Plants and animals activate defenses after perceiving pathogen-associated molecular patterns (PAMPs) such as bacterial flagellin. In Arabidopsis, perception of flagellin increases resistance to the bacterium Pseudomonas syringae, although the molecular mechanisms involved remain elusive. Here, we show that a flagellin-derived peptide induces a plant microRNA (miRNA) that negatively regulates messenger RNAs for the F-box auxin receptors TIR1, AFB2, and AFB3. Repression of auxin signaling restricts P. syringae growth, implicating auxin in disease susceptibility and miRNA-mediated suppression of auxin signaling in resistance. 相似文献

The trichomes of rice leaves are formed by the differentiation and development of epidermal cells. Plant trichomes play an important role in stress resistance and protection against direct ultraviolet irradiation. However, the development of rice trichomes remains poorly understood. In this study, we conducted ethylmethane sulfonate (EMS)-mediated mutagenesis on the wild-type (WT) indica rice ‘Xida 1B’. Phenotypic analysis led to the screening of a mutant that is defective in trichome development, designated lhl1 (less hairy leaf 1). We performed map-based cloning and localized the mutated gene to the 70-kb interval between the molecular markers V-9 and V-10 on chromosome 2. The locus LOC_Os02g25230 was identified as the candidate gene by sequencing. We constructed RNA interference (LHL1-RNAi) and overexpression lines (LHL1-OE) to verity the candidate gene. The leaves of the LHL1-RNAi lines showed the same trichome developmental defects as the lhl1 mutant, whereas the trichome morphology on the leaf surface of the LHL1-OE lines was similar to that of the WT, although the number of trichomes was significantly higher. Quantitative real-time PCR (RT-qPCR) analysis revealed that the expression levels of auxin-related genes and positive regulators of trichome development in the lhl1 mutant were down-regulated compared with the WT. Hormone response analysis revealed that LHL1 expression was affected by auxin. The results indicate that the influence of LHL1 on trichome development in rice leaves may be associated with an auxin pathway. 相似文献

Upland cotton (Gossypium hirsutum L.) is the most important natural textile fiber crop worldwide. Plant height (PH) is a significant component of plant architecture, strongly influencing crop cultivation patterns, overall yield, and economic coefficient. However, cotton genes regulating plant height have not been fully identified. Previously, an HD-Zip gene (GhHB12) was isolated and characterized in cotton, which regulates the abiotic and biotic stress responses and the growth and development processes. In this study, we showed that GhHB12 was induced by auxin. Moreover, overexpression of GhHB12 induces the expression of HY5, ATH1, and HAT4, represses the spatial-temporal distribution, polar transport, and signaling of auxin, alters the expression of genes involved in cell wall expansion, and restrains the plant height in cotton. These results suggest a role of GhHB12 in regulating cotton plant height, which could be achieved by affecting the auxin signaling and cell wall expansion. 相似文献

Plant disease resistance (R) genes trigger innate immune responses upon pathogen attack. RAR1 is an early convergence point in a signaling pathway engaged by multiple R genes. Here, we show that RAR1 interacts with plant orthologs of the yeast protein SGT1, an essential regulator in the cell cycle. Silencing the barley gene Sgt1 reveals its role in R gene-triggered, Rar1-dependent disease resistance. SGT1 associates with SKP1 and CUL1, subunits of the SCF (Skp1-Cullin-F-box) ubiquitin ligase complex. Furthermore, the RAR1-SGT1 complex also interacts with two COP9 signalosome components. The interactions among RAR1, SGT1, SCF, and signalosome subunits indicate a link between disease resistance and ubiquitination. 相似文献

The immunoglobulin MOPC167, which binds the transition state analog p-nitrophenylphosphorylcholine with high affinity, catalyzed the hydrolysis of the corresponding carbonate 1. MOPC167 catalysis displayed saturation kinetics with catalytic constant (kcat) = 0.4 min-1 and Michaelis constant (Km) = 208 microM, showed substrate specificity, and was inhibited by p-nitrophenylphosphorylcholine. The rate of the reaction was first order in hydroxide ion concentration between pH 6.0 and 8.0. The lower limit for the rate of acceleration of hydrolysis by the antibody above the uncatalyzed reaction was 770. This study begins to define the rules for the generation of catalytic antibodies. 相似文献

The optic tectum of zebrafish is involved in behavioral responses that require the detection of small objects. The superficial layers of the tectal neuropil receive input from retinal axons, while its deeper layers convey the processed information to premotor areas. Imaging with a genetically encoded calcium indicator revealed that the deep layers, as well as the dendrites of single tectal neurons, are preferentially activated by small visual stimuli. This spatial filtering relies on GABAergic interneurons (using the neurotransmitter γ-aminobutyric acid) that are located in the superficial input layer and respond only to large visual stimuli. Photo-ablation of these cells with KillerRed, or silencing of their synaptic transmission, eliminates the size tuning of deeper layers and impairs the capture of prey. 相似文献

An ancient date seed (Phoenix dactylifera L.) excavated from Masada and radiocarbon-dated to the first century Common Era was germinated. Climatic conditions at the Dead Sea may have contributed to the longevity of this oldest, directly dated, viable seed. Growth and development of the seedling over 26 months was compatible with normal date seedlings propagated from modern seeds. Preliminary molecular characterization demonstrated high levels of genetic variation in comparison to modern, elite date cultivars currently growing in Israel. As a representative of an extinct date palm population, this seedling can provide insights into the historic date culture of the Dead Sea region. It also has importance for seed banking and conservation and may be of relevance to modern date palm cultivation. 相似文献

The synergistic response of cells to the stimulation of multiple receptors has been ascribed to receptor cross talk; however, the specific molecules that mediate the resultant signal amplification have not been defined. Here a 24-kilodalton single transmembrane protein, designated calcyon, we functionally characterize that interacts with the D1 dopamine receptor. Calcyon localizes to dendritic spines of D1 receptor-expressing pyramidal cells in prefrontal cortex. These studies delineate a mechanism of Gq- and Gs-coupled heterotrimeric GTP-binding protein-coupled receptor cross talk by which D1 receptors can shift effector coupling to stimulate robust intracellular calcium (Ca2+i) release as a result of interaction with calcyon. The role of calcyon in potentiating Ca2+-dependent signaling should provide insight into the D1 receptor-modulated cognitive functions of prefrontal cortex. 相似文献

为了比较妙峰山玫瑰花瓣于采收当天提取和储存4 d后提取所得精油的化学成分变化,通过水蒸气蒸馏法提取玫瑰精油,以GC方法对玫瑰精油中的香叶醇含量进行定量分析,以GC-MS方法对玫瑰精油其他化学成分进行鉴定和定量分析。结果表明:主香成分之一香叶醇的含量,由当日提取的14.91%,冰箱保存4 d后降至6.44%,室温4 d后降至3.11%;GC-MS共检测到89个化合物,鉴定出其中的75个,玫瑰花所含的低沸点成分特别是主体香气成分在储藏4 d后含量明显降低。由此可见:储存对玫瑰精油香气成分有明显影响,冷藏对玫瑰精油成分有一定保护作用。 相似文献

Symmetric application of indole-3-acetic acid, CO(2), or, to a lesser extent, ethylene can substitute for the contact stimulus in inducing coiling in the tendrils of Marah fabaceus. In the case of auxin, treatment of the apical few millimeters results in strong, permanent coiling throughout the length of the tendril. The speed of the response to CO(2) is comparable to that to tactile stimuli. A possible mechanism for thigmotropism is outlined. 相似文献

The ubiquitination of the hypoxia-inducible factor (HIF) by the von Hippel-Lindau tumor suppressor (pVHL) plays a central role in the cellular response to changes in oxygen availability. pVHL binds to HIF only when a conserved proline in HIF is hydroxylated, a modification that is oxygen-dependent. The 1.85 angstrom structure of a 20-residue HIF-1alpha peptide-pVHL-ElonginB-ElonginC complex shows that HIF-1alpha binds to pVHL in an extended beta strand-like conformation. The hydroxyproline inserts into a gap in the pVHL hydrophobic core, at a site that is a hotspot for tumorigenic mutations, with its 4-hydroxyl group recognized by buried serine and histidine residues. Although the beta sheet-like interactions contribute to the stability of the complex, the hydroxyproline contacts are central to the strict specificity characteristic of signaling. 相似文献

Cell-mediated (type-1) immunity is necessary for immune protection against most intracellular pathogens and, when excessive, can mediate organ-specific autoimmune destruction. Mice deficient in Eta-1 (also called osteopontin) gene expression have severely impaired type-1 immunity to viral infection [herpes simplex virus-type 1 (KOS strain)] and bacterial infection (Listeria monocytogenes) and do not develop sarcoid-type granulomas. Interleukin-12 (IL-12) and interferon-gamma production is diminished, and IL-10 production is increased. A phosphorylation-dependent interaction between the amino-terminal portion of Eta-1 and its integrin receptor stimulated IL-12 expression, whereas a phosphorylation-independent interaction with CD44 inhibited IL-10 expression. These findings identify Eta-1 as a key cytokine that sets the stage for efficient type-1 immune responses through differential regulation of macrophage IL-12 and IL-10 cytokine expression. 相似文献

There are compelling reasons for choosing to develop the human as the highest-order experimental system in genetics: an obvious social context that stirs interest, wide medical observation of the population that permits identification of an abundance of genetic defects, and our ability to perceive in the human subtle or complex variations that may not be observable in other species. Various lines of genetic inquiry that are based on research in other systems--cytogenetic analysis, biochemical studies, mapping of defective loci by linkage analysis in affected families, and in vitro techniques such as the creation of transgenic organisms--complement and enrich each other. New phenomena that would not have been predicted from investigations in other organisms have been found in humans, such as the discovery of the "giant" Duchenne muscular dystrophy gene and the identification of recessive cancer genes. Genetic research is yielding insights into human biology that are raising new possibilities for therapy and prevention of disease, as well as challenges to society in the form of ethical decisions about the appropriate application of genetic information. 相似文献

Light generates reducing equivalents in chloroplasts that are used not only for carbon reduction, but also for the regulation of the activity of chloroplast enzymes by reduction of regulatory disulfides via the ferredoxin:thioredoxin reductase (FTR) system. FTR, the key electron/thiol transducer enzyme in this pathway, is unique in that it can reduce disulfides by an iron-sulfur cluster, a property that is explained by the tight contact of its active-site disulfide and the iron-sulfur center. The thin, flat FTR molecule makes the two-electron reduction possible by forming on one side a mixed disulfide with thioredoxin and by providing on the opposite side access to ferredoxin for delivering electrons. 相似文献

以‘湘杂棉14’为材料,采用RT–PCR方法克隆了棉花ABP1基因全长cDNA序列。序列全长792 bp,与GenBank中陆地棉ABP1基因序列的同源性为99%。半定量分析表明,该基因在棉花叶、花及纤维中都有表达,其中叶中的表达量最高,花和纤维中的表达量稍低。ABP1基因在纤维快速伸长期表达水平较高,纤维发育的其他时期的表达水平较低。将ABP1基因cDNA构建成棉花ABP1过表达载体,并以NPT II作为选择标记,经农杆菌介导转化棉花,获得了6株Kan抗性植株,其中3株ABP1表达水平提高。ABP1过表达植株生长发育正常,但纤维长度变化不显著,说明ABP1棉纤维细胞中提高表达水平对其细胞伸长没有显著作用。 相似文献

Proliferation of legume nodule primordia is controlled by shoot-root signaling known as autoregulation of nodulation (AON). Mutants defective in AON show supernodulation and increased numbers of lateral roots. Here, we demonstrate that AON in soybean is controlled by the receptor-like protein kinase GmNARK (Glycine max nodule autoregulation receptor kinase), similar to Arabidopsis CLAVATA1 (CLV1). Whereas CLV1 functions in a protein complex controlling stem cell proliferation by short-distance signaling in shoot apices, GmNARK expression in the leaf has a major role in long-distance communication with nodule and lateral root primordia. 相似文献
