A total of 112 named cultivars and advanced breeding lines were assayed through horizontal starch electrophoresis to test the utility of isozyme loci as a means to objectively identify potato cultivars. Both leaf and tuber tissue were sampled in thirteen enzyme systems. Two buffer systems (Histidine-citrate, pH 5.7 and Lithium-borate, pH 8.3) were used to resolve 16 isozyme loci, of which 14 were scorable without progeny testing. A total of 43 scorable allozymes were detected. All cultivars or advanced breeding lines that were the result of hybridization were discriminated by their sum electrophoretic pattern, whereas, sports (i.e., Russet Burbank vs. Burbank) or line selections (i.e., Norgold Russet “M1” vs. “M2”) have patterns that are identical to the original mother clone. The allelic diversity within and among cultivars indicate that horizontal starch electrophoresis offers an objective means to discriminate sexually-derived potato cultivars.  相似文献

Potato cultivars were assessed for their sensitivity to potato tuber necrotic ringspot disease (PTNRD) induced by two North American isolates (Tu619 and Tu660) of the tobacco veinal necrosis strain of potato virus Y (PVYN) and belonging to the tuber necrosis (PVYNTN) sub-group. Several widely grown cultivars (e.g. Norchip and Ranger Russet) were found to be highly sensitive to PTNRD, while others (e.g. Russet Burbank, Russet Norkotah and Shepody) were not. The foliage symptoms induced by these isolates were distinct from those reported to be produced by European isolates of PVYNTN, in that systemic necrotic rings and chlorophyll figures on the leaves were not produced. Instead, the symptoms were similar to those of normal strains of PVYN. The apparent correlation between resistance to the PVYO (common) strain group and cultivar sensitivity to PTNRD is discussed.  相似文献

Seven primer pairs flanking di- and tri-nucleotide repeat sequences, identified from previously sequenced regions of the potato genome, were examined for their potential use in DNA-fingerprinting of thirty-nineSolanum tuberosum subsp. tuberosum cultivars (released between 1861 and 1988) and one diploidS. phureja breeding line. Of the simple sequence repeats (SSRs), the primers for six SSRs amplified DNA sequences within the potato genome between cultivars for a total of 14 bands. The polymerase chain reaction (PCR)based amplification products generated from each primer pair consisted of 1 to 2 bands per cultivar but band variation among cultivars demonstrated up to 4 bands per SSR. A similarity matrix generated from five SSRs was able to distinguish 24 of the 40 cultivars. However, when the potato cultivars were grouped by tuber type (round white-skinned, long white-skinned, russetskinned, red-skinned, and yellow flesh) only five pairs of cultivars remained indistinguishable: Atlantic/Katahdin, Belchip/Wauseon, Red LaSoda/Bliss Triumph, Red Pontiac/Norland, and Burbank/Spunta. Although SSRs did not generate unique fingerprints for all of the North American genotypes examined, the potential to discriminate most cultivars should increase as additional SSRs are identified in potato.  相似文献

Summary Available information has been reviewed on tuber resistance toP. infestans, its inheritance and breeding procedures used to obtain potato cultivars superior in this character. P. infestans is worldwide the most destructive potato pathogen. Tuber resistance is an essential component of potato resistance as this pathogen is often responsible for tuber rot in storage, and infected tubers of susceptible cultivars provide an important way for its overwintering. In Europe many cultivars and advanced breeders selections have been obtained with tuber resistance toP. infestans, but in the last decades no progress is noted in the mean level of this resistance in cultivars. The expression of tuber resistance depends to a large extent on testing conditions and therefore it can be difficult to evaluate. Consequently making progress in breeding is not easy, and also cultivar assessment data from various countries sometimes differ considerably. It is concluded from published data that it should be easier to make progress in breeding potato cultivars with resistant tubers if more attention is paid to combining the various types of resistance which are already known, and if the genetic determination of durable resistance toP. infestans is better understood. Neither founding clones nor major genetic contributors to modern North American potato cultivars have been systematically identified. Available pedigrees, through 12 generations, were used to identify founding clones and major contributing ancestors (MCAs), to outline relationships among these clones, to determine the genetic contribution of MCAs and exotic germplasm to prominent cultivars, and to draw conclusions about the gene base of prominent North American cultivars. Based on 1996 certified seed acreage, 46 cultivars were identified as prominent, of which 44 had published pedigrees. Using this pedigree data, 12 MCAs consisting of four types were identified: land race types including Daber and Sutton’s Flourball; immediate descendants of land race types including Busola, Early Rose, Garnet Chili, Imperator, Richter’s Jubel, and Triumph; early USDA releases including Chippewa, Earlaine, and Katahdin; and one derivative of a GermanSolanum demissum ×S. tuberosum population designated Germ. No. 3895-13. These clones appeared in the pedigrees of from 61 to 100% of the 44 most prominent North American cultivars. Some MCAs appeared numerous times in the pedigrees of the cultivars with the maximum being Busola, which appeared in the pedigree of NorDonna 94 times. Across the 44 cultivars, the genetic contribution of MCAs averaged from 1.4% for Daber to 23.2% for Katahdin. A positive genetic contribution by exotic germplasm was present in 34 of the 44 cultivars. Krantz, Russet Nugget, Conestoga, and Yukon Gold had exotic germplasm contributions of 49.5, 34.1, 32.3, and 31.3%, respectively.  相似文献

Potato constituents were monitored to evaluate their contribution to potato chip color. Five chipping potato varieties: Snowden, AC Ptarmigan, FL 1625, FL 1815 and ND2471-8 were evaluated. Ascorbic acid, sugars, phenolic acids, and amino acids were determined and quantified by High Performance Liquid Chromatography (HPLC) and the color of potato chips was measured by both CIELab Hunter-ColorQuest and Agtron instruments. Composition and chip color varied among the different varieties. AC Ptarmigan and ND2471-8 produced the darkest chip color (on average L*= 49.0, chroma=19.5, and hue angle= 62.9) compared with FL 1815 (L*= 58.4, chroma= 28.3 and hue angle= 75.7). Reducing sugar concentration did not completely explain or predict color quality when it was present in low concentrations (ca. < 60 mg/100g). Other reactants present in the potato slices played an important role in the final color quality of potato chips. Multiple correlation analysis showed negative association of ascorbic acid (r= -0.7), fructose (r= -0.7), a chlorogenic acid isomer (r= -0.7), glucose (r= -0.7) and glutamine (r= -0.5) with potato chip color. Six cultivars, Atlantic, Eramosa, Kanona, Norchip, Onaway and Saginaw Gold and four selections, MS 700-70, MS 700-83 (Spartan Pearl), MS 716-15 and W-855 (Snowden), were analyzed for sucrose, glucose, chip color, specific gravity and yield at 98 and 138 days in 1988 and 1989, t evaluate their performance to Michigan growing conditions. The average yield of these cultivars at the two harvests for 1988 and 1989 was 46.9 and 54.7 t/ha and 43.1 and 52.3 t/ha, respectively. Eramosa and Onaway were the earliest maturing varieties with low specific gravity, high glucose content and processed into dark colored chips. The glucose and sucrose contents of the remaining eight cultivars at each harvest date were low and produced acceptable chips. The specific gravity of Atlantic, MS 700-70, MS 716-15 and W-855 was in the range of 1.079–1.088 and for Norchip, Kanona and Saginaw Gold the range was 1.071–1.076. The date of harvest, variety and year of production had significant effect (p > 0.01) on yield. The specific gravity, glucose content and chip color were significantly influenced by variety and year of production. The correlation between chip colorvs glucose (r= -0.842, p = 0.000) and harvest datevs yield (r= -0.572, p = 0.000) was also significant.  相似文献

Summary Ten cultivars contrasting in chip quality were grown in the field and in the glasshouse to evaluate three different methods for chip quality assessment. Specific gravity was also measured. The glasshouse culture simulated growth of seedlings and the field culture represented growth of advanced breeding lines. Differences between cultivars for chip quality and specific gravity could be established in both environments. Although ranking of the cultivars in both environments was not identical, both good and bad genotypes could be identified. It seems that mild selection for specific gravity and chip quality among glasshouse-grown seedlings can be exercised. If tubers of glasshouse-grown plants are too small to slice chips, Reflocheck Glucose test strips offer a satisfactory alternative to frying chips. Crip in UK  相似文献

Summary The possibilities of selecting individual seedlings and seedling populations for dry matter content and chip colour were examined. Tuber glucose content as measured with Reflocheck Glucose test strips was strongly correlated with the colour of the chips, but Gluko-Test strips were less reliable. Glasshouse and field determinations of both the dry matter content and the glucose content of the seedling populations were closely related. Therefore it should be possible to select populations for both characters on the basis of the population performance in the glasshouse. Individual seedlings could be reliably selected for dry matter content, but such selection should not be rigorous. Individual seedling selection for chip colour is not recommended. Summary Between 1993 and 1998 205 different potato cultivars and 1220 accessions/genotypes of wild and cultivated potato species from the IPK Genebank Gatersleben were evaluated. Parameters interesting for starch isolation and especially for the use of starch were determined. Altogether, there was a higher variability in wild potato species than in cultivated potatoes for all characteristics investigated: dry matter content, starch content, protein content, amylose content and mean particle diameter of starch granules.  相似文献

Summary Twenty seven cultivars of potato were screened for common scab grown in a commercial field in two successive years (1996–97 and 1997–98). Eight cultivars were least susceptible and the others ranged from medium susceptible to very highly susceptible. None of the cultivars was resistant. Microtuber size, media growth regulators, incubation period, as well as bud maturity and endogenous abscisic acid content of microtubers were evaluated for their effects on length of dormant period of Kennebec, Russet Burbank and Superior microtubers. The dormant period of microtubers was found to be cultivar-specific and a significant correlation was observed betweenin vitro andin vivo dormant periods. Smaller microtubers (≤250 mg) had longer dormant periods than did those greater than 250 mg. Dormant periods were unaffected by addition of coumarin or 2-(chloroethyl)-trimethylammoniumchloride and 6-benzylaminopurine to the culture media or incubation period (28 and 56 days). Developmental age had no effect on individual buds ability to break dormancy and elongate. A positive correlation was observed between tissue levels of abscisic acid and microtuber dormant periods.  相似文献

Summary The growth and duration of crop leaf area determines the amount of solar radiation intercepted by the canopy and therefore influences the extent of photosynthesis, evaporation, transpiration and final dry matter yield. The objective of this study was to develop cultivar specific relationships to estimate the daily leaf area index (LAI) for the potato crop (Solanum tuberosum L.) that included the effects of available soil water. The model is divided into three LAI growth stages, the durations of which are partially related to potato heat units (PHU). The LAI in the first stage is estimated from a cultivar specific leaf area-based radiation use efficiency index with a soil water reduction factor. The second stage involves the maintenance of a constant LAI with the duration related to both PHU and a soil water index that can accelerate senescence. The final stage includes a decrease in the LAI from a maximum to zero in response to a cultivar specific PHU accumulation. Model simulations compared favourably with independent LAI measurements obtained with a LI-COR plant canopy analyzer over two seasons.  相似文献
