109 Pyrus accessions including 92 local Chinese accessions of P. bretschneideri were identified genetically using nine microsatellite loci developed from apple and pear. The nine SSR loci revealed 129 alleles in 109 pear accessions and 114 alleles in 92 Chinese white pears. Among the 92 local Chinese accessions of P. bretschneideri, 70 could be differentiated successfully except for 10 sets of synonymous or mutants. For the 92 accessions, the number of putative alleles per locus ranged from seven to 18, with an average of 12.67; the average values of observed heterozygosity and Shannon’s Information index were 0.60 and 1.85, respectively. A phenogram based on the SSR (simple sequence repeat) genotypes was obtained. The 109 accessions clustered into 11 groups based on geographical origin. The European pears and the Asian pears did not form independent two groups, but three P. communis cultivars grouped together independently. The Japanese P. pyrifolia cultivars mingled together with the Chinese P. bretschneideri cultivars, but four P. ussuriensis cultivars except for one (Jianbali) grouped together independently. The results indicated that the relationship of P. bretschneideri cultivars and P. pyrifolia cultivars was much closer than others.  相似文献

Twenty-five Pyrus communis L. cultivars including eight traditional Portuguese pears, and four commercial Pyrus pyrifolia (Burm.) Nak. (Japanese pear or `nashi') cultivars were analysed by RAPD and AFLP techniques focusing on their molecular discrimination and the assessment of their genetic relatedness. Twenty-five primers generated 324 RAPD markers, among which 271 (84%) were polymorphic. The AFLP technique, using seven primer combinations, revealed a similar level of molecular polymorphisms (87%), representing 418 polymorphic bands among a total of 478 scored in autoradiographs. The high reproducibility of RAPD and AFLP techniques was confirmed comparing DNA samples from different extractions and different digestions of DNA from the same plant. Three genetic similarity matrices and respective dendrograms were elaborated on using RAPD, AFLP or joint RAPD and AFLP data. Both molecular marker techniques proved their reliability to assess genetic relationships among pear cultivars. P. pyrifolia cultivars exhibit a closer genetic relatedness, clustering apart from P. communis cultivars. Within P. communis, `William's', as well as `Doyenne du Comice', cluster close to their hybrids. Most of the Portuguese cultivars tend to cluster together, indicating to constitute a relatively independent genetic pool, which can be of interest in pear breeding programs.  相似文献

The genetic relationships were assessed for the first time in Turkish durum wheat cultivars using AFLP markers. In the analysis, 18 AFLP primer combinations resulted in a total of 189 polymorphic loci. All of the selective primers used are Eco and Mse primers with three nucleotide extentions on the 3 ends. The number of polymorphic markers per primer combination ranged from 4 to 24. The relationships, among nine winter and six spring type durum wheat cultivars, obtained with various algorithms are in accordance with the known pedigree information of the cultivars. Based on `Nei72' genetic distance analysis, the most distant two cultivars are `Berkmen-469' (winter type) and `Diyarbakìr-88' (spring type), and the closest two are `Selçuklu-97' and `Sofu', with the values of 0.793 and 0.115, respectively. Persimmon culture has become an alternative for fruit growers in Italy, Israel and Spain. However, there is some confusion concerning cultivar identification in non-Asian countries were the species was introduced. Authenticating the identity of germplasm resources of persimmon would be a great value for breeding. Due to the floral biology of the persimmon species the expected genetic variability is high. RAPD markers were chosen for this study of persimmon germplasm resources in Spain. Nineteen markers were obtained from 8 primers. The average of 2.4 markers/primer was higher than that obtained in other woody fruit species. The availability of data referenced from isozymes and pomological traits revealed complete agreement among groups identified by RAPDs, isozymes and pomological traits. Results suggest that RAPD technology is adequate for fingerprinting persimmon and the data is consistent with persimmon's hypothetical origins, adaptation history and previous classification by isozymes or pomological traits. Many synonyms have been identified, mostly in cultivars from local surveys, suggesting that a few new genotypes arose in Spain since the species was introduced to Europe. By means of RAPD markers, a better identification of the European persimmon germplasm is available.  相似文献

The genetic variation and relationships among 31 accessions of Phaseolus vulgaris L., and two representatives of Vigna unguiculata L., were evaluated by AFLP analysis. A total of 263 DNA fragments across all materials were scored using nine primer combinations, averaging 32 per primer. More than 95% of the amplification products showed polymorphism, indicating high variation at the DNA level among these accessions. Pair-wise genetic similarity (Jaccard's coefficient) ranged from 0.553 to 0.840, with a mean of 0.765. Twenty-three accessions (70%) clustered into three groups. A majority of the commercial cultivars (91%) clustered within a single group, whereas the landraces were distributed along all the variation. An apparent correlation with phaseolin types was detected. Results of this study suggest that Brazilian landraces truly represent the overall genetic variability of Phaseolus vulgaris, confirming the multiple origins of these materials, and their potential as a source of variation for breeding programs.  相似文献

Increasing the knowledge of the molecular diversity of a crop is essential for extending its genetic base, identifying cultivars and selecting parental varieties for breeding programs. In this sense, Cucurbita maxima Duch. is poorly characterised. Nineteen accessions of this species and 8 related Cucurbita accessions were included in a genetic diversity analysis. For this purpose, Random Amplified Polymorphic DNA markers (RAPDs), which analyse neutral variability, and Sequence-Based Amplified Polymorphism (SBAPs), which preferentially amplify coding regions of the genome, were used. While the UPGMA cluster and the principal coordinates analysis obtained using RAPDs did not group the different accessions according either to fruit morphological criteria or to passport data (origin and agro-climatic conditions), the principal coordinates analysis obtained using SBAPs grouped the different pumpkin accessions fundamentally according to the type of use (human consumption, animal fodder or ornamental). This passport trait is reported to be associated with agronomic breeding characters of interest. The usefulness of both types of markers for discriminating accessions of breeding interest is discussed.  相似文献

Genetic variation and phylogenetic relationships among 50 wild and cultivated accessions of 19 Indian Citrus genotypes were examined through comparison of Directed Amplification of Minisatellite DNA (DAMD) markers and morphological characters. DAMD-PCR analysis with four primers resulted in amplification of a total of 45 bands, of which 35 (78 %) were polymorphic. Morphometric evaluation using 76 morphological characters showed high level of variability ranging from 0.18 to 1.00 (avg 0.39), whereas the Jaccard’s coefficient values of genetic similarity calculated from DAMD data ranged from 0.41 to 1.00 (avg 0.68), indicating moderate genetic divergence among the accessions studied. UPGMA dendrograms generated separately from morphometric and DAMD data segregated all the accessions of Citrus into four main clusters, each containing a true basic species and their probable hybrids. The grouping of individual accessions/genotypes under respective species or cultivars in DAMD dendrogram was based purely on their genetic relationships rather than geographical origin. There was no absolute congruence between the data and dendrograms generated from morphometric and DAMD analyses. The study demonstrates the resolving power of DAMD markers for discrimination of individual genotypes of Citrus under its respective species, hybrid or cultivar groups and inferring their genetic and phylogenetic relationships as well. This is the first report on application of DAMD markers in Citrus.  相似文献

ABSTRACT

Assessing the genetic variation and relationships present in crop germplasm is a pre-requisite for parental selection and breeding. The objective of this study was to determine the genetic relationships present among selected heat and drought tolerant wheat genotypes using simple sequence repeat (SSR) markers, agronomic traits and grain quality parameters to select desirable parents for breeding. Twenty-four agronomically selected wheat genotypes sourced from the International Maize and Wheat Improvement Centre (CIMMYT)’s heat and drought tolerance nursery and four local check varieties were genotyped using 12 selected polymorphic SSR markers. The test genotypes were phenotyped using yield and yield-component traits, and grain protein content (GPC) under non-stressed (NS) and drought-stressed (DS) conditions. Expected heterozygosity mean value of 0.58 indicated moderate genetic diversity for breeding. The studied wheat genotypes were delineated into six genetic groups using cluster analysis. Significant genotypic differences were observed for agronomic traits and GPC under NS and DS conditions. Genetically unrelated breeding parents including LM02, LM13, LM23, LM41, LM44, LM71, LM73 and LM75 were selected for population development and breeding for enhanced grain yield and protein content under heat and drought-stressed environments.  相似文献

Choix, a plant in the tribe Maydeae of the grass family, has been cultivated in Asia for several thousand years. It is a potential gene resource for improvement of other cereal crops because of its nutritional value and tolerance to stress. Genetic variation and relationships among 21 Choix lachryma-jobi L. accessions were characterized by random amplified polymorphic DNA (RAPD) markers. A total of 205 DNA fragments across all materials were amplified with 31 random primers, averaging 6.61 per primer. Among amplified fragments, 115 showed polymorphism averaging 3.71 per primer. Of amplified markers, 56.1% were polymorphic, indicating considerable variation at the DNA level among these accessions. Some fragments were accession-specific. Pair-wise genetic similarity (GS) among 21 accessions ranged from 0.809 to 0.301. The 21 accessions clustered into two major groups. Three exotic Choix accessions clustered together. Three other Choix accessions, collected from Guangxi, China, clustered into a cohesive subgroup. Four wild types of Choix clustered into the same subgroup. These results indicated that the classification by RAPD data reflected the differences in geographic origins and evolution in Choix.  相似文献

Alternative berry crops belonging with the Rosaceae family currently have a great importance in the US and Canadian agribusiness, in this sense, the Mexican serviceberry (Malacomeles denticulata) is a fruit shrub mainly distributed in Mexico that has potential uses like horticultural crop; however, little information about its variability has been reported. That is why this research aims to determine the variability of M. denticulata by SSR markers. The apple SSR markers had a good transferability to Malacomeles (74 %); all transferable SSR are polymorphic and sixteen of them had high values of polymorphic information content, Nei’s expected heterozygosity (He), and Shannon’s information index (I), even the apple SSR loci were not previously proved in this species. Besides it was found high variability in the germplasm of central Mexico mainly within populations (72 %), where three different genetic pool were detected suggesting that a breeding program could be based in individual selection. That is why it is necessary select outstanding individual into population before to make hybridizations among these selected parents.  相似文献

For adding the hulless barley resources of Shangri-la region to the global barley resource library, a basic work was done by us to assess their genetic diversity of this region. The genetic diversity of 60 hulless barley samples collected from three counties in Shangri-la region of Yunnan Province, were studied using SSR (simple sequence repeats) and AFLP (amplified fragment length polymorphism) markers. A total of 70 alleles were detected for 19 pairs of SSR primers, and 525 band containing 464 polymorphic bands were revealed for 5 pairs of AFLP primers. The value of polymorphism information content (PIC) ranged from 0.03 to 0.86 for SSR primers. The total numbers of alleles were 51, 55, 43 in three populations and the polymorphic bands were 188, 205 and 141. The genetic distances and genetic identity among the three populations showed their close relationship. The gene diversity among populations relative to the total population diversity (Gst) was 0.13 for SSR markers and 0.02 for AFLP markers and indicated that just 13 and 2% variations were among populations, respectively. The UPGMA cluster analysis revealed that all of the samples grouped randomly rather than clustered into distinct groups corresponding to their populations, row types and spring/fall types. We concluded that there was high genetic diversity in the population of Shangri-la region and the formation of diversity was related to complex environment and inhabitants’ traditional practices.  相似文献

The genetic diversity of 10 commercial cultivars of common beans, developed in Northern Argentina was analyzed based on RAPD markers. Sixteen primers were assayed and among them only 4 showed polymorphisms. A similarity matrix was generated by applying three different association coefficients, Simple Matching, Jaccard and Dice. By the UPGMA method dendrograms were generated and also the principal coordinate analysis was performed. The similarity values found were higher than 40% suggesting that genetic diversity is low. Both cluster analysis and principal coordinates analysis associated commercial cultivars either to the Andean or the Mesoamerican gene pool.  相似文献

The essential oil composition and genetic variability of six commercial cultivars of thyme (Thymus vulgaris L.), a Mediterranean medicinal and aromatic plant, were analyzed by GC-MS and randomly amplified polymorphic DNA (RAPD), respectively. All evaluated cultivars belong to the thymol chemotype, with differences in the concentrations of thymol, gamma-terpinene, p-cymene, and other minor components. The comparison of the oil components concentration by multivariate analysis allowed separation of the cultivars into two groups. All of the cultivars exhibited characteristic RAPD patterns that allowed their identification. On the basis of the RAPD patterns, the cultivars could be divided into two clusters, which coincides with results obtained by oil GS-MS analysis, with a correlation coefficient of -0.779.  相似文献

RAPD markers were used to analyse the genetic variability among a cole crop cultivar collection from France and to identify possible duplicate accessions for facilitating the germplasm multiplication. We surveyed 24 cauliflower, 24 cabbage and 48 kale populations using 62, 100 and 89 RAPD markers respectively on 40 seed bulk samples per accession. Genetic distances were calculated on the basis of these markers. Using the phylogenetic package PAUP we compared the genetic clusters obtained for the RAPD markers with the groups based on morphologic and agronomic data. For cauliflowers and cabbages a drastic selection by growers led to an extensive differentiation of morphologic and precocity traits related to geographic origin, which matches the grouping based on RAPD markers. Furthermore, RAPDs detected in some cases misclassification of accessions in the collection. Kales formed an homogeneous group probably due to extensive genetic exchanges leading to a continuous diversity. For the other crops, different horticultural types were separated in several subgroups by the RAPD markers suggesting different genetic origins. RAPD markers provide a rapid and informative approach to analyze the genetic variability in a collection. It can be applied to autogamous (spring cauliflower) or allogamous (winter cauliflower) crops, even in cases where the intra-population diversity is extensive (cabbages).  相似文献

The common apricot (Prunus armeniaca L.) is adaptable to the diverse geographical areas throughout northern China where the original center for common apricot is worldwide. The diversity of 94 apricot samples, including 66 typical Chinese local cultivars, four kernel-using apricots, thirteen cultivars of Xinjiang (belong to Central Asian groups), ten exotic cultivars and one black apricot (Prunus dasycarpa Ehrh.) accession, were investigated using 21 simple sequence repeat markers. Totally, 662 genotypes were identified in the set of tested 93 common apricot cultivars, with an average of 31.52 genotypes per locus. The allele diversity per locus, with an average allele number of 15.14, and the heterozygosity rates, with an average expected heterozygosity value for the sample set of 0.792, were found to be higher than those in previous studies, implying that Chinese apricot germplasms presented a high level of genetic diversity. The clustering analysis and principal coordinate analysis outlined the genetic relationships of apricot cultivars tested from different geographical distributions. The results supported the traditional classification of apricot germplasm based on the ecological types, and showed that the richest diversity existed in the northern China and northwestern China local cultivars among common apricot germplasm resources tested. This study will facilitate the understanding of the evolution relationships and the conservation strategies of the genetic diversity in apricot in China.  相似文献
