treatments. At d 43 reduced BWG and highest FCR were seen in NC birds, regardless of coccidiosis control (P < 0.05). Adding DFM+XAP significantly improved BWG and FCR by 5% (2,087 vs. 1,991 g/bird) and 6% (1.803 vs. 1.906), respectively. In birds receiving vaccine (P < 0.05), resulting in BWG (2,125 g/bird) and FCR (1.772) not statistically different to birds fed PC diets. Across anticoccidial strategies, adding DFM+XAP resulted in 3% BWG improvement (P < 0.05) as compared to NC. The net value of DFM+XAP addition, compared to PC, was $15.15/ton feed in vaccinated birds and $14.71 in those treated with ionophores. In conclusion, the addition of DFM+XAP to broiler diets significantly improved performance and profitability, regardless of coccidiosis control program, though response was likely to be greater in vaccinated birds.  相似文献

Twenty European Eimeria spp. field isolates were subjected to an anticoccidial sensitivity test (AST). The anticoccidial drugs tested were diclazuril (Clinacox) and monensin (Elancoban). The assay was performed in a battery cage trial. Infected medicated birds were compared with an unmedicated control group. Coccidial lesion scores and oocyst shedding were used as parameters. The results of the AST show that resistance is common amongst coccidiosis field isolates, especially Eimeria acervulina (68% and 53% resistance for diclazuril and monensin, respectively). Resistance is less frequent amongst Eimeria maxima (38% and 50% resistance for diclazuril and monensin, respectively) and Eimeria tenella isolates (23% and 38% resistance for diclazuril and monensin, respectively). A highly significant influence of the coccidiosis prevention program (live coccidiosis vaccination with Paracox-5 vs. anticoccidial drugs in feed) on the sensitivity patterns of Eimeria spp. field isolates for both diclazuril (P= 0.000) and monensin (P= 0.001) was found. Further, when looking at the single species and each anticoccidial drug level, significantly more sensitivity of E. acervulina for monensin (P= 0.018), E. maxima for diclazuril (P = 0.009), and E. tenella for diclazuril (P = 0.007) was found in isolates originating from vaccinated flocks. Moreover, for E. acervulina and diclazuril, E. maxima and monensin, and E. tenella and monensin a trend toward higher sensitivity of isolates for these products was found when live coccidiosis vaccination was applied. The present study shows that sensitivity for the anticoccidial drugs diclazuril and monensin is more frequent in Eimeria spp. field isolates originating from broiler farms where a coccidiosis vaccination policy is followed.  相似文献

A crude resinous extract from Commiphora swynnertonii was tested against an experimental coccidial infection in local chickens. A total of 80 growing chickens were randomly assigned into five groups, which received different treatments. Chickens in G1 were not infected with coccidian oocysts and therefore served as a negative control. All chickens in G2, G3, G4 and G5 were infected through oral administration of coccidian oocysts suspension at a dosed rate of 1.5?×?104 Eimeria spp. oocysts per bird. Starting from day 3 post-infection (p.i), chickens in different groups were treated for 7 consecutive days as follows: G1 and G2 (positive control) received 5 ml of normal saline as placebo, G3 and G4 were given the extract at 400 and 800 mg/kg bodyweight whereas G5 received anticoccidial drug. Clinical signs, bodyweights, oocysts counts and mortality rates were observed regularly. Results showed that oral administration of the resinous extract to chickens with coccidiosis significantly reduced mortality rate from 94 to 25 % and oocysts counts from 1.03?×?105 to 6.55?×?103 oocysts/g faeces (p?<?0.05). Also a body condition score chart indicated less severe clinical signs of the disease in the groups which received the extract. Mean daily body weights were slightly reduced by the administration of the extract but this effect disappeared by day 7 p.i. These findings clearly indicate that resinous extract from C. swynnertonii has significant anticoccidial effect against experimental Eimeria spp. infection in chickens. A larger field trial to validate the use of the extract in chickens naturally infected with Eimeria spp. is required.  相似文献

Necrotic enteritis is a complex disease condition of broiler chickens, commercial layer pullets, and turkeys and requires the presence of a toxigenic strain of Clostridium perfringens, alteration of bird diets, and damage to the intestinal epithelium. All of these alone, but especially in combination, result in significant alterations of the intestinal microflora. The intestinal microflora is part of a complex ecosystem that is involved in augmenting intestinal development, immune surveillance, and competitive exclusion against pathogenic organisms. However, when the microflora fails to protect the mucosa from pathogens, antimicrobials are used to reduce the numbers of pathogenic organisms or to prevent their colonization of the intestine. We were interested in studying the effects of monensin on the bacterial community within the ileum of chickens because ionophore antimicrobials are commonly used to prevent Eimeria infections in broilers. We used two 16S ribosomal DNA community analysis protocols, terminal restriction fragment length polymorphism analysis combined with 16S rDNA clone libraries. These methods showed that monensin caused significant alterations in the microbial community structure of the ileum. Whereas the ileal bacterial community of control birds primarily consisted of lactobacilli, monensin-treated birds had communities rich in clostridia. None of the birds exhibited signs of intestinal disease or mucosal lesions, which suggested that the clostridia were avirulent. Although the therapeutic benefits of monensin may largely result from its anticoccidial effects, its ability to foster a competitively exclusive bacterial community may contribute to the intestinal health of birds, thus preventing the development of necrotic enteritis.  相似文献

A 42-day broiler floor pen study was conducted comparing the anticoccidial efficacy of toltrazuril (Baycox) as a stand alone treatment and as an additional treatment to in-feed anticoccidial programs. Toltrazuril was administered on days 18 and 19 in the drinking water at 7 mg/kg of body weight. The treatments were 125 ppm nicarbazin (days 0-14) to 66 ppm salinomycin (SAL) (days 15-35) with and without toltrazuril, SAL (days 0-35) with and without toltrazuril, nonmedicated (NM) to SAL with toltrazuril, and NM with and without toltrazuril. The controls were NM noninfected and infected. The treatments were replicated in five blocks of eight pens each in a randomized complete block design. All withdrawal feed was nonmedicated. On day 14, birds, except noninfected, were exposed to coccidial oocysts (Eimeria acervulina, Eimeria maxima, and Eimeria tenella) seeded litter. On days 21, 28, 35, and 42, birds and feed were weighed, four birds per pen were coccidial lesion scored, and litter oocyst counts were performed. The coccidial infection in the NM infected treatment caused a significant (P < 0.05) coccidiosis infection. Coccidiosis was moderately controlled in the anticoccidial treatment birds without toltrazuril. Performance in the NM with toltrazuril was equal to or better (P < 0.05) than the anticoccidial programs without toltrazuril. Toltrazuril was equal to the noninfected birds in performance. Toltrazuril most completely eliminated all coccidial lesions and dramatically reduced oocyst shedding. The performance data, lesion scores, and oocyst counts showed that a 2-day treatment with toltrazuril successfully controlled the coccidiosis with no relapse of infection. Toltrazuril can thus be used for supplemental control with in-feed anticoccidials or as a primary anticoccidial with nonmedicated feed.  相似文献

A trial was conducted to assess the effects of the dietary supplement Yucca schidigera extract on the performance of broilers subjected to different coccidiosis control methods. Six hundred male broiler chicks were randomly allocated to 1 of 5 treatments with 4 replicates each. The treatments were as follows: T1, vaccinated against coccidiosis; T2, nonvaccinated, a dietary coccidiostat; T3, vaccinated against coccidiosis, diet supplemented with Y. schidigera extract (0.01%); T4, coccidiostat plus Y. schidigera extract added to feed (0.01%); and T5, nonvaccinated, nonsupplemented control. The parameters analyzed were bird performance and morphology of the intestinal mucosal surface. The intestinal villi were higher at 6 d of age in broilers treated with coccidiostats or vaccinated against coccidiosis and supplemented with Y. schidigera extract compared with groups receiving the vaccine only or no treatment. Addition of the Y. schidigera extract to the diet of vaccinated birds improved average daily gain and feed conversion rate at 42 d of age compared with the other treatments, suggesting there might be a beneficial, synergistic effect between the coccidiosis vaccine and the Y. schidigera extract.  相似文献

The efficacy and economic benefits of Supercox, a live anticoccidial vaccine were examined and compared with an anticoccidial drug in a trial in broiler chickens under modern commercial conditions in China. In total, 40,660 chickens were used in the present study, half of which were vaccinated with the Supercox vaccine comprising a precocious line of Eimeria tenella and non-attenuated lines of Eimeria maxima and Eimeria acervulina, and the other half were medicated with Diclazuril delivered as feed additive at the dosage of 1mg/kg of feed. The vaccine was administered orally to 7-day-old chickens. No clinical diseases were diagnosed in any of the vaccinated birds. However, clinical coccidiosis occurred in a large proportion of medicated control birds and these chickens had to be treated with anticoccidial drugs (Diclazuril and Toltrazuril). Comparison of production performance between vaccinated birds and medicated control birds revealed that the vaccine Supercox performed better than anticoccidial drugs in terms of mortalities, costs and overall economic benefits (profits). These findings demonstrated that the use of the Supercox vaccine could control clinical coccidiosis in broilers and achieve production performance superior to that using anticoccidial drugs, particularly where drug resistance might result in failure to control clinical diseases.  相似文献

Paracox anticoccidial vaccine was administered to a 7-day-old flock of commercial broiler breeder stock subsequently reared to point-of-lay in the same house. For comparison, three subgroups of another flock of broiler breeders were also vaccinated with Paracox at 7 days of age, reared to 42 days and then transferred to new litter on another farm until point-of-lay. The first subgroup received no further treatment, but the second and third each received a second vaccination with Paracox, either immediately after transfer to the new litter or 42 days after transfer. Using an Eimeria necatrix model, protective immunity was demonstrated by virulent challenge of samples of birds from all groups by the age of 37–40 days (30–33 days after the first vaccination), and was maintained to at least 122–125 days of age, whether the birds remained on the same litter or were transferred to another farm, and whether they received one or two anticoccidial vaccinations. Therefore, there is no disadvantage in transferring birds onto new litter 35 days after a single Paracox vaccination, nor is there any advantage in giving a second vaccination after such a transfer. Vaccinated birds seeded the new litter with oocysts, despite being clinically immune to coccidiosis. A supplementary laboratory experiment showed that birds vaccinated at 8 days of age passed almost no oocysts after a second vaccination at 43 days of age. This indicated that they were not only protected against clinical coccidiosis, but were almost solidly immune to a homologous infection 5 weeks after a single vaccination. Nevertheless, oocysts appeared in the litter of all four groups of commercial breeders throughout the trial, showing that wild-type heterologous infections occurred whether the birds were transferred to new litter or not, but these did not overwhelm the acquired protective immunity and cause clinical coccidiosis.  相似文献

Purpose

Coccidiosis is an important parasitic disease in poultry industry. Owing to the development of drug resistance against Eimeria and concerns about drug residues, attentions toward the alternative compounds including herbal medicines have been increased. This study aimed to examine the prophylactic and therapeutic effects of a new formulation derived from Artemisia sieberi extract on avian coccidiosis.

Methods

The extract was obtained from A. sieberi using petroleum ether, and then, it was formulated into a wet granule. Three hundred and sixty 1-day-old broilers were divided into six groups, each with three replicates (n?=?20); the first group was chosen as the noninfected control group and the remainders were challenged by oral administration of 250,000 oocysts of Eimeria tenella per chick on day 21. Group 2 (as nontreated control) received no treatment, but group 3 (as prophylactic group) received 0.5 mg/kg artemisinin 10 days before challenging with E. tenella. However, groups 4, 5, and 6 received 1 mg/kg artemisinin on the next day and at 24 and 48 h after the challenge, respectively, for 5 days. Mortality rate, bloody diarrhea score, lesion score, and oocysts per gram (OPG) of feces were used to determine the anticoccidial effects of the formulation.

Results

The granule significantly (p?<?0.05) reduced mortality, diarrhea, and lesion score in the treated groups. The OPG output was also significantly (p?<?0.0001) reduced in the groups that received formulation. However, there was no significant difference between the prophylactic and therapeutic groups.

Conclusion

The results suggested that this new formulation is a promising herbal medicine that can be used as a prophylactic or therapeutic product to control avian coccidiosis.  相似文献

Studies were conducted to assess the performance of turkeys fed diets containing lactose. Study 1 consisted of 96, mixed-sex turkey poults that were housed in battery cages and fed diets containing 0 (8% cornstarch), 0.5, 1, 2, 4, or 8% lactose from 0 to 6 wk. Study 2 used 360, mixed-sex turkey poults that were raised in floor pens and fed a commercial diet and diets containing 0 (4% starch), 0.5, 1, 2, or 4% lactose from 0 to 6 wk. Diets and water were offered ad libitum throughout both trials. The objectives were 1) to determine the effect of lactose on turkey performance and 2) to establish the optimum level for lactose inclusion in diets. In Study 1, feed intake, BW, fecal ammonia and nitrogen, and feed efficiency were determined. Diet composition, BW, feed intake, total body and carcass composition, cecal pH, volatile fatty acids (VFA) and lactic acid were measured in Study 2. Turkeys consuming the 4% lactose diet gained the most weight (P < 0.01) compared with other treatments in Study 1, and lactose improved feed efficiency. In Study 2, all lactose diets resulted in heavier BW (P < 0.05) compared with the commercial diet. Turkeys consuming diets with 0.5, 1, 2 or 4% lactose had less total body fat compared with turkeys consuming a commercial or 0% lactose diet (P < 0.05).  相似文献

This study investigated the effects of various coccidiosis control programs in combination with antibiotic growth promoters (AGPs) on growth performance and host immune responses in broiler chickens. The coccidiosis programs that were investigated included in ovo coccidiosis vaccination (CVAC) with Inovocox or in-feed medication with diclazuril as Clinacox (CLIN) or salinomycin (SAL). The AGPs were virginiamycin or bacitracin methylene disalicylate plus roxarsone. As a negative control, chickens were non-vaccinated and fed with non-supplemented diets (NONE). All animals were exposed to used litter from a commercial broiler farm with confirmed contamination by Eimeria parasites to simulate in-field exposure to avian coccidiosis. Broiler body weights in the CVAC group were greater at 14 and 32 days of age, but not at day 42, compared with the NONE, CLIN, and SAL groups. At day 14, the SAL group showed decreased body weight and reduced ConA-stimulated spleen cell proliferation compared with the CLIN and SAL groups. In contrast, at days 34 and 43, splenocyte proliferation was greater in the CVAC and CLIN groups compared with the NONE and SAL groups. Lymphocyte subpopulations and cytokine mRNA expression levels in the intestine and spleen were also altered by the denoted treatments. Collectively, these results suggest that in ovo coccidiosis vaccination or coccidiostat drug medication programs in combination with AGPs influences chicken growth and immune status in an Eimeria-contaminated environment.  相似文献

Anticoccidial vaccine and an anticoccidial drug rotation program were compared to determine which program was more effective in producing coccidia populations sensitive of 1 ppm diclazuril. The study used an anticoccidial drug-sensitivity battery test (AST) to determine the baseline level of diclazuril sensitivity to field isolates of Eimeria spp. from seven broiler complexes that had used diclazuril. Based on percentage reduction in weight gain and lesion scores, 25% or fewer of the isolates were effectively controlled by diclazuril. Following the baseline sampling, four of the complexes switched to a nondiclazuril in-feed anticoccidial drug program and three of the complexes switched to a vaccination program for two broiler grow-out cycles as the sole coccidiosis-control program. This study demonstrated that the vaccine used (Coccivac-B) contained anticoccidial drug-sensitive strains. Eimeria isolates were subsequently collected from the identical houses and diclazuril AST results were compared with the baseline AST results. Following the two grow-out cycles, sensitivity of the isolates to diclazuril from the four complexes that continued to use in-feed anticoccidial drugs remained essentially unchanged. The isolates from the three complexes that switched to the vaccination program demonstrated a marked increase in diclazuril sensitivity, with 60%-100% of the isolates from each complex effectively controlled by diclazuril. Vaccination with the anticoccidial drug-sensitive strains produced a measurable increase in the level of sensitivity to diclazuril.  相似文献

An experiment was carried out to assess the influence of tannic acid (TA) on integrity of the intestine in broiler chicks vaccinated against coccidiosis and challenged with the disease. In a 2 × 2 factorial design, the trial had five groups of 10 chickens each, including positive (group 2) and negative (group 1) controls. The chickens were kept on wood shavings and fed a commercial maize and soybean-based starter-grower diet. From day 1, groups 3 and 5 received TA (10 g kg(-1)) in their diet. On day 4, birds of groups 4 and 5 were vaccinated orally against coccidiosis with anti-coccidial vaccine, Livacox T?. Each dose of the vaccine contained 300-500 sporulated oocysts of each of Eimeria acervulina, Eimeria maxima and Eimeria tenella. On day 18, all experimental groups except for the negative (group 1) were challenged with 10-fold dose of Livacox T? to produce a mild coccidiosis infection. Faecal samples of individual birds were collected on day 23, and the number of faecal oocysts was determined. d-Xylose absorption test was also carried out on all birds on day 23. Immediately after d-xylose absorption test, all birds were killed humanely and the intestinal tract was removed, weighed and examined for gross lesions. Results showed that negative (group 1) and positive controls (group 2) had the highest and lowest levels of plasma d-xylose post-ingestion of the substrate, respectively. Vaccination and/or feeding TA raised the level of plasma d-xylose in infected birds, although this was not significant for TA-fed birds. Vaccination reduced but TA increased the total number of oocysts per gram of faeces. Birds of groups 2-5 had distinct intestinal lesions when compared with group 1. However, vaccination prevented intestinal lesions. Relative weights of intestinal parts were the lowest in group 1 and the highest in group 2. Vaccination but not TA reduced the relative weights of intestinal parts in infected birds. It was concluded that dietary tannins may reduce the efficacy of anticoccidial vaccines and alter the proper development of immunity against the disease.  相似文献

A field trial was conducted on a farm in Michigan with commercial turkeys naturally infected with Ascaridia dissimilis. The birds were treated at approximately 8 and 14 wk of age on a per-barn basis with fenbendazole (Safeguard) in the feed at 16 ppm for 6 d (1 barn), or with levamisole (Prohibit) in the water at 16 mg/kg of BW (2 barns). Six birds were obtained at random and sacrificed in each barn for worm burden determinations before treatments and again soon after treatments. Nematicidal efficacies for fenbendazole and levamisole were shown to be 99.3 to 99.9% and 54.6 to 85.8%, respectively. Despite the far greater reductions in worm burdens by fenbendazole as compared with levamisole, the burdens at the time of retreatment had equalized in all barns, illustrating the role of premunition regarding turkey ascaridiasis, and highlighting the overriding importance of reducing the challenge to provide long-term control of this parasitism.  相似文献

Avian coccidiosis is one of the most important diseases of poultry and it is responsible for a large number of all broiler mortalities worldwide. The control of this disease relies mainly on the use of anticoccidial drugs. However, herbal preparations could be an alternative for the treatment against coccidiosis in chickens. The direct effects of Moringa oleifera acetone extracts on broiler chickens naturally infected with mixed Eimeria species was investigated to determine the relative efficacy of the extracts against coccidiosis in birds. The investigations were carried out in seven groups (ten chickens per group). The birds were given various doses (1.0, 2.0, 3.0, 4.0 and 5.0 g/kg body weight) of acetone extract of leaves of M. toltrazuril® (positive control) and untreated (negative control). The extract was evaluated for anticoccidial activity by means of inhibition of oocyst output in faeces, faecal score, weight gain and mortality. Haematological indices were evaluated by standard methods. The group treated with 1.0 g/ kg body weight Moringa oleifera extract produced the least inhibitory effect on oocyst shed in the faeces (96.4 %), while the groups treated with 2.0 g/kg, 3.0 g/kg, 4.0 g/kg and 5.0 g/kg body weight of the extract produced 97.4, 98.7, 99.1 and 99.8 %, respectively. Body weight gains of infected chickens treated with the extract significantly improved (p?<?0.05), and faecal scores were milder. Packed cell volume, haemoglobin concentration and red blood count of the treated birds were significantly (p?<?0.05) higher than those of the infected untreated group. Moringa oleifera leaves could find application in the treatment of avian coccidiosis in veterinary practice.  相似文献

The aim of this project is to study the clinical signs and lesion of velogenic Newcastle disease (vND) in commercial turkeys, and also to find out if La Sota vaccination offered protection against these signs and lesions. The cockerels were included as positive controls. One hundred and twenty turkey poults and cockerels were divided into eight groups as follows: unvaccinated unchallenged turkeys (UUT), unvaccinated challenged turkeys (UCT), vaccinated unchallenged turkeys (VUT), vaccinated challenged turkeys (VCT), and along the same lines, the cockerel groups were UUC, UCC, VUC and vaccinated challenged cockerels (VCC). Vaccination was at 3 weeks of age while challenge was at 6 weeks of age. The unvaccinated turkeys and cockerels (UCT and UCC) showed different degrees of depression, diarrhoea and later paralysis at challenge. Total mortality was 100% in cockerels within 6 days, but 60% in turkeys. Similar but milder clinical signs were found in the VCC with a total mortality of 13.3%. The VCT showed mild drop in feed and water consumption, and no mortality. All the challenged groups had significant (p < 0.05) loss of weight when compared with their controls. Necropsy showed that while the UCC had severe proventricular haemorrhages, intestinal and caecal tonsil ulcers, the UCT had no digestive tract lesion. There was severe atrophy of the lymphoid organs in all the challenged groups. Histopathological sections of the bursa, spleen and thymus in all the challenged groups with special emphasis on the vaccinated and unvaccinated turkeys with mortalities of 0 and 60%, respectively, had very severe necrosis and depletion of the lymphoid tissue. Virus was isolated from the cloacal swabs. The haemagglutination inhibition antibodies were significantly higher (p < 0.05) in the challenged groups than the unchallenged. The above observations in the intestinal tracts of UCT are of diagnostic significance while the gross and microscopic lesions in the UCT and VCT show that La Sota vaccination may not protect turkeys against the destruction of the lymphoid organs by vND as earlier reported in chickens. This may lead to immunosuppression and production problems in areas where vND is enzootic.  相似文献

Caprine coccidiosis, affecting mainly young goat kids around the weaning period, is worldwide the most important disease in the goat industry. Control of caprine coccidiosis is increasingly hampered by resistances developed against coccidiostatic drugs leading to an enhanced need for anticoccidial vaccines. In the current study we conducted an oral immunization trial with live attenuated sporulated Eimeria ninakohlyakimovae oocysts. Sporulated E. ninakohlyakimovae oocysts were attenuated by X-irradiation technique. The experimental design included a total of 18 goat kids divided into the following groups: (i) animals immunized with attenuated E. ninakohlyakimovae oocysts at 5 weeks of age and challenged 3 weeks later with non-irradiated homologous oocysts (group 1); (ii) animals infected with non-attenuated E. ninakohlyakimovae oocysts at 5 weeks of age and challenged 3 weeks later with non-attenuated homologous oocysts (group 2); (iii) animals primary-infected with untreated E. ninakohlyakimovae oocysts at 8 weeks of age (control of the challenge infection, group 3); (iv) non-infected control animals (group 4). Goat kids immunized with live attenuated E. ninakohlyakimovae oocysts (group 1) excreted significantly less oocysts in the faeces (95.3% reduction) than kids infected with non-attenuated ones (group 2). Furthermore, immunization with live but attenuated oocysts resulted in ameliorated clinical coccidiosis compared to goat kids infected with untreated oocysts (group 2) and resulted in equally reduced signs of coccidiosis after challenge infection compared to acquired immunity driven by non-attenuated oocysts. Overall, the present study demonstrates for the first time that live attenuated E. ninakohlyakimovae oocysts orally administered showed almost no pathogenicity but enough immunogenicity in terms of immunoprotection. Importantly, vaccinated animals still shed low amounts of oocysts, guaranteeing environmental contamination and consecutive booster infections to sustain ongoing immunity.  相似文献
