The time course of cannabinoid accumulation in the leaves of individual plants of three Cannabis accessions was determined by gas-chromatographic analysis in greenhouse-grown plants. The total amounts and the concentration ratios of CBD, THC and CBG were determined; two accessions (an experimental hybrid, (21R × 15R) × NL, and plants from a seized seed lot) were found chemotypically uniform, with all plants belonging to chemotpe II (mixed) and I (high THC) respectively. The Carmagnola accession showed chemotypic heterogeneity, with a majority of plants belonging to chemotype III. The CBD/THC and CBG/CBD ratios were shown to be largely constant in the leaves, since 28 and until 103 days after sowing, and consistent with the ratios determined on mature inflorescences. CBD and THC maximum amounts in the leaves showed a peak in the leaves around 80 days from sowing, and were shown to be simultaneous during the growth period, irrespective of the chemotypes. Callus cultures were obtained from all the five different chemotypes (I, II, III, IV, V), and GC analyses were performed. Independently of the type and amount of cannabinoids in the mother plants, it was confirmed that callus cultures of Cannabis were not able to produce detectable amounts of any cannabinoids.  相似文献

The mechanism that controls the proportion of cannabichromene (CBC), a potential pharmaceutical, in the cannabinoid fraction of Cannabis sativa L. is explored. As with tetrahydrocannabinol (THC) and cannabidiol (CBD), CBC is an enzymatic conversion product of the precursor cannabigerol (CBG). CBC is reported to dominate the cannabinoid fraction of juveniles and to decline with maturation. This ontogeny was confirmed in inbred lines with different mature chemotypes. A consistent CBC presence was found in early leaves from a diverse clone collection, suggesting that CBC synthase is encoded by a fixed locus. Morphological variants possessing a ‘prolonged juvenile chemotype’ (PJC), a substantial proportion of CBC persisting up to maturity, are presented. PJC is associated with a reduced presence of floral bracts, bracteoles, and capitate-stalked trichomes. Genetic factors causing these features were independent of the allelic chemotype locus B that was previously postulated and regulates THC and CBD synthesis and CBG accumulation. In contrast to previously described Cannabis chemotypes, the cannabinoid composition of PJCs showed plasticity in that reduced light levels increased the CBC proportion. The ability of PJC plants to enable the production of pharmaceutical raw material with high CBC purity is demonstrated.  相似文献

Summary In the framework of a comprehensive Dutch research project the feasibility of hemp cultivation for paper pulp production is being investigated. This project also includes a hemp breeding programme which was initiated at CPRO-DLO in 1990. Hemp breeding is primarily aimed at improving bast fibre production since bast fibre is qualitatively superior to woody core fibre for paper pulp production. The progress of the ongoing breeding programme is hampered by the lack of knowledge on the inheritance of agronomic traits.The following traits were examined on 252 parental plants in 1991 and subsequently on their progenies in 1992: fibre content, plant height, date of flowering, stem diameter and cannabinoid contents. For each trait heritabilities were estimated and direct and indirect effects of artificial selection were studied.The heritability of bast fibre content was high and mass selection proved to be an efficient method causing no undesirable influence on other characters. Characters not directly related to bast fibre yield such as date of flowering, plant height and stem diameter were shown to have disadvantages as selection criteria for the improvement of bast fibre yield.The cannabinoids THC and CBD were studied, as the acceptance of hemp cultivation requires a low level. The content of THC, the psychoactive component can be successfully reduced by mass selection, but it is not certain that mass selection is the most efficient method.  相似文献

A genetic factor that blocks the cannabinoid biosynthesis in Cannabis sativa has been investigated. Crosses between cannabinoid-free material and high content, pharmaceutical clones were performed. F1s were uniform and had cannabinoid contents much lower than the mean parental value. Inbred F2 progenies segregated into discrete groups: a cannabinoid-free chemotype, a chemotype with relatively low cannabinoid content and one with relatively high content, in a monogenic 1:2:1 ratio. In our model the cannabinoid knockout factor is indicated as a recessive allele o, situated at locus O, which segregates independently from previously presented chemotype loci. The genotype o/o underlies the cannabinoid-free chemotype, O/o is expressed as an intermediate, low content chemotype, and O/O is the genotype of the high content chemotype. The data suggests that locus O governs a reaction in the pathway towards the phenolic cannabinoid precursors. The composition of terpenoids and various other compound classes of cannabinoid-free segregants remains unaffected. Backcrossing produced cannabinoid-free homologues of pharmaceutical production clones with potential applications in pharmacological research. A new variant of the previously presented allele ‘B 0’, that almost completely obstructs the conversion of CBG into CBD, was also selected from the source population of the cannabinoid knockout factor.  相似文献

Summary The inheritance of resistance to coffee berry disease (CBD) has been studied by applying a preselection test to F2 progenies of a half diallel cross between 11 coffee varieties with different degrees of resistance and to sets of parental, F1, F2, B11 and B12 generations of crosses between resistant and susceptible varieties. True resistance to CBD appears to be controlled by major genes on three different loci. The highly resistant variety Rume Sudan carries the dominant R- and the recessive K-genes. The non-allelic interaction between these two genes is of a duplicate nature. The R-locus has multiple alleles with R 1R1alleles present in Rume Sudan and the somewhat less effective R 2R2alleles in a variety like Pretoria, which also has the K-gene. The moderately resistant variety K7 carries only the recessive K-gene. The arabica-like variety Hibrido de Timor (a natural interspecific arabica x robusta hybrid) carries one gene for CBD resistance on the T-locus with intermediate gene action. It probably inherited this gene from its robusta parent. There is circumstantial evidence that the resistance to CBD is of a stable nature, but it is advisable to accumulate in one genotype as many resistance genes as possible by combining in the breeding programme the resistance of Rume Sudan with that of Hibrido de Timor.  相似文献

Summary Crossability and cytology were examined in F1, F2, B1 and hybridsplants of F1 hybrids of Brassica campestris and three wild relatives of B. oleracea, B. bourgeaui, B. cretica and B. montana, respectively. The F2 plants were obtained after self-and open pollination of the F1 hybrids. The B1 and hybrid plants were produced after the F1 hybrids backcrosses with B. campestris and crossed with B. napus, respectively. After crossing the F1 hybrids, many seeds of the F2, B1 and hybrid plants were harvested. Multivalent formation was high in the chromsome configuration for the PMCs of F2, B1 and hybrid plants, suggesting that crossing over might occur between them. Many different types of aneuploids were obtained in the progenies of the F2, B1 and hybrid plants. It is suggested that different types of normal egg cells may be produced by one-by-one or little-by-little chromosome addition. The possibility is discussed of gene transfer from B. bourgeaui, B. cretica and B. montana, to cultivated plants, B. campestris and B. napus.  相似文献

Summary Pollen grain embryogenesis in anther cultures of Brassica juncea cv. PR-45 was considerably enhanced by treating the donor plants with 4 mll-1 (v/v) of ethrel or delayed sowing of the donor plants, the latter treatment being superior. The anthers derived from plants sown about two months after the normal sowing period showed 18% androgenesis as compared to 3.5% in the control.Pollen grain embryos normally showed very poor germination (10%) on B5 or B5 containing GA3. However, ABA or cold treatment promoted normal germination of these embryos. Exposure of the embryos to 4°C for 6 days, which proved to be the best treatment, induced 66% germination of the embryos.  相似文献

Summary The first backcross and F2 progenies from triploid F1 and tetraploid F1 hybrids between B. napus and 2x and 4x B. oleracea ssp. capitata (cabbage) were studied for their general morphology, resistance to race 2 of the clubroot pathogen, chromosome number and meiotic chromosome behavior. No linkage was apparent between resistance and the major morphological characters. Unreduced gametes played a large part in the successful formation of seed of the B1 and F2 progeny. B1 plants with low chromosome numbers were selected for use in recurrent backcrosses. The potential use of anther culture to extract gametic progenies from resistant B1 and F2 plants with higher chromosome numbers was suggested. The presence of homoeologous pairing observed in all the plants is considered advantageous for selecting suitable progeny in later generations.  相似文献

The genetics of resistance to Phomopsis stem blight caused by Diaporthe toxica Will., Highet, Gams & Sivasith. in narrow-leafed lupin (Lupinus angustifolius L.) was studied in crosses between resistant cv. Merrit, very resistant breeding line 75A:258 and susceptible cv. Unicrop. A non-destructive glasshouse infection test was developed to assess resistance in the F1, F2, selected F2-derived F3 (F2:3) families, and in selfed parent plants. The F1 of Unicrop × 75A:258 (and reciprocal cross) was very resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible), which suggested the presence of a single dominant allele for resistance in 75A:258. In Merrit × Unicrop (and reciprocal), the F1 was moderately resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible). Thus Merrit appeared to carry an incompletely dominant resistance allele for resistance. The F1 of Merrit × 75A:258 (and reciprocal) was very resistant and the F2 segregated in a ratio of 15:1 (resistant: susceptible), which supported the existence of independently segregating resistance alleles for resistance in 75A:258 and Merrit. Alleles at loci for early flowering (Ku) and speckled seeds (for which we propose the symbol Spk) segregated normally and independently of the resistance alleles. Resistant F2 plants gave rise to uniformly resistant or segregating F2:3 families, whereas susceptible F2 plants gave rise only to susceptible F2:3 families. However, the variation in resistance in the F2 and some F2:3 families of crosses involving 75A:258, from moderately to extremely resistant, was greater than that expected by chance or environmental variation. We propose the symbols Phr1 to describe the dominant resistance allele in 75A:258, and Phr2 for the incompletely dominant resistance allele in Merrit. Phr1 appears to be epistatic to Phr2, and expression of Phr1 may be altered by independently segregating modifier allele(s). Summary Life Cycle Assessment (LCA) was used to assess the environmental impacts of field production of fibre hemp and seven other crops in France. The production of 1 ha of hemp yielded a eutrophication potential of 20.5 kg PO4-equivalents, a global warming potential of 2330 kg CO2-equivalents, an acidification potential of 9.8 kg SO2-equivalents, a terrestrial ecotoxicity potential of 2.3 kg 1,4-dichlorobenzene-equivalents, an energy use of 11.4 GJ, and a land use of 1.02 ha.year. A comparison of hemp (low impacts), wheat (intermediate impacts) and sugar beet (high impacts) revealed that the crops were similar for the relative contributions of emitted substances and resources used to impacts, and for the relative contribution of processes to impacts. A reduction of the impacts of hemp production should focus on eutrophication, and consider the reduction of climate change, acidification and energy use as secondary objectives. Given this objective, the overall environmental effect of the substitution of mineral fertiliser by pig slurry is negative. The introduction of reduced tillage is of interest, as it decreases energy use, acidification and climate change. Measures leading to a reduction in NO3 leaching are highly interesting, as they strongly decrease eutrophication. Implications for hemp breeding are discussed.  相似文献

Summary Fibre formation in hemp (Cannabis sativa L.) is a dynamic process. In order to follow this process, the chemical composition of the hemp stem was studied during plant development using the acid and neutral detergent fibre and lignin methods. Additionally, near infra-red spectroscopy was carried out. To predict the chemical composition of the stem samples partial least square (PLS) analysis was carried out. The developed PLS models can predict the level of %ADF and %NDF in the hemp bast and core samples. For %ADL, the models are only suited for either core or bast tissues and can not be used for both tissues simultaneously.  相似文献

Summary The somatic karyotype and meiotic chromosome behavior were studied in an 18-chromosome B1 plant derived from backcrossing a triploid (Brassica napus x B. oleracea ssp. capitata) F1 hybrid to cabbage. It is considered that cabbage chromosomes no. 1 and no. 7 were substituted by two shorter B. napus chromosomes. Meiotic disturbances were more apparent during the late stages of second division. Seed fertility of this plant was largely restored in the second backcrosses with both cabbage and broccoli. 18-chromosome B2 plants resistant to race 2 of Plasmodiophora brassicae were recovered among the progenies.Contribution no. The cytogenetic study was investigated in the intergeneric F1 hybrid, F2and backcross progenies (BC1). The plants used were Brassica juncea(2n=36) and Diplotaxis virgata(2n=18). Three intergeneric F1 hybrids between two species were produced through ovary culture. They showed 36 chromosomes. It might consist one genome of B. juncea and two genomes of D. virgata. The morphology of the leaves resembled that of B. juncea. The color of the petals was yellow that was like in D. virgata. The size of the petal was similar to that of B. juncea. The mean pollen fertility was15.3% and the chromosome associations in the first meiotic division were(0–1)IV+(0–2)III+(8–12)II+(12–20)I. Many F2 and BC1seeds were harvested after open pollination and backcross of the F1 hybrids withB. juncea, respectively. The F2seedlings showed different chromosome constitutions and the range was from 28 to54 chromosomes. Most seedlings had 38chromosomes followed by 36, 40 and 54. The BC1 seedlings also showed different chromosome constitutions and the range was from 29 to 62. Most seedlings had both 40and 54 chromosomes followed by 36, 46 and52. In the first meiotic division of F2 and BC1 plants, a high frequency of bivalent associations was observed in all the various kinds of somatic chromosomes. Many F3 and BC2 seeds were obtained by self-pollination and open pollination of both F2 and BC1 plants, and by backcrossing both F2 and BC1plants with B. juncea, respectively,especially, three type progeny with 36, 40or 54 chromosomes. The somatic chromosomes of the F3 and BC2 plants were further investigated. The bridge plants between B. juncea and D. virgata with 36 chromosomes may be utilized for breeding of other Brassica crops as well as B. juncea. Summary For RFLP mapping of R-genes, determining resistance to specific races of Phytophthora infestans in tetraploid potato, it is necessary to develop well segregating populations at the 2x level. During mapping studies, evidence was obtained that more genetic factor(s) are involved in the expression of R-genes than conventionally believed. Two experiments are described in which such an additional genetic factor was suppressing or enhancing the expression of unknown R nand R ifactors. R nand R iappeared to be present in the investigated plant material, containing R4 and R10, or in one of the susceptible crossing parents. In a third experiment, the expression and the segregation of the well known R1 gene was influenced by an additional genetic factor. In that case there were indications for a dominant suppressor. This was established by the selection of susceptible plants carrying a RFLP allele of probe GP21 closely linked to R1. In three of the four F1 populations, resulting from crosses between such susceptible plants and susceptible tester plants, resistnat progenies were found. The resistance appeared to be R1-specific. This clearly indicates that in three of the four investigated susceptible plants, the R1 gene was still present but not expressed.  相似文献

Summary Dominant genes An 1 for pink flower colour and Br 1 and Br 2 controlling branching of the canes in Rubus coreanus are described. In an F1 progeny and in first backcrosses to red raspberry, some seedlings inherited the ability of R. coreanus to form up to three accessory buds per node. Polygenic systems controlling spine number and spine size are described, number and size being positively correlated. It is postulated that the greater size of spines of h (glabrous-caned) plants is due to linkage of a block of size-controlling genes with the H locus. The greater number of spines of H (hairy-caned) plants is attributed to a pleiotropic effect of the H allele. A new type of dwarf, cauli-flower, which occurred in the F1 and some first backcross progenies, is described.  相似文献

Summary Cytoplasmic male sterility (cms) is described in the F1 hybrids Ribes × carrierei (R. glutinosum albidum × R. nigrum) and R. sanguineum × R. nigrum. In backcrosses to R. nigrum, progenies with R. glutinosum cytoplasm were either all male sterile, or segregated for full male fertility (F) and complete (S) and partial (I) male sterility. Ratios of F:I+S suggested that two linked genes controlled cms, F plants being dominant for one (Rf 1) and recessive for the other (Rf 2).Segregation for cms in relation to three linded genes, Ce (resistance to the gall mite, Cecidophyopsis ribes), Sph 3(resistance to American gooseberry mildew, Sphaerotheca mors-uvae) and Lf 1(one of two dominant additive genes controlling early season leafing out) indicated that Rf 1and Rf 2were in this linkage group. Crossover values of 0.36 for Ce-Lf 1, and 0.15 for Lf 1-Sph 3were estimated from the relative mean differences in season of leafing out between seedlings dominant and recessive for Ce and Sph 3.It is suggested that competitive disadvantage of lf 1-carrying gametes and/or zygotes at low temperatures may be implicated in the almost invariable deficit of plants dominant for the closely linked mildew resistance allele Sph 3. Summary Internal anthocyanin pigmentation (IP) in otherwise normally green cabbage occurs in a number of Oregon State University breeding lines. Extracted pigment, tested for spectral absorption and for color reactions with lead acetate and aluminium chloride, was similar but not necessarily identical to pigment extracted from red cabbage cultivar Redman. When IP line R52 was crossed with normal green line C70, the F1, F2 and backcross progenies indicated that IP at the intensity found in R52 was determined by a single factor in homozygous condition, with intermediate levels of IP expressed by the heterozygous genotypes. Modifying factors also appear to influence the level of IP. In the cross R52 (IP)×R51 (normal green), expression of IP in the F1 was much reduced. The F2 failed to fit the expected 3 IP: 1 green ratio due to an excess of green plants, but instead, closely fit a 9:7 ratio. This may have resulted from incomplete expression of IP because of modifiers, rather than from the effects of a second major gene. Summary Fordhook Zucchini, a squash (Cucurbita pepo) cultivar having intensely colored fruits conditioned by genotype D/D L-1/L-1 L-1/L-2, was crossed with Beirut, a squash cultivar having lightly colored fruits. Beirut was also crossed with Vegetable Spaghetti, a cultivar having lightly colored fruits conditioned by genotype d/d l-1/l-1 l-2/l-2, and with tester stocks of genotypes d/d L-1/L-1 l-2/l-2 and d/d l-1/l-1 L-2/L-2. The results indicated that Beirut, like Vegetable Spaghetti, possesses genotype d/d l-1/l-1 l-2/l-2. However, no F2- or backcross-generation segregates possessing the Light type 1 phenotype, conditioned by recombinant genotype d/d L-1/-l-2/l-2 and heretofore observed when Fordhook Zucchini was crossed with Vegetable Spaghetti, were recovered when Fordhook Zucchini was crossed with Beirut. The results of crossing Beirut with Vegetable Spaghetti and with the tester stocks indicated that both Beirut and Fordhook Zucchini carry a recessive gene, designated pl, which results in Plain light fruit coloration in plants of d/d L-1/- l-2/l-2 genotype. Beirut possesses genotype d/d l-1/l-1 l-2/l-2 pl/pl, Fordhook Zucchini D/D L-1/L-1 L-2/L-2 pl/pl, and Vegetable Spaghetti d/d l-1/l-1 l-2/l-2 Pl/Pl.
