The PLGA ultrafine fibrous scaffold was successfully fabricated by electrospinning. The morphology and properties of the PLGA vascular scaffolds were examined. In particular, the in vitro degradation behavior of the electrospun PLGA vascular scaffolds was investigated by means of morphology, microstructure, mass loss, Mw, and breaking strength characterization. The results showed that electrospun scaffold possessed ultrafine fibrous and porous structure, and had adequate mechanical properties to be developed as a substitute for native blood vessels. In vitro degradation study showed that the PLGA ultrafine fibrous scaffold could biodegrade in the PBS solution, and the mass loss, Mw, and breaking strength studies indicated that degradation rate of the electrospun PLGA nanofibers was greater in the first 2 weeks. After the degradation of 2 weeks, the degradation slowed down. Furthermore, with the extension of the degradation time, the thermal decomposition temperature of the PLGA scaffold decreased gradually. The results indicated that the electrospun PLGA vascular scaffold could be considered as an ideal candidate for tissue-engineered blood vessel.  相似文献

Flexible tubular structures fabricated from solution electrospun fibers are finding increasing use in tissue engineering applications. However it is difficult to control the deposition of fibers due to the chaotic nature of the solution electrospinning jet. By using non-conductive polymer melts instead of polymer solutions the path and collection of the fiber becomes predictable. In this work we demonstrate the melt electrospinning of polycaprolactone in a direct writing mode onto a rotating cylinder. This allows the design and fabrication of tubes using 20 μm diameter fibers with controllable micropatterns and mechanical properties. A key design parameter is the fiber winding angle, where it allows control over scaffold pore morphology (e.g. size, shape, number and porosity). Furthermore, the establishment of a finite element model as a predictive design tool is validated against mechanical testing results of melt electrospun tubes to show that a lesser winding angle provides improved mechanical response to uniaxial tension and compression. In addition, we show that melt electrospun tubes support the growth of three different cell types in vitro and are therefore promising scaffolds for tissue engineering applications.  相似文献

Electrically conductive nanofibers were fabricated from elastic polyurethane (PU) and PU/multiwalled carbon nanotubes (MWCNTs) nanocomposite by electrospinning method. The nanocomposites were electrospun at various MWCNTs loading. Electron microscopy was used to investigate nanofibers morphology and dispersion of MWCNTs in the electrospun nanofibers. The results showed that the presence of the MWCNTs promoted the creation of fibrous structures in comparison with the PU without MWCNTs. On the other hand, increasing the MWCNTs content resulted in a slight increase in the average fiber diameter. TEM micrographs and mechanical properties of the electrospun mats indicated that the homogeneous dispersion of MWCNTs throughout PU matrix is responsible for the considerable enhancement of mechanical properties of the nanofiber mats. Electrical behavior of the conductive mats was also studied, in view of possible sensor applications. Cyclic experiments were conducted to establish whether the electrical properties were reversible, which is an important requirement for sensor materials.  相似文献

In this study, electrospinning of poly(ε-caprolactone) (PCL) and its optimum preparation conditions were examined in detail using various solvent systems, such as formic acid, dichloromethane/dimethyl formamide (DMF), chloroform/DMF, and dichloroethane. The average fiber diameter of the electrospun PCL mat was controlled by the solvent used with a proper concentration of PCL dope solution. Different fiber sizes (0.1, 0.8, 1.9, and 3.4 μm) of uniform PCL mats were fabricated and the effects of fiber size on surface morphology, tensile properties and cell behavior were investigated. Here, we manipulated much broader range of average fiber diameter of the mats, from nano to several micron size of fiber. It was found that the fiber diameter greatly affected topology (surface roughness) and mechanical properties of the electrospun PCL mat and consequently, they influenced the cell behavior (adhesion and proliferation) significantly. We expect that these results will provide more feasible application of electrospun PCL scaffold in tissue engineering through the co-relations in structure and property of PCL fiber mat on cell behavior.  相似文献

A series of PU fibrous membranes were fabricated by using electrospinning method. The microstructure of the membranes was characterized by field-emission scanning electron microscopy, X-ray diffraction and Fourier transform infrared spectrum. Their mechanical properties were tested by dynamic mechanical thermal analysis and stress-strain behaviors. The solution concentration, the applied voltage and the tip-collector distance had an effect on the crystallinity degree and molecular orientation of PU, the size and distribution of the fiber diameter and the point-bonded structures between the fibers, leading to the change in the microstructure and the mechanical properties of the fibrous membrane. Fibers with a smaller diameter had higher strength but lower ductility. The fibrous membranes indicated the similar stress-strain behaviors, which slopes in the initial stage were low and that in the later stage were high. The initial elastic behavior with the low Young’s modulus were attributed to the network structure of the fibrous membranes and that with the high Young’s modulus was from the electrospun PU fibers.  相似文献

In this study, to improve the cellular biocompatibility of PVP-PCL micro- and nanofiber scaffold, a novel electrospun collagen/PVP-PCL micro- and nanofiber scaffold was sucessfully prepared assisted by ultrasonic irradiation using chloroform/ethanol mixtures as solvent. The micro- and nanofibers of the electrospun PCL-PVP scaffolds still presented compact inter-fiber entanglement and three-dimensional netlike network with some certain range of pore space after introducing collagen. The incorporated collagen phase was dispersed as inclusions within the electrospun fibers, and then could be easily released by immersing the scaffold in Hanks simulated body fluid. Meanwhile, the integral triple helix structure of collagen could be maintained after blending with the PVP-PCL mixture due to the weak intermolecular interactions. Furthermore, the suitable mechanical and degradation properties of the PVP-PCL scaffold were still reserved after introducing collagen, and the introduction of collagen could further promote the thermostability of the PVP-PCL scaffold. Above all, the collagen/PVP-PCL scaffold showed no cytotoxicity, better cell proliferation, and improved viability of primary fibroblasts than the PVP-PCL scaffold. In conclusion, blending collagen with the PVP-PCL mixture in this study has potential for promoting the biocompatibility of PVP-PCL micro- and nanofiber scaffolds for tissue engineering.  相似文献

Lotus-leaf-like nanofibrous surfaces were prepared by electrospinning hydrophobic poly(vinylidene fluoride) (PVDF) from a mixed solvent of N,N-dimethylformamide (DMF) and acetone. PVDF fibrous mats with a bead-on-string morphology were generated because the nonpolar acetone decresed the viscosity of the PVDF solution and promoted the evaporation of the solution during electrospinning process. The morphology of the nanofibirous surface was examined by scanning electron microscopy. Micron-sized beads were introduced to the electrospun PVDF mats, resulting in enhanced hydrophobicity of the electrospun mats. The addition of a small amount (0.05 vol%) of acetic acid to the polymer solution effectively improved the bead-on-string morphology of the electrospun mats, and led to a higher water contact angle (WCA). The electrospun PVDF fibrous mat showed a maximum WCA of 148.5° due to the appropriate surface roughness.  相似文献

Electrospinning has been recognized as an efficient technique for the fabrication of neural tissue engineering scaffolds. Many approaches have been developed on material optimization, electrospinning techniques, and physical properties of scaffolds to produce a suitable scaffold for tissue engineering aspects. In this study, structural properties of scaffolds were promoted by controlling the speed of fiber collection without any post-processing. PLGA scaffolds, in two significantly different solution concentrations, were fabricated by the electrospinning process to produce scaffolds with the optimum nerve cell growth in a desired direction. The minimum, intermediate and maximum rate of fiber collection (0.4, 2.4, 4.8 m/s) formed Random, Aligned and Drown-aligned fibers, with various porosities and hydrophilicities. The scaffolds were characterized by fiber diameter, porosity, water contact angle and morphology. Human nerve cells were cultured on fiber substrates for seven days to study the effects of different scaffold structures on cell morphology and proliferation, simultaneously. The results of MTT assay, the morphology of cells and scaffold characterization recommend that the best structure to promote cell direction, morphology and proliferation is accessible in an optimized hydrophilicity and porosity of scaffolds, which was obtained at the collector linear speed of 2.4 m/s.  相似文献

Ionic conductivity and mechanical properties of a mixed polymer matrix consisting of poly(ethylene glycol) (PEG) and cyanoresin type M (CRM) with various lithium salts and plasticizer were examined. The CRM used was a copolymer of cyanoethyl pullulan and cyanoethyl poly(vinyl alcohol) with a molar ratio of 1:1, mixed plasticizer was ethylene carbonate (EC) and propylene carbonate (PC) at a volume ratio of 1:1. The conductive behavior of polymer electrolytes in the temperature range of 298∼338 K was investigated. The PEG/LiClO4 complexes exhibited the highest ionic conductivity of ∼10−5 S/cm at 25°C with the salt concentration of 1.5 M. In addition, the plasticized PEG/LiClO4 complexes exhibited improvement of ionic conductivity. However, their complexes showed decreased mechanical properties. The improvement of ionic conductivity and mechanical properties could be obtained from the polymer electrolytes by using CRM. The highest ionic conductivity of PEG/CRM/LiClO4/(EC-PC) was 5.33×10−4 S/cm at 25°C.  相似文献

In order to fabricate continuously twisted nanofiber yarns, double conjugate electrospinning had been developed using two pairs of oppositely charged electrospinning nozzles. The principle and process of this novel yarn spinning method were analyzed, and the effect of applied voltage, nozzle distance between positive and negative, solution flow rate and funnel rotating speed on the diameters, twist level and mechanical properties of resultant PAN nanofiber yarns were investigated in this paper. The results indicated that electrospun nanofibers aggregated stably and bundled continuously at the applied voltage of 18 kV, the nozzle distance of 17.5 cm between positive and negative, the overall flow rate of 3.2 ml/h and the flow ratio of 5/3 for positive and negative nozzles. The resultant nanofiber yarns had favorable orientation and uniform twist distribution, and the twist level of nanofiber yarns increased with the increase of the ratio of funnel rotating speed and winding speed. The diameters and mechanical properties of nanofiber yarns depended on their twist level. The diameters of prepared PAN nanofiber yarns ranged from 50 µm to 200 µm, and the strength and elongation of PAN nanofiber yarns at break were 55.70 MPa and 41.31%, respectively, at the twist angle of 41.8 °. This method can be also used to produce multifunctional composite yarns with two or more components.  相似文献

The aim of this study was to fabricate a new kind of hybrid fabric composites with the cross-linked electrospun poly(vinyl butyral) (PVB) composite nanofibres. The experiments were performed with the 10 wt.% PVB/ethanol solution for electrospinning where the modified silica nanoparticles (mSiO2), the oxidised single-walled carbon nanotubes (o-SWCNT) and the o-SWCNT/mSiO2 hybrid nanoparticles were added to the solution. The electrospun fibres were crosslinked with glutaraldehyde (GA) afterwards in order to reinforce the composite structure by bonding to the p-aramid fabrics. The chemical and thermo-mechanical properties of the hybrid fabric composites were evaluated. The greatest improvement in thermo-mechanical properties was achieved by the sample which contained the cross-linked PVB fibres with the o-SWCNT/mSiO2 hybrid nanoparticles.  相似文献

The complex nature of spinal cord injuries has provided much inspiration for the design of novel biomaterials and scaffolds which are capable of stimulating neural tissue repair strategies. Recently, conductive polymers have gained much attention for improving the nerve regeneration. In our previous study, a three-dimensional (3D) structure with reliable performance was achieved for electrospun scaffolds. The main purpose in the current study is formation of electrical excitable 3D scaffolds by appending polyaniline (PANI) to biocompatible polymers. In this paper, an attempt was made to develop conductive nanofibrous scaffolds, which can simultaneously present both electrical and topographical cues to cells. By using a proper 3D structure, two kinds of conductive scaffolds are compared with a non-conductive scaffold. The 3D nanofibrous core-sheath scaffolds, which are conductive, were prepared with nanorough sheath and aligned core. Two different sheath polymers, including poly(lactic-co-glycolic acid) PLGA and PLGA/PANI, with identical PCL/PANI cores were fabricated. Nanofibers of PCL and PLGA blends with PANI have fiber diameters of 234±60.8 nm and 770±166.6 nm, and conductivity of 3.17×10-5 S/cm and 4.29×10-5 S/cm, respectively. The cell proliferation evaluation of nerve cells on these two conductive scaffolds and previous non-conductive scaffolds (PLGA) indicate that the first conductive scaffold (PCL/ PANI-PLGA) could be more effective for nerve tissue regeneration. Locomotor scores of grafted animals by developed scaffolds showed significant performance of non-conductive 3D scaffolds. Moreover, the animal studies indicated the ability of two new types of conductive scaffolds as spinal cord regeneration candidates.  相似文献

The present work reports the preparation of Poly L-Lactide (PLLA) and Curcumin loaded Poly L-Lactide (CPLLA) nanofibers by electrospinning. A series of PLLA solution (12 wt %) and C-PLLA (12 wt % PLLA) solution containing Curcumin (0.5 wt % and 1 wt %)) were electrospun into nanofibers. SEM images showed the average diameter of PLLA and C-PLLA in the range of 50?C200 nm. The TEM images showed the dispersion of Curcumin on C-PLLA nanofibers. The XRD pattern indicated decreases of crystallinity with the increase in the amount of Curcumin. The characteristic peak of Curcumin was confirmed by FTIR. The TGA results showed the degradation of PLLA and C-PLLA close to 300 °C. The percentage porosity and the contact angle of PLLA were found to be 90.2 % and 115±3 ° with deionised water, respectively. The water uptake percentage was found to be 17.6 %. The percentage cumulative release of Curcumin at the end of 8th day for 0.5 and 1.0 wt % formulations was 81.4±1.3 and 86.7±1.7 % respectively. The in-vitro biological cytotoxicity studies were performed using C6 glioma cells and NIH 3T3 fibroblast by MTT assay and SEM analysis.  相似文献

In this study, we report the fabrication and evaluation of a hybrid multi-scale basalt fiber/epoxy composite laminate reinforced with layers of electrospun carbon nanotube/polyurethane (CNT/PU) nanofibers. Electrospun polyurethane mats containing 1, 3 and 5 wt% carbon nanotubes (CNTs) were interleaved between layers of basalt fibers laminated with epoxy through vacuum-assisted resin transfer molding (VARTM) process. The strength and stiffness of composites for each configuration were tested by tensile and flexural tests, and SEM analysis was conducted to observe the morphology of the composites. The results showed increase in tensile strength (4–13 %) and tensile modulus (6–20 %), and also increase in flexural strength (6.5–17.3 %) and stiffness of the hybrid composites with the increase of CNT content in PU nanofibers. The use of surfactant to disperse CNTs in the electrospun PU reinforcement resulted to the highest increase in both tensile and flexural properties, which is attributed to the homogeneous dispersion of CNTs in the PU nanofibers and the high surface area of the nanofibers themselves. Here, the use of multi-scale reinforcement fillers with good and homogeneous dispersion for epoxy-based laminates showed increased mechanical performance of the hybrid composite laminates.  相似文献

By combining the organic-inorganic hybridization, wet phase inversion, and electrospinning, novel electrospun polyurethane (PU) membranes with in-situ generated nano-TiO2 were prepared, which satisfied the requirements of an ideal wound dressing. The morphology of the PU-TiO2 mats and the cross sectional morphologies of the membranes were characterized by a scanning electron microscopy (SEM). The average diameter of the individual fibers obtained from the solutions was 341±12 nm. SEM micrographs with higher magnification further showed that the in-situ generated TiO2 particles were well-separated and dispersed homogeneously in the membranes. The average sizes of TiO2 particles were increased from 31 to 57 nm, with the increase of nano-TiO2 concentration. The water vapor transmission rates (WVTRs) of the membranes were in the range of 373.55–3121.86 g/m2·d and decreased gradually with the increase of nano-TiO2 concentration. The water absorption of various PU membranes was in the range of 210.90–397.98 % which was enough to prevent wound beds from exudate accumulation. Shake flask testing indicated that the PU membrane exhibited antibacterial efficiency against Pseudomonas aeruginosa (Ps. aeruginosa) and Staphylococcus aureus (S. aureus) due to in-situ generated of nano-TiO2. These electrospun nanofibrous membranes also had no toxic effect and showed good and immediate adherence to L929 cells.  相似文献

Fabrication of Ceftazidime (CTZ) loaded silk fibroin/gelatin (SF/GT) nanofibers (NFs) without the loss of structure and bioactivity of CTZ was demonstrated by electrospinning method. The structure, morphology and mechanical properties of the electrospun SF/GT nanofibrous mats were characterized using FT-IR, SEM and DSC. The drug release profile of different electrospun fibers was analyzed using spectrophotometric method, and also diffusion method was applied to assess the antibacterial effect of NFs. Cell viability was evaluated by MTT assay. The results show that the average diameter of drug loaded NFs at the optimum polymer to drug feeding ratio (10:1) was 276.55±35.8 nm, while increasing the feeding ratio to 1:1 increases the average diameter to 825.02±70.3 nm. FT-IR of drug loaded NFs was revealed that CTZ was successfully encapsulated into NFs while viability study approved cytocompatibility of SF/GT NFs. CTZ was released from NFs during 6 h, and formation of inhibition zone in diffusion test demonstrated the antibacterial effect of drug loaded NFs. Altogether, the CTZ loaded SF/GT NFs can improve the drug effectiveness particularly in the prevention of post-surgical adhesions and infections for wound dressing.  相似文献

In the present work, polyamide6-room temperature ionic liquid (PA6-RTIL) composite nanofibers and membranes were successfully prepared for the first time by an electrospinning technique. The surface morphology, component analysis, mechanical properties, thermal properties and conductivity of the PA6-RTIL composite membranes were investigated by field-emission scanning electron microscope (FE-SEM), fourier transform infrared spectrometer (FT-IR), tensile testing, thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and digit multimeter, respectively. The morphology, fiber diameter, mechanical strength of the obtained fibers can be controlled by changing experimental parameters for electrospinning, especially the content of RTIL in original electrospun mixture solution. The composite fibrous membranes showed ideal mechanical properties and significantly enhanced conductivity, which may be attributed to intrinsic high mechanical strength of PA6 and conductivity of RTIL.  相似文献

The formation of thrombosis has limited the applications of small diameter vascular in cardiovascular diseases. In order to improve the anticoagulant activities of scaffolds, this study combined fucoidan with CS/PVA and investigated the complete physicochemical and mechanical characterization of the scaffolds to evaluate the feasibility of Fucoidan/CS/PVA scaffolds used in vascular tissue engineering. The SEM graphs show a well defined and interconnected pore structure and the nanofiber diameters are ranging from 341 nm to 482 nm. After immersing in PBS for 5 days, the tensile strength of the crosslinked scaffolds was 722±38 kPa while the elongation at break was 35.5±1.6 %. Besides, added with fucoidan, the scafflolds showed lower rate of plate adhesion (14.75±2.10 %) and markedly prolonged the APTT and TT. Furthermore, owing to the great water uptake ability, sufficient porosity, enhanced drug release and low cytotoxicity, the Fucoidan/CS/PVA scaffolds might be used for vascular tissue engineering with good prospect.  相似文献

In order to realize flexible organic light emitting diode (OLED), it is critically important to protect moisture and oxygen penetration into the flexible device through the polymer substrates because of intrinsically poor stability of organic materials of OLED to moisture. Since complete protection of the moisture penetration is almost impossible, moisture absorbents are used inside OLED. In this study, metal oxide/polymer hybrid nanofiber with mechanical flexibility and efficient moisture absorption capability was fabricated by electrospinning. Hydrophilic poly(vinyl pyrrolidone) (PVP) or hydrophobic poly(vinylidenefluoride) (PVDF) and calcium oxide (CaO) were used as the fiber forming polymers and as the moisture absorbing particle, respectively. Polymers were first dissolved in appropriate solvents and CaO was then dispersed in the prepared polymer solutions. The hybrid solution was then electrospun, producing the hybrid nanofiber web looking like paper with thickness of about 100 µm. It was observed that all CaO particles were uniformly held by the nanofiber. It was also observed that only a small portion of the surface of CaO particles was covered by fiber since the fiber diameter was much smaller than the CaO particle size. This implied effective moisture absorption through the free surface of CaO. The hybrid nanofiber absorbed moisture very efficiently upto about 70 % of the nanofiber mass. Once moisture was absorbed by the nanofiber, it was never released even at 150 °C in vacuum for 2 hours. It is, therefore, expected that the nanofiber may be applied as the flexible thin moisture absorbent to improve stability of the flexible OLED.  相似文献
