We studied seed morphometric characters, oil content and azadirachtin content in a medicinally important and insect repellent tree species Azadirachta indica A.Juss. Ten populations were selected from two central Indian states of Madhya Pradesh and Chhattisgarh and twenty candidate plus trees were selected from each population based on tree attributes. Azadirachtin content was estimated in seeds, in vitro shoots and callus cultures through HPLC. Significant variation(p \ 0.05) was observed in seed morphometric characters, oil content and azadirachtin content among the ten populations. Maximum value for 100 seed weight was obtained in seeds of Raigarh(30.51 g) which was on par with seeds of Bargi(28.37 g)and Chhatarpur(26.36 g). The ratio of highest kernel to seed was also obtained in seeds of Raigarh(60.11%).Kernels from Chhatarpur population recorded the highest oil content(24.20%), which was equal to that in Sihore(23.75%) and Katni(21.75%). We recorded highly significant variation in azadirachtin content in seed samples,in vitro shoot samples and callus cultures. Maximum azadirachtin was present in the seeds of Gwalior(0.882%)which was on par with azadirachtin content in seeds of Chhatarpur, Katni, Sihore, Shahdol and Chhindwara.Maximum azadirachtin in the in vitro shoots was present in the cultures of Gwalior(0.218%) which was statistically on par with Sihore(0.176%). Similarly, in callus cultures of nodal segments maximum azadirachtin was recorded in the Gwalior population(0.033%) which was statistically on par with Sihore(0.028%). The trees of Gwalior and Sihore populations can be produced in large numbers through vegetative propagation to obtain seeds with higher azadirachtin content.  相似文献

Seed morphometric characters and oil content were studied in multiple-use plant, Calophyllum inophyllum L. of two countries, Australia (southern hemisphere) and Sri Lanka (northern hemisphere). Seven provenances were selected which included three from northern Australia and four from Sri Lanka. Twelve Candidate plus trees (CPTs) each were selected from 2 to 3 different locations within each provenance based on the morphometric and qualitative traits (GBH > 100 cm). Seed collection in both hemispheres was carried out from May to August 2008. Calophyllum inophyllum L. provenances showed a distinct hemisphere variation in their seed-related characters and oil content. All provenances differed significantly (P < 0.05) with one another in seed length. Seeds from Anuradhapura, Sri Lanka and seeds from Cardwell Australia recorded the highest (≈57%) and the lowest oil (≈31%) content respectively. Strong correlations were found between seed morphometric characters. Relatively weaker correlations were found between seed morphometric characters and oil content. Variations in seed-related characters were largely attributed by the provenance contribution which shows the significance of the effect of genetic variability on above mentioned seed related characters.  相似文献

Jatropha curcas L. is being promoted by governments for greening marginal areas for rural development and biodiesel production in sub-Saharan Africa, however large gaps in knowledge on its agronomy and yield potential exist. The effect of duration of storage at room temperature, pre-sowing seed treatment of Jatropha seed and fruit colour harvest index on germination and seedling growth parameters was investigated. There was a significant negative correlation between the duration of storage of Jatropha seed and seed oil content (P < 0.001, r = ?0.99), viability (P < 0.01, r = ?0.93), percent germination (P < 0.05, r = ?0.90) and germination index (P < 0.05, r = ?0.89). The relationship between duration of seed storage and seed oil content and seed germination parameters was best described by quadratic and exponential functions which showed rapid declines in seed oil content and germination parameters in the first 3 years of storage and a slower decline between 3 and 8 years of storage. There was a significant negative correlation between duration of storage and seedling height (P < 0.01, r = ?0.97), stem diameter (P < 0.01, r = ?0.93) and dry weight (P < 0.05, r = 0.91) that was best described by quadratic functions. Seed oil content was significantly and positively correlated to seed viability (P < 0.01, r = 0.96), germination per cent (P < 0.01, r = 0.94) and germination index (P < 0.01, r = 0.93). The relationship between seed oil content and seed germination parameters was best described by exponential functions which showed an exponential increase in seed germination parameters as oil content increased above 30% and smaller increases below 30% oil content. Progressive peroxidation of lipids as seed aged explains the loss of oil from the seed. Peroxidation of lipids produced highly reactive radicals that damaged membranes and proteins reducing seed germination and seedling growth parameters as seed aged. Pre-sowing seed treatments (seed soaking and scarification) produced marginal gains in germination and seedling growth parameters in Jatropha seed stored for 3 months to 8 years. Seeds harvested at the yellow fruit maturity index, 60 days after anthesis (DAA), showed the highest seed germination percent while seed harvested at the black fruit maturity index, 75 DAA, had the highest oil content. Harvesting of Jatropha seed for oil expression is therefore recommended at the black maturity index while harvesting for seed is recommended at the yellow maturity stage. The results of this study indicate that Jatropha seed should be stored for as short a period of time as possible to maximize seed germination, seedling growth and oil yield.  相似文献

Cinchona officinalis (Rubiaceae) is an endemic species of the Loja Valley in southern Ecuador with medicinal uses. Because of over-exploitation in the nineteenth century and more recent disturbances to its ecosystem, C. officinalis populations are threatened. Currently, natural regeneration of the populations is low, despite its high plant regeneration and seed formation capacity. In the present study, an efficient protocol for germination, shoot proliferation and plantlets regeneration was developed for this species. Phenolic content and germination rate of C. officinalis seeds were compared with a control species, C. pubescens. Nodal segments from seedlings of C. officinalis were cultured on Gamborg medium supplemented with different combinations of plant growth regulators. Because the phenol content is high in C. officinalis, the phenolic should be removed with hydrogen peroxide or water washes to stimulate germination. Shoots and callus developed from nodal segments within 45 days using most of the tested combinations of plant growth regulators. The best rates of shoot proliferation, callus formation and adventitious buds were obtained in medium supplemented with 5.0 mg L?1 6-benzyl-aminopurine and 3.0 mg L?1 indole-3-butyric acid.  相似文献

Nitraria sibirica Pall. is a shrub that grows in saline-alkali soil and has traditional medicinal value and potential commercial value. The objectives of this study include induction and multiplication of callus, establishment of a suspension cell line, and isolation of protoplasts from cell suspensions. Murashige and Skoog (MS) medium was used for callus induction from mature seeds of N. sibirica. Seed-derived calluses were further multiplied on MS medium augmented with 0.5 mg L?1 6-benzylaminopurine (6-BA) and 1.0 mg L?1 2,4-dichlorophenoxy (2,4-D) acetic acid. Suspension cultures of N. sibirica were initiated by transferring friable calli to the same liquid multiplication medium. Characterization of the suspension culture was assessed based on fresh mass, dry mass, cell viability and pH value of the culture. A typical growth curve was observed after inoculating 1.5 g of callus in 40 mL liquid medium, including a lag phase, an exponential growth phase, a stationary phase, and a negative acceleration phase. The effect of factors such as pre-plasmolysis, enzyme combination, enzymolysis time and mannitol concentration, on the isolation of cell-derived protoplasts were evaluated to determine the usefulness of suspension cultures. The maximum yield (9.79 × 106 cells/g) and highest viability (79.97%) of protoplast were reached when approximately 1 g of cell suspension (cultured for 6 days) was inoculated for 12 h in cell and protoplast washing solution made of 0.8 mol L?1 mannitol mixture solution, cellulose onozuka R-10 2% (w/v), hemicellulose 0.2%, macerozyme R-10 1%, and pectolyase Y-23 0.5%. Protoplast yield was significantly influenced by pre-plasmolysis and cellulose onozuka R-10 (P < 0.05).  相似文献

An evaluation of 30 naturally available candidate-plus trees (CPTs) of Pongamia pinnata was carried out to elucidate the genetic variability and relationship of pod and seed traits and progeny growth traits to select the best planting material with higher productivity. Significant variations were observed in pod and seed characteristics, oil content, and during the progeny trial studies. The highest values were found in 100-pod weight (410.4 gm) and 100-seed weight (195.92 gm) and the phenotypic and genotypic variance was found maximum in 100 pod weight (4364.71 and 4289.93) and 100 seed weight (813.8 and 768.93), respectively. Estimates of the phenotypic coefficient of variance were higher than the genotypic coefficient of variance for all the pod and seed traits and progeny growth traits, which depict the predominant role of the environment. Approximately 99 % of broad-sense heritability was revealed in oil content, followed by 100-pod weight (98.28 %), and canopy growth (96.93 %). Pod thickness shows the highest genetic advance of 56.61 %, followed by pod width (53.43 %) and canopy growth (49.3 %). Pod and seed traits have proven a positive correlation between each other and with progeny growth traits, except a few negative values. Hierarchical clustering by Ward’s minimum variance cluster analysis showed phylogeographical patterns of genetic diversity. K-means clustering revealed that trees from different geographical regions were grouped together in a cluster where as trees from the same geographical area are placed in a different cluster. Seven Pongamia ecotypes (GRP-8, 9, 13, 14, 18, 28, and 29) are found superior in traits, namely 100-pod weight, 100-seed weight, oil content, germination rate, plant height, canopy growth, and collar diameter. The traits 100-pod weight and 100-seed weight are highly correlated with the progeny growth traits and even have higher heritability and genetic advantages.  相似文献

Authentication of seed provenance is an importance issue to avoid the negative impact of poor adaptation of progenies when planted outside their natural environmental conditions. The objective of this study was to evaluate the potential of near infrared (NIR) spectroscopy as rapid and non-destructive method for authentication of Picea abies L. Karst seed provenances. For this purpose, five seed lots from Sweden, Finland, Poland and Lithuania each were used. NIR reflectance spectra were recorded on individual seeds (n = 150 seeds × 5 seed lots × 4 provenances = 3000 seeds) using XDS Rapid Content Analyzer from 780 to 2500 nm with a resolution of 0.5 nm. Classification model was developed by orthogonal projection to latent structures-discriminant analysis. The performance of the computed classification model was validated using two test sets—internal (the same seed lots as the model but excluded during model development; n = 600 seeds) and external (seed lots not included in the model; n = 1158 seeds). For the internal test, the model correctly recognized 99% of Swedish, Finnish and Polish samples and 97% of Lithuanian seeds. For the external test samples, the model correctly assigned 81% of Swedish, 96% of Finnish, 98% of Lithuanian and 93% of Polish seeds to their respective classes. The mean classification accuracy was 99 and 95% for internal and external test set, respectively. The spectral differences among seed lots were attributed to differences in chemical composition of seeds, presumably fatty acids and proteins, which are the dominant storage reserves in P. abies seeds. In conclusion, the results demonstrate that NIR spectroscopy is a very promising method for monitoring putative seed provenances and in seed certification.  相似文献

The germination of freshly collected Juniperus polycarpos (K. Koch) seeds is very low and attributed to the large proportion of nonviable seeds in the seed lots. Thus, the aim of this study was to improve seed lot quality by removing nonviable seeds using two flotation techniques: incubation-drying-separation (IDS) and modified specific gravity (MSG) separation. We examined different IDS conditions (the specific incubation time, subsequent drying time and sorting media) for effectively sorting out nonviable seeds; and tested the feasibility of modified SG separation, which involved soaking seeds in water for a certain period before sorting in different concentrations of sucrose solution. Viable seeds were expected to absorb and metabolically bind more water during soaking than dead seeds could, and hence be sorted effectively depending on the viscosity of the sucrose solution. The viability of the seeds that floated or sank was determined in a topographical tetrazolium chloride (TTC) test. For the IDS trial, 7 days incubation followed by 9 h of drying and sedimentation in pure water or 200 g·L?1 sucrose solution identified, respectively, 75 and 82 % of the seeds as viable (sunken) seeds. For the MSG trail, 77 % viable seeds were recovered in the sunken fraction when seeds were soaked for 48 h then immediately sorted in 600 g·L?1 sucrose solution. In both cases, the loss of viable seeds in the discarded floating fraction was only 4 %. The results demonstrate that both IDS and MSG separation techniques substantially improved seed lot viability, but MSG separation is simple and needs no modern seed handling facility, and its efficacy relies on seed mass (due to the initial soaking) and the specific density and viscosity of the flotation medium.  相似文献

Abstract

The response of common alder [Alnus glutinosa (L.) Gaertn.] and downy birch (Betula pubescens Ehrh.) seeds to germination temperature was examined following chilling and priming. Seeds of two seed lots of each species were subjected to combinations of chilling (4±1°C) and priming (20°C) treatments in fully imbibed (FI) state or a lower target seed moisture content (TMC) level (30% and 35% in alder and birch, respectively). After treatment, the seeds were allowed to germinate for 56 days at constant temperatures of 7.5, 10, 15, 20, 25 or 30°C. The response to temperature and pretreatment differed between species, but the effect was consistent in each seed lot within each species. In alder, the TMC seeds germinated well across the full range of temperatures, whereas there was an optimum temperature (22–23°C) for seeds given the FI pretreatment. Priming had no significant effect on the germination response of the TMC seeds in alder, but priming greatly improved germination in the FI seeds, especially at the lower germination temperatures (optimum 18–19°C). In contrast, in birch, the TMC seeds germinated better across the full temperature range, but the optimal germination temperature (15°C) was the same for all seed pretreatments. Priming improved germination in both the FI and TMC seeds in birch.  相似文献

Ponderosa pine (Pinus ponderosa Dogl. ex. Laws.) is the most planted conifer species in the forest-steppe ecotone of Patagonia, Argentina, because of its adaptability and excellent growth rates. In spite of this, and the increasing demand for this species, local commercial seed lots showed low quality, making hazardous seedling production. Aiming to upgrade germinability of a local seed lot, we set an experiment to determine the duration of the prechilling period (0, 10, 20, 30, 40 and 60 days) that promoted the highest seed germination speed (GE) and percentage (GP). Moreover, part of that lot was IDS treated, in an attempt to separate empty and dead filled seeds from viable seeds. Results showed that after 40 days prechilling, GE reached 62%, and GP 70%, both higher than the values obtained under customary conditions. The application of the IDS technique, after 40 days prechilling, 8 h drying at ambient conditions (16 ± 2°C and 50% HR), and 25% seed moisture content (mc), increased GE and GP to 68% and 89%, respectively. Optimal prechilling periods and the application of the IDS technique successfully improved germinability of ponderosa pine seeds from Patagonian stands.  相似文献

The aim of this study was to determine the efficiency of methods for the artificial scarification of black locust seeds based on the influence of the air and liquid nitrogen temperatures. Three populations were selected for the analyses: a managed tree stand, a selected seed stand and a seed orchard. Samples of 200 seeds (4 × 50) were randomly selected. Before scarification, the seeds were stored at ?70 °C for 24 h. Nine different scarification treatments were evaluated. To calculate the germination capacity and the dynamics of germination, observations were made on days 3, 5, 7, 10 and 14 after treatment. For statistical analyses, we used repeated measures analysis of variance. The obtained results indicated that the highest scarification efficiency for black locust seeds occurred in the treatment in liquid nitrogen and in an air temperature range of 85–90 °C. The provenance of seeds had a significant effect on the dynamics of germination. Thermal methods and the use of liquid nitrogen can be alternatives to the labour intensive methods of cutting the seeds or using measures that require specialised equipment.  相似文献

A nursery experiment was conducted to determine the effects of seed pretreatment methods on the germination of Faidherbia albida at ICRAF, Nairobi from six provenances consisting of Awassa, Taveta, Lake Koka, Maseno, Chinzombo and Wagingombe. Seeds were subjected to five pretreatment methods namely nicking, soaking in acid, hot water, cold water and control. Germination percentages (GP), mean germination time (MGT) and germination index (GI) were calculated and the data was subjected to ANOVA. The study revealed significant (p ≤ 0.05) differences in seed treatments among provenances in all studied parameters. Highest germination among pretreatments in Awassa (99 %) and Wagingombe (80 %) was observed in nicked seeds, Chinzombo (81 %) in nicked and acid treated seeds, Lake Koka (90 %) in acid treated seeds and Taveta (28 %) and Maseno (64 %) in cold water treated seeds. Nicking gave the highest cumulative GP (69.67) while lowest GP was observed in hot water treated seeds (23.17). Acid treatment exhibited lowest MGT (8.85 days) and highest GI (2.29) while highest MGT (24.35 days) and lowest GI (0.31) were observed in control and hot water treatment respectively. Although acid treatment gave a high GP and lowest MGT and GI, nicking and soaking in cold water for 24 h is being recommended as cheaper and less hazardous pretreatment methods to improve germination in F. albida since sulphuric acid is expensive and requires proper handling techniques. Significant correlation between geo-climatic data and germination parameters of seeds subjected to different pretreatments indicates that provenances are as important as pretreatments in germination of the species.  相似文献

The effects of the presence of a medium, seed moisture content, warm and chilling treatments on the germination response of European rowan (Sorbus aucuparia L.) seeds of Irish origin were investigated with the aim of reducing precocious (or premature) germination and maximising germination after pretreatment. After adjusting seed moisture content, seeds of two lots were subjected to warm treatment for 0–6 weeks followed by 0–50 weeks of chilling. The seeds then were allowed to germinate at a constant 15 °C with 8 h lighting per day or 20 (dark)/30 °C (light). Some seeds were treated in the fully imbibed (FI) state in a medium (standard operational treatment). Seed lot effects were evident, but treatment effects were consistent in each lot. Warm treatment greatly increased germination and reduced precocious germination. The presence of a medium was not required to achieve high germination and seed moisture content close to the FI state was needed to maximise germination. Climate change may reduce the ability of this species to regenerate naturally, favouring other species that require less chilling.  相似文献

From four Korean pine (Pinus koraiensis) orchards, 60 clones were selected and analyzed for the fatty acid and amino acid components of the seeds to reveal the variations and correlations of the seed characteristics among the clonal source orchards and clones. The nutritional components of the seeds of the P. koraiensis trees exhibited rich genetic variation; the variation coefficient of the fatty acids was 2.24–66.83 %, while the variation coefficient of the amino acids was 14.70–38.88 %. Relatively high genetic-improvement potential exists for the nutritional components of the seeds. The phenotypic differentiation of the fatty acid and amino acid components reveals that variation within the population (85.18 %) was the primary source for the variation of the fatty acid components; variation among the orchards (63.08 %) was the primary source of the variation of the amino acid components. Data drawn from various clonal source orchards all showed that the seed characteristics were highly controlled by heritability (h2 > 80 %), and the seed characteristics of the P. koraiensis trees exhibited a similar genetic gain trend. The principal components were analyzed to obtain the comprehensive principal component value for each clonal seed orchard. Twelve clones were selected based on a clonal selection rate of 20 %. Correlation and multiple stepwise-regression analyses were conducted, considering different location conditions, to reveal the stable correlations between the seed characteristics to facilitate improvements of the seed yield of P. koraiensis trees and the clonal selection. Species of real characteristics in P. koraiensis were controlled by higher heritability. Genetic gain was obtained by selecting of superior clones.  相似文献

Plants have compensating mechanisms to alleviate the loss of vegetative and reproductive tissues by herbivores. In this study, we evaluated how reproductive traits (size and production of fruit and seed, as well as germination and viability of seeds) of Myrtus communis (myrtle) were affected by goat grazing exclosure of forest understory in the Doñana Natural Park. One hundred and fifty myrtles were selected in three sites with similar environmental characteristics, although with a different length of grazing exclusion: (1) exclosure for 2 years (GE2); (2) exclosure for 3 years (GE3); and (3) long ago grazed plots (GE15): non-accessible to goat for at least 15 years. Fruit production, fruit and seed morphology and seed viability and germination were evaluated. Length of exclosure to goats increased fruit production, mass and volume of the fruits and seed, and endocarp mass. However, seed viability was higher in more recently grazed plots (70–90%) than in GE15 plots (51%). The final balance of viable/germinable seeds produced by an average sized myrtle in each treatment showed that the recently grazed myrtles (GE2) produced nearly five times less seeds (1200) than the myrtles grazed 15 years ago (5500). However, myrtles recovered the ability to produce viable seeds quickly, almost tripling, each year of non-grazing the number of viable/germinable seeds. The results of this study indicated that goat grazing could affect regeneration of myrtle because it reduced reproduction output and the number of viable propagules. Future studies should evaluate other related ecological aspects, such as fruit consumption by potential frugivores and seed dispersal (distance and place of seed deposition).  相似文献

Context

Black poplar (Populus nigra L.) is an alluvial forest tree species whose genetic pool is decreasing in Europe. Poplar trees produce short-lived seeds that do not store well.

Aim

The feasibility of seed storage in conventional and cryogenic conditions after their desiccation from water content (WC) of 0.15 to 0.07 g H2O g?1 dry mass (g g?1) was investigated.

Methods

Seed germinability was evaluated (seeds with a radicle and green cotyledons were counted) after storage of seeds for a period of 3 to 24 months at different temperatures: 20°, 10°, 3°, ?3°, ?10°, ?20° or ?196°C.

Results

Seeds desiccated to a 0.07 g g?1 WC can be stored successfully at ?10 °C and ?20 °C for at least 2 years. A significant decrease in germination was observed only after 12 months of seed storage (WC 0.15 g g?1) at temperatures above 0 °C. We demonstrated that both fresh (0.15 g g?1 WC) and desiccated (0.07 g g?1 WC) seeds can be preserved at ?196 °C for at least 2 years.

Conclusions

Seed storage temperature and time of storage were statistically significant factors affecting seed storability. The presented data provide a foundation for the successful gene banking of P. nigra seeds.  相似文献

The Ecuadorian Government has, since the end of 2011, promoted an initiative for using pure jatropha oil, derived from the seeds of Jatropha curcas L., to generate electricity to supply Floreana Island (one of the Galapagos Islands). In the present study, nine elite accessions of J. curcas from tropical dry forests in Ecuador were cultivated for the purpose of evaluating seed production in relation to the propagation method used (seed, cutting). A split-plot design with three replicates was applied. Seed production, measured over a period of 7 years (2009–2015) showed a large year-to-year variation. However, seed production varied significantly among accessions and depended on the propagation method (seeds or cuttings). Propagation by stem cuttings stabilized the variability in the vegetative and seed production components measured, increasing the strength of their relationships with seed yield. The three highest yielding accessions produced 0.28–0.30 kg of seeds tree?1 year?1. As seed yield was not correlated with annual precipitation, the year-to-year variation in production of accessions indicates the effect of other factors not considered here, such as agronomic practices. The study findings suggest that agroforestry practices could be developed that would enable J. curcas to play a predominant role in a strategy for tropical dry forests aimed at reducing poverty (also known as pro-poor growth strategy).  相似文献

Knowledge of seed desiccation tolerance is fundamental for conservation and use of forest species. The protocol used for classification of seed desiccation tolerance and storage is time consuming and many times limited by the lack of information about optimum conditions for seed germination and treatments to overcome seed dormancy. This study evaluated 66 Brazilian tree species aiming to correlate seed characteristics with desiccation tolerance. For this purpose, a model was established to explain the relationship of tegument/seed mass ratio (SCR), seed mass, and water content of embryo + endosperm with desiccation tolerance. The principal component analysis showed the establishment of two groups, indicating the interaction between desiccation tolerance and seed characteristics. Recalcitrant seeds are more often associated with the water content of embryo + endosperm and water content of tegument + endocarp, while orthodox seeds are more associated with SCR and number of seeds per kilogram. The classification found using the model proposed was significantly correlated with desiccation tolerance and storage, with 92% confidence for the analyzed species. Seeds morphological characteristics can be used for prediction of desiccation tolerance and storage behavior; however, the use of a model that combines more variables increases the chance of accurate classification.  相似文献
