The diversity of arbuscular mycorrhizal (AM) fungi in soils under a yam cropping system in four agroecologies of Nigeria was investigated. Soil samples were collected from yam fields at Onne (humid forest, high rainfall area), Ibadan (derived savanna), Abuja (Guinea savanna) and Ubiaja (humid forest, medium rainfall area). Soil characteristics, AM fungi species, spore abundance, Shannon diversity index, species richness and evenness were determined. A total of 31 AM fungi species was isolated from the four agroecologies with a range of 14–20 species found in a single location. Glomus species were the most abundant among AM fungi species with G. geosporum, G. intraradices and G. mosseae occurring in large populations in all locations. Ubiaja, which had a cassava/natural vegetation sequence before yam, had significantly higher spore abundance and species richness than the other locations, which had a yam/legumes or a maize/legume sequence before yam. However, diversity was significantly higher at Abuja, which had a maize/legume sequence with yam, than Ibadan, which had only a yam/legume sequence. The study revealed significant diversity in AM fungal species across agroecologies in yam-growing regions. Further research on the functional consequences of changing composition of AM fungi species across the region is recommended.  相似文献

Maize (Zea mays L.) is an important crop in central Thailand where fallow is widely practiced and farmers are interested in crop rotation and beneficial soil biota. A pot experiment using a Typic Paleustult (topsoil + subsoil) from the National Corn and Sorghum Research Centre, Nakhonratchasima Province, Thailand was undertaken over three successive crops to evaluate effects of agronomic practices on populations of arbuscular mycorrhizal (AM) fungi and to determine whether reintroduction of a local Glomus was beneficial to maintain maize yield. The three crops and their treatments were: (1) preceding crop: maize grown in all pots; (2) subexperiment 1: agronomic practices [maize, fallow ± soil disturbance, fallow with solarization, non–AM host (cabbage)]; and (3) subexperiment 2: maize ± Glomus sp. 3 at three rates of P fertilization (0, 33, 92 kg P ha–1). The AM‐fungal community was established under the preceding crop. In subexperiment 1, the three fallow treatments decreased (30%–40%) the total AM spore number in the topsoil whereas there was no change under maize or cabbage. Glomus, the dominant genus, showed sensitivity to fallow. In subexperiment 2, inoculation with Glomus sp. 3 enhanced total AM spore number and root colonization when applied following the three fallow treatments. Furthermore, inoculation promoted grain yield; at nil P following fallow ± soil disturbance, at 33 kg P ha–1 following fallow without soil disturbance, and following solarization. Two treatments, maize following maize and maize following cabbage, did not respond to inoculation with Glomus sp. 3. Overall, the results suggest that reintroduction of Glomus sp. 3, a local AM fungus in this soil, may overcome negative effects of fallow and promote effectiveness of P fertilizer. Further work is needed to evaluate the benefits of other indigenous AM species that persist under modern fertilization practices.  相似文献

丛枝菌根(AM)真菌可以与湿地植物共生,并在湿地生态系统的修复与维护过程中具有重要作用。然而,不同环境条件下湿地土壤AM真菌群落分布特征及其影响因素仍然有待明晰。选择鄱阳湖湿地不同水位条件(高水位和低水位)2种典型植物群落(南荻和水蓼)为研究对象,运用高通量测序技术探究水位和植被类型对鄱阳湖湿地土壤AM真菌群落结构和多样性的影响,比较不同环境条件下AM真菌群落结构和多样性差异,分析其与土壤理化性质的关系。结果表明,AM真菌OTU数为21~38,水位和植被类型影响鄱阳湖湿地土壤AM真菌OTU数,高水位下AM真菌OTU数高于低水位,水蓼群落高于南荻群落。被鉴别出的菌属为Glomus、Claroideoglomus和Paraglomus,其中,Glomus是优势属,占比85%以上,其相对丰度在高水位下显著高于低水位(p<0.05),南荻群落高于水蓼群落。AM真菌多样性指数(Simpson和Shannon指数)受水位和植被类型以及二者交互影响(p<0.01),低水位下不同群落之间的AM真菌多样性指数无显著差异,而高水位下水蓼群落AM真菌多样性指数显著高于南荻群落(p<0.05...  相似文献

对贵州务川汞矿区不同水平汞污染植物根际土壤丛枝菌根(Arbuscular Mycorrhizal,AM)真菌资源分布及多样性进行分析,结果发现:本研究选取的务川汞矿区3个样地土壤汞含量在94.1~268.0 mg/kg,远高于国家规定的土壤汞含量,汞污染极其严重。随着汞含量增加,AM真菌侵染率、孢子密度显著降低,AM真菌群落丰富度、多样性及均匀度均有所降低,但差异未达显著水平,而仍有部分对汞污染耐受性较强的AM真菌有较高的分布。另外,管柄囊霉属AM真菌在5个样品中均有较高丰度,球囊霉属AM真菌在火棘根际的相对丰度随着汞含量升高而升高,因此,火棘–管柄囊霉属AM真菌或火棘–球囊霉属AM真菌联合进行汞污染修复,有一定的应用价值。  相似文献

 We studied fluctuations in the numbers of autotrophic ammonium oxidizers, ammonifying microorganisms and denitrifying microorganisms in pot cultures of mycorrhizal and non-mycorrhizal maize. The populations were enumerated after 0, 15, 30, 45 and 60 days of plant growth. Two arbuscular mycorrhizal (AM) fungi belonging to different Glomus species were investigated. Pot cultures with AM-infected maize had significant quantitative and qualitative changes in the root-associated population of N-transforming bacteria compared with the non-mycorrhizal controls. The occurrence of autotrophic ammonium oxidizers in pot cultures of the AM fungi Glomus mosseae and G. fasciculatum was significantly higher than in non-mycorrhizal cultures throughout maize growth. The occurrence of these bacteria was delayed by 15 days in non-mycorrhizal as opposed to Glomus-colonized soil. Ammonifying and denitrifying bacterial populations were significantly decreased in the pot cultures of AM plants compared with the control. The distribution patterns of the physiological groups of bacteria tested were similar for both AM treatments but different from that of the non-mycorrhizal controls. Activity measurements expressed on a per cell basis showed changes with respect to the form of N in the mycorrhizal soil. Intracellular arbuscular mycorrhizal (AM) colonization was compared between nitrogen (NH4NO3) fertilized (10 g N m–2) and nonfertilized tallgrass prairie plots. In the microscopic analyses of host roots, only intracellular coils showed an increasing trend as a result of N fertilization, whereas intracellular colonization by arbuscules, hyphae, or vesicles did not differ between the N treatments. Clone libraries established from pooled PCR products of AM fungi contained exclusively species of Glomus; no other genera were detected indicating that Glomus spp. dominated the host roots. Comparisons between observed and random topologies indicated that cloned sequence placement covaried with N treatment: unique clades within Glomus originated exclusively from N-fertilized or nonfertilized treatments. We conclude that the communities of dominant and most commonly occurring AM fungi changed in response to N amendment, although the root colonization showed minimal or no response.  相似文献

Grapevines form associations with arbuscular mycorrhizal (AM) fungi. These root-dwelling fungi have the potential to contribute to crop vigor, productivity, pathogen protection, and nutrient content in grapes. In this study the arbuscular mycorrhizal fungal communities of grapevines and the surrounding interrow and native vegetation are compared. We found over 40 different taxa associating with both vines and interrow vegetation, but these communities differed based on host plant identity. These differences were apparent even after accounting for differences in soil chemical properties and differences in host plant diversity between vinerows and interrows, indicating that Vitis preferentially interacts with a subset of the viticultural fungal community. Since AM fungal communities play a major role in grapevine health, our results suggest that host identity and the diversity of AM fungal hosts in a vineyard can have strong effects on arbuscular mycorrhizal fungi community structure. A greenhouse study was conducted to study the efficiency of 14 isolates of arbuscular mycorrhizal (AM) fungi isolated from a local agricultural soil on the productivity of sweet potato (Ipomoea batatas). The different AM fungi enhanced the biomass and nutritional status of sweet potato seedlings to different extents. The genus Glomus was more effective than Acaulospora or Scutellospora. Efficiency also varied among isolates of Glomus irrespective of individual host plant or location of origin. Intraspecific differences were sometimes greater than interspecific differences. Benefits deriving from fungal isolates were positively correlated with the root-colonization rate and the abundance of extraradical propagules of the AM fungi. Taking plant yield parameters, nutritional status of the plants, and fungal attributes into consideration, GEGM (Glomus etunicatum together with Glomus mosseae) and GE6 (Glomus etunicatum) were the most effective AM symbionts for sweet potato under the experimental conditions.  相似文献

The interactions between Phaseolus vulgaris, Rhizobium spp. strains nodulating P. vulgaris, and arbuscular mycorrhizal (AM) fungi were assessed under greenhouse conditions in a nonsterilized Typic Haplustalf soil from Cauca, Colombia. Our results indicate a specific involvement of AM fungal species in nitrogen acquisition by the legume plants from symbiotic nitrogen fixation and from soil. A significant specific influence of inoculation with Glomus spp. on the 15N/14N ratio in plant shoots was dependent on the inoculated rhizobial strain, but AM fungal inoculation had no significant effect on shoot dry weight or nodule occupancy in the two different rhizobial strain treatments. The results imply that in low P soils the effects of an improved mycorrhizal symbiosis may include improved symbiotic N2 fixation efficiency and/or improved soil N uptake. The arbuscular mycorrhizal fungi (AMF) communities composition regulate plant interactions and determine the structure of plant communities. In this study we analysed the diversity of AMF in the roots of two perennial gypsophyte plant species, Herniaria fruticosa and Senecio auricula, and an annual herbaceous species, Bromus rubens, growing in a gypsum soil from a semiarid area. The objective was to determine whether perennial and annual host plants support different AMF communities in their roots. The roots were analysed by nested PCR, cloning, sequencing of the ribosomal DNA small subunit region and phylogenetic analysis. Twenty AMF sequence types, belonging to the Glomus group A, Glomus group B, Diversisporaceae, Acaulosporaceae, Archaeosporaceae and Paraglomeraceae, were identified. Both gypsophyte perennial species, H. fruticosa and S. auricula had different compositions of the AMF community and higher diversity than B. rubens. This annual plant species shared the full composition of its AMF community with both perennial plant species. Seasonal variations in the colonisation of AM fungi could explain the observed differences in AMF community composition, but this is still a working hypothesis that requires the analysis of further data obtained from a higher number of both annual and perennial plant species in order to be fully tested.  相似文献

It is suggested that the diversity of arbuscular mycorrhizal fungi (AMF) and their association with distinct plants species are crucial in the early stages of revegetation procedures since the AMF roots colonisation plays an important role improving plant establishment and growth. We carried out a study where we analyse the AMF community composition in the roots of Ephedra fragilis, Rhamnus lycioides, Pistacia lentiscus and Retama sphaerocarpa fourteen months after revegetation in a Mediterranean semiarid degraded area of southeast Spain in order to verify whether different plant species can variably promote the diversity of AM fungi in their rhizospheres after planted. We analysed a portion of approximately 795 bases pairs of the small-subunit ribosomal DNA by means of nested PCR, cloning, sequencing and phylogenetic analyses. Eight fungal sequence types belonging to Glomus group A and B and to the genus Paraglomus were identified. The different plant species had different AM fungal community composition. Thus, R. lycioides harboured the highest number of four fungal sequence types while from E. fragilis only two types could be characterized that were specific for this plant species. P. lentiscus and R. sphaerocarpa harboured each one three sequence types and two of them were shared. All AMF sequence types were found in the natural soil. These results show that one effective way of restoring degraded lands is to increase the number of plant species used, which would increase the AMF diversity in the soil and thus the below-ground, positive interactions.  相似文献

对西北地区5个酿酒葡萄赤霞珠(Vitis vinifera L. cv. Cabernet Sauvignon)葡萄园根际土壤060 cm土层的AM真菌空间分布进行了研究。结果表明,葡萄根系可形成丛枝菌根,且侵染率较高,最高达79%; 在西北地区的5个样地中共分离出AM真菌4属22种,其中球囊霉属(Glomus)15种,无梗囊霉属(Acaulospora)4种,盾巨孢囊霉属(Scutellospora)2种,巨孢囊霉属(Gigaspora)1种。5个样地孢子密度大小顺序为: 陕西泾阳(JY)>山西永济(YJ)>陕西杨凌(YL)>宁夏银川(YC)>甘肃莫高(MG)。各样地葡萄根际土壤中AM真菌种的丰富度不同,陕西泾阳地区最高; 分布于葡萄根际的AM真菌按种类多少排序的属依次是: 球囊霉属无梗囊霉属盾巨孢囊霉属巨孢囊霉属,球囊霉属占据的比例保持着绝对优势; 根内球囊霉、摩西球囊霉、地表球囊霉在不同样地中均为优势菌株,副冠球囊霉,集球囊霉,细凹无梗囊霉是多数样地中的稀有种类。研究表明,葡萄与AM真菌具有良好的共生关系,二者协同进化产生了具有生态环境特异性的菌根真菌多样性; 葡萄根际存在较为丰富的丛枝菌根真菌资源,可供进一步开发利用。  相似文献

为了研究土地利用方式对酸性红壤丛枝菌根真菌(arbuscular mycorrhizal fungi, AMF)群落的影响,调查了酸性红壤4种土地利用方式(草地、玉米、花生和大豆)下非根际和根际土壤AMF群落多样性和组成结构。结果表明:土地利用方式显著影响了AMF群落优势属球囊霉属(Glomus)和巨孢囊霉属(Paraglomus)的相对丰度,但是根际作用影响不明显。土地利用方式而非根际作用显著影响了AMF群落香农指数和物种丰富度,其中大豆地表现出最低的香农指数和物种丰富度。土地利用方式和根际作用都显著影响AMF群落组成结构,但是土地利用方式的作用强度明显高于根际作用。球囊霉属主要解释了不同土地利用方式之间的AMF群落组成差异。土壤p H是影响土壤AMF群落结构的最关键因子。因此,土地利用方式比根际作用表现出对酸性红壤AMF群落更大的影响,展现了土地利用变化在影响土壤AMF群落方面的重要作用。  相似文献

为阐明毛乌素沙地3种典型克隆植物沙鞭[Psammochloa villosa(Trin.)Bor.]、羊柴(Hedysarum leaveMaxim)和油蒿(Artemisia ordosica Krasch.)根际AM真菌多样性,2006年的5月、7月、10月从毛乌素沙地选取东北缘的中国科学院植物研究所鄂尔多斯沙地草地生态研究站和西南缘的陕西榆林珍稀沙生植物保护基地两个样地,按0~10 cm、10~20 cm、20~30 cm、30~40 cm、40~50 cm 5个土层采集3种克隆植物根际土壤样品,研究了其根际AM真菌物种多样性和生态分布。在分离出的4属23种AM真菌中,球囊霉属(Glomus)15种,无梗囊霉属(Acaulospora)5种,巨孢囊霉属(Gigaspora)2种,盾巨孢囊霉属(Scutellospora)1种。摩西球囊霉(G.mosseae)是沙鞭根际的优势种,黑球囊霉(G.melanosporum)是3种克隆植物共同的常见种;不同属种的AM真菌生态分布亦存在差异。AM真菌孢子密度、种的丰度和物种多样性指数均表现为在研究站样地的羊柴根际最高。该研究结果表明,毛乌素沙地的3种典型克隆植物与AM真菌之间形成良好的共生关系,这对开发漠境AM真菌资源和利用菌根生物技术维护沙地生态系统结构的完整性具有重要意义。  相似文献

Purpose

Modern agricultural science has greatly reduced the use of tillage. Monitoring conservation versus conventional tillage effects on soil microbes could improve our understanding of soil biochemical processes and thus help us to develop sound management strategies. The objective of this study was to investigate the effects of conservation tillage on the spore community structure and the diversity of soil arbuscular mycorrhizal (AM) fungi and to find out the main factors that influence these parameters.

Materials and methods

A long-term field experiment established in a sandy loam soil in Northern China has received continuous tillage management treatments for 3?years, including conventional tillage (CT), no tillage (NT), and alternating tillage (AT). Topsoil samples (0?C15?cm) from four individual plots per treatment were collected for the analysis of chemical properties and fungal parameters. AM fungal spores were isolated using the wet-sieving method and identified to species level based on morphology by light microscopy. The community structure and the diversity of AM fungi were evaluated using the following parameters: spore density, relative abundance, species richness, Shannon?CWiener index (H??), evenness (E), and Simpson's index (D). Jaccard index (J) of similarity was calculated to compare AM fungal species composition under different treatments.

Results and discussion

Twenty-eight species of AM fungi within four genera, Glomus, Acaulospora, Scutellospora, and Entrophospora, were recovered from the 12 plots within the three tillage management treatments. Higher spore density, species richness, and species diversity (H??, E, and D) of AM fungi were observed in the two conservation tillage treatments, and the redundancy analysis showed that the species richness significantly correlated to soil organic carbon content (P?<?0.05). The positive effects of NT and AT on the species richness were very close, while the AT had relatively greater beneficial impacts on the spore density and the evenness of AM fungi compared to the NT. The lowest Jaccard index (J) of similarity in species composition was also observed between the AT and CT treatments.

Conclusions

Soil organic carbon, the spore density, and species richness and diversity of AM fungi increased in the two conservation tillage treatments. The species richness of AM fungi significantly correlated to soil organic carbon content (P?<?0.05). Compared with the CT treatment, the AT rather than the NT significantly increased the spore density and the evenness of AM fungi (P?<?0.05). Thus, alternating tillage practice may be more beneficial to agroecosystem in this region.  相似文献

Arbuscular mycorrhizal fungi (AMF) have great potential for assisting metal-hyperaccumulating plants in the remediation of contaminated soils. However, little information is available about the symbiosis and community composition of AMF associated with manganese (Mn) hyperaccumulator, such as Phytolacca americana, growing on Mn-contaminated soils under natural conditions. Therefore, the objective of this study was to analyze AMF diversity and community composition in P. americana roots growing at an Mn mining site. Molecular techniques were used to analyze AMF community composition and phylogenetic relationship in P. americana roots sampled from three Mn mine spoils and one adjacent reference areas. Results obtained showed that mycorrhizal symbionts successfully established even in the most heavily Mn-polluted sites. Root colonization and AMF diversity were significantly negatively correlated with total and extractable Mn concentrations. Principal component analysis (PCA) revealed that Mn contamination impacted AMF diversity, and shaped AMF community structure. Phylogenetic analyses demonstrated that all species were affiliated with Glomus, suggesting that Glomus was the dominant genus in this AMF community. Some unique sequences that occurred exclusively in heavily polluted sites associated with P. americana may belong to symbiotic fungi with great potential for improving the phytoremediation efficiency of Mn-contaminated soils.  相似文献

A mesocosm experiment was conducted to investigate whether communities of arbuscular mycorrhizal (AM) fungi associated with roots of native (Piptatherum miliaceum, Retama sphaerocarpa, Psoralea bituminosa, Coronilla juncea, and Anthyllis cytisoides) and for comparison (Lolium perenne) seedlings in a heavy-metal-contaminated, semiarid soil were affected by the application of composted sugar beet waste. We also investigated whether there were relation between AMF diversity and metal concentration (Al, Cd, Cu, Fe, Mn, Pb and Zn) and total P in shoot as well as some soil parameters (total organic carbon and total N) when the SB waste was added to the soil. We analyzed a portion of approximately 795 base pairs of the small-subunit (SSU) rRNA gene by nested PCR, cloning, sequencing, and phylogenetic analyses. Twelve different AMF sequence types were distinguished: seven of these belonged to Glomus group A, one to Glomus group B, one to Diversispora, one to Archaeospora, and two to Paraglomus. The AM fungal populations colonizing roots in a heavy-metal-polluted soil were quite dependent on the host plant, the highest diversity values being obtained in authochtonous plants recognized as metallophytes, such as P. bituminosa, and in an allochtonous, invasive species (L. perenne). No significant correlation was found between AMF diversity and plant metal concentration and soil parameters. Excepting P. bituminosa, when sugar beet waste was added to soil, the populations of AM fungi in roots increased and the shoot metal concentrations decreased in all host plant species studied. Therefore, the addition of sugar beet waste can be considered a good strategy for the remediation and/or phytostabilization of mine tailing sites.  相似文献

The influence of inoculation of olive trees with arbuscular mycorrhizal (AM) fungi, Glomus (G) intraradices, on microbial communities and sugar concentrations, were examined in rhizosphere of olive trees (Olea europaea L.). Analyses of phospholipid and neutral lipid fatty acids (PLFA and NLFA, respectively) were then used to detect changes in microbial community structure in response to inoculation of plantlets with G. intraradices.Microscopic observations studies revealed that the extraradical mycelium of the fungus showed formation of branched absorbing structures (BAS) in rhizosphere of olive tree. Root colonization with the AM fungi G. intraradices induced significant changes in the bacterial community structure of olive tree rhizosphere compared to non-mycorrhizal plants. The largest proportional increase was found for the fatty acid 10Me18:0, which indicated an increase in the number of actinomycetes in mycorrhizal rhizosphere soil, whereas the PLFAs i15:0, a15:0, i16:0, 16:1ω7 and cy17:0 which were used as indicators of bacteria decreased in mycorrhizal treatment compared to non-mycorrhizal control treatment. A highest concentration of glucose and trehalose and a lowest concentration of fructose, galactose, sucrose, raffinose and mannitol were detected in mycorrhizal rhizosphere soil. This mycorrhizal effect on rhizosphere communities may be a consequence of changes in characteristics in the environment close to mycorrhizal roots.  相似文献

The aim of this study was to evaluate the diversity and distribution of arbuscular mycorrhizal (AM) fungi in the salt affected soils of Varanasi district. Saccharum munja Roxb. was predominate among all the vegetation and shown strong AM fungal colonisation. Soil characteristics revealed their neutral to alkali nature and habituated six genera and eight different species of AM fungi i.e. Acaulospora (1), Cetraspora (1), Entrophospora (1), Funneliformis (2- F. geosporum and F. mosseae), Glomus (1), Rhizolglomus (2- R. intraradices and R. fasciculatum) in the rhizosphere of native salt tolerant vegetation. Rhizolglomus fasciculatum was the most widely distributed species under strongly alkaline condition (52.96%) while Funneliformis mosseae was well distributed under slightly (22.99%) to moderately alkaline condition (35.78%). Soil alkalinity was observed to be detrimental for AMF but members of family Glomeraceae registered their presence more frequently (~65%) with higher spore density showing strong adaptation which might be helpful in restoration of moderately to strongly alkaline disturbed eco-region.  相似文献

Plant growth-promoting rhizobacteria and arbuscular mycorrhizal (AM) fungi represent two main groups of beneficial microorganisms of the rhizosphere. The role of different strains of Azospirillum on AM fungi development was evaluated by measuring the percentage of AM colonisation of the root system in durum wheat and maize plants, grown under both greenhouse and field conditions. The effect of wild-type Azospirillum brasilense strain Sp245 and genetically modified (GM) derivatives overproducing indole-3-acetic acid was assessed at greenhouse level in (1) three different cultivars of durum wheat, in the presence of indigenous AM fungi and (2) maize plants artificially inoculated with Glomus mosseae and Glomus macrocarpum. In addition, the establishment of natural AM fungal symbiosis was evaluated using Azospirillum lipoferum CRT1 in maize plants at field level. Despite the stimulatory effect of the different Azospirillum inocula on root growth, no significant differences in AM colonisation were found, independently of the AM fungus involved, either in wheat or in maize plants. Similarly, GM A. brasilense, which strongly stimulates root development, did not affect AM formation. Although these results were obtained in conditions in which the mycorrhization rate was moderate (15–30%), overall considered they indicate that the use of wild-type or GM Azospirillum phytostimulators does not alter mycorrhization.  相似文献
