An isolate of Puccinia coronata f.sp. avenae with virulence against the oat crown rust resistance gene Pc94 was discovered in oat fields in Córdoba, Spain. In order to identify alternative sources of resistance to this virulent isolate, a collection of 159 Avena accessions, including 107 A. sativa and 29 A. byzantina landraces and 23 commercial A. sativa and A. strigosa cultivars, was screened. Eight resistant landraces and four cultivars were selected according to macroscopic assessment to further characterize the operative defence mechanisms. Histological studies showed a range of defence mechanisms, acting alone or in combination, which impeded fungal development at different stages. Some accessions allowed reduced fungal growth before mesophyll penetration. In others, the fungus was arrested at the penetration stage by mesophyll cell wall strengthening and/or papilla deposition. Mesophyll cells of several accessions were penetrated by the fungus, but then the hypersensitive response (HR) leading to cell death hampered fungal development. In some cases cell death was very fast and colonies aborted early, whereas in other cases necrosis was observed later and associated with numerous secondary hyphae, suggesting a slow HR. Characterization of defence mechanisms will be useful for breeding programmes and for further cellular and molecular studies to unravel the bases of resistance. Commonalities with the resistance of the same oat collection to powdery mildew are discussed.  相似文献

Thirty-nine genotypes ofCucumis melo (plant introduction entries, open-pollinated cultivars and F1 hybrids) were evaluated for resistance to powdery mildew under either natural field conditions or artificial inoculation in growth chambers at the cotyledonary stage and the 2-true-leaf stage. Results confirmed that susceptibility in cotyledons was not necessarily associated with susceptibility in either true leaves in growth chambers or adult plants in the field. However, resistance at the 2-true-leaf stage in growth chambers was highly correlated with resistance of field-grown plants. Results also showed that 20 muskmelon genotypes resistant to race 1 at the cotyledonary stage were also resistant at the 2-leaf-stage and as adult plants in the field. The same was true for ten genotypes with race 2 inoculations. Because muskmelon genotypes expressing resistance in cotyledons were also resistant in true leaves in growth chambers or the field, the use of plants at the cotyledonary stage is recommended for screening for powdery mildew resistance caused by race 1 or race 2 ofS. fuliginea. When cotyledons are susceptible, screening should be done at the 2-true-leaf stage.  相似文献

Powdery mildew caused by Blumeria graminis DC. f. sp. avenae Em. Marchal. is a deleterious foliar disease of common oat which occurs in many parts of the world. Resistance existing in oat cultivars is broken down by new races of this pathogen and because of this fact there is necessity to look for new and effective sources of resistance to powdery mildew in oat. To identify new potential resistance sources to powdery mildew, a total of 67 oat genotypes from five different species Avena sterilis, A. fatua, A. sativa, A. maroccana and A. murphyi and 20 powdery mildew isolates were investigated for disease reactions. Among tested oat accessions the highest level of resistance was associated with A. maroccana genotypes. Genotypes belonging to species of A. murphi and A. sterilis also showed a good level of resistance. Identified genotypes could be used in oat breeding programmes to improve levels of resistance to powdery mildew.  相似文献

Powdery mildew, caused by Blumeria graminis f. sp. hordei (Bgh), is a worldwide disease problem on barley (Hordeum vulgare) with potentially severe impact on yield. Historically, resistance genes have been identified chiefly from cultivated lines and landraces; however, wild barley (H. vulgare subsp. spontaneum) accessions have proven to be extraordinarily rich sources of powdery mildew resistance. This study describes the characterization of a collection of 316 wild barley accessions, known as the Wild Barley Diversity Collection (WBDC), for resistance to powdery mildew and the genetic location of powdery mildew resistance loci. The WBDC was phenotyped for reaction to 40 different Bgh isolates at the seedling stage and genotyped with 10 508 molecular markers. Accessions resistant to all 40 isolates of Bgh were not found; however, three accessions (WBDC 053, 085 and 089) exhibited resistance to 38 of the isolates. Gene postulation analyses revealed that many accessions, while resistant, contained none of the 12 genes present in the Pallas near‐isogenic lines Mla1, Mla3, Mla6, Mla7, Mla9, Mla12, Mla13, Mlk1, MlLa, Mlg, Mlat and Ml(Ru2), suggesting that the accessions carry novel genes or gene combinations. A genome‐wide association study of powdery mildew resistance in the WBDC identified 21 significant marker‐trait associations that resolved into 15 quantitative trait loci. Seven of these loci have not been previously associated with powdery mildew resistance. Taken together, these results demonstrate that the WBDC is a rich source of powdery mildew resistance, and provide genetic tools for incorporating the resistance into barley breeding programmes.  相似文献

Powdery mildew (Blumeria graminis f. sp. tritici) results in serious economic loss in wheat production. Exploration of plant resistance to wheat powdery mildew over several decades has led to the discovery of a wealth of resistance genes and quantitative trait loci (QTLs). We have provided a comprehensive summary of over 200 powdery mildew genes (permanently and temporarily designated genes) and QTLs reported in common bread wheat. This highlights the diverse and rich resistance sources that exist across all 21 chromosomes. To manage different data for breeders, here we also present a bridged mapping result from previously reported powdery mildew resistance genes and QTLs with the application of a published integrated wheat map. This will provide important insights to empower further breeding of powdery mildew resistant wheat via marker-assisted selection (MAS).  相似文献

Abstract

Cercospora leaf spot, caused by Cercospora canescens and powdery mildew, caused by Erysiphe polygoni are two of the most important fungal diseases of mungbeans. Nearly 4000 accessions of the global mungbean collection at the Asian Vegetable Research and Development Center were screened for resistance to these two pathogens. Less than 4% and 12% of the accessions showed resistance to Cercospora leaf spot and powdery mildew, respectively. The level of resistance to Cercospora leaf spot varied and highly resistant lines were not found. A few lines were rated highly resistant to powdery mildew over several years but others were rated moderately resistant or susceptible in other years.  相似文献

为西北农林科技大学小麦新育成品种(系)在黄淮麦区的大面积推广,该研究对83份西农新育成的小麦品种(系)进行苗期抗条锈病和白粉病鉴定,成株期抗条锈病、白粉病、叶锈病和赤霉病鉴定,并在田间自然环境下对其抗性进行鉴定及对相关抗病基因进行分子检测。结果显示,在苗期人工接种鉴定中,有63、29和16份小麦品种(系)分别对条锈菌Puccinia striiformis f.sp.tritici生理小种CYR32、CYR33和CYR34表现出抗性,9份小麦品种(系)对3个条锈菌生理小种均表现出抗性;有10、3和0份小麦品种(系)分别对白粉菌Blumeria graminis f.sp.tritici生理小种E15、E09和A13表现出抗性。在成株期人工接种鉴定中,有23、15、28和62份小麦品种(系)分别对条锈病、白粉病、叶锈病和赤霉病表现出抗性。在83份小麦品种(系)中有6份在苗期和成株期均对小麦条锈病表现出抗性。在田间抗性鉴定中,有57、6、65和40份小麦品种(系)分别对条锈病、白粉病、赤霉病及叶锈病表现出抗性。在83份小麦品种(系)中,3份含有Yr5基因,22份含有Yr9基因,3份含有Yr17基因,2份含有Pm24基因,14份含有Lr1基因,所占比例分别为3.6%、26.5%、3.6%、2.4%和16.8%。  相似文献

The cultivated tomato ( Lycopersicon esculentum) is susceptible to powdery mildew ( Oidium lycopersicum). Six accessions of three related Lycopersicon species show high levels of resistance (Lindhout et al., 1994b). The present research aimed at describing the development of O. lycopersicum on susceptible cv Moneymaker and characterizing the defence response to O. lycopersicum in Lycopersicon accessions by histological analysis. Spore germination and (primary) haustorium formation in resistant accessions were as frequent as in the susceptible L. esculentum cv Moneymaker. A high frequency of necrosis of epidermal cells in which a haustorium was formed appeared to be the major defence response, indicating that resistance to O. lycopersicum in the Lycopersicon genus was predominantly based on the hypersensitive reaction. However, the resistance in L. parviflorum was less associated with hypersensitivity than in other resistant accessions, suggesting the existence of a different but still unknown resistance mechanism. In addition, evidence is provided that the level of resistance could depend on the genetic background and the plant age.  相似文献

Lettuce (Lactuca sativa) is the major leafy vegetable that is susceptible to powdery mildew disease under greenhouse and field conditions. Quantitative trait loci (QTLs) for resistance to powdery mildew under greenhouse conditions were mapped in an interspecific population derived from a cross between susceptible L. sativa cultivar Salinas and the highly susceptible L. serriola accession UC96US23. Four significant QTLs were detected on linkage groups LG 1 (pm‐1.1), LG 2 (pm‐2.1 and pm‐2.2) and LG 7 (pm‐7.1), each explaining between 35 to 42% of the phenotypic variation. The four QTLs are not located in the documented hotspots of lettuce resistance genes. Alleles for the disease resistance at the four QTLs originated from both parents (two from each), demonstrating that even highly susceptible accessions may provide alleles for resistance to powdery mildew. These QTLs appeared to operate during limited periods of time. Results of the field trials with F2:3 and F3:4 families derived from a Soraya (moderately resistant) × Salinas cross demonstrated effective transfer of resistance to powdery mildew in this material. An integrated rating approach was used to estimate relative levels of resistance in 80 cultivars and accessions tested in a total of 23 field and greenhouse experiments. Generally, very low resistance was observed in crisphead‐type lettuces, while the highest relative resistance was recorded in leaf and butterhead types. Comparison of two disease assessment methods (percentage rating and the area under the disease progress steps, AUDPS) for detection of QTLs shows that the two approaches complement each other.  相似文献

Fusarium oxysporum f. sp. pisi (Fop) is an important pathogen of field pea (Pisum sativum) worldwide. The constant evolution of the pathogen drives the necessity to broaden the genetic basis of resistance to Fop. To achieve this, it is important to have a large germplasm collection available and an accurate and efficient method for disease assessment. Here, a detailed evaluation method coupling disease incidence, disease rating over time and its related area under the disease progression curve (AUDPC) was established and used to screen a Pisum spp. germplasm collection against one isolate of Fop race 2. A large variation in the disease response of specific pea accessions ranging from highly resistant to susceptible was observed within the collection, indicating the quantitative expression of the resistance. The repetition of the inoculation experiments on a subset of 19 accessions, including two susceptible accessions, indicated that the scoring method was robust and reproducible and confirmed the highly resistant phenotypes of 11 accessions. To initiate the characterization of resistance mechanisms within these accessions, the external and internal stem symptoms were compared between these selected pea accessions, together with the extent of fungal colonization within plants. All these tests indicated that, in all resistant accessions, the resistance mechanisms efficiently stopped pathogen progression at the crown. Incorporation of these sources of resistance to breeding programmes will contribute to improved Fop resistance in pea cultivars.  相似文献

Triticale is the intergeneric hybrid between wheat and rye. With the expansion of the triticale growing area, powdery mildew has emerged and become a significant disease on this new host. Recent research demonstrated that this ‘new’ powdery mildew on triticale has emerged through a host range expansion of powdery mildew of wheat. Moreover, isolates sampled from triticale still infect their previous host, wheat, but isolates sampled from wheat hardly infect triticale. Race‐specific and adult‐plant resistance have been identified in triticale cultivars. The main objective of this study was to characterize the cellular basis of powdery mildew resistance in triticale. Commonalities with resistance responses in other cereals such as wheat, barley and oat are discussed. A detailed comparative histological study of various resistance responses during cross‐inoculation of either virulent or avirulent wheat and triticale isolates on both hosts was carried out. The present data provide evidence that for incompatible interactions, the formation of non‐penetrated papillae is the predominant resistance response, while the hypersensitive response (HR) acts as a second line of defence, to cut the fungus off from nutrients, if penetration resistance fails. It is not clear yet what causes the slower growth and reduced colony size of triticale isolates when inoculated on wheat. Possibly, post‐penetration resistance mechanisms, other than HR, are switched on during these (semi‐) compatible interactions. Molecular studies on gene expression and gene function of defence‐related genes might reveal further insights into the genetic basis of these resistance responses.  相似文献

Host genetic resistance is the most effective and sustainable means of managing tan spot or yellow spot of wheat. The disease is becoming increasingly problematic due to the adoption of minimum tillage practices, evolution of effector‐mediated pathogenicity, and widespread cultivation of susceptible cultivars from a narrow genetic base. This highlights the importance of broadening the diversity of resistance factors in modern breeding germplasm. This study explored 300 genetically diverse wheat accessions, originally sourced from the N. I. Vavilov Institute of Plant Genetic Resources (VIR), St Petersburg, Russia. The collection was screened for resistance to tan spot at seedling and adult stage under controlled conditions, and in the field across 2 years. The phenotypic datasets, coupled with ToxA bioassay screening, identified a number of accessions with useful sources of resistance. Seedling disease response corresponded well with ToxA sensitivity (= 0.49, < 0.000), but not adult responses (= ?0.02 to ?0.19, < 0.002), and overall reactions to ToxA appeared to show poor correspondence with disease response at the adult stage. ToxA‐insensitive accessions were generally found resistant across different growth stages (all‐stage resistance, ASR) in all experiments (seedling and adult stage under controlled conditions and field). ToxA‐sensitive accessions that were susceptible at seedling stage, but resistant at both adult‐plant stages, were deemed to carry adult‐plant resistance (APR). This study provides detailed information on the degree of tan spot resistance in the Vavilov wheat collection and discusses strategies to harness these sources to boost the diversity of resistance factors in modern wheat breeding germplasm.  相似文献

ABSTRACT In this work, we studied the resistance of 277 Medicago truncatula accessions against powdery mildew and further characterized the defense mechanisms of resistant plants. Ten resistant accessions were selected according to macroscopic assessment. Histological studies showed a range of defense mechanisms, acting alone or combined, that impeded fungal development at different stages. Some accessions allowed a reduced spore germination frequency compared with that of the susceptible control. In others, the fungus was arrested at penetration stage due to papilla formation. Epidermal cells of several accessions were penetrated by the fungus but then hypersensitive response (HR) leading to cell death hampered fungal development. In some cases, cell death was very fast and no haustorium could be observed in epidermal cells, whereas in others, haustoria and secondary hyphae indicated a slow HR. Finally, in some accessions in which no HR was observed, colony growth was restricted through posthaustorial defense mechanisms. Characterization of defense mechanisms will be useful for further cellular and molecular studies to unravel the bases of resistance in this species in particular and in legume-powdery mildew interaction in general.  相似文献

Aegilops sharonensis (Sharon goatgrass) is a wild relative of wheat and a rich source of genetic diversity for disease resistance. The objectives of this study were to determine the genetic basis of leaf rust, stem rust, and powdery mildew resistance in A. sharonensis and also the allelic relationships between genes controlling resistance to each disease. Progeny from crosses between resistant and susceptible accessions were evaluated for their disease reaction at the seedling and/or adult plant stage to determine the number and action of genes conferring resistance. Two different genes conferring resistance to leaf rust races THBJ and BBBB were identified in accessions 1644 and 603. For stem rust, the same single gene was found to confer resistance to race TTTT in accessions 1644 and 2229. Resistance to stem rust race TPMK was conferred by two genes in accessions 1644 and 603. A contingency test revealed no association between genes conferring resistance to leaf rust race THBJ and stem rust race TTTT or between genes conferring resistance to stem rust race TTTT and powdery mildew isolate UM06-01, indicating that the respective resistance genes are not linked. Three accessions (1644, 2229, and 1193) were found to carry a single gene for resistance to powdery mildew. Allelism tests revealed that the resistance gene in accession 1644 is different from the respective single genes present in either 2229 or 1193. The simple inheritance of leaf rust, stem rust, and powdery mildew resistance in A. sharonensis should simplify the transfer of resistance to wheat in wide crosses.  相似文献

In pea, two single recessive genes, er1 and er2, have been identified for resistance to powdery mildew caused by Erysiphe pisi, but little is known about their mode of action. Pea accessions carrying the genes er1 or er2 and other accessions displaying resistance to powdery mildew in the field were studied. In accessions carrying gene er1, epidermal cell penetration was prevented and very few haustoria or colonies were formed. Under controlled conditions, er1 conferred complete or almost complete resistance to the fungal isolates used and this resistance was not associated with macroscopically visible necrosis. Under field conditions these accessions developed a low level of disease. Resistance in line JI2480 (carrying er2) increased with temperature and leaf age, and complete resistance was expressed only at high temperature (25 °C) or in mature leaves. This resistance was based mainly on post-penetration cell death, complemented by a reduction of percentage penetration success in mature leaves. Combining the resistance provided by gene er1 and by line JI2480 into new cultivars is likely to increase their level of resistance and enhance durability of the protection.  相似文献

This study was designed to assess the reliability of grapevine leaf bioassays for predicting disease resistance on fruit in the field. The efficacy of various grapevine quantitative trait loci (QTLs) for conferring resistance to downy and powdery mildew was evaluated in bioassays and in a 2‐year field experiment for downy mildew. The resistance genes studied were inherited from Muscadinia rotundifolia (Rpv1 and Run1) and from American Vitis species through cv. Regent (QTLRgP and QTLRgD). In bioassays, genotypes carrying Run1 blocked powdery mildew development at early stages. Genotypes combining Run1 with QTLRgP displayed no greater level of resistance. For downy mildew, genotypes carrying Rpv1 and/or QTLRgD were more resistant than the susceptible cv. Merlot, and showed a high level of leaf resistance in the field (<10% severity). Disease levels on bunches were much higher than those on leaves, with a high variability between Rpv1 genotypes (1–48%). A Bayesian decision theory framework predicted that an OIV‐452 threshold of 5 in leaf bioassays allowed accurate selection of grapevine genotypes (P = 0·83) with satisfactory disease severity on bunches. Therefore, this study validates that the use of early bioassays on leaves, as currently performed by grapevine breeders, ensures a satisfactory level of resistance to downy mildew of bunches in the field.  相似文献

Thirty barley landraces collected from Morocco in 1985 and 1989, and held in the Polish Gene Bank, IHAR, Radzików, Poland, were screened for resistance to powdery mildew. Fifteen tested landraces (50%) showed powdery mildew resistance reactions and 24 single plant lines were selected. Eighteen lines originating from 13 landraces were tested with 17 isolates of powdery mildew and another six lines originating from six landraces were tested with 23; the isolates were chosen according to their virulence spectra observed on the ‘Pallas’ isolines differential set. Three lines (E 1090-2-2, E 1110-3-2 and E 1077-1-1) showed resistance to all powdery mildew virulence genes prevalent in Europe. In 21 lines, unknown genes alone or in combination with specific ones were detected. Five different resistance alleles(Mlat, Mlal, Mla3, Mlg andMl(CP)) were postulated to be present in the tested lines, alone or in combination:Mlat was postulated to be present in nine (~38%) lines;Mlg andMl(CP) in two lines, andMla1 andMla3 in one tested line each. The use of newly identified sources of resistance in barley breeding as a means of controlling powdery mildew is discussed.  相似文献

Ten oat genotypes were assessed for adult plant resistance (APR) to powdery mildew ( Erysiphe graminis f.sp. avenae ) in small field plots and on detached leaf segments in the laboratory using glasshouse-grown plants. Significant genotype differences were observed in the field and on leaf segments for latent period, infection frequency and spore production, components used to assess APR. The genotype OM1711, which was selected because it expressed a higher level of mildew resistance than its most resistant parent, cv. Maldwyn, showed this transgressive resistance in both field and laboratory tests. By contrast, the genotype OM1387, also selected for high mildew resistance, only showed this enhancement in the field. It is concluded that quantitative assessment of mildew in detached leaves is not sufficiently precise to be used alone in selecting segregants for high levels of APR, and that field data are also needed.  相似文献
