The study objective was to determine the effects of feeding food enriched in antioxidants and a program of environmental/cognitive enrichment on selected ex vivo assays of inflammatory and immune cells in healthy geriatric Beagle dogs (n=21). Four groups of dogs were tested using a 2 x 2 factorial design. The 2-year longitudinal study included both nutritional (control food or antioxidant-fortified food) and behavioral (normal level or cognitive enrichment) interventions. Behavior enrichment included increased exercise, environmental enrichment, and a series of learning tasks. Phagocytosis of opsonized latex-coated beads by peripheral blood neutrophils was measured by flow cytometry and found to be significantly increased in dogs receiving both dietary antioxidants and cognitive enrichment. Simultaneous stimulation of cells with Con A and suppression with Dex resulted in decreased lymphocyte proliferation in dogs receiving both dietary antioxidants and cognitive enrichment, compared to dogs receiving dietary antioxidants or cognitive enrichment alone. There were no significant differences between the groups of dogs for percentages of CD4 and CD8 T-lymphocyte subpopulations before or after lymphocyte stimulation with Con A. These results support our hypothesis that both dietary antioxidants and behavioral enrichment enhance host defense mechanisms. 相似文献

The aim of the present study was to evaluate the impact of dietary fatty acids (FA) during the weaning period on expression of genes involved in the oxidation and metabolism of FA (peroxisome proliferator-activated receptor (PPAR-), stearoyl-CoA-desaturase (SCD), Δ6-desaturase (D6D), acetyl CoA carboxylase (ACC) and fatty acid synthase (FAS)). Liver samples were obtained from littermates, either on day 28 of age just before weaning, or at day 56 after the 4 weeks ad lib. provision of 5% of either animal fat, fish oil or sunflower oil. In conclusion, genes involved in the regulation of FA conversion (SCD, D6D) were influenced by the n-6 to n-3 ratio, whereas the FA oxidation, as indicated by the expression of PPAR-, was highly likely regulated by the hepatic ratio between mono- and poly-unsaturated FA. Furthermore, weaning and/or age affected the hepatic expression of genes involved in FA synthesis and conversion, but not the expression of PPAR-. 相似文献

Background: Fat is the primary source of the volatiles that determine the characteristic flavors of animal products.Because unsaturated fatty acids(UFAs) contribute to changes in flavor as a result of the oxidation process, a feeding trial was performed to investigate the effects of dietary soybean oil or antioxidants on the fatty acid and volatile profiles of the tail subcutaneous(SF) and perirenal fat tissues(PF) of fattening lambs. Thirty-six Huzhou lambs were assigned to four dietary treatments in a randomized block design. The lambs' diets were supplemented with soybean oil(0 or 3 % of DM) or antioxidants(0 or 0.025 % of DM).Results: Neither soybean oil nor antioxidant supplementation had an effect on lamb growth(P 0.05). In regard to tail SF, soybean oil supplementation increased the 18:2n6t(P 0.05) and the total amount of volatile acids,whereas antioxidant supplementation increased the content of C18:2n6c and C18:3n3(P 0.05) but had no effect on the volatiles profile. In regard to PF, dietary soybean oil supplementation increased the C18:0 content(P 0.01);decreased the C18:1(P = 0.01), C22:1 n9(P 0.01) and total UFA(P = 0.03) contents; and tended to decrease the E-2-octenal(P = 0.08), E, E-2, 4-decadienal(P = 0.10), 2-undecenal(P = 0.14) and ethyl 9-decenoate(P = 0.10) contents.Antioxidant supplementation did not affect either the fatty acid content or the volatiles profile in the PF.Conclusions: Tail SF and PF responded to dietary soybean oil and antioxidant supplementation in different ways. For SF, both soybean oil and antioxidant supplementation increased the levels of unsaturated fatty acids but triggered only a slight change in volatiles. For PF, soybean oil supplementation decreased the levels of unsaturated fatty acids and oxidative volatiles, but supplementation with antioxidants had little effect on PF fatty acids and the volatiles profile. 相似文献

Parecoxib is an inactive pro-drug that is rapidly converted to valdecoxib, a selective cyclooxygenase (COX)-2 inhibitor registered for the management of post-operative pain in humans. Recent studies have suggested that parecoxib has excellent clinical efficacy and safety in veterinary species. The aim of the current study was to assess the pharmacokinetics of parecoxib and valdecoxib after intravenous (i.v.) and intramuscular (i.m.) administration. Seven healthy male Beagle dogs received 2.5 mg/kg parecoxib by either the i.v. or i.m. route in a cross-over design, with the alternative route of administration used 1 week later. The plasma concentrations of both analytes were detected according to a previously validated method using high performance liquid chromatography with fluorescence detection (HPLC-FL). No adverse effects were observed in any animal during the study. For both routes of administration, parecoxib was rapidly and almost completely converted to valdecoxib. The parecoxib/valdecoxib area under the curve (AUC) ratio for both routes of administration was 1.4. The half-life of valdecoxib was about 2 h, which was shorter than reported for humans, although the plasma concentrations following both routes of administration were likely to be effective for analgesia. The absolute bioavailability of parecoxib was 66%. The pharmacokinetic features of parecoxib make it suitable for treatment of acute pain in the canine species. 相似文献

In order to compare the stimulation ability of insulin secretion, we determined changes in plasma glucose and insulin concentrations after intravenous administration of various amino acids and essential fatty acids in clinically healthy adult cats. Plasma glucose concentrations were within the normal ranges after injection of amino acids and fatty acids. Plasma insulin concentrations increased rapidly 2 to 4 min after injection of arginine, then decreased to the basal levels at 20 min in all five cats. Insulin peak responses were significantly greater in arginine injections than in normal saline (P<0.01). Areas under the curve (AUC) of plasma insulin concentrations from 0 to 10 min after injection of arginine were significantly larger than after injection of normal saline (P<0.01) and glucose (P<0.05). Increases in AUC of plasma insulin concentration from 0 to 60 min were observed after injection of arginine, leucine, alanine, and fat emulsion. Arginine had a strong insulinotropic effect, and leucine, alanine, and fatty acids had weak ones. Besides, valine, methionine, taurine and glutamine had no stimulant activity of insulin. Given the risk of glucose toxication and required time for testing, the intravenous arginine tolerance test may be useful for estimation of insulin responses in cats. 相似文献

OBJECTIVE: To determine the effect of dietary n-6 to n-3 fatty acid ratios and alpha-tocopheryl acetate concentration on immune functions andT cell subpopulations in healthy dogs. ANIMALS: Thirty-two 7- to 10-year old female Beagles. PROCEDURE: For 17 weeks, dogs were fed food that contained low (1.4:1) or high (40:1) ratios of n-6 to n-3 fatty acids in combination with 3 concentrations of all rac-alpha-tocopheryl acetate (low, 17 mg/kg of food; medium, 101 mg/kg; high, 447 mg/kg). Dogs were inoculated twice with a keyhole limpet hemocyanin suspension at 13 and 15 weeks. RESULTS: After 12 weeks, dogs consuming low concentrations of alpha-tocopheryl acetate had lower percentages of CD8+ T cells, compared with dogs consuming medium or high alpha-tocopheryl acetate concentrations. Also, dogs consuming low alpha-tocopheryl acetate concentrations had higher CD4+ to CD8+ T cell ratios. On day 4 of week 15, the percentage of CD8+ T cells was highest in dogs fed medium concentrations of alpha-tocopheryl acetate, compared with other dogs; however, the CD4+ to CD8+ T cell ratio was higher only in dogs fed low concentrations of alpha-tocopheryl acetate with high concentrations of n-3 fatty acids. Dogs consuming low concentrations of n-3 fatty acids with medium concentrations of alpha-tocopheryl acetate had the largest delayed-type hypersensitivity (DTH) skin test response. CONCLUSIONS AND CLINICAL RELEVANCE: An optimum amount of dietary alpha-tocopheryl acetate concentration, regardless of the dietary n-6 to n-3 fatty acid ratio, stimulates the CD8+ T cell population. Effects of an optimum amount of dietary alpha-tocopheryl acetate concentration on the DTH response are blunted by dietary n-3 fatty acids. 相似文献

1. This study examined the effect of increasing amounts of dietary polyunsaturated fatty acids on the fatty acid composition in serum and antibody production following a standard vaccination programme in growing turkeys. Turkey poults were fed on 5 diets containing 75g/kg added fat made up of different proportions of palm and soyabean oils, and were vaccinated against Newcastle disease, infectious bronchitis and necrotic enteritis according to a standard vaccination programme. Blood samples were taken before and one week after each vaccination.
2. Fatty acid composition in serum reflected the composition of the diets although arachidonic acid concentration was not changed by dietary fatty acid content. Growth, erythrocyte and leukocyte parameters were not affected by the respective diets.
3. Specific antibody production was related quadratically to serum linoleic and total n‐6 polyunsaturated fatty acid concentrations. No correlation was found with linolenic or arachidonic acids.
4. It is concluded that dietary fatty acid composition can augment the specific anti‐vaccine immune response in turkey poults. 相似文献

OBJECTIVE: To determine effects of dietary n-3 fatty acids from Menhaden fish oil on plasma alpha-tocopherol concentrations in Beagles. ANIMALS: 32 female Beagles. PROCEDURE: For 82 days, dogs were fed diets that contained 1 of 2 ratios of n-6:n-3 fatty acids (40:1 [low n-3] and 1.4:1 [high n-3]) and 1 of 3 concentrations of all-rac-alpha-tocopheryl acetate (low, 17 mg/kg of diet; medium, 101 mg/kg; and high, 447 mg/kg) in a 2 X 3 factorial study. RESULTS: Diets high in n-3 fatty acids significantly increased total content of n-3 fatty acids in plasma (17.0 g/100 g of fatty acids), compared with low n-3 diets (2.02 g/100 g of fatty acids). Mean +/- SEM plasma concentration of cholesterol was significantly lower in dogs consuming high n-3 diets (4.59 +/- 0.48 mmol/L), compared with dogs consuming low n-3 diets (5.71 +/- 0.48 mmol/L). A significant interaction existed between the ratio for n-6 and n-3 fatty acids and amount of alpha-tocopheryl acetate in the diet (plasma alpha-tocopherol concentration expressed on a molar basis), because the plasma concentration of alpha-toco-pherol was higher in dogs consuming low n-3 diets, compared with those consuming high n-3 diets, at the 2 higher amounts of dietary alpha-tocopheryl acetate. Plasma alpha-tocopherol concentration expressed relative to total lipid content did not reveal effects of dietary n-3 fatty acids on concentration of alpha-tocopherol. CONCLUSIONS AND CLINICAL RELEVANCE: Plasma alpha-tocopherol concentration is not dependent on dietary ratio of n-6 and n-3 fatty acids when alpha-tocopherol concentration is expressed relative to the total lipid content of plasma. 相似文献

Recent research in our lab has demonstrated a significant association between the incidence of subclinical mastitis and specific polymorphisms of the CXCR2 gene in Holstein dairy cows. This gene encodes a receptor for interleukin-8 (IL-8), a key regulator of neutrophil migration, killing and survival. Because of the importance of this gene in neutrophil function, we hypothesized that differences in neutrophil killing and survival may exist among the CXCR2 genotypes and potentially contribute to the observed variation in intramammary infections. To test this hypothesis, neutrophils were isolated from cows representing each CXCR2 +777 genotype (GG, GC or CC) and tested for suppression of apoptosis, reactive oxygen species (ROS) generation, glutathione levels, and bactericidal activity. A significant increase in survival was observed in neutrophils from cows with a CC genotype when compared to those with a GG genotype in response to IL-8, but not dexamethasone. In contrast, a significant reduction in neutrophil ROS generation in response to phorbol-13-myristate-12 acetate (PMA) was observed in cows with a CC genotype when compared to those with a GG genotype. However, no differences in bactericidal activity or glutathione levels were observed among genotypes. The functional activity of neutrophils from cows heterozygous for this polymorphism was intermediate between those with homozygous genotypes for those assays where differences were observed among homozygous genotypes. In summary, our results suggest that neutrophils from Holstein cows with different CXCR2 genotypes vary in their ability to suppress apoptosis and produce ROS. These differences have the potential to influence overall neutrophil function and may partially explain the variation observed with respect to mastitis in vivo. These results provide a foundation for future research aimed at better understanding the basic differences between dairy cows genetically more or less susceptible to mastitis and has the potential to provide novel preventive and therapeutic measures against inflammatory diseases such as mastitis. 相似文献

OBJECTIVE: To investigate the effects of short-chain fatty acids (SCFAs) and pH on neutrophil oxidative burst, phagocytosis, and morphology after exposure to acetate, propionate, butyrate, or succinate at pH 5.5 and 6.7. SAMPLE POPULATION: Neutrophils isolated from bovine blood samples and Porphyromonas levii, Prevotella spp, and Bacteroides fragilis isolated from lesions of cattle with acute interdigital phlegmon (foot rot). PROCEDURES: Bacteria were cultured in strictly anaerobic conditions. Bacterial SCFA production was measured with high-performance liquid chromatography. Neutrophils were isolated, stimulated with phorbol 12-myristate 13-acetate (PMA) or opsonized zymosan (OZ), and incubated with dihydroethidium or dichlorofluorescein diacetate to measure production of O(2)and H(2)O(2), respectively. Phagocytosis was assessed after exposure to serum-opsonized bacteria. Cellular morphology was assessed with differential staining. RESULTS: All bacteria produced at least 3 of the 4 SCFAs. Production of both O(2) and H(2)O(2) was markedly curtailed in PMA-stimulated neutrophils exposed to SCFA at pH 5.5, compared with production at pH 6.7. Succinate caused a significant dose-dependent decrease in O(2) production at pH 6.7 in OZ-stimulated neutrophils. Monoprotic SCFAs elicited a significant increase in H(2)O(2) production in OZ-stimulated neutrophils at pH 6.7 but a significant decrease at pH 5.5. Monoprotic SCFAs significantly increased phagocytosis at pH 6.7 but decreased phagocytic activity at pH 5.5. Cellular necrosis was observed in cells exposed to SCFAs at pH 5.5. CONCLUSIONS AND CLINICAL RELEVANCE: Establishment and persistence of anaerobic bacteria in cattle with foot rot infection may result in part from neutrophil dysfunction secondary to the effects of bacterially secreted SCFA in acidotic microenvironments. 相似文献

In two experiments with growing-finishing pigs six different dietary fats were added to a conventional diet (control--C) to study the effects of dietary monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA) on the fatty acid composition of backfat and kidney fat at similar amounts of double bonds in feed (Exp. 1:7% pork fat--PF, 4.95% olive oil--OO, 3.17% soybean oil--SO) or a constant amount of 5% of processed fats (Exp. 2: partially hydrogenated fat--SAT, fractionated pork fats: olein--OLE, stearin--STE). Compared with the control, PUFA were only slightly increased in backfat of pigs fed PF, OLE, STE or OO, although dietary PUFA intake was up to 70% higher. With SO PUFA were significantly increased in adipose tissues, predominantly at the expense of MUFA. Consequently, a non-linear relationship was found between PUFA intake and proportion in backfat. MUFA were incorporated at the expense of SFA, therefore, adipose tissues of OO fed animals were lowest in SFA. Despite comparable amounts of double bonds in feed (Exp. 1), the degree of unsaturation measured as fat score (sum of double bonds) was in the order SO > OO > PF > C. In contrast, the proportion of SFA was C > PF = SO > OO. Regarding the decisive role of SFA for fat consistency it may be concluded that MUFA should also be considered in feeding recommendations for pigs. Furthermore, in case of a high dietary supply of MUFA, a simple index of double bonds might not be sufficiently conclusive to judge pig fat quality. 相似文献
