Susceptibility to spinosad (Success®/Tracer®) of beet armyworm (Spodoptera exigua) from the southern USA and Southeast Asia was determined through exposure of second‐ and third‐instar larvae to dipped cotton leaves. LC50 estimates of susceptibility of second‐ and third‐instar larvae of field populations ranged from 0.279 to 6.14 and 0.589 to 14.0 mg spinosad litre−1, respectively. A Thailand population was 22‐ and 24‐fold less susceptible than the six other US field populations evaluated, and 85‐ and 58‐fold less susceptible than a reference laboratory population, respectively. From these results, we initiated experiments to test the hypothesis that the Thailand population was resistant to spinosad. F1 crosses between the resistant Thailand population and a susceptible reference strain yielded individuals that were 22‐fold less sensitive to spinosad than the susceptible parent. This same resistant strain exhibited significantly greater survivorship on plants treated with spinosad in the field. Lastly, selection of an Arizona population resulted in a significant reduction in susceptibility to spinosad, further substantiating the hypothesis of a genetic basis for resistance to spinosad. These findings indicate a vulnerability of this new insecticide to resistance development in beet armyworm and should serve as a warning against excessive use of it. © 2000 Society of Chemical Industry  相似文献

Two strategies for controlling first-instar larvae of gypsy moth (Lymantria dispar (L)), insecticidal bait and contact insecticide applied directly to the tree trunk, were evaluated in the laboratory. Spinosad was selected as a candidate natural-product insecticide that is active both by contact and ingestion. Incorporated into artificial diet-based bait, spinosad was toxic to neonate larvae with a minimal 10-s feeding period, with an LC50 value of 20 (15-26, 95% confidence interval) mg liter-1. It was significant that neonate larvae did not discriminate between spinosad-treated and control diet. Efficacy of diet-based bait in the laboratory, however, was significantly impacted by previous exposure to diet; fed larvae did not stop at the bait and did not incur mortality, as compared to unfed larvae. Oak bark was a suitable substrate from which neonate larvae could contact spinosad residues. Spinosad applied directly to oak bark resulted in significant mortality after 1- and 4-min crawling contact exposure times (LC50 = 24 [20-29, 95% CI] and 8.7 [6.9-11, 95% CI] mg liter-1, respectively) and contact activity persisted for 2 weeks. While contact activity was more potent on glass surfaces than on oak bark, the LC50 values differed only by factors of 2.4 and 3.6, for 1- and 4-min exposures respectively.  相似文献

Laboratory studies were conducted to determine the effect of the naturally derived compound spinosad on Ceratitis capitata Wied. (Diptera, Tephritidae). The organophosphate fenthion was used as a standard. Direct dose-dependent mortality and reduced fecundity were observed in oral treatment of adults with spinosad. The LC90 values 14 h and seven days after treatment were 19·50 and 0·49 mg litre−1 respectively. Fenthion was less active (the LC50 eight days after treatment was 1·17 mg litre−1) and did not affect the fecundity of the fly. Adults were also very susceptible to spinosad and fenthion via residual contact. For spinosad, 100% mortality was recorded 48 h after treatment for a dose of 10 mg litre−1. Spinosad was more effective than fenthion in suppressing larval development when neonate larvae were reared on treated diet supplemented with a range of concentrations from 0·02 to 0·83 mg kg−1 diet. Last-instar larvae were much less susceptible to spinosad or fenthion when exposed via dipping or when they pupated in treated medium and both products had similar performance. A lack of ovicidal activity was observed in direct egg-treatments with spinosad but significant reductions from 1 mg litre−1 onwards were recorded for fenthion.  相似文献

The insecticidal activities of four surfactants (Cide‐kick, Silwet L‐77, M‐Pede and APSA‐80), a dishwashing detergent (New Day), a mineral oil (Sunspray oil), a cotton seed oil and a vegetable oil, alone or in combination, were tested against nymphs of Bemisia argentifolii Bellows & Perring on collards and tomato. Silwet L‐77 was more effective (>95% mortality) than Cide‐Kick or APSA‐80 at rates from 0.25–1.00 g AI litre−1 but caused severe phytotoxicity to tender tomato leaves at all but the lowest rate. New Day dish detergent at 2.0 ml litre−1 caused mortality (95%) comparable to M‐Pede insecticide soap at 10‐fold greater concentration. A New Day ingredient, cocamide DEA, was considerably more active than the other ingredients or the commercial mixture. Additional surfactants added to Sunspray oil increased efficacy in some treatments, but not others. Toxic responses of 2nd‐ and 3rd‐ instar whiteflies to vegetable oil and cotton seed oil at 5.0 and 10.0 ml litre−1 plus 0.4 g AI litre−1 APSA‐80 ranged from 22.1 to 79.9% and 66.3–88.7% mortality, respectively. Whitefly mortality was greater on tomato than on collard in six of seven instances when differences between host plants were significant. Eleven sesquiterpene lactone derivatives of parthenin ( 1 ), obtained from wild feverfew, Parthenium hysterophorus, were prepared by chemical and photochemical transformations. The compounds tested were a pyrazoline adduct ( 2 ) of parthenin, its cyclopropyl ( 3 ) and propenyl ( 4 ) derivatives, anhydroparthenin ( 5 ), a dihydro‐deoxygenated product ( 6 ), a formate ( 7 ) and its corresponding alcohol ( 8 ) and acetate ( 9 ), a rearranged product ( 10 ), lactone ( 11 ) and hemiacetal ( 12 ). All these derivatives, along with parthenin, were tried for their antifeedant action against sixth‐instar larvae of Spodoptera litura, for insecticidal activity against the adults of store grain pest Callosobruchus maculatus, for phytotoxic activity against Cassia tora, and for nematicidal activity against the juvenile stage‐II (J2) of the root knot nematode Meloidogyne incognita. Antifeedent bioassay revealed that parthenin is moderately antifeedant. Among the derivatives, the saturated lactone ( 11 ) was found to be about 2.25 times more active than parthenin. The pyrazoline adduct ( 2 ) was found to be the most effective as an insecticide, with LC50 values after 24, 48 and 72 h of 96, 43 and 32 mg litre−1, respectively, which are comparable with neem extract. Compound 4 was found to be the most effective inhibitor of germination and seedling growth of C tora, with ID50 values for germination, plumule length and radicle length of 136, 326 and 172 compared with 364, 738 and 427 mg litre−1, respectively, for parthenin. Compound 10 was found to be the most effective in terms of nematicidal activity. Base‐line susceptibility for six‐day‐old larvae of the diamondback moth, Plutella xylostella, against Bacillus thuringiensis var kurstaki (Biobit®) was studied by a cabbage leaf disc dip bioassay technique. Diamondback moth from 13 locations in seven different states spread over a distance of about 3000 km longitudinally was used for these studies. Forty‐eight‐hour LC50 values varied from 1.0 to 10.97 mg AI litre−1. Further investigations on the development of resistance under laboratory conditions showed an increase in LC50 from 2.76 (for unselected F1 generation) to 5.28 mg AI litre−1 (for selected F9 generation), using a selection concentration of 6.4 mg AI litre−1. This suggested a possibility of the development of resistance under field conditions if there were to be extensive and indiscriminate use of B thuringiensis. These findings are discussed in relation to integrated pest management and the mechanisms of resistance in resistance management tactics. © 2000 Society of Chemical Industry  相似文献

The pine processionary moth Thaumetopoea wilkinsoni Tams is a serious pest on pine trees in southwestern Turkey. The efficacy of a naturally derived insecticide, spinosad, on fourth-fifth instar larvae of T. wilkinsoni was studied under laboratory conditions. The product exhibited strong larvicidal activity and at doses above 5 mg litre(-1) caused > 90% mortality in the fourth-fifth larval stages of the species after 72 h. At 72 h the LD50 and LD90 values were 3.26 and 5.69 mg litre(-1) respectively. The results showed that spinosad is highly effective on T. wilkinsoni larvae.  相似文献

BACKGROUND

Tuta absoluta (Lepidoptera: Gelechiidae) is difficult to control by means of foliar insecticides, partly because of the endophytic feeding behavior of its larvae. The biopesticide spinosad is applied as a foliar spray for control of T. absoluta and has systemic properties when applied as a soil drench to the growing medium of tomato plants. The aims of this study were to determine the: (i) instar-dependent tolerance of larvae to spinosad; (ii) efficacy of spinosad drench application for the control of larvae; (iii) residual period of systemic activity of spinosad in leaves and fruit after drenching; and (iv) effect of spinosad drenching on tomato plant growth parameters.

RESULTS

The estimated LC50 value (Lethal Concentration at which 50% of the larvae died) differed between instars. The LC50 for second-instar larvae (0.41 ppm) to spinosad was significantly lower than that for third- (0.64 ppm) and fourth-instar (0.63 ppm) larvae. The LC80 value (Concentration at which 80% of the larvae died) for fourth-instar larvae (2.48 ppm) was 2.6- and 1.7-fold higher than that for the second- and third-instar larvae, respectively. The spinosad concentration recorded in leaves at 25 days after treatment (DAT; 0.26 μg g−1) was significantly lower than that in leaves sampled at 3, 10 and 15 DAT. High larval mortalities were, however, recorded for the duration of the experiment, which lasted 25 days (equivalent to one T. absoluta generation).

CONCLUSION

Systemic spinosad effectively controlled T. absoluta larvae over a prolonged period. However, drenching this insecticide violates the recommendation of the Insecticide Resistance Action Committee to avoid treating consecutive insect generations with the same mode of action and can therefore result in the evolution of insecticide resistance. © 2023 The Authors. Tuta absoluta (Meyrick) resistance to insecticides has become a significant problem in many tomato production areas in South America. New insecticides are now available for the management of this pest (i.e. spinosad), however there is scarce information about their efficacy on field populations.With the aim of determining the susceptibility of T. absoluta to spinosad we evaluated the response of second instar larvae, from five field populations (Azapa 1, Azapa 2, Lluta, Colín and Valdivia) and a laboratory reference strain (S), to a diagnostic concentration of the insecticide. We also determined the activity of the detoxifying enzymes mixed-function oxidases (MFO), glutathione-S-transferases (GST) and esterases (EST) in the same larval stage. Larval mortality in field populations was significantly lower in Azapa 1 (50.0%), Azapa 2 (44.9%), Lluta (39.9%) and Colín (53.5%) when compared to the laboratory strain (91.7%). MFO activities in field populations were between 1.8 and 4.6 times higher than those observed in the S strain, while for EST, the ratio varied from 1.7 to 14.7. The lowest ratios were observed for the GST (0.5-2.7), however, significant differences were detected for the three enzyme systems. We conclude that the evaluated mechanisms would be involved in spinosad resistance of populations of T. absoluta, presenting an increased MFO activity in all populations.  相似文献

BACKGROUND: The immediate lethality caused by spinosad has been widely studied on Spodoptera exigua (Hübner). However, long‐term effects can also provide valuable information on insecticide toxic action. Here, the persistence of spinosad on Capsicum annuum L. foliage and the lethal and sublethal effects of greenhouse‐aged foliar residues of this insecticide on third instars of S. exigua are reported. RESULTS: Foliage was collected at 0, 3, 5, 10, 20, 30, 40 and 50 days after application, and spinosad residues were measured. Residues decreased over time according to first‐order kinetics. The average rate constant and half‐life of disappearance were 4.44 × 10?3 and 156 days and 5.80 × 10?3 and 120 days for 60 and 120 mg L?1 respectively. Larval mortality gradually decreased, corresponding to the residues, but was still appreciable (35 and 65% for 60 and 120 mg L?1 respectively) when the larvae were fed with foliage collected 50 days after treatment. Subsequently, pupal development was reduced and varied between 20 and 60% and between 21 and 41% for 60 and 120 mg L?1, respectively, in all ages of leaf residues that were bioassayed. At all time points, the consumption rate by the larvae was reduced between 62 and 84% for both concentrations that were bioassayed. CONCLUSION: It is concluded that, under the present greenhouse conditions, the degradation of spinosad was slower than that reported by other authors in the field, and, because of that, its residues could cause lethal and sublethal effects to S. exigua larvae. The effects of DPX-MP062 [methyl 7-chloro-2,3,4a,5-tetrahydro-2-[methoxycarbonyl(4-trifluoromethoxyphenyl)carbamoyl] indeno[1,2-e][1,3,4] oxadiazine-4a-carboxylate] a broad-spectrum insecticide with a novel mode of action, on the Egyptian cotton leafworm, Spodoptera littoralis, were studied in laboratory experiments. Egg hatch was affected by high concentrations (125 mg AI litre-1) of DPX-MP062. Larvae that hatched from treated eggs were significantly affected at concentrations of 12·5 mg AI litre-1 and greater. Larvae were fed castor bean leaves treated with DPX-MP062; 1st-instar larvae were the most susceptible development stage. Pupation and adult formation were determined in assays with 5th-instar larvae. There was strong suppression of adult formation; 65 and 91% at 0·5 and 0·75 mg AI litre-1, respectively. Highly affected larvae died before pupation; slightly affected ones reached pupation 2–4 days later, were smaller than larvae in the untreated control, and were sometimes unable to develop into normal adults. Comparatively high concentrations (50 and 100 mg AI litre-1) of the test compound were necessary to affect adults by ingestion, but no effects from contact application could be determined at a concentration of 100 mg AI litre-1. © 1998 Society of Chemical Industry  相似文献

为筛选防治金银花尺蠖Heterolocha jinyinhuaphaga Chu幼虫的高效、低残留杀虫剂,采用浸虫法和浸叶法分别测定了13种杀虫剂对金银花尺蠖幼虫的触杀毒力和胃毒毒力,筛选出高活性药剂,并对筛选的药剂进行田间防效试验及检测其在金银花中的农药残留量。毒力测定结果表明,甲维盐对金银花尺蠖2龄和4龄幼虫的毒力最高,触杀毒力LCct50分别为0.291、0.391 mg/L,胃毒毒力LCst50分别为0.081、0.275 mg/L;多杀菌素、氟铃脲、氯虫苯甲酰胺药剂对金银花尺蠖幼虫也表现出较高的致毒作用。田间药效试验表明,有效成分用量2.25 g (a.i.)/hm2的甲维盐、有效成分用量11.25 g (a.i.)/hm2的多杀菌素对金银花尺蠖幼虫均有极好的防治效果,药后7 d防治效果分别为96.39%和93.13%,表明甲维盐和多杀菌素是防治金银花尺蠖幼虫的特效药剂。残留检测分析结果表明,有效成分用量6、12 g (a.i.)/hm2的甲维盐喷雾处理3 d后,金银花中农药残留量分别为0.0219、0.0725 mg/kg,7 d后分别为0.0070、0.0168 mg/kg。  相似文献

The tobacco whitefly, Bemisia tabaci Gennadius (Homoptera: Aleyrodidae) which occurs in various parts of the world, has developed a high degree of resistance against several chemical classes of insecticide, including organophosphates, carbamates, pyrethroids, insect growth regulators and chlorinated hydrocarbons. The present studies were done in order to monitor the susceptibility of whitefly populations in southern Spain to insecticides commonly used there. Systemic bioassays using Spanish field populations of B tabaci collected in 1994, 1996 and 1998 indicated an increase, albeit a slow one, in resistance to imidacloprid over this period. Comparative studies of other neonicotinoids using the same bioassay revealed a high degree of cross‐resistance to acetamiprid and thiamethoxam. Leaf‐dip bioassays with adult females from these populations revealed a high level of resistance to cyfluthrin, endosulfan, monocrotophos, methamidophos, and pymetrozine, each at 200 mg litre−1. Buprofezin and pyriproxyfen were tested against second‐instar nymphs and eggs, respectively. Buprofezin also showed a lower efficacy against ESP‐98, a strain of B tabaci received from Almeria in 1998, but pyriproxyfen resistance was not obvious when tested against eggs of strain ESP‐98. Field trials in 1998 revealed good efficacy of imidacloprid in one farm in the Almeria region and two greenhouses in Murcia and Sevilla, but a loss of activity by imidacloprid in another farm in the Almeria region. BACKGROUND: Upon emergence from their pupal cells, bollworm, Helicoverpa zea (Boddie), adults actively seek and feed on plant exudates before they disperse and reproduce on suitable host plants. This nocturnal behavior of the bollworm may be exploited as a pest management strategy for suppression of the insect by using an attractant/stimulant mixed with an insecticide to induce feeding to cause adult mortality or reproductive reduction/inhibition. This study aimed to determine in the laboratory whether or not spinosad when mixed with sucrose solution as a feeding stimulant and ingested by bollworm could influence mortality and reproduction of the insect. RESULTS: Sublethal concentrations of spinosad fed to laboratory‐reared females confined with males significantly reduced percentage hatch of eggs at 0.1 mg L?1, and it was reduced to near zero at 2.5 mg L?1 when compared with females fed 2.5 M sucrose solutions only. The lethal concentration (LC99) for males captured from the field in sex‐pheromone‐baited traps was 73 mg L?1 for 24 h response. Proboscis extension response was not inhibited significantly even at 10 g L?1. In spite of a 137‐fold increase in lethal dose concentration, spinosad did not inhibit feeding. CONCLUSION: A detailed study of laboratory‐reared and field‐collected bollworm adults relative to mortality and reproduction after ingestion of spinosad indicates that spinosad would be useful in an attract‐and‐kill strategy to control the insect when mixed with a feeding attractant/stimulant. 对甜菜夜蛾的毒力测定结果表明,多杀菌素能显著降低其初孵幼虫的存活率、成虫的产卵量和成虫寿命。经6.25 μg/mL药液处理的卵,其初孵幼虫存活率仅为7.48%,而对照则为76.72%;取食12.50 μg/L药液的成虫,其产卵量和成虫寿命仅为取食蜂蜜水的1/6和1/2.5。随虫龄的增加,甜菜夜蛾幼虫对多杀菌素的敏感性显著降低,3龄、4龄幼虫的敏感性仅为2龄幼虫的1/9和1/16,且增效剂氧化胡椒基丁醚(PBO)与磷酸三苯酯(TPP)表皮点滴处理不表现增效作用。多杀菌素对甜菜夜蛾卵的孵化率、蛹重以及蛹的羽化率没有明显影响。亚致死剂量处理3龄幼虫后,其体内超氧化物歧化酶、过氧化氢酶和过氧化物酶等主要保护酶系的活性在处理后0~24 h与对照差异不明显,保护酶系与甜菜夜蛾对多杀菌素敏感性之间没有明显关系。  相似文献

BACKGROUND: The most common ways to control dengue vector Aedes aegypti (L.) are larval source reduction in domestic habitats and ground application of small quantities of aerosol insecticide (ultralow volume). Nevertheless, these actions have been shown repeatedly to be ineffective in controlling Ae. aegypti populations. RESULTS: The efficacy of a new smoke‐generating formulation containing pyriproxyfen and permethrin was evaluated in the laboratory. Smoke‐generating tablets containing each insecticide individually or combined were prepared, and the recovery of the insecticides from the smoke was determined. Recovery values of over 90% were obtained for pyriproxyfen, and around 50% for permethrin. The biological efficacy of pyriproxyfen released in the smoke was evaluated in the laboratory, on late third‐instar or early fourth‐instar Ae. aegypti larvae, using different concentrations of pyriproxyfen and exposure times. Adult emergence inhibition (EI) values of 100% were obtained at 30 min, and a dose‐dependent effect was observed at 5 min. The effect of pyriproxyfen released in the smoke was due to direct contact with the larvicide in the water rather than by inhalation of the fumes. The efficacy of permethrin released in the fumes was also evaluated as knockdown effect (KT50) on adults for a tablet containing permethrin alone or permethrin plus pyriproxyfen. There was no significant difference in KT50 values obtained for permethrin (KT50 = 19.9 min) and permethrin plus pyriproxyfen (KT50 = 19.4 min). CONCLUSION: The excellent laboratory performance of this new formulation on immature stages and adults indicates that a smoke‐generating tablet containing pyriproxyfen and permethrin could be a new tool for controlling mosquitoes. BACKGROUND: Azinphos‐methyl is the main insecticide used to control codling moth on apple and pears in Northern Patagonia. The aim of this study was to evaluate the toxicological and biochemical response of diapausing larvae of codling moth in orchards subjected to different insecticide selection pressure. RESULTS: Dose–mortality assays with azinphos‐methyl in diapausing larvae of Cydia pomonella L. showed significant differences between the LD95 from a population collected in one untreated orchard (2.52 µg moth?1) compared with that in a laboratory‐susceptible population (0.33 µg moth?1). Toxicity to azinphos‐methyl in field populations of diapausing larvae collected during 2003–2005 was evaluated by topical application of a discriminating dose (2.5 µg moth?1) that was obtained from larvae collected in the untreated orchard (field reference strain). Significantly lower mortality (37.71–84.21%) was observed in three out of eight field populations compared with that in the field reference strain. Most of the field populations showed higher esterase activity than that determined in both the laboratory susceptible and the field reference strains. Moreover, there was a high association between esterase activity and mortality (R2 = 0.64) among the field populations. On the other hand, a poor correlation was observed between glutathione S‐transferase activity and mortality (R2 = 0.33) among larvae collected from different orchards. CONCLUSIONS: All the field populations evaluated exhibited some degree of azinphos‐methyl tolerance in relation to the laboratory susceptible strain. Ephestia elutella larvae in diapause were exposed at 25°C to methyl bromide at 12 mg litre?1 for 3.5 or 7.5 h and then immediately exposed to a lower concentration. The minimum effective concentration (that at which Haber's rule, concentration × time = k, a constant for mortality, still applied) was about 3 mg litre?1 in tests with no previous exposure toa high concentration, but it was about 2.5 mg litre?1 for individuals surviving a 3.5 h exposure to 12 mg litre?1, and was about 1.6 mg litre?1 for those surviving a 7.5 h exposure to 12 mg litre?1. These exposures to 12 mg litre?1, respectively, killed 2–20% and 50–75% of larvae exposed, and hence the smaller the proportion of survivors of exposure to a high concentration, the lower the minimum effective concentration needed against them. Thus the low concentration persisting at the end of a practical fumigation should contribute significantly to the success of the treatment and be much more effective than any similar low concentration present soon after the introduction of gas.  相似文献

为明确转Bt基因玉米对草地贪夜蛾Spodoptera frugiperda的抗性,采用室内玉米离体组织生测法测定取食转Cry1Ab-ma基因抗虫玉米CM8101心叶、花丝和籽粒后草地贪夜蛾1龄和2龄幼虫存活数量、存活幼虫的体重、蛹重、化蛹和羽化等参数,分析转Cry1Ab-ma基因抗虫玉米CM8101三种组织对草地贪夜蛾1龄和2龄幼虫的抗性。结果显示,转Cry1Ab-ma基因玉米CM8101心叶可完全杀死草地贪夜蛾1龄幼虫,2龄幼虫有存活,存活率为10.0%,但不能化蛹与羽化;花丝可杀死大部分草地贪夜蛾1龄幼虫,存活率为3.3%,并阻断其生活史,但对2龄幼虫杀伤力弱,存活率为73.3%,存活幼虫化蛹率和羽化率分别为20.0%和13.3%;籽粒可完全杀死草地贪夜蛾1龄幼虫,2龄幼虫有存活,存活率为13.3%,有少量可化蛹,化蛹率为3.3%、有少量羽化,羽化率为3.3%。表明转Cry1Ab-ma基因玉米CM8101在短期内对草地贪夜蛾1龄和2龄幼虫有良好的控制作用。  相似文献

The residual effect of two biopesticides, abamectin and spinosad, on the predatory bugOrius laevigatus (Fieber) (Hemiptera: Anthocoridae) was tested using a laboratory and an extended laboratory method. Experiments were conducted in the spring and summer 2000. The LD50s on the 2nd and 5th nymphal instar and the adult stage, were lower for abamectin than for spinosad. The toxicity of each product on the 2nd instar and adults was significantly greater than on the 5th nymphal instar. Abamectin was much more persistent in spring than in summer. Spray deposits (at the recommended rate of 10 ppm a.i. for leafminer control) were toxic for 1 month in spring, whereas in summer they were no longer toxic after 2 weeks.
