The palatine tonsils play a key role in initiating immune responses against antigens entering the body through the mouth. They are also replication sites of some pathogens. There is no data available about the structure of the palatine tonsils of the Egyptian water buffalo. Therefore, palatine tonsils of 14 clinically healthy buffalo bulls (2–3 years old) were examined macroscopically and microscopically using light, and transmission electron microscopes. The tonsils had an elongated kidney shape with a central invagination (tonsillar fossa) containing a single macroscopic opening leading to a small central cavity (tonsillar sinus). A number of macroscopic crypts originated from this sinus (internal crypts). Besides the tonsillar fossa, also small macroscopic crypts (external crypts) were present. The tonsils were enclosed by a thin connective tissue capsule and septa divided the tonsils into incomplete lobes. Within these encapsulated organs mucous glands were very obvious. Each crypt was highly branched and lined with stratified squamous non-keratinized epithelium. Several lymphoid cells infiltrated between the epithelial cells forming patches of lymphoepithelium. The crypt lumen contained lymphocytes, neutrophils and erythrocytes. Lymph nodules with clear germinal centers extended under the epithelial surface. Diffusely distributed lymphocytes were found in the narrow interfollicular region. High endothelial venules, interdigitating dendritic cells, macrophages and plasma cells were observed among the diffuse lymphocytes. Lymphatics filled with lymphocytes drained the tonsils.  相似文献

The total number, distribution and size of the optic axons of the buffalo (Bos bubalis) were studied in five optic nerves using light and transmission electron microscopes. Semithin sections were cut at 0.40 μm with an ultramicrotome and stained with 0.3% toluidine blue for light microscopic analysis. The ultrathin sections were cut at 70 nm, mounted on copper mesh grids (No. 200) and stained with saturated solution of uranyl acetate dihydrate and examined with the transmission electron microscope. The optic axons were assessed in 10 regions of roughly equal area. The total number of optic fibers was estimated on semithin and ultrathin sections. The measurements of the axonal diameters were made on electron micrographs taken from various regions across the optic nerve. The mean value of the total number of optic axons calculated was 1 397 099 ± 10 995 and 1 584 255 by light and electron microscopes, respectively. There was very little difference in the density of the optic fibers between the 10 regions of the optic nerve. The optic nerve area ranged from 18.2 to 22.4 mm2 (mean, 21.3 mm2). The diameters of the optic axons ranged from 0.5 to 12.3 μm (mean, 3.3 μm).  相似文献

The present study was planned to investigate the prevalence of Sarcocystis spp. among slaughtered water buffaloes (Bubalus bubalis) at Alexandria province, Egypt. Three hundred blood samples were collected from slaughtered buffaloes (5–7 years old). Two techniques were used to evaluate the seroprevalence of Sarcocystis spp., enzyme-linked immunosorbent assay (ELISA) and indirect haemagglutination assay (IHA). It was revealed that 203 (67.6 %) and 191 (63.6 %) of the tested serum samples were seropositive to Sarcocystis spp. by ELISA and IHA, respectively. The results of sensitivity and specificity of IHA relative to ELISA were 94 and 100 %, respectively. For molecular characterization of inter- and intra-species genetic polymorphism within Egyptian isolates of Sarcocystis spp. of water buffaloes, polymerase chain reaction (PCR) and polymerase chain reaction-restriction length polymorphisms (PCR-RFLPs) were performed on four macroscopic isolates. The isolates represented two different geographical regions of Egypt, Alexandria and Assuit provinces. Alexandria isolates (large and small-sized cyst of the same host) and Assuit isolates (large and small-sized cyst of the same host) were used. The 18S rDNA of the macroscopic cysts were characterized, in tandem, by four restriction endonucleases, RsaI, MboI, SspI and DraI. RsaI and MboI enzymes did not show any restriction sites for all isolates, leaving the amplified fragments without cutting. SspI showed two fragments in Alexandria and Assuit small-sized isolates cut by the enzyme at 600–700-bp fragments, while Alexandria and Assuit large-sized cysts amplicons were not digested by this enzyme. The fourth enzyme, DraI, cut the PCR product of Alexandria large-sized cysts into two fragments (420–780 bp), while Assuit large-sized amplicon was not cut. It could be concluded that there was a far distance between the two local isolates (small and large sized), but there were no differences between the large-sized isolates.  相似文献

Pre‐scapular, femoral and mesenteric lymph nodes from five buffalo calves and five buffalo bulls were studied using light and transmission electron microscopy. The nodes were surrounded with a thin capsule of dense connective tissue and smooth muscles. Subcapsular and trabecular lymphatic sinuses were lined with endothelial cells resting on a basement membrane. The cortex was formed by lymphoid follicles and inter‐follicular lymphocytes. Primary and secondary follicles were observed. The medulla was made up of medullary cords of lymphocytes separated by lymphatic sinuses. These sinuses were lined with a discontinuous epithelium and interestingly crossed by reticular fibres. High endothelial venules were found in the paracortical area. Several lymphocytes were observed infiltrating the wall of these venules. The lymph nodes of the Egyptian water buffalo showed a typical structure compared with the majority of mammals, with no age‐related structural variation.  相似文献

The distribution of retinal blood vessels in the eye of buffalo was studied macroscopically and microscopically in twenty-two eyes of healthy animals. After macroscopic observation, 12 of 22 eyes were used for histological study. Ten eyes were stained with hematoxylin and eosin and two eyes with PAS stain. The present findings revealed that the eye of the buffalo was characterized by the complex network of retinal blood vessels (holangiotic or euangiotic). The central retinal artery and vein pierced the eye through the optic disc and gave off several branches. There were four pairs of primary vessels that were named dorsal, ventral, nasal and temporal retinal arteries and veins. The veins anastmosed with each other at the optic disc to form a somewhat circle. Three patterns of the distribution of blood vessels were described. The study aimed at identifying the occurrence of hydatidosis in slaughter buffaloes and helminth eggs, particularly Taenia spp. eggs in stray dogs' fecal samples in Kathmandu Valley, Nepal. The study period was from November, 2004 to April, 2005. A total of 500 buffalo carcasses in a slaughterhouse, at the riversides and individual butchers'sites were examined for the presence of hydatid cysts. The cyst fluid was collected and microscopically examined to determine if it was fertile or not. Of the 500 carcasses examined, 10.6% had hydatid cysts. Specifically, the slaughterhouse carcasses had a 6.7% occurrence of hydatid cysts, whereas those examined at riversides and individual butcher places had 10.0% and 12.7%, respectively. Distributions of the hydatid cysts by specific organs showed single occurrence of 6.4% in lungs, 2.4% in livers and 1.8% in both, livers and lungs. In a total of 53 infected buffaloes, 58.5% were fertile and 41.5% sterile cysts. Overall, 366 faecal samples from stray dogs were collected around buffaloes slaughtering areas (slaughterhouse and individual butcher places), riversides, garbage sites and temple sites. These samples were examined by formalin-ether concentration technique to investigate the presence of different helminth eggs. The total prevalence of Taenia, hookworms, Trichuris/Capillaria,Toxocara, Spirometra and Physaloptera eggs were 12.8%, 18.3%, 19.1%, 12.8%, 1.4% and 1.9%, respectively.  相似文献

The objective of this study was to determine the prevalence of antibodies to Neospora caninum and Toxoplasma gondii among 500 cattle (Bos indicus) and 500 buffaloes (Bubalus bubalis) using the indirect fluorescent antibody test (IFAT) technique. Blood samples from were collected from water buffalo and cattle in 10 municipalities in the northern region of Brazil. The frequency of cattle and water buffaloes seropositive for Neospora caninum in Pará state, Brazil, was 55% and 44%, respectively, and the frequency of cattle and water buffaloes seropositive for Toxoplasma gondii was 52% and 39%, respectively. Seropositivity for both N. caninum and T. gondii was detected in 10.6% of the cattle samples and 14.8% of the buffalo samples. The frequency of cattle positive for N. caninum and T. gondii was significantly (p < 0.05) higher than that of buffalo in two and three provinces, respectively. Buffaloes had a lower seroprevalence for N. caninum or T. gondii in all of the provinces studied. These results suggest that both species, when exposed to the same risks for N. caninum and T. gondii infection, have a high serological prevalence. Cattle showed a higher probability of being seropositive when exposed to the same risks for N. caninum and T. gondii. Our study, which included an extensive number of blood samples, provides important epidemiological information pertinent to buffalo production in tropical countries that can be used as a basis for disease-management practices in Latin America.  相似文献

The aim of the present study was to evaluate the validity of Theileria annulata surface protein (TaSP)-ELISA, in comparison with traditional microscopic test, for the diagnosis of T. annulata infection among Egyptian baladi cattle (Bos taurus) and water buffaloes (Bubalus bubalis). Molecular confirmation of infection using T. annulata merozoite surface (Tams-1) target amplification by PCR was used as a gold standard. A total of 76 clinically suspected animals including 64 baladi cattle and 12 water buffaloes were investigated in the current study by the three methods. Based on the PCR-confirmed results, the evaluation study revealed higher sensitivity of TaSP-ELISA (72.9% and 75%) as compared to microscopic examination (58.3% and 50%) among cattle and buffaloes, respectively. On the other hand, the specificity of TaSP-ELISA in diagnosis of T. annulata infection was higher (87.5%) in baladi cattle as compared to water buffaloes (37.5%). In conclusion, TaSP-ELISA was shown to be suitable for the diagnosis of T. annulata infection in cattle under field conditions.  相似文献

Semen quality, testis size and efficiency of sperm production in Bali cattle (Bos sondaicus) and hybrids with Bos indicus and Bos taurus were determined. Mean (+/- SEM) daily sperm production per gram of testis parenchyma (DSPG) in six purebred Bali bulls was 12.2 +/- 0.7 x 10(6). F1 B sondaicus cross B taurus bulls and F1 B sondaicus cross B indicus bulls were sterile. Spermatogenesis was arrested at the late primary spermatocyte stage. In 11 B sondaicus cross B indicus crosses, mean DSPG was lower than in the purebred B sondaicus, although four (one 1/4 B sondaicus, one 3/4 B sondaicus, one 5/8 B sondaicus inter se and one 3/8 B sondaicus inter se) exhibited DSPG levels similar to the foundation stock. Semen from those crossbreeds which exhibited complete spermatogenesis was more variable in terms of spermatozoal concentration, percentage of spermatozoa exhibiting progressive motility and levels of spermatozoal abnormalities. In crossbreeds where sperm production was reduced or absent, there was seminiferous epithelial dysfunction, manifested as an increased frequency of degenerative late pachytene and diplotene primary spermatocytes and germinal cells occurring later in the cycle, or in extreme cases, as complete arrest of spermatogenesis at the late primary spermatocyte stage.  相似文献

SUMMARY Differences occur between female Bos indicus and Bos taurus cattle in various aspects of reproductive physiology and behaviour. These may be associated with different natural and human selection pressures, compounded by strong genotype-environment interactions. B indicus cattle are better adapted for tropical environments (despite overall poor cattle reproductive rates in these regions) which tend to be more stressful for B taurus genotypes. Conversely, B taurus cattle generally show superior reproductive and productive traits under more favoured, temperate conditions. Despite genotype-environment effects, B indicus females are generally considered to take longer to achieve puberty and to have longer gestation lengths, exhibit prolonged postpartum anoestrus, show greater seasonality of reproductive traits (tending to be long-day breeders), display a shorter, less overt oestrus as well as less tendency to allow riding behaviour by subordinate females. Some groups appear to have increased losses both during pregnancy and in the neonatal period. On the positive side, B indicus females respond well to managerial and nutritional interventions, tend to have greater reproductive longevity and they generally exhibit strong maternal traits. Culling of infertile females and selection for greater male scrotal circumference and sex-drive, in conjunction with the use of target weights, body condition scoring and weaning stratagems can improve reproductive rates in B indicus females.  相似文献

In the current study, milk protein variation was examined in cattle (Bos indicus), mithun (Bos frontalis), yak (Bos grunniens) and their hybrid populations in Bhutan to estimate genetic variability, conduct genetic characterization and assess the possibility of gene flow between mithun and cattle. Isoelectric focusing of 372 milk samples from 11 populations detected four molecular types of β‐lactoglobulin (A, B, E and M), five molecular types of αS1‐casein (A, B, C, E and X) and three molecular types of k‐casein (A, B and X). Mithun and yak shared alleles but were found to exhibit different allele frequencies for the proteins studied. The degree of genetic variability within populations was measured by average heterozygosity and ranged from 24–40% in cattle, 26% for yak and 33% for mithun. We also resolved the traditional mithun and cattle hybridization system via principal component analysis. Our results suggested secondary introgression of mithun genes to the village Thrabum population, and a close genetic relationship between Bhutanese indigenous cattle and Indian cattle.  相似文献

Delayed pubertal development and low fertility of Bos indicus x Bos taurus crossbred male cattle and domestic buffaloes is hardly understood hence, a sensitive enzymeimmunoassay (EIA) was developed using the second antibody-coating technique and testosterone-3-O-carboxymethyloxime-horseradish peroxidase conjugate as a label for determination of testosterone in blood plasma. The EIA was validated by standard criteria. Blood samples were collected by venipuncture from growing male cattle (Karan Fries and Sahiwal) and buffalo (Murrah) and testosterone was estimated using the EIA procedure. Plasma testosterone concentrations increased significantly (P < 0.05) with advancing age. Testosterone concentrations were significantly (P < 0.01) higher in Sahiwal males in comparison to Karan Fries males. The low testosterone levels in crossbred than Sahiwal could imply that crossbred males have either not stabilized genetically or not adapted well in Indian climatic conditions resulting in poor libido and poor semen quality. The low testosterone levels in Murrah buffalo males may be the possible reason for delayed maturity in this species. The direct, sensitive EIA validated for estimating the plasma testosterone concentration was reliable for studying the testosterone profile in blood plasma of males. The results suggest that there could be a requirement for higher testosterone secretion by males during early stages of growth for attaining early sexual maturity.  相似文献
