To study the behavior and mutation of Ralstonia solanacearum in Solanum toxicarium, which is resistant to bacterial wilt, S. toxicarium was grown in aseptic culture and inoculated with R. solanacearum. Although 60%–80% of the inoculated plants were wilting after 2 to 3 days, most wilted plants had recovered by 20 days after inoculation. The pathogen was reisolated from over 98% of inoculated plant stems, but the percentage of recovery decreased the closer the isolation sites were toward the upper stem sections. Three colony types, characterized as fluidal white, nonfluidal red, and a mixture of fluidal white and nonfluidal red, were reisolated from the stems. Nonfluidal red colonies were less virulent on tomato plants than fluidal white colonies.  相似文献

Laboratory rearing of insects for scientific research ensures a constant supply of insects. However, laboratory colonies may diverge genetically and/or phenotypically from wild populations due to selection pressures in their artificial environment. This study examined how variation in a colony ofTrichoplusia ni influenced reproducibility over time of bioassays of toosendanin (a limonoid extracted from the bark ofMelia azedarach). In four of five experiments insect growth was reduced significantly (P=0.05) by the presence of toosendanin in the diet. The fifth experiment showed the same trend, but the effect was not significant. Mean larval mass of both control and treated insects also varied greatly among experiments. The variation and distribution of virulent phenotypes ofBremia lactucae Regel, the causal agent of lettuce downy mildew, were studied during 2002–2003 in lettuce fields (Lactuca sativa) in Israel. A total of 21 isolates ofB. lactucae were collected from nine locations in three regions of Israel: Galilee, the Coastal Plain, and the Shefela. The isolates were examined for the presence of 21 virulence factors (v-factors) and their combinations with differential sets of lettuce lines/varieties. There were clear differences in v-factors, and a broad diversity of v-phenotypes among the isolates was found. Although 17 different v-phenotypes and 20 v-factors were detected, a composite of similar v-phenotypes generally occurred between isolates within the three regions. They differed mostly in the presence or absence of only a few v-factors. The Coastal Plain region averaged the highest virulence complexity (0.63), significantly different from that of the Shefela (0.45) and of Galilee (0.4). The pathogenic fungus Verticillium fungicola, responsible for dry bubble disease of the common mushroom Agaricus bisporus, causes various symptoms on its host, bubbles (undifferentiated spherical masses), bent and/or split stipes (blowout) and spotty caps. Host DNA quantification by real-time PCR was used to observed relationships between the type of symptom and the relative amount of A. bisporus and V. fungicola in diseased mushrooms. Verticillium fungicola is involved in bubble formation but does not appear to regulate its growth. Quantifications in bubbles and stipe-bubbles (morphology between bubble and sporophore with stipe blowout) showed that the pathogen has no effect on the growth of undifferentiated host hyphae but prevents morphological differentiation if not initiated and stops it when initiated hyphae are affected. Mushrooms with stipe blowout exhibiting both mature and abortive lamellae reveal that V. fungicola has a restricted area of action in host tissues. Despite their visual aspect, healthy looking parts of mushrooms showing spots or stipe blowout were actually contaminated. Discolouration and symptom development are two distinct events. The colour of the tissues was correlated to the percentage of A. bisporus DNA, suggesting that discolouration is not an efficient defensive mechanism, and occurs at the time V. fungicola developed enough to induce tissues necrosis.  相似文献

Alstroemeria plants were surveyed for viruses in Japan from 2002 to 2004. Seventy-two Alstroemeria plants were collected from Aichi, Nagano, and Hokkaido prefectures and 54.2% were infected with some species of virus. The predominant virus was Alstroemeria mosaic virus, followed by Tomato spotted wilt virus, Youcai mosaic virus (YoMV), Cucumber mosaic virus (CMV), Alstroemeria virus X and Broad bean wilt virus-2 (BBWV-2). On the basis of nucleotide sequence of the coat protein genes, all four CMV isolates belong to subgroup IA. CMV isolates induced mosaic and/or necrosis on Alstroemeria. YoMV and BBWV-2 were newly identified by traits such as host range, particle morphology, and nucleotide sequence as viruses infecting Alstroemeria. A BBWV-2 isolate also induced mosaic symptoms on Alstroemeria seedlings.  相似文献

Phytophthora nicotianae and P. palmivora are the most important soil-borne pathogens of citrus in Florida. These two species were detected and identified in singly and doubly infected plants using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) of internal transcribed spacer (ITS) regions of ribosomal DNA. The sensitivity of the PCR-RFLP was analyzed and the usefulness of the method evaluated as an alternative or supplement to serological methods and recovery on semi-selective medium. In a semi-nested PCR with universal primers ITS4 and ITS6, the detection limit was 1 fg of fungal DNA, which made it 1000× more sensitive than a single-step PCR with primers ITS4 and DC6. The sensitivity of detection for P. nicotianae was shown to be ten-fold lower than for P. palmivora, limiting its detection with restriction profiles in plants infected by both fungal species. Phytophthora nicotianae was detected with species-specific primers in all samples inoculated with this species despite the absence of species-specific patterns in RFLP. In contrast, the incidence of detection of P. palmivora in the presence of P. nicotianae was considerably lower using plating and morphological detection methods. Due to its high sensitivity, PCR amplification of ribosomal ITS regions is a valuable tool for detecting and identifying Phytophthora spp. in citrus roots, provided a thorough knowledge of reaction conditions for the target species is established prior to the interpretation of data.  相似文献

Tomatoes grown in soilless systems can be seriously damaged byFusarium oxysporum Schlect f.sp.radicis lycopersici (Forl) causing Fusarium crown and root rot (FCRR). FCRR suppression can be achieved through the use of chemicals, selected substrates, composts and artificially introduced antagonistic microorganisms. This study evaluated the natural capacity of a used rockwool to suppress FCRR infections. New and used rockwool, sampled from closed soilless systems, was either autoclaved or not, either artificially inoculated withForl or not and, finally, sown with tomato seeds cv. ‘Cuore di Bue’. The effects of autoclaved/non-autoclaved and used/new rockwool on FCRR incidence were assessed by evaluating the symptoms of crown rot on the root — shoot transition zone of tomato seedlings. Non-autoclaved and inoculated used rockwool significantly reduced FCRR incidence when compared with non-autoclaved and inoculated new rockwool. Autoclaved and inoculated used rockwool did not suppress FCRR, similarly to new and inoculated rockwool. These findings are in accordance with other research that, on a cucumber/Pythium host/pathogen complex in a closed rockwool soilless system, demonstrated the key role of resident microflora in suppressing the root rot disease. http://www.phytoparasitica.org posting Dec. 8, 2006.  相似文献

The susceptibility ofThaumetopoea wilkinsoni larvae toBacillus thuringiensis (Bt) formulations was screened in 2003 and 2004. Eggs and larvae were collected from pine forests in 11 geographical locations in Israel. Larval mortality bioassays were conducted with commercial formulations (Delfin WG, Dipel DF and Foray 48B) at concentrations ranging from 0.001% to 0.1%. Significant differences in susceptibility toBt were recorded among populations that were treated withBt intensively, frequently, or never. The mortality recorded in a population that was never treated withBt was twice that in an intensivelyBt-treated population. Twelve isolates of Japanese Phytophthora infestans, which differed from the major genotypes US-1, JP-1, JP-2, and JP-3, were analyzed for RG57 fingerprints, mtDNA haplotypes, two allozyme genotypes, and mating types. Genotypes JP-1.1, JP-2.1, JP-2.2, JP-3.1, and JP-4 were newly defined. JP-1.1 and JP-2.1 were isolated discontinuously from potato fields in several years, and JP-1.1 was found in Hokkaido and Kagoshima. These results show that some minor genotypes can overwinter and disperse from their original site.  相似文献

Freshly harvested Striga asiatica L. seeds will germinate in response to a stimulant only after the passage of time, an after-ripening period, and exposure to moisture at a suitable temperature, a conditioning period. To investigate the role of seed moisture content in the regulation of the after-ripening period, seeds were placed in chambers having specific relative humidity of 6%, 14%, 33%, 75% and 91% for 30, 60, 90 and 150 days. The seeds were then conditioned and germination percentage, response to tetrazolium and seed moisture contents were measured. Seeds at moisture contents less than 10% at the start of conditioning had germination of greater than 93%. Seeds at moisture contents over 10% at the start of conditioning could germinate between 60% and 3%, with germination decreasing as seed moisture content at the start of conditioning increased. The highest moisture content (17%) and lowest germination percentage (3%) occurred in seeds stored at 91% relative humidity for 150 days. There was a linear relationship of a high degree of correlation (0.997) between a positive tetrazolium test and germination capacity. Germination capacity of seeds could be changed from 90% to 3% by prolonged storage in water (dilute benomyl solution), causing `wet dormancy', then returned to 90% germination by returning to dry storage. Seed moisture content at the beginning of conditioning appears to control the responsiveness of the seeds to germination stimulants. The implications of these findings to the control of the parasite are discussed.  相似文献

Different sets of wheat genotypes were tested under field conditions by spraying inocula of isolates of seven Fusarium spp. and Microdochium nivale (formerly F. nivale) in the period 1998–2002. The severity of Fusarium head blight (FHB), Fusarium-damaged kernels (FDK), the yield reduction and the deoxynivalenol (DON) contamination were also measured to describe the nature of the resistance. The degrees of FHB severity of genotypes to F. graminearum, F. culmorum, F. avenaceum, F. sporotrichioides, F. poae, F.␣verticillioides, F. sambucinum and M. nivale were very similar, indicating that the resistance to F.␣graminearum was similar to that for other Fusarium spp. listed. This is an important message to breeders as the resistance relates not only to any particular isolate of F. graminearum, but similarly to isolates of other Fusarium spp. This holds true for all the parameters measured. The DON contamination refers only to DON-producers F. graminearum and F. culmorum. Highly significant correlations were found between FHB, FDK, yield loss and DON contamination. Resistance components such as resistance to kernel infection, resistance to DON and tolerance were identified in the more susceptible genotypes. As compared with western European genotypes which produced up to 700 mg kg−1 DON, the Hungarian genotypes produced only 100 mg kg−1 at a similar FDK level. This research demonstrates the importance of measuring both FDK and DON in the breeding and selection of resistant germplasm and cultivars.  相似文献

Mycosphaerella pinodes is one of the most devastating pea pathogens. Pea cultivars with adequate levels of resistance to control the disease are not so far available. However, promising levels of resistance have been identified in wild accessions of pea. In the present investigation the inheritance of resistance to M. pinodes was studied in two crosses between the susceptible pea cv. ‘Ballet’ and the partially wild resistant accessions P665 (Pisum sativum subsp. syriacum) and P42 (P. sativum subsp. sativum var. arvense). Both additive and dominant effects were important in control of resistance and susceptibility dominated over resistance.  相似文献

During a 6-year study, grapevine propagation materials and young grapevines were analysed to evaluate the presence of internal wood discolouration and the occurrence of fungal species involved in Petri disease. The intensity of wood discolouration increased with the ageing of the plants. The maximum incidence of dark streaks was observed in the rootstock while necrosis originating from buds or nodes were notably present in the trunk and cordon of older vines. In contrast, the highest levels of brown-red halo symptoms, defined as discoloured areas around the pith, were recorded in the early growth stages. Phaeoacremonium spp. and Phaeomoniella chlamydospora were usually isolated from the rooted-grafts and the 3-year old plants, respectively. The number of infected grapevines increased with age. Most of the P. chlamydospora strains were isolated from dark streaks or dots, while Phaeoacremonium spp. were detected in brown-red halo symptoms and other symptomatic or asymptomatic wood. The greatest incidence of the two fungal taxa was recorded in the lower parts of the grapevine, including the roots and rootstock.  相似文献

Xanthomonas oryzae pv. oryzae strain T7174R is lysed by bacteriophage OP1h and OP1h2. Three mutants tolerant to both OP1h and OP1h2 were isolated by transposon mutagenesis. The mutants had an insertion of the transposon in XOO1687, which is predicted to encode a TonB-dependent receptor gene. Plasmid pHMIroNB that contained XOO1687 of T7174R was constructed, and the mutant was transformed with the plasmid. The transformant recovered sensitivity to OP1h and OP1h2. Electron microscopic analysis demonstrated that OP1h and OP1h2 can adsorb to the wild type and the transformant, but they could not adsorb to the phage-tolerant mutant. These results suggest that the TonB-dependent receptor gene relates to adsorption and infection of T7174R by OP1h2 and OP1h. Y. Inoue and S. Tsuge have contributed equally to this work.  相似文献

The influence of the root-knot nematode Meloidogyne incognita (Kofoid & White) Chitwood on the competitive relationships between pepper (Capsicum annuum L.) and Solanum nigrum L. (black nightshade) was investigated in a glasshouse experiment. Seven competition treatments were set up: two intraspecific for the crop and the weed and five interspecific treatments in which the emergence of S. nigrum plants was progressively delayed with respect to that of the pepper. Nematodes reproduced in every inoculated plant and their multiplication rates were high in both pepper and S. nigrum. The parasite reduced all growth and yield parameters of the crop, but did less harm to the weed. The negative effect of S. nigrum on pepper peaked in the treatment in which the weed and pepper plants emerged together. S. nigrum was a stronger competitor than pepper under both nematode-infested and nematode-free conditions. The effect of nematodes on pepper yield was less than that of competition, but both appeared to be additive.  相似文献

The red spider miteTetranychus evansi (Acari: Tetranychidae) Baker and Pritchard was recorded for the first time in Greece, in the area of Tympaki (south-central Crete) onSolanum nigrum. T. evansi is a pest of crops of the family Solanaceae, which are grown extensively in Crete. The species identification was based on both morphological and molecular data. The second internal transcribed spacer was PCR amplified and sequenced in samples from Crete. Sequences were compared with the sequence ofT. evansi from Brazil and with the ITS2 sequences (retrieved from GenBank) of the two closely related tetranychid species most commonly found in Greece,Tetranychus turkestan andTetranychus urticae.  相似文献
