Farmed grayling, Thymallus thymallus (L.), are susceptible to atypical Aeromonas salmonicida (aAS) infections. Interactions between bacteria and parasites were studied using grayling subjected to concomitant exposure to aAS bacteria and the digenean parasite Diplostomum spathaceum. Atypical AS was detected from fish by a combination of bacterial cultivation and polymerase chain reaction techniques. A detection level of 17 aAS cells per 100 mg intestine tissue sample was obtained. Concomitant bacterial exposure did not enhance the severity of grayling eye rupture and nuclear extrusion induced by D. spathaceum, but D. spathaceum invasion into grayling increased the proportion of fish carrying aAS in their heart tissue. However, the number of aAS cells detected in heart tissue was low. Atypical AS did not cause acute disease or mortality during 15 days post-exposure. There was a higher prevalence of aAS in grayling heart samples than in intestinal samples, indicating that the intestine is not favoured by aAS. We suggest that heart tissue would be a good organ from which to isolate aAS when tracing latent carrier fish. We conclude that penetrating diplostomids can enhance bacterial infections in fish and that diplostomids can cause serious eye ruptures in grayling.  相似文献

Abstract Artificially hatched and reared juvenile European grayling, Thymallus thymallus (L.), from three natural populations were released into an experimental stream. Two of the populations were from lakes and the third population was from a river. Post-stocking dispersal was studied by recapturing fish in traps and by electric fishing after each of the stocking exercises. On all three occasions the first fish to be recaptured were caught in a trap about 400 m downstream from the release site, within 5–12 min of release. Proportions of stocked grayling juveniles recaptured by electric fishing in the fast-flowing section of the experimental stream (the rapids) were 9.9%, 46.7% and 16.6% after the first, second and third stocking, respectively. No significant population-specific difference was found between fish caught in the downstream trap and fish recaptured in the rapids. The uppermost part of the rapids (stretch 1) and the catch in the upstream trap was dominated by fish of riverine origin. Post-stocking migration was influenced by sex but not by body size. Females showed a greater propensity to stay in the rapids compared with males. Implications of the results for stocking grayling in natural streams and regulated rivers are discussed.  相似文献

Arctic charr ( Salvelinus alpinus L.) were fed diets based on a commercial recipe supplemented with either linseed, soybean or marine oil prior to cohabitant challenge with Aeromonas salmonicida ssp. s almonicida . Mortality varied significantly between the three dietary groups. Highest mortality (48%) was observed in fish fed the marine oil and the lowest mortality (20%) was in the group fed soybean oil. Transmission electron microscopy (TEM) examination of the digestive tract of uninfected fish demonstrated substantial numbers of bacterial cells between microvilli. However, only a few bacteria were recovered that were associated with the microvilli of infected fish. Immunocytochemical staining/labelling investigations using TEM and an immunogold method were performed on mid-gut segments of fish fed the marine oil diet and showed augmentation of goblet cells and the presence of A. salmonicida ssp. salmonicida in the gastrointestinal tract of diseased fish after challenge with the pathogen. It is suggested that the gastrointestinal tract could be an infection route of A. salmonicida ssp. salmonicida. The greater prevalence of goblet cells supports the suggestion that sloughing off mucus is a protective response against bacterial infections. These results make an important contribution to our understanding of how nutrition can affect the disease resistance of fish.  相似文献

The metalloendopeptidase AsaP1 is one of the major extracellular virulence factors of A. salmonicida subsp. achromogenes, expressed as a 37‐kDa pre‐pro‐peptide and processed to a 19‐kDa active peptide. The aim of this study was to construct mutant strains secreting an AsaP1‐toxoid instead of AsaP1‐wt, to study virulence of these strains and to test the potency of the AsaP1‐toxoid bacterin and the recombinant AsaP1‐toxoids to induce protective immunity in Arctic char. Two A. salmonicida mutants were constructed that secrete either AsaP1E294A or AsaP1Y309F. The secreted AsaP1Y309F‐toxoid had weak caseinolytic activity and was processed to the 19‐kDa peptide, whereas the AsaP1E294A‐toxoid was found as a 37‐kDa pre‐pro‐peptide suggesting that AsaP1 is auto‐catalytically processed. The LD50 of the AsaP1Y309F‐toxoid mutant in Arctic char was significantly higher than that of the corresponding wt strain, and LD50 of the AsaP1E294A‐toxoid mutant was comparable with that of an AsaP1‐deficient strain. Bacterin based on AsaP1Y309F‐toxoid mutant provided significant protection, comparable with that induced by a commercial polyvalent furunculosis vaccine. Detoxification of AsaP1 is very hard, expensive and time consuming. Therefore, an AsaP1‐toxoid‐secreting mutant is more suitable than the respective wt strain for production of fish bacterins aimed to protect against atypical furunculosis.  相似文献

Abstract Arctic charr populations in southern latitudes are nonmigratory, with all life‐stages limited to freshwater lakes and in‐ or out‐flowing tributaries. Although many of these populations are reported to also spawn in lake littorals, little is known about the physical characteristics of putative spawning grounds. A total of 23 discrete spawning sites within three Irish lakes were located by fyke netting of spawning adults and snorkelling in littoral habitats. Spawning sites were found to be long, narrow strips running parallel to the shore at a maximum depth of 124 cm. Spawning sites were limited to areas of coarse mineral substrate with an adequate (c. 8 cm) depth of clean interstitial spaces. In individual lakes, combined areas of spawning sites made up 0.4–0.7% of available littoral. Egg densities varied considerably between sites (33–900·eggs m?2) and were significantly correlated with gradient and width of spawning sites. No evidence of redd digging was found. The shallow, localised and restricted nature of spawning grounds makes such populations vulnerable to anthropogenically induced postoviposition changes in surface water level, eutrophication processes such as increased lake sedimentation and elevated nutrient status.  相似文献

Muscle carotenoid concentrations were measured in immature and maturing 2+ Arctic charr, Salvelinus alpinus (L.), sampled from April until ovulation started in November. In immature fish, muscle carotenoid concentration increased steadily and had reached about 10 mg kg?1 by the end of the experiment. In mature fish of both sexes, pigment concentrations decreased during the autumn and muscle carotenoid concentrations were significantly lower than in the immature fish during the latter stages of the trial. No significant difference in muscle pigmentation was observed between sexes. Carotenoid concentrations were lowest in the cranial region of the fillet, but decreased in all parts of the fillet when the charr became mature. In females, the decrease seemed to be most marked in the caudal region. At the termination of the experiment, fillets accounted for about one-half of the carotenoid content in mature fish of both sexes. High concentrations of pigment were recorded in the skin and fins of mature males. In mature female charr, pigment was almost equally distributed among skin, fins and ovaries  相似文献

Abstract– The cannibalistic response of four domesticated Arctic charr stocks were compared experimentally using a radiographic technique. Charr from Arctic Spitsbergen (80°N) consistently exhibited a much higher frequency of cannibalistic response than fish from three charr strains from mainland Norway (70°N). The results indicate that the extent of cannibalistic feeding in Arctic charr is population specific and support the hypothesis that charr from Arctic populations have stronger cannibalistic tendencies than do fish from more temperate regions.  相似文献

Abstract Dominance hierarchy formation in groups of fish is thought to be a major reason for the development of size heterogeneity. The objective of the current experiment was to examine the influence of different feeding frequencies on growth and changes in size distribution in groups of Arctic charr Salvelinus alpinus (L.). The fish were fed equivalent excess rations continuously for 24 h a day, for 2 h twice a day (early morning and evening), for 2 h once a day in the morning or for 2 h once every other day also in the morning. Growth was measured by individual weighing of fish four times during the 130‐day experiment. Coefficient of variation in size within rearing groups was used to assess size dispersion and the possible development of hierarchies within the groups. Feeding frequency did not influence variation in size within groups, and there were no signs of hierarchy formation.  相似文献

Embryo somatic tissues, non‐somatic yolk‐sac materials, and whole, individual fingerlings (age 0+) of Arctic charr, Salvelinus alpinus (L.), as well as a commercial trout diet, were analysed for a wide spectrum of amino acids. Analytical material consisted of prefeeding swim‐up fry that were separated into discrete yolk sac and somatic embryo tissue samples. Amino acid concentrations in fry somatic tissue and whole fingerlings were generally very similar to each other, but were lower than those measured in yolk materials. Higher correlations were observed between the majority of specific amino acid concentrations in the trout diet when compared with fingerling data (r2 = 0.91) and fry somatic tissue data (r2 = 0.89), than when correlated with fry yolk sac material (r2 = 0.76). These results indicate that the essential amino acid profiles of fry somatic tissue and whole fingerlings are closer to that of a commercial feed than they are to the endogenous profiles found in the embryonic yolk sac material itself. The dietary ratios of individual essential amino acids were also compared with the total essential amino acid concentrations (A/E ratios) in whole fingerling tissues, and these ratios could be used to accurately estimate the apparent essential amino acid requirements of Arctic charr. The rationale for using carcass amino acid composition data to estimate the dietary essential amino acid requirements of Arctic charr is discussed.  相似文献

This study examined the long-term effects of a potential feeding deterrent, oxytetracycline (OTC), and a putative feeding stimulant, squid extract, on feed intake, growth and dry matter (DM) digestibility in Arctic charr, Salvelinus alpinus (L.). Fish were fed one of four diets for 9 weeks: 1. commercial feed formulation (basic); 2. BM (basic plus 20 g kg−1 OTC); 3. BMS (BM plus 10 g kg−1 squid extract); 4. BS (basic plus 10 g kg−1 squid extract). The feed intake of Arctic charr was not reduced by the addition of OTC to the basic feed, and addition of squid extract to the medicated feed did not enhance feed consumption. DM digestibility and growth tended to be reduced in groups of fish fed diets supplemented with the antibiotic (BM and BMS). This suggests that OTC may have interfered with nutrient digestion and absorption. OTC does not seem to be a feeding deterrent for Arctic charr, possibly indicating that the charr is a rather unselective feeder. Because the medicated feeds were readily accepted, there may be little to be gained by addition of feeding stimulants to these types of feeds for charr.  相似文献

Changes in lipid class composition of tissues of anadromous Arctic charr, Salvelinus alpinus, were studied in relation to seasonal patterns of lipid deposition and lipid mobilisation. Charr of a north Norwegian population were collected in late May when fish were migrating towards the sea, and in mid-July when the fish re-entered fresh water. Some maturing fish captured in mid-July were held in captivity until late September to assess lipid dynamics associated with maturation. The carcass (head, fins, skeleton and skin) accounted for approx. 50% of the total body lipid content when the fish re-entered fresh water from the sea. Triacylglycerols (TAG) comprised 75-80% of the carcass lipid in ascending charr, confirming that the carcass is a major lipid depot. The skeletal muscle also represented a major lipid store: muscle of ascending charr contained 35-40% of the body lipids, with TAG being dominant (approx. 75% of muscle lipid). The gut (gastrointestinal tract and visceral lipid) and liver of ascending charr each held only 4-5%; of the body lipid, TAG being the dominant lipid class in both of these depots (Gut TAG: 60-65%; Liver TAG: 50-55%). Body lipid decreased 30-40% during maturation: lipids were depleted from all depots, but mobilisation of from the carcass and muscle was quantitatively the most important. In mature females, the ovaries held almost 25% of the body lipid, with the proportion of TAG exceeding that of polar lipids (PL)(TAG: 62%; PL 12.4%). In the mature males, the testes accounted for less than 3% of the total lipids. TAG tended to dominate the testis lipids (TAG: 29.5%), but there were also quite high proportions of free fatty acids, cholesterol and polar lipids (FFA: 23.5%; C: 15%; PL 9.7%). Females lost approx. 80% of their body lipid during spawning and overwintering, the majority of this being TAG. Males also lost large amounts of TAG during the winter, but in contrast to the females lipid depletion amounted to only 50-55% of that present at the time of ascent into fresh water.  相似文献

Abstract – Owing to limited knowledge of the habitat use and diet of juvenile Arctic charr from the High Arctic, particularly young‐of‐the‐year (YOY), we assembled data obtained from samples taken in and around Lake Hazen, Nunavut, Canada, to assess juvenile habitat use and feeding. Juvenile charr demonstrated a preference for stream environments, particularly those fed by warm upstream ponds. Charr occupying both stream and nearshore lake habitats were found to feed similarly, with chironomids occurring most frequently in diets. Some older stream‐dwelling charr preyed on smaller, younger Arctic charr. Preferred stream occupancy is likely mediated by physical barriers created mainly by water velocity, and by distance from the lake, lake‐ice dynamics, low water depth, and turbidity. Water velocities resulted in stream habitat segregation by size, with YOY mainly found in low‐velocity pools and back eddies adjacent to stream banks, but not in water velocities >0.1 m·s?1. Greatest charr densities in streams were found in small, shallow, slow‐flowing side channels, which are highly susceptible to drought. Under predicted climate change scenarios, streams fed by small ponds will be susceptible to intermittent flow conditions, which could result in increased competition among juvenile charr for the remaining stream habitats. In addition, glacier‐fed streams are likely to experience increased flow conditions that will exacerbate physical barriers created by water velocity and further reduce the availability of preferred stream habitat.  相似文献

The influence of feeding high levels of polyunsaturated fatty acids (PUFA) on muscle fatty acid composition and indices of oxidative damage was examined in Arctic charr, Salvelinus alpinus (L.). All diets contained 100 g kg?1 lipid of dry weight. Two diets contained marine fish oils giving a PUFA level of 250 g kg?1 and 500 g kg?1 of lipid. The remaining two diets contained vegetable oils high in either 18:2n-6 or 18:3n-3, giving a PUFA level of more than 500 g kg?1 of dietary lipid. The charr were maintained at 8°C until their weight doubled, and were then transferred to 0.8°C for 30 days. Growth was similar in all groups. The fatty acid compositions of muscle were influenced by dietary PUFA but were less diverse than those of the diets. The overall pattern of fatty acid compositions indicated preferential desaturation and elongation of n-3 PUFA coupled with selective oxidation of 18:2n-6. Total n-3 PUFA content in TAG was always lowered compared with the diet, suggesting a specific mechanism for the removal of these fatty acids. Subjecting the fish to low temperature increased PUFA content in muscle of charr fed the 250 g kg?1 marine n-3 PUFA diet, but had no effect on the other treatments. For fish at 8°C, no significant differences were found between groups in terms of haematocrit, plasma alanine aminotransferase (ALAT), and plasma and muscle thiobarbituric acid reactive substances (TBARS), although there was a tendency towards increased levels of TBARS in the group receiving 500 g kg?1 marine n-3 PUFA of lipid. Subjecting the muscle to forced oxidative conditions resulted in increases in TBARS in all groups, particularly those fed 500 g kg?1 marine n-3 PUFA. Lowering the environmental temperature corresponded with a further increase in the plasma ALAT and muscle TBARS in this group. It is concluded that feeding diets containing high levels of long-chain n-3 PUFA may be detrimental to the fish's health and flesh quality, particularly at low environmental temperatures.  相似文献

To compare the rates of digestion and absorption of individual fatty acids, Arctic charr, Salvelinus alpinus (L.), were fed isoenergetic diets containing 40 g kg?1 coconut oil, and various combinations of 10 g kg?1 of polyunsaturated fatty acids (PUFA) (18:2n-6 or 18:3n-3) and monounsaturated fatty acids (MONO) (20:1n-9 or 22:1n-9) in the form of free fatty acids (FFA) or triacylglycerol (TAG). The average lipid digestibility for all diets measured by use of the chromic oxide method in the pyloric caeca area, midgut and hindgut were 72%, 83% and 88%, respectively, showing that lipid digestion and absorption occur mainly in the pyloric caeca area, but also extend throughout the intestinal tract. Analyses of digesta present in the intestinal segments suggest the predominance of non-specific lipolytic activity producing primarily FFA and glycerol from dietary TAG. Comparisons of the fatty acid composition of the lipid classes in the digesta showed that the utilization of dietary TAG was dependent both on the rate of release of the individual fatty acids from TAG, and their subsequent rate of absorption. When supplied as either FFA or TAG, the levels of PUFA (18:2n-6 or 18:3n-3) in the digesta were very low, indicating almost complete utilization. Both MONO used (20:1n-9 or 22:1n-9) were absorbed less efficiently than PUFA, but the rate of release from TAG seemed to be rate limiting only for 22:1n-9, which accumulated in the digesta. The rates of absorption of 20:1n-9 and 22:1n-9 when fed as FFA were the same. Comparisons of the levels of fatty acids in the dietary coconut oil TAG with those of the digesta lipids showed that 12:0 was a good substrate for intestinal lipase and was quickly absorbed. The lipolysis of 14:0 and 16:0 was intermediate while the longer-chain 18:0 appeared very resistant to digestion and was a major component of TAG, diacylglycerols and monoacylglycerols present in particularly the hindgut digesta. The absorption of 18:0 also appeared to be very low. The results suggest that PUFA are released very rapidly from dietary TAG by intestinal lipases in Arctic charr, and are specifically absorbed compared with long-chain saturated and monounsaturated fatty acids. The rate of lipolysis may be the rate-limiting step in the digestion of very long chain monounsaturated fatty acids such as 22:1n-9, while both the rate of lipolysis and absorption may be rate limiting for long-chain saturated fatty acids such as 18:0.  相似文献

The effects of three different temperatures on the growth and maturation of the offspring of cultured versus wild populations of juvenile arctic charr (Salvelinus alpinus L.) were investigated. The fish (start weight 17 g) were reared for 5 months at constant temperatures of 12, 14 and 16°C under a continuous light regime (LD24:0). Growth performance was significantly influenced by both temperature and source population. The offspring from the farmed fish displayed significantly higher mean weights at all temperatures compared with the offspring from the wild fish. The results indicate that the optimal temperature for growth (Topt) decreases with increasing fish size in the offspring of both cultivated and wild fish. Significant differences in length–weight relationship were found, with the offspring of wild fish displaying higher condition factor throughout the experiment. The results demonstrate that the offspring from multiple generation farmed population of arctic charr are better adapted to high temperatures compared with the offspring (F1 generation) from a wild population of arctic charr, and the former are also able to maintain growth at higher temperatures. There was a significant difference in maturation between the two populations, as the offspring from cultured fish displayed significantly lower level of maturation: 0%, 4% and 2% mature compared with 24%, 40% and 42% in the offspring from wild fish at temperatures of 12, 14 and 16°C respectively. The offspring from farmed strains of arctic charr thus appear to be the most suitable population for aquaculture.  相似文献

1. This paper highlights the recent extinctions of two native Arctic charr populations in the West of Ireland, one population from the Lough Conn catchment, and a second from the Lough Corrib catchment, respectively. 2. The distribution of charr populations in the two catchments is reviewed. The current distribution information is drawn from recent fish stock surveys (Lough Corrib in 1996, and Lough Conn in 1997 and 1998). Previous records of charr in the literature and unpublished data are described to place the current distribution of charr within these catchments into a historical perspective. 3. The importance of the charr population in Lough Mask (Lough Corrib catchment) is emphasized, as it is one of the last extant examples of charr from a large Irish alkaline lake. 4.
