A kinetic method has been developed for the determination of 1-naphthylacetic acid by means of micellar-stabilized room temperature phosphorescence (MSRTP) using the stopped-flow mixing technique. The main feature of this system is that it diminishes the time required for the deoxygenation of the micellar medium and for the phosphorescence development. Phosphorescence enhancers such thallium(I) nitrate, sodium dodecyl sulfate (SDS), and sodium sulfite were optimized to obtain maximum sensitivity. The pH was also optimized as it strongly affects the luminescent properties of 1-naphthylacetic acid. A pH of 6.6 was selected as adequate for the phosphorescence development. The kinetic curve of 1-naphthylacetic acid phosphorescence was scanned at lambda(ex) = 278 nm and lambda(em) = 490 nm, and the maximum rate of phosphorescence was taken as the analytical signal. This was obtained by calculating the maximum slope of the curve in an interval of 3.6 s as it provided a good noise-to-signal ratio. This method permitted the determination of 1-naphthylacetic acid throughout a concentration range of 100-1800 ng mL(-1) with high precision (relative standard error = 0.91% and relative standard deviation = 2.30%; 1-naphthylacetic acid concentration = 800 ng mL(-1)). According to the Clayton criterion, the detection limit was 45 ng mL(-1). The same limit resulted in 39.3 ng mL(-1) when the error propagation theory was applied. The applicability of the method was successfully demonstrated by determining 1-naphthylacetic acid in different kind of samples, such as phytosanitary products, soils, pears, and apples. Recovery values not significantly different from the nominal content or the spiked amount were found for these determinations.  相似文献

Valuable information about possible types of linkages, reaction mechanisms, and sequences for oxidative coupling of phenolic compounds in planta is available from in vitro model systems. Ferulate oligomers were generated in a system using ethyl ferulate, peroxidase, and hydrogen peroxide under various conditions. A molar ferulate/H2O2 ratio of 1:1, an ethanol level of 30% in an aqueous sodium phosphate buffer (pH 6.0), and a reaction time of 10 min were considered to be ideal to produce maximal proportions of ferulate trimers and tetramers from ethyl ferulate as starting material. The dominant trimer and tetramer were each isolated from the reaction mixture and identified as 8-O-4/8-5(cyclic)-dehydrotriferulic acid triethyl ester and 8-5(cyclic)/4-O-5/8-5(cyclic)-dehydrotetraferulic acid tetraethyl ester. The structure of the 8-O-4/8-5(cyclic)-dehydrotriferulic acid triethyl ester revealed that a third ferulate unit is bound to a preformed 8-O-4-diferulate dimer, a surprising reaction sequence considering the dominance of 8-5-coupled dimers among dehydrodiferulates in H2O2/peroxidase-based model reactions. As 4-O-5-coupling is not favored in the dimerization process of ferulates, the main tetramer isolated in this study is probably formed by 4-O-5-coupling of two preformed 8-5(cyclic)-diferulates, a logical step in analogy with reactions occurring in lignin biosynthesis.  相似文献

A total of 82 grapevine genotypes were sampled from several areas of the Italian region of Sicily where vineyards are widely spread. The grapevines were characterized using six microsatellite markers (VVS2, VVMD5, VVMD7, VVMD27, VrZAG62 and VrZAG79) to evaluate genetic diversity. Thirty-seven of the 82 cultivars sampled had their names quoted in historical and literary sources, while 45 cultivars from old vineyards did not have their names reported in ancient literature. According to their genetic profiles at SSR loci, 70 different cultivars were found, while interesting cases of synonymies (Regina and Moscato bianco, Alicante and Dolcetta or among different clones of Zibibbo and Catarratto) and cases of homonymy (Frappato and Nerello Mascalese) were discovered. Several genetic parameters were calculated to assess the efficacy of the loci chosen in this work. Pairwise genetic distances between all cultivars were calculated. A dendrogram representing the genetic similarities among cultivars was depicted using the UPGMA method to investigate their relationships, explaining them from a historical point of view. The cluster distribution of cultivars clearly does not reflect their current geographical distribution, suggesting successive introductions of cultivars in Sicily from different areas of origin.  相似文献

Hydroxycinnamic acids are natural antioxidants found in fruits, vegetables, and cereals. In this study, the antioxidant activity of various types of hydroxycinnamoyl glycoside esters that mimic the structure of polymeric carbohydrates was studied in different model systems prone to oxidation, namely, liposomes and emulsions. In addition, radical scavenging activity against the stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was tested. It was found that the esterification in the primary hydroxyl group of the glycoside resulted in the improved radical scavenging activity of both sinapoyl and feruloyl glycosides compared to conjugation to the secondary hydroxyl group. Increased activity was also observed, particularly in the case of feruloyl glucosides in inhibiting the oxidation of liposomes emulsions. The results showed that sinapic and ferulic acid glycoside esters were as effective or more efficient antioxidants than their free forms. In conclusion, the strength of their antioxidant effect depends on the nature of conjugation.  相似文献

Phospholipid fatty acid (PLFA) and total soil fatty acid methyl esters (TSFAME), both lipid-based approaches used to characterize microbial communities, were compared with respect to their reliable detection limits, extraction precision, and ability to differentiate agricultural soils. Two sets of soil samples, representing seven crop types from California's Central Valley, were extracted using PLFA and TSFAME procedures. PLFA analysis required 10 times more soil than TSFAME analysis to obtain a reliable microbial community fingerprint and total fatty acid content measurement. Although less soil initially was extracted with TSFAME, total fatty acid (FA) content g−1 soil (DW) was more than 7-fold higher in TSFAME- versus PLFA-extracted samples. Sample extraction precision was much lower with TSFAME analysis than PLFA analysis, with the coefficient of variation between replicates being as much as 4-fold higher with TSFAME extraction. There were significant differences between PLFA- and TSFAME-extracted samples when biomarker pool sizes (mol% values) for bacteria, actinomycetes, and fungi were compared. Correspondence analysis (CA) of PLFA and TSFAME samples indicated that extraction method had the greatest influence on sample FA composition. Soil type also influenced FA composition, with samples grouping by soil type with both extraction methods. However, separate CAs of PLFA- and TSFAME extracted samples depicted strong differences in underlying sample groupings. Recommendations for the selection of extraction method are presented and discussed.  相似文献

The effect of riboflavin or fatty acid methyl esters on cholesterol photooxidation was studied. Samples containing cholesterol, either alone or in combination with riboflavin or fatty acid methyl esters, were illuminated at 25 degrees C in an incubator for 28 days. The various cholesterol oxidation products (COPs) and cholesterol were analyzed by gas chromatography-mass spectrometry (GC-MS), and riboflavin was determined by HPLC. Results showed that the presence of riboflavin or fatty acid methyl esters facilitated production of COPs and degradation of cholesterol, and the degradation fits a first-order model. The COPs formed during light storage included 7 alpha-OH, 7 beta-OH, 7-keto, 3,5-cholestadien-7-one, 5,6alpha-EP, and 5,6beta-EP. The addition of riboflavin caused formation of 3,5-cholestadien-7-one through dehydration of 7-keto, whereas in the presence of docosahexaenoic acid methyl ester, the formation of 5,6alpha-EP or 5,6beta-EP was favored. Riboflavin was more effective for generation of COPs than fatty acid methyl esters.  相似文献

Ethyl esters (FAEE) and trideuterium-labeled methyl esters (d3-FAME) of fatty acids were prepared and investigated regarding their suitability as internal standards (IS) for the determination of fatty acids as methyl esters (FAME). On CP-Sil 88, ethyl esters of odd-numbered fatty acids eluted approximately 0.5 min after the respective FAME, and only coelutions with minor FAME were observed. Depending on the problem, one or even many FAEE can be added as IS for the quantification of FAME by both GC-FID and GC-MS. By contrast, d3-FAME coeluted with FAME on the polar GC column, and the use of the former as IS requires application of GC-MS. In the SIM mode, m/z 77 and 90 are suggested for d3-methyl esters of saturated fatty acids, whereas m/z 88 and 101 are recommended for ethyl esters of saturated fatty acids. These m/z values give either no or very low response for FAME and can thus be used for the analysis of FAME in food by GC-MS in the SIM mode. Fatty acids in sunflower oil and mozzarella cheese were quantified using five saturated FAEE as IS. Gravimetric studies showed that the transesterification procedure could be carried out without of loss of fatty acids. GC-EI/MS full scan analysis was suitable for the quantitative determination of all unsaturated fatty acids in both food samples, whereas GC-EI/MS in the SIM mode was particularly valuable for quantifying minor fatty acids. The novel GC-EI/MS/SIM method using fatty acid ethyl esters as internal standards can be used to quantify individual fatty acids only, that is, without determination of all fatty acids (the common 100% method), although this is present. This was demonstrated by the exclusive quantification of selected fatty acids including methyl-branched fatty acids, erucic acid (18:1n-9trans), and polyunsaturated fatty acids in cod liver oil and goat's milk fat.  相似文献

The mass and infrared spectra of the methyl esters of 9 chlorophenoxy acid herbicides are presented. Ultraviolet data are discussed and proton magnetic resonance spectra are tabulated. Because of the sensitivity of the technique, the mass spectra are most useful for the identification of those compounds in residues, especially by combined gas chromatography-mass spectrometry. The pure herbicides used for the recording of the spectra were obtained by synthesis and recrystallization.  相似文献

Approaches for the capillary gas chromatographic (GC) based analysis of intact plant stanyl esters in enriched foods were developed. Reference compounds were synthesized by enzyme-catalyzed transesterifications. Their identities were confirmed by means of mass spectrometry. Using a medium polar trifluoropropylmethyl polysiloxane stationary phase, long-chain plant stanyl esters could be separated according to their stanol moieties and their fatty acid chains. Thermal degradation during GC analysis was compensated by determining response factors; calibrations were performed for ten individual plant stanyl esters. For the analysis of low-fat products (skimmed milk drinking yogurts), the GC separation was combined with a "fast extraction" under acidic conditions. For fat-based foods (margarines), online coupled LC-GC offered an elegant and efficient way to avoid time-consuming sample preparation steps. The robust and rapid methods allow conclusions on both, the stanol profiles and the fatty acid moieties, and thus provide a basis for the authentication of this type of functional food ingredients.  相似文献

生物柴油可作为改善低硫柴油润滑性能的天然添加剂。该文将豆蔻酸甲酯(C14:0)、棕榈酸甲酯(C16:0)、硬脂酸甲酯(C18:0)、油酸甲酯(C18:1)、亚油酸甲酯(C18:2)、亚麻酸甲酯(C18:3)、蓖麻醇酸甲酯(C18:1 OH)及蓖麻油甲酯和餐饮废油甲酯按照0.5%、1.0%、1.5%和3.0%的体积分数添加到低硫柴油中,在高频往复试验机(high-frequency reciprocating rig,HFRR)上进行润滑性能测试,探究脂肪酸甲酯的碳链长度、不饱和度及含羟基等结构特征对润滑性能的影响。结果表明,长碳链脂肪酸甲酯一般比短链润滑效果好;碳链长度为十八的脂肪酸酯中,不饱和程度即碳碳双键数目越高则润滑性能越好;而在相同碳链长度和不饱和度条件下,含羟基的蓖麻醇酸甲酯的润滑改善效果优于油酸甲酯。由多种脂肪酸酯构成的混合物生物柴油的润滑性能要优于某单一的纯脂肪酸甲酯。在低硫柴油中,当某饱和脂肪酸甲酯的体积分数比例达3.0%时,或不饱和酯的体积分数达到1.5%时,或生物柴油的体积分数达1.0%时,可使低硫柴油的润滑性能指标满足相关标准。研究脂肪酸甲酯的各种结构特征对其润滑性能的影响及作用机制,有助于筛选合适的生物柴油组分及其添加浓度作为低硫柴油的润滑添加剂。  相似文献

The esterified fatty acid composition of cheese (YC) from yak ( Bos grunniens), reared in the highlands of the Nepalese Himalayas, was studied using capillary gas-liquid chromatography and compared with that of dairy cow Cheddar cheese (DC) purchased in a local market. The YC was collected from Dolakha, Nepal. The YC had a lower (P<0.001) myristic acid (C14:0; 6.7 vs 10.3%, YC vs DC, respectively) and palmitic acid content (C16:0; 23.3 vs 29.2%, YC vs DC, respectively) compared to DC. The YC had a lower (P<0.01) total medium-chain saturated fatty acids (C10:0-C16:0) content compared to DC (36.7 vs 47.3%, YC vs DC, respectively). On the other hand, the YC had a 24.8% higher (P<0.01) level of total long-chain saturated fatty acids (C17:0-C26:0) and a 3.2 times higher (P<0.001) content of total n-3 PUFA than DC. The ratio of n-3 PUFA to n-6 PUFA in YC was 0.87 compared to 0.20 in DC. YC had a 2.8 times higher (P<0.001) total trans-18:1 (9.18 vs 3.31%, YC vs DC, respectively) content. The percentage of vaccenic acid ( trans-11-C18:1) in YC was 4.6 times higher (6.23 vs 1.35% of total fatty acids, YC vs DC, respectively) than in DC. Vaccenic acid constituted 67.9% of total trans-C18:1 in YC. The Delta9-desaturase index for YC was lower than that of DC. The total conjugated linoleic acid (CLA) content in YC was 2.3% of total fatty acids compared to 0.57% in DC. The cis-9, trans-11 CLA isomer in YC constituted 88.5% of the total CLA. The results suggest that cheese from yak, grazed on Himalayan alpine pastures, may have a more healthful fatty acid composition compared to cheese manufactured from dairy cattle fed grain-based diets.  相似文献

An assay-guided isolation gave three antioxidants including two newly identified compounds from the rhizomes of Alpinia speciosa, which is used as an important plant in the food culture of the Okinawa area of Japan. Spectroscopic analysis of the two new compounds revealed them to be new glucoside esters of ferulic acid. The antioxidant activity of the esters was measured using two different methods. Both compounds showed greater activity than that of Trolox in the TLC method; however, one of the compounds showed weaker inhibitory activity than that of Trolox and epicatechin against AMVN-induced methyl linoleate oxidation.  相似文献

Two brands of stainless steel penicylinders, S&L Metal Products and Fisher Scientific, were simultaneously tested to determine if they provide comparable results when used in the AOAC use-dilution method of disinfectant testing. Results showed consistently more positive tubes for the Fisher brand of penicylinders than for the S&L, regardless of the surface finish of the test cylinders.  相似文献

The objective was to evaluate alternatives to the peroxide value method of choice in the dairy industry, the method issued by the International Dairy Federation. Furthermore, the study evaluated the feasibility of alternative solvents for extracting lipids and subsequent peroxide value determinations. Packaged cheeses were stored under illuminated display at 4 degrees C to obtain samples with various peroxide contents but with uniform gross composition. The hydroperoxide contents were measured during 3 weeks of storage by applying two lipid extraction methods, Folch and Bureau of Dairy Industry (BDI) extractions, and three different hydroperoxide extraction solutions [chloroform/methanol (7:3, v/v), hexane/2-propanol/methanol (5:7:2, v/v/v), and methanol/decanol/hexane (3:2:1, v/v/v)], prior to standard colorimetric measurements. Extraction yields of fat from Havarti cheeses using the Folch and BDI extraction methods were approximately 109 and 61%, respectively, of the yields obtained by the International Dairy Federation gravimetric reference method. Although differences in fat extraction yields were compensated for, significantly higher peroxide values resulted from the Folch extraction method than from the BDI extraction method. The peroxide values obtained by the three methods were all in the same range, and pronounced linear correlations between peroxide contents determined using the three solutions were noted (r (2) in the range of 0.951-0.983). Peroxide value levels were not significantly different in samples stored in the dark or exposed to light.  相似文献

菜心对邻苯二甲酸酯(PAEs)吸收途径的初步研究   总被引:16,自引:2,他引:16  
采用玻璃室处理和污染土壤覆盖原土壤来控制PAEs来源进行盆栽试验,应用GC/MS联机检测技术初步研究了菜心对PAEs的吸收途径。结果表明:污染土壤处理与污染土壤上覆盖原土壤处理相比,前者菜心茎叶中DBP和DEHP的含量均高于后者,但相差不大,表明菜心茎叶可以吸收污染土壤中挥发出来的DBP和DEHP,而根系吸收运移是菜心茎叶中DBP和DEHP的主要来源途径。玻璃室处理增加了菜心茎叶和根系中DBP的含量,而对DEHP的影响趋势不明显。DBP与DEHP相比,前者更易被菜心根系吸收并向地上部(茎叶)运移,后者主要滞留在根部。  相似文献

通过室内盆栽试验,研究了甜菜与黑麦草、苜蓿、苏丹草分别间作及4种植物各自单作对土壤中邻苯二甲酸二乙酯(DEP)、邻苯二甲酸二丁酯(DBP)、邻苯二甲酸丁基苄基酯(BBP)和邻苯二甲酸(2-乙基己基)二酯(DEHP)4种邻苯二甲酸酯类(PAEs)的植物修复效果。结果表明:与空白对照相比,种植植物的修复效果更好;苜蓿单作与间作都有较好的修复效果,其中甜菜/苜蓿间作PAEs的去除率最高,可达66.48%;植物单作与间作相比,间作的修复效果高于单作,间作增强土壤中过氧化氢酶和磷酸酶的活性,从而促进了根际微生物对PAEs的降解;就单一污染物来说,DBP和DEHP在污染土壤和植物茎叶中的浓度较其他两种污染物高,两者在土壤中的去除率也较高,其中DEHP为最高,均可达50%以上,DBP的去除率也在40%以上;DEHP在植物茎叶中的生物富集系数明显较低,且单作低于间作,而DBP和BBP的生物富集系数较高。可选择苜蓿作为土壤中PAEs修复的一种高效修复植物,植物间作相对于单作有更好的修复效率,也可更高效地利用土地资源,因此可优先选择作为植物修复的一种种植模式。  相似文献
