Factors influencing boar sperm cryosurvival   总被引:1,自引:0,他引:1  
Optimal sperm cryopreservation is a prerequisite for the sustainable commercial application of frozen-thawed boar semen for AI. Three experiments were performed to identify factors influencing variability of postthaw sperm survival among 464 boar ejaculates. Sperm-rich ejaculate fractions were cryopre-served using a standard freezing-thawing procedure for 0.5-mL plastic straws and computer-controlled freezing equipment. Postthaw sperm motility (assessed with a computer-assisted semen analysis system) and viability (simultaneously probed by flow cytometry analysis after triple-fluorescent stain), evaluated 30 and 150 min postthaw, were used to estimate the success of cryopreservation. In the first experiment, 168 unselected ejaculates (1 ejaculate/boar), from boars of 6 breeds with a wide age range (8 to 48 mo), were cryopreserved over a 12-mo period to evaluate the predictive value of boar (breed and age), semen collection, transport variables (season of ejaculate collection, interval between collections, and ejaculate temperature exposure), initial semen traits, and sperm quality before freezing on sperm survival after freezing-thawing. In Exp. 2, 4 ejaculates from each of 29 boars, preselected according to their initial semen traits and sperm quality before freezing, were collected and frozen over a 6-mo period to evaluate the influence of interboar and intraboar ejaculate variability in the survival of sperm after cryopreservation. In Exp. 3, 12 ejaculates preselected as for Exp. 2, from each of 15 boars with known good sperm cryosurvival, were collected and frozen over a 12-mo period to estimate the sustainability of sperm cryosurvival between ejaculates over time. Boar and semen collection and transport variables were not predictive of sperm cryosurvival among ejaculates. Initial semen traits and sperm quality variables observed before freezing explained 23.2 and 10.9%, respectively, of the variation in postthaw sperm motility and viability. However, more that 70% of total variance observed in postthaw sperm quality variables among ejaculates was explained by boar. This indicates that boar is the most important (P < 0.001) factor explaining the variability among ejaculates in sperm cryosurvival, with most (14 of the 15 boars in Exp. 3) showing consistent (P > 0.05) sperm cryosurvival over time.  相似文献

Sperm quality is often evaluated through computer-assisted semen analysis (CASA) and is an indicator of boar fertility. The aim of this research was to study the relationship between CASA motility parameters and fertility results in pigs. Insemination records and semen parameters from a total of 45,532 ejaculates collected over a 3-yr period were used. The statistical model for analysis of fertility data from these inseminations included factors related to sow productivity. The boar- and semen-related variance (direct boar effect) were corrected for the effects of individual boar, genetic line of the boar, age of the boar, days between ejaculations, number of sperm cells in an ejaculate, number of sperm cells in an insemination dose, and AI station. The remaining variance was analyzed if semen motility parameters had a significant effect. This analysis revealed significant (P < 0.05) effects of progressive motility, velocity curvilinear, and beat cross frequency on farrowing rate (FR). Total motility, velocity average path, velocity straight line, and amplitude of lateral head displacement affected (P < 0.05) total number of piglets born (TNB). Boar- and semen-related parameters explained 5.3% of the variation in FR and 5.9% of the variation in TNB. Motility parameters, measured by CASA, explained 9% of the boar- and semen-related variation in FR and 10% of the boar- and semen-related variation in TNB. Individual boar and genetic line of the boar affected (P < 0.0001) the variation in FR and TNB. No differences (P > 0.05) were observed between effects of AI stations on fertility outcome, underscoring the objectivity of the CASA system used. Motility parameters can be measured with CASA to assess sperm motility in an objective manner. On the basis of the motility pattern, CASA enables one to discriminate between the fertilizing capacity of ejaculates, although this depends on the genetic line of the boar used in AI stations.  相似文献

The aim of the study was to investigate the effect of the antioxidant astaxanthin on boar semen. Twenty ejaculates from 10 boars (two ejaculates/boar) were extended and split in three groups: semen control (SC), solvent control (C; semen with dimethyl sulfoxide, the diluent of astaxanthin) and semen with astaxanthin (A) in concentration 0.5 μmol/L. Sperm quality parameters (motility and kinetics, morphology, viability, functional integrity of sperm plasma membrane by Hypo‐Osmotic Swelling Test [HOST] and DNA integrity) were assessed at 0, 24 and 48 hr of storage at 17°C (experiment I), before (0 hr) and after (1 hr) of sperm thermal resistance assay at 37°C (experiment II) and finally before (0 hr) and after (1 hr) sperm in vitro incubation (38.5°C, 5% CO2, maximum humidity [experiment III]). In experiment I, group A performed overall better than group SC and as a tendency better than group C regarding viability. Total motility, rapid spermatozoa and HOST remained constant across time in group A, whereas they decreased in the remaining groups. In experiment II, regarding motility and viability, group A displayed better results across time than the other two groups. In experiment III, viability and total motility decreased in groups SC and C, while in group A, these parameters were not significantly different between the examination time points. In conclusion, astaxanthin has a beneficial and protective effect on boar semen quality under the investigated conditions.  相似文献

The sperm chromatin structure assay is a method for assessment of sperm DNA fragmentation, a parameter reported to be negatively related to field fertility in several mammal species. This method calculates a DNA fragmentation index (DFI) whose high values indicate abnormal chromatin structure. In this study, running from March 2010 until June 2017, the aim was to assess sperm DFI in stored liquid extended semen from two different pig breeds, Norwegian Landrace (NL; n = 693) and Norwegian Duroc (ND; n = 655), and to evaluate the influence on total number of piglets born (TNB). There was a significantly higher median DFI (p < 0.0001) in ejaculates from the 478 ND boars compared to the 452 NL boars. Data from 19,496 NL litters and 3,877 ND litters of the same boars were retrieved. For either breed, sow herd (p < 0.0001), parity (p < 0.05) and DFI (p < 0.05) showed significant effects on TNB. The DFI was negatively correlated to TNB in both breeds. The boars with the 5% lowest TNB had a least square means DFI of 3.05% and 2.24% in NL and ND, respectively, compared to 1.67% and 1.23% for the boars with the 5% highest TNB (p < 0.01). The DFI and the motility of the same semen samples were negatively correlated (p < 0.0001), and the high and low TNB groups showed significant differences in motility. However, this difference could not be used for practical prediction of TNB group (92.1% vs. 89.7%; p = 0.0038 and 92.3% vs. 89.5%; p = 0.018; NL and ND, respectively). In conclusion, our results indicate that sperm DNA integrity in semen with good motility and morphology may be an additional prediction parameter for fertility in pigs.  相似文献

The reactive oxygen species (ROS) which are produced during storage of boar semen are causing oxidative stress and leads to poor fertility. Also, tropical and sub-tropical weather condition adversely impacts the physicomorphological quality and fertility of boar sperm. The aim of this study was to examine the effects of feeding linseed oil to boar on its seminal attributes, sperm kinetics, biomarkers of antioxidant, fatty acid profile of seminal plasma (SP) and sperm and in vivo fertility. Six Hampshire crossbreed boars were fed with 90 ml linseed oil (LIN) whereas six Hampshire crossbreed boars were fed 90 ml canola oil (CON) for 16 weeks. Sperm quality was evaluated (60 ejaculates for each group; a total of 120 ejaculates) for motility, livability, abnormal morphology, acrosomal membrane integrity, hypo-osmotic swelling test (HOST) and sperm kinetic parameters by computer assisted semen analysis (CASA) at 0 h and at 72 h of storage at 17°C. Biomarkers of antioxidant (glutathione peroxidase; GPx, catalase; CAT, total antioxidant capacity; TAC) and malondialdehyde (MDA) were measured in SP and serum. Gas chromatography–mass spectrometry (GC–MS) was used for the estimation of fatty acid composition of SP and sperm. Boars fed with linseed oil had higher semen volume (p < .01) and more total sperm numbers (p < .01). Feeding linseed oil to boar enhanced seminal attributes (p < .05) at 0 h as well as at 72 h of storage. Linseed oil feeding (p < .01) improved biomarkers of antioxidants and significantly (p < .01) lowered the lipid peroxidation in serum and SP. Linseed oil feeding (p < .05) increased the proportion of alpha linolenic (ALA), arachidonic and docosahexaenoic (DHA) fatty acids in SP. The ratio of n-6 to n-3 fatty acids in sperm increased significantly (p < .01) in treatment group. Farrowing rate was significantly (p < .05) higher in treatment group. In conclusion, feeding linseed oil to boar improved the in vivo fertility, enhanced antioxidant capacity and increased the DHA content of SP and sperm.  相似文献

This study compares three fluorescence methods for the evaluation of boar sperm viability and examines the interrelationships between fluorescence viability and motility of liquid semen, stored for 7 days, and fertility. One artificial insemination dose from each of 102 boars was stored for 7 days at 20°C. Plasma membrane integrity was evaluated in three different ways. First, a combination of two fluorophore probes, calcein acetylmethyl ester and ethidium homodimer-1 (EH), was used to stain samples for fluorescence microscopic analysis by making semen smears. Secondly, semen samples were stained with EH with and without the addition of a detergent (ATP-releasing reagent), and fluorescence intensities were measured with a fluorometer. Thirdly, fluorometric evaluation was carried out in the same pattern as with EH, by means of a more permeable, less specific DNA fluorochrome (more interference with RNA), Hoechst 33258. Computer-assisted motility assessment gave the values for total, rapid and progressive motility and path velocity in semen stored for 7 days. Fertility of the boars was determined by the nonreturn (NR%) rate within 60 days of first inseminations and litter size of multiparous farrowings. The results showed that all three fluorescence methods were strongly intercorrelated. All plasma membrane integrity parameters correlated significantly with motility parameters and with fertility parameters (NR% and litter size of multiparous farrowings), but motility parameters did not correlate with NR%. It seems that fluorometric measurement could prove useful for plasma membrane integrity studies in liquid boar semen. Use of the objective and fast fluorometer-based viability assay is thus suitable for several applications in sperm studies.  相似文献

The economic profitability of a boar station largely depends on semen quantity and quality traits. However, genetic analysis of semen traits has not yet been done in the boar population in China. In this study, we aimed to estimate genetic parameters for semen traits and the influence of seasons on these traits by using data of Duroc, Landrace and Yorkshire boars in South China. The following four semen traits were analysed: semen volume (ml; VOL), sperm concentration (106/ml; DEN), sperm motility (MOT) and percentage of abnormal sperm (ABN). Genetic parameters and season effects were estimated simultaneously for each breed by using a multiple‐trait (4 × 4) repeatability animal model. The four traits had a moderate heritability with average estimates of 0.23, 0.28, 0.26 and 0.17 across the three breeds, respectively. The estimates of genetic correlations among four traits differed in the three breeds. In particular, in Yorkshire, the four traits were nearly genetically independent. The season of collecting semen had a significant impact on these four semen traits except ABN in Duroc (Bonferroni adjusted p < 0.05/6). The moderate heritabilities indicate the possibility of effective selection of boars for semen traits. Different genetic correlations for the three breeds suggest that the selection strategy for the four traits should be investigated separately for each breed. Some necessary actions should be taken to reduce the influence of seasons on semen traits.  相似文献

The aim of this study was to investigate whether there is a relationship of young boar testes size with semen traits and with productive traits. The dimensions (length, width and volume) of each testis and semen traits (semen volume, percentage of sperm with progressive motility, sperm concentration, total number of sperm in semen, percentage of sperm with normal acrosome, percentage of sperm with major and minor morphological defects, osmotic resistance test value and activity of aspartate aminotransferase in seminal plasma) were determined on 120 young boars aged 6 months. At 180 day of age, the boars backfat thickness and leanness (by ultrasonic apparatus) and body weight were also measured. The average daily gain was determined in the period from 70 to 180 days of age of the boars. The results showed that the sperm concentration, total number of sperm in semen and percentage of progressive motile sperm were a significantly positively correlated with width and volume of the left (p ≤ 0.01) and right testis (p ≤ 0.05) and with total volume of both testes (p ≤ 0.01). But the highest values of correlation coefficients were found between the width of the left testis and sperm concentration, total number of sperm in semen and percentage of progressive motile sperm. A correlations of dimensions (length and width) and volume of testes with other semen traits were very low and statistically non‐significant. The volume of testes (left and right testis and total testes) was significantly positively correlated with body weight at 180 days of age and daily gain (p ≤ 0.01), but lower correlation coefficient was between left testis and daily gain (p ≤ 0.05), whereas correlations were low and non‐significant with leanness and backfat thickness.  相似文献

An investigation involving seven boars, active in artificial insemination, and 1,350 multiparous sows was conducted at a private farm and aimed at examining the relationship between sperm quality traits and boar fertility in terms of farrowing rate and litter size. This experiment was done for 6 months. The semen samples were evaluated for subjective sperm motility and concentration. Ejaculates with at least 1 × 108 sperm/mL and 70% sperm progressive motility were extended with a commercial medium to 30 × 106 sperm/mL and used for artificial insemination (AI). AI dose was 100 mL semen containing 3 × 109 spermatozoa. Aliquots of diluted semen were assessed for live morphologically normal spermatozoa (LMNS, eosin-nigrosin stain exclusion assay) and sperm chromatin instability (SCI, acridine orange assay). Farrowing rates according to different boar sperm varied (p < 0.001) from 59.3 to 88.92%. The mean values of LMNS (47.2~76.5%) and SCI (0.16~4.67%) differed significantly among boars. LMNS (r = 0.79, p < 0.05) and SCI (r = -0.90, p < 0.02) accounted for 62.2 and 81.7% of the variability in farrowing rates, respectively. After the combination of sperm traits, the relationship between percentage of LMNS with stable chromatin structure and farrowing rate was significant (r = 0.86, p < 0.05). The number of live piglets per parturition was not significantly correlated with sperm quality attributes. In conclusion, boar fertility after AI with freshly diluted semen can be predicted based on the evaluation of sperm morphology and chromatin integrity.  相似文献

Motility is the most widely used indicator of sperm quality. Computer‐Assisted Semen Analysis (CASA) allows the objective evaluation of sperm motility parameters. CASA technology is a common tool to predict semen doses in farm animal reproduction. The kinds of video cameras used until now for image acquisition have presented limited frame rates (FR), which have a negative influence on the quality of the obtained data. The aim of the present work was to define the optimal frame rate for a correct evaluation of boar sperm motility and its subpopulation structure. Eighteen ejaculates from nine mature boars of the Pietrain breed were used. Using the ISAS®v1 CASA‐Mot system, with a video camera working up to 200 Hz, six FRs (25, 50, 75, 100, 150 and 200 fps) were compared. ISAS®D4C20 counting chambers, warmed to 37°C, were used. FR affected all the kinematic parameters, with curvilinear velocity (VCL) and BCF the most sensitive ones. All the parameters showed differences among animals. Non‐linear correlation showed the asymptotic level for VCL at 212 fps, being the highest FR for all the parameters. For future studies based just on progressive motility, almost 100 fps FR for 0.5 s must be used, while when kinematics must be considered, almost 212 fps for one‐second should be analysed. Three principal components were obtained (velocity, progressivity and oscillation), being similar at 50 and 200 fps. Cells were grouped in four subpopulations but with different kinematic and cellular distribution at both FRs.  相似文献

为研究不同品种、采精月份、采精月龄和采精间隔等因素对加系公猪精液品质的影响,以及品种、初次采精周龄对精液质量稳定性的影响,本研究以江西某种公猪站79头加系大白猪、长白猪、杜洛克猪种公猪为试验群体,收集2018年12月至2020年12月3 921条精液采集与精液质量数据,通过混合线性模型与方差分析探究各因素对精液量、精液密度、精子活力、总精子数及其稳定性的影响。结果显示,从不同品种对精液质量的影响来看,长白猪精液量和总精子数均高于大白猪、杜洛克猪,但杜洛克猪精液密度高于长白猪、大白猪,杜洛克猪精子活力最低;从不同月份来看,1~3月采精精液密度最高,4~6月采精精子活力最高,10~12月采精精液量和总精子数最高,精液量呈现秋冬多、春夏少的季节变化规律。公猪不同月龄采精,精液质量指标也存在差异,月龄越小精液量越低,但精液密度偏高,精子活力相对较好,在19~24月龄黄金期总精子数最高。不同采精间隔对精液质量有较大影响,采精间隔越长,精液量、精液密度、精子活力和总精子数相对较好,采精间隔为5 d时综合性能最佳,但过长的采精间隔导致精子活力降低。品种影响总精子数稳定性,长白猪、大白猪总精子数稳定性显著优于杜洛克猪(P<0.05)。本研究结果表明,品种、采精月份、采精月龄和采精间隔均会影响公猪精液质量,关注这些因素有助于公猪站制定更完善的生产计划,提高公猪利用率。  相似文献

【目的】探究品种、公猪出生胎次、公猪同窝仔猪数、公猪乳头数、采精季节、采精月龄和采精间隔对猪精液品质的影响,以及不同品种对精液质量稳定性的影响。【方法】选取909头杜洛克、长白、大白种公猪为试验群体,收集2021年4月至2022年4月27 408条精液测定记录,采用混合线性分析模型和方差分析探究品种、公猪出生胎次、公猪同窝仔猪数、公猪乳头数、采精季节、采精月龄和采精间隔对精液体积、精液密度、精子活力、直线前进运动精子比例、精子畸形率、总精子数及各精液性状稳定性的影响。【结果】从不同品种对精液品质的影响来看,长白猪精液体积和总精子数均显著高于大白猪、杜洛克猪(P<0.05),杜洛克猪精液密度显著高于大白猪和长白猪(P<0.05),杜洛克猪、大白猪精子活力均显著高于长白猪(P<0.05),大白猪直线前进运动精子比例显著高于长白猪和杜洛克猪(P<0.05),长白猪和大白猪精子畸形率均显著低于杜洛克猪(P<0.05);从不同公猪出生胎次对精液品质的影响来看,1~3胎出生的公猪具有较高的精液品质;从不同采精季节对精液品质的影响来看,精液密度、精子活力和总精子数秋、冬季显著高于春、夏季(P<0.05);从不同采精月龄对精液品质的影响来看,16~25月龄公猪具有较高的精液品质;从不同采精间隔对精液品质的影响来看,4~5 d为最佳采精间隔,采精间隔过长会导致精子畸形率上升;从不同公猪总乳头数来看,乳头数13~16个时,各精液性状品质都处于中等水平,有利于公猪生产应用。3个品种各精液性状间稳定性趋势不一,杜洛克猪和大白猪精液体积和精子活力的稳定性均显著高于长白猪(P<0.05),杜洛克猪精液密度、精子畸形率和总精子数的稳定性均显著高于大白猪和长白猪(P<0.05),大白猪直线前进运动精子比例的稳定性显著高于长白猪和杜洛克猪(P<0.05)。各品种中,长白猪稳定性较差;各精液性状中,精子活力稳定性最好。【结论】品种、公猪出生胎次、采精季节、采精月龄和采精间隔均会影响公猪精液品质,可根据不同品种公猪制定更完善的选择方案,提高精液质量,加速公猪遗传改良。  相似文献

This study aimed to investigate the effects of bovine serum albumin (BSA) on boar sperm quality during liquid storage at 17°C. Boar semen samples were collected and diluted with Modena containing different concentrations (0, 1, 2, 3, 4, 5 and 6 g/l) of BSA, and sperm motility, plasma membrane integrity, acrosome integrity, total antioxidative capacity (T‐AOC) activity and malondialdehyde (MDA) content were measured and analysed. The results showed that Modena supplemented with 3, 4 and 5 g/l BSA could improve boar sperm motility, effective survival time and plasma membrane integrity (p < 0.05), decrease MDA content (p < 0.05), while no statistical difference was observed for sperm acrosome integrity and T‐AOC activity among these three groups (p > 0.05). The semen sample diluted with Modena containing 4 g/l BSA could achieve optimum effect, and sperm survival time was 7.5 days. After 7 days preservation, sperm motility, plasma membrane integrity and acrosome integrity were 54%, 49% and 78%, respectively. T‐AOC activity and MDA content were 1.03 U/ml and 17.5 nmol/ml, respectively. In conclusion, Modena supplemented with BSA reduced the oxidative stress and improved the sperm quality of boar semen during liquid storage at 17°C, and 4 g/l BSA was the optimum concentration. Further studies are required to obtain more concrete results on the determination of antioxidant capacities of BSA in liquid preserved boar semen.  相似文献

The semen evaluation techniques used in most commercial artificial insemination centers, which includes sperm motility and morphology measurements, provides a very conservative estimate of the relative fertility of individual boars. As well, differences in relative boar fertility are masked by the widespread use of pooled semen for commercial artificial insemination (AI) in many countries. Furthermore, the relatively high sperm numbers used in commercial AI practice usually compensate for reduced fertility, as can be seen in some boars when lower numbers of sperm are used for AI. The increased efficiency of pork production should involve enhanced use of boars with strong reproductive efficiency and the highest genetic merit for important production traits. Given that the current measures of semen quality are not always indicative of fertility and reproductive performance in boars, accurate and predictive genetic and protein markers are still needed. Recently, significant efforts have been made to identify reliable markers that allow for the identification and exclusion of sires with reduced reproductive efficiency. This paper reviews the current status of proteomic and genomic markers of fertility in boars in relation to other livestock species.  相似文献

The present study determined the effect of different types of sugars (lactose, fructose, glucose and sorbitol) used in egg yolk-based extender on the post-thawed boar semen quality. Twenty-two ejaculates from 6 fertility-proven Yorkshire boars were cryopreserved by liquid nitrogen vapor method. Sperm motility, viability, acrosome integrity and intact functional plasma membrane were determined at 0, 2 and 4 hr after thawing. It was found that the lactose-based extender resulted in a higher percentage of post-thawed sperm motility, viability, intact acrosome and functional plasma membrane than sorbitol-based extender (P<0.05) and fructose-based extender yielded a higher post-thawed sperm motility and viability than sorbitol-based extender (P<0.05). It could be concluded that sorbitol was not an effective sugar for the cryopreservation in boar semen.  相似文献

The purpose of this study was to estimate the genetic parameters of semen traits of Landrace boars in South China and to analyze the influence of the age and season of semen collection on semen traits, so as to provide theoretical basis for formulating reasonable boar breeding program. The Asreml-R was used to analyze the 107 221 semen data of 1 605 Landrace boars from 2 AI stations in Southern China, the single-trait repeatability animal model was used to estimate the variance components, heritability and repeatability of each semen traits, two-trait repeatability animal model was used to estimate genetic correlation and phenotypic correlation of semen volume, semen density, sperm motility and percentage of abnormal sperm. The general linear model of R language program was used to analyze the influence of age and season of semen collection on semen traits. The results showed that the semen volume and percentage of abnormal sperm had medium heritability (0.23 and 0.38), and the variation coefficient of percentage of abnormal sperm was 85.42%, while the other traits had low heritability (0.07-0.19). Semen volume-semen density and sperm motility-percentage of abnormal sperm had extremely significantly negative genetic correlation(-0.77 and -0.90, respectively). Semen density-sperm motility showed extremely significantly positive genetic correlation (0.50). The influence of the age of semen collection on semen traits was significant(P<0.05). After boars reached sexual maturity, semen volume showed a significant increase, semen density and percentage of abnormal sperm showed a general decrease, and total sperm number and functional sperm number were significantly higher in the 13-18-month of age group than those in the other groups (P<0.05). The semen density was the highest in spring, and the total sperm number and functional sperm number were significantly higher in autumn and winter than those in spring and summer (P<0.05). In conclusion, the semen volume and percentage of abnormal sperm of Landrace boars can be selected as candidate traits. In the boar production management, boar whose semen production after 36 months of age decline, will be considered to update. Boar semen quality was significantly higher in autumn and winter than those in spring and summer. It is suggested that cooling work should be done in advance in Southern China in summer.  相似文献

The aim of the study was to determine the relation between the semen quality, frequency of sperm defects, sperm dimensions and shape, and the ejaculate volume of Large White and Landrace boars. A total of 648 ejaculates collected from 31 Large White and 30 Landrace boars were divided into three groups according to the criterion of the ejaculate volume. In this study Landrace boars produced ejaculates with higher volume, sperm concentration, and total numbers of spermatozoa than Large White boars. Landrace boars also showed a lower frequency of sperm with morphological abnormalities (P < 0.05). Landrace boars sperm had larger heads, which were by 0.15 μm longer, and by a larger perimeter and area (P < 0.05). Landrace boar spermatozoa also had a longer flagellum and were generally larger and by 2.07 μm longer than Large White boar sperm (P < 0.05). Significant differences were also found in the shape of sperm of the two breeds (P < 0.05). Landrace boars sperm had more elongated heads, and the ratio of head size to flagellum length was lower than in Large White boars sperm (P < 0.05). Semen volume, sperm concentration, percentage progressive motility, percentage of abnormal spermatozoa and total number of sperms from 215 830 ejaculates were analysed. The ejaculates were collected between 2000 and 2005 and originated from 3675 boars of different breeds and crossbred combinations from 23 AI centres of the Czech Republic. Genetic parameters were estimated using multiple- and single-trait animal models. Factors included in the models were: breed or crossbred combination, year–month, age class of the boar, interval between subsequent semen collections, joint effect of AI centre and year, permanent environment and additive genetic effect of the boar. The estimated heritabilities for semen volume and sperm concentration were approximately 0.20, whereas the estimates were somewhat lower for motility, percentage of abnormal sperm and total number of sperms. High negative genetic correlations were estimated between semen volume and sperm concentration (−0.68 for dam breeds, −0.69 for sire breeds) and between motility and percentage of abnormal sperm (−0.93 for dam breeds and −0.59 for sire breeds). The correlations between both semen volume or sperm concentration and motility or percentage of abnormal sperm were mostly small and negligible. Repeatabilities of 0.43–0.46, 0.37–0.38, 0.29–0.35, 0.42–0.50 and 0.29–0.30 were estimated for semen volume, sperm concentration, motility, percentage of abnormal sperm and the total number of sperms, respectively. On the basis of the estimated genetic parameters presented here, effective selection on sperm characteristics, especially for volume and concentration, should be possible using an animal model.  相似文献

旨在估计华南地区长白种公猪精液性状遗传参数以及分析公猪采精月龄和季节对精液性状的影响,为合理制定种公猪育种方案提供理论依据。本研究采用Asreml-R对华南地区两个公猪站1 605头长白公猪107 221条精液数据进行统计分析,利用单性状重复力动物模型估计公猪精液各性状的方差组分、遗传力和重复力,利用两性状重复力动物模型对精液体积、精液密度、精子活力和精子畸形率等性状进行遗传相关和表型相关估计;利用R语言程序中的一般线性模型分析采精月龄和季节对精液性状的影响。结果表明,精液体积和精子畸形率属于中等遗传力(0.23和0.38),其中精子畸形率的变异系数较大,为85.42%,其余性状都为低遗传力(0.07~0.19);精液体积与精液密度以及精子活力与精子畸形率为极显著遗传负相关(-0.77和-0.90);精液密度与精子活力呈极显著的遗传正相关(0.50)。采精月龄对精液性状影响显著(P<0.05),在公猪达到性成熟后,精液体积呈显著上升趋势,精液密度和精子畸形率总体呈下降趋势,精子总数和有效精子数在13~18月龄组显著高于其他组(P<0.05)。在春季精液密度最高,精子总数和有效精子数在秋、冬季显著优于春、夏季(P<0.05)。综上,长白公猪精液体积和精子畸形率具有较大的选育潜力,可作为候选性状进行选择。在公猪生产管理方面,公猪36月龄后精液产量下降,考虑更新淘汰。公猪精液品质在秋、冬季显著优于春、夏季,在中国南方夏季应提前做好降温工作。  相似文献

Contents Determination of the osmotic resistance of boar spermatozoa and the relations to sperm freezing and long storage The osmotic behaviour of fresh boar spermatozoa (482 ejaculates from 34 A.I. boars) was examined in a standard medium but with different osmotic pressures ranging from 150 mosm/kg till 500 mosm/kg. The percentage of spermatozoa with normal acrosomal ridge (NAR) was determined after incubation of 0.2 ml fresh semen in 3 ml of the osmotic test solutions at 39° C for 15 and 120 min. In the non-isotonic media % NAR decreased and could be quantified depending to incubation time and to the different osmolalities; NAR decreased strongest at 150 or 500 mosm/kg and after 120 min incubation time. Differences in % NAR were large between ejaculates and between boars but low within boars and statistically significant correlations had been found to % NAR and motility in froren/thawed semen samples or stored samples. Still higher correlations were found by calculation of an osmotic resistance test-value (ORT-value) which is defined as follows: ORT = 1/2 % NAR in isotonic medium/(after 15 min) +% NAR in 150 or 500 mosm/kg/(after 15 or 120 min) ORT-values can be calculated for hypotonic (150 mosm/kg) as well as for hypertonic (500 mosm/kg) test solutions and for 15 and 120 minutes incubation time. The hypotonic ORT-value for 120 min. is the most accurate; the values varied from 34 till 83 between ejaculates and from 44 till 76 between boars with a low variability within boars. Grouping of boars by their ORT-values in “good”, “moderate” and “poor” corresponded to a similar grouping of these boars by their semen quality (% NAR and motility) in preserved semen samples. Also a grouping of single ejaculates by their ORT-values corresponded to a similar qualitative classification of these ejaculates. ORT seems to become a good and practicable assessment test for the selection of boars suitable dor deep freezing or long storage of their spermatozoa. On the other hand preservation techniques too can be examined by ORT. But more investigations with this test must be done especially its relation to the fertilization capacity of semen samples with different ORT-values.  相似文献
