The influence of training on blood lactate concentrations during treadmill exercise and a 40-minute inactive recovery period was examined in seven trained and seven detrained thorough-bred horses. Lactate concentrations were measured in venous blood collected at the end of each exercise state, and at intervals for 40 minutes afterwards. Measurements were made of maximum oxygen uptake (V̇O2max, ml kg−1 min−1), VLA4 (velocity at which blood lactate concentration was 4 mmol litre−1); LA8 (lactate concentration [mmol litre−1] during exercise at 8 m sec−1), peak lactate (highest lactate concentration after exercise), LA40 (lactate concentration 40 minutes after exercise), the time of peak lactate concentration (minutes after exercise) and the rate of disappearance of blood lactate (Rtd). The trained horses had a significantly lower LA8 (2·1 ± 0·1 vs 6·5 ± 1 mmol litre−1, P<0·01), higher VLA4 (9·8 ± 0·2 vs 5·8 ± 0·6 m sec−1, P<0·01) and higher V̇02max (156·3 ± 3·8 vs 107·1 ± 3·9 ml kg−1 min−1, P<0·001). The value of Rtd and the time of peak lactate concentration were not significantly different.  相似文献

The effects of feeding diets with different milliequivalents (mEq) of dietary ([Na+ + K+] − [Cl + SO4=]) to dairy cows during the last seven weeks of pregnancy on bone morphology at parturition were studied. Nine monozygotic twin pairs of pregnant cows (five pairs of parity 1 or 2 and four pairs of parity 3 or more) were allocated to two diets which were formulated to provide either −4 mEq (anion diet) or +572·5 mEq (cation diet) of ([Na+ + K+] − [Cl + SO4=]) kg−1 dietary dry matter. Bone biopsies were taken from the tuber coxae between three and eight hours after parturition. The plasma concentrations of calcium and inorganic phosphorus, the total plasma alkaline phosphatase activity and the urinary hydroxyproline:creatinine ratio were not significantly affected by diet during the experimental period. In low parity (2 or less) cows the percentage trabecular bone volume, the percentage osteoclast surface and the mean number of osteoclasts per microscopic field (identified by Goldner staining) were lower on the anion diet than on the cation diet (P<0-02). In the high parity cows, the percentage osteoid volume (P<0·05) and the ratio of percentage osteoid volume to percentage osteoid surface (P<0·001) were greater in the cows fed the anion diet than in the cows fed the cation diet. The results show that reducing the mEq of dietary ([Na+ + K+] − [Cl + SO4=]) to −4 mEq kg−1 dietary dry matter affected some of the parameters of bone formation but did not enhance bone resorption.  相似文献

These studies were undertaken to examine the systemic and renal effects of the pharmacological inhibition of endothelium-derived nitric oxide (EDNO) in cats. In six healthy cats, the intravenous infusion of nitro-L-arginine at a dose of 100 μg kg−1 bodyweight min−1 resulted in a marked increase (P<0·001) in mean arterial pressure from the control value of 116·7 ± 4·6 mmHg to 154·2 ± 6·8 mmHg and an increase (P<0·05) in renal vascular resistance from the control value of 3·69 ± 0·33 mmHg min ml−1 to 6·83 ± 1·15 mmHg min ml−1. The increase in renal vascular resistance was generalised, with comparable increments in preglomerular and postglomerular vascular resistance. Mean values for glomerular capillary pressure (61·1 ± 61·9 vs 1·9 ± 1·6 mmHg), calculated from the sum of arterial colloid osmotic pressure plus proximal tubule stop-flow pressure, did not change in response to the infusion of nitro-L-arginine. However, there was a marked reduction in renal blood flow (29·4 ± 3·1 to 16·9 ± 2·3 ml min−1, P<0·01) and glomerular filtration rate (5·22 ± 0·57 to 3·52 ± 0·45 ml min−1, P<0·01). These results provide evidence that EDNO plays an important role in the basal regulation of systemic arterial blood pressure and renal haemodynamics in cats.  相似文献

Background

Canine pyometra is a life-threatening disease common in countries where spaying of dogs is not routinely performed. The disease is associated with endotoxemia, sepsis, systemic inflammatory response syndrome (SIRS) and a 3–4% mortality rate. Blood lactate analysis is clinically valuable in predicting prognosis and survival, evaluating tissue perfusion and treatment response in human and veterinary critical care settings. The aims of the present study were to investigate 1) the blood lactate levels of female dogs with pyometra by a hand-held analyser and 2) if these levels are related with the clinical status or other biochemical or hematological disorders.

Methods

In total 31 female dogs with pyometra admitted for surgical ovariohysterectomy and 16 healthy female control dogs were included in the present study. A complete physical examination including SIRS-status determination was performed. Blood samples for lactate concentrations, hematological and biochemical parameters, acid-base and blood gas analysis and other laboratory parameters were collected and subsequently analysed. The diagnosis pyometra was verified with histopathological examination of the uterus and ovaries. Increased hospitalisation length and presence of SIRS were used as indicators of outcome.

Results

In the pyometra group the median blood lactate level was 1,6 mmol l-1 (range <0.8–2.7 mmol l-1). In the control group the median lactate level was 1,2 mmol l-1 (range <0.8–2.1 mmol l-1). Of the 31 bitches 19 (61%) fulfilled 2 or more criteria for SIRS at inclusion, 10 bitches (32%) fulfilled 3 of the SIRS criteria whereas none accomplished more than 3 criteria. Lactate levels did not differ significantly between the pyometra and control group, or between the SIRS positive and SIRS negative dogs with pyometra. Increased lactate concentration (>2.5 mmol l-1) was demonstrated in one female dog with pyometra (3%), and was not associated with longer hospitalisation or presence of SIRS. Lactate measurement was not indicative of peritonitis. None of the bitches died during or within two months of the hospital stay. The measurements of temperature, heart rate, respiratory rate, percentage bandforms of neutrophilic granulocytes, α2-globulins, creatinin, pvCO2, TCO2 and base excess showed significant differences between the SIRS positive and the SIRS negative pyometra cases.

Conclusion

Increased blood lactate concentrations were demonstrated in 3% (1/31), and SIRS was present in 61% (19/31) of the female dogs with pyometra. Preoperative lactate levels were not related with presence of SIRS or prolonged hospitalisation. Lactate measurement was not indicative of peritonitis. The value of a single and repeated lactate analysis in more severely affected cases remains to be determined.  相似文献

Near infrared spectroscopy (NIBS) was used to detect changes in brain oxygenation in five tranquilised calves that were placed on a hypoxic gas mixture (10 per cent O2/90 per cent N2) and hyperoxic gas mixture (30 per cent O2/70 per cent N2) for five minutes at each concentration. A NIRO 500 (Hamamatsu, Japan) was used for the NIBS, with the incident light source and separate detector (optodes) placed on shaved skin on the most dorsal surface of the frontal bone. Sequential arterial blood gas sample analyses provided confirmation of the appropriate change in systemic oxygenation status. By the end of the five-minute-period of breathing 10 per cent oxygen, NIBS of the calf head detected highly significant changes in haemoglobin oxygenation reflective of hypoxaemia, with oxyhaemoglobin decreasing by 23·5 units (P<0·01) and deoxyhaemoglobin increasing by 45·6 units, (P<0·01) from the baseline of breathing room air. Total haemoglobin (oxyhaemoglobin + deoxyhaemoglobin) showed a significant increase of 22·1 units (P<0·05) but there was no significant change in LAIRS determined cytochrome aa3 oxygenation. Concomitant blood gas alterations included significant decreases in Pa)2 (−27·8 mmHg, P<0·01), haemoglobin saturation (−29·0 per cent, P<0·05), and PaCO2 (−7·8 mmHg, P<0·05) and significantly increased blood pH (0·059, P<0·05). At the end of the five minutes of breathing 30 per cent oxygen NIBS of the calf head detected significantly increased oxyhaemoglobin (13·1 units, P<0·01) and decreased deoxyhaemoglobin (−13·7 units, P<0·05) when compared with baseline breathing of room aim. Total haemoglobin and cytochrome aa3 were unchanged from baseline. The accompanying arterial blood gas changes included significant increases in PaO2 (30·9 mmHg, P<0·05), arterial O2 saturation (11·7 per cent, P<0·05), and significantly decreased pH (−0·026, P<0·05), This study showed that NIRS can be used to continuously and non-invasively detect cerebral oxygenation changes in the live calf in response to both increased and decreased systemic arterial oxygen. Additionally, despite induction of profound hypoxaemia, cytochrome aa3 in the brain did not appear to become reduced.  相似文献

This study characterised the peripheral plasma concentration of PSP-60 throughout gestation, and examined the effect of stage of gestation and foetal number on this protein in Holstein cows after non-surgical embryo transfer. Cows (n=12) were divided into two groups; Group 1 contained single embryo recipient cows (n=5), Group 2 contained twin-embryo recipient cows (n=7). Blood was collected approximately every third day from day 0 (first day of standing oestrus), then daily for the last 10 days of gestation and until one day post-partum. Two of the twin-embryo recipient cows had abnormal pregnancies, consequently data from them was considered separately. In both groups PSP-60 increased progressively from about day 20 post-oestrus to 20 days pre-partum (from 0·9 ± 0·2 to 49·7 ± 8·7 ng ml−1, and from 1·3 ± 0·6 to 115 ± 34·9 ng ml−1 (mean ± SEM), in singleton and twin-bearing groups, respectively). The mean concentrations between 20 and 10 days pre-partum increased dramatically by about six-fold (P<0·001) in singleton-bearing cows (from 49·7 ± 8·7 ng ml−1 to 2838 ± 73·7 ng ml−1) to over two-fold in twin-bearing cows (from 115 ± 34·9 ng ml−1 to 284 ± 98·2 ng ml-1). The mean concentrations of the two groups were indistinguishable between 10 days pre-parturn and parturition. Cows giving birth prematurely to stillborn calves or to a schistosomus reflexus calf exhibited abnormal PSP-60 profiles. Our findings indicate that peripheral plasma PSP-60 concentrations are correlated to the stage of gestation and foetal number, and assist in predicting foeto-placental viability.  相似文献

Forty multiparous Holstein dry cows on d 21 prepartum were randomly allocated to four blocks of 10 cows to examine the effects of reducing the dietary cation–anion difference (DCAD) on calcium homeostasis, acid–base balance, health status, and subsequent lactation performance. The reduced DCADs (Na + K − Cl − S, mEq/kg DM) of + 150,+ 50, − 50, and − 150 were obtained by addition of anionic salts. Reducing DCAD resulted in mild metabolic acidosis as indicated by the sharp decline in urinary pH, and minor reductions in blood pH and HCO3 concentration. Greater plasma calcium concentration was observed in cows fed diets of − 50 and − 150 DCAD (< 0.05) than those on + 50 and + 150 DCAD diets. The nadir of plasma calcium level on the day of calving was lower (< 0.05) than the highest level on d 14 prepartum (8.33 vs. 9.30 mg/dL). Composite colostrum calcium concentration was decreased (< 0.05) with time on d 1 relative to d 2 postpartum (0.212 vs. 0.174%), and feeding of diet − 150 DCAD induced higher (< 0.05) composite colostrum calcium content than other three DCAD diets. No case of milk fever occurred for any diets, but feeding the two negative DCAD diets reduced (< 0.05) retained placenta incidence compared with diet of + 150 DCAD. Dry matter intake, milk yield and compositions of fat, protein, and lactose were non-significantly affected (> 0.05) by dietary treatments. In conclusion, urinary pH is an effective indicator of extracellular fluid acid–base balance, and feeding negative DCAD in late gestation period is beneficial for dairy cows in blood calcium homeostasis and improvement of health status.  相似文献

The aim of the experiment was to study the relationship between plasma lactate and allantoin accumulation in horses undergoing five exercises differing in intensity and length. Twenty-five adult trotter horses were used (18 males, two castrated, and five females), housed in three training centers. The horses were assigned to five groups: slow trot, over 2000 m (Group 1); slow trot over 1600 m (Group 2); fast trot over 1600 m (Group 3); fast trot over 2000 m (Group 4); fast trot over 2400 m (Group 5). Plasma was obtained from blood sampled at rest, at the end of the bout of exercise and after 15 and 45 minutes from the end of the bout of exercise and analyzed for glucose, lactate, uric acid, free fatty acids (FFA) and allantoin concentrations. Accumulations of plasma lactate and allantoin (mmol/sec) were calculated as difference between end of exercise and rest and between 45 minutes sample and rest, respectively.Ranking the intensity of exercise using the lactate concentrations at the end of exercise, the level of exertion was highest for Group 3 horses and lowest for Group 5 horses (20.9 and 2.8 mmol/l, respectively). At the end of exercise, glucose concentrations were much higher for horses undertaking the more intensive exercise (Groups 3 and 4 compared to Group 2). FFA concentrations were highest at the end of exercise for Groups 2 and 3 and after 15 minutes for Groups 4 and 5. Plasma uric acid and allantoin concentrations peaked 15 and 45 minutes from the end of exercise, respectively, independently of exercise intensity. The relationship between accumulation of plasma allantoin (y, dependent variables) and lactate (x, independent variable) was non-linear: y=0.15−2.61*x+68.3*x2 (r2=0.900; se=0.19). This suggests that allantoin accumulation could be used together with plasma lactate to calibrate the workload to muscle conditions to prevent muscle injury.  相似文献

Lactate kinetics in whole blood of horses was investigated after exercise of differing velocities and duration. The following categories of exercise were used: A: <11 m/second and >180 seconds (n=35), B: >11 m/second and <180 seconds (n=17) and C: <11 m/second and <180 s (n=10). The mean peak lactate concentration determined in horses in category A was 4.49 ± 2.21 mmol/1, in B, 16.32 ± 4.81 mmoVl and in C, 4.58 ± 1.59 mmol/l. While the maximum lactate concentrations in categories A and C were always found immediately after the exercise, the peaks in category B were measured between the first and tenth minute after exercise. Mean lactate concentrations measured at 2-minute intervals after bouts of category-B exercise tended to stabilize 3 to 10 minutes after exercise; however, mean lactate concentrations measured during the intervals before and after the peak value differed significantly. The lactate concentration returned to pre-exercise levels within 20 minutes after exercise bouts of category C, but remained above pre-exercise levels up to 60 minutes after bouts of category-A and -B exercise. It was concluded that, for an evaluation of lactate data after intensive anaerobic exercise, sequential blood sampling at 2-minute intervals for a period of up to 12 minutes after exercise is necessary. Less frequent sampling may be a reason for the often described irreproducibility of lactate concentrations in horses. After aerobic or mild anaerobic exercise, one sample is sufficient, but it has to be taken as soon as possible after exercise.  相似文献

Endotoxaemia is a leading cause of death among horses. Thrombocytopenia is a common finding in clinical and experimentally-induced cases of endotoxaemia and can lead to coagulopathies, including disseminated intravascular coagulopathy which is usually fatal. In this study it was shown that endotoxin (3 ng ml−1 to 25 μg ml−1) can aggregate equine platelets in heparinised whole blood in vitro. The endotoxin-induced aggregation (EIA) was shown to be dependent on the presence of leucocytes in the blood and did not occur when detoxified endotoxin was used, suggesting that lipid A was necessary for the response. Aspirin (1 mmol litre−1) had no effect on EIA whereas apyrase (40 μg ml−1) completely abolished it and CV3988 (3 to 30 μmol litre−1) (a competitive antagonist of platelet-activating factor) inhibited the response in a concentration-dependent manner. It is concluded that endotoxin activates equine platelets at low concentrations through an indirect mechanism that involves calcium, leucocytes, adenine nucleotides and platelet-activating factor.  相似文献

The study aimed to evaluate the effect of feeding Borrena hirticulata (BH), Ficus hirta (FH), rice straw (RS) and concentrate-based total mixed ration (TMR) on nutrient utilization, rumen fermentation and growth in mithun. Growing male mithun calves were randomly allotted to 2 feeding groups (6 in each), TMR1 and TMR2. The TMRs consisted of RS 300 g kg− 1, concentrate 400 g kg− 1 and BH 300 g kg− 1 (TMR1) or FH 300 g kg− 1 (TMR2) on a dry matter (DM) basis. Both TMRs were fed ad libitum to the animals for 121 d and a digestibility study was conducted during the last 7 d of the experiment. To assess rumen fermentation, rumen fluid was collected at 2 h interval for 24 h. Apparent digestibility of DM, crude protein (CP) and crude fibre (CF) did not differ significantly between the TMRs. Nevertheless, apparent digestibility of ether extract was found to be significantly (< 0.01) greater in TMR2 (0.59) compared to TMR1 (0.54). Body weight gain (BWG; g d− 1), DM intake (kg d− 1), CP intake (g d− 1) and feed efficiency (kg feed kg− 1 gain) were found to be significantly (P < 0.05) greater in TMR1 (548, 5.14, 713 and 9.28) compared to TMR2 (496, 4.91, 703 and 10.03). An insignificant positive association (r = 0.35) between DM intake and BWG, but a significant (P < 0.01) positive association (r = 0.74) between CP intake and BWG were evident. Rumen pH (5.71 to 7.18) and ammonia-nitrogen (8.0 to 25.0 mg/dl) did not differ significantly between the TMRs, but differed significantly (P < 0.01) at different h post-feeding. In contrast, rumen total volatile fatty acid (42 to 105 mM) and total nitrogen (40.4 to 90.3 mg/dl) differed significantly (P < 0.05) between the TMRs and at different h post-feeding. The study revealed that BH, FH, RS and concentrate-based TMRs may be fed to mithun for satisfactory growth.  相似文献

Blood acid-base and electrolyte status was studied in four sedentary Miniature Horses treated with 200, 300, 400 and 500 mg of sodium bicarbonate (NaHCO3) per kg of body weight (BW). Arterial blood was collected before treatment with NaHC03 and each hour for 5 h after treatment. All treatments resulted in an increase in blood pH, bicarbonate (HCO3) concentration and base excess (BE) by 1 h post-dosage, which continued through the 5th hour (P < .05). Treatment with 200 mg NaHC03/kg BW resulted in less elevated blood HCO3 concentrations (P < .03) and BE values (P < .01) when compared to the other treatments. Following dosing with NaHCO3, plasma Na+ concentrations increased among all treatments but declined to initial values by 3 h post-treatment. The 200 mg NaHCO3/kg BW dosage resulted in the smallest increases in plasma Na+ concentrations (P < .03). Both plasma K+ and Ca++ concentrations were lower (P < .05) among all treatment groups 1 h post-dosage but returned to initial values by 5 h and 3 h posttreatment, respectively, with no differences (P >.05) among treatments. All NaHCO3 dosages increased blood buffering capacity as indicated by increased blood pH, HCO3 concentration and BE. Maximum blood pH, HCO3 concentration and BE was reached using a dosage of 300 mg NaHCO3/kg BW. Also, all treatments altered the plasma electrolyte concentrations.  相似文献

Short chain fatty acids (SCFA) are the main products of indigestible carbohydrates undergoing bacterial fermentation in the hindgut, which are related to some physiological functions. This study was designed to investigate the effects of SCFA infusion by ileum on the carcass traits, meat quality and lipid metabolism of growing pigs. In a 28-day study, 24 growing barrows fitted with a T-cannula in distal ileum were divided into 4 treatments: 1) Control, 2) antibiotics (AB), 3) AB + 300 mL of SCFA1 solution (ABS1), 4) AB + 300 mL of SCFA2 solution (ABS2). The concentrations of acetate, propionate and butyrate in SCFA1 solution were respectively 61.84, 18.62 and 12.55 mmol/L, and in SCFA2 were respectively 40.08, 15.41 and 9.78 mmol/L. The results showed that the SCFA infusion increased the average daily feed intake and average daily gain of pigs (P < 0.05). Meanwhile, the SCFA treatments increased longissimus dorsi area (P < 0.05) and carcass weight (P = 0.058), decreased the drip loss of longissimus dorsi (P = 0.059), and reduced serum concentrations of triglyceride, total cholesterol and urea nitrogen (P < 0.05). Besides, the SCFA administration inhibited the mRNA expressions of fatty acid synthase (FAS) and acetyl-CoA carboxylase in longissimus dorsi (P < 0.05), the mRNA expression of FAS in the liver (P < 0.05), and the mRNA expression of hormone-sensitive lipase in abdominal fat (P < 0.05). Short chain fatty acid infusion also enhanced the mRNA expression of carnitine palmitoyltransferase-1α in the liver (P < 0.05), the mRNA expressions of peroxisome proliferator activated receptor gamma and lipoprotein lipase in abdominal fat (P < 0.05), and the mRNA expressions of free fatty acid receptor 2, glucose-6-phosphatase and phosphoenolpyruvate carboxykinase 1 in the colon (P < 0.05). These results suggested that SCFA administration in the ileum could improve the carcass traits and meat quality of growing pigs, which was possibly due to the fact that SCFA modulated lipid metabolism.  相似文献

The aim of this study was to examine whether increased frequency of luteinizing hormone (LH) pulses influences luteal progesterone (P4) secretion by measuring progesterone concentrations at the secreted (caudal vena cava) and circulating levels (jugular vein) in lactating dairy cows. Cows received six intravenous administrations of 2.5 μg of GnRH (gonadorelin acetate, n=4) or 2 ml saline (n=3) at 1-h intervals on 12.4 ± 0.4 (mean ± SE) days after ovulation. Blood samples were collected from the caudal vena cava and jugular vein every 12 min for 12 h (6 h before and after treatment). During the 6 h after treatment, frequency of LH pulses (5.3 ± 0.3 and 3.0 ± 0.0 pulses/6 h) and mean LH concentration (0.50 ± 0.06 and 0.38 ± 0.05 ng/ml) were greater (P<0.05) in GnRH-treated cows than in saline-treated cows. Mean P4 concentration and amplitude of P4 pulses in the caudal vena cava during the 6 h after treatment were greater (P<0.05) in GnRH-treated cows than in saline-treated cows, but the frequency of P4 pulses was not different between the groups. Mean P4 concentration in the jugular vein during the 6 h after treatment was also higher (P<0.05) in GnRH-treated cows than in saline-treated cows (7.0 ± 1.3 and 5.4 ± 0.9 ng/ml). These results indicate that the increased frequency of LH pulses stimulates progesterone secretion from the functional corpus luteum and brings about higher P4 concentrations in the circulating blood in lactating dairy cows.  相似文献

Muscle ATP loss with exercise has implications both to the causes of fatigue and muscle damage. To study this at the single muscle fibre level, five trained thoroughbred horses performed consecutive 90 second gallops on an inclined treadmill followed by a final gallop to fatigue. Biopsies of the m. gluteus medius were taken at rest, post-exercise and during 24 hour recovery. Blood lactate was 20·0 mmol litre−1 or more, and plasma NH3 300–800 μmol litre−1, following the final gallop. Minimal changes occurred in the plasma markers, and . ATP loss with exercise was 32·2 ( 12·2) per cent. Following exercise single fibre contents showed a much broader distribution than at rest, with contents in some close to zero. Following five and 24 hour recovery, however, frequency distribution curves were close to those seen at rest. There was no difference in the ATP contents of types I, IIa and IIb at rest of with exercise or recovery. The results pointed to marked heterogeneity between individual fibres in their biochemical response with exercise, independent of fibre type.  相似文献

The aim of the study was to investigate the effect of duration of feeding linseed (rich in n-3 PUFA) on peroxisome proliferator-activated receptor γ (PPARγ) and tumor necrosis factor (TNF-α) gene expression, and muscle mass of growing–finishing barrows. Two isoenergetic and isonitrogenous diets were formulated, and one of which was the basal diet and another one was the linseed diet including linseed at the level of 10%. Twenty-four Landrace × Yorkshire barrows weighing 35 ± 3.7 kg were randomly assigned to four treatments with six individuals per treatment. Pigs in treatment 1 (T1) fed the control diet throughout the experimental period, while pigs in T2, T3 and T4 fed the control diet except for 30, 60, and 90 d prior to slaughter when the linseed diet were fed. The experiment was conducted for 90 days. The longissimus muscle mass and each muscle mass in the hind leg were weighted. PPARγ and TNF-α mRNA expression levels in muscle, spleen and adipose tissue, and plasma concentrations of TNF-α data were measured and analyzed. The results showed that the longissimus muscle mass, quadriceps femoris muscle mass and semitendinosus muscle mass increased linearly (< 0.01) as prolonged the time of feeding linseed diet. The expression of PPARγ in longissimus muscle and spleen increased (< 0.01) linearly as prolonged the time of feeding linseed diet, while the expression of PPARγ in adipose tissue were not affected (= 0.095). Duration of linseed addition linearly decreased (< 0.01) TNF-α gene expression levels in the longissimus dorsi muscle, adipose and spleen, and serum concentration of TNF-α as well. The expression levels of PPARγ negatively correlated with the expression of TNF-α in muscle (R2 = 0.70, < 0.001) and spleen (R2R2 = 0.77, < 0.001) respectively. Likewise, PPARγ expression level in spleen (R2R2 = 0.59, < 0.01) or muscle (R2R2 = 0.52, < 0.05) negative correlated with serum TNF-α concentration, while there were significant quadratic relation between muscular PPARγ (R2R2 = 0.80, < 0.01) or muscular TNF-α (R2R= 0.87, < 0.01) expression and the longissimus dorsi muscle mass. These data suggested that duration of feeding linseed diet lead to a linear decrease of TNF-α gene expression, which may increase the muscle mass in growing–finishing barrows, at least in part, through a PPARγ-dependent mechanism.  相似文献

The purpose of this, trial was to determine the effect of hyperkalemic periodic paralysis (HYPP) on exercise tolerance in Quarter Horses. Five HYPP affected and five nonaffected horses were matched for age, size, gender and reproductive status. HYPP status was diagnosed by DNA analysis and potassium chloride challenge testing. Plasma lactate concentration and heart rate were used as indicators of work intensity. Serum potassium concentrations were also monitored. Two exercise experiments were conducted, the first being forty-five minutes of slow, aerobic exercise (hacking) and the other being moderate, partially anaerobic exercise (galloping). Post-exercise the horses were cooled out by randomly assigning them to either forty minutes, of standing still or forty minutes of walking. Heart rates of HYPP affected and unaffected horses were not significantly different during exercise or recovery. Plasma lactate concentrations changed slightly following slow exercise and were significantly higher for HYPP affected horses (P=0.01).At the end of exercise, values were 1.4±0.2 mmol/L and 1.0±0.1 mmol/L for HYPP affected and unaffected horses, respectively. Following moderate exercise, plasma lactate concentrations were much greater, and the difference (P<0.001) between affected and unaffected horses was more marked: immediately following exercise concentrations were 10.6±1.8 and 6.2±1.0 mmol/L in affected and unaffected horses, respectively. The higher post-exercise plasma lactate concentrations in affected horses indicates increased anaerobic muscle metabolism. Serum potassium concentrations rose following exercise and significantly higher values were seen in horses that were walked rather than stood still post-exercise.Hyperkalemic periodic paralysis (HYPP) is a dominant autosomal genetic defect occurring in American Quarter Horses and related breeds. 1–5 The condition is widely geographically distributed and has been estimated to affect 0.4% of all Quarter Horses.6 Signs include sporadic attacks of muscle fasciculation, muscle spasm, sweating and weakness. Oral administration of potassium chloride produces a more severe hyperkalemia in HYPP affected horses and induces clinical attacks.1 Electromyography reveals widespread continuous, spontaneous, muscle contraction.2 Some people believe that HYPP affected horses are suitable for riding7; others have expressed doubts about the safety of this practice and the exercise tolerance of affected horses. HYPP affected horses have an unstable muscle membrane potential causing random muscle fiber contractions,2,8 which could antagonize purposeful movement. This, in combination with increases in extracellular potassium concentration occurring during exercise,9–11 may cause affected horses to stumble or collapse while being ridden, posing a danger to both horse and rider. In HYPP affected people, potassium concentration rises approximately one to two hours after exercise during which time HYPP attacks can occur.9,10 For these reasons, the exercise tolerance of HYPP affected horses and the effects of exercise on spontaneous HYPP attacks deserves investigation.We standardized the exercise test and chose heart rate and plasma lactate concentrations as indicatoors, of exercise tolerance and energy metabolism.12 Heart rate is the major determinant of oxygen delivery to muscle and the rate rises with exercise intensity until it reaches a plateau at high velocities.13,14 Several lines of investigation indicate that plasma lactate concentrations reflect muscle work. Lactate release from resting muscle is minimal but large amounts are released during strenuous exercise.15–17 Lactate release increases with increasing muscle work.16,18 During exercise, muscle oxygen consumption increases until eventually a plateau is reached after which no further increase in oxygen consumption in response to increased work loads is possible.18 At this point the mitochondrial electron chain transport system is operating at its maximal possible rate, energy production can only be supplemented by the use of inefficient anerobic glycolytic metabolism and the muscle starts to release lactate. If the concentrations of plasma lactate are high, the muscles are depending on anaerobic metabolism and are working closer to exhaustion16,18–21The objectives of this experiment were twofold. One was to determine if HYPP adversely affects exercise performance. Serum testosterone concentrations and the spermiograms of prepuberal bulls fed two levels of protein diets were investigated at 7, 10, 14 and 18 months of age. Scrotal circumference, body condition score and total sperm counts of those animals on a high protein diet were significantly (P <0.05) higher than those on a low protein diet. However, sperm motility, total dead sperm and abnormal sperm did not differ between the treatment groups (P <0.05). One bull fed a high protein diet had significantly higher testosterone concentrations (basal and peak) than a bull fed low protein throughout the four sampling periods (P <0.05). Testosterone concentrations, scrotal circumference, volume of semen, sperm concentration and sperm output of bulls on low and high protein diets increased significantly with age (P <0.05). Peak testosterone concentrations ranged from 1.1. ng ml−1 at 7 months to a maximum of 5.3 ng ml−1 at 18 months. The 24 h secretory patterns of testosterone were episodic, pulsatile or temporal in nature. The peaks occurred mostly in the morning hours and ranged from one to five in number. Protein intake in prepubertal bulls could have significant influence on spermiogram and testosterone production.  相似文献

To find the effect of journey duration on the stress levels of weaned piglets a combination of physiological welfare indicators was used. One hundred and thirty-six piglets were monitored in two replicates of a short (0.6 h) and a long (8.3 h) commercial transport from a rearing to a growing–finishing farm. Variables studied were haematocrit, red blood cell count (RBC), total white blood cell count (WBC), differential WBC counts, serum pig's major acute phase protein (Pig-MAP), haptoglobin (Hp), cortisol, glucose, creatine phosphokinase (CPK), lactate dehydrogenase (LDH), total protein, and albumin before loading and immediately after unloading. A marked increase in leucocytes (< 0.01) and a tendency to lower eosinophil concentrations (= 0.086) were observed in those piglets transported during the short journeys. On the other hand, the decrease in lymphocytes was particularly remarkable in the long journeys (< 0.01). No increase in Hp concentrations after the transports was found, but an increase in Pig-MAP concentrations was observed at the end of the long journeys (< 0.001). Cortisol concentrations did not show significant changes in either short or long journeys. According to CPK (< 0.05) and LDH activities, some physical fatigue was detected at the end of transports, particularly in the short journeys. The decrease in haematocrit values in the short journeys (< 0.05), and the absence of variations in RBC, albumin, and total protein concentrations would indicate that the loss of body water during both transport durations was reduced. Sex did not affect the response of piglets to transport, which could be attributable to the young age of the animals. Changes in albumin concentrations (< 0.05), together with the tendencies to higher WBC (= 0.092) and neutrophil counts (= 0.076), suggest a more marked response to the stress of transport in heterozygous for the halothane gene (Nn) piglets than in homozygous negative (NN) piglets. Overall results indicate that, although transports were generally stressful, it is unclear which of the two durations was more detrimental for piglet welfare, and suggest a different stress response to transport with respect to that of older pigs.  相似文献

The objective of this experiment was to study the effects of interactions between medium quality grass silage (GS1) and maize silage (MS) as well as between low-quality grass silage (GS2) and MS on ad libitum intake, digestibility and N retention in wether sheep. Two grass silages (GS1 and GS2) were ensiled in round bales, without additives, from the primary growth of orchard grass (Dactylis glomerata L.) harvested at two different maturity stages. The study consisted of seven feeding treatments incorporating GS1, GS2 and MS fed alone and forage mixtures of GS1 and MS as well as GS2 and MS (67:33% and 33:67%, respectively, DM (dry matter) basis).Delayed harvesting lowered (P < 0.05) the crude protein (CP) concentration in GS2 compared to GS1. The DM content (g kg− 1 fresh sample) and starch concentration (g kg− 1 DM) of MS were 264 and 211, respectively.Inclusion of MS in the GS1-based ration had positive linear effects on CP and starch digestibility (P < 0.05 and P < 0.01, respectively) and N intake (P < 0.01) while a negative effect on neutral detergent fibre (NDF) and acid detergent fibre (ADF) digestibility (P < 0.05 and P < 0.01, respectively). A positive associative response of GS1 and MS was observed for DM ad libitum intake (g kg− 1 M0.75 day− 1) (quadratic, P < 0.05), CP digestibility (quadratic, P < 0.01), N intake (quadratic, P < 0.01) and N balance (quadratic, P < 0.05). Inclusion of MS into the GS2-based ration had a positive linear effect on the ration fresh matter ad libitum intake (kg day− 1 and g kg− 1 M0.75 day− 1) (P < 0.01 and P < 0.001, respectively), NDF ad libitum intake (kg day− 1 and g kg− 1 M0.75 day− 1) (P < 0.01), digestibility of DM (P < 0.01), organic matter (OM) (P < 0.01), ADF (P < 0.05), starch (P < 0.001), digestibility of OM in DM (D-value) (P < 0.001), and N intake (P < 0.01). Positive associative effects of GS2 and MS were observed on all the intake and digestibility parameters measured, N intake (quadratic, P < 0.001) and N balance (quadratic, P < 0.05). It was concluded that, as expected, a positive associative response of GS2 and MS was recorded for all the measured parameters while that of GS1 and MS for a limited number of parameters, probably due to lower quality of MS (lower starch concentration) than required for improved utilization of the GS1-based ration.  相似文献
