Pinus pinaster Ait. is one of the main forest tree species planted in Spain, Portugal and France. Due to its high economic relevance, there is considerable interest in developing techniques for vegetative breeding aimed at mass propagation. In this study we present a mini-propagation protocol in order to define an efficient method to propagate families or clones of P. pinaster. We carried out three experiments using mini-cuttings of 3–5 cm in length with the aim of evaluating the effects of temperature (4°C vs. 25°C), plant growth regulator (IBA) and shoot age on rooting ability. Percentage of rooted cuttings and morphological root variables were recorded. The percentage of rooted cuttings per treatment ranged from 68 to 97%. Treatment with IBA significantly influenced the rooting process at 25°C but not at 4°C. The number of apexes, length, area and volume of roots were all positively affected by temperature treatment. Shoot age also had a positive effect on rooting capacity of cuttings, with the cuttings from the youngest shoots (70 days after pruning) having higher rooting percentages, ranging from 84.7 to 98.3%. The use of juvenile material, good environmental conditions and IBA all benefited the rooting of clonal material, resulting in high rooting capacity. This study presents an innovative propagation protocol for P. pinaster that can be used as a tool in breeding programs.  相似文献

Mesquite (Prosopis alba Grisebach), an important species in arid and semiarid regions, is currently commercially propagated by seeds and so there is great interest in developing techniques for species propagation, domestication and improvement. Cloning through mini-cuttings is a new and promising technique for the production of clonal seedlings of forest species. The main objective of this work was to evaluate vegetative propagation using the mini-cutting technique and indolebutyric acid (IBA) at different concentrations (0, 3,000, 4,500, 6,000 and 7,500 mg L?1) on rooting of P. alba clones. Rooting was achieved in 98–100 % of the mini-cuttings at all concentrations tested. Increasing IBA concentration resulted in an increase in the number of leaves and leaflets, as well as in fresh matter weight and number and length of roots of clone seedlings, until an optimum point (between 3480 and 4800 mg L?1) was reached. Plants were also vegetatively propagated in the field via conventional clonal garden cuttings under the same rooting conditions to compare the efficiency of the two propagation techniques. Mini-cuttings showed higher rooting and survival percentages than cuttings. A correlation analysis conducted between characteristics of stock plants (height, diameter, number of shoots) and shoots (height, diameter) showed a positive correlation between rooting and height of shoots with total root length, leaflet number, and height of clonal seedlings. An efficient production system, which can quickly produce high quality plants, is shown in this study of Liquidambar styraciflua. The mini-cuttings technique was used for ex vitro clonal propagation of three sweetgum clones. The semi-hydroponics system for production of propagules was also tested, and the concentration of indole-3-butyric acid (IBA) for rooting was evaluated. Three adult clones, produced according to conventional cutting processes, were used as mini-stumps and maintained in semi-hydroponics systems of medium size sand. Ten collections of sprouts were prepared to evaluate the production of semi-hydroponics systems (mini-clonal hedges). Sprouts from collection number six were used at different concentrations of IBA solution (0; 500; 1,000; 3,000 and 6,000 mg l−1) to test the mini-cuttings technique. The high survival rates of mini-stumps after ten collections (96.3%), and high annual production per square meter (~2,953 mini-cuttings) showed that the mini-clonal hedges are viable to conduct sweetgum mini-stumps. The formation of 62.5% of healthy plants after hardening under outdoor conditions showed that sweetgum could be successfully propagated by the mini-cuttings technique.  相似文献

Eucalyptus benthamii is a forest species of economic interest that has difficulty with seed production and also is considered to have difficulty with adventitious rooting using propagation techniques,such as cutting or mini-cutting.We aimed to assess the adventitious rooting percentage under different storage times in low temperatures and at various IBA(indole-3-butyric acid) concentrations to determine the optimal time of permanence for rooting Eucalyptus benthamii minicuttings in a greenhouse.Shoots collected from mini-stumps cultivated in a semi-hydroponic system were used to obtain the mini-cuttings.For the first experiment,the mini-cuttings were stored at 4℃ for 0(immediate planting),24,48,72,96 and 120 h.The second experiment evaluated the rooting dynamic to determine the optimal time of permanence for minicuttings in a greenhouse.The basal region of the mini-cutting was treated with various IBA solutions:0(free of IBA),1,000,2,000,3,000 and 4,000 mg·L-1.Every seven days(0(immediate planting),7,14,21 and 28 days),destructive sampling of the mini-cuttings was performed to evaluate the histology of the adventitious rooting.Eucalyptus benthamii minicuttings should be rooted immediately after the collection of the shoots.The 2,000 mg·L-1 IBA concentration induced a greater speed and percentage of adventitious rooting,and an interval of 35 to 42 days was indicated for permanence of the mini-cuttings in the greenhouse.Exposure to low temperature induced adventitious root formation with diffuse vascular connections.  相似文献

Khaya anthotheca is a hardwood species from Africa and recently introduced to Brazil. This species yields high-quality wood for diverse applications, but little has been done toward an effective propagation method for large-scale production in nurseries. We evaluated the effects of different concentrations of macro and micronutrients (i.e., 100, 50 and 25% of the concentration of a referenced solution) combined with indole-3-butyric acid (IBA) at 0 and 2 g L?1 on the survival rate of mini-stumps and the survival and adventitious rooting of K. anthotheca mini-cuttings. The mini-stumps were grown from a source of seeds imported from Ghana, West Africa. The mini-stumps survived at a high rate (97%). Consistently high shoot yields were obtained from the mini-stumps (average of 1.8 per mini-stump). High survival percentage of mini-cuttings and adventitious rooting were observed when solutions with 100 and 50% of the nutrients concentration were combined with 2 g L?1 IBA. Histological sections analyzed through optical microscopy indicated the tissues from mini-cutting stems were juvenile, which might have stimulated adventitious rooting. These results have important implications for further work aimed at establishing propagation strategies for K. anthotheca, which are of prime importance for assisting breeding programs of this species.  相似文献

Eucalyptus globulus Labill and hybrids thereof have low lignin content, favoring cellulose extraction, but are often recalcitrant to clonal propagation. This work analyzed biochemical and morphological changes during adventitious rooting of mini-cuttings of E. globulus × maidenni obtained from mini-stumps cultured in drip fertigated sand bed or intermittent flooding tray commercial propagation systems. Morphological (% rooting, root number and length, mean rooting time) and biochemical parameters (peroxidase activity, total phenolic content and flavonoid content) were monitored to characterize the rooting phases. All of the rooting parameters were equivalent in both systems, indicating comparable efficiency of both methods in clonal propagation. Kinetic profiles of biochemical parameters were also similar, although the activity of peroxidases was an order of magnitude higher and the phenolic content about three times lower in cuttings derived from intermittent flooding-grown mini-stumps than in those derived from sand bed-grown mini-stumps. Taken together, results suggest that rooting phases were similar in both systems: induction before day 5, formation from day 5 to 15, and elongation from day 15 to 45. These data may contribute to the development of rooting phase-specific mineral nutrient solutions to maximize clonal propagation and plant survival.  相似文献

In the Nordic countries, Norway spruce (Picea abies) is a major species in tree breeding. In order to facilitate breeding work and availability of highly bred forest regeneration material, the time required for breeding and implementation of results should be shortened. This could be done by accelerating production of clonal material for field testing, and possibly for planting stock, by combining production of rooted cuttings with somatic embryogenesis (SE). This would allow efficient production of numerous plants of the same genotype, with equal age and propagation history between genotypes. In the present work, we studied the rooting potential of cuttings from Norway spruce emblings. Altogether 36 clones from 12 families representing elite breeding materials and ornamental forms were examined under different rooting conditions (container type and rooting media) in 2015 and 2016. Our results show that Norway spruce emblings are good donors for cuttings. Best combination (peat–vermiculite mixture and Plantek 81f containers) resulted in 91% rooting, variation among the tested clones of elite breeding materials being 55–100% per treatment. The rooting variation between families is acceptable for breeding purposes. High rooting (87–96%) of ornamental forms indicates propagation potential with the combination of SE and rooted cuttings.  相似文献

Many studies have assumed that plant terpenes favor fire due to their enormous flammability. However, only a few of them, all performed on green leaves, have demonstrated this. In the present work we investigated the question of whether litter terpene content can be used to estimate flammability and temperatures reached during fire. Epiradiator and burn table tests were used to compare flammability of leaf litter of P. pinaster, P. halepensis, P. pinea, C. albidus, C. ladanifer, C. laurifolius and the mixture of litter of P. pinaster with that of the other five species (e.g. P. pinaster + P. halepensis). Tests with burn table showed increasing spread rates and shorter combustion times under higher terpene contents. Flame height was triggered both with higher a terpene content and bed thickness, whereas the percentage of burned biomass was only significantly correlated to bed height. Epiradiator tests indicated that terpene concentration in leaf litter was positively correlated to flame height and negatively correlated to both flame residence time and ignition delay. Flammability was high for P. pinaster, P. halepensis, and hence for P. pinaster + P. halepensis, intermediate for C. albidus, P. pinea and P. pinaster combined with each of these species, and low for C. laurifolius, C. ladanifer and P. pinaster combined with them. Accordingly, their terpene content was high, intermediate and low. We concluded that plants might influence fire intensity, by having stored terpenes in their dead leaves, in addition to having developed traits to survive fire. Thus, a correct management of dead aboveground fuels rich in terpene concentrations, such as those of P. pinaster and P. halepensis, could prove helpful in reducing the hazard of fire.  相似文献

The main objective of our clonal propagation program is the production of high quality timber and faster tree growth. Teak (Tectona grandis Linn. f.) is an important plantation species in the tropics but the demand of teak timber is increasing globally. Teak planting stock is still produced from seeds of unselected elite genotypes which may lead to poor stand establishment. The number of seeds per fruit and their germination ability are also variable and limited by several factors. Therefore an alternative method i.e., clonal propagation is required for mass multiplication of elite trees. It is an established fact that the clonal propagation or adventitious root formation in cuttings is markedly affected by several external and internal factors. Considering these, an experiment was conducted on leafy soft wood shoot cuttings of 3-year-old hedged teak stock plants grown in a hedge garden to study the effects of branch position and auxin treatment on adventitious root formation. A factorial completely randomized design was used for experimentation. Coppice shoots were collected from basal, middle and upper positions of the hedge garden; and mono-nodal leafy cuttings were prepared. These cuttings were treated with different concentrations of IBA and NAA; and were cultured in a mist chamber for rooting where the relative humidity was maintained at 85 ± 2% with maximum and minimum day-night temperature at 32 ± 1°C and 26 ± 1°C respectively. After 45 days the cuttings were removed from the rooting medium and observations were made. It was observed that the rooting parameters viz., percent rooting and percent sprouting, mean number of leaves, mean number of shoots and length, and mean number of roots and length per cutting were significantly influenced by the branch position and auxin treatment. Treatment with 4000 ppm IBA increased percent rooting and percent sprouting whereas NAA suppressed it. Treatment with 4000 ppm IBA also increased the mean number of leaves and shoots, and the length, and the mean number of roots per cutting. The highest mean number of roots and length were observed in cuttings taken from the branch emerging at the upper position followed by branches of middle and lower position. Cuttings of branches at a middle position showed the highest values of percent rooting, percent sprouting, mean number of leaves and shoots and mean shoot length per cutting. Furthermore, the interactive effect of branch position and auxin treatment on rooting and sprouting response was also found to be significant. Overall, the findings of this investigation suggest that middle branch position in hedge plants and 4000 ppm IBA can be made to induce maximum rooting and to produce high quality planting stock material for clonal forestry program of teak.  相似文献

Abstract

One-year-old Pinus sylvestris donor plants were top-pruned in early May, allowing cuttings of three types to develop: shoots from adventitious buds near the top of the plant (T), weaker shoots from adventitious buds emerging further down the stem (M) and shoots from normal buds on branches (B). Cuttings from 46 half-sib families were collected at the beginning of August, treated with indole-butyric acid, immediately inserted in containers with porous substrate and put in a greenhouse, where soil was heated and high air humidity was maintained. The rooting frequency, estimated by counting living plants 10 months after insertion, was 54%. The family variance component for rooting was small and non-significant, while the clone-within-family component was strongly significant and accounted for 21% of the total random variance. Type M and type B cuttings showed significantly better rooting ability than type T cuttings. Including only plants with at least four acceptable shoots in the propagation experiment, 61% of the potential donor plants remained, without loss of any family. If a more restricted selection criterion was applied, including only plants with at least eight acceptable shoots, 21% remained, with a loss of 13 families. It was concluded that rooting ability of P. sylvestris can be brought to a level that could justify its inclusion in a breeding programme. The lack of family variation was also favourable in this respect, as no families were lost owing to low rooting response. However, the number of suitable cuttings produced by the donor plants needs to be increased.  相似文献

By propagating lodgepole pine (Pinus contorta) cuttings in vivo, we obtained after 7 growing cycles (ca 3.5 years) in a greenhouse, sufficient number of cuttings from most families to establish clonal progeny tests. Twenty-one full-sib families with approximately 20 clones per family were studied for five years. Years when cuttings were set, families within latitude and clones within families differed significantly in rooting percentages, with the variance components 4.2%, 8.2% and 9.5%, respectively. One way to get a frequent and uniform rooting is to take cuttings from non-leading shoots since they have higher rooting percentage than leading shoots. Neither total length of the cuttings nor length of the primary needles were significantly correlated to rooting percentage. With appropriate management of the ortets and the cuttings during rooting, most clones could be included in a cutting propagation program.  相似文献

We assessed different rootstocks and grafting techniques to clonally propagate mastic tree (Pistacia lentiscus var. chia). Mastic tree is a dioecious tree but the quantity and quality of the resin secreted by female trees are very low; therefore, male trees that are clonally propagated are used to establish commercial plantations for mastic production. Conventional long branch cuttings for clonal production requires too much materials and has a very low rooting rate. Rooting with tissue culture and green cuttings has also failed outdoors. Grafting of mastic on other Pistacia species has not been tried so far; therefore, this study was aimed at developing grafting methods for the clonal propagation of mastic. P. atlantica and P. lentiscus rootstocks were grafted from 15 February to 15 October every 15 days using three grafting methods and a two-factor randomized block trial design. Early spring (15 February–15 March) proved to be the best time for grafting using either of the rootstocks. The highest grafting success was attained using the scions grafted on P. atlantica from 15 February to 15 March. The best bud-burst percentage was obtained using P. atlantica and P. lentiscus rootstocks.  相似文献

Total dry mass significantly differed between watering treatments but it did not among populations. Populations could be clustered into four main groups.     Seedlings of four populations each of Pinus pinaster, P. halepensis, P. canariensis and P. pinea were grown in controlled conditions to evaluate both inter- and intra-specific differences in response to watering. We submitted half of the plants to a moderate water stress and after 22 weeks, we recorded height, stem diameter and root, stem and leaves dry weight. Patterns and amounts of phenotypic changes, including changes in biomass allocation, were analysed. We found a scant response in P. canariensis, P. pinaster and P. halepensis presented high population divergence for phenotypic changes, and P. pinea showed marked allocational shifts and no population divergence. The phenotypic changes observed within species are interpreted as a plastic response. The variation encountered within P. halepensis and P. pinaster may be indicative of specialisation to either resource-rich or resource-poor habitats, being populations from favourable sites more plastic. P. pinea exhibited a very uniform plastic response, indicating generalist behaviour.  相似文献

    Propagation of subtropical eucalypts is often limited by low production of rooted cuttings in winter. This study tested whether changing the temperature of Corymbia citriodora and Eucalyptus dunnii stock plants from 28/23°C (day/night) to 18/13°C, 23/18°C or 33/28°C affected the production of cuttings by stock plants, the concentrations of Ca and other nutrients in cuttings, and the subsequent percentages of cuttings that formed roots. Optimal temperatures for shoot production were 33/28°C and 28/23°C, with lower temperatures reducing the number of harvested cuttings. Stock plant temperature regulated production of rooted cuttings, firstly by controlling shoot production and, secondly, by affecting the ensuing rooting percentage. Shoot production was the primary factor regulating rooted cutting production by C. citriodora, but both shoot production and root production were key determinants of rooted cutting production in E. dunnii. Effects of lower stock plant temperatures on rooting were not the result of reduced Ca concentration, but consistent relationships were found between adventitious root formation and B concentration. Average rooting percentages were low (1–15% for C. citriodora and 2–22% for E. dunnii) but rooted cutting production per stock plant (e.g. 25 for C. citriodora and 52 for E. dunnii over 14 weeks at 33/28°C) was sufficient to establish clonal field tests for plantation forestry.  相似文献

    The pinewood nematode (PWN), Bursaphelenchus xylophilus, the causal agent of the pine wilt disease, has been detected in several regions of Portugal affecting Pinus pinaster, a coniferous species of a great economic value. The nematodes, migrating through resin canals and feeding on parenchyma cells, induce rapid metabolic changes in ray parenchyma cells, cavitation areas, and denaturation and necrosis of parenchyma and cambial cells. To understand how anatomic changes and biochemical incidences of tree defense reactions affect the technological parameters of the wood, the gross calorific value (GCV) and chemical composition of PWN-infected and -uninfected P. pinaster wood were evaluated. The GCV was determined using Parr 6300 Automatic Isoperibol Calorimeter, and chemical composition analysis was performed by determining the contents of carbon, hydrogen, nitrogen, sulfur and Ash on complete and instant oxidation of samples by “flash” combustion. The Student’s t test with Welch correction was used for statistical data analysis. The difference between the GCV and chemical composition for PWN-infected and -uninfected P. pinaster wood was statistically significant for the GCV and for hydrogen and nitrogen contents. The carbon, oxygen, sulfur and Ash contents did not differ statistically. The GCV of PWN-infected wood varied between the highest value of hardwood and the lowest value of softwood. This interdisciplinary study stresses the important technological and economic aspects, namely the impact of PWN on wood properties and the suitability of infected P. pinaster wood for use in the wood-processing and energy industries.  相似文献

    To assess the potential to integrate somatic clones (SC) of desired characteristics in production of high genetic quality seed, controlled crosses between different SCs of black spruce (Picea mariana (Mill) B.S.P.) were used to assess their suitability for the production of viable pollen, cones, seeds and seedlings. These SC produced male and female strobili at an early stage. Pollen, cones and seeds produced were characterized (mass, size, germination); their characteristics were similar to those produced by trees in natural forests or seed orchards. A maternal effect was demonstrated for the cone size and seed mass. Although seeds had excellent germination rates, the somatic biparental crosses were divided into three distinct groups with different germination curves using the Weibull function. Seeds from controlled crosses between different SC enabled the production of high morpho-physiological quality seedlings in a forest nursery. Using black spruce as a model, we showed, for the first time, that SC can be used as seed producers. These encouraging results open new perspectives on the tangible integration of somatic embryogenesis (SE) in the chain of seed, vegetative propagation (cuttings and SE) and production of plants for high productivity plantations. Controlled crosses can be made between SC with the desired characteristics (fewer large branches, fewer nodes, good growth, high wood density, performance, improved yield, etc.), vegetative propagules produced and deployed to clonal tests. After elimination of the worst performing SC, clonal tests can be converted into seed orchards that produce a new generation of seeds of high genetic quality. This will allow the rapid introduction of new materials in elite breeding programs of forest species.  相似文献

    The results of bulk propagation of Norway spruce for one year are presented. The number of cuttings produced per seedling ranged from 21 to 38 between different families, with a large within-family variation. No clear relationship between family field performance and production of cuttings could be seen. Established cuttings produced more propagules per plant than seedlings in the first propagation. Misting gave about 92% rooting, with no significant differences between different rooting media used. A bulk propagation programme is proposed with a potential production of about 1000–1500 plants per germinated seed within 3 years of sowing.  相似文献

    Pterocarpus santalinoides is a multipurpose tree species, native to Africa and South America. However, excessive pruning for fodder is jeopardising its seed production leading to poor regeneration in the Sahel region. Efficient vegetative propagation techniques may overcome this concern. To address this issue, four experiments were conducted to determine the effects of substrate (Experiment 1), cutting type (Experiments 2 and 3), genotype (Experiment 4) and auxins (Experiments 1, 2, and 4) on rooting ability in a non-mist propagating unit in Mali. Among the four tested substrates, no significant differences were found in rooting parameters, including the percentage of rooted cuttings, the number of roots and secondary roots per rooted cutting, and the length of the longest root per rooted cutting. Semi-hardwood cuttings rooted significantly better than softwood cuttings with a higher percentage of rooted cuttings (31.7?±?6.5% vs. 9.2?±?2.7%), greater number of roots (8.2?±?4.8 vs 1.5?±?0.7), greater number of secondary roots (16.7?±?4.2 vs 4.6?±?1.8) and longer root length (17.8?±?12.0?cm vs 2.3?±?1.0?cm) per rooted cutting. In addition, one-node cuttings cut below the second and third nodes, respectively, had the highest rooting success (88% and 90%, respectively) followed by cuttings cut below the fourth node (68%) and the first node (62%). Significant differences in the percentage of rooted cuttings were found among 12 randomly selected genotypes with rooting varying from 2% to 75%.The results also showed that auxin application, especially IBA, had a significant effect on rooting success, the number of roots and secondary roots per rooted cutting. Single-node cuttings, and cuttings of 10?cm length with multiple softwood and semi-hardwood nodes, can be used with or without auxin treatment for successful clonal propagation of P. santalinoides. The results of the present study indicated that P. santalinoides is amenable to clonal propagation using low-cost propagators and sand-based rooting media.  相似文献
